Description
Foreliggende oppfinnelse angår fremstilling av nye 2'-halometylidenderivater som er anvendbare som antivirale og antineoplastiske midler.
Foreliggende oppfinnelse angår således en analogifremgangsmåte for fremstilling av terapeutisk aktive forbindelser av formelen
hvori
X± er halogen,
eller farmasøytisk akseptable salter derav.
Som anvendt her, angir uttrykket "halogen" eller "halo-" et fluor-, klor-, brom- eller jodatom.
Analogifremgangsmåten ifølge oppfinnelsen er kjenne-tegnet ved at
a) et 3,5-0-tetraisopropyldisiloksan-l,3-diyl-2-ketonukleosidderivat av formel
omsettes med et arylsulfonylhalometylidenfosforylid av formel under dannelse av et 3,5-0-tetraisopropyldisiloksan-l,3-diyl-2-arylsulfonylhalometyliden-nukleosidderivat av formel b) 3,5-0-tetraisopropyldisiloksan-l,3-diyl-2-arylsulfonylhalometyliden-nukleosidderivatet av formel omsettes med et fluoridanion eller syre under dannelse av det tilsvarende 2-arylsulfonylhalometyliden-nukleosidderivat av formel c) 2-arylsulfonylhalometyliden-nukleosidderivatet av formel omsettes med et aluminium/kvikksølvamalgam, et natrium/kvikk-sølvamalgam, eller tributyltinnhydrid/azobisisobutylnitril, etterfulgt av vandig syrebehandling, og d) 2-arylsulfonylhalometyliden-nukleosidderivatet omsettes eventuelt med base.
En generell synteseprosedyre for fremstilling av forbindelser av formel (1), hvori X1 er hydrogen, er vist i reaksj onsskj emaet
I trinn a omsettes ketonderivatet (2), f.eks. 4-etoksy-1-[(3,5-0-tetraisopropyldisiloksan-l,3-diyl)-B-D-erytro-pentofuran-2-keto-osyl]-2(lH)-pyrimidon, i en Wittig-reaksjon med et fosforylid som generelt beskrevet for reaksjonsskjerna A. Når en forbindelse av formel (1) ønskes, hvori en av X1 og X2 er hydrogen, kan (2) omsettes med et arylsulfonylhalometylidenfosforylid, under dannelse av det tilsvarende 2-arylsulfonylhalometylidenderivat (5), slik som 2-fenylsul-fonylhalometylidenderivatet.
Det egnede arylsulfonylhalometylidenfosforylid kan fremstilles i henhold til prosedyrer som er velkjente innen faget. Hvor f.eks. en forbindelse av formel (1) er ønsket hvori en av Xx og X2 er hydrogen, kan det egnede keton (2) omsettes med et arylsulfonylhalometylidenfosforylid fremstilt ved omsetning av et halofosfat (slik som dietylklorfosfat) med et halometylarylsulfon i nærvær av en base (slik som litiumdiisopropylamid).
Når en forbindelse av formel (1) ønskes, hvori en av Xx og X2 er fluor og den andre er hydrogen, kan nærmere bestemt det egnede keton (2) omsettes med et arylsulfonylfluormetyl-idenfosforylid, fremstilt ved omsetning av et halofosfat (slik som dietylklorfosfat) med fluormetylarylsulfon i nærvær av en base (slik som litiumdiisopropylamid).
I trinn b fjernes den tetraisopropyldisiloksanbeskyt-tende gruppe av (5), som beskrevet for reaksjonsskjerna A (trinn b), under dannelse av det tilsvarende avblokkerte 2-arylsulfonylhalometyliden-substituerte derivat (6). Eksempelvis kan et fluoridsalt, slik som tetrabutylammoniumfluprid, anvendes for å fjerne den tetraisopropyldisiloksan-blokkerende gruppe.
I trinn c fjernes arylsulfonylgruppen av (6) og erstattes med et hydrogenatom for å tilveiebringe det tilsvarende 2-halometylidenderivat (7). Dette kan utføres i henhold til prosedyrer velkjente innen faget, slik som omsetning av (6) med et aluminium/kvikksølvamalgam eller en natrium/kvikk-sølvamalgam eller tributyltinnhydrid/azobisisobutylnitril (AIBN) i tilbakeløpskokende benzen, etterfulgt av vandig syrebehandling.
Det 2-arylsulfonylhalometyliden-substituerte derivat (6) er en ny forbindelse som er anvendbar som et kjemisk mel-lomprodukt ved syntese av forbindelser av formel (1), hvori en av Xx eller X2 er hydrogen. Betegnelsen "Ar" i forbindelsene av formel (6) angir en C6-C12-arylgruppe, som tidligere definert. De foretrukne forbindelser av formel (6) er de hvori Ar er fenyl.
Hvor et 4-alkoksy-substituert pyrimidon, slik som 4-etoksy-substituert pyrimidon, anvendes som utgangsmaterialet (2) i trinn a, kan 4-alkoksygruppen av pyrimidonbasen omdannes i trinn c til 4-ketogruppen under dannelse av det tilsvarende uridin- eller tymidinderivat (7), eller kan omdannes til 4-aminogruppen under dannelse av det tilsvarende cytidinderivat (7) . Disse reaksjoner kan utføres i henhold til prosedyrer velkjente innen faget.
Som det klart fremgår for fagmannen, eksisterer 2'-halometylidenderivatet, representert ved forbindelsen (7) i reaksjonsskjemaet, som to geometriske isomerer som kan angis som (Z)- og (E)-isomerene. Disse isomerer kan separeres under anvendelse av konvensjonelle separasjonsteknikker velkjente innen faget. Eksempelvis kan de geometriske isomerer oppløses ved kolonnekromatografi under anvendelse av en Dowex 1-X2 (0H~~ <f>orm)-harpiks.
Det etterfølgende eksempel representerer en typisk syntese som beskrevet ved reaksjonsskjemaet.
Eksempel
( Z)- og ( E)- 2'- deoksy- 2'- fluormetylidencvtidin
Trinn a: 4- etoksv- l- f( 3, 5- 0- tetraisopropyldisiloksan- l, 3-divl)- B- D- erytro- pentofuran- 2-( 2- fluor- 2- fenylsulfonylmetylid-en) osyl1- 2( 1H)- pyrimidon
Fremstill dietylfluormetylfenylsulfonylfosfonat som følger: Tilsett til en løsning av 500 mg (2,87 mmol) fluor-metylfenylsulfon i 30 ml tørr THF, som er blitt avkjølt til ca. -60°C i en tørr 3-hals kolbe på 100 ml utstyrt med røres-tav, argon innløpsventil, termometer og gummiforsegling, 500 mg (0,42 ml, 2,87 mmol) dietylklorfosfat via en sprøyte. Tilsett til denne blanding en løsning av 3,48 ml (5,74 mmol) 1,65 M litiumdiisopropylamid i cykloheksan via sprøyte, og følg dannelsen av dietylfluormetylfenylsulfonylfosfonat ved gass-væskekromatografi (GLC).
Tilsett til den ovenfor angitte dietylfluormetyl-fenylsulfonylfosfonatløsning en løsning av 732 mg (2 mmol) 4-etoksy-1-[(3,5-0-tetraisopropyldisiloksan-l,3-diyl)-2-keto-B-D-erytro-pentofuranosyl]-2(1H)-pyrimidon i 5 ml tørr THF og la reaksjonsblandingen få oppvarmes til romtemperatur over natten under en argonatmosfære. Hell blandingen over i en mettet, iskald løsning av ammoniumklorid og ekstraher blandingen 3 ganger med 75 ml etylacetat hver gang. Kombiner de organiske lag, tørk med vannfritt magnesiumsulfat og fordamp til tørr-het. Kromatografer residuet på en silikagelflashkolonne og eluer med etylacetat/heksan (1/1, v/v) under dannelse av tittelforbindelsen.
Trinn b: 4- etoksy- l- rB- D- erytro- pentofuran- 2-( 2- fluor- 2- fenyl-sulf onvlmetyliden) osvl1- 2( 1H)- pyrimidon
Tilsett til en løsning av 2,2 ml (2,2 mmol) 1,0 M tetrabutylammoniumfluorid i THF 668 mg (1 mmol) 4-etoksy-l-[(3,5-0-tetraisopropyldisiloksan-l,3-diyl)-B-D-erytro-pentofuran-2-(2-fluor-2-fenylsulfonylmetyliden)osyl]-2(1H)-pyrimidon og omrør blandingen ved romtemperatur i 2 timer. Nøytrali-ser blandingen med eddiksyre, tilsett flashsilikagel til blandingen og fordamp til tørrhet i vakuum. Anbring residuet på en flashsilikagelkolonne og eluer med kloroform/etanol (20/1, v/v) under dannelse av tittelforbindelsen.
Trinn c: ( Z)- og ( E)- 2'- deoksy- 2'- fluormetvlidencytidin
Tilsett til en løsning av 854 mg (2 mmol) 4-etoksy-l-[B-D-erytro-pentofuran-2-(2-fluor-2-fenylsulfonyImetyliden) - osyl]-2(1H)-pyrimidon i 100 ml 10 % vandig THF under en nitro-genatmosfære, aluminiumamalgam (fremstilt fra 0,04 g aluminium i 2 % vandig HgCl2). Omrør og ventiler blandingen mens den kokes under tilbakeløpskjøling i 2 timer. Filtrer blandingen og fordamp mesteparten av THF i vakuum. Ekstraher residuet 3 ganger med 25 ml etylacetat hver gang, kombiner de organiske lag og tørk med vannfritt Na2S04. Fordamp til tørrhet i vakuum og anbring residuet på en flashsilikagelkolonne og eluer med kloroform/etanol (9/1, v/v) under dannelse av (Z)- og (E)-4-etoksy-1-[B-D-erytro-pentofuran-2-(2-fluormetyliden)osyl]-2(1H)-pyrimidon, som en blanding av geometriske isomerer.
Oppvarm en løsning av 858 mg (3 mmol) (Z)- og (E)-4-etoksy-1-[B-D-erytro-pentofuran-2-(2-fluormety1iden)osyl]-2(1H)-pyrimidon i metanolisk ammoniakk (10 ml, mettet ved 0°C) i et forseglet rør til 100°C i 2 dager. Fordamp løsningen til tørrhet og separer (Z)- og (Ef-isomerene av tittelforbindelsen ved kromatografi ved å anbringe residuet på en kolonne pakket med Dowex 1-X2 (0H"-form) og eluering med metanol.
( Z)- 2'- deoksy- 2' fluormetvlidencytidin

Utgangsmaterialer for anvendelse ved den generelle synteseprosedyre vist i reaksjonsskjemaet, er lett tilgjenge-elige ved anvendelse av syntesemetoder og prosedyrer som er velkjente innen faget.
En metode for behandling av en pasient angrepet av en neoplastisk sykdomstilstand omfatter administrering dertil av en terapeutisk effektiv antineoplastisk mengde av en forbindelse av formel (1). Uttrykket "neoplastisk sykdomstilstand" som anvendt her, angir en unormal tilstand eller en tilstand karakterisert ved en hurtig formerende cellevekst eller neo-plasma. Neoplastiske sykdomstilstander, for hvilke behandling med en forbindelse av formel (1), (la) eller (lb) vil være særlig anvendbar, innbefatter: Leukemier, slik som akutt lym-foblastisk, kronisk lymfocytisk, akutt myloblastisk og kronisk mylocytisk; Karsinomer, slik som de i cervix, oesophagus, mage, tynntarm, tykktarm og lunger; Sarkomer, slik som oester-oma, osteosarcoma, lepoma, liposarcoma, hemangioma og heman-giosarcoma; Malanomer, innbefattende amelanotisk og melanot-isk; og blandetyper av neoplasiaer, slik som karsinosarkom, lymfoid vevstype, follikulær retikulum, cellesarkom og Hodgkins sykdom.
Inhiberende effekt av forbindelser på HeLa- celleproliferering
Den inhiberende effekt av forskjellige forbindelser av formel (1) på HeLa celleproliferering in vitro ble bestemt i henhold til metoden beskrevet av Sunkara, et al. [J. Nati. Cancer Insit. 70, 505-509 (1983)]. Eksponensielt voksende HeLa celler ble inkubert i nærvær av forskjellige konsentrasjoner av testforbindelsene i 96 timer. IC50 verdier ble beregnet som representerer konsentrasjonen av testforbindelsen ved 50 % inhibering av celleveksten. Resultatene av denne studie er angitt i tabell 1.

En terapeutisk effektiv antineoplastisk mengde av en forbindelse av formel (1) angir en mengde som er effektiv ved
enkel eller multippel dose-administrering til pasienten, til å regulere veksten av neoplasmen eller til å forlenge overlevel-sesevnen for pasienten utover den som er forventet i fravær av slik behandling. Som anvendt her angir "regulering av veksten" av neoplasmen en nedsettelse, avbrytelse eller stopping av
dens vekst og metastaser, og indikerer ikke nødvendigvis en total eliminering av neoplasmen.
Som anvendt her, angir uttrykket "terapeutisk effektiv mengde" en terapeutisk effektiv antineoplastisk av en forbindelse av formel (1). En terapeutisk effektiv mengde kan lett bestemmes av legen som fagmann ved anvendelse av kjente teknikker og ved observering av resultater erholdt under analoge omstendigheter. Ved bestemmelse av den terapeutisk effektive mengde eller dose, tas et utall av faktorer i betraktning av legen, innbefattende arten av pattedyr, dets størrelse, alder og generell helse; den spesifikke sykdom som er involvert, graden av, eller innbefattelsen eller streng-heten av, sykdommen; responsen hos den individuelle pasient; den bestemte forbindelse som administreres; administreringsmåte; biotilgjengelighetskarakteristikkene for det admini-strerte apparat; det valgte dosesystem; anvendelse av led-sagende medikamentering, og andre relevante omstendigheter.
En terapeutisk effektiv mengde av en forbindelse av formel (1), forventes å variere fra 0,1 mg pr. kg kroppsvekt pr. dag (mg/kg/dag) til 100 mg/kg/dag. Foretrukne mengder forventes å variere fra 0,5 til 10 mg/kg/dag.
Ved utøvelse av behandlingen av en pasient angrepet med en sykdomstilstand som beskrevet ovenfor, kan en forbindelse av formel (1), administreres i en hvilke som helst form eller på en hvilke som helst måte som gjør forbindelsen bio-tilgjengelig i effektive mengder, innbefattende oral og parenteral administrering. Eksempelvis kan forbindelsene av formel (1), administreres oralt, subkutant, intramuskulært, intravenøst, transdermalt, intranasalt, rektalt og lignende. Oral administrering er generelt foretrukket. Fagmannen vedrør-ende fremstilling av formulering, kan lett velge den riktige form og administreringsmåte, avhengig av de bestemte karakter-istika for den valgte forbindelse, den sykdomstilstand som skal behandles, graden av sykdom og andre relevante omstendigheter .
Forbindelsene kan administreres alene eller i form av et farmasøytisk preparat, eller i kombinasjon med farmasøytisk akseptable bærere eller eksipienser, hvor mengden og arten av disse bestemmes av løseligheten og de kjemiske egenskaper av den valgte forbindelse, den valgte administreringsmåte og sta-ndard farmasøytisk praksis. Selv om forbindelsen ifølge oppfinnelsen i seg selv er effektive, kan de formuleres og administreres i form av deres farmasøytisk akseptable syreaddis-jonssalter pga. av stabilitet, hensiktsmessig krystallisering, øket løselighet og lignende.
De farmasøytiske preparater fremstilles på kjent måte innen det farmasøytiske fag. Bæreren eller eksipiensen kan være et fast, halvfast eller væskeformig materiale som kan tjene som en bærer eller medium for den aktive bestanddel. Egnede bærere eller eksipienser er velkjente innen faget. Det farmasøytiske preparat kan være tilpasset for oral eller parenteral bruk, og kan administreres til pasienten i form av tabletter, kapsler, stikkpiller, løsninger, suspensjoner eller lignende.
Forbindelsene fremstilt ifølge oppfinnelsen kan administreres oralt, eksempelvis med et inert fortynnings-middel eller med en spiselig bærer. De kan være innelukket i y gelatinkapsler eller presset til tabletter. For oral terapeutisk administrering, kan forbindelsene være inkorporert med eksipienser og anvendes i form av tabletter, pastiller, kapsler, eliksirer, suspensjoner, siruper, kjeks, tyggegummi og lignende. Disse preparater skal inneholde minst 4 % av forbindelsen ifølge oppfinnelsen, den aktive bestanddel, men kan varieres avhengig av den bestemte form og kan hensiktsmessig være mellom 4 % til 70 vekt-% av enheten. Mengden av forbindelsen tilstedeværende i preparatene, er slik at en egnet dose vil bli erholdt. Foretrukne preparater ifølge oppfinnelsen fremstilles slik at en oral doseringsenhetsform inneholder mellom 5,0-300 mg av en forbindelse ifølge oppfinnelsen.
Tablettene, pillene, kapslene, pastillene og lignende, kan også inneholde en eller flere av følgende adjuvanser: bindemidler, slik som mikrokrystallinsk cellulose, gummitragant eller gelatin; eksipienser, slik som stivelse eller laktose, oppbrytende midler, slik som algininsyre, Primogel, maisstivelse og lignende; smøremidler, slik som mag-nesiumstearat eller Sterotex; glidemidler, slik som kolloidal silisiumdioksyd; og søtningsmidler, slik som sukrose eller
sakkarin kan tilsettes, eller et smaksgivende middel, slik som peppermynte, metylsalisylat eller appelsinsmak. Når doserings-enhetsformen er en kapsel, kan den i tillegg til materialer av den ovenfor angitte type, inneholde en væskeformig bærer, slik som polyetylenglykol eller en fettolje. Andre doseringsenhets-former kan inneholde forskjellige andre materialer som modifi-serer den fysikalske form av doseringsenheten, f.eks. som be-legg. Således kan tabletter eller piller, være belagt med sukker, skjellakk eller andre enteriske beleggingsmidler. En sirup kan, i tillegg til de nye forbindelser, inneholde sukrose som et søtningsmiddel og visse konserveringsmidler, fargestoffer og smaksgivende midler. Materialer anvendt ved fremstilling av disse forskjellige preparater skal være farmasøytisk rene og ikke-toksiske i de anvendte mengder.
For parenteral terapeutisk administrering, kan forbindelsene inkorporeres i en løsning eller suspensjon. Disse preparater skal inneholde minst 0,1 % av en forbindelse fremstilt ifølge oppfinnelsen, men kan varieres til å være mellom 0,1 og 50 vekt% derav. Mengden av forbindelsen tilstedeværende i slike preparater er slik at en egnet dose vil bli erholdt. Foretrukne preparater fremstilles slik at en parenteral doser-ingsenhet inneholder mellom 5,0 og 100 mg av forbindelsen.
Løsningene eller suspensjonene kan også innbefatte en eller flere av følgende adjuvanser: sterile fortynningsmidler, slik som vann for injeksjon, saltvannsløsning, herdede oljer, polyetylenglykoler, glyserol, propylenglykol eller andre syn-tetiske løsningsmidler; antibakterielle midler, slik som ben-zylalkohol eller metylparaben; antioksydanter, slik som askor-binsyre eller natriumbisulfitt; gelatinerende midler, slik som etylendiamintetraeddiksyre; buffere, slik som acetater, citra-ter eller fosfater, og midler for justering av tonisiteten, slik som natriumklorid eller dekstrose. Det parenterale preparat kan være innelukket i ampuller, engangssprøyter eller multiple doseampuller fremstilt av glass eller plast.
Claims (1)
- Analogifremgangsmåte for fremstilling av terapeutisk aktive forbindelser av formelenhvori Xx er halogen,eller farmasøytisk akseptable salter derav, karakterisert ved at a) et 3,5-0-tetraisopropyldisiloksan-l,3-diyl-2-ketonukleosidderivat av formelomsettes med et arylsulfonylhalometylidenfosforylid av formelunder dannelse av et 3,5-0-tetraisopropyldisiloksan-l,3-diyl-2-arylsulfonylhalometyliden-nukleosidderivat av formel b) 3,5-0-tetraisopropyldisiloksan-l,3-diyl-2-arylsulfonylhalometyliden-nukleosidderivatet av formelomsettes med et fluoridanion eller syre under dannelse av det tilsvarende 2-arylsulfonylhalometyliden-nukleosidderivat av formel c) 2-arylsulfonylhalometyliden-nukleosidderivatet avformelomsettes med et aluminium/kvikksølvamalgam, et natrium/kvikk-sølvamalgam, eller tributyltinnhydrid/azobisisobutylnitril, etterfulgt av vandig syrebehandling, og d) 2-arylsulfonylhalometyliden-nukleosidderivatet omsettes eventuelt med base.
