Titel: 4-Carbamoyloxy-oxazaphosphorine, Verfahren zu ihrer Herstellung und diese enthaltende pharmazeutische Zubereitungen

Aus den DE-OS 2 231 311 und 2 552 135 ist bekannt, daß man durch Einführung einer Hydroperoxygruppe -00H in die 4-Stellung des Moleküls der bekannten Cytostatika 2-^Bis-(2-chlorethyl)-amino7“2-oxo-tetrahydro-2H-1,3,2-oxaza-phosphorin (Cyclophosphamide), 3-(2-Chlorethylaminö)-2-/bis-(2*-chlorethyl)-amino/-2-oxo-tetrahydro-2H-1,3,2-oxaza-; Phosphorin (Trofosfamid), 3-(2-Chlorethylamino)-2-(2‘- ; chlorethylamino)-2-oxo-tetrahydro-2H-1,3,2-oxazaphosphorin (Ifosfamid), 3-(2-Chlorethylamino)-2-(2‘-methansulfonylethyl-! amino)-2-oxo-tetrahydro-2H-1,3,2-oxazaphosphorin (Sufosfamid) und anderer Cyclophosphamide ähnlicher Strukturformel Verbin-j düngen mit wertvollen cytostatischen Eigenschaften schaffen kann, diese Verbindungen jedoch eine so geringe Stabilität besitzen, daß ihre Verarbeitung zu pharmazeutischen Zubereitungen, wie es in der Humanmedizin notwendig ist, nicht möglich ist. Es ist daher Aufgabe der vorliegenden Erfindung, in 4-Stellung mit einer weiter abgewandelten Hydroxygruppe substituierte Cyclophosphamide zu schaffen, die sich insbesondere durch hohe cystatische Wirksamkeit und verbesserte Stabilität auszeichnen.

Eine andere Gruppe bevorzugter Verbindungen der Formel I sind diejenigen, in denen X Sauerstoff ist, und , : die gleich oder voneinander verschieden sind, Wasserstoff oder ein 2-Chlorethyl-Rest sind, R^, Rg und Rg Wasserstoff darstellen und R? Der Reaktionsablauf läßt sich gemäß den erfindungsgemäßen Verfahren dünnschichtchromatographisch verfolgen. Die Isolierung von dünnschichtchromatographisch einheitlichen - 6 - ν »-

Substanzen gelingt durch übliche Aufarbeitungsverfahren für derartige Produkte, insbesondere durch Kristallisation oder chromatographische Reinigung. Die StrukturSicherung erfolgt durch Schmelzpunkt, Dünnschichtchromatographie, Elementaranalyse und/oder IR- und ^H-NMR-Spektralanalyse,
Die im erfindungsgemäßen Verfahren als Ausgangsmaterial verwendeten Verbindungen sind weitgehend bekannt, können kristallin oder als Rohprodukt eingesetzt werden und lassen sich wie folgt in an sich bekannter Weise synthetisieren; 4-Hydroxy-oxazaphosphorine werden durch Reduktion der 4-Hydroperoxy-Derivate erhalten (z.B. A. Takamizawa u.a., J.Med.Chem. 18, 376 (1975)). 4-Methoxy- bzw. 4-Ethoxy--oxazaphosphorine bilden sich unter saurer Katalyse aus den 4-Hydroxy-Derivaten in Methanol bzw. Ethanol oder in inerten Lösungsmitteln, die Methanol bzw. Ethanol enthalten. Die Hydroxyharnstoff-Derivate werden durch die Umsetzung von entsprechend substituierten Isocyanaten bzw. Carbamidsäurechloriden mit Hydroxylamin bzw. N-monosubsti-tuierten Hydroxylaminen hergestellt. In ähnlicher Weise lassen sich die N-Hydroxy-carbamidsäureester (Hydroxy-carbamate) durch die Umsetzung der entsprechenden Chlorkohlensäureester mit Hydroxylamin bzw. N-monosubstituierten Hydroxylaminen gewinnen. Für die Synthese von Hydroxyharn-stoff- bzw. N-Hydroxycarbamidsäureestern mit einer zusätzlichen Hydroxy-, Carboxy- oder Sulfonsäure-Gruppe ist ein Syntheseweg mit Schutzgruppe empfehlenswert.
Die Oxime der Formel IV sind auf verschiedenen Synthesewegen zugänglich. So können sie durch die Umsetzung der entsprechenden 4-Hydroxy-oxazaphosphorine mit Hydroxylamin bzw. mit Hydroxylamin-Salzen erhalten werden.

Von den erfindungsgemäßen 4-Carbamoyloxy-oxazaphosphorinen lassen sich die racemischen cis- und trans-Isomeren hersteilen (cis « 2R, 4R / 2S, 4S; trans = 2R, 4S / 2S, 4R).

Die Zuordnung ist in Übereinstimmung mit den IUPAC-Nomen-klaturregeln und mit der Literatur über entsprechende Oxazaphosphorin-Derivate. Die cis- bzw, trans-Form läßt sieb je nach Wahl der Reaktionsbedingungen auch gezielt hersteilen. Pharmakologisch zeigen die Isomere keine signifikanten Unterschiede.
Die erfindungsgemäßen Verbindungen zeichnen sich durch bemerkenswerter chemotherapeutisch wertvolle Eigenschaften aus. Im Vergleich mit der vorbekannten Bezugssubstanz Cyclophosphamid besitzen sie an experimentellen Transplan-tationstumoren der Ratte eine annähernd gleich starke kanzerotoxische chemotherapeutische Wirksamkeit. Sie wirken in wässriger Lösung direkt alkyliered und besitzen in vitro eine hohe Zytotoxizität, während ' Cyclophosphamid einer enzymatischen Aktivierung bedarf und in vitro praktisch nicht zytotoxisch wirkt. Die akute Toxizität der erfindungsgemäßen Verbindungen ist ca. 4 mal geringer als die der Bezugssubstanz Cyclophosphamid, die therapeutische Breite ist damit deutlich erhöht. Auch im Hinblick auf organotoxische Nebenwirkungen, wie Leukozytendepression und Immunsuppression, weisen die erfindungsgemäßen Verbindungen deutliche Vorzüge gegenüber der BezugsSubstanz Cyclophosphamid auf.

Die erfindungsgemäßen Verbindungen sind zur Behandlung maligner Tumore und ähnlicher maligner Krankheiten wie Leukämie beim Menschen geeignet. Dabei kommen Mengen im Bereich von 0,01 - 100 mg/kg zur Anwendung. Die hierbei eingesetzten pharmazeutischen Zubereitungen werden nach üblichen Methoden unter Verwendung üblicher Zusatz- und Trägerstoffe hergestellt.

Die folgenden Beispiele dienen der weiteren Erläuterung der vorliegenden Erfindung, ohne sie hierauf zu beschränken.
Beispiel 1 2-(Bis-(2-chlorethyl)-amino)-2-oxo-tetrahydro-2H-1,3.2-oxazaphosphorin-4-yl-oxv-harnstoff 15 g (54 mmol) 4-Hydroxycyclophosphamid (d.h. 2-(Bis-(2-chlorethyl)-amino)-4-hydroxy-tetrahydro-2H-1,3,2-oxaza-phosphorin-2-oxid) und 4,4 g (58 mmol) Hydroxyharnstoff wurden in 70 ml DMF gelöst, mit TriChloressigsäure angesäuert (pH 3-4) und 20 h bei 0°C im Kühlschrank stehen gelassen. Danach wurde der Kristallbrei mit 70 ml Essigester verdünnt und nach 2 h abgesaugt, gewaschen, getrocknet und aus Methanol umkristallisiert.
Ausbeute: 11,3 g (62 % d.Th.) der cis-Form, F.: 139-143°C (Zers.)
Beispiel 2 2-(Bis-(2-chlorethyl)-amino)-2-oxo-tetrahydr0-2H-1,5.2-oxazaphosphorin-4-yl-oxv-hamstoff ♦ cis-Form 1,1 g (4,0 mmol) 4-Hydroxycyclophosphamid wurden in Methanol gelöst, mit einer Spur Trichloressigsäure versetzt, über Nacht bei -25°C stehengelassen, dann schonend Methanol abgezogen, der Rückstand in wenig Methylenchlorid aufgenommen, getrocknet und zu 1,2 g 4-Methoxycyclophosphamid (d.h. 2-(Bis-(2-chlorethyl)-amino)-4-methoxy-tetrahydro-2H-1,3,2-oxazaphosphorin-2-oxid) eingeengt. Anschließend , - 9 - ' » * %jî wurden 1,2 g 4-Methoxycyclophosphamid und 304 mg Hydroxy-j harnstoff in 3 ml DMF gelöst, und 20 h bei -25°C im Kühl- ] schrank aufbewahrt. Der Kristallbrei wurde mit 3 ml Essig- ! ester verdünnt, abgesaugt, gewaschen, getrocknet und aus
Methanol umkristallisiert.
Ausbeute: 670 mg (50 % d.Th.) des gleichen Produktes wie in Beispiel 1.
Beispiel 5 2-(Bis-(2-ohlorethyl)-amino)-2~oxo-tetrahydro-2H-1» 3 « 2- i oxazaphosphorln-4-yl-oxy-harnstoff, trans-Form 'i *s | I 16 g (58 mmol) 4-Hydroxycyclophosphamid und 5,2 g (68 mmol) j Hydroxyharnstoff wurden in 160 ml Wasser gelöst, mit Tri- ] chloressigsäure angesäuert (pH 3-4) und 20 h bei 0°C im | Kühlschrank stehengelassen. Danach wurde der Kristallbrei i abgesaugt, mit Wasser gewaschen, über ^2% lm Hochvakuum | getrocknet und aus Methanol/Chloroform umkristallisiert.
I Ausbeute: 12,7 g (65 % d.Th.) der trans-Form des Produktes I Λ | in Beispiel 1, F.: 148 C (Zers.).

Dann I wurde mit Trichloressigsäure angesäuert (pH 3-4) und 5 h t * l a ij ' V k } ' - 10 - Îi 1 » , * \\ , bei ~15°C gerührt* Nach Stehen über Nacht bei 0°C wurde die Reaktionslösung mit der 2-fachen Menge Wasser verdünnt.
Anschließend wurde 4 mal’mit je 300 ml Essigester/Methanol (10j1) ausgeschüttelt, die vereinigten Essigesterphasen 2 mal mit Wasser gewaschen, über Natriumsulfat getrocknet und im Vakuum zu 22 g öl eingeengt. Nach dem Aufnehmen in
Essigester/Methanol kristallisierten 4,2 g (Schmp. 106 - 110°C) aus. Die Mutterlauge wurde säulenchromatographisch an Kieselgel mit Chloroform/Methanol (10:1) aufgetrennt und zusammen mit dem 1. Kristallisat aus Essigester/Methanol umkristallisiert.
Ausbeute: 7,0 g (35 % d.Th.), F.: 115-116°C (Zers.),
Beispiel 5 3~Benzyl~1-/2-(bis~(2-chlorethyl)-aroino)-2-oxc-tetrahTdro- 1,3.2-oxazaphosphorin-4-yl~oxv7~harnstoff -
Zu 900 mg (3,25 mmol) 4-Hydroxycyclophosphamid in 1 ml Methylenchlorid wurden 540 mg (3,25 mmol) -3-Benzyl-1-hydroxyharnstoff in 40 ml Aceton und eine katalytische Menge Trichloressigsäure gegeben. Das Gemisch wurde über Nacht bei -25°C aufbewahrt, die Kristalle anschließend abgesaugt, mit Aceton und Äther gewaschen und aus Essigester umkristallisiert.
Ausbeute: 500 mg (40 % d.Th.), F.: 122-123°C (Zers.).

Beispiel 6 % • t 5-(o-Bromphenyl)-1-/2-(bis-(2-chlorethyl)-amino)-2-oxo-tetrahyàro-1♦3.2-oxazaphosphorin-4-yl-oxy7-harnstoff 560 mg (2 mmol) 4-Hydroxycyclophosphamid in 10 ml Aceton wurden mit 460 mg 3-o-Bromphenyl--1-hydroxy haras toff und einer katalytischen Menge Trichloressigsäure versetzt und bei -25°C stehengelassen. Nach 2 Tagen wurden die Kristalle abgesaugt und aus Aceton umkristallisiert. Ausbeute: 320 mg (32 % d.Th.), F.: 110-111°C (Zers.).
Beispiel 7 N-Z£-(Bis-(2-chlorethyl)-amino)-2-oxo-tetrahydro-1,3.2-oxazaphosphorin-4-yl-oxv7“4-morpholihocarboxamid 1,2 g (4,3 mmol) 4-Hydroxycyclophosphamïd in 15 ml Aceton wurden mit 630 mg (4,3 mmol) N-Hydroxy-morpholino-carbox-amid und einer Spur Trichloressigsäure versetzt und bei -25°C aufbewahrt. Nach 4 Tagen wurden die Kristalle abgesaugt und aus Aceton umkristallisiert.
Ausbeute: 780 mg (45 % d.Th.), F.: 123-124°C (Zers.).

Ethyl-2-(bis-(2-chlorethyl)-amino-2-oxo-tetrahydro-2H“1>3,2- oxazaphosphorln-4-yl-oxy-carbamat
Beispiel 71 - 18 - a % * % k 550 mg (2 mmol) 4-Hydroxycyclophosphamid und 210 mg (2 mmol) Ethylhydroxycarbamat (Hydroxyurethan) wurden in 5 ml trockenem, alkoholfreiem Methylenchlorid gelöst, mit einer katalytischen Menge Trichloressigsäure versetzt, Molekularsieb 4 A zugegeben und bei -25°C 3 Tage stehengelassen. Anschließend wurde vom Molekularsieb abgetrennt, im Scheidetrichter einmal mit verdünnter Natriumhydrogencarbonat-Lösung gewaschen, die über Natriumsulfat getrocknete Methylenchloridphase i.V. eingeengt und mit Ether verdünnt. Nach 20-stündigem Stehen bei ~25°C wurden die Kristalle abgesaugt, gewaschen und getrocknet.
Ausbeute: 290 mg (40% d. Th.),Fp. 96°C.
Beispiel 72
Benzyl-2-(bis-(2-chlorethyl)-amino-2-oxo-tetrahydro-2H~1,3,2-oxazaphosphorin-4-yl-oxy-carbamat 750 mg (2,7 mmol) 4-Hydroxycyclophosphamid und 450 mg (2,7 mmol) Benzylhydroxycarbamat wurden in 6 ml alkoholfreiem Methylen-Chlorid gelöst, mit etwas Trichloressigsäure versetzt , bei -25°C im Kühlschrank stehengelassen, nach 3 Tagen abfiltriert, die Mutterlauge mit 5 ml Chloroform verdünnt, mit Wasser, ‘ mit verdünnter Natriumhydrogencarbonat-Lösung und nochmals mit Wasser ausgewaschen, über Natriumsulfat getrocknet und s im Vakuum eingeengt. Der ölige Rückstand wurde aus
Essigester/wenig Methanol umkristallisiert,
Ausbeute: 680 mg (59% d. Th.), Fp. 112 - 114°C

Beispiel 73 V* % · a) 3-Hydroxy-ureido-essigsäure 56,5 g (0,28 Mol) Glycinbenzylester-Hydrochlorid wurden in 300 ml Toluol suspendiert und unter Rühren bei einer Badtemperatur von 140°C zwei Stunden lang trockenes Phosgen-Gas eingeleitet. Danach wurde das Reaktionsgemisch im Vakuum eingeengt und der Rückstand im Hochvakuum destilliert.
Ausbeute: 51 g (95% d, Th.) Benzyl-isocyanato-acetat Κρ.Λ 100 - 102°C.
Zu 28,7 g (0,15 Mol) Benzyl-isocyanato-acetat in 50 ml Dioxan wurden unter Rühren und zeitweiliger Kühlung 6,6 g (0,2 Mol) Hydroxylamin in 200 ml Dioxan getropft. Nach 1-stündigem Nachrühren bei Raumtemperatur wurde i.V. eingeengt und der Rückstand aus Essigester umkristallisiert,
Ausbeute: 28,1 g (83,6% d. Th.) Benzyl-3-hydroxy-ureido-acetat Fp. 113 - 120°C.
22,4 g (0,1 Mol) Benzyl-3-hydroxy-ureido-acetat in 300 ml - Methanol wurden mit 5 g Palladium-Kohle versetzt und in einer
Schüttelente hydriert. Nach etwa 20 Minuten war die entsprechende Wasserstoff-Aufnähme beendet. Der Katalysator wurde abgesaugt, das Filtrat i.V. eingeengt und der feste Rückstand aus Dioxan umkristallisiert.
Ausbeute: 9,8 g (73% d. Th.) 3-Hydroxy-ureido-essigsäure,
Fp. 135°C.
- 20 - « * » b) 2,4 g (18 mmol) 3-Hydroxy-ureido-essigsäure in 10 ml Wasser und 25 ml Aceton wurden mit 6,1 g (2 mmol) 4-Hydroxycyclophosphamid versetzt und über Nacht bei -25°C stehengelassen. Anschließend wurde 25 ml Aceton und 1,8 g (18 mmol) Cyclohexylamin in 10 ml Aceton zugegeben, nach 2-stündigem Stehen abgesaugt und aus Aceton mit wenig Methanol umkristallisiert.
Ausbeute: 3,1 g (44% d. Th.), P.: 107 - 108°C.
Beispiel 74.
3-/N ,N-(Bis-(2-chlorethyl)-diamino-phosphf ny.l-oxy7~ proplonaldehyd-oxim (Aldophosphamid-Oxim) 4,0 g (13,7 mmol) 4-Hydroperoxycyclophosphamid wurden in 50 ml Wasser unter Eiskühlung suspendiert und mit 500 mg Natriumthiosulfat x 5 Mol Wasser versetzt. Unter Rühren bei 5-10°C der pH-Wert mit einem pH-Meter kontrolliert, mit 2 normaler Schwefelsäure zwischen pH 4,5 und 5,5 gehalten und solange eine konzentrierte Lösung von Natriumthiosulfat zugetropft, bis der pH-Wert nicht mehr anstieg« Man rührte eine halbe Stunde bei ca. 10°C nach, tropfte eine wässrige Lösung von 950 mg Hydroxylamin-Hydrochlorid zu, hielt den pH-Wert mit 2 normaler Natronlauge auf 5, ließ über Nacht das Reaktionsgemisch im Kühlschrank bei 5°C stehen, extrahierte viermal mit je 50 ml Essigester und engte die organischen Extrakte nach dem Trocknen über Natriumsulfat im Vakuum bei 30°C ein. Der Rückstand wurde in Methylenchlorid aufgenommen und die Kristalle nach 1 Tag abgesaugt.
Ausbeute: 3,4 g (85% d. Th.), F.: 79 -81°C.
* * 3-p-Brom-1-Z2~(bis-(2-chlorethyl)-amino)-2-oxo-tetrahydro- 1,3,2-oxazaphosphorin-4-yl-oxy7-harnstoff - 21 -
Beispiel 75 5,8 g (20 mmol) Aldophosphamid-oxim in 60 ml Aceton wurden mit 4g (20 mmol) p-Bromphenylisocyanat in 40 ml Aceton versetzt, 5 Stunden unter Kühlung gerührt. Nach 2 Stunden wurde das abgesaugte Kristallisat i.V. am RotationsVerdampfer ' bei 40°C getrocknet und in Methanol umkristallisiert.
Ausbeute: 8,1 g (82,8% d. Th.), F.: 118 - 120°C.
Beispiel 76 m-Trifluormethylphenyl-1-Z2-(bis-(2-chlorethyl)-amino)-2-oxo-tetrahydro-1,3,2-oxazaphosphorin-4-yl-oxy7-harnstoff 7,3 g (25 mmol) Aldophosphamidoxim in 80 ml Aceton wurden mit 4,7 g (25 mmol) m-Trifluormethylphenylisocyanat in 40 ml Aceton versetzt. 3 Stunden wurde bei 0°C gerührt, über Nacht im Kühlschrank bei -25°C stehengelassen, mit 150 ml Petrolether versetzt und nach weiterem Stehen im Kühlschrank über Nacht bei -25°C abgesaugt. Das Kristallisat wurde bei 30°C getrocknet und aus Isopropanol umkristallisiert.
Ausbeute: 9,2 g (76,8% d. Th.), F.: 91 - 93°C.
Beispiel 77 3-Cyclohexyl-1-/2-(bis-(2-chlorethyl)-amino)-2-oxo-tetrahydro-1,3,2“Oxazaphosphorin-4-y.l-oxy7-harnstof f 5g (18 mmol) Aldophosphamid-oxim und 2,2 g Cyclohexyl-isocyanat wurden in je 10 ml Aceton gelöst, bei 0°C zusammengegeben, nach 2 Stunden die Kristalle absaugt und mit Aceton/Ether nachgewaschen.
Ausbeute: 4,2 g (56% d. Th.), F.: 113°C.

Beispiel 78 * ' 5 g (18 mmol) Aldophosphamid-oxim und 1,2 g Ethylisocyanat " wurden in je 15 ml Aceton gelöst, und bei ca. 0°C zusammen gegeben. Nach 5 Stunden wurde das Kristallisat abgesaugt und mit Aceton/Ether nachgewaschen.
Ausbeute 3,5 g (54% d. Th.), F.: 101°C.
Beispiel 79 2- (Fluoren-2-yl)-1-/2-(bis-(2-chlorethyl)-amino)-2-oxo-tetrahydro-1,3,2-oxazaphosphorin-4-yl-oxy7-harnsto£f 2,9 g (10 mmol) Aldophosphamid-oxim in 30 ml Aceton wurden mit 2,1 g (10 mmol) Fluorenyl-2-isocyanat in 20 ml Aceton bei 0°C versetzt. Am nächsten Tag wurde das ausgefallene Produkt abgesaugt, i.v, bei 6Q°C getrocknet und aus Isopronal/Methanol umkristallisiert.
Ausbeute: 2,5 g (40,1% d. Th.), F.: 114°C.
Beispiel 80 3- Benzoyl~l-/2-(bis-(2-chlorethyl)-amino)-2-oxo-tetrahydgo-1,3,2-oxazaphosphor in-4-y l-oxy7-harnstof f 5,8 g (20 mmol) Aldophosphamid-oxim in 60 ml Aceton wurden mit 2,9 g (20 mmol) Benzoylisocyanat in 40 ml Aceton versetzt. 5 Stunden lang wurde im Eisbad unter Stickstoffat-mosphäre gerührt, nach 2 Stunden der Feststoff abgesaugt, am RotationsVerdampfer bei 30°C getrocknet und aus Methanol umkristallisiert.
Ausbeute: 2,4 g (27,3% d. Th.), F.: 124 - 125°C.

Beispiel 81 - 23 - Λ ' ri χ « 3-P-Nitrophenyl-l-/2-(bis-(2-chlorethyl)-amino)-2-oxo-tetrahydro-1,3,2-oxazaphosphorin-4-yl-oxy7-harnstoff
Zu 5,8 g (20 mmol) Aldophosphamid-oxim in 60 ml Aceton wurde " eine Lösung von 3,3 g (20 mmol) p-Nitrophenylisocyanat in 40 ml Aceton gegeben, nach 2 Stunden das Festprodukt abgesaugt, am RotationsVerdampfer bei 40°C getrocknet, und aus Dimethyl-formamid/Ethanol umkristallisiert.
Ausbeute: 6,7 g (73,5% d.Th.), P.s 117 - 118°C.
Pharmazeutische Anwendungsbeispiele
Beispiel 82 (magensaftresistente Filmtabletten) 100 g der Verbindung entsprechend Beispiel Nr. 5 werden mit 7,0 g Aerosil durch ein Sieb gegeben und gemischt. Dieser Mischung fügt man 76,0 g Avicel PH 105, 10 g Maisstärke und 7,0 g Stearinsäure zu und mischt bis zu homogener Verteilung aller Bestandteile. Diese Masse wird in bekannter Weise zu Kernen von 200 mg mit 100 mg Wirkstoff gepreßt.
Diese Kerne werden mit einem magensaftresistenten Filmüberzug, z.B. aus geeigneten Cellulosederivaten oder einem vollsynthetischen Filmbildner in organischer Lösung oder wässriger Dispersion, der Zusätze wie Weichmacher, Farbstoffe, Geschmacksstoffe oder Entschäumer enthalten kann, überzogen.
Beispiel 83 (magensaftresistente Gelatinesteckkapseln) * 24 ·* * ** T * 250 g der Verbindung entsprechend Beispiel Nr. 5 werden mit 7,5 g Aerosil durch ein Sieb gegeben und gemischt. Dieser Mischung fügt man 40 g Lactose und 2,5 g Magnesium-stearat zu und mischt bis zu homogener Verteilung aller V1 Bestandteile.
Diese Masse wird in Einzeldosen von 300 mg in Gelatinesteckkapseln abgefüllt.
Eine Kapsel enthält 250 mg Wirkstoff.
Die Gelatinekapseln werden entsprechend Beispiel 1 mit einem magensaftresistenten Filmüberzug versehen.

- 5. Verfahren zur Herstellung der 4-Carbamoyloxy-oxaza-phosphorin-Derivate gemäß Ansprüchen 1 bis 4, dadurch gekennzeichnet, daß man ein 4-Hydroxy- oder 4-Methoxy- oder
- 4-Ethoxy-oxazaphosphorin der allgemeinen Formel II R. OY V I3 I r4 N N U- H R' > y II 2 ^ \ /\R4 4 \ < ψ » 4 ** worin R^, R2, R3 und R^ die gleiche Bedeutung wie in Formel ' ' I haben und Y Wasserstoff, Methyl oder Ethyl ist, mit einem äydroxycarbamat-Derivat der allgemeinen Formel - III Rc X I5 II HO - N - C - Z III worin Z, Rj. und X die gleiche Bedeutung wie in Formel I haben, in Gegenwart eines inerten Lösungsmittels, gegebenenfalls unter Kühlen oder Erwärmen und/oder gegebenenfalls in Anwesenheit eines sauren Katalysators, umgesetzt wird,
- 6. Verfahren zur Herstellung von Verbindungen der Formel I gemäß Anspruch 1, worin Z mit Rg = H und R,.
