Claims (48)
- 돌연변이 IL-2 부분에 융합된 비-IL-2 부분을 포함하는 융합 단백질에 있어서, 상기 융합 단백질이 고친화도 수용체를 발현하는 세포에 대해 참고 단백질보다 더 큰 선택성을 나타내며, 상기 참고 단백질에는 비-돌연변이 IL-2 부분에 융합된 비-IL-2 부분이 포함되고, 상기 선택성은 IL-2Rβγ수용체를 발현하는 세포의 활성화에 대한 IL-2Rαβγ수용체를 발현하는 세포의 활성화의 비로 측정되는 단백질.
- 제 1 항에 있어서, 돌연변이 IL-2 부분에 아미노산 치환, 아미노산 결실 및 아미노산 개질로 이루어진 군으로부터 선택되는 아미노산 돌연변이가 포함되는 융합 단백질.
- 제 1 항에 있어서, 상기 선택성이 성숙한 인간 IL-2 단백질의 위치 88 에 N 에서 R 로의 아미노산 치환을 갖는 돌연변이 인간 IL-2 부분에 융합된 비-IL-2 부분을 포함하는 참고 융합 단백질의 선택성의 약 0.1% 내지 약 100% 인 융합 단백질.
- 제 1 항에 있어서, 상기 선택성이 성숙한 인간 IL-2 단백질의 위치 88 에 N 에서 R 로의 아미노산 치환을 갖는 돌연변이 인간 IL-2 부분에 융합된 비-IL-2 부분을 포함하는 참고 융합 단백질의 선택성의 약 0.1% 내지 약 30% 인 융합 단백질.
- 제 1 항에 있어서, 상기 선택성이 성숙한 인간 IL-2 단백질의 위치 88 에 N 에서 R 로의 아미노산 치환을 갖는 돌연변이 인간 IL-2 부분에 융합된 비-IL-2 부분을 포함하는 참고 융합 단백질의 선택성의 약 1% 내지 약 20% 인 융합 단백질.
- 제 1 항에 있어서, 상기 선택성이 성숙한 인간 IL-2 단백질의 위치 88 에 N 에서 R 로의 아미노산 치환을 갖는 돌연변이 인간 IL-2 부분에 융합된 비-IL-2 부분을 포함하는 참고 융합 단백질의 선택성의 약 2% 내지 약 10% 인 융합 단백질.
- 제 1 항에 있어서, IL-2Rαβγ수용체를 발현하는 세포가 CTLL-2, Kit 225 및 성숙한 T-세포로 이루어진 군으로부터 선택되는 융합 단백질.
- 제 1 항에 있어서, IL-2Rβγ수용체를 발현하는 세포가 TF-1β세포 및 NK 세포로 이루어진 군으로부터 선택되는 융합 단백질.
- 제 1 항에 있어서, 성숙한 인간 IL-2 단백질 보다 더 긴 혈청 반감기를 갖는 융합 단백질.
- 제 1 항에 있어서, 상기 비-IL-2 부분이 알부민인 융합 단백질.
- 제 1 항에 있어서, 상기 비-IL-2 부분에 항체 도메인이 포함되는 융합 단백질.
- 제 11 항에 있어서, 항체 도메인이 KS-1/4, dI-KS, dI-KS(γ4h)(FN>AQ), huKS, huKS(N 에서 Q 로), NHS76(γ2h), NHS(γ4h), NHS76(γ2h)(FN>AQ), NHS76(γ4h)(FN>AQ) 및 14.18 로 이루어진 군으로부터 선택되는 융합 단백질.
- 제 2 항에 있어서, 상기 돌연변이가 IL-2Rαβγ수용체에 대한 단백질의 친화도에 비해 IL-2Rβγ수용체에 대한 융합 단백질의 친화도 상에 차별적 효과를 갖는 융합 단백질.
- 제 1 항에 있어서, 차별적 효과가 2 배 초과이며, 하기와 같이 정의되는 융합 단백질:
- 제 14 항에 있어서, 차별적 효과가 약 5 배 내지 약 10 배인 융합 단백질.
- 제 14 항에 있어서, 차별적 효과가 약 10 배 내지 약 1000 배인 융합 단백질.
- 제 14 항에 있어서, 돌연변이 IL-2 부분에 아미노산 치환 N88R 또는 D20T 가 포함되는 융합 단백질.
- 제 1 항에 있어서, 성숙한 인간 IL-2 단백질의 위치 88 에 N 에서 R 로 변화되는 아미노산 치환이 포함되는 융합 단백질.
- 제 18 항에 있어서, 성숙한 인간 IL-2 단백질의 위치 85 에 L 에서 T 로, 및 위치 86 에서 I 에서 T 로 변화되는 아미노산 치환이 추가 포함되는 융합 단백질.
- 제 1 항에 있어서, 성숙한 인간 IL-2 단백질의 위치 20 에 D 에서 T 로 변화되는 아미노산 치환이 포함되는 융합 단백질.
- 제 1 항에 있어서, 상기 돌연변이 IL-2 부분에 K8, Q13, E15, H16, L19, D20, Q22, M23, N26, H79, L80, R81, D84, N88, I92 및 E95 로 이루어진 군으로부터 선택되는 아미노산 위치에 돌연변이를 갖는 성숙한 인간 IL-2 단백질이 포함되는 융합 단백질.
- 제 1 항에 있어서, 상기 돌연변이 IL-2 부분에 L25, N31, L40, M46, K48, K49, D109, E110, A112, T113, V115, E116, N119, R120, I122, T123, Q126, S127, S130 및 T131 로 이루어진 군으로부터 선택되는 아미노산 위치에 돌연변이를 갖는 성숙한 인간 IL-2 단백질이 포함되는 융합 단백질.
- 제 21 항에 있어서, 아미노산 치환이 N88R 인 융합 단백질.
- 제 21 항에 있어서, 아미노산 치환이 D20T 인 융합 단백질.
- 제 22 항에 있어서, 아미노산 치환이 Q126D 인 융합 단백질.
- 제 21 항 또는 제 22 항에 있어서, 하나 이상의 상기 아미노산 치환이 IL-2Rαβγ수용체에 대한 상기 단백질의 친화도에 비해 IL-2Rβγ수용체에 대한 상기 융합 단백질의 친화도 상에 차별적 효과를 갖는 융합 단백질.
- 제 21 항 또는 제 22 항에 있어서, 차별적 효과가 2 배 초과이며, 하기와 같이 정의되는 융합 단백질:
- IL-2 부분 및 비-IL-2 부분을 포함하는 융합 단백질에 있어서, 상기 비-IL-2 부분에 IL-2Rαβγ수용체에 대한 상기 융합 단백질의 친화도에 비해 IL-2Rβγ수용체에 대한 상기 융합 단백질의 친화도 상에 차별적 효과를 갖는 돌연변이가 포함되는 융합 단백질.
- 제 28 항에 있어서, 상기 차별적 효과가 약 5 배 내지 약 10 배인 융합 단백질.
- 제 28 항에 있어서, 상기 차별적 효과가 약 10 배 내지 약 1000 배인 융합 단백질.
- 제 28 항에 있어서, 상기 비-IL-2 부분에 항체 도메인이 포함되는 융합 단백질.
- 제 31 항에 있어서, 상기 항체 도메인에 IgG 감마 1 도메인, IgG 감마 2 도메인 또는 IgG 감마 4 도메인이 포함되는 융합 단백질.
- 제 31 항에 있어서, 상기 돌연변이가 상기 항체 도메인 내에 존재하는 융합 단백질.
- 제 33 항에 있어서, 상기 돌연변이가 서열 번호 1 에서 N 을 상이한 아미노산으로 변화시키는 융합 단백질.
- 제 34 항에 있어서, 상기 N 이 Q 로 변화되는 융합 단백질.
- 제 32 항에 있어서, 상기 돌연변이가 서열 번호 2 에서 FN 을 AQ 로 변화시키는 융합 단백질.
- 제 31 항에 있어서, 서열 번호 35 및 서열 번호 36 의 아미노산 서열이 포함되는 융합 단백질.
- 제 11 항에 있어서, 상기 항체 도메인에 IgG 감마 1 도메인, IgG 감마 2 도메인 또는 IgG 감마 4 도메인이 포함되는 융합 단백질.
- 제 11 항에 있어서, 상기 항체 도메인에 돌연변이가 포함되는 융합 단백질.
- 제 39 항에 있어서, 상기 돌연변이가 서열 번호 1 에서 N 을 상이한 아미노산으로 변화시키는 융합 단백질.
- 제 40 항에 있어서, 상기 N 이 Q 로 변화되는 융합 단백질.
- 제 41 항에 있어서, 상기 돌연변이가 서열 번호 2 에서 FN 을 AQ 로 변화시키는 융합 단백질.
- 제 11 항에 있어서, 서열 번호 35 및 서열 번호 36 의 아미노산 서열을 포함하는 2 개 펩티드의 이량체인 융합 단백질.
- 하기를 포함하는, 세포 표면 상의 별개의 수용체에 결합하는 2 개 이상의 단백질 도메인을 포함하는 치료적 융합 단백질의 개선 방법:i. 상기 별개의 수용체에 특이적인 2 개 이상의 시험관 내 분석에서의 활성에 대해 상기 융합 단백질을 시험하고,ii. 돌연변이, 또는 화학적 또는 생화학적 개질에 의해 상기 융합 단백질을 개질시키고,iii. 상기 개질된 융합 단백질을 상기 2 개 이상의 분석에서 시험하고,iv. 상기 비-개질된 융합 단백질에 비해 상기 분석에서 이들의 활성에 상이하게 영향을 미치는 개질된 융합 단백질을 동정하고,V. 상기 동정된 개질 융합 단백질을 동물에서 활성에 대해 시험함.
- 제 44 항에 있어서, 상기 융합 단백질의 상기 개질이 상기 부분에 대한 수용체와 융합 단백질 내 부분의 상호작용에 영향을 미치는 방법.
- 제 45 항에 있어서, 상기 수용체가 하나 이상의 상기 시험관 내 분석에서 존재하는 방법.
- 제 46 항에 있어서, 상기 시험관 내 분석이 세포 기반 분석인 방법.
- 제 47 항에 있어서, 상기 분석이 IL-2Rαβγ또는 IL-2Rβγ또는 둘 다를 발현하는 세포를 이용하는 방법.
