Claims (1)
- 【特許請求の範囲】 1.エンテロバクターから得られるビオチン合成用遺伝子bioB、bioF、bioC、bioD およびbioAまたはそれらと機能的に同等の遺伝子的変異体および突然変異体 を含むDNA断片において、これらの遺伝子が転写単位中で、ある転写方向に配置 されており、共通のプロモーターにより転写されることを特徴とするDNA断片。 2.エンテロバクターが、Escherichia、SalmonellaおよびCitrobacter属のグ ループから選択されることを特徴とする請求項1のDNA断片。 3.エンテロバクターが、Escherichia Coli種から選択されることを特徴とす る請求項1または2のDNA断片。 4.共通のプロモーターがtac-プロモーターであることを特徴とする請求項1 〜3のいずれかのDNA断片。 5.転写単位の、bioB-遺伝子と直接結合している遺伝子調節要素が、 配列 を含むことを特徴とする請求項1〜4のいずれかのDNA断片。 6.転写単位の、bioB-遺伝子と直接結合している遺伝子調節要素が、 配列 を含むことを特徴とする請求項1〜4のいずれかのDNA断片。 7.転写単位の、bioB-遺伝子と直接結合している遺伝子調節要素が、 配列 を含むことを特徴とする請求項1〜4のいずれかのDNA断片。 8.転写単位中で連続しているbioDおよびbioAの間隔が50bp以下である ことを特徴とする請求項1〜7のいずれかのDNA断片。 9.bioDおよびbioAが、bioD-遺伝子の3’-末端がbioA-遺伝子のためのリボ ソーム結合位置を含むように配置されていることを特徴とする請求項1〜8のい ずれかのDNA断片。 10.請求項1〜9のいずれかのDNA断片を含むことを特徴とするプラスミド 。 11.寄託番号DSM 7246、DSM 7247、場合によってはDSM 8554でE.Coli XL1-B lue、E.Coli BM4062またはE.Coli ED8767中に寄託してあることを特徴とする プラスミドpBO30A-15/9。 12.寄託番号DSM 8555でAgrobacterium/Rhizobium sp.HK4中に寄託してあ ることを特徴とするプラスミドpBO47。 13.請求項1〜12のいずれかのDNA断片またはプラスミドを含むことを特 徴とする微生物。 14.寄託番号DSM 7246、DSM 7247、場合によってはDSM 8554で寄託してある 、それぞれプラスミドpBO30A-15/9を含むことを特徴とするE.Coli XL1-Blue、E .Coli BM4062およびE.Coli ED8767、ならびにそれらの遺伝子的変異体および 突然変異体。 15.寄託番号DSM 8555で寄託してある、プラスミドpBO47を含むことを特徴 とするAgrobacterium/Rhizobium sp.HK4、ならびにそれらの遺伝子的変異体お よび突然変異体。 16.物質交代可能な炭素供給源が、請求項13〜15のいずれかの微生物の 作用により発酵し、ビオチンになることを特徴とする生物工学的なビオチンの合 成方法。 17.ビオチン生成酵素を使用し、無細胞系でデチオビオチンをビオチンへ変 換することを含むビオチンの製造方法において、変換を、チアミンピロリン酸塩 、NADPH、S-アデノシルメチオニン、Fe++イオン、システイン、およびアスパラ ギン、アスパラギン酸、グルタミンおよびセリンのグループから選択された少な くとも1種の他のアミノ酸の存在下に行なうことを特徴とする方法。 18.変換をフラボドキシンおよびフェレドキシン(フラボドキシン)- NADP+-還元酵素の存在下に行なうことを特徴とする請求項17の方法。 19.変換が、Escherichia Coli細胞抽出物の45%飽和における硫酸アンモニ ウム沈殿により得られるタンパク質画分の存在下に行なわれることを特徴とする 請求項17または18の方法。 20.Escherichia Coli細胞抽出物の45%飽和における硫酸アンモニウム沈殿 により得られる、ビオチン生成酵素刺激作用を有するタンパク質画分。 21.無細胞系でのビオチンの製造方法における、請求項20のタンパク質画 分の使用。
