Abstract
본 발명은 유전자들이 전사 단위체에 배열되어있는, 장내세균으로부터의 바이오틴 생합성에 관여할 수 있는 바이오B, 바이오F, 바이오C, 바이오D 및 바이오A를 포함하는 DNA단편 또는 그의 동일한 기능의 유전적 변종 및 돌연변이에 관한 것이다. 본 발명은 또한 상기 DNA단편 및 플라스미드를 함유하는 미생물, 및 상기 미생물을 사용하는 바이오틴 제조방법에 관한 것이다.
Description

제1도는 바이오틴 생합성의 대사 경로의 효소를 나타낸다.
제2도는 플라스미드 pBO30의 구조도를 나타낸다.
제3도는 플라스미드 pBO30, pBO30-9 및 pBO30A-15의 바이오D 유전자의 3'말단 부분 및 바이오A 유전자의 5'말단(파선 화살표;바이오A 출발 코돈을 밑줄긋고, 바이오D 정지 코돈은 점선으로 나타낸다) 대한 DNA 서열과 함께 플라스미드 구조에서 상대적인 제한 효소의 절단위치, 및 바이오A 유전자의 샤인-달가노(Shine-Dalgano, SD)서열을 나타낸다. 잠재적인 "스템-루프"구조는 실선 화살표로 표시한다.
Claims (23)
- 유전자들이 전사 단위체에 배열되어있는, 장내세군으로부터의 바이오틴 생합성 유전자인 바이오B, 바이오F, 바이오C, 바이오D 및 바이오A를 함유하는 DNA단편 또는 그의 동일한 기능의 유전적 변종 및 돌연변이.
- 제1항에 있어서, 장내세균이 에쉐리키아소, 살모넬라속 및 시트로박터속으로 구성된 군으로부터 선택된 DNA단편.
- 제1항 또는 제2항에 있어서, 장내세균이 에쉐리키아 콜라이(Escherichia coli)종으로부터 선택된 DNA단편.
- 제1항 내지 제3항중의 어느 한 항에 있어서, 전사 단위체의 유전자-조절인자가 tac프로모터를 포함하는 DNA단편.
- 제1항 내지 제4항중의 어느 한 항에 있어서, 바이오B 유전자에 직접 연관된 유전자-조절인자가 하기의 서열을 포함하는 DNA단편 : AAGCTTACTC CCCATCCCCC TGTTGACAAT TAATCATCGG CTCGTATAAT GTGTGGAATT GTGAGCGGAT AACAATTTCA CACAGGAAAC AGGATCGGTA CCTTAGGAGG TGACTAGTC.
- 제1항 내지 제4항중의 어느 한 항에 있어서, 바이오B 유전자에 직접 연관된 유전자-조절인자가 하기의 서열을 포함하는 DNA단편: AAGCTTACTC CCCATCCCCC TGTTGACAAT TAATCATCGG CTCGTATAAT GTGTGGAATT GTGAGCGGAT AACAATTTCA CACAGGAAAC AGGATCGGTA CCTAAGGAGG TTTACTAGTC.
- 제1항 내지 제4항중의 어느 한 항에 있어서, 바이오B 유전자에 직접 연관된 유전자-조절인자가 하기의 서열을 포함하는 DNA단편: AAGCTTACTC CCCATCCCCC TGTTGACAAT TAATCATCGG CTCGTATAAT GTGTGGAATT GTGAGCGGAT AACAATTTCA CACAGGAAAC AGGATCGGTA CCTAAGGAGA CTAGTC.
- 제1항 내지 제7항중의 어느 한 항에 있어서, 전사 단위체에서 지속성인 바이오D 및 바이오A 유전자 사이의 거리가 50bp이하인 DNA단편.
- 제1항 내지 제8항중의 어느 한 항에 있어서, 바이오D 및 바이오A 유전자가 바이오D 유전자의 3' 말단이 바이오A 유전자의 리보좀 견합부위를 포함하도록 배열된 DNA 단편.
- 제1항 내지 제9항중의 어느 한 항에 기재된 DNA 단편을 함유하는 플라스미드.
- 수탁번호가 각각 DSM 7246, DSM 7247 및 DSM 8554로 기탁된 에쉐리키아 콜라이 XL1-블루(Blue), 에쉐리키아 콜라이 BM4062 또는 에쉐리키아 콜라이 ED8767 내의 플라스미드 pBO30A-15/9.
- 수탁번호 DSM 8555로 기탁된 애그로박테리움/리조비움종 HK4 내의 폴라스미드 pBO47.
- 제1항 내지 제12 항중의 어느 한 항에 기재된 DNA 단편 또는 플라스미드를 함유한 미생물.
- 수탁번호가 각각 DSM 7246, DSM 7247 및 DSM 8554로 기탁된, 플라스미드 pBO30A-13/9를 각기 함유하는, 에쉐리키아 콜라이 XL1-블루, 에쉐리키아 콜라이 BM4062 및 에쉐리키아 콜라이 ED8767 및 그의 유전적 변종 및 돌연변이.
- 수탁번호 DSM 8555로 기탁된, 플라스미드 pB047을 함유하는, 애그로박테리움/리조비움종 HK4 및 그의 유전적 변종 및 돌연변이.
- 대사될 수 있는 탄소원이 제13항 내지 제15항중의 어느 한 항에 기재된 미생물에 의해 바이오틴으로 발효되는 바이오틴합성을 위한 생물공학적 방법.
- 전환이 티아민 피로포스페이트, NADPH, S-아데노실메티오닌, Fe2+이온, 시스테인, 및 아스파라긴, 아스파르트산, 글루타민 및 세린으로 구성된 군으로부터의 하나 이상의 다른 아미노산의 존재하에서 수행되는, 세포-프르 시스템에서의 바이오틴 합성효소에 의한 탈티오바이오 틴에서 바이오틴으로의 전환을 포함하는 바이오틴 제조방법.
- 제17항에 있어서, 전환이 플라보독신 및 페레독신(플라보독신)-NADP+리덕타제의 존재하에서 수행되는 바이오틴 제조방법.
- 제17항 또는 제18항에 있어서, 전환이 에쉐리키아 콜라이의 세포 추출물의 45% 포화된 황산암모늄 침전물에 의해 수득될 수 있는 단백질 분획의 존재하에서 수행되는 바이오틴 제조방법.
- 에쉐리키아 콜라이의 세포 주출물의 45% 포화된 황산암모늄 침전물에 의해 수득될 수 있는, 바이오틴 합성효소-촉진 작용을 갖는 단백질 분획.
- 세포-프리 시스템에서의 바이오틴 제조방법에 있어서 제20항에 기재된 단백질 분획의 용도.
