Description
本発明は、ビオチン物質交代方法の遺伝子を発現させるための組換え体遺伝子材料を含む微生物であるエンテロバクターのビオチン物質交代方法の遺伝子を発現させるための組換え体遺伝子材料、およびそのような微生物の、ビオチン製造のための生物工学的方法における使用に関する。本発明はさらに、無細胞系におけるビオチン生成酵素によるデチオビオチンの変換を含む、ビオチンの製造方法に関する。
したがって、ビオチンは、食品および飼料に有用な添加物質である。
合成有幾化学の方法でビオチンを製造するには手数と費用がかかる。この理由から、微生物の助けを借りて、グルコースのような安価な出発物質からビオチンを合成できる生物工学的方法がますます注目されている。
これらの方法は、グルコースから出発して行なわれる。特許文献1(EP-B-236429)は、たとえばE.Coliのビオチン−オペロンで形質転換された微生物を記載しているが、その際、宿主はそのbirA/bioR−遺伝子において突然変異している。
特許文献3(EP-A-449724)は、ビオチン−オペロンで形質転換した微生物を開示しているが、これらの微生物はさらに突然変異を示し、その結果グルコース消費が少なくなっている。
しかし、これまで知られている生物工学的方法により得られる収率は、経済的な観点からは満足できるものではない。
本発明のDNA断片およびベクターを構築するために、まずビオチン−オペロンの遺伝子を好適な微生物の染色体から分離し、続いてプロモーターおよびリボソーム結合位置のような遺伝子調節要素を管理しながら、それらが単一の転写単位中に組織されて存在するように結合するのが有利である。bio-遺伝子を分離するための出発材料として、エンテロバクター科の細菌品種、例えばEscherichia、SalmonellaまたはCitrobacter属を使用することができる。出発材料は、Escherichia coli種の微生物が最も有利である。
通常の様式で培地中の栄養素を変えることにより、および発酵条件をそれぞれの微生物に適合させることにより、ビオチンの収率をさらに改善することができる。
本発明を下記の実施例によりさらに説明する。
一般的な方法
制限エンドヌクレアーゼは、製造者の指示にしたがって、3-5単位/μgDNAで使用した。たとえばDNA/DNAハイブリッド化のためのプローブとして、および配列化反応のための「プライマー」として使用するための、制限切断位置を取り入れるためのDNAリンカー(Boehringer Mannheim,BRDから入手)の、および合成オリゴヌクレオチド(Microsynth,Windisch,CHから入手)の標識付けおよび加リン酸は、Sambrookら(Molecular Cloning:実験室マニュアル、第2版、Cold Spring Harbour Laboratory,Cold Spring Harbour,NY;11.31および5.68;1989)によるT4−ポリヌクレオチド−キナーゼで行なった。リゲーシヨン反応は、製造者の指示にしたがってT4-DNA-リガーゼで行なった。
単一の転写単位におけるE.Coliビオチン−オペロンのクローニング
1.1 pBO1およびM13bioDの構築
bio伝子をクローニングするために、E.Coli DSM 498(K12「宿主型」、ドイチェ・ザムルング・フュア・ミクロオルガニズメン・ウント・ツェルクルトゥーレン・GmbH)の染色体DNAを分離した。分離は本質的にHahnおよびHennecke(mol.Gen.Genet.193:46-52;1984)により行なった。続いて2μgのE.Coli DSM 498の全DNAを制限酵素PstIで切断した。DNA断片を水平な0.7%アガロースゲル中で通常の様式(Sambrookら、1989、上記文献6.19〜6.9)で電気泳動により分離し、"Gene Screen"薄膜(NEN-Du Pontのナイロン薄膜)上に移した(Southern,J.Mol.Biol.,98:503-517;1975)。DNAを真空オーブン中、80℃で2時間培養することにより、乾燥したフィルター上に固定した。bio-オペロンを含むDNA断片を識別するために、bioB遺伝子の5'末端から得た配列(Otsuka,A.J.,Dissertation,カリフォルニア大学San Diego,CA.;1978)に相当する5'-GGCTCACCGCCCACGCTGGACATTG-3'配列の25ヌクレオチド長の合成オリゴヌクレオチドを、プローブとして、フィルターに結合したDNAとハイブリッド化した。このために、まずこのオリゴヌクレオチド40pMolをT4-ポリヌクレオチドおよびγ-[32P]-ATP(75μCi)で標識を付けた。フィルターに結合したDNAと放射性標識を付けたプローブのハイブリッド化は、(Sambrookら、1989、上記文献9.52〜9.55)により行なった。このために、DNAをまず5x Denhardt-溶液(1x Denhardt-溶液:0.02%牛血清、0.02%Ficoll,0.01%ポリビニルピロリドン)、6x SSC-緩衝液(1x SSC:150 mM NaCl、15 mMクエン酸ナトリウム、pH7.2)および150μg/ml Lachssperm-DNAで2時間予備ハイブリッド化し、続いて2x Denhardt-溶液、6x SSC、0.5%SDS、150μg/ml Lachssperm-DNAで18時間ハイブリッド化し、2時間洗浄し、最後に2x SSC、0.1% SDSで各30分間
4回洗浄した。温度はすべての工程で65℃であった。この「サザーンブロット」により、標識を付けたオリゴヌクレオチドを5.4 kb長のPstI-断片でハイブリッド化した。
程を4x Denhdrdt-溶液、6x SSC,100μg/mlサケの精子DNA中で行ない、続いて2x SSC中で6x洗浄した。温度は65℃であった。3クローンがbioB-オリゴヌクレオチドを結合し、このクローンの一つから、5.4kb長のPstI-断片(図2)を含むプラスミドpBO1を分離した。制限分析および公開されているデータ’SzybalskiおよびSzybalski、Gene 19:93-103;1982)との比較により、pBO1は、ビオチン−オペロンのbioD以外のすべての遺伝子を含むことがわかった。
これには、E.Coli DSM 498のDNA 30μgをSspIで切断し、0.7%アガロースゲル上で分離した。0.9kb〜1.3kbの大きさの断片を切り取り、電気透析により分離した。この断片0.5μgをSmaIで切断したフアージヘクターM13mp19(Yanisch-Perronら、1985、上記文献)で結紮した。この結紮物で、E.Coli JM109(Yanisch-Perronら、1985、上記文献)のトランスフェクションをMessing(MethodsEnzymol.,101:20-79;1983)にしたがって行なった。挿入物(Phaenotyp LacZ-)を分離し、NYB培地中で増殖させた。E.Coli細胞を遠心分離した後、上澄み液各50μl中のファージを、Schleicher&Schuell "minifold I"装置で「ドットブロット」としてニトロセルロース-フィルター(Schleicher&Schuell BA 85)上に置いた。ファージを変性させるために、このフィルターを0.1M NaOH/1.5M NaCl-緩衝液で5分間処理し、続いて0.5M トリス-HCl,pH 7.5/2.5 M NaClで(5分間)中和した。DNAを80℃で培養(2時間)することによりフィルター上に固定した。フィルターを、記載されているように(Sambrookら,1989、上記文献、9.52-9.55)、放射性標識を付けた520 pb長のSphI/PstI-断片で60℃でハイブリッド化した。
それぞれ0.5μgのプラスミドpBO1および0.5μgのファージM13bioDを制限酵素SnoIおよびHindIIIで切断し、再結紮した。E.Coli RR28をこの結紮物で形質転換した後、組換えたプラスミドを制限分析により検査した。ベクターpHE3の本質的な配列ではなく、bioD遺伝子の一部を含む、約1.5 kb長の、pBOlのSnol/られているプラスミドpBO2(図2)を選択した。分析により、プラスミドpBO2は、E.Coli中に存在するように、完全なbio-オペロンを、uvrB-プロモーター(Sancarら、Cell,28;523-530;1982)の配列とともにbioDの下流に含むことがわかった。
pBO2を含むE.Coli RR28は、NA-プレート上で、pBO1を含むものよりも発育が悪く、明らかにより小さなコロニーを形成することが観察された。この理由は、pBO2中のurvB配列であろう。このurvB配列を欠失させるために、20μgのpBO2-DNAをHindIIIで切断し、150μlのBal31-緩衝液(600 mM NaCl、12.5 mM MgCl2、12.5 mM CaCl2、1mM EDTA、20 mM トリス-HCl、pH7.2)に入れた。次いで段階的に短縮するために、直線状プラスミドBal31(Alteromonas espejiani,Boehringer Mannheim,BRD)を加えた。30℃で3、6、9、12および15分間培養した後、各30μlのアリコートを取りだし、各2μlの0.5 M EGTA(エチレングリコール-ビス-(2-アミノエチル)-テトラアセテート)、pH7.5を加え、続いてフェノール抽出により、Bal31-反応を停止させた。次いで、これらのアリコートを40μlのMung Beanヌクレアーゼ緩衝液(30mM酢酸ナトリウム、50 mM NaCl、1mM ZnCl2、5%グリセリン、pH 4.6)に入れ、対形成していない1本鎖末端を除去し、非特異的な、先のない末端を造るために、Mung Beanヌクレアーゼ(Boehringer Mannheim,BRD)で、37℃で10分間処理した。
1.4.1 pBO22:bioB前部のプロモーターの構築
遺伝子bioB前部の適切なプロモーターを組み込むためには、遺伝子bioBFCDの好ましくない宿主型プロモーターを除去しなければならない これはNcoIで切断することにより達成できるが、これによってbioB-遺伝子のスタートコドンも除去される。この場合、プロモーターとしてtac-プロモーター(RuessellおよびBennett,1982,上記文献)を選択できるが、これはこのプロモーターが構成または誘導性プロモーターとして使用できE.Coli中のみならず、他の多くのグラム陰性菌中で非常に良好な活性を有するためである。
最後に4μgのベクターpHE3をPstIおよびAatIIで切断し、P15A-レプリコンをpHE3から(Henneckeら、1982、上記文献)分離した。
5μgのpBO22をPstIで切断し、突き出したPstI-末端をMung Beanヌクレアーゼで処理することにより、平滑な末端に短縮した。次いで、SnoIで切断し、得られた6.8 kb長のDNA-断片を分離した。bioD-遺伝子の3'-末端を有する0.76 kb-DNA-断片を5μgのpBO3から、ClaIにより制限し、突き出したClaI-末端をクレノーポリメラーゼで充填し、SnoIで制限した後、分離した。両DNA-断片をT4-DNA-リガーゼで結紮し、次いでこの結紮物でE. Coliを形質転換した。クロラムフェニコール上で選別した後、CmRを含む形質転換物から、制限分析によりプラスミドpBO27が得られた。このプラスミドはtac-プロモーターを、遺伝子bioB、bioF、bioCおよび完全なbioD-遺伝子とともに転写単位中に含んでいる(図2)。
bioA-遺伝子の前の好ましくない宿主型プロモーターを除去するために、まず5μgのpBO3をBglIIおよびKpnIで制限し、遺伝子bioB、bioF、bioAおよびORFIを含む4.4 kb-断片を分離した。この断片0.5μgを、BamHIおよびKpnIで切断したファージベクターM13mp18(Yanisch-Perronら、1985, 上記文献)0.5μgで結紮した。
転写単位中で遺伝子bioDおよびbioAを配置するために、5μgのプラスミドpBO6(図2)をSphIおよびSalIで切断した。得られた、遺伝子bioDおよびbioD-遺伝子の上流にあるDNAのSalI-末端までの72bpを含む0.97 kb長のDNA断片を分離した。
bio-遺伝子の前のtac-プロモーターを有する転写単位を構築するために、M13bioDAのDNA 5μgをEcoRIで切断し、突き出したEcoRI-末端を充填するために上記のようにクレノーポリメラーゼで処理し、次いでSnoIで切断した。得られた2.6 kb長の、遺伝子bioD、bioAおよびORFIを含むDNA-断片を分離した。5μgのプラスミドpBO28(図2)をSalIで切断し、突き出したSalI-末端を裂くためにMungBeanヌクレアーゼで処理し、次いで同様にSnoIで切断した。6.4 kb長の、ベクター-DNA、tac-プロモーターおよび遺伝子bioBFCを有するDNA-断片を分離した。
1.5.1 pBO30A-9およびpBO30A-15の構築
プラスミドpBO30を含むE. Coli DS410(DouganおよびSheratt, Mol. Gen. Genet. 151:151-160; 1977)のミニ細胞中では、bioA-遺伝子によりコード化されたDAPA-アミノトランスフェラーゼは、ビオチン合成用の他の酵素よりも発現が本質的に弱かった。bioA-遺伝子の発現を改良するための試験で、「ステム-ループ」構造のような妨害の可能性がある配列を除去するために、bioD-遺伝子とbioA-遺伝子の間隔をエキソヌクレアーゼBal31で短縮した。そのために、25μgのpBO30をSalIで切断し、次いで上記のようにエキソヌクレアーゼBal31およびクレノー-ポリメラーゼで処理した。配列5'-CGTCGACG-3'の合成オリゴヌクレオチド、SalI-リンカーでリゲーションすることにより、SalI-切断位置を再発生させた。次いで、DNAをSalIおよびSnoIで切断し、約640 bp長の、3'-末端で短縮したbioD-断片を分離した。これらの断片を、プラスミドpBO30をSalIおよびSnoIで切断した後で分離することができ、変化していないbioA-遺伝子を含む、8.25kbの大きさの断片で結紮した。
bioDA-領域のDNA-配列およびそこから派生する、プラスミドpBO30、pBO30A-9およびpBO30A-15のアミノ酸配列を図3に示す(Seq ID No 9-16)。
pBO30における発現が、たとえばbioD-遺伝子の発現よりも明らかに弱いbioB-遺伝子の翻訳を改良するために、pBO30中のbioB-遺伝子の上流の、tac-プロモーター、およびクローニングされたtac-プロモーター断片中に含まれるリボソーム結合位置を含む配列を変性させた。このために、合成された、さまざまな配列を含む、いわゆる混合されたオリゴヌクレオチドを遺伝子bioBの前に置いた。好適なリボソーム結合位置を簡単に選択するために、翻訳性bioB::lacZ遺伝子融合、pbioB::lacZ-2、を使用した。pbioB::lacZ-2は、ベクター部分において、リボソーム結合位置を含むtac-プロモーターにおいて、および遺伝子bioBの5'-末端においてプラスミドpBO22と同等である(図2)。しかし、bioB-構造遺伝子のヌクレオチド326の後のNruI-切断位置では、bioB-遺伝子の3'-末端および残りのbioB-遺伝子が欠失し、E. ColiのlacZ-遺伝子(Casadabanら、Methods Enzymol. 100:293-308; 1983)が、bioBおよびlacZがbioB::lacZ融合タンパク質を発現させるための正しい leseraster中に融合され、NruI-切断位置が再発生するように組み込まれた。
bor, N.Y., 352-355; 1972)にしたがって測定した。これにはまず、液体培地中でE. Coli-品種をbio::lacZ-プラスミドで600 nm(OD600)における光学密度約0.5まで濃縮した。
bio-遺伝子を含む転写単位に最適なリボソーム結合位置を組み込むために、各5μgのプラスミドpbioB::lacZ/985E、pbioB::lacZ/16またはpbioB::lacZ/9をClaIおよびNruIで切断し、tac-プロモーター、それぞれのリボソーム結合位置およびbio-遺伝子の5'-末端を含む約550bp長のDNA-断片を分離した。同時に5μgのプラスミドpBO30ΔA(図5)をClaIおよびNruIで切断し、7.7 kb長のDNA-断片を分離した。pBO30から派生したpBO30ΔAでは、bio-遺伝子の大部分および妨害となるNruI-切断位置を含むSalI/BamHI-断片が欠失している(図5)。両断片を結紮し、組み替えたプラスミドを含むクローンを分離した。このようにして、プラスミドpBO30ΔA/9、pBO30ΔA/16およびpBO30ΔA/985を得た。図5は、pbioB::lacZ/9からのリボソーム結合位置を含むpBO30ΔA/9の例におけるそのような構造を示す。
次いで、ベクター-DNA、tac-プロモーター、リボソーム結合位置を改良したbioB-遺伝子および遺伝子bioFCを含む、それぞれ6.6 kb長のDNA-断片を分離した。
2μgのプラスミドpBO30ΔA/9を上記のようにSnoIおよびKpnIで切断し、6.6 kb長のDNA-断片を分離した。4μgのpBO30A-15をSspIで切断した。得られた直線状DNAを配列5'-CGGTACCG-3'のKpnI-リンカーでリゲーシヨンすることにより、bioA-遺伝子の下流に新しいKpnI-位置を挿入した。SnoIで切断した後、遺伝子bioDAを含む2.1 kb-断片を分離した。分離したDNA-断片を結紮し、E. Coli RR28をリゲーシヨン混合物で形質転換した。遺伝子bioBFCDAを含む組換えプラスミドを制限分析で確認した。このようにして、ORFI-遺伝子が欠失したpBO30A-15/9ΔorfIが得られた(図5)。
5μgのプラスミドpBO30A-15/9を制限酵素XbaIおよびEcoRIで切断した。得られた5.8 kb大の、tac-プロモーターおよびビオチン−オペロンを含む制限断片を分離し、続いて同様にXbaIおよびEcoRIで切断した「広宿主領域」プラスミドpRK29OX(Alvarez-Moralesら、Nucl. Acid. Res. 14,4207-4227,1986;XhoI制限位置を欠失させ、その位置にXbaI-位置を挿入することにより変性)で結紮した。その結紮物でE. Coli S17-1(Simonら、Biotechnology 1:784-791;1983)を形質転換した。 組み替えたプラスミドを制限分析で検査し、このようにして、pRK290X中にビオチン−オペロンを一体化したプラスミドpBO47が得られた。
bioB-遺伝子が欠失したプラスミドpBO74ΔBの構築は、プラスミドpBO74-13から出発して行なった(図7)。プラスミドpBO74-13はpBO30(図2)のような同じDNA-構成要素からなる。しかし、プラスミドpBO74-13中のbio-遺伝子の順序は異なっている。
2.1 Escherichia coli生産-品種によるビオチンの生体内発酵
pBO30A-15/9(DSM 7246)を含むE. Coli-品種XL-1Blueを、20 l MBR-発酵装置中、グリセリン最少培地(培養開始時に3%グリセリン)で、バッチ供給法により、650 nmにおける光学密度(OD650)が20になるまで37℃で30時間培養した。プラスミドpBO30A-15/9の存在は、発酵の前培地(31グリセリン-最少培地)およびバッチ相にクロラムフェニコール(50μg/ml)を加えることにより確認した。グルコースまたはコハク酸塩(バッチ発酵相の開始時に0.4%)のような他の炭素供給源も同様に実用的である。細胞の物質交代活性は、その特異的な酸素吸収率により追跡した。発酵中のビオチン生産は、発酵装置−培地のビオチン値を滴定することにより、Lactobacillus plantarumによる生物学的定量で追跡した(E. DeMollおよびW. Shive,Anal. Chem. 158:55-58,1986)。
100μg/mlのアンピシリン(ナトリウム塩、Fluka)および50μg/mlのクロラムフェニコール(fluka)。
2.2 Agrobacterium/Rhizobium-生産-品種 HK4/pBO47によるビオチンの生体内発酵
ビオチン生産プラスミドpBO47(DSM 8555)を含むビオチン-オキソトローフ品種Agrobacterium/Rhizobium sp HK4の細胞を、21のMBR-発酵装置中、L-グルタミン酸/ベタイン最少培地中で、バッチ供給法により30℃でOD650が70になるまで培養した。HK4/pBO47は、極めて遅い成長(「維持成長」)でも著しく安定したビオチン合成速度が特徴である。したがって、この実験ではバイオマスの培養後、炭素「供給」を大幅に減少させた状態で長い維持期間(500時間)が続いた。
脱イオン水1.25リットルに下記の成分を溶解させるか、ないしは加えた。

(生体内ビオチン生成酵素反応の測定)
3. 1 E.Coli-細胞抽出物の製造
プラスミドpBO30A-15/9を含むE.Coli XLI-Blue(DSM 7246)からの細胞抽出物(抽出物Z)、およびプラスミドpBO74ΔBを含むE. Coli XLI-Blueからの細胞抽出物(DSM 7245、抽出物W)を製造した。これには、微生物の細胞を、20 g/l栄養肉汁、5 g/l酵母抽出物および20 mg/l Cmを含む培地中、37℃、OD600 2、体積800 l で培養した。細胞は濾過により収穫し、続いて5000 x gで15分問遠心分離した。
この生体内試験では、酵素ビオチン生成酵素により、14C-標識を付けたデチオビオチン(0.1 μCi; 1.95 nmol)を14C-標識を付けたビオチンに変換する反応、または標識を付けてないデチオビオチンを35S-標識を付けたシステイン(20μCi;1.32 nmol)で35S-標識を付けたビオチンに変換する反応で試験した。その際形成される14Cビオチンまたは35S-ビオチンは、抽出後、定量HPLCにより「オン−ライン」放射線化学検出器で、または薄層クロマトグラフィーおよびそれに続くオートラジオグラフィーでX線フィルムを載せることにより半定量的に、容易に測定することができる。
説塩した無細胞抽出物Zを、実施例3.2と同様にデチオビオチンおよびコファクターSAH,TPP,NADPHおよびFe++グルコン酸塩で培養したとき、デチオビオチンのビオチンへの変換はまったく観察されなかった。システイン(332μM)およびアスパラギン(15mM)またはシステインおよびアスパラギン酸塩(15mM)またはシステインおよびグルタミン(15mM)またはシステインおよびセリン(15mM)を実施例3.4によるコフアクターとともにこの無細胞抽出物に加えたところ、ビオチンの生産が確認された。
実施例3.3に記載したものと同じ脱塩した細胞抽出物を、L-システイン、アスパラギン酸、デチオビオチン、SAM、TPP,NADPHおよびFe++-グルコン酸塩で培養した場合、デチオビオチンはビオチンに変換された。 これらのコフアクターのビオチン生成酵素反応に対する影響を試験するために、これらを個別に、および相互の組合せで使用した。 これらすべてのコフアクターの唯一の組合せだけがビオチン生成酵素活性を示した。 コフアクターのーつが欠けても、ビオチン生成酵素活性は測定できなかった、すなわちすべてのコフアクターがビオチン生成酵素活性に必要である(実施例3.3、表II)。
デチオビオチンのビオチンへの変換には、ビオチン生成酵素に加えて多くのタンパク質が関与していることの証拠に、まず、無細胞抽出物Zを硫酸アンモニウム分別を行なった。これは4℃で30分間撹拌しながら25%硫酸アンモニウムの飽和で行なった。次いで、10'000 x gで30分間遠心分離し、得られたペレットを廃棄した。得られた上澄みを70%硫酸アンモニウムで飽和させたところ、ビオチン生成酵素が沈殿した。この沈殿物を少量の100 mM HEPES-緩衝液(pH 7.5)中に再分散させ、脱塩し(Sephadex G25M PD-10)、次いでアニオン交換体タロマトグラフイー(Q-Sepharose Fast-Flow, Pharmacia)を使用し、100 mM-1M HEPES-緩衝液(pH 7.5)の連続勾配で精製した。ビオチン生成酵素活性を有する画分を濃縮(Amicon Ultrafiltrationszelle, YM-10 Membr-an)し、すでに説明したように脱塩し、続いてQ-Sepharose”Hi-Load”アニオン交換体-クロマトグラフィ−カラムで再度クロマトグラフィーにかけた(Pharmacia; 20 mM 卜リス緩衝液 (pH 7.5)1 mM DTTおよびO-1M NdCl-勾配を有する)。ビオチン生成酵素活性が高い画分を一つに合わせ、濃縮し、脱塩した。これらの画分では、ビオチン生成酵素は、ビオチン生成酵素活性に必要な他のタンパク質で最早汚染されていない。
このために、45%および55%飽和の硫酸アンモニウムで抽出物を順次沈殿させた。
硫酸アンモニウムを加えた後、全体を4℃で30分間撹拌し、続いて10' 000 x gで30分間遠心分離した。45%硫酸アンモニウム飽和で得られた沈殿物を100 mMの HEPES緩衝液(pH 7.5)中に再分散させた。続いて45%-沈殿物、55%-沈殿物および55%-上澄みからアリコー卜を採取し、脱塩した(Sephadex G25M PD-10カラム)。 個々の画分を個別に、および相互の組合せで実施例3.2により試験した。
-硫酸アンモニウム45%飽和からの沈殿
-硫酸アンモニウム55%飽和からの上澄み液
が得られた。
抽出物W(実施例2.2)から硫酸アンモニウム55%飽和の後に得られた上澄み液を脱塩し(Sephadex G25M PD-10カラム)、続いてアニオン交換体クロマトグラフィーカラム(Q-Sepharose Fast-F1ow (Pharmacia) )上に載せた。このカラムは、1 mM DTTを含む20 mM 卜リス緩衝液(pH 7.5)であらかじめ平衡化した。結合していない物質を、この緩衝液で洗浄することにより除去した。カラムに結合したタンパク質は、連続NaCl勾配(0-1M)で溶離させた。溶離したタンパク質画分を一つに合わせ、濃縮し(Amicon限外濾過セル、YM-10薄膜)、脱塩し(Sephadex G25M PD-10)、続いて20 mMトリス緩衝液(pH7.0、1mM DTTを含む)で平衡化したMono Q-アニオン交換体クロマトグラフィーカラムで精製した。次いで精製した画分をSDS-PAGEで試験した。
抽出物Wをアニオン交換体クロマトグラフィーカラム(Q-Sepharose Fast-Flow(Pharmaeia))上に載せた。このカラムは、1mM DTTを含む20 mM トリス緩衝液(pH 7.5)であらかじめ平衡化した。カラムに結合したタンパク質は、連続NaCl勾配(0-1M)で溶離させた。溶離したタンパク質画分を一つに合わせ、実施例3.7と同様に濃縮し、説塩し、Mono Q-アニオン交換体クロマトグラフィーカラム上に載せた。このカラムに結合したタンパク質は、連続NaCl勾配(20 mMトリス緩衝液0-0.4M)で溶離させた。続いて、一つに合わせた溶離タンパク質抽出物(すでに説明したように濃縮し、脱塩した)をSuperose 12 Prep.ゲル濾過-クロマトグラフィーカラム(pharmacia、20 mMトリス-緩衝液で平衡化)上に、次いでSephacryl HR100-ゲル濾過-カラム(pharmacia、20 mMトリス緩衝液で平衡化)上に載せた。20mMトリス緩衝液で溶離させた後、別のタンパク質が均質な状態で得られた(SDS-PAGEで検査)。このタンパク質を含む画分を実施例3.7の試験システムと同様にして同定した。ビオチン生成酵素活性は、この画分を加えた後にのみ測定した。
精製したビオチン生成酵素(実施例3.2)は、精製したフラボドキシンおよびフェレドキシン(フラボドキシン)-NADP+-還元酵素、および必要なコファクターならびにアミノ酸と、ビオチン生成酵素活性を有していなかった。活性を得るために、45%硫酸アンモニウム画分中のさらに別の1種以上のタンパク質を探した。
無細胞抽出物Zに、フラボドキシン、フェレドキシン(フラボドキシン)NADP+-還元酵素およびビオチン生成酵素反応に関与する1種以上のタンパク質を加えた。
タンパク質、コフアクターおよびアミノ酸を加えることにより、ビオチン生成酵素反応が強化された(表III)。
これらの成分の、精製したビオチン生成酵素によるビオチン生成酵素反応に対する影響を試験するために、これらを個別に、または組合せて使用した。コファクターは実施例3.4と、アミノ酸は実施例3.3と同じ量で使用した。これらの成分がすべて存在する場合、精製したビオチン生成酵素によりデチオビオチンはビオチンに完全に変換された。これらの成分の一つが欠けると、活性はまったく測定されなかった。そのため、デチオビオチンの変換にはこれらのすべての成分が必要である(表IV)。
Claims (5)
- ビオチン生成酵素(biotin synthase)を使用し、無細胞系でデチオビオチンをビオチンへ変換することを含むビオチンの製造方法において、前記変換を、
a)チアミンピロリン酸塩、
b)NADPH、
c)S-アデノシルメチオニン、
d)Fe2+イオン、
e)システイン、および
f)アスパラギン、アスパラギン酸、グルタミンおよびセリンからなる群より選択される少なくとも1種の他のアミノ酸
の存在下に行なうことを特徴とする方法。 - 前記変換が、フラボドキシンおよびフェレドキシン(フラボドキシン)-NADP+-還元酵素の存在下で行われることを特徴とする請求項1に記載の方法。
- 前記変換が、Escherichia Coli細胞抽出物の45%飽和における硫酸アンモニウム沈殿により得られるタンパク質画分の存在下で行われることを特徴とする請求項1または2に記載の方法。
- 前記ビオチン生成酵素(biotin synthase)が、精製されたビオチン生成酵素であることを特徴とする請求項1または2に記載の方法。
- 前記変換が、pH6〜9および温度4〜50℃で行われることを特徴とする請求項1〜4の何れか1項に記載の方法。
