Description
本出願は、2009年4月30日に出願された米国仮出願第61/174,473号の優先権の利益を主張し、当該内容は引用によりその全てが本明細書に組み込まれる。
本発明は、1,3-ブタンジオール(1,3-BDO)の産生を触媒する酵素をコードする遺伝子を発現する非天然微生物に一部関する。本明細書に開示された1,3-ブタンジオールの産生のための経路は、3つの前駆体:(i)D-アラニン、(ii)アセトアセチル-CoA、及び(iii)4-ヒドロキシブチリル-CoAに基づいている。これらの経路を成功裏に操作することは、十分な活性及び特異性を有する適切なセットの酵素を同定すること、該経路の相応する遺伝子を産生宿主へとクローニングすること、発酵条件を最適化すること、及び発酵後の産物形成についてアッセイすることを必要とする。

(アセトアセチル-CoAを中間体として使用する1,3-BDO合成)
本実施例は、アセトアセチル-CoAを前駆体として使用する1,3-ブタンジオールを産生することのできる微生物の作出を説明する(図2における工程G、H、及びI)。
(4-ヒドロキシブチリル-CoAを中間体として使用する1,3-BDO合成)
本実施例は、4-ヒドロキシブチリル-CoAを前駆体として使用して1,3-ブタンジオールを産生することのできる微生物の作出を説明する(図3における工程A、工程B、及び工程E)。
Claims (9)
(a) 前記3-ヒドロキシブチリル-CoA還元酵素(アルデヒド形成)が、acr1、sucD、bphG、bld、adhE、Msed_0709、mcr、asd-2、Saci_2370、Ald、及びeutEからなる群から選択される1つ以上の遺伝子によってコードされる、
(b) 前記3-ヒドロキシブチルアルデヒド還元酵素が、alrA、ADH2、yqhD、bdh I、bdh II、adhA、4hbd、adhI、P84067、mmsb、dhat、及び3hidhからなる群から選択される1つ以上の遺伝子によってコードされる、又は
(c) 前記アセトアセチル-CoA還元酵素(ケトン還元)が、thrA、akthr2、hom6、hom1、hom2、fadB、fadJ、Hbd2、Hbd1、hbd、HSD17B10、phbB、phaB、Msed_1423、Msed_0399、Msed_0389、Msed_1993、adh、adhA、adh-A、mdh、ldhA、ldh、及びbdhからなる群から選択される1つ以上の遺伝子によってコードされる、前記非天然大腸菌。
前記非天然大腸菌が、培地中にあり、該培地中の酸素の量が、溶存酸素について飽和の10%未満である、前記非天然大腸菌。
前記少なくとも3つの外来性核酸のうちの少なくとも1つが、異種性核酸である、前記非天然大腸菌。
