Description
本発明は医薬品製剤の分野、より詳しくは増殖性疾患及び自己免疫及び/又は炎症の要素を有する疾患を治療する場合に使用するための拮抗剤抗CD40抗体を含む安定な液体医薬組成物に関する。
遺伝子操作技術の開発における近年の進歩は薬品としての使用のために十分に多くの種々の生物学的に活性なポリペプチドを提供している。しかしながらポリペプチドは変性及び可溶性及び不溶性の凝集体の形成を包含する物理的不安定性、及び、加水分解、酸化及び脱アミド化のような種々の化学的不安定性の結果として生物学的活性を損失する場合がある。液体医薬品製剤中のポリペプチドの安定性は、例えばpH、イオン強度、温度、凍結−解凍の反復サイクル、及び加工中に起こるもののような機械的剪断力への曝露のような要因により影響される場合がある。凝集体形成及び生物学的活性の損失はまた、保存バイアル内部の溶液中及び気液界面での物理的攪拌及びポリペプチド分子相互作用の結果としても起こり得る。別の立体構造変化は、輸送中の攪拌又は別様に起因する界面の圧縮−伸長の間に気液及び固液界面に吸着したポリペプチドにおいて起こり得る。そのような攪拌はタンパク質をもつれさせ、凝集させ、粒子形成させ、最終的には他の吸着されたタンパク質とともに沈潜させる場合がある。タンパク質医薬品の安定性に関する一般的検討は例えばManning等(1989)Pharm.Res.6:903−918及びWang and Hanson(1988)J.Parenteral Sci.Tech.42:S14を参照できる。
即ち、CD40LによるCD40の係留とその後のCD40シグナリングの活性化は正常な免疫応答のための必要な工程であるが;CD40シグナリングの脱調節は疾患をもたらす場合がある。CD40シグナリング経路は自己免疫疾患に関与することが分かっている(Ichikawa等(2002)J.Immunol.169:2781−2787及びMoore等(2002)J.Autoimmun.19:139−145)。更に又、CD40/CD40L相互作用は炎症プロセスにおいて重要な役割を果たしている。例えば、CD40とCD40Lの両方がヒト及び実験的なアテローム性動脈硬化症患部で過剰発現される。CD40刺激はマトリックス分解酵素の発現及び組織因子発現をアテローム関連細胞型、内皮細胞、平滑筋細胞及びマクロファージにおいて誘導する。更に又、CD40刺激はプロ炎症性サイトカイン、例えばIL−1、IL−6及びIL−8及び接着分子、例えばICAM−1、E−セレクチン及びVCAMの生産を誘導する。CD40/CD40L相互作用の抑制は動物モデルにおいてアテローム形成を防止する。移植モデルにおいて、CD40/CD40L相互作用のブロッキングは炎症を防止する。CD40/CD40L結合はアルツハイマーのアミロイドベータペプチドと相乗作用的に作用して、小グリア細胞の活性化を増進することにより、神経毒をもたらす。慢性関節リューマチ(RA)を有する患者においては、CD40発現は関節の軟骨細胞において増大しており、即ち、CD40シグナリングは損傷性のサイトカイン及びマトリックスメタロプロテイナーゼの生産に寄与していると考えられる。Gotoh等(2004)J.Rheumatol.31:1506−1512を参照できる。

治療上又は予防上活性な成分としての拮抗剤抗CD40抗体を含む安定な液体医薬組成物及びその調製において有用な方法を提供する。これらの組成物は拮抗剤抗CD40抗体、約pH5.0〜約pH7.0に組成物のpHを維持するための緩衝剤、及び液体組成物をほぼ等張性とするために十分な量の塩酸アルギニンを含む。1つの実施形態において、緩衝剤はクエン酸塩/クエン酸緩衝液であり、拮抗剤抗CD40抗体はCHIR−12.12又はCHIR−5.9拮抗剤抗CD40抗体又はその抗原結合フラグメントであり、組成物は等張性付与剤として塩酸アルギニンを含み、組成物は更にノニオン性界面活性剤及び/又はL−メチオニンを追加的安定化剤として含む。本発明の安定な液体拮抗剤抗CD40抗体含有医薬組成物は、増殖性疾患及び自己免疫及び/又は炎症の要素を有する疾患を治療するための方法において使用される。
本発明の好ましい実施形態では、例えば以下の液体医薬組成物などが提供される:

安定な液体医薬組成物であって、以下:
(a)治療上又は予防上活性な成分としての拮抗剤抗CD40モノクローナル抗体であって、該モノクローナル抗体はヒトB細胞の表面上に発現されるヒトCD40抗原に特異的に結合することができ、該モノクローナル抗体は該B細胞の表面上に発現されるCD40抗原に結合している場合に有意なアゴニスト活性を有さない、抗体;
(b)該組成物をほぼ等張性とするために十分な量の酸性形態でのアルギニン(塩酸アルギニン);及び、
(c)約pH5.0〜約pH7.0の範囲内に該組成物のpHを維持するための緩衝剤であって、該緩衝剤はクエン酸塩/クエン酸緩衝剤である、緩衝剤;
を含む、安定な液体医薬組成物。
(項目2)
前記組成物が約240mmol/kg〜約360mmol/kgの浸透圧重量モル濃度を有する項目1記載の組成物。
(項目3)
前記緩衝剤の濃度が約5mM〜約100mMである項目1記載の組成物。

h)配列番号10又は配列番号12に示されるヒトCD40配列の残基82〜89を含むエピトープに結合するモノクローナル抗体;
i)競合的結合試験においてモノクローナル抗体CHIR−5.9又はCHIR−12.12と競合するモノクローナル抗体;
j)前述の項目a)のモノクローナル抗体又は前述の項目c)〜i)の何れか1つのモノクローナル抗体であって、該抗体は組み換え産生される、抗体;及び、

a)モノクローナル抗体CHIR−5.9又はCHIR−12.12;
b)ハイブリドーマ細胞株5.9又は12.12により生産されるモノクローナル抗体;c)配列番号6に示される配列、配列番号7に示される配列、配列番号8に示される配列、配列番号6及び配列番号7に示される配列の双方、及び配列番号6及び配列番号8に示される配列の双方よりなる群から選択されるアミノ酸配列を含むモノクローナル抗体;

本発明の医薬組成物は抗CD40抗体、特にCD40受容体をターゲティングし、ADCCを調節し、CD40シグナリング、特にCD40のCD40リガンド(CD40L)との相互作用又は両方により媒介されるCD40シグナリング経路に干渉する拮抗剤抗CD40抗体又はその抗原結合フラグメントを含む。「CD40抗原」、「CD40細胞表面抗原」、「CD40受容体」又は「CD40」とは、腫瘍壊死因子(TNF)受容体ファミリーに属する膜貫通糖タンパク質を意図している(例えば5,674,492及び4,708,871;Stamenkovic等(1989)EMBO8:1403;Clark(1990)Tissue Antigens36:33;Barclay 等(1997)The Leucocyte Antigen Facts Book(第2版;Academic Press,San Diego)を参照できる)。この遺伝子のオルタナティブスプライシング転写物変異体によりコードされているヒトCD40の2つのアイソフォームが発見されている。第1のアイソフォーム(「長鎖アイソフォーム」又は「アイソフォーム1」としても知られている)は、最初の19残基により示されるシグナル配列を有する277アミノ酸前駆体ポリペプチド(配列番号11(ゲンバンクアクセッション番号X60592及びNM_001250参照)によりコードされる配列番号12(最初はゲンバンクアクセッション番号CAA43045として報告され、ゲンバンクアクセッション番号NP_001241においてアイソフォーム1として識別されている))として発現される。第2のアイソフォーム(「短鎖アイソフォーム」又は「アイソフォーム2」としても知られている)は、最初の19残基により示されるシグナル配列も有する203アミノ酸前駆体ポリペプチド(配列番号9(ゲンバンクアクセッション番号NM_152854)によりコードされる配列番号10(ゲンバンクアクセッション番号NP_690593))として発現される。ヒトCD40のこれらの2つのアイソフォームの前駆体ポリペプチドはそれらの最初の165残基を共有している(即ち配列番号10及び配列番号12の残基1〜165)。短鎖アイソフォームの前駆体ポリペプチド(配列番号10に示す)は翻訳のフレームシフトをもたらすコーディングセグメントを欠いている転写物変異体(配列番号9)によりコードされており;結果として生じるCD40アイソフォームはより短く、CD40の長鎖アイソフォームに含有されるもの(配列番号12の残基166〜277に示されるC末端)とは異なるC末端(配列番号10の残基166〜203)を含有している。本発明の目的のためには、「CD40抗原」、「CD40細胞表面抗原」、「CD40受容体」又は「CD40」という用語はCD40の短鎖及び長鎖アイソフォームの両方を包含する。
FcγRIIA(活性化受容体)及びFcγRIIB(抑制受容体)を包含し、これらはその細胞質ドメインにおいて主に異なる同様のアミノ酸配列を有する。活性化受容体FcγRIIAはその細胞質ドメインに免疫受容体チロシン系活性化モチーフ(ITAM)を含有している。抑制受容体FcγRIIBはその細胞質ドメインに免疫受容体チロシン系抑制モチーフ(ITIM)を含有している(Daeron(1997)Annu.Rev.Immunol.15:203−234参照)。FcRはRavetch and Kinet(1991)Annu.Rev.Immunol.9:457−492(1991);Capel等(1994)Immunomethods 4:25−34;and de Haas等(1995)J.Lab.Clin.Med.126:330−341において考察されている。他のFcR、例えば将来発見されるものも本明細書における「FcR」という用語に包含される。用語は又、母動物IgGの胎仔への転移を担っている生仔受容体FcRnも包含する(Guyer等(1976)J.Immunol.117:587 and Kim等(1994)J.Immunol.24:249(1994))。
本発明の医薬組成物において使用するための抗体、例えば本明細書に開示した拮抗剤抗CD40抗体は当該分野で知られた何れかの抗体製造方法を用いて製造できる。即ち、ポリクローナル血清は従来の方法により製造してよい。一般的に、目的の抗原、CD40抗原を含有する溶液を先ず使用して適当な動物、好ましくはマウス、ラット、ウサギ、又はヤギを免疫化する。ウサギ又はヤギは入手できる血清の量及び標識された抗ウサギ及び抗ヤギ抗体の入手し易さのため、ポリクローナル血清の製造のために好ましい。
本発明の医薬組成物は当該分野で知られている種々の拮抗剤抗CD40抗体を用いて製剤できる。そのような変異体は親抗体の所望の結合特性を保持していることになる。即ち、例えば製剤すべき拮抗剤抗CD40抗体が親のCHIR−12.12又はCHIR−5.9抗体の変異体である場合、変異体抗体は親CHIR−12.12又はCHIR−5.9抗体の結合特性を保有することになる。抗体変異体を作成するための方法は一般的に当該分野で使用できるものである。
本発明の医薬組成物はCD40発現細胞が関連する癌又は前悪性の状態を有する対象を治療する場合に、又はCD40発現細胞が関連する炎症性疾患及び/又は自己免疫疾患を治療するために使用される。「治療」とは本明細書においては、対象に対する拮抗剤抗CD40抗体を含む医薬組成物の適用又は投与、又は、対象に由来する単離された組織又は細胞系統に対する拮抗剤抗CD40抗体を含む医薬組成物の適用又は投与として定義され、ここで対象は疾患、疾患の症状、又は疾患に至る素因を有し、ここで目的は疾患、疾患の症状、又は疾患に至る素因の治癒、療養、軽減、緩和、改変、緩解、改良、改善又は影響を及ぼすことである。
約3mg/kg〜約30mg/kg、約3mg/kg〜約25mg/kg、約3mg/kg〜約20mg/kg、約5mg/kg〜約15mg/kg、又は約7mg/kg〜約12mg/kgの範囲である。投与方法は拮抗剤抗CD40抗体又はその抗原結合フラグメントの治療有効用量の単回投与または治療有効用量の多数回投与を含んでよい。
溶液の条件(例えばpH、緩衝物質種、及びイオン強度)及び賦形剤(例えば界面活性剤及び安定化剤)は液体製剤中のタンパク質の安定性のための重要な要因である。CHIR−12.12の物理化学的安定性はpH5.5において最適となる。しかしながらCHIR−12.12タンパク質は望ましくない溶液条件下では凝集形成及びフラグメント化を介して分解する場合があり;更に又、特に過酸化物の夾雑物及び/又はTweenのような生原料賦形剤と共に導入される金属の痕跡量の存在下において、酸化する場合もある。以下の実験は最適pH5.5において製剤した場合に、凝集形成、フラグメント化及び酸化に対抗してCHIR−12.12を安定化させるための最良の鑑賞物質種及び適切な賦形剤を発見するために実施した。
試験のためのCHIR−12.12薬品物質(DS)ロットはCHO誘導精製バルクのロット#CD021105A及びロット#PD010705Aであった。DSロットはXoma,Ltd(Berkeley,CA)において製造された。本試験のための製剤試料は該当する緩衝溶液に対するDSの透析とその後のTween所望量の強制添加により調製した。全試料中のCHIR−12.12タンパク質の濃度は約20mg/mlであった。
サイズ排除クロマトグラフィー(SEC)
SEC−HPLCは分子量が低下する順序で分子を分離する。その結果、CHIR−12.12凝集体はHPLCカラムから溶出する最初のものであり、その後、単量体が続き最後にフラグメントが溶出する。CHIR−12.12の純度、凝集及びフラグメント化は、0.7mL/分の流量において移動相として50mMリン酸ナトリウム、200mMNaCl、pH7.0を用いながらTosohaasTSK−GEL3000SWX1カラムを有するWaters AllianceHPLCにより分析した。
CHIR−12.12の酸化はWaterAlliance HPLCシステムを用いながらToshoTSKゲルブチル−NPRカラム、移動相Aとしての2M硫酸アンモニウム/20mMTris、pH7.0、及び移動相Bとしての20mMTris、pH7.0を用いて流量1.0ml/分で測定した。CHIR−12.12抗体をパパインで消化することによりFabおよびFcフラグメントが生じる。CHIR−12.12の酸化生成物は酸化Fcフラグメント(metSO)であり、これはHPLCカラムから主要なFab物質種と主要なFc物質種の間に溶出する前Fc物質種である。
凝集及びフラグメント化に対するクエン酸塩緩衝液の安定化作用
DSロット#PD010705Aに由来するCHIR−12.12を150mMNaCl、0.1%(w/v)Tween80及びpH5.5において、10mMのクエン酸塩、酢酸塩、コハク酸塩、又はリン酸塩の緩衝溶液中20mg/mlで製剤した。製剤試料は3ccのガラスバイアル内に1.2ml溶液として充填し、5℃、25℃および40℃において保存した。CHIR−12.12安定性試料はSEC試験により所定の時点において分析した。
CHIR−12.12安定性試料を0.1%及び0.2%(w/v)のTween80を用いながらクエン酸塩、酢酸塩及びコハク酸塩中に調製した。試料を5℃、25℃および40℃において保存し、所定の時点において酸化に関して疎水性相互作用クロマトグラフィー(HIC)により分析した。CHIR−12.12の酸化生物はPreFcピーク物質種の合計のパーセント、すなわちPre−Fc%として測定した。表1に示す結果はクエン酸塩系の製材がコハク酸塩及び酢酸塩の緩衝製剤よりも少ない酸化生成物を形成し、クエン酸塩緩衝液がCHIR−12.12の酸化を最小限としたことを示している。これらの結果は、クエン酸が恐らくはキレート形成剤として作用することによりCHIR−12.12タンパク質の微量金属誘導酸化を抑制したことを示唆している。
DSロット#CD010705Aを10mMクエン酸ナトリウム/クエン酸、150mMNaCl、0.1%Tween80又はTween20、並びに種々の量(0〜5mM)のL−メチオニン中、20mg/mlに製剤した。製剤試料は3ccのガラスバイアル内に2.5mlを充填し、40℃において保存した。表2は初期及び40℃3か月における試料に関するHIC結果を示す。初期においては全製剤中のCHIR−12.12の酸化は元のバルク薬品物質(DS)ロット#CD010705Aと同等であった。L−メチオニンを含有しない製剤における酸化のレベルは40℃3か月では倍加を超過していた。しかしながらL−メチオニンを含有する製剤における酸化のレベルは40℃における3か月の保存中を通して殆ど変化していなかった。
以下の試験は静脈内注入を介した投与を意図した液体医薬組成物として製剤されたCHIR−12.12の長期保存安定性のために張力付与剤および安定化剤を選択することを目的とした。タンパク質の非経腸製品に対してはNaClが最も一般的に使用されている張力付与剤であるが、抗体治療薬に対しては最適な安定化作用を有していない。本試験は水性製剤中のCHIR−12.12に対する塩化ナトリウム及びアルギニンの酸性形態(塩酸アルギニン)の比較対照させた安定化作用に関して報告する。
本試験のために使用したCHIR−12.12薬品物質(DS)はCHO誘導精製バルクのロット#CD021105Aであった。DSロットはXoma Ltd(Berkeley,CA)において製造された。

分析方法
示差走査熱量測定(DSC)
CHIR−12.12製剤試料の立体構造的安定性は、MicroCalVP−DSCを用いながら、1℃/分で15℃〜90℃に加熱することにより評価した。
CHIR−12.12の純度、凝集、及びフラグメント化は、0.7mL/分の流量において移動相として50mMリン酸ナトリウム、200mMNaCl、pH7.0を用いながらTosohaasTSK−GEL3000SWXLカラムを有するWaters AllianceHPLCにより分析した。
CHIR−12.12の純度もまた非還元及び還元条件下において12%Tris−グリシンゲルを用いながら評価した。タンパク質はクーマシーブルー染色により検出した。
CHIR−12.12の電荷変化関連脱アミド化は、Dionex PropacWCX−10カラムを有するWaterAlliance HPLCシステム、移動相Aとしての50mMHEPES、pH7.3及び移動相Bとしての500mMNaCl含有50mMHEPES、pH7.3を用いながら、0.8ml/分の流量で測定した。

図7は上記「材料」のセクションにおいて記載した2種の製剤中のCHIR−12.12のDSCセオグラムを示す。CHIR−12.12の熱的展開(thermal unfolding)は少なくとも2つの熱遷移を呈し、恐らくはそれぞれFab及びFcドメインの展開/融解を示していると考えられる。より高温においては、タンパク質はおそらくは凝集し、DSCシグナルの損失をもたらす。本試験においては、最低の熱遷移温度は融解温度Tmとして定義した。塩酸L−アルギニン含有製剤はNaCl含有製剤よりも高値のTmを呈し、塩酸L−アルギニンがNaClを用いた場合よりも高い立体構造的安定性を有するCHIR−12.12を与えることが示唆された。
6か月まで5℃でインキュベートしたのち、SEC−HPLCによれば塩酸L−アルギニン及びNaCl含有製剤中ではタンパク質の凝集体およびフラグメントの量は無視できる程度であり(<0.5%)、2種の製剤の間には安定性にほとんど差がなかった(データ示さず)。加速保存条件下、即ち25℃6か月においては、塩酸L−アルギニン含有製剤は図8に示す通りより高値のパーセントの単量体を含有していた。単量体に費やされたため、凝集体およびフラグメントの含量は両方とも保存時間とともに緩徐に増大した。しかしながら、塩酸L−アルギニン含有製剤が発生した凝集体及びフラグメンはそれぞれ図9及び10に示す通りNaCl含有製剤よりも少量であった。同様に、40℃で保存した場合、塩酸L−アルギニン含有製剤はそれぞれ図11、12及び13に示す通り、NaCl含有製剤の場合よりも高値のパーセントの単量体及び低値のパーセントの凝集体及びフラグメントを示していた。40℃4カ月保存により、塩酸L−アルギニン含有製剤では残存単量体91.8%、凝集体1.7%、及びフラグメント6.5%であったのに対し、NaCl含有製剤の場合は、単量体87.9%、凝集体2.2%、及びフラグメント9.9%であった。SEC−HPLCの結果は塩酸L−アルギニンはNaClと比較して、CHIR−12.12タンパク質の安定性を向上させることを示している。
表3は非還元(NR)及び還元(R)条件下に分析した塩酸L−アルギニン及びNaCl含有製剤に関するSDS−PAGEの結果を示す。CHIR−12.12の純度は非還元条件下の主要バンドのパーセントとして、又は、還元条件下の重鎖及び軽鎖の合計のパーセントとして測定した。ゼロ時を除き、塩酸L−アルギニン含有製剤は非還元及び還元条件下の両方においてNaCl含有製剤の場合よりも高値の純度を示した。SDS−PAGEの観察結果はNaClよりも塩酸L−アルギニンがCHIR−12.12の安定性を増大させたSEC−HPLCの結果と合致していた。
CIEX−HPLCは酸性の変異体が主要ピーク物質種の前に溶出し、、塩基性の変異体が主要ピーク物質種の後に溶出するように電荷に基づいて分子を分離する。CHIR−12.12の純度及びその脱アミド化産物の含有量をそれぞれ主要ピークのパーセント及び酸性変異体のパーセントとして測定した。
CHIR−12.12バルク薬品物質の凍結保存は幾つかの理由、例えば向上した製品の安定性及びシェルフライフ、低下した微生物生育、並びに輸送中の発泡の排除などのために、液体保存よりも好ましい。しかしながら凍結及びその後の解凍は、氷/液体の界面及び溶液の濃度勾配をもたらすことにより、タンパク質溶液にストレスをもたらす場合がある。ストレスはタンパク質を変成させ、凝集、及び悪い例においては目視可能な粒子又は沈殿物の形成をもたらす場合がある。タンパク質の凝集体は低減した薬剤の力価及び増大した免疫原性を伴う場合が多いため、タンパク質製剤の成分及び凍結解凍条件を最適化することにより凝集を最小限とすることは極めて重要である。
CHIR−12.12バルクDSロット#UA7870、#TC23−2、#UB1291、#PD010705A、及び#CD083005Aを本試験のために使用した。Tween80、Tween20、Brij35、及びプルロニックF68はそれぞれ、Sigma、J.T.Baker、Alfa Aesar及びMediaTechCellgroより購入した。CHIR−12.12薬品物質の凍結保存用のポリカーボネート(PC)ビンはNalgeneから購入した。
対照試料を除いて、全ての他の試料は−20℃及び−60℃における完全凍結とその後の多数サイクル反復する周囲温度における完全解凍に付した。
凍結解凍試験のための試料はCHIR−12.12薬品物質ロット#UA7870及び#TC23−2から作成した。全試料は10mMクエン酸ナトリウム/クエン酸、150mMNaCl、及びpH5.5の緩衝溶液中、透析とその後の種々のパーセント(0〜0.5%w/v)の以下のノニオン系界面活性剤、即ちTween80、Tween20、Brij35、及びプルロニックF68の1つを添加することにより製剤した。2.5mlの各試料をガラスバイアル中に充填し、−60℃で一夜凍結し、最大8サイクルまで周囲温度において完全解凍した。初期(ゼロ時)及び各凍結解凍サイクル後の試料を透明性及び目視可能な沈殿/凝集体に関して目視的に検査した。
CHIR−12.12薬剤物質ロット#UB1291及び#PD010705Aを溶液(20mg/mlCHIR−12.12、10mMクエン酸ナトリウム/クエン酸、150mMNaCl、pH5.5)中に製剤し、その後0〜0.5%(w/v)のTween80又はTween20を添加した。製剤試料20mlの小分量ずつを125ccのポリカーボネートのビンに充填し、−60℃凍結及び周囲温度解凍に付した。凍結解凍5サイクルの後、HIAC−Royco液体粒子計数システムを用いて≧10μm及び≧25μmの目視可能に至らない凝集体に関して試料を測定した。
5サイクルの凍結解凍の前後において、製剤(20mg/mlCHIR−12.12、10mMクエン酸ナトリウム/クエン酸、150mM塩酸L−アルギニン、5mML−メチオニン、0〜0.2%Tween20及びpH5.5)を動的光散乱分析器を用いて凝集体に関して評価した。
目視可能な粒子の評価
表4は目視による観察の結果を総括したものである。凍結解凍サイクルを開始する前であるゼロ時において、試料は全て僅かに不透明であるが目視可能な凝集体/沈殿は含有していなかった。1回の凍結解凍サイクルの後、界面活性剤無添加の製剤全てにおいて、Tween80を0〜0.05%(w/v)含有する製剤において、及び0〜0.1%(w/v)Brij35を含有する製剤において、並びに0〜0.5%(w/v)プルロニックF68を含有する試料において、数個の目視可能な凝集体/沈殿が形成していた。0.0%〜0.5%(w/v)Tween20を含有する試料には凍結解凍8サイクル全体に渡って如何なる凝集体及び沈殿も観察されなかった。このことは、Tween20は多数回の凍結解凍サイクルに起因する大型の不溶性凝集体の形成を防止することにおいてTween80よりも有効であることを示唆している。Brij35及びプルロニックF68はTween80及びTween20よりもはるかに低有効性であった。
表5は種々の濃度のTween80を含有するクエン酸塩/クエン酸緩衝製剤のml当たりの目視可能に至らない凝集体の計数値を示す。Tween80濃度の上昇に伴って目視可能に至らない粒子の計数値の低下傾向が観察され、目視可能に至らない粒子の計数の低下はTween80が0.1%(w/v)を超えている場合に顕著さが低下し、Tween80の使用に関する適切な濃度は0.1〜0.2%(w/v)であることが示唆された。
表8はCHIR−12.12の粒子の平均の流体力学的直径、多分散性及び単量体物質種のパーセント強度を示す。動的光散乱分析は単量体物質種の100%強度で示されるとおり、凍結解凍の5サイクルの前後における全試料において唯一の単量体物質種のみを検出している。このことは全試料が主に単量体よりなるものであったことを示唆している。凍結乾燥試験5サイクルの後、流体力学的直径及び多分散性の増大により示される通り、Tween20を含有しない、0.005%(w/v)Tween20を含有する試料中では、数個の凝集体(恐らくは2量体又は3量体)が単量体と共存していると考えられる。0.025〜0.1%(w/v)Tween20を含有する試料は流体力学的直径及び多分散性の値の変化を殆ど示しておらず、それらがそれほど凝集体形成をすることなく単量体の以前のレベルであり続けたことを示している。
抗CD40抗体の拮抗剤活性を評価するために以下の試験を使用することができる。これらの試験のためのヒトB細胞は例えば本質的にDe Groot等(1990)Lymphokine Research(1990)9:321に記載されている通り、扁桃腺切除術を受けた個体から得られた扁桃腺から単離することにより得ることができる。慨すれば、組織をメスの刃を用いて分散させ、貪食細胞及びNK細胞を5mML−ロイシンメチルエステル処理により枯渇させ、臭化2−アミノエチルイソチオウロニウムで処理したヒツジ赤血球(SRBC)とのロゼット形成1サイクルによりT細胞を除去する。得られたBリンパ球プレパレーションの純度は抗(CD20)mAbB1(Coulter Clone,Hialeah,FA)又は抗(CD3)mAbOKT3(Ortho,Raritan,NJ)及びウサギ抗(マウスIg)のFITC共役F(ab’)2フラグメント(Zymed,San Francisco,CA)を用いた間接的免疫蛍光標識及びFACS分析により確認できる。
B細胞(ウェル当たり4x104)を平底96穴マイクロプレート中10%うし胎児血清を添加した200μlのIMDM中で培養する。固定化抗(IgM)抗体(イムノビーズ;5μg/ml;BioRad,Richmond,California)を添加することによりB細胞を刺激する。所望により100U/mlの組み換えIL−2を添加する。種々の濃度の被験モノクローナル抗体(mAb)をマイクロ培養物発生時に添加し、第3日に18時間パルス処理後の(3H)チミジンの取り込みの測定により増殖を評価する。
Banchereau等(1989)Science251:70に記載されたものと類似の培養系においてB細胞増殖を刺激する抗CD40モノクローナル抗体の能力に関して試験するために、ヒトFcγRIIのHR対立遺伝子型を発現するマウス3T6トランスフェクション体を使用する。B細胞(ウェル当たり2x104)を10%ウシ胎児血清及び100U/ml組み換えIL−4を添加した200μlのIMEM中、1x104のトランスフェクション体細胞(5000Radで照射)の存在下に平底マイクロウェル中で培養する。B細胞添加の前に、3T6細胞を少なくとも5時間培養プラスチックに接着させる。抗CD40mAbを15ng/ml〜200ng/mlの種々の濃度で添加し、B細胞の増殖を第7日に[3H]チミジンによる18時間パルス処理後のチミジン取り込みの測定により評価する。
拮抗剤抗CD40モノクローナル抗体(mAb)はまた上記したB細胞増殖試験を用いながらS2C6(正常B細胞の増殖のCD40刺激のアゴニストとも報告されているSGN−14としても知られている;Francisco等(2000)Cancer Res.60:3225−3231)のような抗CD40抗体によるB細胞増殖の刺激を抑制するそれらの能力により特性化することもできる。ヒト扁桃腺B細胞(4x104)をセファロースビーズにカップリングさせた抗IgM(5μg/ml)及び抗CD40mAbS2C6(1.25μg/ml)の存在下にマイクロウェル中200μl中で培養する。種々の濃度の目的の抗CD40を添加し、[3H]チミジン取り込みを3日後に試験する。対照として抗(グルコセレブロシダーゼ)mAb8E4を同濃度において添加できる。Barneveldら(1983)Eur.J.Biochem.134:585。拮抗剤抗CD40抗体は例えば少なくとも75%以上mAbS2C6による抗IgM誘導ヒトB細胞増殖の同時刺激を抑制できる(即ち、拮抗剤抗CD40抗体存在下のS2C6刺激増殖は拮抗剤抗CD40抗体非存在下に観察されるものの25%以下である)。これとは対照的に、β−グルコセレブロシダーゼに対して指向された非関連のmAb8E4の等量を用いた場合には有意な抑制は観察されない。Barneveld等、上出。このような結果は、抗CD40mAbはヒトB細胞の増殖のための刺激シグナルは送達しないが、逆に、別のmAbでCD40をトリガーすることにより生じる刺激シグナルを抑制することができることを示している。
Zubler等(1985)J.Immunol.(1985)134:3662の観察によれば、EL4B5として知られているマウス胸腺腫EL−4系統の突然変異体サブクローンは、ネズミ及びヒトの両方の起源のB細胞が増殖して免疫グロブリン分泌プラズマ細胞に分化することをインビトロで強力に刺激することができる。この活性化は、抗原非依存性であり、MHC制限性ではないことがわかっている。ヒトB細胞の最適な刺激のためには、活性化ヒトT細胞由来の上澄みの存在が必要であったが、B細胞応答は又EL4B5細胞をホルボール−12−ミリステート13−アセテート(PMA)又はIL−1で予備活性化した場合にも起こっている。Zubler等(1987)Immunological Reviews 99:281;及びZhang等(1990)J.Immunol.144:2955。この培養系におけるB細胞活性化は効率的であり、限界希釈法による実験ではヒトB細胞の大多数が活性化されて増殖し、抗体分泌細胞に分化した。Wen等(1987)Eur.J.Immunol.17:887。
拮抗剤抗CD40抗体はB細胞による免疫グロブリン生産の拮抗剤として機能できる。抗CD40抗体はこの型の拮抗剤活性に関して、T細胞ヘルパー試験において活性化されたT細胞により接触依存性の態様において刺激されているB細胞による免疫グロブリン生産を抑制する抗体の能力を評価することにより試験できる。この態様において、96穴組織培養プレートを抗CD3mAbCLB−T3/3の復水1:500希釈物でコーティングした(CLB、Amsterdam、The Neterlands)。記載通り、」同時刺激性のmAb、即ち、抗CD2mAbCLB−T11.1/1及びCLB−T11.2/1た(CLB、Amsterdam、The Neterlands)、両方の復水1:1000、及び抗CD28mAbCLB−28/1(CLB、Amsterdam、The Neterlands)を添加する。その後、扁桃腺T細胞(照射、3000Rad;ウェル当たり105)、扁桃腺B細胞(ウェル当たり104)、及びrIL−2(20U/ml)を添加した。各細胞培養物の最終容量は200μlとする。8日後、細胞を回転沈降させ、細胞非含有上澄みを採取する。(希釈した)試料中のヒトIgM及びIgGの濃度は後述する通りELISAにより推定する。
ヒトIgM及びIgGの濃度はELISAにより推定される。96穴ELISAプレートを4℃で16時間インキュベートすることにより4μg/mlのマウス抗ヒトIgGmAbMH16−01(CLB,Amsterdam,The Netherlands)又は1.2μg/mlのマウス抗ヒトIgMMab4102(Tago,Burlinga,e.CA)でコーティングする。プレートをPBS−0.05%Tween20(PBS−Tween)で3回洗浄し、1時間BSAで飽和させる。2回洗浄後、プレートを被験試料の種々の希釈度で37℃1時間インキュベートする。3回洗浄後、結合したIgを1μg/mlのパーオキシダーゼ標識マウス抗ヒトIgGmAbMH16−01(CLB)又はマウス抗ヒトIgMmAbMH15−01(CLB)とともに、37℃で1時間インキュベートすることにより検出する。プレートを4回洗浄し、結合パーオキシダーゼ活性を気質としてO−フェニレンジアミンを添加することにより顕在化する。ヒト標準血清(H00,CLB)を用いて各試験に関する標準曲線を確立する。
Claims (55)
- 安定な液体医薬組成物であって、以下:
(a)治療上又は予防上活性な成分としての拮抗剤抗CD40モノクローナル抗体であって、該モノクローナル抗体はヒトB細胞の表面上に発現されるヒトCD40抗原に特異的に結合することができ、該モノクローナル抗体は該B細胞の表面上に発現されるCD40抗原に結合している場合に有意なアゴニスト活性を有さない、抗体;
(b)該組成物を240mmol/kg〜360mmol/kgの浸透圧重量モル濃度を有するものとするために十分な量の等張化剤であって、該等張化剤が50mM〜300mMの濃度の塩酸アルギニンである、等張化剤;及び、
(c)pH5.0〜pH7.0の範囲内に該組成物のpHを維持するための緩衝剤であって、該緩衝剤はクエン酸塩/クエン酸緩衝剤である、緩衝剤;
を含み、ここで、該拮抗剤抗CD40モノクローナル抗体が、以下:
(i)配列番号2に示される可変領域および定常領域の配列と、配列番号4に示される可変領域および定常領域の配列とを含む、モノクローナル抗体;
(ii)配列番号2に示される可変領域および定常領域の配列と、配列番号5に示される可変領域および定常領域の配列とを含む、モノクローナル抗体;
(iii)配列番号2に示されるリーダー領域、可変領域および定常領域の配列と、配列番号4に示されるリーダー領域、可変領域および定常領域の配列とを含む、モノクローナル抗体;ならびに
(iv)配列番号2に示されるリーダー領域、可変領域および定常領域の配列と、配列番号5に示されるリーダー領域、可変領域および定常領域の配列とを含む、モノクローナル抗体;
よりなる群から選択される、安定な液体医薬組成物。 - 前記拮抗剤抗CD40モノクローナル抗体が、特許寄託番号PTA−5543としてATCCに寄託されたハイブリドーマ細胞株から得られるモノクローナル抗体CHIR−12.12である、請求項1記載の組成物。

(a)配列番号2に示される可変領域および定常領域の配列と、配列番号4に示される可変領域および定常領域の配列とを含む、モノクローナル抗体;
(b)配列番号2に示される可変領域および定常領域の配列と、配列番号5に示される可変領域および定常領域の配列とを含む、モノクローナル抗体;
(c)配列番号2に示されるリーダー領域、可変領域および定常領域の配列と、配列番号4に示されるリーダー領域、可変領域および定常領域の配列とを含む、モノクローナル抗体;ならびに

- 液体医薬組成物中の抗CD40モノクローナル抗体の安定性を増大させるための方法であって、該方法が、該抗CD40モノクローナル抗体、該組成物を240mmol/kg〜360mmol/kgの浸透圧重量モル濃度を有するものとするために十分な量の等張化剤であって、該等張化剤が該組成物中50mM〜300mMの濃度の塩酸アルギニンである、等張化剤、及びpH5.0〜pH7.0の範囲内に該組成物のpHを維持するための緩衝剤を組み合わせることにより該組成物を製剤することを含み、該緩衝剤はクエン酸塩/クエン酸緩衝剤であり、ここで、該抗CD40モノクローナル抗体が、以下:
(a)配列番号2に示される可変領域および定常領域の配列と、配列番号4に示される可変領域および定常領域の配列とを含む、モノクローナル抗体;

(d)配列番号2に示されるリーダー領域、可変領域および定常領域の配列と、配列番号5に示されるリーダー領域、可変領域および定常領域の配列とを含む、モノクローナル抗体;
よりなる群から選択される、方法。 - 前記抗CD40モノクローナル抗体が、特許寄託番号PTA−5543としてATCCに寄託されたハイブリドーマ細胞株から得られるモノクローナル抗体CHIR−12.12である、請求項44又は45記載の方法。
- 前記抗CD40モノクローナル抗体が、CHO細胞株において生産される、請求項44又は45記載の方法。
- 前記緩衝剤の濃度が5mM〜100mMであり、50mM〜200mMの塩酸アルギニンが該組成物中にある、請求項44〜47の何れか1項に記載の方法。
- 前記緩衝剤の濃度が5mM〜20mMであり、100mM〜175mMの塩酸アルギニンが該組成物中にある請求項48記載の方法。
- 前記緩衝剤の濃度が10mMであり、150mMの塩酸アルギニンが該組成物中にある請求項49記載の方法。
- 前記緩衝剤がクエン酸ナトリウム/クエン酸緩衝剤である請求項44〜50の何れか1項に記載の方法。
- 前記組成物が5.5のpHを有する請求項51記載の方法。
- 前記組成物が更にメチオニン、ポリソルベート20、又はメチオニンとポリソルベート20の両方を含むように製剤され、該メチオニンは、存在する場合には、0.5mM〜20.0mMの濃度において該組成物中に存在し、該ポリソルベート20は、存在する場合には、0.025%〜0.1%(w/v)の濃度において該組成物中に存在する請求項44〜52の何れか1項に記載の方法。
- 前記抗CD40モノクローナル抗体が0.1mg/ml〜50.0mg/mlの濃度において前記組成物中に存在する請求項44〜53の何れか1項に記載の方法。
- 前記抗CD40モノクローナル抗体が10.0mg/ml〜35.0mg/mlの濃度において前記組成物中に存在する請求項54記載の方法。
