Description
V1が炭素である場合のW1およびV2が炭素である場合のW2は以下の、
(式中R3およびR4は水素であるか、あるいは、これらの1つがOHである;)
X1およびX2は水素、低級アルキルまたはハロゲンであり、これらの少なくとも1つはハロゲンである;
Zは炭素、酸素、硫黄、または窒素である;
R2は水素またはアルキルである;
Yは非置換またはオキソ、ハロゲン、アルキル、ヒドロキシル、アリール、または複素環基で置換された、飽和または不飽和C2−10炭化水素鎖である;
Aは−CH2OH、−COCH2OH、−COOHまたはその官能性誘導体である;そして、
R1は非置換またはハロゲン、オキソ、ヒドロキシ、低級アルコキシ、低級アルカノイルオキシ、低級シクロアルキル、低級シクロアルキルオキシ、アリール、アリールオキシ、複素環基、または複素環−オキシ基で置換された直鎖または分枝鎖を形成する飽和または不飽和低級炭化水素;低級シクロアルキル;低級シクロアルキルオキシ;アリール、アリールオキシ、複素環基または複素環−オキシ基である;
C−13位とC−14位の間の結合は二重結合または単結合である;そして、
C−15はR、Sまたはその混合物の立体配置を有する]。
(i)薬剤の親水性または浸透圧によって腸管内に水と電解質が保持され、それによって腸管内容物の体積が増加し、その結果間接的に内容物がより速く移動する。
(ii)薬剤が腸粘膜に作用して電解質および水の正常な吸収の総量が減少して水分含量が増加し、その結果間接的に腸管内容物がより速く移動する。
(iii)薬剤が腸粘膜に作用して電解質および水の正常な分泌の総量が増加して水分含量が増加し、その結果直接的および/または間接的に腸管内容物がより速く移動する。
(iv)薬剤が第一に腸管運動に作用して移動を速くし、その結果間接的に水および電解質の吸収のための時間が短くなるために水および電解質の正味の吸収が減少する。
好ましいW2はR3およびR4の両方が水素原子である。
好ましいYは非置換の6−8の炭素原子を含む飽和または不飽和炭化水素鎖である。
好ましいR2は水素原子である。

移動相:アセトニトリル(液体クロマトグラフグレード)/メタノール(液体クロマトグラフグレード)/酢酸アンモニウム水溶液(0.05mol/L)(4:11:5)の混合溶液
トリメチル−β−、またはヒドロキシプロピル−β−シクロデキストリン)、分枝シクロデキストリン(例えばグルコシル−、マルトシル−シクロデキストリン)、ホルミル化シクロデキストリン、硫黄含有シクロデキストリン、リン脂質などの安定剤を含んでいてもよい。上記のシクロデキストリンを用いる場合、シクロデキストリンに封入される化合物は安定性が増強した形になることがある。リン脂質は、リポソームを形成するために用いられることがあり、その結果安定性が増強する。
本発明の組成物における、C−16位にハロゲン原子を有するハロゲン化−二環式化合物の効果を例証するために、以下の実施例を用意し、試験した。
本発明の、一般式(I)のZが酸素原子であって、C−9位にケトンが存在する、単環式/二環式構造の比による、組成物の生物活性が、以下の実施例により理解されるであろう。C−16位のフッ素原子の数と単環式/二環式構造の比を表2に示す。
式(I)のZが酸素であって、C−9位にケトンが存在し、5,6−炭素間が二重結合である、単環式/二環式構造の比による、組成物の生物活性を、以下に示す。
3から9名の健康な男性ヴォランティアを以下の単環式/二環式構造(CDCl3中)を4:96の比で含有する組成物で処置した。
Claims (9)
- さらに6−14の炭素原子を有する脂肪酸のトリグリセリドである中鎖脂肪酸トリグリセリドを含有する、請求項1または2に記載の組成物。
- 中鎖脂肪酸トリグリセリドが、該二環式化合物1重量部に対して1−1,000,000重量部存在する、請求項3に記載の組成物。
- 中鎖脂肪酸トリグリセリドが、該二環式化合物1重量部に対して5−500,000重量部存在する、請求項4に記載の組成物。
- 中鎖脂肪酸トリグリセリドが、該二環式化合物1重量部に対して10−200,000重量部存在する、請求項5に記載の組成物。
- 中鎖脂肪酸トリグリセリドが、カプリル酸トリグリセリドおよび/またはカプリン酸トリグリセリドである、請求項6に記載の組成物。
- ヒト便秘患者における便秘の軽減または予防のための、請求項1−7のいずれかに記載の組成物。
- 腸洗浄のための、請求項1−7のいずれかに記載の組成物。
