Description
- Es ist seit langem bekannt, dass Hausstaub allergische Reaktionen wie Asthma und Rhinitis beim Menschen auslösen kann. Es wurde bereits 1928 berichtet, dass die Staubmilben im Hausstaub die Hauptursache der allergischen Reaktion wären, aber erst seit den 60er Jahren schätzten Forscher deren Bedeutung richtig ein.
- Es wird angenommen, dass der Kot von zwei besonderen Hausstaubmilben-Spezies, Dermatophagoides farinae (bekannt als Der-f) und Dermatophagoides pteronyssinus (bekannt als Der-p), die Immunreaktionen des Körpers auslösen, wodurch die bekannten allergischen Symptome verursacht werden.
- Eine Übersicht darüber ist bereitgestellt in Experimental and Applied Acarology, 10 (1991) S. 167–186 in einem Artikel mit dem Titel „House dust-mite allergen": Eine Übersicht von L. G. Arlian.
- Beide Arten, Der-f und Der-p, sind weltweit verbreitet. In einigen Gebieten ist Der-f die einzige Dermatophragoiden-Spezies. In anderen Gebieten ist Der-p die einzige Spezies. In noch anderen Gebieten kommen beide Spezies vor, wobei im Allgemeinen die eine oder die andere überwiegt.
- Eine Möglichkeit der Bewältigung dieser allergischen Reaktion war das häufige und gründliche Saugen der Oberflächen wie Teppichen, die Hausstaubmilben und ihren Kot enthalten, jedoch ist dies sowohl zeitaufwändig (d.h. es muss regelmäßig durchgeführt werden, wenn man eine allergenfreie Umgebung schaffen will) als auch stark abhängig von der Wirksamkeit des verwendeten Staubsaugers und Filterbeutels, z.B. Mikrofilters oder Zweischichtenstaubsaugerbeutels.
- WO 93/15774 beschreibt die Dekontamination und Entgiftung als Mittel zum Keimfrei stellen des Heims unter Verwendung eines in Aerosolform im Raum zerstäubten Produkts. Das Produkt kann eine Formulierung von zehn Bestandteilen sein, die Thymol in einer Mindestenge von 4% einschließen können.
- Chemical Abstracts, Bd. 69, Nr. 19, 4. November 1968, Abstract-Nr. 75913, Honma, Shoichi, "Effect of Various Chemicals on Mites", beschreibt die Verwendung von Chemikalien zum Abtöten von Milben. Thymol war zur Behandlung von Milbenbefall von Interesse.
- Wir entdeckten eine Anzahl von allergendeaktivierenden Mitteln, die gegen beide Spezies, sowohl Der-f als auch Der-p, wirksam, sind. Überraschenderweise entdeckten wir, dass einige dieser deaktivierenden Mittel für den behandelten Staubmilbenallergentyp spezifisch sind. Zum Beispiel wirken gegen Der-f-Allergene wirksame Mittel nicht automatisch auch gegen Der-p-Allergene.
- Erfindungsgemäß wird ein Verfahren zum Deaktivieren eines Der-f- und/oder Der-p-Allergens bereitgestellt, das das Inkontaktbringen des Allergens mit einer wirksam deaktivierenden Menge eines deaktivierenden Mittels umfasst, wobei das Verfahren eine Zusammensetzung verwendet, die das deaktivierende Mittel in einer Menge von 0,01% bis 3% enthält, wobei das deaktivierende Mittel 6-Isopropyl-m-cresol ist.
- 6-Isopropyl-m-cresol ist allgemein als Thymol bekannt.
- In Verfahren zur Behandlung von Decken und Teppichen zum Deaktivieren der Allergene darin beträgt die vorliegende Menge des deaktivierenden Mittels etwa 16 g bis 170 g pro 10 m2, vorzugsweise etwa 32 g pro 10 m2.
- Ferner wird erfindungsgemäß eine Aerosolzusammensetzung bereitgestellt, die 0,01 bis 3% 6-Isopropyl-m-cresol als deaktivierendes Mittel, ein Treibgas und wahlweise ein Lösungsmittel enthält.
- Vorzugsweise beträgt die Treibgasmenge in einer solchen Zusammensetzung 4% bis 50%, stärker bevorzugt 4% bis 30%.
- Vorzugsweise beträgt die Lösungsmittelmenge in einer solchen Zusammensetzung 0% bis 90% und besonders bevorzugt 20% bis 90%.
- Vorzugsweise ist das Treibgas ausgewählt aus handelsüblichen Treibgasen, z.B. C1-4-Alkanen, Chlorfluorkohlenwasserstoffen und komprimierten Gasen wie Stickstoff, Luft und Kohlendioxid.
- Vorzugsweise ist das Lösungsmittel ausgewählt aus C1-6-Alkoholen (z.B. Ethanol) oder Wasser.
- Zusätzlich können Zusammensetzungen dieser Erfindung auch eine oder mehrere der Folgenden enthalten:
einen Duftstoff, vorzugsweise in einer Menge von 0% bis 5%, stärker bevorzugt von 0% bis 2%;
eine antimikrobielle Verbindung, z.B. Alkyldimethylbenzylammoniumsaccharinat, vorzugsweise in einer Menge von 0,01% bis 1%;
ein oberflächenaktives Mittel, z.B. Dow Corning 193, vorzugsweise in einer Menge von 0,1% bis 1%;
einen Korrosionshemmstoff z.B. Natriumnitrit, Natriumbenzoat, Triethanolamin und Ammoniumhydroxid, vorzugsweise in einer Menge von 0,1% bis 10%;
und
ein milbentötendes Mittel wie Benzylbenzoat, Pyrethroid Permethrin, d-Allethrin und wahlweise ein Synergetikum wie Piperonylbutoxid, vorzugsweise in einer Menge von 0,1% bis 10%. - Die Erfindung wird nun durch die folgenden Beispiele veranschaulicht.
- Beispiele
- Das Testverfahren in den Beispielen 1 und 2 lautet folgendermaßen und ist als ELISA-Protokoll bekannt.
- Das ELISA-Protokoll für Der-f und Der-p wurde folgendermaßen als Messung der denaturierenden Eigenschaft für denaturierende Mittel entwickelt.
- ELISA Protokoll 1
-
- 1. Der Staub wird aus HooverTM-Staubsaugerbeuteln gesammelt und durch eine Reihe von Sieben bis herunter auf 63 Mikron gesiebt.
- 2. Saubere Petrischalen werden mit der zu testenden Chemikalie gekennzeichnet (auf der Grundfläche). Drei Wiederholungen werden für jede Behandlung verwendet.
- 3. Jede Petrischale wird mit Filterpapier ausgekleidet, und 0,2 g Staub wird in jede Petrischale auf das Filterpapier gegeben. Der Deckel (oder der Boden, falls die Schalen umgedreht sind), wird wieder auf die Schale gesetzt, und die Schale wird geschüttelt, um den Staub gleichmäßig über das Filterpapier zu verteilen.
- 4. 2%ige, wässrige Lösungen des deaktivierenden Mittels wurden verwendet. Das deaktivierende Mittel wurde mit einem Sorbitoleat als oberflächenaktives Mittel (d.h. Tween) emulgiert.
- 5. Der Staub wird mit der entsprechenden Behandlung besprüht, zwei Sprühstöße sind für eine ausreichend Bedeckung erforderlich (1 Sprühstoß = 1,5 ml).
- 6. Man lässt dies unbedeckt bei Raumtemperatur stehen, bis das Filterpapier trocken ist. Das kann bis zu vier Stunden dauern.
- 7. Der Staub wird in gemäß der Behandlung beschriftete Eppendorf-Reaktionsgefäße geleert.
- 8. Zu jedem Eppendorf-Reaktionsgefäß wird 1 ml 5%iges Rinderserumeiweiß in Phosphatgepufferter Kochsalzlösung mit Tween BSA-PBS-T geben (das 5-fache Gewicht des Staubes) (20 ml BSA-PBS-T = 1 g BSA in 20 ml PBS-T) gegeben.
- 9. Dies wird über Nacht in den Kühlschrank gestellt.
- 10. Es wird für eine Dauer von 5 Minuten bei 13.000 UpM zentrifugiert.
- 11. Der Überstand wird in ein neues gemäß der Behandlung beschriftetes Eppendorf-Reaktionsgefäß dekantiert.
- 12. Es wird erneut für eine Dauer von 5 Minuten bei 13.000 UpM zentrifugiert.
- 13. Es werden Verdünnungen von 1:10 und 1:100 hergestellt indem man 100 μl der unverdünnten Lösung zu 900 μl einer 1%igen BSA-PBS-T-Lösung gibt (1:10). Dies wird unter Verwendung von 100 μl der 1:10-Verdünnung durch Zugabe von 900 μl der 1%igen BSA-PBS-T-Lösung auf eine 1:100-Verdünnung wiederholt.
- ELISA-Protokoll 2 für Der-f und Der-p: Innenraum-Biotechnologien

- 1. Proben und Verdünnungen werden wie im Protokoll hergestellt.
- 2. 500 ml eines 50 mM Carbonat/Bicarbonat-Puffers werden hergestellt, indem man 0,795 g Na2CO3 und 1,465 g NaHCO3 in 500 ml destilliertem Wasser löst. Es wird überprüft, ob der pH-Wert bei 9,6 liegt. (Diese Lösung wird in einem Erlenmeyerkolben im Kühlschrank aufbewahrt).
- 3. Monoklonaler Antikörper (aufbewahrt im Gefrierschrank) muss unter Verwendung des folgenden Verfahrens zur Pufferlösung gegeben werden (1 μg pro Mulde; 11 ml werden benötigt), angewendet auf der ELISA-Platte.
- – 11 ml des Carbonat/Bicarbonat-Puffers werden zur Mischplatte gegeben.
- – –11 μl des monoklonalen Der-f1- oder Der-f2-Antikörpers (aufbewahrt im Gefrierschrank, Eppendorf-Reaktionsgefäß in Verwendung ist im Kühlschrank) wird zur Pufferlösung gegeben. Um sicher zu stellen, dass die gesamten Antikörper von der Spitze abgewaschen sind, wird die Pipettenspitze gewaschen, indem man die Pufferlösung auf- und absaugt und zum gründlichen Mischen leicht rührt.
- 4. 100 μl der Antikörper-Lösung werden in jede Mulde der Mikrotiter-Platte pipettiert, die Platte wird abdeckt und über Nacht bei 4°C stehengelassen.
- 5. Die Platte geleert, indem man sie schnell über dem Ausguss umdreht, dann getrocknet, indem man sie auf einem Stapel Papiertüchern ausklopft.
- 6. In jede Mulde werden 200 μl Waschpuffer gegeben: PBS/0/05% Tween (PBS-T).
- 7. Die Schritte 5 und 6 werden wiederholt (indem insgesamt zwei Waschungen durchgeführt werden).
- 8. Es wird sicher gestellt, dass alle Mulden trocken sind, dann werden 100 μl 1%ige BSA-PBS-T-Lösung zugesetzt. Die Platte wird wieder abgedeckt und für eine Dauer von 1 Stunde bei Raumtemperatur inkubiert*.
- 9. Die Schritte 5 bis 7 werden wiederholt (zwei Waschungen).
- 10. *Während der Inkubationszeit werden die Allergen-Standards in Verdünnungen zwischen 125 und 1 μg/ml Der-f1 oder Der-p1 hergestellt:
- – 25 μl Allergen-Standard (aufbewahrt im Gefrierschrank in einer Styropor-Verpackung) werden zu 475 μl der 1%igen BSA-PBS-T-Lösung geben und vollständig gemischt – gekennzeichnet als „125".
- – 250 μl der 1%igen BSA-PBS-T-Lösung werden zu 7 weiteren Eppendorf-Reaktionsgefäßen gegeben, die gekennzeichnet sind mit 62,5, 31,25, 15,63, 7,61, 3,9, 1,95 und 0,98.
- – 250 μl werden aus dem ersten Eppendorf-Reaktionsgefäß (gekennzeichnet als 125) genommen, und in das nächste (gekennzeichnet mit 62,5) überführt. Dies wird gründlich gemischt.
- – Unter Verwendung einer neuen Pipettenspitze werden 250 μl aus dem Eppendorf-Reaktionsgefäß mit der Kennzeichnung 62,5 in dasjenige mit der Kennzeichnung 31,25 überführt. Dieses Verfahren wird bis zur Kon zentration hinab auf 0,98 fortgesetzt (125, 62,5, 31,25, 15,63, 7,61, 3,9, 1,95, 0,98)
- – Insgesamt werden 475 + (250 × 7) = 2,3 ml: 0,023 g BSA zu 2,3 ml PBS-T gegeben.
- 11. Aliquote von 100 μl der Allergenprobe werden zusammen mit den Standard-Allergen-Proben für die Eichkurve in doppelter Ausführung auf die Platte gegeben. Die Standards kommen gewöhnlich in die ersten beiden Säulen auf der linken Seite, mit der niedrigsten Konzentration oben. Es wird für eine Dauer von einer Stunde inkubiert.
- 12. Es wird den Schritten 5 bis 6 gefolgt, indem 5 Waschungen durchgeführt werden.
- 13. 11 ml 1%ige BSA-PBS-T-Lösung (0,11 g BSA zu 11 ml PBS-T) werden zur Mischplatte geben. Dazu werden 11 μl des biotinylierten monoklonalen Antikörpers (Kühlschrank) geben und gründlich gemischt.
- 14. 100 μl werden in jede Mulde pipettiert und für eine Dauer von 1 Stunde bei Raumtemperatur inkubiert.
- 15. Die Platte wird geleert und wie in Schritt 12 beschrieben (5 Waschungen) gewaschen.
- 16. 11 μl Streptavidin (Gefrierschrank) werden zu 11 ml 1%iger BSA-PBS-T-Lösung geben. In jede Mulde werden 100 μl pipettiert und 30 Minuten inkubiert. Jegliche übrige Lösung wird in einem Fläschchen aufbewahrt.
- 17. Die Platte wird geleert und wie in Schritt 12 beschrieben (5 Waschungen) gewaschen.
- 18. Eine OPD-Lösung wird hergestellt, indem man die beiden Tabletten (in Silber- und Goldfolie) in 20 ml destilliertes Wasser gibt (in einem Glasfläschchen). Es wird ziemlich heftig im Dunklen geschüttelt, bis die Tabletten sich gelöst haben (das Fläschchen wird entweder in Alufolie oder ein Papiertuch gewickelt).
- 19. Eine kleine Menge wird zur Restlösung von Schritt 16 geben. Eine Farbänderung wird abgewartet (positive Reaktion). 200 μl werden zu jeder Mulde geben, und es wird für eine Dauer von mindestens 30 Minuten im Dunklen inkubiert.
- 20. Die Platte wird bei 450 nm/405 nm abgelesen, falls kein Filter verfügbar ist.
- Beispiel 1
- Das in der folgenden Tabelle dargelegte deaktivierende Mittel wurde gemäß dem vorstehenden Verfahren gegen Der-f-Allergene verwendet, und die Ergebnisse lauten wie nachstehend angegeben. Gerbsäure wurde als Vergleichssubstanz verwendet. Was nach der Behandlung mit dem deaktivierenden Mittel und Gerbsäure gemessen wurde, war die Menge aktiv gebliebenen Allergens nach der Behandlung. Das Verhältnis der Menge an aktivem Allergen nach Behandlung mit 6-Isopropyl-m-cresol verglichen mit Gerbsäure ist ebenso angegeben.
- Beispiel 2
- Das nachstehend erwähnte deaktivierende Mittel wurde gemäß dem vorstehenden Verfahren gegen Der-p-Allergene verwendet, und die Ergebnisse lauten wie nachstehend angegeben. Was gemessen wurde, waren die Menge der Allergene nach der Behandlung mit dem deaktivierenden Mittel und die Menge der Allergene nach Saugen ohne Behandlung mit deaktivierendem Mittel.
- Beispiele 3–6
- Die folgenden Formulierungen können als Träger-Zusammensetzungen zur Verwendung in einem Aerosol zum Deaktivieren von Der-f- und Der-p-Allergenen hergestellt werden.
Claims (10)
- Verfahren zum Deaktivieren eines Der-f- und/oder Der-p-Allergens, umfassend das Inkontaktbringen des Allergens mit einer deaktivierenden, wirksamen Menge eines deaktivierenden Mittels, wobei im Verfahren eine Zusammensetzung verwendet wird, die das deaktivierende Mittel in einer Menge von 0,01 bis 3% umfasst und das deaktivierende Mittel 6-Isopropyl-m-cresol ist ist.
- Verfahren nach Anspruch 1, wobei das Allergen ein Der-f-Allergen ist.
- Verfahren nach Anspruch 1, wobei das Allergen ein Der-p-Allergen ist.
- Verfahren nach einem der vorangehenden Ansprüche, wobei das deaktivierende 6-Isopropyl-m-cresol auf Stoffmaterialien, ausgewählt aus Wolldecken, Teppichen und Polstermöbeln, mit einer Auftragungsmenge von 16 g bis 170 g deaktivierendes Mittel pro 10 Quadratzentimeter aufgebracht wird, wodurch Allergene, die mit von Staubmilben ausgeschiedenen Fäkalienteilchen verbunden sind, auf der Oberfläche von Stoffmaterialien bekämpft werden.
- Verfahren nach einem der vorangehenden Ansprüche, wobei das deaktivierende Mittel in einer Aerosolzusammensetzung beinhaltet ist, die auch ein Treibgas und gegebenenfalls ein Lösungsmittel enthält.
- Verfahren nach Anspruch 5, in welchem die Menge des vorliegenden Treibgases 4 bis 50% und die Menge des vorliegenden Lösungsmittels 0 bis 90% beträgt, wobei alle Prozentgehalte auf das Gewicht bezogen sind.
- Verfahren nach Anspruch 5 oder 6, in welchem das Treibgas ausgewählt ist aus einem C1-4-Alkan und Kohlendioxid.
- Verfahren nach einem der Ansprüche 5 bis 7, wobei das Lösungsmittel ausgewählt ist aus C1-6-Alkoholen und Wasser.
- Verfahren nach Anspruch 8, in welchem das Lösungsmittel Ethanol ist.
- Verfahren nach einem der Ansprüche 5 bis 9, in welchem die Zusammensetzung einen oder mehrere der folgenden Bestandteile enthält: einen Duftstoff, ein oberflächenaktives Mittel, ein antimikrobielles Mittel, einen Korrosionshemmstoff, ein milbentötendes Mittel.
