Description
- Die vorliegende Erfindung betrifft Adamantanderivate, ein Verfahren zu ihrer Herstellung, diese Verbindungen enthaltende pharmazeutische Zusammensetzungen, ein Verfahren zur Herstellung der pharmazeutischen Zusammensetzungen und ihre Verwendung bei der Therapie.
- Der P2X7-Rezeptor (früher unter der Bezeichnung P2Z-Rezeptor bekannt), bei dem es sich um einen ligandengesteuerten Ionenkanal handelt, ist auf verschiedenen Zelltypen, größtenteils denjenigen, die bekanntlich am Entzündungs-/Immunprozeß beteiligt sind, im einzelnen Makrophagen, Mastzellen und Lymphozyten (T und B), anzutreffen. Die Aktivierung des P2X7-Rezeptors durch extrazelluläre Nucleotide, insbesondere Adenosintriphosphat, führt zur Freigabe von Interleukin-1β (IL-1β) und Riesenzellenbildung (Makrophagen/Mikrogliazellen), Degranulation (Mastzellen) und L-Selectin-Abspaltung (Lymphozyten). P2X7-Rezeptoren befinden sich auch auf antigenpräsentierenden Zellen (APC), Keratinozyten, speichelproduzierenden Acinuszellen (Parotiszellen), Hepatozyten, Erythrozyten, Erythroleukämiezellen, Monozyten, Fibroblasten, Knochenmarkzellen, Neuronen und renalen Mesangialzellen. Die WO 99/29660 und die WO 99/29661 betreffen Adamantanderivate, die P2X7-Rezeptorantagonistenwirkung zeigen.
- Wünschenswert wäre die Herstellung von Verbindungen, die als P2X7-Rezeptorantagonisten wirksam sind, zur Verwendung bei der Behandlung von Entzündungs-, Immun- oder Herz-Kreislauf-Erkrankungen, bei deren Ätiologie der P2X7-Rezeptor eine Rolle spielen kann.
- Gegenstand der vorliegenden Erfindung ist daher eine Verbindung der allgemeinen Formel worin D für CH2 oder CH2CH2 steht;
E für NHC(O) steht;
R1 und R2 jeweils unabhängig voneinander für Wasserstoff, Halogen (z.B. Fluor, Chlor, Brom oder Iod), Amino (NH2), Nitro NO2, C1-C6-Alkyl oder Trifluormethyl stehen, aber R1 und R2 nicht beide gleichzeitig für Wasserstoff stehen dürfen;
R3 für eine Gruppe der Formel steht;
R4 für eine C1-C6-Alkylgruppe steht;
X für eine NR13-Gruppe steht;
R5 für Wasserstoff steht und
R13 für Wasserstoff steht;
oder ein pharmazeutisch annehmbares Salz oder Solvat davon. - Im Rahmen der vorliegenden Beschreibung kann ein Alkylsubstituent oder ein Alkylteil in einer Substituentengruppe linear oder verzweigt sein, sofern nicht anders vermerkt. Im Rahmen der vorliegenden Erfindung kann eine Alkylgruppe oder ein Alkylteil bis zu 6 Kohlenstoffe enthalten, beispielsweise Methyl, Ethyl, n-Propyl, Isopropyl, n-Butyl, Isobutyl, t-Butyl, n-Pentyl und n-Hexyl. Eine C2-C6-Alkenylgruppe kann linear oder verzweigt sein. In einer Di-C1-C6- alkylaminogruppe können die Alkylteile gleich oder verschieden sein.
- Vorzugsweise stehen R1 und R2 jeweils unabhängig voneinander für ein Wasserstoff- oder Halogenatom oder eine Amino-, Nitro-, C1-C4-Alkyl- oder Trifluormethylgruppe (aber R1 und R2 dürfen nicht beide gleichzeitig für ein Wasserstoffatom stehen).
- Besonders bevorzugt stehen R1 und R2 jeweils unabhängig voneinander für ein Wasserstoff-, Chlor- oder Bromatom oder eine Amino-, Nitro-, C1-C3-Alkyl- oder Trifluormethylgruppe (aber R1 und R2 dürfen nicht beide gleichzeitig für ein Wasserstoffatom stehen).
- Ganz besonders bevorzugt stehen R1 und R2 jeweils unabhängig voneinander für ein Wasserstoff- oder Chloratom (aber R1 und R2 dürfen nicht beide gleichzeitig für ein Wasserstoffatom stehen).
- R4 steht für eine C1-C6-Alkylgruppe, beispielsweise eine lineare C1-C6-Alkylgruppe, wie CH2, (CH2)2, (CH2)3 oder (CH2)4.
- Bevorzugte erfindungsgemäße Verbindungen sind u.a.:
5-(2-Aminoethyl)-2-chlor-N-(tricyclo [3.3.1.13,7]dec-1-ylmethyl)benzamid
und pharmazeutisch annehmbare Salze und Solvate davon. - Gegenstand der vorliegenden Erfindung ist ferner ein Verfahren zur Herstellung einer Verbindung der Formel (I) gemäß obiger Definition, bei dem man:
- (a) wenn X für ein Sauerstoff- oder Schwefelatom oder eine NR13-Gruppe steht, eine Verbindung der allgemeinen Formel worin L1 für eine Abgangsgruppe (z.B. ein Halogenatom oder eine Trifluormethansulfonatgruppe) steht und D, E, R1, R2 und R4 die bei Formel (I) angegebene Bedeutung besitzen, gegebenenfalls in Gegenwart eines geeigneten Silbersalzes (z.B. Silbertrifluormethansulfonat) mit einer Verbindung der allgemeinen Formel worin X' für ein Sauerstoff- oder Schwefelatom oder eine NR13-Gruppe steht und R5 die bei Formel (I) angegebene Bedeutung besitzt, umsetzt; oder
- (b) wenn X für eine NR13-Gruppe steht, eine Verbindung der allgemeinen Formel worin R20 für eine Bindung oder eine C1-C5-Alkylgruppe steht und D, E, R1 und R2 die bei Formel (I) angegebene Bedeutung besitzen, in Gegenwart eines Reduktionsmittels (z.B. Natriumtriacetoxyborhydrid) mit einer Verbindung der allgemeinen Formel worin R5 und R13 die bei Formel (I) angegebene Bedeutung besitzen, umsetzt;
- Die erfindungsgemäßen Verfahren können zweckmäßigerweise in einem Lösungsmittel, z.B. einem organischen Lösungsmittel, wie Dichlormethan, 1,2-Dichlorethan oder Tetrahydrofuran, bei einer Temperatur z.B. im Bereich von 0 bis 200°C, vorzugsweise im Bereich von 0 bis 150°C, durchgeführt werden.
- Zur Herstellung von Verbindungen der Formel (V), worin R20 für eine Bindung steht, kann man eine Verbindung der allgemeinen Formel worin Hal für ein Halogenatom, wie Brom, steht und D, E, R1 und R2 die bei Formel (I) angegebene Bedeutung besitzen, mit einer Base, wie t-Butyllithium, und dann mit einem Formylierungsmittel, wie Dimethylformamid, umsetzen.
- Zur Herstellung von Verbindungen der Formel (VII) setzt man zweckmäßigerweise eine Verbindung der allgemeinen Formel worin R25 für NH2 oder CO2H steht und D die bei Formel (I) angegebene Bedueutung besitzt, mit einer Verbindung der allgemeinen Formel worin R30 für CO2H oder NH2 steht und R1, R2 und Hal die bei obiger Formel (VII) angegebene Bedeutung besitzen, um.
- Zur Herstellung von Verbindungen der Formel (V), worin R20 für eine C1-C5-Alkylgruppe steht, kann man beispielsweise eine entsprechende Verbindung der Formel (V), worin R20 für eine Bindung steht, in Gegenwart einer Base mit (Methoxymethyl)diphenylphosphinoxid oder mit einer Verbindung der allgemeinen Formel worin n für 0, 1, 2 oder 3 steht und R und R' unabhängig voneinander für C1-C6-Alkylgruppen stehen, umsetzen und dann hydrieren.
- Alternativ dazu kann man zur Herstellung von Verbindungen der Formel (V), worin R20 für eine C2-C5-Alkylgruppe steht, eine Verbindung der Formel (VII) in Gegenwart eines Palladiumkatalysators mit einem Alkenol (z.B. 2-Propen-1-ol (Allylalkohol), But-3-enol, Pent-4-enol oder Hex-5-enol) umsetzen und danach gegebenenfalls hydrieren und oxidieren, beispielsweise mit Dess-Martin-Periodinan-Reagens (diese beide letzten Schritte sind nicht erforderlich, wenn es sich bei dem Alkenol um Allylalkohol handelt).
- Als weitere Alternative kann man zur Herstellung von Verbindungen der Formel (V), worin R20 für eine C2-C5-Alkylgruppe steht, eine Verbindung der Formel (VII) in Gegenwart eines Palladiumkatalysators, wie Palladiumacetat, mit einem Alkenoatester (z.B. Methylacrylat oder Ethylacrylat) umsetzen und danach die Estergruppe zu einer Hydroxylgruppe reduzieren und dann mit einem Oxidationsmittel (z.B. Dess-Martin-Periodinan-Reagens) zum Aldehyd oxidieren.
- Verbindungen der Formel (I), worin E für eine NHC(O)-Gruppe steht, können aus einer Verbindung der allgemeinen Formel worin R1, R2 und R3 die bei Formel (I) angegebene Bedeutung besitzen und X für NR13 steht, durch Umsetzung mit Adamantylmethylamin oder Adamantylethylamin in Gegenwart eines Kupplungsmittels, wie 1,1'-Carbonyldiimidazol, hergestellt werden.
- Verbindungen der Formel (XI) können aus einer Verbindung der allgemeinen Formel worin L2 für eine Abgangsgruppe (wie ein Halogenatom oder eine Trifluormethansulfonatgruppe) steht und R1, R2 und R4 die bei Formel (I) angegebene Bedeutung besitzen, mit einer Verbindung der Formel (IV) gemäß obiger Definition, gegebenenfalls in Gegenwart eines Silbersalzes, wie Silbertrifluormethansulfonat, hergestellt werden.
- Verbindungen der Formeln (III), (IV), (VI), (VIII), (IX), (X), (XIII) und (XIV) sowie Verbindungen HO-R7Z, HS-R7Z und H2N-R7Z sind im Handel erhältlich, in der Literatur gut bekannt oder nach bekannten Methoden leicht zugänglich.
- Wie für den Fachmann leicht ersichtlich ist, müssen bei den erfindungsgemäßen Verfahren bestimmte funktionelle Gruppen, wie Hydroxyl-, Carboxyl-, Aldehyd-, Carbonyl- oder Aminogruppen, in den Ausgangsreagentien oder Zwischenverbindungen möglicherweise durch Schutzgruppen geschützt werden. Somit kann die Herstellung der Verbindungen der Formel (I) in einer bestimmten Stufe die Abspaltung einer oder mehrerer Schutzgruppen umfassen.
- Die Schützung und Entschützung funktioneller Gruppen wird in „Protective Groups in Organic Chemistry", Herausgeber J.W.F. McOmie, Plenum Press (1973), und „Protective Groups in Organic Synthesis", 2. Auflage, T.W. Greene und P.G.M. Wuts, Wiley-Interscience (1991), beschrieben.
- Die Verbindungen der obigen Formel (I) können in ein pharmazeutisch annehmbares Salz oder Solvat davon, vorzugsweise ein Säureadditionssalz, wie ein Hydrochlorid, Hydrobromid, Phosphat, Acetat, Fumarat, Maleat, Tartrat, Citrat, Oxalat, Methansulfonat oder p-Toluolsulfonat, oder ein Alkalimetallsalz, wie ein Natrium- oder Kaliumsalz, umgewandelt werden.
- Bestimmte Verbindungen der Formel (I) können in stereoisomeren Formen existieren. Die Erfindung umfaßt selbstverständlich alle geometrischen und optischen Isomere der Verbindungen der Formel (I) und Gemische davon einschließlich Racemate. Tautomere und Gemische davon bilden ebenfalls einen Aspekt der vorliegenden Erfindung.
- Die erfindungsgemäßen Verbindungen sind insofern vorteilhaft, als sie pharmakologisch wirksam sind und zur Verwendung als Modulatoren der P2X7-Rezeptoraktivität verwendet werden können. Sie sind daher als Pharmazeutika zur Verwendung bei der Behandlung oder Prävention von rheumatoider Arthritis, Osteoarthritis, Psoriasis, allergischer Dermatitis, Asthma, Hyperreaktivität der Atemwege, chronischer obstruktiver Lungenerkrankung (COPD), Bronchitis, septischem Schock, Glomerulonephritis, Reizkolon, Crohn-Krankheit, Colitis ulcerosa, Atherosklerose, Wachstum- und Metastasenmalignazellen, Myoblastenleukämie, Diabetes, neurodegenerativen Krankheiten, Alzheimer-Krankheit, Meningitis, Osteoporose, Brandverletzungen, ischämischer Herzkrankheit, Schlaganfall, peripherer Gefäßkrankheit und Krampfadern indiziert.
- Gegenstand der vorliegenden Erfindung ist demgemäß eine Verbindung der Formel (I) oder ein pharmazeutisch annehmbares Salz oder Solvat davon gemäß obiger Definition zur Verwendung bei der Therapie.
- Einen weiteren Gegenstand der Erfindung bildet die Verwendung einer Verbindung der Formel (I) oder eines pharmazeutisch annehmbaren Salzes oder Solvats davon gemäß obiger Definition bei der Herstellung eines Arzneimittels zur Verwendung bei der Therapie.
- Im Rahmen der vorliegenden Beschreibung schließt der Begriff „Therapie" auch „Prophylaxe" ein, sofern nicht anders vermerkt. Der Begriff „therapeutisch" ist entsprechend aufzufassen.
- Es wird erwartet, daß die Prophylaxe für die Behandlung von Personen, bei denen die entsprechende Krankheit bzw. das entsprechende Leiden bereits einmal aufgetreten ist oder bei denen sonst ein erhöhtes Risiko der entsprechenden Krankheit bzw. des entsprechenden Leidens angenommen wird, besonders relevant ist. Zu den Patienten, bei denen das Risiko des Auftretens einer bestimmten Krankheit bzw. eines bestimmten Leidens besteht, gehören im allgemeinen diejenigen, bei denen die Krankheit bzw. das Leiden bereits in der Familie aufgetreten ist, oder diejenigen, die gemäß genetischen Tests oder genetischem Screening für die Krankheit bzw. das Leiden besonders anfällig sind.
- Die erfindungsgemäßen Verbindungen können bei einem Verfahren zur Erzielung von Immunsuppression (z.B. bei der Behandlung von rheumatoider Arthritis, Reizkolon, Atherosklerose, Psoriasis, Lungenkrankheiten, z.B. COPD oder Bronchitis, oder Erkrankungen des Zentralnervensystems, z.B. Alzheimer-Krankheit oder Schlaganfall) verwendet werden, bei dem man einem Patienten eine therapeutisch wirksame Menge einer Verbindung der Formel (I) oder eines pharmazeutisch annehmbaren Salzes oder Solvats davon gemäß obiger Definition verabreicht.
- Für die obigen therapeutischen Anwendungen variiert die verabreichte Dosierung natürlich mit der eingesetzten Verbindung, der Verabreichungsart, der gewünschten Behandlung und der indizierten Krankheit bzw. dem indizierten Leiden. Zur Erzielung von Immunsuppression liegt die Tagesdosis der Verbindung der Formel (I) in der Regel im Bereich von 0,001 mg/kg bis 30 mg/kg.
- Die Verbindungen der Formel (I) und pharmazeutisch annehmbare Salze und Solvate davon können für sich alleine verwendet werden, werden aber im allgemeinen in Form einer pharmazeutischen Zusammensetzung verabreicht, in der die Verbindung der Formel (I) bzw. das Salz bzw. das Solvat (Wirkstoff) zusammen mit einem pharmazeutisch annehmbaren Hilfsstoff, Verdünnungsmittel oder Träger vorliegt. Je nach dem Verabreichungsweg enthält die pharmazeutische Zusammensetzung vorzugsweise 0,05 bis 99 Gew.-% (Gewichtsprozent) und besonders bevorzugt 0,10 bis 70 Gew.-% Wirkstoff und 1 bis 99,95 Gew.-% und besonders bevorzugt 30 bis 99,90 Gew.-% eines pharmazeutisch annehmbaren Hilfsstoffs, Verdünnungsmittels oder Trägers, wobei sich alle Gewichtsprozentangaben auf die Gesamtzusammensetzung beziehen.
- Gegenstand der vorliegenden Erfindung ist somit auch eine pharmazeutische Zusammensetzung, enthaltend eine Verbindung der Formel (I) oder ein pharmazeutisch annehmbares Salz oder Solvat davon gemäß obiger Definition zusammen mit einem pharmazeutisch annehmbaren Hilfsstoff, Verdünnungsmittel oder Träger.
- Gegenstand der Erfindung ist ferner ein Verfahren zur Herstellung einer erfindungsgemäßen pharmazeutischen Zusammensetzung, bei dem man eine Verbindung der Formel (I) oder ein pharmazeutisch annehmbares Salz oder Solvat davon gemäß obiger Definition mit einem pharmazeutisch annehmbaren Hilfsstoff, Verdünnungsmittel oder Träger vermischt.
- Die erfindungsgemäße pharmazeutische Zusammensetzung kann topisch (z.B. in die Lungen und/oder die Atemwege oder auf die Haut) in Form von Lösungen, Suspension, Heptafluoralkan-Aerosolen und Trockenpulverformulierungen, oder systemisch, z.B. durch orale Verabreichung in Form von Tabletten, Kapseln, Sirupen, Pulvern oder Granulaten, oder durch parenterale Verabreichung in Form von Lösungen oder Suspensionen, oder durch subkutane Verabreichung oder durch rektale Verabreichung in Form von Suppositorien oder transdermal verabreicht werden.
- Die vorliegende Erfindung wird nun unter Bezugnahme auf die folgenden Ausführungsbeispiele näher erläutert.
- Beispiel 1
- a) 5-(Brommethyl)-2-chlor-N-(2-tricyclo[3.3.1.13,7]dec-1-ylmethyl)benzamid
- Eine Lösung von 2-Chlor-5-(brommethyl)benzoesäure (1,0 g) in Dichlormethan (25 ml) wurde bei 0°C mit Dimethyl formamid (0,05 ml) gefolgt von Oxalylchlorid (0,52 ml) versetzt. Der Ansatz wurde auf Raumtemperatur kommen gelassen und 30 min gerührt. Der nach Abziehen der flüchtigen Bestandteile unter Vakuum verbleibende Rückstand wurde unter Hochvakuum getrocknet. Der Rückstand wurde in Dichlormethan (20 ml) gelöst und bei 0°C zu einer Lösung von 2-Adamantanmethylamin-Hydrochloridsalz (0,95 g) in Dichlormethan (20 ml) und Diisopropylamin (2 ml) gegeben. Der Ansatz wurde auf Raumtemperatur kommen gelasen und 2 h gerührt. Die organischen Substanzen wurden mit Wasser (20 ml) und dann mit gesättigter wäßriger Ammoniumchloridlösung gewaschen, wonach die organische Schicht über Magnesiumsulfat getrocknet und filtriert wurde. Das Filtrat wurde unter vermindertem Druck aufkonzentriert, was einen Feststoff ergab. Die Rohsubstanz wurde aus Dichlormethan/Hexan umkristallisiert, was die im Untertitel aufgeführte Verbindung in Form eines weißen Feststoffs (1,3 g) ergab.
- b) 2-Chlor-5-(cyanomethyl)-N-(tricyclo[3.3.1.13,7]dec-1-ylmethyl)benzamid
- Eine Lösung von 5-(Brommethyl)-2-chlor-N-(2-tricyclo[3.3.1.13,7]dec-1-ylmethyl)benzamid (Beispiel 1a, 1,8 g) in Ethanol (50 ml) wurde mit Kaliumcyanid (525 mg) und 18-Krone-6 (150 mg) versetzt, wonach die Mischung 24 h unter Rückfluß erhitzt wurde. Nach dem Abkühlen wurde die Reaktionsmischung bis zur Trockne eingedampft, wonach der Rückstand zwischen Essigsäureethylester und gesättigter Natriumhydrogencarbonatlösung verteilt wurde. Die organischen Substanzen wurden abgetrennt, mit Kochsalzlösung gewaschen, über Magnesiumsulfat getrocknet, filtriert und eingedampft. Durch Reinigung mittels Chromatographie an Siliciumdioxid (unter Verwendung eines Gradienten von Isohexan/Essigsäureethylester von 4:1 bis 6:4 als Elutionsmittel) wurde die im Untertitel aufgeführte Verbindung in Form eines rosafarbenen Feststoffs (1,0 g) erhalten. - Eine gekühlte Lösung von 2-Chlor-5-(cyanomethyl)-N-(tricyclo[3.3.1.13,7]dec-1-ylmethyl)benzamid (Beispiel 1b, 2,1 g), (1,1-Dimethylethoxy)carbonylkohlensäure-1,1-dimethylethylester (2,67 g) und Cobalt(II)-chlorid (1,6 g) in Methanol (100 ml) wurde portionsweise mit Natriumborhydrid (1,6 g) versetzt. Nach 1,5 h wurde das Lösungsmittel abgedampft und der Rückstand zwischen Essigsäureethylester und gesättigter Natriumhydrogencarbonatlösung verteilt, wonach die unlöslichen Cobaltsalze abfiltriert wurden. Die organische Phase wurde abgetrennt, mit Kochsalzlösung gewaschen, über Natriumsulfat getrocknet, filtriert und eingedampft, was die im Untertitel aufgeführte Verbindung in Form eines Schaums (2,2 g) ergab.

- Eine Lösung von [2-[4-Chlor-3-[[(tricyclo[3.3.1.13,7]dec-1-ylmethyl)amino]carbonyl]phenyl]ethyl]carbamidsäure-1,1-dimethylethylester (2,2 g, Beispiel 1c) in Methanol/Dichlormethan (1:1) (50 ml) wurde mit Salzsäure (4 M in 1,4-Dioxan, 3,0 ml) versetzt. Nach 24 h wurde das Lösungsmittel abgedampft, wobei die Titelverbindung in Form eines Schaums (1,85 g) zurückblieb.

- Bestimmte Verbindungen, wie Benzoylbenzoyladenosintriphosphat (bbATP) sind bekanntlich Agonisten des P2X7-Rezeptors und bewirken die Bildung von Poren in der Plasmamembran (Drug Development Research (1996), 37 (3), S. 126). Folglich beobachtet man bei der Aktivierung des Rezeptors mit bbATP in Gegenwart von Ethidiumbromid (einer fluoreszierenden DNA-Sonde) einen Anstieg der Fluoreszenz von intrazellulär DNA-gebundenem Ethidiumbromid. Der Fluoreszenzanstieg kann als Maß für die Aktivierung des P2X7-Rezeptors und daher zur Quantifizierung der Wirkung einer Verbindung auf den P2X7-Rezeptor verwendet werden.
- Auf diese Art und Weise wurde jede der Titelverbindungen der Beispiele auf Antagonistenwirkung am P2X7-Rezeptor geprüft. So wurde die Prüfung in Flachboden-Mikrotiterplatten mit 96 Vertiefungen durchgeführt, wobei die Vertiefungen mit 250 μl Testlösung, die 200 μl einer Suspension von THP-1-Zellen (2,5 × 106 Zellen/ml) mit 10–4 M Ethidiumbromid, 25 μl einer kaliumreichen Pufferlösung mit 10–5 M bbATP und 25 μl der kaliumreichen Pufferlösung mit 3 × 10–5 M Testverbindung enthielt, gefüllt wurden. Die Platte wurde mit einer Kunststoff-Folie abgedeckt und 1 Stunde bei 37°C inkubiert. Dann wurde die Platte in einem Fluoreszenzplattenlesegerät von Perkin-Elmer, Anregung 520 nm, Emission 595 nm, Spaltbreiten: Anr. 15 nm, Em. 20 nm, ausgelesen. Zu Vergleichszwecken wurden bbATP (ein P2X7-Rezeptor-Agonist) und Pyridoxal-5-phosphat (ein P2X7-Rezeptor-Antagonist) bei der Prüfung separat als Kontrollen verwendet. Aus den erhaltenen Ablesungen wurde für jede Testverbindung ein pIC50-Wert berechnet, bei welchem es sich um den negativen Logarithmus der zur Verringerung der bbATP-Agonistenwirkung um 50% benötigten Konzentration der Testverbindung handelt. Jede der Verbindungen der Beispiele zeigte Antagonistenwirkung mit einem pIC50-Wert > 5,0.
Claims (15)
- Verbindung der allgemeinen Formel worin D für CH2 oder CH2CH2 steht; E für NHC(O) steht; R1 und R2 jeweils unabhängig voneinander für Wasserstoff, Halogen, Amino, Nitro, C1-C6-Alkyl oder Trifluormethyl stehen, aber R1 und R2 nicht beide gleichzeitig für Wasserstoff stehen dürfen; R3 für eine Gruppe der Formel steht; R4 für eine C1-C6-Alkylgruppe steht; X für eine NR13-Gruppe steht; R5 für Wasserstoff steht und R13 für Wasserstoff steht; oder ein pharmazeutisch annehmbares Salz oder Solvat davon.
- Verbindung nach Anspruch 1, in der D für CH2 steht.
- Verbindung nach Anspruch 1 oder 2, in der R1 und R2 jeweils unabhängig voneinander für ein Wasserstoff-, Chlor- oder Bromatom oder eine Amino-, Nitro-, C1-C3-Alkyl- oder Trifluormethylgruppe stehen.
- Verbindung der Formel (I) oder ein pharmazeutisch annehmbares Salz oder Solvat davon nach Anspruch 1, bei dem es sich um 5-(2-Aminoethyl)-2-chlor-N-(tricyclo[3.3.1.13,7]dec-1-ylmethyl)benzamid handelt.
- Verfahren zur Herstellung einer Verbindung der Formel (I) nach Anspruch 1, bei dem man (a) eine Verbindung der allgemeinen Formel worin L1 für eine Abgangsgruppe steht und D, E, R1, R2 und R4 die bei Formel (I) angegebene Bedeutung besitzen, gegebenenfalls in Gegenwart eines geeigneten Silbersalzes mit einer Verbindung der allgemeinen Formel worin R5 und R13 die bei Formel (I) angegebene Bedeutung besitzt, umsetzt; oder (b) eine Verbindung der allgemeinen Formel worin R20 für eine Bindung oder eine C1-C5-Alkylgruppe steht und D, E, R1 und R2 die bei Formel (I) angegebene Bedeutung besitzen, in Gegenwart eines Reduktionsmittels mit einer Verbindung der allgemeinen Formel (VI) gemäß obiger Definition unter (a) umsetzt; und gegebenenfalls nach (a) oder (b) die erhaltene Verbindung der Formel (I) in ein pharmazeutisch annehmbares Salz oder Solvat davon umwandelt.
- Pharmazeutische Zusammensetzung, enthaltend eine Verbindung der Formel (I) oder ein pharmazeutisch annehmbares Salz oder Solvat davon nach einem der Ansprüche 1 bis 4 zusammen mit einem pharmazeutisch annehmbaren Hilfsstoff, Verdünnungsmittel oder Träger.
- Verfahren zur Herstellung einer pharmazeutischen Zusammensetzung nach Anspruch 5, bei dem man eine Verbindung der Formel (I) oder ein pharmazeutisch annehmbares Salz oder Solvat davon nach einem der Ansprüche 1 bis 6 mit einem pharmazeutisch annehmbaren Hilfsstoff, Verdünnungsmittel oder Träger vermischt.
- Verbindung der Formel (I) oder ein pharmazeutisch annehmbares Salz oder Solvat davon nach einem der Ansprüche 1 bis 4 zur Verwendung bei der Therapie.
- Verbindung der Formel (I) oder ein pharmazeutisch annehmbares Salz oder Solvat davon nach einem der Ansprüche 1 bis 4 zur Verwendung bei der Behandlung von rheumatoider Arthritis.
- Verbindung der Formel (I) oder ein pharmazeutisch annehmbares Salz oder Solvat davon nach einem der Ansprüche 1 bis 4 zur Verwendung bei der Behandlung von chronischer obstruktiver Lungenerkrankung.
- Verbindung der Formel (I) oder ein pharmazeutisch annehmbares Salz oder Solvat davon nach einem der Ansprüche 1 bis 4 zur Verwendung bei der Behandlung von Osteoarthritis.
- Verwendung einer Verbindung der Formel (I) oder eines pharmazeutisch annehmbaren Salzes oder Solvats davon nach einem der Ansprüche 1 bis 4 bei der Herstellung eines Arzneimittels zur Verwendung bei der Therapie.
- Verwendung einer Verbindung der Formel (I) oder eines pharmazeutisch annehmbaren Salzes oder Solvats davon nach einem der Ansprüche 1 bis 4 bei der Herstellung eines Arzneimittels zur Verwendung bei der Behandlung von rheumatoider Arthritis.
- Verwendung einer Verbindung der Formel (I) oder eines pharmazeutisch annehmbaren Salzes oder Solvats davon nach einem der Ansprüche 1 bis 4 bei der Herstellung eines Arzneimittels zur Verwendung bei der Behandlung von chronischer obstruktiver Lungenerkrankung.
- Verwendung einer Verbindung der Formel (I) oder eines pharmazeutisch annehmbaren Salzes oder Solvats davon nach einem der Ansprüche 1 bis 4 bei der Herstellung eines Arzneimittels zur Verwendung bei der Behandlung von Osteoarthritis.
