Description
Die vorliegende Erfindung betrifft die Cyclosporinderivate
der allgemeinen Formel I,
worin A =
(nachstehend als
Cyclosporin D
bezeichnet),
oder
(nachstehend als
Dihydrocyclosporin D
bezeichnet),
bedeutet,
Verfahren zu ihrer Herstellung dadurch gekennzeichnet, daß man
bedeutet,
Verfahren zu ihrer Herstellung dadurch gekennzeichnet, daß man
- a) Cyclosporin D erhält, indem man einen Cyclosporin D produzierenden Stamm der Pilzspecies Tolypocladium inflatum Gams in Gegenwart eines Nährmediums züchtet und Cyclosporin D isoliert, oder daß man
- b) Dihydrocyclosporin D erhält, indem man Cyclosporin D hydriert,
und pharmazeutische Zubereitungen, die mindestens eine Verbindung
gemäß Anspruch 1 enthalten.
In der Literatur (z. B. Helvetica Chimica Acta, 59, 1076 (1976)
werden Cyclosporinderivate mit antiarthritischer Wirksamkeit und
starker Hemmung der zellulären Immunität beschrieben, z. B. der
Hauptmetabolit Cyclosporin A. Es wurde nun gefunden, daß das
Wirkungsprofil der neuen Verbindungen Cyclosporin D bzw. Dihydrocyclosporin
D sich von Cyclosporin A sowohl qualitativ wie auch
quantitativ unterscheidet. So zeigen Cyclosporin D und Dihydrocyclosporin
D in den meisten Prüfungen auf Immunosuppression im
Gegensatz zu Cyclosporin A nur eine schwache Wirkung. Hingegen
sind Cyclosporin D und Dihydrocyclosporin D gut aktiv im Modell
der sich entwickelnden oder bereits etablierten Freund-Adjuvans-
Arthritis.
Die Feststellung dieser im Hinblick auf den Stand der Technik
unerwarteten Eigenschaften: Geringe immunosuppressive Aktivität
bei deutlicher Wirkung in der Freund-Adjuvans-Arthritis bedeutet
offensichtlich einen äußerst wichtigen Fortschritt auf dem Weg
zur Entwicklung eines Antiarthritikums, das Lymphozyten nicht
beschädigen würde.
Die Züchtung nach Verfahren a) läßt sich nach an sich bekannten
Methoden für die Züchtung von analogen Stämmen, z. B. wie in
Beispiel 1 beschrieben, durchführen.
Ein bevorzugter Cyclosporin D produzierender Stamm ist der frei
zugängliche Stamm NRRL 8044 der Pilzspecies Tolypocladium
inflatum Gams. Eine Kultur davon wurde beim United States Department
of Agriculture (Northern Research and Development Division),
Peoria, Ill., USA deponiert. Dieser Stamm wurde vormals der
Pilzspecies Trichoderma polysporum (Link ex. Pers.) zugeordnet
und ist z. B. in der DE-OS 24 55 859 beschrieben.
Für die Herstellung von Cyclosporin D lassen sich auch Stämme der
Pilzspecies Tolypocladium inflatum Gams verwenden, wie sie z. B.
durch Selektion oder Mutation des Pilzstammes NRRL 8044 unter der
Einwirkung von Ultraviolett- oder Röntgenstrahlen oder durch
Anwendung anderer Maßnahmen, z. B. durch Behandlung von
Laboratoriumskulturen mit geeigneten Chemikalien, gewonnen werden
können.
Cyclosporin D kann auf an sich bekannte Weise isoliert werden,
z. B. wie in Beispiel 1 beschrieben. Hierbei kann Cyclosporin D
abgetrennt werden von gleichzeitig in größeren Mengen vorhandenen
Naturprodukten, z. B. das etwas polarere Cyclosporin A
(auch bekannt als S 7481/F-1), das polarere Cyclosporin B (auch
bekannt als S 7481/F-2) und das noch polarere Cyclosporin C.
Das erfindungsgemäße Verfahren b) läßt sich nach an sich bekannten
Methoden ausführen, z. B. durch katalytische Hydrierung.
Als Lösungsmittel kommen vorzugsweise niedere aliphatische
Alkohole, wie z. B. Methanol, Äthanol, Isopropanol, oder Äthylacetat
in Frage. Die Hydrierung erfolgt zweckmäßigerweise im
neutralen Bereich bei Temperaturen zwischen 20 und 30°C und bei
Atmosphärendruck oder wenig erhöhtem Druck. Als Katalysator kommt
vorzugsweise Palladium, z. B. Palladium auf Kohle, in Frage.
Cyclosporin D und Dihydrocyclosporin D zeichnen sich durch
interessante chemotherapeutische und pharmakologische Eigenschaften
aus und können daher als Heilmittel verwendet werden.
Sie zeichnen sich durch eine schwache immunosuppressive Wirkung
bei deutlicher Wirkung in der Freund-Arthritis aus.
Die immunosuppressive Wirkung sowie die Wirkung in der Freund-
Adjuvans-Arthritis von Cyclosporin D, Dihydrocyclosporin D sowie
von Cyclosporin A kann an den nachstehenden Testmodellen (1-5)
wie folgt gezeigt werden:
Bei nachstehenden Testverbindungen hemmen die Concanavalin
stimulierte DNA-Synthese (Hemmung des H³-Thymidin-Einbaues),
die Zellproliferation und die Blastogenese an Mäusemilzlymphozyten
bei folgenden Konzentrationen:

Mäusemilzzellen werden während 3 bis 4 Tagen in Gegenwart von
Antigen (Schaferythrocyten, SE) und der Testsubstanz gezüchtet.
Die Zellen werden geerntet, gewaschen und plattiert
mit frischem Antigen (SE) in halbfestem Agar. Nach Inkubation
während 60 Minuten wird Komplement hinzugefügt und die Inkubatinon
während 90 Minuten fortgesetzt. Die Empfindlichkeit
der Mauslymphozyten gegenüber dem Antigen während der Primärkultur
führt zu einer Antikörperfreisetzung. Die Gegenwart
von Komplement und dem ausgeschiedenen gegenüber SE spezifischen
Antikörper führt bei den Schaferythrocyten zur
Plaque-Bildung. Die Suppression der Plaque-bildenden Zellen
wird bei Verwendung der Testsubstanzen in den folgenden
Konzentrationen beobachtet:

Mäuse werden mit DNP-KLH immunisiert und bekommen nach
3 Wochen einen Booster mit demselben Antigen. Ein bis
4 Wochen nach dem "Challenge" werden die Milzen entnommen und
eine Zellkultur bereitet. Durch Zugabe von Antigen (DNP-KLH)
werden spezifische Antikörper gegen dieses Antigen sezerniert
und können im Überstand durch ELISA-Technik gemessen werden.
Die Testsubstanzen werden bei der in vitro-Inkubation in verschiedenen
Konzentrationen der Zellkultur zugegeben. Die
Antikörperentwicklung in Abwesenheit der Testsubstanzen wird
als Kontrolle verwendet und wird als 100% Reaktion angenommen.
Die Reaktion in Gegenwart der Testsubstanzen wird in
der prozentualen Änderung der Reaktion, verglichen mit 100%
der Kontrollreaktion ausgedrückt.
Die Suppression der Reaktion (Immunantwort) wird beobachtet
bei der Verwendung einer Testsubstanz bei folgender Konzentration:

Lymphozyten (Mäusemilzzellen) reagieren bei Co-Inkubation mit
allogenischen Zellen durch Proliferation und Differenzierung.
Die Reaktion wird in Gegenwart und in Abwesenheit der Testsubstanzen
gemessen. Die Reaktion in Abwesenheit der Testsubstanzen
wird als Kontrolle verwendet und wird als 100%
Reaktion angenommen. Die Reaktion in Gegenwart der Testsubstanzen
wird in der prozentualen Änderung der Reaktion,
verglichen mit100% der Kontrollreaktion ausgedrückt.
Die Suppression der Reaktion wird beobachtet bei der Verwendung
der Testsubstanzen in folgenden Konzentrationen:

Die entzündungshemmende Wirkung kann mit dem Freund-Adjuvans-
Arthritis-Test an der Ratte gezeigt werden. Bei diesem Test
wird die Adjuvans-Arthritis nach der Methode von Pearson und
Wood, "Arthr. Rheum.", 2, 440 (1959) induziert.
Die Testsubstanzen zeigten sich bei diesem Test als wirksam
bei der sich entwickelnden Arthritis und bei der etablierten
Arthritis in folgenden Dosen:
Cyclosporin D und Dihydrocyclosporin D sind insbesondere indiziert
als Antiarthritica.
Als Heilmittel können Cyclosporin D und Dihydrocyclosporin D
allein oder in geeigneter Arzneiform mit pharmakologisch
indifferenten Hilfsstoffen verabreicht werden.
In den nachfolgenden Beispielen erfolgen alle Temperaturangaben
in Celsiusgraden.
500 Liter einer Nährlösung, die pro Liter 40 g Glucose, 2,0 g
Natriumcaseinat, 2,5 g Ammoniumphosphat, 5 g MgSO₄ · 7 H₂O, 2 g
KH₂PO₄, 3 g NaNO₃, 0,5 g KCl, 0,01 g FeSO₄ und entmineralisiertes
Wasser enthält, werden mit 50 Liter einer Vorkultur des Stammes
NRRL 8044 angeimpft und in einem Stahlfermenter unter Rühren
(170 UPM) und Belüftung (1 Liter Luft/Min./Liter Nährlösung)
13 Tage bei 27° inkubiert (siehe DE-OS 24 55 859).
Die Kulturbrühe wird mit der gleichen Menge n-Butylacetat ausgerührt,
nach Abtrennung der organischen Phase wird diese im Vakuum
konzentriert und der Rohextrakt durch 3stufige Verteilung
zwischen Methanol-Wasser (9 : 1) und Petroläther entfettet. Die
methanolische Phase wird abgetrennt, im Vakuum konzentriert und
das Rohprodukt durch Zugabe von Wasser ausgefällt. Das nach der
Filtration gewonnene Material wird an Kieselgel mit Hexan-Aceton
(2 : 1) als Elutionsmittel chromatographiert, wobei die zuerst
eluierten Fraktionen vorwiegend Cyclosporin A und Cyclosporin D
enthalten, die später eluierten Anteile vorwiegend Cyclosporin C.
Zur weiteren Reinigung werden die Cyclosporin A- und D-haltigen
Fraktionen aus der 2- bis 2,5fachen Menge Aceton bei -15°
kristallisiert und das Kristallisat wird anschließend durch
zweimalige Chromatographie an Kieselgel weiter aufgetrennt, wobei
die mit Essigester wassergesättigt zuerst eluierten Fraktionen
Cyclosporin D in stark angereicherter Form enthalten. Diese
werden in der doppelten Menge Aceton gelöst und bei -15°
kristallisieren lassen. Das dabei erhaltene Rohkristallisat von
Cyclosporin D wird zur weiteren Reinigung in der 10fachen Menge
Aceton gelöst, mit 2 Gewichtsprozent Aktivkohle versetzt und
während 5 Minuten auf 60° erwärmt. Das nach Filtration über Talk
erhaltene klare und beinahe farblose Filtrat wird auf ein Drittel
des Volumens eingeengt und auf Raumtemperatur erkalten lassen,
wobei Cyclosporin D spontan auskristallisiert. Durch Stehenlassen
bei -17° wird die Kristallisation vervollständigt. Die durch
Abfiltrieren gewonnenen Kristalle werden mit wenig eiskaltem
Aceton gewaschen und anschließend im Hochvakuum bei 80°C
während 2 Stunden getrocknet.
Charakterisierung von Cyclosporin D:
Farblose, prismatische Kristalle; Smp. 148-151°
[α] = -245° (c = 0,52 in Chloroform)
[α] = -211° (c = 0,51 in Methanol)
[α] = -245° (c = 0,52 in Chloroform)
[α] = -211° (c = 0,51 in Methanol)
400 mg Palladium-Kohle (10% Palladium) werden in 15 ml Äthanol
während 20 Minuten vorhydriert. Zu dieser Suspension des
Palladiumkatalysators wird die Lösung von 3,66 g Cyclosporin D in
30 ml Äthanol zugegeben und darauf bei 24° und einem Druck von
736 mm Quecksilbersäule bis zur beendeten Wasserstoffaufnahme
hydriert. Anschließend filtriert man vom Katalysator ab und
dampft das Filtrat im Vakuum bei 20 bis 40° zur Trockne ein.
Dabei fällt das dünnschichtchromatographisch einheitliche
Dihydrocyclosporin D als farbloses amorphes Pulver an, das im
Hochvakuum während 4 Stunden bei 70° getrocknet wird.

Claims (3)
1. Cyclosporinderivate der allgemeinen Formel I,
worin A =
(nachstehend als
Cyclosporin D
bezeichnet),
oder
(nachstehend als
Dihydrocyclosporin D
bezeichnet),
bedeutet.

2. Verfahren zur Herstellung der Verbindungen gemäß Anspruch 1,
dadurch gekennzeichnet, daß man
- a) Cyclosporin D erhält, indem man in an sich bekannter Weise einen Cyclosporin D produzierenden Stamm der Pilzspecies Tolypocladium infatum Gams in Gegenwart eines Nährmediums züchtet und Cyclosporin D isoliert, oder indem man
- b) Dihydrocyclosporin D erhält, indem man in an sich bekannter Weise Cyclosporin D hydriert.
3. Pharmazeutische Zubereitungen, die mindestens eine Verbindung
gemäß Anspruch 1 enthalten.
