一般而言,与任何其它植物相比,蘑菇含有较低的脂肪,但是含有较高的蛋白和糖类含量。糖类也包括海藻糖、甘露醇、树胶醛糖等及多聚糖,多聚糖难于吸收进入人的肠胃,其主要成分是不消化的膳食纤维。因此,与按食物分析计算的食物原料相比,蘑菇是具有较低热量的食物原料。此外,每个个体中通常包括100~800mg量的麦角固醇和钙,当干燥时它们可转化成维生素D2。除了所述的成分之外,蘑菇还含有维生素B1、维生素B2及烟酸,不含维生素A和C。此外,关于矿物质,其中K比Na占更高的份数,P、Ca和Fe次之。蘑菇的调味成分主要是核酸及谷氨酸、琥珀酸、苹果酸、尿醇等的组合物。因此,蘑菇不仅是具有较低热量并能发挥生理功能的食物原料,而且具有极佳的风味、味道和口味。
然而,蘑菇含有大量的水和氮化合物,因此其会腐烂,并且由于其软组织的原因微生物易于繁殖。由此,收获蘑菇后其保存期较短。蘑菇可分为活产品和干燥产品的形式。由于蘑菇被用作配菜或调味品,所以其可在提供基本营养方面起作用。
近来,蘑菇已被公认为是抑制癌症和变异性、降低血清中脂质的形成、增强对疾病的免疫力、抑制衰老及预防成人病的有效原料。到目前为止,已将蘑菇的用途扩展到了医药领域。已预期的药用蘑菇,如灵芝(Ganderma lucidum)、香菇(Lentinus edodes)、平茹(Pleurotus osteratus)、Elfvingia applanata、蘑菇(agaricus)、黑木耳(Auricularia auricula)及石耳(Umbilicaria esculenta)。特别地,据报道包括在灵芝提取物内的多聚糖蛋白配合物表现出对癌细胞增殖的抑制、对原发性高血压的治疗、对过氧化脂质形成的抑制等。此外,众所周知香菇具有抗癌性质,使胆固醇、肌张力、多尿下降的性质,及可治疗高血压、肾炎、哮喘、胃溃疡等,据报道其提取物有降低血清和肝中脂质及抑制肝损害的作用。此外,据报道来源于平菇的多聚糖提取物具有降低血清中胆固醇的作用及对由四氯化碳引起的肝损害具有抑制作用。此外,据报道从平菇的果肉和菌丝体得到的提取物具有抗氧化作用。
乳酸菌是使用诸如葡萄糖、乳糖的碳水化合物可产生乳酸的细菌,并从公元3,000前就用于发酵牛奶和干酪。由于由乳酸菌发酵的牛奶可抑制消化器官内有害菌的生长并预防人衰老,所以乳酸菌发酵的牛奶已在世界范围内作为货物出售至现在。已研究了乳酸菌对人健康的许多有益作用。
乳酸菌的此类作用包括肠道调节(即预防痢疾和便秘)、对肠癌增殖的抑制、及通过抑制有害菌的生长抑制衰老、促进维生素形成的生长、通过控制胆固醇预防成人病及增强免疫力等。
乳酸菌典型的例子包括链球菌、小球菌、乳酸链球菌、乳酸杆菌、vipidus等。在自然界中例如人和动物的消化道和几乎所有的蔬菜内发现有乳酸菌。保加利亚细菌、酸奶酪细菌及嗜温细菌已被用于生产酸奶酪。酸奶酪细菌、乳酪菌(casei bacteria)及嗜酸菌被用于生产含有乳酸菌的饮料。乳酪菌和牛奶链球菌已被用于生产干酪。牛奶链球菌已被用于生产发酵的黄油。每种特定的乳酸菌已被用于不同种食物的制造过程。
乳酸菌可抑制肠上皮细胞及其新陈代谢。乳酸菌通过乳酸菌的发酵可分泌乳酸、(低级)脂肪酸、细菌素、H2O2等,以致于可抑制有害菌的生长并通过HMG(羟基甲基戊二酸)、乳清酸、尿酸等降低胆固醇的形成。特别地,嗜酸乳酸杆菌可直接分解胆固醇。乳酸菌可活化在免疫系统中的小噬细胞侦测杆菌,从而可侦测细菌和病毒的出现,抑制由于淋巴细胞分裂引起的癌细胞增殖,并提高血液中的抗体IgA的生产,促进γ-干扰素的生产。这样一系列的功能提高了免疫力,增加了食物的营养价值,抑制了内生传染,抑制了肠内致癌物质的形成,并导致有害菌的死亡。
近来饮食的改变可导致较高可能性的脑血管系统疾病和循环系统疾病,如心脏病、高血压、高脂血症、动脉硬化及恶性瘤。此类慢性退行性疾病与活体内的脂质新陈代谢紊乱相关。近来,对可改善人健康的生理活性原料进行了有力的调查和研究,其结果是发现并报道了对脂质和氧化作用有效的天然成分。其中,可食用和药用的蘑菇作为抗氧化原料已引起了注意。
由于活体内的氧化压力,自由基的形成可使生物膜脂质过氧化,并且增加的过氧化脂质可损害组织和器官,导致新陈代谢紊乱。因此由于在活体内自由基形成的原因,能够抑制氧化损害的生理活性材料被期望有助于降低循环系统疾病和慢性疾病,例如癌。
此外,近来饮食的改进和生活方式的变化导致由于摄取高热量食物并缺少适度锻炼引起的肥胖。工业的发展和社会复杂关系是引起压力的原因,而医药的发展延长了生命。这些因素是各种疾病的原因,特别地,糖尿病是引起慢性血管疾病的原因,并且提高了死亡率。
糖尿病是一种特征在于胰腺内胰岛素分泌不足或在每个组织中胰岛素受体紊乱及高血糖水平的疾病。糖尿病分成胰岛素相关糖尿病(1型糖尿病)和非胰岛素相关糖尿病(2型糖尿病)。当由于免疫系统的紊乱分泌胰岛素的胰腺β-细胞被破坏时,出现胰岛素相关糖尿病,当由于遗传或肥胖的原因,胰岛素受体如肌肉细胞紊乱时出现非胰岛素相关糖尿病。
近来对探求各种对于降低血糖水平的生理活性原料如茶对叶、薏苡和桑树叶进行了持续的研究。但是这些研究主要集中在对1型糖尿病的治疗。
根据这些统计资料,在韩国这两种类型中1型糖尿病患者呈逐年增长的趋势,95%患有糖尿病的患者是2型糖尿病。可降低血糖水平的药剂口服给药至2型糖尿病患者并配合饮食治疗。最后,在饭后将阿卡波糖(acarbose)和伏格列波糖(voglibose)给药至患者以降低血糖水平,但它们价格较高。
为克服这些缺点,已对可降低血糖水平的商业上可得到的及可食用的食物或各种天然来源的生理活性原料进行了研究。韩国专利第165,939号公开了一种用于降低血糖水平并含有枸杞提取物的组合物及其制备方法。韩国专利第195,886号公开了一种用于治疗糖尿病并含有Crdycepsspp.、牛黄、枸杞、野葛根等的药物组合物。然而,在所述专利中公开的用于治疗糖尿病的制剂其味道、风味、口味不适合,并且其制备过程非常复杂。
因此,本领域需要可有效治疗2型糖尿病的食物组合物及具有适合的味道、风味、口味的制剂。
如上所述,蘑菇和乳酸菌被用作保健食品的原料,但是其协同作用仍不清楚。
发明揭露
由此,本发明人已对蘑菇和乳酸菌发酵溶液的药理作用进行了广泛的调查研究。结果我们发现蘑菇乳酸发酵溶液表现出有效的协同作用,然后实现了本发明。
因此,本发明的目的是提供一种制备具有极佳味道、风味及口味的蘑菇乳酸发酵溶液并可缩短发酵期的方法,及由此制备的蘑菇乳酸发酵溶液。
本发明的另一目的是提供一种制备能够抑制过氧化脂质形成的蘑菇乳酸发酵溶液的方法,及由此制备的蘑菇乳酸发酵溶液。
本发明的再一目的是提供一种制备能够有效降低血糖水平的蘑菇乳酸发酵溶液的方法,及由此制备的蘑菇乳酸发酵溶液。

实施本发明的最佳方式
根据本发明制备蘑菇乳酸发酵溶液的方法包括如下步骤:
a)制备含有蘑菇成分的培养基;
b)将乳酸菌株接种在该培养基上;
c)培养该接种了菌株的培养基;及
d)老化该经培养的培养基。
此外,所述的步骤(a)包括如下步骤i)通过研磨或提取,从蘑菇的果肉或菌丝体得到蘑菇成分;ii)将重量为0.1-10%的得到的蘑菇成分、重量为1-50%,优选为1-20%的脱脂乳、重量为0.1-20%的白糖和平衡重量的纯化水加到该含有蘑菇成分的培养基中,并使此混合物均匀化以制备乳酸菌培养基;iii)在75-110℃的温度范围热处理该含有蘑菇成分的培养基15-40分钟;及iv)将该热处理的培养基冷却至35-40℃的温度范围。
可用在所述的i)中的蘑菇例子包括所有的食用和药用蘑菇,例如姬松茸(Agarics blazei)、灵芝、灰树花(Grifola frondosa)、Elfvingia applanata、平菇、洋菇(Agaricus bisporus)、金针菇(Flammulina velutipes)、香茹及Crdyceps spp.,但不限于这些。可用的蘑菇部分是其果肉和菌丝体。如果是姬松茸、灵芝、灰树花、Elfvingia applanata可以使用从它们的果肉或菌丝体得到的蘑菇成分。如果是平菇、洋菇、金针菇、香茹及Crdyceps spp.,蘑菇成分是从它们的果肉得到的。此外,蘑菇成分优选是蘑菇粉末和提取物的混合物。特别地,用于降低血糖水平的蘑菇成分优选是灵芝的提取物。
在步骤b)中,将基于含有所述蘑菇成分的培养基总重量计,重量为1-10%的冷藏乳酸菌或热处理的乳酸菌接种于在步骤a)的iv)中被冷却的培养基上。考虑到发酵期,接种的乳酸菌株优选是热处理的乳酸菌。热处理是通过将冷藏的菌株放进培养箱,并将该菌株培养至25-40℃的温度范围而实现的。
在步骤c)中,当保持培养箱的温度在35-40℃的范围内的同时,培养3-20小时。如果接种的是热处理的乳酸菌株,那么培养时间优选在3-6小时的范围内。
在步骤d)中,在3-5℃的温度下老化预定的时间。
本发明也提供由所述的制备蘑菇乳酸发酵溶液的方法制备的蘑菇乳酸发酵溶液。
研究了由此得到的蘑菇乳酸发酵溶液的性质、对过氧化脂质形成的抑制作用及降低血糖水平的作用。
姬松茸、灵芝、平菇、洋菇、金针菇、灰树花、香茹、Elfvingia applanata及Crdyceps spp.可用于本发明的蘑菇乳酸发酵溶液,但是本发明的范围不限于这些。所有的食用和药用蘑菇都能用于制备本发明的蘑菇乳酸发酵溶液。
首先,参照图1,示意性地说明了制备本发明的蘑菇乳酸发酵溶液的方法。
a)制备含有蘑菇成分培养基的步骤
i)得到蘑菇成分
选取姬松茸、灵芝、灰树花、Elfvingia applanata的蘑菇果肉或菌丝体及平菇、洋菇、金针菇、香茹及Crdyceps spp.的果肉中的一种或多种,洗涤,在60℃的热空气干燥器中干燥,并研磨,得到蘑菇干燥粉末。
选取姬松茸、灵芝、灰树花、Elfvingia applanata的蘑菇果肉或菌丝体及平菇、洋菇、金针菇、香茹及Crdyceps spp.的果肉中的一种或多种,洗涤,以常规的方式使用适当的溶剂在高压杀菌器中提取,得到蘑菇提取物。
ii)制备乳酸菌培养基
通过将重量为0.1-10%的得到的蘑菇成分、重量为1-50%,优选为1-20%的脱脂乳、重量为0.1-20%的白糖和平衡重量的纯化水加到在所述的i)中得到的含有蘑菇成分的培养基中,并使此混合物均匀化以制备乳酸菌培养基。在该乳酸菌培养基中还可包括其它有效的成分,如低聚糖、糊精、维生素和矿物质。
iii)~iv)热处理及冷却
在75-110℃的温度范围热处理该含有蘑菇成分的培养基15-40分钟后,将该热处理的培养基冷却至35-40℃的温度范围。
b)将乳酸菌株接种在该培养基上的步骤
将在培养箱中培养至25-40℃温度范围的冷藏乳酸菌或热处理的乳酸菌接种于在步骤a)的iv)中被冷却的培养基上。此时,基于含有所述蘑菇成分的培养基总重量计,接种的乳酸菌株的量可被选择在重量为1-10%的范围内。
c)培养该接种了菌株的培养基的步骤
当保持培养箱的温度在35-40℃内的同时,将接种了菌株的乳酸菌培养基培养3-20小时。如果接种的是热处理的乳酸菌株,那么培养时间优选在3-6小时的范围内。
d)老化该培养的培养基的步骤
在3-5℃的温度范围内将所述步骤c)中培养的菌株老化10-20小时。
下文将说明本发明的优选实施方案,但是优选实施方案仅是说明性的,本发明不限于此。
实施例
原料
在实验室培养姬松茸、灵芝、灰树花、Elfvingia applanata、平菇、洋菇、金针菇、香茹及Crdyceps spp.的菌丝体,从市场得到姬松茸、灵芝、灰树花、Elfvingia appalnata的果肉。
实施例1-4:姬松茸乳酸发酵溶液的制备
实施例1
研磨姬松茸的果肉和菌丝体,得到干燥粉末。将重量为5%的得到的粉末、重量为10%的脱脂乳、重量为2%的白糖和平衡重量的纯化水的混合物加到培养基中并均匀化。在100℃的温度热处理该含有蘑菇成分的培养基20分钟,然后冷却至37℃。
将基于含有所述蘑菇成分的培养基总重量计,重量为3%的冷藏保加利亚乳酸杆菌(Lactobacillus bulgaricus)接种于该培养基上。制备6个接种了菌株的含有蘑菇成分的培养基试样。当保持培养箱的温度在37℃的同时,将试样分别培养1、2、3、4、5、6小时,并测定它们的pH和酸度。
随后,在4℃的温度下将该培养的试样老化12小时。在均化器中均匀化该老化的试样以制备姬松茸的乳酸发酵溶液。
实施例2
除了用姬松茸提取物替代干燥粉末外,按与实施例1相同的过程制备姬松茸乳酸发酵溶液。
实施例3
研磨姬松茸的果肉和菌丝体,得到干燥粉末。将重量为5%的得到的粉末、重量为10%的脱脂乳、重量为2%的白糖和平衡重量的纯化水的混合物加到培养基中并均匀化。在100℃的温度热处理该含有蘑菇成分的培养基20分钟,然后冷却至37℃。
将冷藏的保加利亚乳酸杆菌在37℃的培养箱中培养1小时。
将基于含有所述蘑菇成分的培养基总重量计,重量为3%的经培养的保加利亚乳酸杆菌接种于该培养基上。制备6个接种了菌株的含有蘑菇成分的培养基试样。当保持培养箱的温度在37℃的同时,将试样分别培养1、2、3、4、5、6小时,并测定它们的pH和酸度。
随后,在4℃的温度下将培养的试样老化12小时。在均化器中均匀化老化的试样以制备姬松茸的乳酸发酵溶液。

除了用姬松茸提取物替代干燥粉末外,按与实施例3相同的过程制备姬松茸乳酸发酵溶液。

除了用从灵芝的果肉和菌丝体得到的重量为0.1%的干燥粉末和重量为5.0%的提取物的混合物替代从姬松茸的果肉和菌丝体得到的干燥粉末外,按与实施例1相同的过程制备灵芝乳酸发酵溶液。
实施例39:混合蘑菇乳酸发酵溶液的制备
将从香菇、平菇和灵芝(重量比=4∶4∶2)得到的重量为1%的干燥粉末和重量为1%的提取物的混合物、重量为13%的脱脂乳、重量为10%的低聚糖、重量为1%的糊精和重量为74%的纯化水均匀化以制得含有蘑菇成分的培养基。通过在80℃的温度下加热该含有蘑菇成分的培养基30分钟来杀菌,然后冷却至37℃。
将基于含有所述蘑菇成分的培养基总重量计,重量为3%的保加利亚乳酸杆菌接种于由此得到的乳酸菌培养基上。将该培养基在37℃的培养箱中培养12小时,然后在4℃的温度下老化12小时。在均化器中均匀化该老化的试样以制备混合蘑菇乳酸发酵溶液。
实施例40-42,对比实施例1和2:灵芝乳酸发酵溶液的制备
将1000ml的蒸馏水(总重量的10%)加到100g的灵芝中,并使用高压杀菌器在低于100℃的温度下提取1小时。过滤得到的提取物,加入100ml的蒸馏水,再次在高于121℃的温度下提取1小时以得到灵芝提取物。
将基于每一个培养基总重量计,分别将重量为0.01%(对比实施例1)、重量为0.1%(实施例40)、重量为0.5%(实施例41)、重量为5%(实施例42)及重量为10%(对比实施例2)的得到的灵芝提取物的加到含有重量为8%的脱脂乳、重量为10%的低聚糖、重量为1%的糊精和平衡重量的纯化水的每一个培养基中,并均匀化。
将基于含有所述蘑菇成分的培养基总重量计,重量为3%的保加利亚乳酸杆菌接种于每一个培养基上。当保持培养箱的温度在37℃的同时,将每一培养基培养12小时,并在4℃的温度下老化12小时以制备灵芝乳酸发酵溶液。
实验实施例
实验实施例:pH、酸度及流变学
为测量从实施例1-36得到的蘑菇乳酸发酵溶液的pH、酸度及流变学(硬度、断裂能及弹性分子)随培养时间的变化,将从每一个实施例得到的乳酸发酵溶液在1000rpm的均化器中均匀化30秒。
用pH计测定pH,通过将每种蘑菇乳酸发酵溶液的一部分(10ml)用三级蒸馏水以1∶1的比例稀释,再加入0.1%的酚酞溶液,并用0.1N NaOH滴定来测定酸度。

如表1和表2所示,从实施例1-36得到的蘑菇乳酸发酵溶液具有较低的pH和较高的酸度。此外,如图2-5所示,可确保乳酸有效的形成速率,并且硬度、断裂能及弹性分子的流变学表现出软组织和极佳的性质(例如粘性)。
特别地,如表2所示,当热处理的乳酸菌接种在含有蘑菇成分的培养基上时,发酵时间缩短约33%。

实验实施例2:对过氧化脂质形成的抑制作用
为研究本发明的蘑菇乳酸发酵溶液对过氧化脂质的影响,将乳酸菌发酵的牛奶、香菇,平菇及灵芝的混合粉末、在实施例39中得到的蘑菇乳酸发酵溶液分别加到含有胆固醇的饮食中。
从加入起的4周内,研究对雌性大鼠体内过氧化脂质的影响。
①原料
从Jinju Mushroom Agricultural Association得到香菇、灵芝及平菇的干燥产品。将干燥产品切成块,研磨并粉碎以得到可通过20筛目筛子的细颗粒形式的粉末。
②实验动物、条件及饮食组成
在22±2℃的温度、50±5%的相对湿度及每12小时光暗交替的条件下培育作为实验动物体重约为180g的雌性大白鼠(Sprague Dawley)。以固体饮食的形式喂养1周,然后用正常饮食喂养4天。将大鼠分成5组,每组6只大鼠。实验饮食组由正常饮食组(ND)、胆固醇饮食组(CD)(通过将0.5%(w/w)的胆固醇和0.125%(w/w)的胆酸钠加到正常的饮食组得到)、添加有乳酸菌发酵牛奶组(CDFM)(通过将乳酸菌发酵牛奶加到胆固醇饮食中得到)、添加有蘑菇粉末组(CDMP)(通过将蘑菇粉末加到胆固醇饮食中得到)及添加有蘑菇乳酸发酵溶液组(CDFMMP)(通过将蘑菇乳酸发酵溶液加到胆固醇饮食中得到)。此时,CDMP组的蘑菇粉末是香菇、平菇和灵芝的混合物,并将4%量的混合物加到该组中。在CDFM组中,乳酸菌发酵牛奶是通过在30℃的温度下用保加利亚乳酸杆菌培养12小时并将其冻干而得到的,将其以13.5%的量加到饮食中。在CDFMMP组中,乳酸菌发酵牛奶是通过将香菇、平菇和灵芝4∶4∶2的混合物以4%的量加到保加利亚乳酸杆菌培养的溶液中、并在30℃的温度下培养12小时且冻干而得到的,将其以1 7.5%的量加到饮食中。表3表明这些实验饮食组的实验饮食组成。在4周内自由供给实验饮食及饮水。每天在固定时间监测饮食供给量,每周测定它们的体重。

③试样
在实验的最后一天,实验动物禁食12小时,用乙醚轻度麻醉,然后从下腔静脉取血样。将血样在室温3,000rpm下离心15,得到血清。用冷的0.9%的生理盐水洗涤切开的组织,干燥,称重,得到用于分析过氧化脂质浓度的试样。
④血清、肝脂质及血糖水平的分析
使用胆固醇C-测试wako药盒(Wako Junyaku,Osaka,Japan)测定血清中的总胆固醇,使用HDL-胆固醇E-测试wako药盒(Wako Junyaku,Osaka,Japan)测定血清中的HDL-胆固醇,使用甘油三酸酯E-测试wako药盒(Wako Junyaku,Osaka,Japan)测定血清中的中性脂质,使用磷脂C-测试wako药盒(Wako Junyaku,Osaka,Japan)测定血清中的磷质,根据葡萄糖氧化酶方法使用商业上可得到的药盒(Wako Junyaku,Osaka,Japan)测定血清中葡萄糖浓度,根据Folch’s法提取肝组织中的脂质并用与测定血清中脂质同样的方法测定其浓度。
⑤每一组织中的匀浆、微粒体及线粒体部分
将1.15%KCl-10mM的磷酸盐缓冲液(pH7.4)加到每一个切开的组织中,然后用均化器均匀化。取出溶液的一部分作为匀浆部分,从剩余的溶液中分别分离出微粒体部和线粒体部分。
⑥组织部分中过氧化脂质浓度的测量
通过如下的方法测量每一个组织中过氧化脂质浓度。首先,将2ml硫代巴比妥酸(TBA)试剂分别加到1ml的匀浆、微粒体及线料体部分溶液中,然后彻底混合。在水浴中加热此混合物15分钟,冷却,在3,000rpm下离心15分钟。取出上层清液,测量535nm处的吸光率。组织中过氧化脂质的浓度表达为nmol/g丙二醛。
⑦统计学
使用血液化学分析仪(Vitalab,Spectra II,Merck)测量血清中的ALT和AST值。基于t-试验检测每组测试结果的统计显著性,并且当其P值小于5%时认为结果具有统计显著性。
通过单因子变异数分析法(one-way ANOVA)将实验结果表达为平均值和标准偏差,基于邓肯多元区间测试(Duncan’s multiple range test)检测每组实验结果的统计显著性,并且当其P值小于5%时认为结果具有统计显著性。

致动脉硬化指数(AI)指的是总胆固醇-HDL胆固醇/HDL胆固醇,及具有不同符号的值具有p<0.05的统计显著性。
从表4可以得知,与正常饮食组(ND)相比,在胆固醇饮食组(CD)中血清总胆固醇的浓度增加4.6倍,表明较高的胆固醇高血糖。然而,与胆固醇饮食组(CD)相比,在添加有乳酸发酵牛奶组中(CDFM)中总胆固醇浓度下降44.6%,在添加有蘑菇提取物组(CDMP)中下降57.4%,在添加有蘑菇乳酸发酵溶液组(CDFMMP)中下降72.0%。
在CDMP和CDFMMP组中血清HDL-胆固醇浓度增加,但是在CDFM中没有变化。此外,在CDFMMP组中相对于CDMP组有显著的增长。从此结果可看出,乳酸菌含有可增加HDL-胆固醇浓度的生量活性成分。
根据Framinghan心脏病研究,当动脉硬化指数不超过3.5时,冠状动脉病的存在是安全的。此外,此研究建议该指数应被保持在不超过4.5。在表3中,比较实验组间的动脉硬化指数,在胆固醇饮食组(CD)中的动脉硬化指数与正常饮食组(ND)相比明显增加。与添加有乳酸发酵牛奶组(CDFM)相比,胆固醇饮食组(CD)中的动脉硬化指数轻微下降。然而,在添加有蘑菇提取物组(CDMP)和添加有蘑菇乳酸发酵溶液组(CDFMMP)中,它们的动脉硬化指数分别下降68.4%和83.3%。
(2)在生物膜中的过氧化脂质的形成
由于组织细胞中过氧化压力的原因导致自由基形成的增加和活体内抗氧化防御力的下降而引起生物膜中脂质的过氧化作用。表明动物内生物膜过氧化脂质形成度的TBARS水平列在表5中。

表5揭示出在正常饮食组和胆固醇饮食组中每个组织中的过氧化脂质的相对含量为肝、肾、心脏、脾的顺序。然而,在用胆固醇自由喂养4周的雄性大鼠中,每个组织中的TBARS相对含量为大脑、肾、心脏、肝、脾的顺序。此外,在用正常饮食喂养的雄性大鼠(6个月大)中,每个组织中的TBARS相对含量为心脏、肝、大脑、肾的顺序。这些结果表明,组织中过氧化脂质的形成取决于种类、年龄和饮食组成的不同。
实验实施例3:血清葡萄糖水平的下降作用
①方法
为检测蘑菇乳酸发酵溶液对降低血清葡萄糖水平的影响,在患有糖尿病的患者中进行下面的方法。
首先,从正常饮食组中选定10个患有高血糖症(平均:315mg/dL)的男性(年龄25-67,平均年龄45),用正常饮食供应3周。在饭前7点钟测量血清葡萄糖水平。结果列在下面的表6中。

将每天200g的普通酸奶酪产品和正常饮食供应给低血清葡萄糖水平的患者达3周。每天清晨饭前测量血清葡萄糖水平。结果列在下面的表7中。供应一周后血清葡萄糖水平增加到262mg/dL(n=9),供应两周后血清葡萄糖水平增加到315mg/dL(n=7),供应三周后血清葡萄糖水平增加到355mg/dL(n=6)。综上所述,可以确定在酸奶酪饮食组中没有血清葡萄糖水平下降作用。

将每天200g的在实施例42中得到的蘑菇乳酸发酵溶液和正常饮食供应给作为对照组并表现出相同血清葡萄糖水平(321mg/dL)的患有糖尿病的10个男性达3周。每天饭前七点钟测量血清葡萄糖水平。结果列在下面的表8中。供应一周后血清葡萄糖水平降低到207mg/dL(n=10),供应两周后血清葡萄糖水平降低到166mg/dL(n=7),供应三周后血清葡萄糖水平降低到150mg/dL(n=10)。此外,10个中第8位糖尿病患者的平均血清葡萄糖水平下降到121mg/dL(n=8),特别地,供应5天后下降到123mg/dL。
综上所述,结果表明本发明的蘑菇乳酸发酵溶液对于血清葡萄糖水平的下降有明显作用。

实验实施例4:对于降低血清葡萄糖水平有效的蘑菇提取物的含量
为测定蘑菇提取物的适合量,用实施例40-43和对比实施例1-2中得到的蘑菇乳酸发酵溶液对每组中的3个人进行实验。
如下面的表9和图6a-6e所示,在0.01%到0.5%的浓度范围内,血清葡萄糖水平的下降作用不明显,但是在小于0.1%的浓度时,血清葡萄糖水平不再增加。此外,在5%水平时的药理作用最佳,但是在不小于10%水平时会导致严重的低血糖症。由此,可以断定加入的蘑菇提取物的最佳量相应于0.1%到7%的范围为约200g。然而,其可随着性别、年龄、体重和糖尿病的严重程度而变化。

稳定摄取本发明的蘑菇乳酸发酵溶液有助于使大多数患有2型糖尿病的患者保持正常的血清葡萄糖水平。
实验实施例5:蘑菇提取物和乳酸菌的协同作用
为验明是否在实验实施例中表现出的血清葡萄糖水平下降是由蘑菇提取物和乳酸菌的协同作用引起的,连续将蘑菇提取物和本发明的蘑菇乳酸发酵溶液供应给患有糖尿病的患者。在测量血清葡萄糖水平后但是在早饭和晚饭前摄取本发明的蘑菇乳酸发酵溶液和蘑菇提取物。每两天的七点钟测量血清葡萄糖水平。首先,将在实施例42中得到的蘑菇乳酸发酵溶液供应给患者达36天,然后切断供应达6天。随后,当血清葡萄糖水平增加时,将与蘑菇乳酸发酵溶液相同量的蘑菇提取物供应给患者。以预定的间隔时间(2天)监测血清葡萄糖水平的变化。
如下面的表10和附图7所示,在10天内血清葡萄糖水平急剧下降,然后稳定在100(mg/dL)约20天(参见图7中的“A”)。然而,由于切断了供应,血清葡萄糖水平增加到276(mg/dL)(参见图7中的“B”)。随后,当将与蘑菇乳酸发酵溶液相同量的蘑菇提取物供应给患者时,血清葡萄糖水平缓慢降低。此后,血清葡萄糖水平没有明显的变化(参见图7中的“C”)。

从上述结果可以看出,蘑菇提取物和乳酸菌对于血清葡萄糖水平的下降有协同作用。
按照本发明方法制得的蘑菇乳酸发酵溶液具有极佳的味道、风味及口味,并且对于抑制过氧化脂质的形成和使血清葡萄糖水平下降是有效的。由此,本发明适用于药物和食品工业。
Claims (18)
1.一种制备蘑菇乳酸发酵溶液的方法,包括如下步骤:
(a)制备含有蘑菇成分的培养基;
(b)将乳酸菌株接种在所述的培养基上;
(c)培养所述接种了菌株的培养基;及
(d)老化所述经培养的培养基。
2.如权利要求1所述的方法,其中所述的步骤(a)包括如下步骤:i)通过研磨或提取,从蘑菇的果肉或菌丝体得到蘑菇成分;ii)制备乳酸菌培养基;iii)在75-110℃的温度范围热处理所述含有蘑菇成分的培养基15-40分钟;及iv)将所述的热处理培养基冷却至35-40℃的温度范围。
3.如权利要求1或2所述的方法,其中所述的蘑菇选自姬松茸、灵芝、灰树花、Elfvingia applanata、平菇、洋菇、金针菇、香茹及Crdycepsspp.。
4.如权利要求2所述的方法,其中在步骤i)中所述的蘑菇提取物是灵芝提取物。
5.如权利要求2所述的方法,其中在步骤i)中所述的蘑菇成分是从香菇、平菇及灵芝的混合物得到的。
6.如权利要求2所述的方法,其中在步骤ii)中所述的乳酸菌培养基由重量为0.1-10%的蘑菇成分、重量为1-50%的脱脂乳、重量为0.1-20%的白糖和平衡重量的纯化水组成。
7.如权利要求1所述的方法,其中基于含有所述蘑菇成分的培养基总重量计,在步骤b)中将重量为1-10%的冷藏乳酸菌或热处理的乳酸菌接种。
8.如权利要求7所述的方法,其中所述的欲接种菌株是保加利亚乳酸杆菌。
9.如权利要求7所述的方法,其中所述的菌株是热处理的乳酸菌。
10.如权利要求9所述的方法,其中所述的热处理是通过将冷藏的菌株放进培养箱,并将所述的菌株培养至25-40℃的温度范围而实现的。
11.如权利要求1所述的方法,其中所述的培养是在35-40℃的温度范围内进行3-20小时。
12.如权利要求11所述的方法,其中所述的培养进行3-6小时。
13.如权利要求1所述的方法,其中所述的老化是在3-5℃的温度范围内进行10-20小时。
14.由权利要求1至13任一项所述方法制得的蘑菇乳酸发酵溶液。
15.如权利要求14所述的蘑菇乳酸发酵溶液,包括可使血糖水平下降的有效成分。
16.如权利要求15所述的蘑菇乳酸发酵溶液,其中所述的有效成分是从灵芝提取物得到的。
17.如权利要求14所述的蘑菇乳酸发酵溶液,包括可使血清中过氧化脂质形成下降的有效成分。
18.如权利要求17所述的蘑菇乳酸发酵溶液,其中所述的有效成分是从香菇、平菇及灵芝的混合物得到的。
