Verfahren zur Herstellung eines neuen basisch substituierten Fettsäure-b' & / (2-halogen-6-methyl-anilids).
Ctegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung eines neuen basisch substituierten Fettsäure- (2-halogen-6- xnethyl-anilids), welches dadurch gekennzeich- net ist, dass man auf eine Verbindung der Formel

in welchen Formeln X und Y reaktionsfähige, bei der Reaktion sich abspaltende Reste bedeuten, einwirken lässt.
Als Verbindungen der Formel I kommen iür einen Umsatz beispielsweise in Frage : 2-Chlor-6-methyl-anilin, eventuell in Form seiner Salze, wie des Hydrochlorids oder des Sulfates, weiter die N-Halogen-magnesyl-Ver- bindungen (X=Mg. Hal) usf.
Als Verbindungen der Formel II kommen beispielsweise in Frage : Di-n-butyl-aminoessig- s : ure selbst und ihre reaktionsfähigen funktionellen Derivate wie ihre Halogenide, ihre Ester, ihre Anhydride, beispielsweise die reinen und auch die gemischten Anhydride mit Phosphorsäure, Schwefelsäure und Kohlensäure usf.
Man kann beispielsweise 2-Chlor-6-methyl a. nilin oder auch ein Salz desselben in Gegenwart eines für diese Zweeke geeigneten wasserabspaltenden Mittels wie Phosphorpent oxyd, Phosphoroxychlorid usf. mit Di-n-butylaminoessigsäure behandeln.
Weiter kann man auch 2-Chlor-6-methylanilin bzw. ein Salz desselben mit einem Di-n-butyl-aminoessigsäurehalogenid (bzw. einem Salz eines solchen) oder einem Anhydrid zur Reaktion bringen.
Es ist weiterhin auch möglich, 2-Chlor-6methyl-anilin mit einem Di-n-butyl-amino- essigsäureester, vorzugsweise einem Arylester, bei erhöhter Temperatur zu acylieren oder beispielsweise ein Halogen-magnesyl-2-chlor6-methyl-anilin (erhalten durch Umsetzen der Base mit einem Alkylmagnesiumhalid) mit einem Di-n-butyl-aminoessigsäurealkylester umzusetzen.
Das auf diese Weise erhaltene Di-n-butylamino-essigsäure-2-chlor-6-methyl-anilid) bildet ein farbloses, unter 0, 06 mm bei 145 siedendes Ö] ; dessen Hydrochlorid schmilzt bei 145-1. 47 . Das neue Anilid soll als Lokal anästhetikum und als Zwischenprodukt zur Herstellung weiterer Derivate Verwendung finden.
Beispiel 1 :
25 Gewichtsteile Di-n-butyl-aminoessig- säurechlorid-hydrochlorid in Benzol werden mit einer Mischung von 14 Gewichtsteilen 2-Chlor-6-methyl-anilin und 20 Gewichtsteilen Triäthylamin erwärmt. Anschliessend wird das gebildete Triathylaminhydroclilorid durch Ausschütteln mit Wasser entfernt und die Benzollösung nach dem Trocknen verdampft.
Der Rüekstand stellt das Di-n-butyl-aminoessigsäure-(2-chlor-6-methyl-anilid) dar, das durch Vakuumdestillation oder Überführen in das Hydrochlorid gereinigt werden kann.
Die Reaktion kann auch ohne Verdün nungs-und Kondensationsmittel durchgeführt werden, indem man beispielsweise das Di-n-butyl-aminoessigsäurechlorid-hydroehlorid mit 2-Chlor-6-methyl-anilin troeken er hitzt, anschliessend die Reaktionsmasse mit Wasser verdünnt und in üblicher Weise auf- arbeitet.
Beispiel 2 :
Ein Mol Brom-magnesyl-2-chlor-6-methyl- anilin (gebildet aus 2-Chlor-6-methyl-anilin und Atlylmagnesiumbromid) in Äther, wird mit 1, 1 bis 1, 2 Molen Di-n-butyl-amino-essigsäure-äthylester umgesetzt. Man gelangt so ebenfalls zu dem gewünschten Anilid.
