Als Verbindungen der Formel I kommen für einen Umsatz beispielsweise in Frage: 2-Clilor-6-metliyl-anilin, eventuell in Form seiner Salze, wie des Hydrochlorids oder des Sulfates, weiter die NT-Halogen-ma.gnesiTl-Ver- bindungen (X<B>=</B> Mg. Hal) usf.
Als Verbindungen der Formel II kommen beispielsweise in Frage: Pyrrolidinoessigsäure selbst und ihre reaktionsfähigen funktionellen Derivate wie ihre Halogenide, ihre Ester, ihre Anhy dride, beispielsweise die reinen und auch die gemischten Anhy dride mit Phosphorsäure, Schwefelsäure und Kohlensäure usf.
Man kann beispielsweise 2-Chlor-6-methyl- anilin oder auch ein Salz desselben in Gegen- wart eines für diese Zwecke geeigneten was serabspaltenden Mittels wie Phosphorpent- oxyd, Phosphoroxyehlorid usf, mit Pyrroli- dinoessigsäure behandeln.
Weiter kann man auch 2-Chlor-6-methy 1- anilin bzw. ein Salz desselben mit einem Pyr- rolidinoessigsäurehalogenid (bzw. einem Salz eines solchen) oder einem Anhydrid zur Re aktion bringen.
Es ist weiterhin auch möglich, 2-Chlor-6- methyl - anilin mit einem Pyrrolidinoessig- säureester, vorzugsweise einem Arylester, bei erhöhter Temperatur zu acylieren oder bei spielsweise ein Halogen-magnesyl-2-chlor-6- methyl-anilin (erhalten durch Umsetzen der Base mit einem Alkylmagnesiumhal.id) mit. einem Pyrrolidinoessigsäurealkylester umzu setzen.
Das auf diese Weise erhaltene Pyrr olidino- essigsäure-(2-chlor-6-methyl-anilid) bildet ein farbloses, bei S4-85 schmelzendes Kristall pulver; dessen Hydrochlorid schmilzt bei 1.94 bis 195 . Das neue Anilid soll als Lokal anästhetikum und als Zwischenprodukt zur Verstellung weiterer Derivate Verwendung finden, <I>Beispiel .1:
</I> 20 Gewichtsteile Pyrrolidinoessigsäurechlo- rid-hydrochlorid in Benzol werden mit einer Mischung von 1.4 Gewichtsteilen 2-Chlor-6- methyl-anilin und 20 Gewichtsteilen Triäthyl- amin erwärmt. Anschliessend wird das gebil dete Triäthylaminhydrochlorid durch Aus- schütteln mit Wasser entfernt und die Denzol- lösung nach dem Trocknen verdampft.
Der Rückstand stellt das Pyr rolidinoessigsäure- (2.. ehlor-6-methyl-anilid) dar, das durch Va kuumdestillation oder Überführen in das Hy- drochlorid gereinigt werden kann.
Die Reaktion kann auch ohne Verdün- nungs- und Kondensationsmittel durchge führt werden, indem man beispielsweise das PyrrolidinoessigsäLire-elilorid-hyclroclilorid mit ?-Clilor-6-methyl-anilin trocken erhitzt, an schliessend die Reaktionsmasse mit Wasser verdünnt und in üblicher Weise aufarbeitet.
Beispiel <I>2:</I> Ein Mol Brom-magnesy 1-2-chlor-6-methyl- anilin (gebildet aus 2-Chlor-6-metlnyl-anilin und Äthylmagnesiumbromid) in Äther, wird mit<B>1,1</B> bis 1,2 Molen Py rrolidinoessigsäure- äthylester umgesetzt. Man gelangt so ebenfalls zu dem gewünschten Anilid.
