Beispiel 2 :
Ein Mol Brom-magnesyl-2-chlor-6-methylanilin (gebildet aus 2-Chlor-6-methyl-anilin und Äthylmagnesiumbromid) in Äther, vTird mit 1, 1 bis 1, 2 Molen ss-Diäthylamino-propion säureäthylester umgesetzt. Man gelant so ebenfa. lls zu dem gewunschten A. nilid.
Process for the production of a new basic substituted fatty acid (2-halogen-6-methyl-anilide).
The subject of the present patent is a process for the preparation of a new basic substituted fatty acid (2-halo-6-methyl-anilide), which is characterized by a compound of the formula
EMI1.1
a compound of the formula
EMI1.2
in which formulas X and Y mean reactive radicals which split off during the reaction, can act.
Compounds of the formula I that can be used for conversion are, for example: '-chloro-6-methyl-aniline, possibly in the form of its salts, such as the hydrochloride or sulfate, and the N-halomagnesyl compounds (X = Mg . Hal) etc.
Examples of possible compounds of formula II are: β-diethylamino-propionic acid itself and its reactive functional derivatives such as its halides, ilii-e esters, their anhydrides, for example the pure and mixed anhydrides with phosphoric acid, sulfuric acid and carbonic acid, etc. .
For example, 2-chloro-6-methyl-aniline or a salt thereof can be treated with β-diethylamino-propionic acid in the presence of a dehydrating agent suitable for this purpose, such as phosphorus pentoxide, phosphorus oxychloride, etc.
In addition, 2-chloro-6-methyl aniline or a salt thereof can also be reacted with an β-diethylamino-propionic acid halide (or a salt thereof) or an anhydride.
It is also possible to acylate 2-chloro-6methyl-aniline with an β-diethylamino-propionic acid ester, preferably an aryl ester, at elevated temperature or, for example, a halomagnesyl-2-chloro-6-methyl-aniline (obtained from Reacting the base with an alkylmagnesium halide) with an s-diethylamino-propionic acid alkyl ester.
The β-diethylamino-propionic acid (2-chloro-6-methyl-anilide) obtained in this way forms a colorless oil that does not boil undecomposed even in a high vacuum; its hydrochloride melts at 113-114. The new anilide is to be used as a local anesthetic and as an intermediate product for the production of further derivatives.
Example 1 :
20 parts by weight of fi-diethylamino-propionic acid-ehlorid-hydroehlorid in benzene are warmed with a mixture of 14 parts by weight of 2-chloro-6-methyl-aniline and 20 parts by weight of triethylamine. The triethylamine hydrochloride formed is then removed by shaking it out with water and the benzene solution is evaporated after drying.
The residue is the β-diethylamino-propionic acid (-chloro-6-methyl-anilide), which can be purified by converting it into the hydrocholide.
The reaction can also be rotated without using diluents and condensation agents, for example by heating the-diethylamino-propionic acid chloride hydrochloride dry with 2-chloro-6-methyl-aniline, then diluting the reaction mixture with water and pouring it in usually worked up.
Example 2:
One mole of bromomagnesyl-2-chloro-6-methylaniline (formed from 2-chloro-6-methyl-aniline and ethylmagnesium bromide) in ether is reacted with 1, 1 to 1, 2 moles of ß-diethylamino-propionic acid ethyl ester. You just get there. lls to the desired A. nilid.
