Verfahren zur Herstellung eines neuen basisch substituierten Fettsäure (2-halogeu-6-methyl-anilids).
Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung eines neuen basisch substituierten Fettsäure- (2-halogen-6- rncthyl-anilids), welches dadurch geliennzeich- net ist, dass man auf eine Verbindung der Formel

in welchen Formeln X und Y reaktionsfähige, bei der Reaktion sich abspaltende Reste bedeuten, einwirken lässt.
Als Verbindungen der Formel I kommen für einen Umsatz beispielsweise in Frage: 2-Chlor-6-methylanilin, eventuell in Form seiner Salze, wie des Hydroc.hlorids oder des Sulfates, weiter die N-Halogen-magnesyl-Verbindungen (X = Mg. Hal) usf.
Als Verbindungen der Formel II kommen beispielsweise in Frage: ss-Dimethylamino-pro pionsäure selbst und ihre reaktionsfähigen funktionellen Derivate wie ihre Halogenide, ihre Ester, ihre Anhydride, beispielsweise die reinen und auch die gemischten Anhydride mit Phosphorsäure, Schwefelsäure und Soh- lensäure usf.
Man kann beispielsweise 2-Chlor-6-methylanilin oder auch ein Salz desselben in Gegenwart eines für diese Zwecke geeigneten wasserabspaltenden Mittels wie Phosphorpentoxyd, Phosphoroxychlorid usf. mit yS-Dimethyl- amino-propionsäure behandeln.
Weiter kann man auch 2-Chlor-6-methylanilin bzw. ein Salz desselben mit einem ss-Dimethylamino-propionsäurehalogenid (bzw. einem Salz eines solchen) oder einem Anhydrid zur Reaktion bringen.
Es ist weiterhin auch möglich, 2-Chlor-6- methvl-anilin mit einem B-Dimethylamino-propionsäureester, vorzugsweise einem Arylester, bei erhöhter Temperatur zn acylieren oder beispielsweise ein Halogen-magnesyl-2-ehlor-6- methyl-anilin (erhalten durch Umsetzen der Base mit einem Alkylmagnesiumhalid) mit einem 8-Dimethylamino- propionsäurealkyl- ester umzusetzen.
Das auf diese Weise erhaltene ss-Dimethyl- amino-propionsäure- (2-ehlor-6-methyl-anilid) bildet ein farbloses, auch im Hochvakuum nicht unzersetzt siedendes Ö1; dessen Hydrochlorid schmilzt bei 77,5-79,5". Das neue Anilid soll als Lokalanästhetikum und als Zwischenprodukt zur Herstellung weiterer Derivate Verwendung finden.
Beispiel 1:
18 Gewichtsteile ss-Dimethylamino-propionsäurechlorid-hydrochlorid in Benzol werden mit einer Mischung von 14 Gewichtsteilen 2-Chlor-6-methyl-anilin und 20 Gewichtsteilen Triäthylamin erwärmt. Anschliessend wird das gebildete Triäthylaminhydrochlorid durch Ausschütteln mit Wasser entfernt und die Benzol lösung nach dem Trocknen verdampft.
Der Rückstand stellt das ss-Dimethylaminopropionsäure- (2-chlor-6-methyl-anilid) dar, das durch Überführen in das Hydrochlorid gereinigt werden kann.
Die Reaktion kann auch ohne Verdün nungs und Kondensationsmittel durchgeführt werden, indem man beispielsweise das ssDi- methylamino- propignsänre- chlorid-hydrochlo- rid mit 2-Chlor-6-methyl-anilin trocken erhitzt, anschliessend die Rcalftionsmasse mit Wasser verdünnt und in üblicher Weise aufarbeitet.
Beispiel 2:
Ein Mol Brom-magnesyl-2-chlor-6-methylanilin (gebildet aus 2-Chlor-6-methyl-anilin und Äthylmagnesiumbromid) in Äther, wird mit 1,1 bis 1,2 Molen ss-Dimethylamino-pro- pionsäureäthylester umgesetzt. Man gelangt so ebenfalls zu dem gewünschten Anilid.

Compounds of the formula I that can be used for conversion are, for example: 2-chloro-6-methylaniline, possibly in the form of its salts, such as hydrochloride or sulfate, and the N-halogenomagnesyl compounds (X = Mg. Hal) etc.
Examples of possible compounds of the formula II are: ß-dimethylamino-propionic acid itself and its reactive functional derivatives such as its halides, its esters, its anhydrides, for example the pure and also the mixed anhydrides with phosphoric acid, sulfuric acid and sole acid, etc.
For example, 2-chloro-6-methylaniline or a salt thereof can be treated with γS-dimethylamino-propionic acid in the presence of a dehydrating agent suitable for this purpose, such as phosphorus pentoxide, phosphorus oxychloride, etc.

The β-dimethylamino-propionic acid (2-chloro-6-methyl-anilide) obtained in this way forms a colorless oil that does not boil undecomposed even in a high vacuum; its hydrochloride melts at 77.5-79.5 ". The new anilide is said to be used as a local anesthetic and as an intermediate for the production of other derivatives.
Example 1:
18 parts by weight of β-dimethylamino-propionic acid chloride hydrochloride in benzene are heated with a mixture of 14 parts by weight of 2-chloro-6-methyl-aniline and 20 parts by weight of triethylamine. The triethylamine hydrochloride formed is then removed by shaking with water and the benzene solution is evaporated after drying.
The residue is ss-dimethylaminopropionic acid (2-chloro-6-methyl-anilide), which can be purified by converting it into the hydrochloride.
The reaction can also be carried out without a diluent and condensing agent, for example by heating the ss-dimethylamino-propignsane chloride hydrochloride with 2-chloro-6-methyl-aniline to dryness, then diluting the calcination mass with water and in the usual way worked up.
Example 2:
One mole of bromagnesyl-2-chloro-6-methylaniline (formed from 2-chloro-6-methyl-aniline and ethylmagnesium bromide) in ether is reacted with 1.1 to 1.2 moles of ethyl β-dimethylamino-propionate. The desired anilide is also obtained in this way.
