Abstract

Reivindicación 1: Un método para modificar el genoma de una célula vegetal de algodón en un sitio predefinido, caracterizado porque comprende los pasos de a) inducir la ruptura de ADN de cadena doble en la cercanía o en dicho sitio predefinido, donde dicha ruptura de cadena doble es inducida por la introducción en dicha célula de una enzima endonucleasa de clivaje raro que reconoce una secuencia de reconocimiento en la cercanía de o en dicho sitio predefinido; b) seleccionar una célula vegetal en donde dicha ruptura de ADN de cadena doble ha sido reparada dando como resultado una modificación en el genoma en dicho sitio preseleccionado, donde dicha modificación se selecciona entre i) un reemplazo de al menos un nucleótido; ii) la supresión de al menos un nucleótido; iii) una inserción de al menos un nucleótido; o iv) cualquier combinación de (i) - (iii); que se caracteriza porque dicha célula está comprendida dentro de los callos embrionarios.Reivindicación 20: El método de acuerdo con reivindicación 19, caracterizado porque dicho gen de interés que se puede expresar en plantas se selecciona del grupo de un gen de tolerancia a herbicidas, un gen de resistencia a insectos, un gen de resistencia a enfermedades, un gen de resistencia al estrés abiótico, una enzima relacionada con la biosíntesis de aceite, la biosíntesis de carbohidratos, una enzima relacionada con la fuerza de las fibras o con la longitud de las fibras, una enzima relacionada con la biosíntesis de metabolitos secundarios. Reivindicación 26: Una planta o fibra o semilla de de algodón o un material propagador de la misma, caracterizada porque comprende una modificación en un sitio predefinido del genoma, obtenida mediante el método de acuerdo con la reivindicación 25 o que consiste esencialmente de las células vegetales de algodón de la reivindicación 25. Reivindicación 28: Un método para producir una planta que comprende una modificación en un sitio predefinido del genoma, caracterizado porque comprende el paso de cruzar una planta que consiste esencialmente de las células vegetales de la reivindicación 25 o una planta de acuerdo con la reivindicación 26 con otra planta o consigo misma y opcionalmente cosechar las semillas.
