Abstract

Reivindicación 1: Un método para introducir una molécula de ADN extraño en un sitio predefinido en un genoma de una célula vegetal, caracterizado porque comprende los pasos de a) inducir una ruptura de ADN de cadena doble en dicho sitio predefinido; b) introducir dicha molécula de ADN extraño en dicha célula vegetal; y c) seleccionar una célula vegetal en donde dicho ADN extraño se introduce en dicho sitio predefinido; y caracterizado porque dicho sitio predefinido está comprendido dentro una región de ADN que codifica una fosfinotricina-cetiltransferasa codificada por Streptomyces hygroscopicus (región de codificación de bar) y que dicha ruptura de ADN de cadena doble es inducida por introducción de una meganucleasa de cadena simple o por un par de meganucleasas que reconoce o reconocen conjuntamente dicho sitio predefinido e induce o inducen dicha ruptura de cadena doble.Reivindicación 21: Una planta, caracterizada porque dicho ADN extraño se introdujo en dicho sitio predefinido, obtenido mediante el método de acuerdo con la reivindicación 19. Reivindicación 22: Una semilla o una parte de propagación de una planta, caracterizada porque comprende dicho ADN extraño integrado en dicho sitio predefinido, de la planta de la reivindicación 21. Reivindicación 23: Un método para cultivar una planta de acuerdo con la reivindicación 21, caracterizado porque comprende el paso de aplicar un compuesto químico a dicha planta o sustrato en donde se cultiva dicha planta. Reivindicación 24: Un proceso para producir una planta que comprende ADN extraño integrado en la región de codificación de bar, caracterizado porque comprende el paso de cruzar una planta que consiste esencialmente de las células vegetales de la reivindicación 20 con otra planta o consigo misma y opcionalmente cosechar las semillas. Reivindicación 28: Uso de una meganucleasa personalizada, caracterizado porque es para introducir un ADN extraño de interés en un sitio predefinido en una célula vegetal.
