(12)α-ヒドロキシ酸類:グリコール酸、クエン酸、リンゴ酸、酒石酸、乳酸等、乳酸ナトリウム等。
(20)収斂剤:コハク酸、アラントイン、塩化亜鉛、硫酸亜鉛、酸化亜鉛、カラミン、パラフェノールスルホン酸亜鉛、硫酸アルミニウムカリウム、レゾルシン、塩化第二鉄、タンニン酸(カテキン化合物を含む)等。
(21)抗酸化剤:ヒドロキシチロソール、パラヒドロキシアニソール、没食子酸プロピル、セサモール、セサモリン、ゴシポール、プロポリス等。
(22)抗炎症剤:インドメタシン、サリチル酸、サリチル酸ナトリウム、アセチルサリチル酸、塩酸ジフェンヒドラミン、d-カンフル、dl-カンフル、ヒドロコルチゾン、グアイアズレン、カマズレン、マレイン酸クロルフェニラミン、グリチルリチン酸又は甘草エキス等。
試薬購入(chemsigma社製)したロツンゲン酸0.89 mgを1mlのメタノールに溶解し、下記条件でLC/MS分析を行った。
分析機器:ACQUITY UPLC Class H(Waters社製)
質量分析計:Xevo G2-S-QTof(Waters社製)
フォトダイオードアレイ:(210~500nm)
カラム:ACQUITY UPLC BEH C18 (50×2.1mm、I.D.2.1μm)
流速:0.8 mL/min
カラム温度:55℃
溶媒;A液:0.1%ギ酸水溶液、B液:0.1%ギ酸アセトニトリル溶液
グラジエント;
0min;%A液/%B液=95/5 → 6min;%A液/%B液=0/100
6min;%A液/%B液=0/100 → 7min;%A液/%B液=0/100
結果は図1に示した通り、ポジティブでm/z:977.7、ネガティブでm/z:487.2であった。ロツンゲン酸の分子量(文献値)は488.7(C30H48O5)であることから、ポジティブで検出した質量は[2M+H]+、ネガティブで検出した質量は[M-H]-であった。
ロツンゲン酸(chemsigma社製)13.0mgをC5D5N(600 μL)に溶かし、13C-NMR測定を行った。さらに上記ロツンゲン酸溶液をC5D5Nで6倍に薄め1H-NMR測定も行った。
結果は表1に示した化学シフトの値を示し、*既報(Phytochemistry 28(5) 1479-1482(1989))に記載された値と一致したことから化合物はロツンゲン酸であることが確認された。
ロツンゲン酸(chemsigma社製)を常法により、ロツンゲン酸のナトリウム塩、カルシウム塩、を調製した。
B16メラノーマ細胞(RIKEN BRC)を60mmのdishにD-MEM(Wako社)にて、24時間培養後、試料(ロツンゲン酸(chemsigma社製)、ウルソール酸(INDOFINE Chemical社製)をそれぞれ終濃度0.8μMとなるように、コントロール(比較対照)はDMSOを終濃度1%となるように)を添加して1時間培養を行い、その後α-MSH(SIGMA-ALDRICH社)を終濃度50ng/mLとなるように添加した。α-MSHの添加後24時間で細胞からRNeasy Mini Kit(QIAGEN社)を用いて抽出を行い、得られたRNAを鋳型にPrimeScript RT reagent Kit(タカラバイオ社)を用いてcDNAを合成した。このcDNAを用いて定量PCR法によりチロシナーゼ遺伝子の発現量を測定した。
結果は、図2に示した通りである。α-MSHの添加でTyrosinase遺伝子の発現が亢進された条件下で、ウルソール酸に遺伝子発現の抑制効果が認められなかった。一方、ロツンゲン酸は、0.8μMの濃度にてTyrosinase遺伝子発現の大きな抑制効果が認められた。なお、ロツンゲン酸が0.1μMの条件でも、0.8μMの濃度の場合に比べて若干低いものの当該抑制効果が認められた。また、試験3により得たロツンゲン酸のナトリウム塩、カルシウム塩についても終濃度0.8 μMで添加し、試験4を行ったところ、同様な効果が得られた。
(1)被検動物及び群構成
ヒトと同様に紫外線照射により色素沈着が生じる動物種である有色モルモットを用いて、株式会社日本バイオリサーチセンターにて紫外線誘発による色素沈着抑制試験を行った。試験サンプル及び紫外線照射方法の異なる以下の6つの群に分けた。

第5群 0.009%ロツンゲン酸(照射)、媒体(照射) 6
富有柿の蔕を粉末化し、有機溶媒にて抽出、分画及び精製して真空乾燥粉末を取得した。上記試験1及び2と同様の方法でHPLCピーク及びNMR分析を行った結果、純粋なロツンゲン酸であることを確認した。エタノール:プロピレングリコール:精製水=6:2:2の媒体にそれぞれ、重量比で0.9%、0.09%及び0.009%となるように溶解し、ロツンゲン酸を含む被験サンプルとして試験に供した。また、コントロールには、60%(W/W)エタノール、20%(W/W)プロピレングリコール水溶液を被験サンプルとして使用した。
各群6匹のモルモットを使用し、それぞれの背部を電気バリカン及び電気シェバーで除毛し、正中を挟んで左右の、2×2cmの正方形2か所を測定部位とした。紫外線照射には紫外線照射装置にて、UV-B(波長250~350nm)を最小紅斑量照射した。紫外線照射は、初回投与日(この日を0日とする。)と、それ以降は初回投与後2日及び4日の計3回行った。
投与濃度は0.1mL/部位とした。投与回数は、いずれも1日1回、投与期間は28日間連続とした。なお、紫外線照射日は紫外線照射後に、色素沈着の測定部は測定後に投与を行った。
初回投与日の投与前(照射前)、投与14日、21日、28日に、色彩色差計を用いて、照射部位についてLab表色系で表示し、L値(明度)を評価に用いた。ΔL値(観察日のL値-照射前のL値)を求めた。また、各群のL値を用いて、群ごとで対応のあるt検定により有意差検定を行った
被験モルモットの紫外線(UV-B)照射時の皮膚色(ΔL値)の推移を図3に示した。紫外線照射開始後、色素沈着によるL値の低下が認められた。紫外線照射量の変動による測定を避けるため、各群において媒体のみを塗布したコントロールとの比較を行った。本発明に係るロツンゲン酸を0.9重量%含む被験サンプルを塗布した第3群は、コントロールの媒体に比べて有意にメラニン色素沈着抑制の効果を示した(p<0.05)。また、ロツンゲニン酸を0.09重量%又は0.009重量%含む被験サンプルを塗布した第4群及び第5群は、t検定では有意差が認められなかったものの、濃度依存的にL値の低下が改善されていること、及び上述した試験4の結果から0.00004重量%(約0.8μM)でもチロシナーゼ発現抑制作用を示したことから、0.009~0.9重量%の範囲内でメラニン生成抑制作用を有すると判断した。
ロツンゲン酸(chemsigma社製)、ラウリル硫酸Na、精製水を30:3:67の重量比で分散させ、高圧ホモジナイザーによって湿式粉砕した。粉砕条件は処理圧力:200MPa、処理回数:80回で行い、得られた懸濁液を処方例6をもとに、下記表2に示した処方a~c3種の美白クリームを作製し、専門パネラーによる使用感の評価を行った。なお、使用感の評価項目は塗布時ののび、塗布後のなめらかさである。
結果は下記表3の通り、ロツンゲン酸の質量濃度が1%、5%では平均スコアが3以上であるが、7%では平均スコアが2以下となり、外用剤中のロツンゲン酸の質量濃度が5%以下であれば、使用感のよい外用剤が得られるものと予測する。

1:良好な使用感はなかった
本発明の処方例6をもとに、下記表4に示した処方A~D4種の美白クリームを作製した。成人男女20名に対して、作製した美白クリームを顔に1日2回塗布、1ヶ月間の使用試験を行った。なお、被験者は5人ずつにわけ、シミや色素沈着について使用前に比べて評価した。
結果は下記表5の通り、ロツンゲン酸を配合した処方が対照品よりも高く、特に0.1%配合した処方Bが他の処方よりも高いことが認められた。
有 効 :使用前よりもシミや色素沈着が改善された。
やや有効 :使用前よりもシミや色素沈着がやや改善された。
無 効 :使用前と変化なし。
Claims (10)
- ロツンゲン酸またはその塩を、美白用有効成分として0.00004~5重量%含有する美白剤。
- ロツンゲン酸またはその塩を、美白用有効成分として0.00004~5重量%含有する美白用皮膚外用剤。
- 前記ロツンゲン酸またはその塩の含有量が、少なくとも0.009重量%である請求項1に記載の美白剤。
- 前記ロツンゲン酸またはその塩の含有量が、少なくとも0.009重量%である請求項2に記載の美白用皮膚外用剤。
- ロツンゲン酸またはその塩を有効成分として含む組成物の有効量を、その必要のある対象者に投与することを含む皮膚の美白方法。
- ロツンゲン酸またはその塩を少なくとも0.009重量%含む組成物で皮膚を処理することを含む皮膚の美白方法。
- 前記皮膚の処理が、前記皮膚が紫外線に暴露される前またはその直後に行われる請求項6に記載の美白方法。
- 美白剤または美白用皮膚外用剤の有効成分として使用するための、ロツンゲン酸またはその塩を含む組成物。
- ロツンゲン酸またはその塩を有効成分として含む組成物の、皮膚の美白処理のための使用。
- 美白剤または美白用皮膚外用剤の製造における、ロツンゲン酸またはその塩の使用。
