AAは、アミノ酸残基を示す。
本明細書中、AAで示される「アミノ酸残基」とは、アミノ酸から1個のアミノ基と1個のカルボキシ基を除いた二価の基を意味する。当該アミノ酸残基におけるアミノ酸としては、アミノ基とカルボキシ基を有する限り特に制限されず、α-アミノ酸、β-アミノ酸、γ-アミノ酸等のいずれでもよい。α-アミノ酸としては、グリシン、アラニン、バリン、ロイシン、イソロイシン、セリン、スレオニン、システイン、メチオニン、グルタミン酸、アスパラギン酸、リジン、アルギニン、ヒスチジン、グルタミン、アスパラギン、フェニルアラニン、チロシン、トリプトファン、シスチン、オルニチン、チロキシン、プロリン、3,4-ジヒドロキシフェニルアラニン等;
β-アミノ酸としては、β-アラニン等;
γ-アミノ酸としては、γ-アミノ酪酸等;が挙げられる。側鎖に官能基を有する場合、当該官能基は、糖アミノ酸の物性(特に水溶性、水中安定性、苦味等)に悪影響を与えない範囲で保護/修飾されていてもよい。
上記アミノ酸は、D体、L体、DL体のいずれでもよい。
本明細書中、Gで示される「全ての水酸基が保護も修飾もされていない糖残基」とは、全ての水酸基がフリーである糖からヘミアセタール水酸基を除いた部分を意味する。当該糖残基は、全ての水酸基がフリーである限り、修飾/改変されていてもよい。「全ての水酸基が保護も修飾もされていない糖残基」としては、グルコース、グルコサミン、N-アセチルグルコサミン、マンノース、ガラクトース、フルクトース、リボース、リキソース、キシロース、アラビノース等の単糖;これらの単糖からなる多糖等の糖類から、ヘミアセタール水酸基を除いた部分が挙げられる。
中でも、グルコース残基、グルコサミン残基およびN-アセチルグルコサミン残基が好ましく、グルコース残基およびN-アセチルグルコサミン残基がより好ましく、グルコース残基が特に好ましい。
上記糖類は、D体、L体のいずれでもよいが、自然界に多く存在するD体が好ましい。
Rで示される「アルキル基」としては、C1-10アルキル基が挙げられ、より好ましくはC1-6アルキル基である。好適な具体例としては、メチル、エチル、プロピル、イソプロピル、ブチル、イソブチル、sec-ブチル、tert-ブチル、ペンチル、ヘキシル等が挙げられる。
Rは、好ましくは水素原子である。

原料化合物は、特に述べない限り、市販されているものを容易に入手できるか、あるいは、自体公知の方法またはこれらに準ずる方法に従って製造することができる。
以下の各方法で得られる化合物の収率は用いる反応条件によって異なりうるが、これらの生成物から通常の手段(再結晶、カラムクロマトグラフィー等)によって単離・精製し、次いで、溶液温度を変化させる手段や溶液組成を変化させる手段等によって沈殿化することができる。
また、各反応において、原料化合物であるアミノ酸が側鎖にヒドロキシ基、アミノ基、カルボキシ基、カルボニル基等を有する場合、これらの基にペプチド化学等で一般的に用いられるような保護基が導入されていてもよく、反応後に必要に応じて保護基を除去することにより目的化合物を得ることができる。
当該工程は、化合物(1)またはその塩のアミノ基をイソシアナト基に変換して、化合物(2)を得る工程である。
当該反応は、通常、反応に影響を及ぼさない溶媒中、化合物(1)またはその塩を塩基の存在下、二炭酸ジtert-ブチル(Boc2O)と反応させることにより行われる。
二炭酸ジtert-ブチルの使用量は、化合物(1)またはその塩1モルに対して、通常0.7~5モル、好ましくは1~2モルである。
塩基としては、4-(ジメチルアミノ)ピリジン等が挙げられる。
塩基の使用量は、化合物(1)またはその塩1モルに対して、通常0.5~3モル、好ましくは1~2モルである。
溶媒としては、反応が進行する限り特に限定されるものではないが、例えば、炭化水素類(例、ベンゼン、トルエン、キシレン、ヘキサン、ヘプタン等)、ハロゲン化炭化水素類(例、クロロホルム、ジクロロメタン等)、エーテル類(例、ジエチルエーテル、ジイソプロピルエーテル、tert-ブチルメチルエーテル、テトラヒドロフラン、1,4-ジオキサン、1,2-ジメトキシエタン等)あるいはそれらの混合物が用いられる。中でも、ジクロロメタンが好ましい。
反応温度は、通常-100~100℃、好ましくは-30~50℃であり、反応時間は、通常0.5~30時間、好ましくは1~5時間である。
反応終了後、化合物(2)は、単離せずに反応混合物のまま次の工程に供される。
なお、化合物(1)が酸付加塩の形態であるときは、塩基で処理して遊離体に変換した後、当該工程に供するか、過剰の塩基の存在下に反応させればよい。
当該工程は、化合物(2)をG1-OHと反応させることにより、化合物(3)を得る工程である。G1-OHはヘミアセタール水酸基以外の水酸基が全て保護された糖である。
当該反応は、通常、反応に影響を及ぼさない溶媒中、化合物(2)をG1-OHと反応させることにより行われる。
G1-OHの使用量は、化合物(2)1モルに対して、通常0.7~5モル、好ましくは1~2モルである。
溶媒としては、反応が進行する限り特に限定されるものではないが、例えば、炭化水素類(例、ベンゼン、トルエン、キシレン、ヘキサン、ヘプタン等)、ハロゲン化炭化水素類(例、クロロホルム、ジクロロメタン等)、エーテル類(例、ジエチルエーテル、ジイソプロピルエーテル、tert-ブチルメチルエーテル、テトラヒドロフラン、1,4-ジオキサン、1,2-ジメトキシエタン等)あるいはそれらの混合物が用いられる。中でも、ジクロロメタンが好ましい。
反応温度は、通常-100~100℃、好ましくは-30~50℃であり、反応時間は、通常3~40時間、好ましくは10~30時間である。
こうして得られる化合物(3)は、公知の分離精製手段、例えば、濃縮、減圧濃縮、溶媒抽出、晶出、再結晶、転溶、クロマトグラフィー等により、単離精製することができる。また、化合物(3)は、単離せずに次の反応に用いてもよい。
当該工程は、化合物(3)のカルボキシ基の保護基とG1に存在する水酸基の保護基を除去して、化合物(Ia)またはその塩を得る工程である。
カルボキシ基の保護基の除去とG1に存在する水酸基の保護基の除去は、同時に行っても、別工程で行ってもよく、後者の場合は、その順序は問わないが、同時に行う方が簡便である。その場合は、これらの保護基は、同じ条件で除去できるように選択される。例えば、R1で示されるカルボキシ基の保護基がメチルまたはエチルであり、G1に存在する水酸基の保護基がアセチルである場合、これらはアルカリ加水分解で除去される。
アルカリとしては、水酸化リチウム、水酸化ナトリウム、水酸化カリウム、水酸化バリウム等が挙げられ、中でも、水酸化リチウムが好ましい。
溶媒としては、反応が進行する限り特に限定されるものではないが、例えば、水、アルコール類(例、メタノール、エタノール、イソプロピルアルコール、tert-ブチルアルコール等)、エーテル類(例、ジエチルエーテル、ジイソプロピルエーテル、tert-ブチルメチルエーテル、テトラヒドロフラン、1,4-ジオキサン、1,2-ジメトキシエタン等)、ハロゲン化炭化水素類(例、ジクロロメタン等)あるいはそれらの混合物が用いられる。中でも、水とアルコール類(例、メタノール、エタノール、イソプロピルアルコール、tert-ブチルアルコール等)の混合物が好ましい。
反応温度は、通常-100~100℃、好ましくは-30~35℃であり、反応時間は、通常5~10時間、好ましくは0.5~2時間である。
こうして得られる化合物(Ia)またはその塩は、公知の分離精製手段、例えば、濃縮、減圧濃縮、溶媒抽出、晶出、再結晶、転溶、クロマトグラフィー等により、単離精製することができる。
Glc-XXXは、アミノ基がD-グルコピラノシルオキシカルボニル基でカルバメート化されたアミノ酸(XXX)を意味し、
4Ac-Glc-XXXは、アミノ基が2,3,4,6-テトラ-O-アセチル-D-グルコピラノシルオキシカルボニル基でカルバメート化されたアミノ酸(XXX)を意味し、

以下の実施例中の「室温」は通常約10℃ないし約35℃を示す。混合溶媒において示した比は、特に断らない限り容量比を示す。
1H-NMR(プロトン核磁気共鳴スペクトル)はフーリエ変換型NMRで測定した。ヒドロキシ基、カルボキシ基、アミノ基等のプロトンが非常に緩やかなピークについては記載していない。
L-ロイシンメチルエステル塩酸塩(293 mg, 1.61 mmol)をテトラヒドロフラン(3.5 ml)に懸濁させ、氷浴を用いて冷却した。この溶液にトリエチルアミン(4.3 ml, 30.8 mmol)を加えた後、室温に昇温して30分間攪拌した。反応溶液をろ別し、濃縮してL-ロイシンメチルエステル(232 mg, 1.61 mmol)を得た。
Boc2O(493 mg, 2.26 mmol)をジクロロメタン(10 ml)に溶解させ、氷浴を用いて冷却した。この溶液に4-(ジメチルアミノ)ピリジン(198 mg, 1.62 mmol)を溶かしたジクロロメタン(7 ml)溶液とL-ロイシンメチルエステル(232 mg, 1.61 mmol)を溶かしたジクロロメタン(7 ml)溶液を加え、室温にて1時間攪拌した。再び氷浴を用いて反応溶液を冷却し、2,3,4,6-テトラ-O-アセチル-D-グルコース(787 mg, 2.26 mmol)を溶かしたジクロロメタン(10 ml)溶液を加え、18時間攪拌した。反応溶液を減圧濃縮し、残渣をシリカゲルカラムクロマトグラフィー(グラジエント;ヘキサン:酢酸エチル=85:15→60:40)にて精製し、4Ac-Glc-Leu-OMe(698 mg, 1.34 mmol, 収率83%,α:β比=1:1)を白色粉末として得た。

4Ac-Glc-Leu-OMe(300 mg, 0.577 mmol)をメタノール(6 ml)と水(3 ml)に溶解させ、恒温槽を用いて-10℃に冷却した。この溶液に1N水酸化リチウム水溶液(2.89 ml, 2.89 mmol)を加え、10分間攪拌した。反応溶液に水(15 ml)を加え、20分間攪拌した。反応液を強酸性樹脂(Amberlite IR-120)で処理し、続いて樹脂をろ別した。ろ液を減圧濃縮し、Glc-Leu(199 mg, 収率quant.,α:β比=1:1)を白色粉末として得た。

L-フェニルアラニンメチルエステル塩酸塩(319 mg, 1.48 mmol)をテトラヒドロフラン(4 ml)に懸濁させ、氷浴を用いて冷却した。この溶液にトリエチルアミン(4 ml, 28.6 mmol)を加えた後、室温に昇温して1時間攪拌した。反応溶液をろ別し、濃縮してL-フェニルアラニンメチルエステル(254 mg, 1.42 mmol)を得た。
Boc2O(431 mg, 1.98 mmol)をジクロロメタン(10 ml)に溶解させ、氷浴を用いて冷却した。この溶液に4-(ジメチルアミノ)ピリジン(172 mg, 1.40 mmol)を溶かしたジクロロメタン(7 ml)溶液とL-フェニルアラニンメチルエステル(254 mg, 1.42 mmol)を溶かしたジクロロメタン(8 ml)溶液を加え、室温にて1時間攪拌した。再び氷浴を用いて反応溶液を冷却し、2,3,4,6-テトラ-O-アセチル-D-グルコース(682 mg, 1.96 mmol)を溶かしたジクロロメタン(10 ml)溶液を加え、18時間攪拌した。反応溶液を減圧濃縮し、残渣をシリカゲルカラムクロマトグラフィー(グラジエント;ヘキサン:酢酸エチル=85:15→57:43)にて精製し、4Ac-Glc-Phe-OMe(769 mg, 1.39 mmol, 収率94%,α:β比=1:1)を白色粉末として得た。

4Ac-Glc-Phe-OMe(394 mg, 0.711 mmol)をメタノール(15 ml)に溶解させ、恒温槽を用いて-10℃に冷却した。この溶液に1N水酸化リチウム水溶液(3.55 ml, 3.55 mmol)を加え、15分間攪拌した。反応溶液に水(40 ml)を加え、40分間攪拌した。反応液を強酸性樹脂(Amberlite IR-120)で処理し、続いて樹脂をろ別した。ろ液を減圧濃縮し、Glc-Phe(262 mg, 0.705 mmol, 収率99%,α:β比=1:1)を白色粉末として得た。

N-ε-(ベンジルオキシカルボニル)-L-リジンメチルエステル塩酸塩(2.71 g, 8.21 mmol)をテトラヒドロフラン(16 ml)に懸濁させ、氷浴を用いて冷却した。この溶液にトリエチルアミン(22.8 ml, 163 mmol)を加えた後、室温に昇温して1時間攪拌した。反応溶液をろ別し、濃縮してN-ε-(ベンジルオキシカルボニル)-L-リジンメチルエステル(2.48 g)を得た。
Boc2O(2.48 g, 11.3 mmol)をジクロロメタン(30 ml)に溶解させ、氷浴を用いて冷却した。この溶液に4-(ジメチルアミノ)ピリジン(1.01 g, 8.28 mmol)を溶かしたジクロロメタン(30 ml)溶液とN-ε-(ベンジルオキシカルボニル)-L-リジンメチルエステル(2.48 g)を溶かしたジクロロメタン(30 ml)溶液を加え、室温にて1時間攪拌した。再び氷浴を用いて反応溶液を冷却し、2,3,4,6-テトラ-O-アセチル-D-グルコース(4.00 g, 11.5 mmol)を溶かしたジクロロメタン(30 ml)溶液を加え、18時間攪拌した。反応溶液を減圧濃縮し、残渣をシリカゲルカラムクロマトグラフィー(グラジエント;ヘキサン:酢酸エチル=70:30→40:60)にて精製し、4Ac-Glc-Lys(Z)-OMe(3.75 g, 6.63 mmol, 収率81%,α:β比=1:1)を白色粉末として得た。

(2) Glc-Lys(Z);N-α-(α/β-D-グルコピラノシルオキシカルボニル)-N-ε-(ベンジルオキシカルボニル)-L-リジン
4Ac-Glc-Lys(Z)-OMe(682 mg, 1.02 mmol)をメタノール(5.1 ml)に溶解させ、恒温槽を用いて-10℃に冷却した。この溶液に1N水酸化リチウム水溶液(5.1 ml, 5.1 mmol)を加え、15分間攪拌した。反応溶液に水(10 ml)を加え、15分間攪拌した。反応液を強酸性樹脂(Amberlite IR-120)で処理し、続いて樹脂をろ別した。ろ液を減圧濃縮し、Glc-Lys(Z)(510 mg, 1.04 mmol, 収率quant.,α:β比=1:1)を白色粉末として得た。

(3) Glc-Lys;N-α-(α/β-D-グルコピラノシルオキシカルボニル)-L-リジン
Glc-Lys(Z)(53.0 mg, 0.109 mmol)をメタノール(2 ml)に溶解し、2%パラジウム炭素触媒(26.3 mg, 50%(w/w))を加え、水素雰囲気下(大気圧)、室温にて3時間半攪拌を行った。反応終了後、触媒をろ別した。ろ液を減圧濃縮し、Glu-Lys(35.0 mg, 0.993 mmol, 収率91%, α:β比=2:3)を白色粉末として得た。

(2) 4Ac-Glc-Glu-OMe;N-(2,3,4,6-テトラ-O-アセチル-α/β-D-グルコピラノシルオキシカルボニル)-L-グルタミン酸α-メチルエステル
4Ac-Glc-Glu(OBn)-OMe(582 mg;糖原料との混合物)をメタノール(13 ml)に溶解し、2%パラジウム炭素触媒(582 mg, 100%(w/w))を加え、水素雰囲気下(大気圧)、室温にて3時間半攪拌を行った。反応終了後、触媒をろ別した。ろ液を減圧濃縮し、残渣をシリカゲルカラムクロマトグラフィー(グラジエント;ヘキサン:酢酸エチル=1:1→1:3)にて精製し、4Ac-Glc-Glu-OMe(251 mg, 0.470 mmol, 収率60%(2 steps),α:β比=1:1)を白色粉末として得た。

メタノール(50 ml)を、恒温槽を用いて-5℃に冷却し、塩化チオニル(5 ml, 68.9 mmol)を滴下した。続いて3,4-ジヒドロキシ-L-フェニルアラニン(10.0 g, 50.7 mmol)を少しずつ加え、5分間攪拌した。室温に戻し、50℃に加熱し、14時間攪拌した。反応溶液を濃縮し、DOPA-OMe塩酸塩(14.3 g, 57.7 mmol, 収率quant.)を油状物質として得た。

(2) Boc-DOPA-OMe;N-(tert-ブトキシカルボニル)-3,4-ジヒドロキシ-L-フェニルアラニンメチルエステル
DOPA-OMe塩酸塩(1.26 g, 5.11 mmol)をテトラヒドロフラン(10 ml)に溶解させ、飽和重層水(8 ml)を加え、氷浴を用いて冷却した。この溶液にBoc2O(1.00 ml, 4.35 mmol)を加え、室温に昇温し、1時間半攪拌した。反応液を減圧濃縮し、残渣にジクロロメタン(10 ml)および水(5 ml)を加え、2回ジクロロメタンで抽出した。有機層を10%クエン酸水溶液(10 ml)、15%食塩水(10 ml)の順で洗浄後、硫酸マグネシウムで乾燥した。乾燥剤をろ別後、ろ液を減圧濃縮し、得られた残渣をシリカゲルカラムクロマトグラフィー(グラジエント;ヘキサン:酢酸エチル=9:1→1:1)にて精製し、Boc-DOPA-OMe(980 mg, 3.15 mmol, 収率76%)を淡桃色の飴状物質として得た。

(3) Boc-DOPA(OBn)2-OMe;N-(tert-ブトキシカルボニル)-3,4-ジベンジルオキシ-L-フェニルアラニンメチルエステル
Boc-DOPA-OMe(984 mg, 3.15 mmol)をN,N-ジメチルホルムアミド(20 ml)に溶解させ、氷浴を用いて冷却した。この溶液に炭酸カリウム(1.37 g, 9.92 mmol)、ベンジルブロミド(0.860 ml, 7.24 mmol)を加え、室温に昇温後、50℃に加熱して1時間攪拌した。ジエチルエーテル(50 ml)および水(100 ml)を加え、ジエチルエーテルにて2回抽出した。有機層を15%食塩水(50 ml)で洗浄後、硫酸マグネシウムで乾燥した。乾燥剤をろ別後、ろ液を減圧濃縮し、Boc-DOPA(OBn)2-OMe(1.37 g, 2.89 mmol, 収率88%)を白色粉末として得た。

(4) DOPA(OBn)2-OMe塩酸塩;3,4-ジベンジルオキシ-L-フェニルアラニンメチルエステル塩酸塩
Boc-DOPA(OBn)2-OMe(570 mg, 1.16 mmol)をジクロロメタン(6 ml)に溶解させ、氷浴を用いて冷却した。この溶液に4N塩酸/1,4-ジオキサン(2 ml)を加え、室温に昇温し、4時間攪拌した。反応溶液を濃縮し、DOPA(OBn)2-OMe塩酸塩(450 mg, 1.05 mmol, 収率91%)を白色粉末として得た。

実施例1の工程(1)と同様にして、DOPA(OBn)2-OMe塩酸塩(450 mg, 1.15 mmol)より、4Ac-Glc-DOPA(OBn)2-OMe(808 mg)を糖原料との混合物として得た。
(6) 4Ac-Glc-DOPA-OMe;N-(2,3,4,6-テトラ-O-アセチル-α/β-D-グルコピラノシルオキシカルボニル)-3,4-ジヒドロキシ-L-フェニルアラニンメチルエステル
実施例4の工程(2)と同様にして、4Ac-DOPA(OBn)2-OMe(808 mg;糖原料との混合物)より、4Ac-Glc-DOPA-OMe(298 mg, 0.509 mmol, 収率44%(2 steps), α:β比=1:1)を油状物質として得た。

実施例1の工程(2)と同様にして、4Ac-Glc-DOPA-OMe(298 mg, 0.509 mmol)より、Glc-DOPA(183 mg, 0.453 mmol, 収率89%, α:β比=1:1)を淡黄色粉末として得た。

L-バリンメチルエステル塩酸塩(5.37 g, 32.1 mmol)をテトラヒドロフラン(64 ml)に懸濁させ、氷浴を用いて冷却した。この溶液にトリエチルアミン(89 ml, 641 mmol)を加えた後、室温に昇温して30分間攪拌した。反応溶液をろ別し、濃縮してL-バリンメチルエステル(3.29 g, 25.1 mmol, 収率78%)を得た。
Boc2O(7.70 ml, 35.1 mmol)をジクロロメタン(85 ml)に溶解させ、氷浴を用いて冷却した。この溶液に4-(ジメチルアミノ)ピリジン(3.37 g, 27.6 mmol)を溶かしたジクロロメタン(85 ml)溶液とL-バリンメチルエステル(3.30 g, 25.1 mmol)を溶かしたジクロロメタン(85 ml)溶液を加え、室温にて1時間攪拌した。再び氷浴を用いて反応溶液を冷却し、2,3,4,6-テトラ-O-アセチル-D-グルコース(11.9 g, 35.1 mmol)を溶かしたジクロロメタン(85 ml)溶液を加え、18時間攪拌した。反応溶液を減圧濃縮し、残渣をシリカゲルカラムクロマトグラフィー(グラジエント;ヘキサン:酢酸エチル=85:18→1:1)にて精製し、4Ac-Glc-Val-OMe(3.95 g, 7.83 mmol, 収率31%, α:β比=3:2)淡黄色粉末として得た。
1H-NMR(400 MHz, CDCl3)δ:0.88-0.94 (m, 3H), 0.96-0.99 (m, 3H), 2.02-2.09 (m, 12H) 2.17-2.24 (m, 1H), 3.75 (s, 1.2H), 3.77 (s, 1.8H), 4.08-4.16 (m, 1H), 4.23-4.35 (m, 2H), 5.06-5.17 (m, 2H), 5.26 (t, 0.4H, J=9.4 Hz), 5.41(t, 1H, J=9.7 Hz), 5.50 (t, 1H, J=9.9 Hz), 5.66 (d, 0.4H, J=8.2 Hz), 6.24 (d, 0.6H, J=3.7 Hz).
ESIMS(m/z): 523.2([M+NH4]+), 528.2([M+Na]+), 522.0([M+Cl]-).
(2) Glc-Val;N-(α/β-D-グルコピラノシルオキシカルボニル)-L-バリン
4Ac-Glc-Val-OMe(154 mg, 0.310 mmol)をメタノール(1.6 ml)に溶解させ、恒温槽を用いて-10℃に冷却した。この溶液に1N水酸化リチウム水溶液(1.54 ml, 1.54 mmol)を加え、10分間攪拌した。反応溶液に水(3.3 ml)を加え、20分間攪拌した。反応液を強酸性樹脂(Amberlite IR-120)で処理し、続いて樹脂をろ別した。ろ液を減圧濃縮し、Glc-Val(98.0 mg, 0.290 mmol, 収率98%, α:β比=3:7)を淡黄色粉末として得た。
1H-NMR(400 MHz, CD3OD)δ:0.96-1.06 (m, 6H), 2.13-2.24 (m, 1H), 3.30-3.91 (m, 6H), 4.08-4.12 (m, 1H), 5.35 (d, 0.7H, J=7.9 Hz), 5.96 (d, 0.3H, J=3.8 Hz).
ESIMS(m/z): 322.2([M-H]-), 645.2([2M-H]-).
L-イソロイシンメチルエステル塩酸塩(3.00 g, 16.5 mmol)をテトラヒドロフラン(33 ml)に懸濁させ、氷浴を用いて冷却した。この溶液にトリエチルアミン(46.0 ml, 330 mmol)を加えた後、室温に昇温して1時間攪拌した。反応溶液をろ別し、濃縮してL-イソロイシンメチルエステル(2.13 g, 14.7 mmol)を得た。
Boc2O(4.48 g, 20.5 mmol)をジクロロメタン(69 ml)に溶解させ、氷浴を用いて冷却した。この溶液に4-(ジメチルアミノ)ピリジン(1.97 g, 16.1 mmol)を溶かしたジクロロメタン(69 ml)溶液とL-イソロイシンメチルエステル(2.13 g, 20.5 mmol)を溶かしたジクロロメタン(69 ml)溶液を加え、室温にて1時間攪拌した。再び氷浴を用いて反応溶液を冷却し、2,3,4,6-テトラ-O-アセチル-D-グルコース(7.17 g, 20.5 mmol)を溶かしたジクロロメタン(69 ml)溶液を加え、18時間攪拌した。反応溶液を減圧濃縮し、残渣をシリカゲルカラムクロマトグラフィー(グラジエント;ヘキサン:酢酸エチル=82:18→50:50)にて精製し、4Ac-Glc-Ile-OMe(5.72 g, 11.0 mmol, 収率75%,α:β比=1:1)を白色シロップ状物質として得た。
1H-NMR(400 MHz, CDCl3)δ:0.89-0.95(t, 6H, J=7.0 Hz), 1.13-1.27(m, 1H), 1.37-1.46(m, 1H), 1.86-1.94(m, 1H), 2.01-2.09(m, 12H), 3.73(s, 1.5H), 3.76(s, 1.5H), 3.80-3.84(m, 0.5H), 4.07-4.14(m, 1.5H), 4.25-4.36(m, 2H), 5.08-5.17(m, 2H), 5.25(t, 0.5H, J=5.2 Hz), 5.41(dd, 1H, J=5.4, 9.1 Hz), 5.47(t, 0.5H, J=5.5 Hz), 5.65(d, 0.5H, J=8.3 Hz), 6.24(d, 0.5H, J=3.7 Hz).
ESIMS(m/z): 542.2([M+Na]+), 558.1([M+K]+).
(2) Glc-Ile;N-(α/β-D-グルコピラノシルオキシカルボニル)-L-イソロイシン
4Ac-Glc-Ile-OMe(1.01 g, 1.95 mmol)をメタノール(9.6 ml)に溶解させ、氷浴を用いて-10℃に冷却した。この溶液に1N水酸化リチウム水溶液(9.75 ml, 9.75 mmol)を加え、10分間攪拌した。反応溶液に水(19 ml)を加え、15分間攪拌した。反応液を強酸性樹脂(Amberlite IR-120)で処理し、続いて樹脂をろ別した。ろ液を減圧濃縮し、再度同様の作業を計3回(1N水酸化リチウム水溶液(6.9 ml, 14.1 ml, 20.0 ml))行った。ろ液を減圧濃縮し、Glc-Ile(626 mg, 1.85 mmol, 収率95%,α:β比=1:1)を白色粉末として得た。
1H-NMR(400 MHz, CD3OD)δ: 0.91-0.98(m, 6H), 1.21-1.29(m, 1H), 1.44-1.56(m, 1H), 1.82-1.94(m, 1H), 3.35-3.42(m, 1H), 3.53(dd, 0.5H, J=3.8, 9.7 Hz), 3.66-3.85(m, 4.5H), 4.10-4.17(m, 1H), 5.33(d, 0.5H, J=7.9 Hz), 5.94(d, 0.5H, J=3.7 Hz).
ESIMS(m/z):336.1([M-H]-), 673.2([2M-H]-).
L-チロシンメチルエステル塩酸塩(2.00 g, 6.22 mmol)をテトラヒドロフラン(30 ml)に懸濁させ、氷浴を用いて冷却した。この溶液にトリエチルアミン(17.3 ml, 124 mmol)を加えた後、室温に昇温して1時間攪拌した。反応溶液をろ別し、濃縮してL-チロシンメチルエステル(1.81 g, 6.35 mmol)を得た。
Boc2O(1.94 g, 8.89 mmol)をジクロロメタン(20 ml)に溶解させ、氷浴を用いて冷却した。この溶液に4-(ジメチルアミノ)ピリジン(853 mg, 6.99 mmol)を溶かしたジクロロメタン(20 ml)溶液とL-チロシンメチルエステル(1.81 g, 6.35 mmol)を溶かしたジクロロメタン(20 ml)溶液を加え、室温にて1時間攪拌した。再び氷浴を用いて反応溶液を冷却し、2,3,4,6-テトラ-O-アセチル-D-グルコース(3.10 g, 8.89 mmol)を溶かしたジクロロメタン(20 ml)溶液を加え、16.5時間攪拌した。反応溶液を減圧濃縮し、残渣をシリカゲルカラムクロマトグラフィー(グラジエント;ヘキサン:酢酸エチル70:30→40:60)にて精製し、4Ac-Glc-Tyr(OBn)-OMe(3.10 g, 4.70 mmol, 収率76%,α:β比=3:2)を淡黄色シロップ状物質として得た。
1H-NMR(400 MHz, CDCl3)δ: 2.00 (s, 1.5H), 2.01 (s, 1.5H), 2.03-2.04 (m, 6H), 2.08 (s, 1.5H), 2.09 (s, 1.5H), 3.01-3.13 (m, 2H), 3.72 (s, 1.5H), 3.74 (s, 1.5H), 4.09-4.15 (m, 2H), 4.24-4.34 (m, 1H), 4.53-4.64 (m, 1H), 5.02-5.50 (m, 6H), 5.65 (d, 0.4H, J=8.4 Hz), 6.24 (d, 0.6H, J=3.6 Hz), 6.87-6.96 (m, 2H), 7.00-7.05 (m, 2H), 7.31-7.44 (m, 5H).
ESIMS(m/z): 682.2([M+Na]+), 698.2([M+K]+).
(2) 4Ac-Glc-Tyr(OBn)-OMe;N-(2,3,4,6-テトラ-O-アセチル-α/β-D-グルコピラノシルオキシカルボニル)-L-チロシンメチルエステル
4Ac-Glc-Tyr(OBn)-OMe(3.09 g, 4.69 mmol)をメタノール:酢酸エチル=1:1の混合溶液(60 ml)に溶解させ、脱気した。5%パラジウム/炭素(3.00 g, 100%(w/w))を加えた後、水素で容器内を置換し、1時間攪拌した。出発物質の残存を確認したため、再度5%パラジウム/炭素(1.50 g, 50%(w/w))を加えた後、水素で容器内を置換し、さらに2.5時間攪拌した。パラジウム/炭素を濾別し、ろ液を減圧濃縮してGlc-Tyr-OMe(2.78 g, 4.88 mmol, 収率92%,α:β比=1:1)を得た。
1H-NMR(400 MHz, CDCl3)δ:2.00 (s, 1.5H), 2.01 (s, 1.5H), 2.03 (s, 3H), 2.05 (s, 1.5H), 2.06 (s, 1.5H), 2.09 (s, 1.5H), 2.10 (s, 1.5H), 2.91-3.13 (m, 2H), 3.70-3.81 (m, 0.5H), 3.76 (s, 1.5H), 3.76 (s, 1.5H), 4.06-4.28 (m, 2.5H), 4.53-4.63 (m, 1H), 5.06-5.49 (m, 4H), 5.59 (d, 0.5H, J=8.4 Hz), 6.23 (d, 0.5H, J=3.6 Hz), 6.71-6.80 (m, 2H), 6.96-7.00 (m, 2H).
ESIMS(m/z): 592.2([M+Na]+).
(3) Glc-Tyr;N-(α/β-D-グルコピラノシルオキシカルボニル)-L-チロシン
4Ac-Glc-Tyr-OMe(1.00 g, 1.76 mmol)をメタノール(8.8 ml)に溶解させ、恒温槽を用いて-5℃に冷却した。この溶液に1N水酸化リチウム水溶液(17.6 ml, 17.6 mmol)を加え、25分間攪拌した。反応液を強酸性樹脂(Amberlite IR-120)で処理し、続いて樹脂をろ別した。ろ液を減圧濃縮し、Glc-Tyr(702 mg, 1.81 mmol, 収率quant.,α:β比=3:2)を白色粉末として得た。
1 H-NMR(400 MHz, CD3OD)δ:2.87-2.95 (m, 1H), 3.10 (dd, 1H, J=5.1, 14.0 Hz), 3.50-3.54 (dd, 3H, J=3.8, 9.9 Hz), 3.64-3.85 (m, 3H), 4.34-4.39 (m, 1H), 5.31 (d, 0.4H, J=8.1 Hz), 5.91 (d, 0.6H, J=3.8 Hz), 6.71 (d, 0.6H, J=8.5 Hz),6.72 (d, 0.4H, J=8.6 Hz), 7.07(d, 0.4H, J=8.6 Hz), 7.08(d, 0.6H, J=8.5 Hz).
ESIMS(m/z): 386.1([M-H]-), 773.2([2M-H]-).
Glc-Phe、Glc-Leu、Glc-LysおよびGlc-Gluをそれぞれ人工胃液(第15改定日本薬局方)で処理し、生成するアミノ酸量を測定した。各化合物を表1の比率で人工胃液に溶解後、37℃の湯浴中で攪拌し、HPLCにて分析した。その結果を図1~4に示す。Glc-PheおよびGlc-Leuの場合には、それぞれ、4日目には原料の4割程度、15日目には8-9割程度のアミノ酸が遊離した。Glc-Lysの場合には4日目には3割程度、8日目には5割程度のリジンが遊離した。Glc-Gluの場合には4日目には4割程度、9日目には7割程度のグルタミン酸が遊離した。

流速:Glc-Leu, Glc-Phe:1.5 ml/min, Glc-Lys, Glc-Glu:1.0 ml/min
検出:フォトダイオードアレイ検出器 測定波長210 nm
注入量:10 μL
Glc-Phe、Glc-Leu、Glc-Lys、Glc-GluおよびGlc-DOPAを、それぞれ表2の比率でリン酸緩衝液(pH 5.5)に溶解させ、表2の量のα/β-グルコシダーゼを添加後、37℃の湯浴中で攪拌した。1%リン酸水溶液にて2倍希釈後、HPLCにて分析した結果を図5~9に示す。Glc-Pheについては、酵素添加直後からフェニルアラニンが7%程度遊離していることが確認され、30分後にはGlc-Pheがほぼ消失し、対応するフェニルアラニンが遊離した。また、Glc-Leuについては1時間後にロイシンが8割程度、Glc-Lysについては1時間後にリジンが2割程度、Glc-Gluについては1時間後にグルタミン酸が4割程度、Glc-DOPAについては1時間後に3,4-ジヒドロキシフェニルアラニンが7割程度遊離した。
LeuおよびGlc-Leuを、それぞれ35℃の湯浴中で攪拌した水(内温34℃)に添加し、溶解速度を測定した。添加した試料の量および測定結果は以下に示す通りである(n=2)。Leuに比べてGlc-Leuは等重量では46倍、等モル量では17倍速く溶けた。
25℃の恒温槽中で水(1 ml)にLeuまたはGlc-Leuが溶解しなくなるまで添加し、2日間攪拌することで溶解度を測定した。HPLCにて濃度を測定した結果、Leuに比べてGlc-Leuの溶解度は63倍向上した。
Glc-DOPAおよびGlc-Tyrについても同様に、25℃の恒温槽中で水(0.5 ml)に1~1.5 g程度添加したところ、いずれも水に溶解している状態であったが、この時点で粘性が高く攪拌が困難であったため、サンプルを希釈し、HPLCにて溶解度を測定した。Glc-DOPAはDOPAに比べて640倍以上、Glc-TyrはTyrに比べて4670倍以上の溶解度であった。

注入量:10 μL
ロイシンには特有の苦味があるが、Glc-Leuには苦味のマスキング効果があるか、官能試験にて調べた。まず3名の被験者A、B、Cは、食品添加物用ロイシンを水に0.5% (5000 ppm)の濃度で溶解した溶液を、マイクロピペットにて0.1 ml量り取り、舌に滴下後、吐き出すことで、ロイシンの苦味の強度を確認した。続いて3名の被験者A、B、Cは、Glc-Leuを水に0.5% (5000 ppm)の濃度で溶解した溶液を、マイクロピペットにて0.1 ml量り取り、舌に滴下後、吐き出すことで、先に確認したロイシンの苦味の強度と比較した。結果は以下の通りとなり、いずれの被験者もロイシンで確認した苦味を感じなかった。

- Gで示される、全ての水酸基が保護も修飾もされていない糖残基の糖が単糖である、請求項1に記載の化合物またはその塩。
- Gで示される、全ての水酸基が保護も修飾もされていない糖残基の糖が、グルコース、グルコサミンまたはN-アセチルグルコサミンである、請求項1に記載の化合物またはその塩。
- 式G-O-で表される部分構造がβ-アノマー構造である、請求項1~3のいずれかに記載の化合物またはその塩。
- AAで示されるアミノ酸残基のアミノ酸がα-アミノ酸である、請求項1~4のいずれかに記載の化合物またはその塩。
- AAで示されるアミノ酸残基のアミノ酸が、バリン、ロイシンまたはイソロイシンである、請求項1~4のいずれかに記載の化合物またはその塩。
- AAで示されるアミノ酸残基のアミノ酸が、フェニルアラニン、チロシンまたは3,4-ジヒドロキシフェニルアラニンである、請求項1~4のいずれかに記載の化合物またはその塩。
- Rが水素原子である、請求項1~7のいずれかに記載の化合物またはその塩。
- アミノ酸前駆体である、請求項1~8のいずれかに記載の化合物またはその塩。
- 生体内でアミノ酸に変換される、請求項1~9のいずれかに記載の化合物またはその塩。
- 請求項1~10のいずれかに記載の化合物またはその塩を含有する、水性組成物。
- 請求項1~10のいずれかに記載の化合物またはその塩を含有する、経口剤。
- アミノ酸のアミノ基に式G-O-C(O)-(式中、Gは、全ての水酸基が保護も修飾もされていない糖残基を示す。)で表される基を導入することを含む、アミノ酸の苦味を低減する方法。
- Gで示される、全ての水酸基が保護も修飾もされていない糖残基の糖が単糖である、請求項13に記載の方法。
- Gで示される、全ての水酸基が保護も修飾もされていない糖残基の糖が、グルコース、グルコサミンまたはN-アセチルグルコサミンである、請求項13に記載の方法。
- 式G-O-で表される部分構造がβ-アノマー構造である、請求項13~15のいずれかに記載の方法。
