Abstract
본 발명은 펩타이드가 결합된 안정한 아스코르빈산 유도체, 그의 제조방법 및 그를 유효성분으로 포함하는 화장료 조성물을 제공한다. 본 발명의 펩타이드가 결합된 아스코르빈산 유도체는 멜라닌 생성 억제를 통한 미백 효과 및 콜라겐 생성 활성화를 통한 주름개선 효과를 동시에 나타내는 이중복합 기능을 가지며, 화장료 조성물에 사용될 수 있다.
Description
본 발명은 우수한 주름개선 및 미백의 이중복합 효능을 갖는 펩타이드가 결합된 안정한 아스코르빈산 유도체, 그의 제조방법 및 그를 유효성분으로 포함하는 화장료 조성물에 관한 것이다.
일반적으로, 아스코르빈산은 환원제, 콜라겐합성 촉진제, 항산화제로 작용하고 소장에서 철분의 흡수를 도우며 카르니틴의 생합성 및 면역기능에 관여한다. 체내에 아스코르빈산이 부족하면 괴혈병, 콜라겐 합성의 이상으로 결체조직의 이상, 뼈의 통증, 골절, 설사 등이 일어나며, 과량의 아스코르빈산이 있을 때에는 메스꺼움, 복통, 설사 등의 위장관 장애가 일어날 수 있는 것으로 알려져 있다. 특히, 아스코르빈산의 뛰어난 항산화 효과는 피부 내 멜라닌 생성을 억제하여 기미, 주근깨와 같은 비정상적인 색소 침착을 방지하는 것으로 알려져 있기 때문에 아스코르빈산은 피부외용제 재료로서 각광받고 있다.
그러나, 상기한 바와 같이 아스코르빈산은 다양한 효과를 나타내지만, 공기와 접촉시켜 열을 가하면 대부분 파괴되고 알칼리에 불안정하며 수용액 상태에서는 쉽게 산화되어 그러한 효과를 상실하게 된다는 문제점이 있다. 따라서 이러한 아스코르빈산의 불안정성을 개선하기 위하여 다양한 아스코르빈산 유도체들이 개발되어 왔다.
기 개발된 아스코르빈산 유도체들의 형태를 살펴보면, 첫째로 인산화된 아스코르빈산 또는 인산화된 아스코르빈산 금속염 형태의 유도체가 있다. 이러한 유도체는 다른 유도체에 비해 인체가 사용할 수 있는 형태인 L-아스코르빈산 형태로의 전환은 쉽지만 음의 전하를 가지므로 피부 흡수가 어렵다는 단점이 있다. 둘째로 지방산이 결합된 형태의 유도체로 아스코르빌 팔미테이트, 아스코르빌 라우레이트, 아스코르빌 스테아레이트 등 다양한 유도체들이 알려져 있는데 이 중에서 아스코르빌-6-팔미테이트가 가장 널리 사용되고 있다 [참조: 미국특허 제5,409,693호]. 하지만 이들 유도체 화합물의 경우 피부에 흡수는 되지만 L-아스코르빈산 형태로의 전환이 어렵다는 단점이 있다. 셋째로 콜라겐 생성 펩타이드가 결합된 형태의 아스코르빈산 유도체로 아스코르빈산의 5번 탄소 또는 6번 탄소의 하이드록시기에 에스테르 결합으로 숙시노일기가 연결되고 아미드 결합으로 콜라겐생성 펩타이드가 연결된 유도체가 알려져 있다 [참조: 대한민국 등록특허 KR 10-0459679호]. 그러나 이들 유도체의 경우 아스코르빈산에 비해 우수한 효능을 보이지만 안정성이 떨어진다는 단점이 있다.
위에서 설명한 바와 같이 현재까지 만들어진 거의 모든 아스코르빈산 유도체들은 순수 아스코르빈산에 비해 효능이나 안정성 측면에서 개선되지 않았으며, 펩타이드가 연결된 아스코르빈산 유도체들의 경우에도 주로 아스코르빈산의 미백 효능만을 목표로 하는 경우가 대부분이었므로 화장품 용도로 널리 사용되기에 한계가 있다.
이에 본 발명자들은 종래 아스코르빈산 유도체들의 문제점을 해결함과 동시에 주름개선 및 미백의 이중복합 효능을 가지는 아스코르빈산 유도체를 개발하기 위해 예의 연구를 수행한 결과, 아스코르빈산 아미노프로판올 인산 디에스테르 화합물에 2 내지 5개의 배열을 갖는 펩타이드 화합물을 결합시키면 주름개선 및 미백의 이중복합 효능을 갖는 안정한 아스코르빈산 유도체를 얻을 수 있음을 확인하고 본 발명을 완성하게 되었다.
따라서, 본 발명의 목적은 하기 화학식 I의 펩타이드가 결합된 아스코르빈산 유도체 또는 그의 화장료로 허용되는 염을 제공하는 것이다.
본 발명의 또 다른 목적은 하기 화학식 I의 펩타이드가 결합된 아스코르빈산 유도체 또는 그의 화장료로 허용되는 염의 제조방법을 제공하는 것이다.
본 발명의 또 다른 목적은 하기 화학식 I의 펩타이드가 결합된 아스코르빈산 유도체 또는 그의 화장료로 허용되는 염을 유효성분으로 포함하는 화장료 조성물 및 그의 피부 주름개선 및 미백 용도를 제공하는 것이다.

본 발명에서 '화장료로 허용되는 염'은 무독성 무기산염 및 유기산염 모두를 포함하며, 예를 들어 염산염, 황산염, 질산염, 인산염, 아세테이트산염, 트리플루오로아세트산염, 벤젠설포네이트산염, 시트레이트산염 등을 포함하나 이에 한정되는 것은 아니다.
또한, 본 발명은 상기 화학식 I의 펩타이드가 결합된 아스코르빈산 유도체의 제조방법에 관한 것으로, 본 발명의 제조방법은 하기 화학식 II의 아스코르빈산 아미노프로판올 인산 디에스테르 화합물과 하기 화학식 Ⅳ의 2 내지 5개의 배열을 갖는 펩타이드 화합물을 축합반응시킨 다음, 보호기가 존재하는 경우 탈보호화 반응시키는 단계를 포함한다.

상기 화학식 IV의 화합물의 카르복실산 보호기로는 메틸, 에틸, t-부틸, 2,2,2-트리클로로에틸 등을 사용할 수 있으나 이에 한정되는 것은 아니다. 또한 화학식 IV의 화합물의 카르복실산 보호기의 탈보호화 반응은 수산화 리튬 또는 트리플루오로아세트산을 사용하여 수행할 수 있으나 이에 한정되는 것은 아니다.
상기 축합반응은 축합제의 존재 하에 수행될 수 있으며, 축합제로는 디시클로헥실카르보디이미드(DCC), 디페닐포스포릴아지드(DPPA), 디에틸 시아노포스포네이트(DEPC), 벤조트리아졸-1-일-옥시-트리스(디메틸아미노)포스포늄헥사플루오로포스페이트(BOP 시약) 등을 사용할 수 있으나 이에 한정되는 것은 아니다. 상기 축합반응시, 필요하다면 트리에틸아민 또는 디이소프로필에틸아민(DIPEA)과 같은 유기 염기를 축합제와 함께 사용할 수 있으며, 필요하다면 4-(디메틸아미노)피리딘(DMAP), N-히드록시 숙신이미드(HOSu)와 같은 활성화 물질을 함께 사용할 수 있다.
상기 축합반응에서 반응용매로는 클로로포름 및 디클로로메탄과 같은 할로겐화 지방족 탄화수소, 에틸아세테이트, 테트라히드로퓨란(THF), 디메틸포름아미드(DMF), 아세토니트릴, 1,4-디옥산, 메탄올 등을 사용할 수 있으며, 반응온도는 -10 ℃ 내지 50 ℃, 바람직하게는 0 ℃ 내지 25 ℃이다.
상기 화학식 II의 화합물은 공지된 방법[참조: Bull. Korean Chem. Soc., 2003, Vol. 24, No. 8, pp 1169-1171]에 따라 합성하거나, 또는 시판 중인 제품을 구입하여 사용할 수 있다.
상기 화학식 IV의 화합물의 펩타이드(R2가 존재하지 않는 경우) 또는 지방산-펩타이드(R2가 팔미틸, 라우릴 또는 스테아릴인 경우)는 고체상 합성법을 이용하여 펩타이드를 합성하거나 펩타이드 합성 후 지방산과의 중합반응을 이용하여 지방산-펩타이드를 합성하여 사용할 수 있으며, 또는 시판 중인 제품을 구입하여 사용할 수 있다.
이하, 본 발명에 따른 상기 화학식 I의 펩타이드가 결합된 아스코르빈산 유도체의 제조방법을 하기 반응식 I을 참조하여 상세히 설명한다. 하기 반응식 I에 기재된 방법은 대표적으로 사용된 방법을 예시한 것일 뿐, 단위조작의 순서, 반응시약, 반응조건 등은 경우에 따라 얼마든지 변경될 수 있다.

본 발명의 제조방법은,
(i) 상기 화학식 III의 화합물에 t-부톡시카르보닐(t-Boc)과 같은 아미노 보호기 및 t-부틸과 같은 카르복실산 보호기를 가하고 반응하여 아미노기 및 카르복시기가 보호화된 화학식 IV의 화합물을 수득하는 단계 또는 상기 화학식 III의 화합물의 C-말단 카르복시기에 수산화리튬과 같은 염기를 사용하여 탈보호화하는 과정을 통해 화학식 IV의 화합물을 수득하는 단계; 및
(ii) 상기 화학식 IV의 화합물에 상기 화학식 II의 화합물(3-아미노프로필 (5-((S)-1,2-디히드록시에틸)-4-히드록시-2-옥소-2,5-디히드로퓨란-3-일)히드로겐 포스페이트)을 디시클로헥실카르보디이미드(DCC)와 같은 축합제의 존재하에 축합반응시키고 염산 또는 트리플루오로아세트산으로 아미노산 잔기 곁가지의 아미노기 및 카르복시기를 탈보호화 반응시키는 과정을 통해 화학식 I의 화합물을 수득하는 단계를 포함한다.
상기 화학식 III의 화합물이 상기 제조방법 중 (i)의 보호화 과정 또는 탈보호화 과정이 불필요한 경우에는 단계 (i)를 거치지 않고 단계 (ii)를 수행하여 화학식 I의 화합물을 합성할 수 있다.
상기 방법에 따라 제조된 본 발명의 펩타이드가 결합된 아스코르빈산 유도체는 안정할 뿐만 아니라, 우수한 주름개선 및 미백의 이중복합 효능을 나타낸다(시험예 1, 2 및 3 참조).
또한, 본 발명은 상기 화학식 I의 펩타이드가 결합된 아스코르빈산 유도체 또는 그의 화장료로 허용되는 염 및 화장료로 허용 가능한 기제를 포함하는 화장료 조성물에 관한 것으로, 본 발명의 화장료 조성물은 특히 피부 주름개선 및 미백의 이중복합 효능을 갖는다.
본 발명의 화장료 조성물은 화장료 조성물 총 중량에 대하여 0.0001 내지 2 중량%, 바람직하게는 0.01 내지 0.20 중량%의 화학식 I의 화합물을 유효성분으로 포함한다.
본 발명의 화장료 조성물은 그 제형이 특별히 한정되는 것은 아니며, 제조하고자 하는 제형에 따라 발명이 속하는 기술 분야에서 통상적으로 사용되는 화장료 조성물 배합 성분을 포함할 수 있다. 본 발명의 화장료 조성물은 화장수, 유액, 영양크림, 팩, 미용액, 에센스 등의 제형으로 제조할 수 있으며, 제조하고자 하는 제형에 따라 추가로 유분, 물, 계면활성제, 보습제, 저급알콜, 증점제, 킬레이트제, 색소, 방부제, 향료 등을 선택하여 배합 첨가할 수 있다. 또한 멜라닌 형성의 주요 요인이 자외선임을 고려할 때, 본 발명의 화장료 조성물은 자외선 차단제, 광산란제 등을 포함할 수 있으며, 그 제형 및 첨가성분이 상기 내용에 한정되는 것은 아니다.
본 발명의 펩타이드가 결합된 아스코르빈산 유도체는 신규 합성 화합물로, 멜라닌 생성 억제를 통한 미백 효과 및 콜라겐 생성 활성화를 통한 주름개선 효과를 동시에 나타내는 이중복합 기능을 가지므로 화장료 조성물에 유용하게 사용될 수 있다.
이하, 제조예 및 실시예에 의해 본 발명을 보다 구체적으로 설명하고자 한다. 이들 제조예 및 실시예는 오직 본 발명을 설명하기 위한 것으로, 본 발명의 범위가 이들 제조예 및 실시예에 국한되지 않는다는 것은 당업자에게 있어서 자명하다.

제조예 1-1에서 수득한 아미노기가 보호화된 화합물 Ⅳ-1(31.2 g, 38.4 mmol)을 디메틸포름아미드 250 mL에 녹이고 N,N'-디사이클로헥실카보디이미드(DCC) (9.5 g, 46.1 mmol)와 N-히드록시숙신이미드(5.3 g, 46.1 mmol)를 가하여 실온에서 3시간 반응시킨다. 반응 후 부산물로 생성된 N,N'-디사이클로헥실우레아를 감압여과로 제거하고 얻어진 여액에 화합물 II의 3-아미노프로필(5-((S)-1,2-디히드록시에틸)-4-히드록시-2-옥소-2,5-디히드로퓨란-3-일) 히드로겐 포스페이트(Vitagen, 13.2 g, 42.2 mmol)와 디이소프로필에틸아민(13.5 mL, 77 mmol)을 가하고 실온에서 20시간 동안 교반시켰다. 반응 완결 후 용매를 감압농축하여 얻어진 잔사에 MeOH, 디클로로메탄, 이소프로필에테르를 순차적으로 가하여 재결정하고 여과하여 화합물 Ⅰ- 1 의 아미노기가 보호화된 화합물(47 g, 112 %)을 얻었다.

수득한 화합물 Ⅰ- 1 의 아미노기가 보호화된 화합물(47 g, 37.9 mmol)을 1,4-디옥산 160 mL에 용해시키고 4 M의 염산 1,4-디옥산 용액 110 mL(440 mmol)를 가하여 실온에서 15시간 동안 교반하였다. 반응 완결 후 용매를 감압농축하여 얻어진 잔사에 아세토나이트릴 600 mL를 가하여 생성된 고체를 여과하여 표제 화합물(33.0 g, 89%)을 수득하였다.

제조예 1-2에서 수득한 아미노기 및 카르복시기가 보호화된 화합물 Ⅳ-2(2.46 g, 3.5 mmol)를 디메틸포름아미드 20 mL에 녹이고 N,N'-디사이클로헥실카보디이미드(DCC) (0.72 g, 3.5 mmol)와 N-히드록시숙신이미드(0.4 g, 3.5 mmol)를 가하여 실온에서 1시간 반응시킨다. 반응 후 부산물로 생성된 N,N'-디사이클로헥실우레아를 감압여과로 제거하고 얻어진 여액에 화합물 II의 3-아미노프로필 (5-((S)-1,2-디히드록시에틸)-4-히드록시-2-옥소-2,5-디히드로퓨란-3-일) 히드로겐 포스페이트(Vitagen, 1.32 g, 4.2 mmol)과 디이소프로필에틸아민 (1.22 mL, 7.0 mmol)을 가하고 실온에서 2시간 동안 교반시켰다. 반응 완결 후 용매를 감압농축하여 얻어진 잔사에 메탄올, 디클로로메탄, 이소프로필에테르를 순차적으로 가하여 재결정하고 여과하여 화합물 Ⅰ-2의 아미노기 및 카르복시기가 보호화된 화합물 (11S,17S)-t-부틸 6,11-비스((t-부톡시카르보닐)아미노)-17-((3-((((5-((S)-1,2-디히드록시에틸)-4-히드록시-2-옥소-2,5-디히드로퓨란-3-일)옥시)(히드록시)포스포릴)옥시)프로필)카바모일)-2,2-디메틸-4,12,15-트리옥소-3-옥사-5,7,13,16-테트라아자노나데-6-켄-19-오에이트(3.28 g, 94%)를 얻었다.

화합물 Ⅰ-2 의 아미노기 및 카르복시기가 보호화된 화합물(3.28 g, 3.29 mmol)을 1,4-디옥산 35 mL 에 용해시키고 4 M의 염산 1,4-디옥산 용액 8.3 mL(33 mmol)를 가하여 실온에서 15시간 동안 교반하였다. 반응 완결 후 용매를 감압농축하여 얻어진 잔사에 아세토니트릴, 디클로로메탄, 이소프로필에테르를 순차적으로 가하여 재결정하고 여과하여 표제 화합 (2.25 g, 91 %)을 얻었다.

시험예
1: 멜라닌 억제 효능 분석
멜라노마 세포주인 B16F1 세포(ATCC CRL-6323)를 10 % 우태아 혈청과 100 U/㎖ 페니실린, 100 ㎍/㎖ 스트렙토마이신이 보충된 DMEM 배지에 첨가하여 37 ℃, 5 % CO2조건에서 배양하였다. 배양 후 배지를 제거하고 DMEM 배지(Invitrogen-Gibco-BRL, 세포/웰), 각 시험 화합물의 최종 농도가 0 ㎍/㎖, 10 ㎍/㎖, 20 ㎍/㎖, 40 ㎍/㎖가 되도록 적정량의 시료액을 처리하였다. 72시간 후에 배지를 제거한 세포를 PBS (phosphated buffer saline)로 세척한 후 세포를 회수하여 60 ℃에서 건조하였다. 다음으로 멜라닌 추출용액(1 N NaOH + 10 % DMSO)을 첨가하여 80 ℃에서 세포를 용해시킨 후 합성 멜라닌을 표준품으로 하여 멜라닌 양을 정량하였고, 단백질량을 정량하여 단위 단백질당 멜라닌 양을 계산한 후 시료 무처리군(control)과 비교하여 % control 값을 산출하였다.
그 결과를 하기 표 1에 나타내었다.
시험예
2: 콜라겐 활성 효능 분석
사람의 정상 섬유아세포(Human dermal fibroblast neonatal, Cascade Co.)를 10 %의 우태아 혈청이 보충된 DMEM 배지를 이용하여 24-웰 마이크로 플레이트에 접종하고(5×104세포/웰), 37 ℃, 5% CO2 조건에서 24시간 동안 배양하였다. 혈청이 들어 있지 않은 DMEM 배지로 교체하여 24시간 동안 배양한 후, 각 시험 화합물의 최종 농도가 1 ㎍/㎖, 5 ㎍/㎖가 되도록 적정량의 시료액을 처리하였다. 48시간 배양 후 세포배양액을 채취하여 후 타카라사(Takara Shuzo Co., Ltd. 일본국)의 콜라겐 측정키트를 사용하여 프로콜라겐 양을 측정하였다. 먼저 1차 콜라겐 항체가 균일하게 도포된 96-웰 마이크로 플레이트에 채취한 세포배양액을 넣고 37 ℃에서 3시간 동안 항원-항체 반응이 일어나도록 하였다. 웰 내의 세포배양액을 제거하고 PBS로 4회 세척하였다. 각 웰에 발색유발물질을 넣고 상온에서 15분간 반응시킨 후, 1 N 황산용액을 첨가하여 반응을 중지시켰다. 분광광도계로 파장 450nm에서의 흡광도를 측정하였다. 표준용액을 이용하여 표준곡선을 작성하고, 상기의 방법으로 얻은 흡광도를 표준곡선에 대입하여 각 시험 화합물이 첨가된 세포배양액의 프로콜라겐 생성량을 산출하였다.
그 결과를 하기 표 2에 나타내었다.
시험예
3: 수분 및 열에 대한 안정성 시험
화장품 조성물에 사용되는 활성성분의 수분 및 열에 대한 안정성은 조성물의 제조공정은 물론 조성물의 장기간 보관에 있어서 중요한 요소이므로, 상기 실시예 1 및 실시예 2에서 각각 제조된 화합물 I-1 및 I-2 에 대한 수분 및 열에 대한 안정성을 측정하였다. 구체적으로, 상기한 화합물 I-1 및 I-2 각각을 40 ℃의 온도 및 75 %의 상대습도인 가혹조건에서 밀봉상태로 보관하면서 각각 0일, 3일, 7일, 14일 및 28일이 지난 후의 시료에 대해서 초기 활성성분 값에 대한 잔사율(%)을 고성능액체크로마토그래피(HPLC)로 분석하였다.
그 결과를 하기 표 3에 나타내었다.
상기 표 3에서 보는 바와 같이, 화합물 I-1 및 I-2 각각을 28 일 동안 가혹조건에서 실험한 결과, 시판 중인 LAAP(Vitagen)와 대등하게 우수한 안정성을 보여주고 있음을 확인할 수 있었다.
시험예 4: DNA methyltransferase(DNMT) 활성 억제 효과
화합물 I-1의 프로콜라겐 합성 촉진 효과에 대한 기전을 확인하기 위해, Hela cell에서 분리한 Nuclear extract와 DNA methyltransferase activity/inhibitor screening assay kit를 이용하여 DNMT 활성 억제효과를 확인하였다. Cystosine-rich DNA가 코팅된 strip에 DNMT를 포함하는 Hela cell nuclear extract, 화합물 I-1과 Adomet를 첨가하였다. 양성대조군으로 5-Aza-2'-Deoxycytidine (5-azadC)를 사용하였다. Methylation된 DNA와 결합하는 capture antibody와 detection antibody를 순차적으로 결합시킨 후 developing solution을 첨가하여 발색 반응 후 stop solution를 첨가하여 반응을 종료하였다. 450nm에서 흡광도를 측정하여 하기 수학식 1로 DNMT 활성 억제 효과를 계산하여, 그 결과를 하기 표 4에 나타내었다
상기 표 4에서 보는 바와 같이, 화합물 I-1은 5 ug/ml 이상의 농도에서 DNA methylation을 50% 이상 억제하였으며, 1~10 ug/ml 범위에서 농도 의존적으로 DNA methylation을 억제하는 효과를 나타내었다. 따라서, 화합물 I-1은 콜라겐 유전자의 methylation을 억제하여 프로콜라겐을 증가시키는 효능을 갖는 물질임을 알 수 있다.

상기의 시험예를 근거로 하여, 하기 표에 기재된 조성대로 실시예에 의한 펩타이드가 결합된 아스코르빈산 유도체를 포함하는 화장료 조성물을 제조하였다. 하기 제형예는 본 발명을 보다 구체적으로 설명하기 위한 예시로, 본 발명이 하기 예시에만 한정되는 것은 아니다.

하기 표 5에 기재된 조성대로 실시예에 의한 펩타이드가 결합된 아스코르빈산 유도체를 포함한 스킨을 제조하였다.
<제조방법>
(1) 수상과 가용화상 각각 균일하게 혼합 및 용해시켰다.
(2) 수상에 가용화상을 넣고 혼합시켜 수중유형 유화를 얻었다.
(3) 이후 펩타이드가 결합된 아스코르빈산 유도체를 용해시킨 상으로 가용화 한 첨가상 I을 투입하여 혼합시킨 후 첨가상 II를 혼합하여 완료하였다.

2. 로션의 제조
하기 표 6에 기재된 조성대로 실시예에 의한 펩타이드가 결합된 아스코르빈산 유도체를 포함한 로션을 제조하였다.
<제조방법>
(1) 수상과 유상을 가온하여 각각 균일하게 혼합 및 용해시켰다.
(2) 75 ℃에서 수상에 유상을 넣고 혼합시켜 가용화시켰다.
(3) 이후 50 ℃까지 냉각한 후 펩타이드가 결합된 아스코르빈산 유도체를 용해시킨 상으로 가용화 한 첨가상 I을 투입하고 혼합시킨 후 첨가상 II를 혼합하였다.

3. 크림의 제조
하기 표 7에 기재된 조성대로 실시예에 의한 펩타이드가 결합된 아스코르빈산 유도체를 포함한 크림을 제조하였다.
<제조방법>
(1) 수상과 유상을 가온하여 각각 균일하게 혼합 및 용해시켰다.
(2) 75 ℃에서 수상에 유상을 넣고 혼합시켜 가용화시켰다.
(3) 이후 50 ℃까지 냉각한 후 펩타이드가 결합된 아스코르빈산 유도체를 용해시킨 상으로 가용화 한 첨가상 I을 투입하고 혼합시킨 후 첨가상 II를 혼합하였다.

- 제 2항에 있어서,
n이 3 내지 5의 정수인 것을 특징으로 하는 펩타이드가 결합된 아스코르빈산 유도체 또는 그의 화장료로 허용되는 염.
- 제 4항에 있어서,
하기 화합물 중에서 선택되는 것을 특징으로 하는 펩타이드가 결합된 아스코르빈산 유도체 또는 그의 화장료로 허용되는 염:
(6S,9S,12S)-6,12-비스(4-아미노부틸)-9-이소프로필-5,8,11,14-테트라옥소-4,7,10,13-테트라아자노나코실(5-((S)-1,2-디히드록시에틸)-4-히드록시-2-옥소-2,5-디히드로푸란-3-일) 히드로겐 포스페이트(Ⅰ-1); 및
(3S)-3-(2-((S)-2-아미노-5-((디아미노메틸렌)아미노)펜탄아미도)아세트아미도)-4-((3-((((5-((S)-1,2-디히드록시에틸)-4-히드록시-2-옥소-2,5-디히드로퓨란-3-일)옥시)(히드록시)포스포릴)옥시)프로필)아미노)-4-옥소부탄산(Ⅰ-2).
- 하기 화학식 II의 화합물과 하기 화학식 Ⅳ의 화합물을 축합반응시킨 다음, 보호기가 존재하는 경우 탈보호화 반응시키는 단계를 포함하는 하기 화학식 I의 펩타이드가 결합된 아스코르빈산 유도체의 제조방법:

는 천연 또는 비-천연 아미노산 중에서 선택되는 동일하거나 상이한 아미노산 잔기가 아미드 결합된 펩타이드이고,
R1은 상기 아미노산 잔기의 곁가지이고,
R1´은 R1이거나 또는 아미노기 및 카르복시기가 보호화된 R1이고,
X는 수소 또는 카르보닐(C=O)이며,
R2는 X가 수소일 때 존재하지 않으며, X가 카르보닐일 때 팔미틸, 라우릴 또는 스테아릴이고,
n은 2 내지 5의 정수이다.
- 제 1항 내지 제 5항 중 어느 한 항에 따른 펩타이드가 결합된 아스코르빈산 유도체 또는 그의 화장료로 허용되는 염 및 화장료로 허용 가능한 기제를 포함하는 화장료 조성물.
- 제 7항에 있어서, 상기 화장료 조성물이 피부 주름개선 및 미백 용도를 갖는 것을 특징으로 하는 화장료 조성물.
- 제 7항에 있어서, 상기 펩타이드가 결합된 아스코르빈산 유도체 또는 그의 화장료로 허용되는 염이 화장료 조성물 총 중량에 대하여 0.0001 내지 2 중량%로 포함되는 것을 특징으로 하는 화장료 조성물.
