Abstract
1. Мультиспецифичное антитело, включающее антигенсвязывающий домен, который специфично связывается с EGFR и HER3, где токсичность антитела меньше, чем дерматологическая токсичность антагониста EGFR.2. Антитело по п. 1 или 2, где токсичность:(а) отсрочена или понижена в плане тяжести в клеточной модели in vitro и не-человеческой животной модели in vivo; или(b) измеряется путем введения указанного антитела первому яванскому макаку и введения эквивалентного количества антагониста EGFR второму яванскому макаку и сравнения образования повреждений кожи у первого яванского макака и у второго яванского макака,где (i) задержка образования повреждений кожи у первого яванского макака указывает на то, что указанное антитело имеет меньшую дерматологическую токсичность, чем указанный антагонист EGFR, или (ii) понижение тяжести повреждений кожи у первого яванского макака указывает на то, что указанное антитело имеет меньшую дерматологическую токсичность, чем указанный антагонист EGFR.4. Claims (33)
1. Мультиспецифичное антитело, включающее антигенсвязывающий домен, который специфично связывается с EGFR и HER3, где токсичность антитела меньше, чем дерматологическая токсичность антагониста EGFR.
2. Антитело по п. 1, которое представляет собой биспецифическое антитело.
3. Антитело по п. 1 или 2, где токсичность:
(а) отсрочена или понижена в плане тяжести в клеточной модели in vitro и не-человеческой животной модели in vivo; или
(b) измеряется путем введения указанного антитела первому яванскому макаку и введения эквивалентного количества антагониста EGFR второму яванскому макаку и сравнения образования повреждений кожи у первого яванского макака и у второго яванского макака,
где (i) задержка образования повреждений кожи у первого яванского макака указывает на то, что указанное антитело имеет меньшую дерматологическую токсичность, чем указанный антагонист EGFR, или (ii) понижение тяжести повреждений кожи у первого яванского макака указывает на то, что указанное антитело имеет меньшую дерматологическую токсичность, чем указанный антагонист EGFR.
4. Антитело по п. 1, где антагонист EGFR представляет собой цетуксимаб.
5. Антитело по п. 1, где антитело ингибирует биологическую активность, по меньшей мере, одного из EGFR и HER3.
6. Антитело по п. 1, где антитело ингибирует биологическую
активность как EGFR, так и HER3.
7. Антитело по п. 1, где антитело ингибирует связывание EGF с EGFR.
8. Антитело по п. 1, где антитело ингибирует индуцированное TGF-α фосфорилирование EGFR.
9. Антитело по п. 1, где антитело ингибирует связывание херегулина с HER3.
10. Антитело по п. 1, где антитело ингибирует индуцированное херегулином фосфорилирование HER3.
11. Антитело по п. 1, где антитело ингибирует индуцированное херегулином фосфорилирование HER3 и индуцированное TGF-α фосфорилирование EGFR.
12. Антитело по п. 1, где антитело ингибирует рост опухолевой клетки.
13. Антитело по п. 1, где антитело связывается с доменом III EGFR.
14. Антитело по п. 1, где антитело связывается с доменом III HER3.
15. Антитело по п. 1, где антитело связывается с доменом III EGFR и доменом III HER3.
16. Антитело по п. 1, где антитело связывается с EGFR и HER3 с Kd менее чем 10-6 M.
17. Антитело по п. 1, где антитело связывается с EGFR с Kd менее чем 10-6 M и связывается с HER3 с Kd менее чем 10-7 M.
18. Антитело по п. 1, где антитело включает
(a) HVR-H1, содержащий аминокислотную последовательность LSGDWIH (SEQ ID NO: 48);
(b) HVR-H2, содержащий аминокислотную последовательность VGEISAAGGYTD (SEQ ID NO: 51); и
(c) HVR-H3, содержащий аминокислотную последовательность ARESRVSFEAAMDY (SEQ ID NO: 53); и
(d) HVR-L1, содержащий аминокислотную последовательность NIATDVA (SEQ ID NO: 55);
(e) HVR-L2, содержащий аминокислотную последовательность SASF (SEQ ID NO: 56); и
(f) HVR-L3, содержащий аминокислотную последовательность SEPEPYT (SEQ ID NO: 57).
19. Антитело по п. 18, содержащее
(a) вариабельный домен тяжелой цепи, характеризующийся, по меньшей мере, 95% идентичности последовательности с аминокислотной последовательностью SEQ ID NO: 30;
(b) вариабельный домен легкой цепи, характеризующийся, по меньшей мере, 95% идентичности последовательности с аминокислотной последовательностью SEQ ID NO: 29; или
(c) вариабельный домен тяжелой цепи, как в (a), и вариабельный домен легкой цепи, как в (b).
20. Антитело по п. 18, содержащее вариабельный домен тяжелой цепи из SEQ ID NO: 30.
21. Антитело по п. 18, содержащее последовательность вариабельного домена легкой цепи SEQ ID NO: 29.
22. Антитело по п. 1, где антитело представляет собой полноразмерное антитело IgG1.
23. Антитело по п. 22, где антитело ингибирует активность ADCC.
24. Антитело по п. 1 для применения в качестве лекарственного средства.
25. Выделенная нуклеиновая кислота, кодирующая антитело по любому из пп. 1-23.
26. Клетка-хозяин, содержащая нуклеиновую кислоту по п. 25.
27. Способ получения антитела, включающий культивирование клетки-хозяина по п. 26 таким образом, что производится антитело.
