Description
Oppfinnelsen vedrører en fremgangsmåte ved fremstillingen av et 2-(6-substituert-l,3-dioksan-4-yl)eddiksyrederivat med formel 1
hvor X står for en utgående gruppe og Ri, R2 og R3 hver uavhengig står for en alkylgruppe med 1-3 karbonatomer, startende fra en forbindelse med formel 2 hvor X er som definert over, hvor anvendelse gjøres av et passende acetaliseringsmiddel, i nærvær av en syrekatalysator. Oppfinnelsen vedrører også de nye forbindelser med formel 1 og 2, samt salter og syrer med formel 3 som kan erholdes derfra
hvor Ri og R2 har de ovennevnte betydninger og hvor Y står for et alkali(jord)metall eller en substituert eller usubstituert ammoniumgruppe.
Søker har overraskende funnet at 2-(6-substituert-l,3-dioksan-4-yl)eddiksyrederivatet av formel (C) kan erholdes selektivt og i et høyt utbytte fra den tilsvarende forbindelse med formel (2), hvor det er mulig å fremstille disse produkter, som er relativt lite stabile, under milde betin-gelser. Dette er desto mer interessant siden dette gir en enkel rute via det tilsvarende salt, den tilsvarende t-butylester og den 2-hydroksymetylsubstituerte forbindelse som intermediater i fremstillingen av HMG-CoA-reduktaseinhibi-torer. Eventuelt går konversjonen (avhengig av de valgte reaksjonsbetingelser) via et mellomliggende salt eller ester, med ringen i forbindelsen i henhold til formel (2) som åpnes.
En ytterligere fordel ved fremgangsmåten i henhold til oppfinnelsen er at både startforbindelsene med formel (2) og produktene med formel (3) er funnet å være krystallinske forbindelser. Dette er fordelaktig for å erholde produkter med en (kjemisk eller stereokjemisk) høy renhet. Dette er viktig spesielt i lys av den tiltenkte farmasøytiske anvendelse. For den tiltenkte anvendelse er spesielt (4R,6S)-2-(6-substituert-l,3-dioksan-4-yl)eddiksyrederivatet viktig. Det kan fremstilles fra den tilsvarende 6-substituerte-2,4,6-trideoksy-D-erytroheksose. Oppfinnelsen vedrører derfor også startforbindelsene med formel 1, spesielt hvor X = Cl. Partikler av slike forbindelser kan ha mer enn 90 vekt% med et lengde/diameterforhold mellom 1:1,5 og 1:6, fortrinnsvis mellom 1:2 og 1:4,4 og en partikkellengde mellom 0,05 og 2 mm, spesielt mellom 0,1 og 1 mm. Forbindelsen med formel II gir klare krystallinske partikler med et skarpt smeltepunkt på 73-74 °C. Produktene med formel 3 avledet fra (4R,6S)-2-(6-substituert-l,3-dioksan-4-yl)ed-diksyrederivatet med formel 1 kan i henhold til oppfinnelsen fremstilles med et enantiomert overskudd (e.e.) på mer enn 95%, spesielt mer enn 99,5%, og med et diastereomert overskudd (d.e.) på mer enn 90%, spesielt mer enn 99,5%. Eksempler på passende utgående grupper X som kan anvendes i fremgangsmåten i henhold til oppfinnelsen er halogener, spesielt Cl, Br eller I; tosylatgrupper; mesylatgrupper; acyloksygrupper, spesielt acetoksy- og benzyloksygrupper; en aryloksy-, spesielt benzyloksy-, eller en nitrosubstituert benzensulfonylgruppe. Av praktiske grunner velges fortrinnsvis Cl som utgående gruppe.
Gruppene Ri, R2 og R3 står hver separat for en alkylgruppe med 1-3 karbonatomer, fortrinnsvis metyl eller etyl. I praksis er Ri=R2=R3=metyl mest foretrukket.
Eksempler på egnede acetaliseringsmidler som kan anvendes i fremgangsmåten i henhold til oppfinnelsen er dialkoksypro-panforbindelser, hvor alkoksygruppene hver fortrinnsvis har 1-3 karonatomer, for eksempel 2,2-dimetoksypropan eller 2,2-dietoksypropan; alkoksypropen, hvor alkoskygruppen fortrinnsvis har 1-3 karbonatomer, for eksempel 2-metoksypro-pen eller 2-etoksypropen. Mest foretrukket er 2,2-dimetoksypropan. Denne kan eventuelt dannes in situ fra aceton og metanol, fortrinnsvis med fjerning av vann.
Som syrekatalysator kan det gjøres anvendelse av syrekata-lysatorene kjent for acetaliseringsreaksjoner, fortrinnsvis ikke-nukleofile sterke syrer, for eksempel sulfonsyrer, spesielt p-toluensulfonsyre, metansulfonsyre av kamfersul-fonsyre; uorganiske syrer med et ikke-nukleofilt anion, for eksempel svovelsyre, fosforsyre; sure ionebyttere, for eksempel DOWEX; eller faste syrer, for eksempel de såkalte heteropolysyrer.
Acetaliseringen kan utføres uten å anvende et separat løsningsmiddel; om ønsket kan reaksjonen også utføres i et organisk løsningsmiddel. Eksempler på egnede organiske løsningsmidler er ketoner, spesielt aceton, hydrokarboner, spesielt aromatiske hydrokarboner, for eksempel toluen, klorerte hydrokarboner, for eksempel metylenklorid. Temperaturen ved hvilken acetaliseringsreaksjonen utføres ligger fortrinnsvis mellom -20 °C og 60 °C, spesielt mellom 0 °C og 30 °C. Acetaliseringsreaksjonen utføres fortrinnsvis under en inert atmosfære.
Det molare forhold av acetaliseringsmiddel til startforbindelse med formel (2) ligger fortrinnsvis mellom 1:1 og 20:1, spesielt mellom 3:1 og 5:1. Ved anvendelse av et organisk løsningsmiddel er det molare forhold spesielt mellom 1:1 og 2:1.
Det molare forhold av syrekatalysator til startforbindelse med formel (2) ligger fortrinnsvis mellom 1:1 og 0,001:1, spesielt mellom 0,01:1 og 0,05:1.
Det resulterende 2-(6-substituerte-l,3-dioksan-4-yl)eddik-syrederivat kan deretter hydrolyseres i nærvær av en base og vann for å danne det tilsvarende salt med formel 3
hvor Y står for et alkalimetall, et alkalijordmetall eller en substituert eller usubstituert ammoniumgruppe, fortrinnsvis Na, Ca eller en tetraalkylammoniumforbindelse.
Hydrolysen av forbindelsen med formel (3) utføres fortrinnsvis med minst 1 baseekvivalent, spesielt 1-1,5 base-ekvivalenter, relativt til forbindelsen med formel (3). Prinsipielt kan et større overskudd anvendes, men i praksis gir dette vanligvis ingen fordeler.
Reaksjonen utføres fortrinnsvis ved en temperatur mellom
-20 °C og 60 °C, spesielt mellom 0 °C og 30 °C.
Hydrolysen kan for eksempel utføres i vann, et organisk løsningsmiddel, for eksempel en alkohol, spesielt metanol eller etanol, et aromatisk hydrokarbon, for eksempel toluen, eller et keton, spesielt aceton eller metylisobu-tylketon (MIBK), eller en blanding av et organisk løs-ningsmiddel og vann, eventuelt katalysert av en faseover-føringskatalysator (PTC) eller tilsetning av et koløsnings-middel.
Hydrolysen kan også utføres enzymatisk, den ønskede dia-stereomer hydrolyseres eventuelt selektivt.
Eksempler på enzymer som passende kan anvendes i fremgangsmåten i henhold til oppfinnelsen er enzymer med lipase-eller esteraseaktivitet, for eksempel enzymer fra Pseudomonas, spesielt Pseudomonas fluorescens, Pseudomonas fragi; Burkholderia, for eksempel Burkholderia cepacia; Chromobacterium, spesielt Chromobacterium viscosum; Bacillus, spesielt Bacillus thermocatenulatus, Bacillus licheniformis; Alcaligenes, spesielt Alcaligenes faecalis; Aspergillus, spesielt Aspergillus niger, Candida, spesielt Candida antarctica, Candida rugosa, Candida lipolytica, Candida cylindracea; Geotrichum, spesielt Geotrichum candidum; Humicola, spesielt Humicola lanuginosa; Penicillium, spesielt Penicillium cyclopium, Penicillium roquefortii, Penicillium camembertii; Rhizomucor, spesielt Rhizomucor javanicus, Rhizomucor miehei; Mucor, spesielt Mucor javanicus; Rhizopus, spesielt Rhizopus oryzae, Rhizopus arhizus, Rhizopus delemar, Rhizopus niveus, Rhizopus japonicus, Rhizopus javanicus; lipase fra svinepankreas, hvetekimli-pase, lipase fra bovin pankreas, griseleveresterase. Fortrinnsvis gjøres det bruk av et enzym fra Pseudomonas cepacia, Pseudomonas sp., Burkholderia cepacia, svinepankreas, Rhizomucor miehei, Humicola lanuginosa, Candida rugosa eller Candida antarctica eller subtilisin. Hvis et enantio-selektivt enzym anvendes, realiseres enda mer enantiomer anrikning i løpet av hydrolysen. Slike enzymer kan erholdes ved å anvende vanlig kjente teknologier. Mange enzymer fremstilles i en teknisk skala og er kommersielt tilgjenge-lige .
De erholdte forbindelser er nye. Oppfinnelsen vedrører derfor også disse produkter med formel 3
hvor X står for et halogen, spesielt Cl, Br eller I, en to-sylat- eller mesylatgruppe, en acyloksygruppe med 3-10 karbonatomer, eller en nitrosubstituert benzensulfonylgruppe, et alkali(jord)metall, eller en substituert eller usubstituert ammoniumgruppe.
Det resulterende salt med formel 3 kan deretter konverteres til den tilsvarende t-butylester (formel la med R 3 = t-butyl) på en måte kjent per se.
I fremgangsmåten i henhold til oppfinnelsen kan forbindelsen med formel (3) for eksempel foresteres for å danne den tilsvarende tert-butylester ved å anvende de følgende meto-der, som generelt er beskrevet i litteraturen: reaksjon med isobuten og sterk syre, for eksempel para-toluensulfonsyre (pTS), svovelsyre eller en sterk sur ione-bytter (US-A-3325466);

Den resulterende t-butylester av 2-(6-substituert-l,3-dioksan-4-yl)eddiksyre kan deretter konverteres til 2-(6-hy-droksymetyl-1,3-dioksan-4-yl)eddiksyren, for eksempel som beskrevet i US-A-5594153 eller i EP-A-1024139, i nærvær av et tetraalkylammoniumhalogenid og/eller en karboksylsyre i saltet, via konversjon til en forbindelse med formel la med R3 = t-butyl og X = en acyloksy-, for eksempel en acetoksy-gruppe. Acyloksygruppen kan deretter konverteres via sol-volyse, på en måte forøvrig generelt kjent, til en hydrok-sylgruppe. Solvolysen kan gjennomføres ved å anvende en base (Na2CC>3, K2CO3, eller natriummetanolat i metanol), eventuelt ved samtidig destillasjon av det dannede metyl-acetatet.
t-butylesteren av 2-(6-hydroksymetyl-l,3-dioksan-4-yl)eddiksyre er et ønskelig intermediatprodukt i fremstillingen av forskjellige statiner, for eksempel ZD-4522, som beskrevet i Drugs of the future, (1999), 24(5), 511-513 av M. Watanabe et al., Bioorg. & Med. Chem. (1997), 5(2), 437-444. Oppfinnelsen tilveiebringer derfor en ny, interessant rute til disse mellomliggende produkter og til sluttpro-duktene, spesielt statiner.
Startforbindelsene med formel 2 kan for eksempel oppnås som beskrevet i WO-A-96/31615.
Oppfinnelsen vil bli belyst med referanse til de følgende eksempler, imidlertid uten å bli begrenset av disse.
Eksempel I
Fremstilling av ( 4R, 6S)- 4- hydroksy- 6- klormetyl- tetrahydro-pyran- 2- on ( forbindelse II; dekket av formel 2)
Ved romtemperatur ble 2,1 ml brom tilsatt i løpet av 45 minutter til en blanding av 6,7 g (40 mmol) 6-klor-2,4,6-trideoksy-D-erytro-heksose (forbindelse I; fremstilt i henhold til fremgangsmåten beskrevet i WO-A-96/31615) og 6,7 g natriumbikarbonat i 40 ml metylenklorid og 10 ml vann. CO2-gass unnslapp, mens pH forble på 5. Etter omrøring i en time hadde, i henhold til gassvæskekromatografi (GLC), startmaterialet blitt fullstendig konvertert. Bromoverskuddet ble nøytralisert med fast Na2S2C>3. Etter faseseparasjon ble vannfasen ekstrahert med to ganger 100 ml etylacetat. De kombinerte organiske faser ble tørket over Na2SC>4 og filtrert. Etter rotasjonsinndamping ble 5,5 g gul olje erholdt (82 % utbytte av forbindelsen med formel (2) med X = Cl relativt til forbindelse I).

Eksempel II
Fremstilling av ( 4R, 6S)- 4- hydroksy- 6- klormetyl- tetrahydro-pyran- 2- on ( forbindelse II; dekket av formel 2)
Til en løsning av 75 g (450 mmol) av forbindelse I i 390 ml vann ble 114 g (715 mmol) brom tilsatt ved 15-25 °C over 3 timer. pH i reaksjonsblandingen ble holdt ved 5-6 via den samtidige tilsetning av natriumkarbonat (88 g total meng-de) . Bromoverskuddet ble nøytralisert med natriumbisulfitt. Produktet ble ekstrahert fra vannfasen med etylacetat (mot-strømsekstraksjon).
Produktet ble krystallisert fra etylacetat/heptan (125 g/ 62 g). Etter avkjøling til 0 °C ble krystallene filtrert, vasket med 50 ml heptan/etylacetat (vekt:vekt =9:1) og tørket, hvilket ga 49,2 g (67 % relativt til forbindelse I) av forbindelse II som fargeløse nåler (smp. 73-74 °C) .
Eksempel III
Fremstilling av ( 4R- cis)- 6-( klormetyl)- 2, 2- dimetyl- l, 3- dioksan- 4- yl- eddiksyre- metylester ( forbindelse III; dekket ved formel 1)
5,5 g av forbindelse II som erholdt i eksempel I ble tilsatt til 20 ml kommersiell dimetoksypropan og 100 mg p-toluensulfonsyremonohydrat ved romtemperatur. Etter omrøring i en time ved romtemperatur viste GLC-analyse at fullstendig konversjon hadde funnet sted og en klar løsning hadde blitt dannet. Etter tilsetning av 500 mg NaHCC>3 fant omrøring sted i 30 minutter ved romtemperatur. Etter fil-trering og rotasjonsinndamping ble 7,1 g av forbindelse III erholdt som en lysegul olje (91 % relativt til forbindelse
II) •

Eksempel IV
Fremstilling av ( 4R- cis)- 6-( klormetyl)- 2, 2- dimetyl- l, 3 — dioksan- 4- yl- eddiksyre- metylester ( forbindelse III; dekket ved formel 1)
Til en løsning av 49,2 g (300 mmol) av forbindelse II i 100 ml toluen ble 47 g (450 mmol) dimetoksypropan og 850 mg p-toluensulfonsyremonohydrat (4,5 mmol) tilsatt. Etter omrøring i en time ved romtemperatur viste GLC-analyse fullstendig konversjon av forbindelse II. Toluenfasen ble vasket med 50 ml av en 0,2 N NaOH-løsning i vann. Etter inndamping ble 67 g av forbindelse III erholdt som en lysegul olje (94 % relativt til forbindelse II).
Eksempel V
( 4R- cis)-( 6- klormetyl)- 2, 2- dimetyl- l, 3- dioksan- 4- yl- eddiksyre, natriumsalt ( forbindelse IV; dekket ved formel 3) 55 g (233 mmol) av forbindelse III ble tilsatt til 200 ml vann. Ved romtemperatur ble 20 g av en 50 % NaOH-løsning i vann tilsatt dråpevis over 2 timer ved pH = 12. Hydrolysen ble overvåket ved å anvende GLC. Etter 20 g forble pH kon-stant. Konsentrert saltsyre ble anvendt for å senke pH til 10. Vannfasen ble vasket med 100 ml etylacetat og dampet inn ved å anvende en rotasjonsinndamper. Den dannede olje ble tørket ved å strippe med absolutt etanol og toluen. Det faste stoff ble omrørt i 200 ml aceton, filtrert og vasket med kald aceton. Utbytte etter vakuumtørking: 45,6 g = 80 % Na-salt relativt til forbindelse III.
<1>H NMR (200 MHz, CDC13/CD30D) : 5 1,21 (dt, 1H) ; 1,36 (s, 3H); 1,49 (s, 3H); 1,79 (dt, 1H); 2,25 (dd, 1H); 2,45 (dd, 1H); 3,46 (m, 2H); 4,11 (m, 1H); 4,36 (m, 1H).
Eksempel VI
( 4R- cis)-( 6- klormetyl)- 2, 2- dimetyl- l, 3- dioksan- 4- yl- eddiksyre, natriumsalt ( forbindelse IV; dekket ved formel 3)
Startene fra 49,2 g av forbindelse I ble en løsning av forbindelse III i toluen fremstilt som beskrevet i eksempel IV. 5 g metanol og 25 ml vann ble tilsatt. Ved romtemperatur ble 25 g av en 50 % løsning av NaOH i vann tilsatt dråpevis over 1 time. Etter omrøring i 4 timer ved romtemperatur indikerte GLC-analyse fullstendig hydrolyse. Base-overskuddet ble nøytralisert til pH 8,5-9,5 med 33 % HC1-løsning i vann. Vannfasen ble separert og tørket via azeo-trop destillasjon ved å anvende 470 ml toluen, hvilket ga 65 g av forbindelse IV som en 16 vekt/vekt% suspensjon i toluen med KF < 0,1 %.
Suspensjonen kan anvendes for syntesen av forbindelse V.
Eksempel VII
( 4R- cis)-( 6- klormetyl)- 2, 2- dimetyl- l, 3- dioksan- 4- yl- eddiksyre, t- butylester ( forbindelse V; dekket ved formel la)
45,5 g IV, natriumsalt (186 mmol) ble tilsatt til en løsning av 159 g ditert-butyldikarbonat i 1400 ml tørr tert-butanol. Etter tilsetning av 6,8 g dimetylaminopyridin fant omrøring sted i 16 timer ved 40 °C. Reaksjonsblandingen ble helt ut i 1500 ml etylacetat og 1.000 ml mettet ammoniumklorid. Vannfasen ble ekstrahert igjen med 1.500 ml etylacetat. De kombinerte organiske faser ble vasket med 600 ml mettet NaCl-løsning. Den organiske fase ble tørket over Na2SC>4, filtrert og deretter dampet inn under vakuum, hvilket ga 51,9 g gul olje (100 % relativt til forbindelse
IV) .
<X>H NMR (200 MHz, CDC13) : 1,15-1,33 (m, 1H) ; 1,40 (s, 3H) ; 1.45 (s, 3H); 1,47 (s, 9H); 1,77 (dt, 1H); 2,33 (dd, 1H); 2.46 (dd, 1H); 3,40 (dd, 1H); 3,49 (dd, 1H); 4,08 (m, 1H); 4,28 (m, 1H).
Eksempel VIII
( 4R- cis)- 6-[( acetoksy) metyl]- 2, 2- dimetyl- l, 3- dioksan- 4- yl-eddiksyre, t- butylester ( forbindelse VI; dekket ved formel la)
Startende fra 33 g av forbindelse V erholdt i henhold til eksempel VII ble på 16 timer 29 g av forbindelse VI erholdt ved 100 °C i henhold til US-A-5457227 (ved å anvende 40 g tetra-n-butylammoniumacetat og i 200 ml DMF), som et fast stoff etter krystallisasjon fra 75 ml heptan.
<X>H NMR (200 MHz, CDCI3) : 5 1,1-1,3 (dt, 1H) ; 1,39 (s, 3H) ; 1,45 (s, 9H); 1,47 (s, 3H); 1,57 (dt, 1H); 2,08 (s, 3H); 2,32 (dd, 1H); 2,46 (dd, 1H); 4,0-4,2 (m, 3H); 4,3 (m, 1H).
Eksempel IX
( 4R- cis)- 6-[ hydroksymetyl]- 2, 2- dimetyl- l, 3- dioksan- 4- yl-eddiksyre, t- butylester ( forbindelse VII)
Startende fra 29 g av forbindelse VI i henhold til eksempel V ble 25,0 g av forbindelse VII erholdt som en lysegul olje med e.e. = 100 %, d.e. = 99,9 % (ifølge GLC) i henhold til US-A-5457227 (det ble anvendt 6,9 g kaliumkarbonat i 300 ml metanol).
<X>H NMR (200 MHz, CDCI3) :
spektrumet var helt på linje med litteraturen (Synthesis 1014, 1995).
Claims (16)
1. Fremgangsmåte ved fremstillingen av et 2-(6-substituert-l, 3-dioksan-4-yl) eddiksyrederivat med formel 1
hvor X står for en utgående gruppe, og Ri, R2 og R3 hver uavhengig representerer en alkylgruppe med 1-3 karbonatomer startende fra en forbindelse med formel 2
hvor X er som definert over, som anvender et acetaliseringsmiddel, i nærvær av en syrekatalysator.
2. Fremgangsmåte ifølge krav 1, hvor X er klor.
3. Fremgangsmåte ifølge krav 1 eller krav 2, hvor Ri=R2=R3=CH3.
4. Fremgangsmåte ifølge ethvert av kravene 1 til 3, hvor det erholdte 2-(6-substituerte-l,3-dioksan-4-yl)eddiksyre-derivatet deretter hydrolyseres i nærvær av en base og vann for å danne det tilsvarende salt med formel 3 hvor X, Ri og R2 er som definert over og Y står for et alkalimetall, et alkalijordmetall eller en substituert eller usubstituert ammoniumgruppe.
5. Fremgangsmåte ifølge krav 4 hvor Y står for Na, Ca eller en tetraalkylammoniumforbindelse.
6. Fremgangsmåte ifølge krav 4 eller 5, hvor fremgangsmåten ytterligere omfatter det erholdte salt deretter konverteres til eddiksyren ifølge formel 3, med Y = H.
7. Fremgangsmåte ifølge ethvert av kravene 4-6, hvori det (4R,6S)-2-(6-substituerte-l,3-dioksan-4-yl)eddiksyre-derivat med formel 3 fremstilles med et enantiomert og et diastereomert overskudd som begge er høyere enn 99 %.
8. Fremgangsmåte ifølge et hvilket som helst av kravene 4 - 7, hvor fremgangsmåten ytterligere omfatter den etter-følgende konversjon av det erholdte salt eller den erholdte syre til den tilsvarende ester med formel la, hvor R3 = t-butyl
9. Fremgangsmåte ifølge krav 8, hvor fremgangsmåten ytterligere omfatter den etterfølgende konversjon av den resulterende ester med formel la, hvor R3 står for t-butyl, til t-butylesteren av 2-(6-hydroksymetyl-l,3-dioksan-4-yl)-eddiksyre.
10. Fremgangsmåte ifølge krav 9, hvor fremgangsmåten vi-dere omfatter den etterfølgende konversjon av den erholdte t-butylester til et statin.
11. Forbindelse med formel (2),
hvor X er brom, en tosylatgruppe, en mesylatgruppe, en acyloksygruppe, en aryloksysubstituert benzensulfonylgruppe eller en nitrosubstituert benzensulfonyl, og hvormed forbindelsen er i (4R,6S)-formen.
12. Forbindelse med formel 1,
hvor X representerer halo, acyloksy, en tosylatgruppe, en mesylatgruppe, en aryloksysubstituert benzensulfonylgruppe, eller en nitrosubstituert benzensulfonylgruppe; Ri, R2, og R3 hver seg uavhengig representerer en alkylgruppe med 1-3 karbonatomer.
13. Forbindelse ifølge krav 12 hvor Ri, R2 og R3 alle er CH3
14. Forbindelse med formel 3
hvor Ri og R2 hver seg uavhengig representerer en alkylgruppe med 1-3 karbonatomer, X representerer en forlatende gruppe, Y representerer et alkalimetall, jordalkalimetall eller en substituert eller ikke-substituert ammoniumgruppe.
15. Forbindelse ifølge krav 14, hvor Y er Na, Ca eller en tetraalkylammoniumgruppe.
16. Forbindelse ifølge ethvert av kravene 12-14, hvor X er Cl.
