Description
Foreliggende oppfinnelse angår en fremgangsmåte for rensing av N-L-cx-aspartyl-L-fenyl-alanin-l-metylester fra en blanding av a- og P-isomerer oppnådd ved omsetning av N-beskyttet L-aspartinsyreanhydrid med L-fenylalanin-metylester i en blanding av enten etylacetat eller diklormetan og eddiksyre.
Aspartam har søtningsegenskaper tilsvarende roe- og rørsukk-er, og benyttes som søtningsmiddel for næringsmidler og drikker som beskrevet i US-PS 3 492 131.
Aspartam er et dipeptid og som sådant er det dannet med en amidbinding mellom en aktivert karboksylgruppe på en aminosyre og aminogruppen på en annen aminosyre. Aktivering er nødvendig for å øke kondensasjonens hastighet og utbytte. Det ønskede rene peptidet krever beskyttelse av alle andre funksjonelle grupper som ikke er involvert i peptidbind-ingsdannelsen. Til slutt fjernes de beskyttende grupper.
Aspartam kan fremstilles ved omsetning av N-beskyttet-L-aspartinsyreanhydrid med L-fenyl-alanin-metylester. Dette gir en blanding av N-beskyttet a-L-aspartyl- og p-L-aspartyl-L-fenylalanin-metylester som krever N-beskyttelsesfjerning av forbindelsen fulgt av separering av de to isomerer.
De N-beskyttende grupper er de vanlige N-beskyttende grupper som benyttes i peptidkjemien slik som benzyl-oksykarbonyl- og formylgruppen. Det er meget godt kjent at reaksjonen for å fjerne disse N-beskyttende grupper er komplisert fordi spalting av peptidbindingen skjer samtidig med dannelse av uønskede biprodukter slik som diketopiperazin.
Den N-avbeskyttende reaksjon som gjennomføres ifølge kjente teknikker som vanligvis benyttes gjennomføres i nærvær av en sterk syre, se US-PS 4 071 511, eller i nærvær av hydrok-sylamin, se US-PS 4 021 418. Selv om disse prosesser ansees å være egnet ut fra et industrielt synspunkt påpekes det flere mangler som for eksempel lave utbytter, kostbare reagenser, forestring av p-karboksygruppen og hydrolyse av ester- eller peptidbindinger. Videre er produktet som således oppnås ikke rent idet det er blandet med vesentlige mengder biprodukter.
Det er derfor nødvendig etterpå å rense produktet med den derav resulterende økning av fremstillingsomkostningene.
Videre er det kjent at en blanding av a- og 3-isomerer dannes under prosessen for fremstilling av aspartam og deretter er det nødvendig med et ytterligere trinn for separering av a-og p-isomerene fordi P-isomeren ikke har søtende egenskaper. Dette fører til en ytterligere økning av produksjonsom-kostningene .
Det er nu uventet funnet en ny og meget enkel prosess som ikke oppviser de ovenfor angitte mangler.
Det tilveiebringes således en fremgangsmåte for fjerning av N-formylgruppen fra L-aspartyl-L-fenyl-alaninmetyl esteren hvori aminogruppen i aspartyldelen er beskyttet med en formylgruppe, hvilken prosess muliggjør samtidig å separere den ønskede ot-isomer fra p-isomeren.
I henhold til dette angår foreliggende oppfinnelse en fremgangsmåte av den innledningsvis nevnte art og fremgangsmåten karakteriseres ved at den omfatter å behandle den oppløste blanding med minst 85$-ig fosforsyre og en lavere alkylakohol fo>r derved å felle ut det lite oppløselige aspartamfosf at,, å oppsamle dette ved filtrering, å oppløse aspartamfosfatet i vann med etterfølgende nøytralisering av aspartamfosfatet med en organisk base for derved å danne det rensede aspartam i et utbytte på 73-90$ beregnet på N-beskyttet aspartam, idet man benytter en mengde av fosforsyre fra 1,2 til 5 mol N-beskyttet aspartam som skal deformuleres, Idet reaksjonen gjennomføres ved en temperatur fra romtemperatur til 60°C i et tidsrom fra 4 til 12 timer.'
Fortrinnsvis benyttes metylalkohol som laverealkylalkohol.
Det spontant separerte a-isomerfosfat samles ved filtrering og omdannes til fri aspartam ved behandling med en base.
Fjerning av formylgruppen direkte i kondensasjonsblandingen unngår isolering av N-formyl aspartam og reduserer produk-sj onsomkostningene.
Den industrielle prosess for fremstilling av a-L-aspartyl-L-fenylalanin-metylester blir videre forenklet ved oppfinnelsen : det er ikke nødvendig med et spesielt trinn for å separere det ønskede peptid fra den 3-isomer som dannes ved kondensasjon, da kun aspartamfosfat krystalliserer fra reaksjonsblandingen etter at beskyttelsesfjerningen er fullført.
Den milde sure hydrolyse ifølge foreliggende fremgangsmåte forhindrer at peptidbindingen spaltes og unngår fjerning av alkoholdelen fra esteren eller forestring av den frie p-karboksylgruppe.
Til slutt unngår betingelsene som benyttes ifølge oppfinnelsen dannelse av uønsket biprodukt diketopiperazin. Under henvisning til forsøksbetingelsene dannes kondensasjonsblandingen der N-deformyleringen direkte gjennomføres av et organisk oppløsningsmiddel og en organisk syre, fortrinnsvis etylacetat eller dikloretan samt eddiksyre.
Fosforsyren som benyttes ved fremgangsmåten ifølge oppfinnelsen kan ha en $-konsentrasjon i vann innen området 70-99 og fortrinnsvis 85$.
Fosforsyren som skal benyttes anvendes i en mengde fra 1,2 til 5 mol/mol N-formyl aspartam som skal deformyleres.
Hva angår alkoholen som tilsettes sammen med fosforsyren til reaksjonsblandingen har denne fra 1 til 4 karbonatomer og er fortrinnsvis metylalkohol. Deformyleringen gjennomføres ved en temperatur fra romtemperatur til 60°C ifra 4 til 12 timer.
Den ønskede rene forbindelse samles som fosfat fra reaksjons-produktblandingen ved avkjøling av reaksjonsblandingen etter at reaksjonen er ferdig.
Aspartamfosfatet som således ble oppnådd omdannes til fri aspartam ved nøytralisering av fosfatet under anvendelse av en base slik som natriumkarbonat, natriumhydroksyd eller ammoniakk i et vandig oppløsningsmiddel.
Foreliggende oppfinnelse tilveiebringer en kommersielt meget brukbar prosess for fremstilling av aspartam fordi det rene ønskede peptid ifølge oppfinnelsen kan fremstilles under unngåelse av to trinn, nemlig isolering av N -formylaspartam og separering av cx- og p-isomerer, med isolasjonsutbytter høyere enn de som oppnås i henhold til de konvensjonelle kjente prosesser.
Oppfinnelsen skal illustreres ytterligere ved følgende eksempler.
Eksempel 1
Til en oppløsning av 100 g N-formyl cx, P-L-aspartyl-L-fenylalaninmetylester med et cx:<p->isomer forhold på 8:2 i 160 ml dikloretan og 40 ml eddiksyre settes 360 ml metylalkohol og 35,2 ml 85$-ig vandig fosforsyre ved romtemperatur.
Blandingen ble oppvarmet til 40°C i 8 timer og deretter avkjølt.
a-L-aspartyl-L-fenylalaninmetylesterfosfatet som krystall-iserte ut ble samlet ved filtrering og tørket.
Utbyttet var 85$, beregnet på basis av N-formyl aspartam.
Eksempel 2
51 g cx-aspartyl-L-fenylalanin-metylesterfosfat ble oppløst i 300 ml vann. Den resulterende oppløsning ble justert til pH 5,2 med 20$ vandig NaOH omrørt i 1 time ved romtemperatur, avkjølt og det utfelte fri aspartam ble samlet ved filtrering.
Det ble oppnådd 27,9 g ren forbindelse i et utbytte av 73$ og med et smeltepunkt på 233 - 235°C under dekomponering, [a]§<2=> + 33.2 (c=l, eddiksyre).
Eksempel 3
Til en oppløsning av 100 g N-f ormyl-a ,cxb-L-aspartyl-L-fenylalanin-metylester med et a:p-isomerforhold på 8:2 i 380 ml etylacetat og 40 ml eddiksyre ble det tilsatt 400 ml metylalkohol og 70,4 ml 85$-ig vandig fosforsyre.
Ved å arbeide som i eksempel 1 ble aspartamfosfat oppnådd i et utbytte av 90$.
Eksempel 4
Ved å arbeide som i eksempel 1 men ved å bruke 99$-ig fosforsyre istedet for 85$-ig ble aspartamfosfat oppnådd i et utbytte av 75$.
Eksempel 5
Ved å arbeide som i eksempel 1 men ved å bruke vandig 50$-ig fosforsyre istedet for 85$-ig ble aspartamfosfat oppnådd i et utbytte av 65$.
Claims (2)

Fremgangsmåte for rensing av N-L-cx-aspartyl-L-fenylalanin-1-metylester (aspartam) fra en blanding av a- og p<->isomerer av N-formyl L-aspartyl-L-fenyl-alanin metylester, oppnådd ved kondensasjon mellom N-formyl-L-aspartinsyreanhydrid og L-fenylalanin metylester, oppløst i et organisk oppløsnings-middel slik som etylacetat eller dikloran og eddiksyre, karakterisert ved at den omfatter å behandle den oppløste blanding med minst 85^-ig fosforsyre og en lavere alkylakohol for derved å felle ut det lite oppløselige aspartamfosfat, å oppsamle dette ved filtrering, å oppløse aspartamfosfatet i vann med etterfølgende nøytralisering av aspartamfosfatet med en organisk base for derved å danne det rensede aspartam i et utbytte på 73-90% beregnet på N-beskyttet aspartam, idet man benytter en mengde av fosforsyre fra 1,2 til 5 mol N-beskyttet aspartam som skal deformuleres, idet reaksjonen gjennomføres ved en temperatur fra romtemperatur til 60° C i et tidsrom fra 4 til 12 timer.
