Abstract
Un aspecto de la invencion es proporcionar construcciones genéticas novedosas para la expresion de ARN inhibitorio en el citoplasma de células eucarioticas. El ARN inhibitorio puede ser un ARN de anti sentido o bien un ARN co-supresor. Las construcciones genéticas de la invencion pueden replicarse en el citoplasma de una célula eucariotica y comprenden una region promotora en combinacion funcional con un polinucleotido de codificacion. Las construcciones genéticas pueden ser diseñadas para replicarse en el citoplasma de células vegetales, células de levadura, y células de mamíferos. Cuando la célula eucariotica de interés es una célula vegetal, la construccion genética se deriva de preferencia de un virus de ARN vegetal. El virus de ARN vegetal derivado de construcciones genéticas puede emplear un promotor subgenomico de virus de planta, incluyendo promotores subgenomicos de tobamovirus en combinacion funcional con la region de codificacion de ARN. Otro aspecto de la presente invencion es proporcionar células que comprenden las construcciones genéticas de la invencion y organismo que comprende una pluralidad de tales células. Otro aspecto de la presente invencion es proporcionar métodos para reducir la expresion de un gen de interés en células eucarioticas, es decir, métodos para producir células eucarioticas que presentan niveles reducidos de expresion de un gen de interés. Los métodos de la presente invencion comprenden el paso de transfectar una célula con una construccion genética de la invencion en donde la region que codifica el ARN es específica para el gen de interés. Otro aspecto de la presente invencion es proporcionar células vegetales que producen niveles elevados de fitoeno carotenoide. Los niveles elevados de fitoeno se logran mediante la inhibicion de la expresion en la enzima fitoeno desaturasa usando los vectores la invencion.
