Abstract

Un aspecto de la invencion es proporcionar construcciones genéticas novedosas para la expresion de ARN inhibitorio en el citoplasma de células eucarioticas. El ARN inhibitorio puede ser un ARN de anti sentido o bien un ARN co-supresor. Las construcciones genéticas de la invencion pueden replicarse en el citoplasma de una célula eucariotica y comprenden una region promotora en combinacion funcional con un polinucleotido de codificacion. Las construcciones genéticas pueden ser diseñadas para replicarse en el citoplasma de células vegetales, células de levadura, y células de mamíferos. Cuando la célula eucariotica de interés es una célula vegetal, la construccion genética se deriva de preferencia de un virus de ARN vegetal. El virus de ARN vegetal derivado de construcciones genéticas puede emplear un promotor subgenomico de virus de planta, incluyendo promotores subgenomicos de tobamovirus en combinacion funcional con la region de codificacion de ARN.
