Abstract

본 발명은 블럭 공중합체 미셀을 이용한 약물전달체 및 이에 약물을 봉입하는 방법에 관한 것으로, 생분해성 소수성 고분자로서 폴리락티드, 폴리글리콜리드, 폴리락티드글리콜리드, 폴리카프로락톤 및 이들의 유도체로 구성되는 그룹으로부터 선택된 적어도 하나와, 친수서 고분자로서 폴리알킬렌옥시드로 구성된 2중 또는 3중 블럭 공중합체로부터 약물 전달 미셀을 형성하고, 여기서 물에 난용성인 약물을 교반, 가열, 초음파조사, 용매증발, 투석 등의 물리적인 방법을 통하여 봉입하는 것으로 구성되며 소수성 약물의 용해성을 향상시켜 효과적으로 체내 전달이 이루어지고 약물의 약리효과를 현저히 증대시키기 위한 약물 전달 시스템으로 이용될 수 있다.

블록 공중합체 미셀을 이용한 약물전달체 및 이에 약물을 봉입하는 방법

제1도는 본 발명에 따른 폴리락티드-폴리에틸렌글리콜-폴리락티드 3중 블럭 공중합체(EL-3L-1)의 겔투과 크로마토그래피 결과를 나타낸 그래프이고,
제2A 및 2B도는 각각 폴리락티드-폴리에틸렌글리콜-폴리락티드 3중 블럭 공중합체 (EL-3L-1)와 폴리에틸렌글리콜-폴리락티드 2중 블럭 공중합체 (EL-2L-2) 미셀(micelle)의 직경 분포 상태를 동적 광산란법 (dynamic light scattering method)에 의해 측정한 결과를 나타낸 그래프이고.
제3A 및 3B도는 각각 상기 EL-3L-1과 EL-2L-2의 농도 변화와 교반법에 의해 봉입된 탁솔의 양의 관계를 나타낸 그래프이며;
제4A 및 4B도는 각각 3중 블럭 공중합체 및 2중 블럭 공중합체의 종류와 교반법에 의해 봉입된 탁솔의 양의 관계를 나티낸 그래프이고,
제5A 및 5B도는 각각 탁솔이 봉입된 폴리락티드-폴리에틸렌글리콜-폴리락티드 3중 블럭 공중합체 (EL-3L-1)와 폴리에틸렌글리콜-폴리락티드 2중 블럭 공중합체 (EL-2L-2) 미셀의 직경 분포 상태를 동적 광산란법 (dynamic light scattering method)에 의해 측정한 결과를 나타낸 그래프이다.

Claims (9)

  1. 생분해성 소수성 고분자로서 폴리락티드, 폴리글리콜리드, 폴리락티드글리콜리드, 폴리카프로락톤 및 이들의 유도체로 구성되는 그룹으로부터 선택된 적어도 하나와, 친수서 고분자로서 폴리알킬렌옥시드로 구성된 2중 또는 3중 블럭 공중합체로부터 형성되는 미셀에 적어도 하나의 소수성 약물이 봉입된 고분자 미셀형 약문 절단체.
  2. 제1항에 있어서, 상기 블럭 공중합체가 하기 식 (Ⅰ) 또는 (Ⅱ)인 것을 특징으로 하는 약물전달체 :
    여기에서, R1은 수소, 또는 탄소수 1 내지 20의 직쇄 또는 측쇄 알킬 그룹이고, m은 2내지 3,000의 정수이고, X는 100내지 100,000의 분자량을 갖는, 폴리락티드, 폴리글리콜리드, 폴리락티드글리콜리드, 폴리카프로락톤 및 이들의 유도체로 구성되는 그룹으로부터 선택된 적어도 하나이다.
  3. 제1항에 있어서, 상기 블럭 공중합체가 친수성 부분으로서 폴리에틸렌옥시드와 소수성 부분으로서 폴리락티드를 포함하는 하기 식 (Ⅲ) 또는 (Ⅳ)인 것을 특징으로 하는 약물전달체 :
    여기에서 i, j 및 k는 각각 독립적으로 2 내지 1,000의 정수이며, m은 2 내지 3,000의 정수이다.
  4. 제1항에 있어서, 상기 소수성 약물이 항암제, 소염진통제, 면역억제제, 간장 질환 치료제, 호르몬제, 항생제, 화학요법제, 대사성 의약품, 소화기 질환 치료제, 호흡기 질환 치료제, 항알러지약, 중추신경계 질환 치료제, 말초 질환 치료제, 순화기 질환 치료제로 이루어진 그룹중에서 선택된 적어도 하나인 것을 특징으로 약물전달체.
  5. 제1항 또는 제4항에 있어서, 상기 소수성 약물이 탁솔(taxol), 아드리아마이신(adriamycin) 테니포사드(teniposide), 에토포사이드(etoposide), 다우노마이신 (daunomycin), 메토트레세이트(methotrexate) 또는 미토마이신 C(mitomycin C), 인도메타신 (indomethacin), 이부프로팬 (ibuprofen), 사이클로스포린(cyclosporine), 비페닐디메틸카르복실레이트로 이루어진 그룹중에서 선택된 적어도 하나인 것을 특징으로 하는 약물전 달체.
  6. 제1항 또는 제4항에 있어서, 상기 소수성 약물이 탁솔, 사이클로스포린 및 아드리아마이신으로 이루어진 그룹중에서 선택된 적어도 하나인 것을 특징으로 하는 약물전달체.
  7. 제1항에 있어서, 동결건조후 수용액을 가했을 때 투명하게 재분산되는 것을 특징으로 하는 약물전달체.
  8. 1) 생분해성 소수성 고분자로서 폴리락티드, 폴리글리콜리드, 폴리락터드글리콜리드, 폴리카프로락톤 및 이들의 유도체로 구성되는 그룹으로부터 선택된 적어도 하나와, 친수성 고분자로서 폴리알킬렌옥시드로 구성된 2종 또는 3종 블럭 공중합체를 용매에 용해시켜 공중합체 용액을 만드는 단계; 2) 상기 공중합체 용액에 소수성 약물을 혼합하여 상기 공중합체로된 미셀에 상기 소수성 약물이 봉입되도록 하는 단계를 포함하는, 블럭 공중합체 미셀에 소수성 약물을 봉입시키는 방법.
  9. 제8항에 있어서, 상기 공중합체 용액에 상기 소수성 약물을 혼합시킨 후 교반, 가열 초음파조사, 용매 증발 또는 투석하여 소수성 약물을 봉입시키는 것을 특징으로 하는 방법,
