Abstract

본 발명은 식물내에서 유전자를 발현시키는 신규의 방법에 관한 것으로서, 식물의(+)가닥 RNA 비루스의 RNA 증식 특성에 기초하는 것으로, 키메라서 멀티시스트론성 유전자가 식물 프로모터, 비루스 복제 오리진, 비루스 이동 단백질 유전자 및 하나 이상의 외부 유전자를 함유하도록 비루스 서브게놈성 프로모터의 조절하에 제조되며, 하나 이상의 상기 재결합 RNA 전사체를 함유하는 식물이 헬퍼 비루스로 접종되고, 헬퍼 비루스의 존재시, 재결합 전사체가 외부 유전자 RNA를 높은 수준으로 생성시키면서 복제되고, 담배비루스 그룹으로부터 유래된 헬퍼 비루스와 이동 단백질 유전자로 레플리콘 RNA 증식에 의하여 담배 식물내에서 효소 클로람페니콜 아세틸트랜스 퍼라아제를 높은 수준으로 발현시키는 시퀀스가 제공되는 것을 특징으로 한다.

Claims (22)

  1. 식물 세포의 염색체내로 결합된 트랜스 유전자로부터 전사되고, (+)의 단일 가닥 RNA 식물 비루스인 실질적인 시퀀스를 함유하는 복제 오리진; 및 (+)의 단일 가닥 RNA 식물 비루스에 비천연인 적어도 하나의 유전자를 코딩하여 복제에 필요한 적어도 하나의 단백질을 코딩하지 않는 것을 특징으로 하는 레플리콘.
  2. 제1항에 있어서, 상기 비-천연 유전자의 발현이 복제를 위한 레플리콘에 의하여 필요한 단백질을 코딩하는 헬퍼 비루스에 의하여 조절되는 것을 특징으로 하는 레플리콘.
  3. 제1항에 있어서, 비천연 유전자를 코딩하는 시퀀스가 레플리콘의 5´ 내지 3´레플리콘 오리진에 위치하는 것을 특징으로 하는 레플리콘.
  4. 제2항에 있어서, 상기 레플리콘이 시스템성 감염을 위한 헬퍼 비루스에 의하여 필요한 유전자를 코딩하는 것을 특징으로 하는 레플리콘.
  5. 제4항에 있어서, 헬퍼 비루스에 의하여 필요한 유전자는 비루스 이동 단백질인 것을 특징으로 하는 레플리콘.
  6. 제5항에 있어서, 상기 비루스 이동 단백질이 레플리콘의 3´내지 5´복제 오리진에 위치하는 것을 특징으로 하는 레플리콘.
  7. 제6항에 있어서, 비천연 유전자를 코딩하는 시퀀스가 레플리콘의 5´내지 3´복제 오리진에 위치하는 것을 특징으로 하는 레플리콘.
  8. 제5항에 있어서, 상기 비천연 유전자가 복제를 위한 레플리콘에 의하여 필요한 단백질을 코딩하는 헬퍼 비루스의 존재시 시스템적으로 발현되는 것을 특징으로 하는 레플리콘.
  9. 제5항에 있어서, 상기 이동 단백질이 담배비루스에 천연적인 것을 특징으로 하는 레플리콘.
  10. 제5항에 있어서, 상기 이동 단백질이 TMV 균주 비루스에 천연적인 것을 특징으로 하는 레플리콘.
  11. 단백질이 (+)의 단일가닥 RNA 식물 비루스에 비천연적인 유전자에 의하여 코딩되는 제1항에 따른 레플리콘을 사용하는 것을 특징으로 하는 식물 세포내에서 발현되는 단백질.
  12. 단백질이 (+)의 단일가닥 RNA 식물 비루스에 비천연적인 유전자에 의하여 코딩되는 제5항에 따른 레플리콘을 사용하는 것을 특징으로 하는 식물 세포내에서 발현되는 단백질.
  13. 제1항에 따른 레플리콘을 사용하는 것을 특징으로 하는 식물 세포내에서 발현되는 RNA 시퀀스.
  14. 제5항에 따른 레플리콘을 사용하는 것을 특징으로 하는 식물 세포내에서 발현되는 RNA 시퀀스.
  15. 제1항에 따른 레플리콘에 의하여 코딩되는 비천연 유전자를 발현시킨 결과 형질감염된 식물의 조직내에 축적되는 것을 특징으로 하는 1차 또는 2차 대사물질.
  16. 제5항에 따른 레플리콘에 의하여 코딩되는 비천연 유전자를 발현시킨 결과 형질감염된 식물의 조직내에 축적되는 것을 특징으로 하는 1차 또는 2차 대사물질.
  17. (+)의 단일 가닥 RNA 식물 비루스인 실질적인 시퀀스를 함유하는 복제 오리진과 (+)의 단일 가닥 RNA 식물 비루스에 비천연인 적어도 하나의 유전자를 코딩하며, 복제에 필요한 적어도 하나의 단백질을 코딩하지 않는 레플리콘을 코딩하는 식물세포의 염섹체내로 결합된 트랜스 유전자를 특징으로 하는 유전자전환성 식물.
  18. a)(+)의 단일 가닥 RNA 식물 비루스인 실질적인 시퀀스를 함유하는 복제 오리진; 및 (+)의 단일 가닥 RNA 식물 비루스에 비천연인 적어도 하나의 유전자를 코딩하며 복제에 필요한 적어도 하나의 단백질을 코딩하지 않는 것을 특징으로 하는 레플리콘을 코딩하는 트랜스유전자를 식물세포의 염색체내로 결합시키고, b) 복제를 위한 레플리콘에 의하여 필요한 단백질을 코딩하는 헬퍼 비루스로 식물 세포를 감염시키는 것을 특징으로 하는 식물내에 유전자를 발현시키는 방법.
  19. 제18항에 있어서, 시스템성 감염을 위하여 레플리콘이 헬퍼 비루스에 의하여 필요한 유전자를 코딩하는 것을 특징으로 하는 방법.
  20. 제19항에 있어서, 헬퍼 비루스에 의하여 필요한 유전자가 이동 단백질을 코딩하는 것을 특징으로 하는 방법.
  21. 제20항에 있어서, 상기 이동 단백질이 담배 비루스에 천연적인 것을 특징으로 하는 방법.
