Claims (28)
- 아데노바이러스 게놈을 포함하는 재조합 서브그룹 B 아데노바이러스 입자로, 여기서 상기 게놈은 E1-영역에 결실을 포함하고, 그 아데노바이러스 게놈은 기능성 E1B 55K 유전자 생성물을 코딩하지 않고, 상기 아데노바이러스 게놈은 발현 서열의 조절하에 기능성 pIX 코딩 서열을 포함하고, E1B 55K 코딩 서열의 3'말단의 일부를 포함하는 상기 발현 서열은 야생형 서브그룹 B 아데노바이러스의 pIX 개방해독 프레임의 바로 상류에 있는 아데노바이러스의 150 내지 600 뉴클레오티드를 함유하고, 그리고 상기 발현 서열은, E1B 55K 정지 코돈과 pIX의 개시 코돈 사이의 서열이 앞에 있고 E1B 55K 코딩 서열의 3'말단으로부터의 뉴클레오티드가 없을 때, 상기 pIX 코딩 서열의 발현에 비해 패키징 세포의 pIX 코딩 서열의 발현을 증가시킬 수 있는 것인 재조합 아데노바이러스 입자.
- 제1항에 있어서, 상기 발현 서열은 야생형 서브그룹 B 아데노바이러스의 pIX 개방 해독 프레임의 바로 상류에 있는 아데노바이러스 서열의 150 내지 250 뉴클레오티드를 함유하는 것인 재조합 아데노바이러스 입자.
- 제1항에 있어서, 상기 발현 서열은 E1B 55K 개방 해독 프레임 서열의 3' 말단의 166 염기쌍을 함유하는 것인 재조합 아데노바이러스 입자.
- 제1항에 있어서, 상기 발현 서열은 야생형 서브그룹 B 아데노바이러스의 pIX 개방 해독 프레임의 바로 상류에 있는 아데노바이러스 서열의 150 내지 200 뉴클레오티드를 함유하는 것인 재조합 아데노바이러스 입자.
- 제1항에 있어서, 상기 아데노바이러스 입자는 아데노바이러스 혈청형 35 입자 또는 아데노바이러스 혈청형 11 입자인 재조합 아데노바이러스 입자.
- 제1항에 있어서, 추가로 아데노바이러스 서브그룹 C 기원의 기능성 E4-orf6 단백질을 코딩하는 서열을 포함하는 것인 재조합 아데노바이러스 입자.
- 제1항의 재조합 아데노바이러스 입자의 게놈을 포함하는 단리된 핵산.
- 제1항의 재조합 아데노바이러스 입자 및 적합한 담체 또는 보조제를 포함하는 백신.
- 제1항의 재조합 아데노바이러스 입자를 포함하는 재조합 아데노바이러스 패키징 세포.
- pIX 코딩 서열을 포함하고 E1-영역에 결실을 갖는 아데노바이러스 게놈을 포함하는 재조합 서브그룹 B 아데노바이러스 입자로, 상기 아데노바이러스 입자는 기능성 E1B 55K 유전자 생성물을 코딩하지 않고, 여기서 상기 pIX 코딩 서열은 이종성 프로모터의 조절하에 있고, 여기서 상기 이종성 프로모터는 E1B 55K 정지 코돈과 pIX의 개시 코돈 사이의 서열이 앞에 있고 E1B 55K 코딩 서열로부터의 뉴클레오디드가 없을 때, 상기 pIX 코딩 서열의 발현에 비해, 패키징 세포에서 pIX 코딩 서열의 발현을 증가시킬 수 있는 것인 재조합 아데노바이러스 입자.
- 제10항에 있어서, 상기 이종성 프로모터는 비-내인성 아데노바이러스 근접의 pIX 프로모터, 비-아데노바이러스 바이러스성 프로모터, 세포성 프로모터, 합성 프로모터 및 하이브리드 프로모터로 이루어진 군으로부터 선택되는 핵산 서열을 포함하는 것인 재조합 아데노바이러스 입자.
- 제11항에 있어서, 상기 이종성 프로모터는 Ad5 근접 pIX 프로모터 또는 라우스 사르코마 바이러스(Rous Sarcoma Virus) 프로모터로 이루어진 군으로부터 선택되는 것인 재조합 아데노바이러스 입자.
- 아데노바이러스 게놈이 E1-영역에 결실을 갖고 그리고 상기 게놈은 기능성 E1B 55K 유전자 생성물을 코딩하지 않는, 아데노바이러스 게놈을 포함하는 재조합 서브그룹 B 아데노바이러스 입자의 안정성 또는 패키징 수용량을 증가시키는 방법으로, 상기 방법은아데노바이러스 게놈 내 pIX 코딩 서열은 E1B 55K 정지 코돈과 pIX의 개시 코돈 사이의 서열이 앞에 있고 E1B 55K 코딩 서열의 3'말단으로부터의 뉴클레오티드가 없을 때 얻어진 pIX 유전자 생성물의 수준에 비해, 상기 패키징 세포에서 pIX 유전자 생성물의 상승된 수준의 존재하에 패키징 세포 중에 재조합 아데노바이러스 입자로 상기 아데노바이러스 게놈의 생산 및 조립에 필요한 성분들을 발현시키는 단계를 포함하는 방법.
- 제13항에 있어서, 상기 pIX 유전자 생성물의 상승된 수준은 아데노바이러스 게놈 중의 야생형 서브그룹 B 아데노바이러스의 pIX 개방 해독 프레임의 바로 상류에 있는 아데노바이러스 서열의 150 내지 600 뉴클레오티드를 함유하는 E1B 55K 코딩 서열의 3'말단의 일부를 유지하거나 또는 재도입하여 생산되는 것인 방법.
- 제13항에 있어서, 상기 pIX 유전자 생성물의 상승된 수준은 이종성 프로모터의 조절하에 pIX 코딩 서열의 발현에 의해 생산되는 것인 방법.
