Description
本発明において、E1B 55Kの停止コドンまでにE1領域中に欠失を有する組み換えグループBアデノウイルスは、類似したAd5組み換えアデノウイルスよりも少ない外来配列を提供できることが示されている。これは、pIXコード領域の先に、E1B 55K停止コドンとpIX開始コドンの間の配列のみがあるときに、所定のパッケージング細胞中で前記グループBのウイルス中でのpIX遺伝子の発現が比較的低いことに依るようである。そのようなウイルスにより高い安定性を付与すること、および/または、より多くの外来配列を入れることができることが示され、そのようなウイルス中にE1B 55Kコード領域由来の配列を含むことによってpIXプロモーターが少なくとも部分的に回復した時か、あるいはpIXを制御するための異種プロモーターを使用することのいづれかにより、所定のパッケージング細胞中においてpIXの正常な、あるいは相対的に過剰であるような発現さえ達成される。

)。応用微生物学研究センター(Applied Microbiology and Research)の欧州動物細胞培養コレクションにNo. 96022940として寄託されたPER.C6(登録商標)は、そのために、組み換えアデノウイルスを増殖させるための非常に適切なパッケージング細胞である。複製能を有するアデノウイルスの産生を減らすための他の方法も認識されており、例えば、パッケージング細胞とベクターの中に存在する配列の間の相同性を減らすために、アデノウイルスの配列を操作するための関心事であった(例えば、Hehir et al., 1996; Robert et al., 2001)。例えばE2, E4などの一部分または全部など、使用された組み換えアデノウイルス中でこれらの機能が機能的に欠損しているとき、必須のE1領域に加えて、パッケージング細胞は、他のアデノウイルスの機能を補うためのアデノウイルス配列を備える。パッケージング細胞中の相補情報は、ゲノム中に組み込まれているか、あるいは例えばプラスミド、ベクター、コスミドなど、細胞外のコピーとしてかのいずれかで存在することができる。他の方法では、組み換えアデノウイルス中では欠損している遺伝情報を備える、所謂ヘルパーウイルスが使用される。組み換えアデノウイルスベクターは、大部分または全てのアデノウイルス遺伝子が除去されたゲノムを含む、組み換えアデノウイルスの産生のために使用される所謂ヘルパーウイルスとしても使用される(ガッツレス(gutless)ベクターまたはヘルパー依存性アデノウイルス)。そのようなガッツレスアデノウイルスの最終作製において、ヘルパーアデノウイルスのパッケージングを避ける必要がある。本技術分野の当業者はこれを達成する方法に精通しており、例えば部位特異的な組み換え酵素をパッケージングシグナル中の加工部位に使用してこのパッケージングシグナルを除去する。しばしば、CsClグラジエント分離を用いて、望みのガッツレスウイルスから残っているヘルパーウイルスを分離する必要がある。ヘルパーおよびガッツレスウイルスのゲノム長さが最大限に異なっているときには、これを達成することは容易となる。そこで大きなヘルパーウイルスは、小さいものよりも好適である。本技術分野の当業者にとって明らかであるように、pIXの発現が増加した組み換えヘルパーウイルスを使用することによって組み換えアデノウイルスの安定性を増加させるために、本発明を同様に適用することが可能であり、それは本発明で述べられた方法により達成できる。組み換えアデノウイルスを相補するのに使用できる遺伝情報を含んでいる任意の細胞を、組み換えアデノウイルスを相補するために使用して、組み換えウイルス微粒子を産生することが可能であり、それはパッケージング細胞の意味の範囲内に含まれるという意図である。本発明によって得られる利点は使用されるパッケージング細胞に依存していないことは、本技術分野の当業者に明らかであろう。
PER.C6培養培地[10%までのFBS(ギブコBRL)及び10mMのMgCl2(4.9Mのストック溶液、シグマ)を補ったDMEM(ギブコBRL)]中で、3x106個の細胞/培養皿の密度で、10cmの培養皿にPER.C6細胞を播いた。2日後、ScaIで直鎖状化したpIG35.55KのDNA(下に記載)の1μgで9つの培養皿をトランスフェクトし、ScaIで直鎖状にしたpIG35.55KのDNAの1.5μgで9枚の培養をトランスフェクトした。別の対照の培養皿では、ScaIで直鎖状にしたpAdApt35.LacZ(WO 00/70071に記載)の1μg又は1.5μgでトランスフェクトし、形質質移入効率をモニターし、また、ScaIで直鎖状にしたpcDNA.nlsLacZの1μg又は1.5μgでトランスフェクトした。pcDNA.nlsLacZ(WO99/55132に記載)は、CMVプロモーターによって駆動されるnlsLacZ遺伝子を有するpcDNA3を基にしたプラスミド(インビトロゲン)である。pcDNA.nlsLacZはまた、neor発現カセットを含有する。陰性対照として、余分の培養皿1枚を、neor選抜遺伝子を欠くコンストラクトである直鎖状にしたpAdApt35.LacZでトランスフェクトした。全てのトランスフェクションは、製造元の指示書に従って、DNAμg当たり5mlのリポフェクトアミン試薬を用いて、リポフェクトアミン形質移入キット(インビトロゲン/ライフテクノロジーズ)により行った。トランスフェクション混合物と共に細胞を4時間インキュベートし、その後、培地をPER.C6培養培地に置き換えた。翌日、1μg又は1.5μgのpAdApt35.LacZでトランスフェクトした2枚の培養皿を除いて、培地を、0.5mg/mlのG418(ギブコBRL)を含有する培養培地に置き換えた。これらの後者の培養皿は、トランスフェクトして2日のLacZの発現をモニターするのに用いた。これらの培養物をX−gal染色した後、形質移入効率は、1.5μgのDNAでトランスフェクトした培養皿の中のやや青色が多い細胞により、およそ40%であると概算された。トランスフェクトされた残りの培養皿では、選抜培地を週に2回取り替えた。選抜培地を最初に添加した後の2週間以内に、陰性対照の培養皿(1.5μgのpAdApt35.LacZでトランスフェクトした)における細胞はほとんど死滅した。pcDNA.nlsLacZでトランスフェクトした培養皿では、細胞クローンが見えるようになった。pIG35.55Kでトランスフェクトした細胞の方がG418に対してさらに耐性を有すると思われたので、トランスフェクトの3週間後、濃度を0.75mg/mlに上げた。3日後及び7日後に、pIG35.55Kでトランスフェクトした培養皿から合計196の細胞クローンを選び、96穴プレートの別々のウエルに播いた。
ヒトのアデノウイルスの初期領域−3は、アデノウイルスの感染に対する宿主免疫反応を妨害するタンパク質のための複数のコーディング領域を含有する。アデノウイルスベクターをワクチンのキャリアとして使用する場合、そのような妨害は望ましくない。従って、我々は、E3領域を欠くAd35骨格のコスミドを構築した。
異なったインサートを含有するAd35に基づく、E1欠失及びE1/E3欠失のベクターを作成し、それは、PER55Kクローン#16細胞(実施例1を参照のこと)を、

組換えAd35ウイルスのゲノム長が増えるにつれて、導入遺伝子に欠失が生じるという知見は、pIX欠失Ad5ウイルス(Ghosh-Choudhury et al., 1987)との類似において、カプシドの安定性の問題があることを示唆している。全長36.7kbのAd35.E1B+AdApt.Lucウイルス(E1Aの配列がAdApt.Lucのカセットに置きかえられている)を作製することができるという事実は、E1Bの欠失が役目を担っていることを示唆する。これは、E1Bタンパク質自体の1つの機能を介して、又はほかのアデノウイルスタンパク質の発現に影響を有するこの領域における未知の調節性配列の機能を介して、のいずれかであってもよい。E1Bタンパク質自体がアデノウイルスのパッケージング容量に影響を与えることは、我々の知識では記載されていない。しかしながら、E1B−21Kタンパク質はトランスフェクトされたDNA(Herrman and Mathews, 1989)を非特異的に安定化し、21Kにおける突然変異は結果として、感染の間、細胞性DNA及びウイルス性DNAの分解を生じる(Pilder et al., 1984; White et al., 1984)ことが記載されている。Ad5のE1B−21Kタンパク質は、PER.C6細胞で発現しているので、これらの知見は、我々の所見に説明を提供していない。pIX遺伝子は、E1B−55Kのコーディング領域の3’に直接位置する。Ad5については、pIXとE1Bがポリアデニル化シグナルを共有するので、pIXのプロモータとコーディング配列がE1B転写領域の範囲内に位置することが知られている。Ad5の場合、pIXの発現に必要な最小プロモーター配列が検討されている(Babiss and Vales, 1991)。上流のSp1部位及びTATAボックス配列を含有するプロモーター配列は、pIXの発現に十分であることが示された。Sp1部位とTATAボックスとの間の間隔ならびにTATAボックス自体の配列が、pIXの発現の程度に影響することが示された。Ad35における相当する領域は、安定なウイルスにおいて十分高い程度にpIXを発現を駆動するかどうかは判っていない。配列の比較は、Sp1部位及びTATA配列の双方が、Ad5のpIXプロモーターで見い出されるものとは異なっていることを示した。ジェンバンクから利用可能な配列情報を用いて、異なったサブグループの抗原型の近位pIXの上流配列(すなわち、E1B−55Kの停止コドンとpIX遺伝子の開始コドンの間)の比較を行った。比較には、配列番号及びジェンバンク参照配列を持つ以下のアデノウイルスを用いた:Ad2(配列番号45;ジェンバンクNC_001405)、Ad5(配列番号46;ジェンバンクM73260)、Ad12(配列番号47;ジェンバンクNC_001460)、Ad9(配列番号48;ジェンバンクAF099665)、Ad40(配列番号49;ジェンバンクL19443)、Ad4(配列番号50;ジェンバンクNC_003266)、サル25(配列番号51;ジェンバンクAF394196)、Ad7(配列番号54;ジェンバンクAD7001)。Ad35の配列(配列番号52)はWO 00/70071に公開されている。Ad11の配列(配列番号53)は、以前は公開されていなかったが、本願明細書で提供される。図3Aは、E1B−55Kタンパク質の停止コドン(すべての配列の最初の3つのヌクレオチド)とpIXタンパク質の開始コドン(最後の3つのヌクレオチド)の間の上述の配列のアラインメントを示す。Ad2及びAd5におけるSp1部位及びTATA配列を四角で囲う。ほとんどの場合他の配列においては、Sp1及びTATAボックスを直接指すには不十分な相同性しかない。従って、Bucher,P(1990年)によって公開されたようなGC−及びTATAボックスに対するコンセンサス配列を用いて、種々の配列における推定上のSp1及びTATAボックスを同定した。図3bは、推定上のSp1及びTATAボックスの配列及びそれらの間の間隔を示す。それぞれサブグループA、D、及びFに属するAd12、Ad9及びAd40は、コンセンサス配列にかなり一致するSp1及びTATAボックスの配列を有する。しかしながら、2つのボックスの間の距離は、Ad5及びAd2に対するよりも小さい。Ad5のE1Bプロモーターも11ヌクレオチドの間隔を持つSp1ボックスとTATA配列を含有するので、これは特異なことではない。しかしながら、Sp1とTATAボックスの間のAd5pIXプロモーター配列における(20のうちの)9ヌクレオチドの欠失は、pIXレベルの低下を生じた(Babiss and Vales, 1991)。サブグループBの抗原型、Ad35、Ad11及びAd7は、サブグループEのウイルスAd4と同様に、多様なTATAボックス配列及び推定上のSp1配列とTATAボックスの間の異なった間隔を有する。ヒトのアデノウイルス4型における近位pIX領域は、サルのアデノウイルス25(CV68)におけるものと同一であり、最近治療用ベクター(Farina et al., 2001)として提案された抗原型である。従って、非ヒト型アデノウイルスに基づく複製不全のベクターについても、pIXの発現は安定なカプシドには不十分である可能性がある。
pAdApt535は、Ad5のpIXのプロモーターの一部を有するAd35アダプタープラスミドであるが、そうでなければ、A35アダプタープラスミドpAdApt35IP1(WO 00/70071を参照のこと)と同一である。その構築を以下に記載する。
上述のようなPER55Kクローン16細胞において、アダプタープラスミド及びAd35ベクターの骨格コスミドをトランスフェクトすることにより組換えウイルスを生成した。この点に関して以下のセットのプラスミドを使用した。

上記で示すように、コスミドpWE.Ad35.pIX−rITRにおけるアデノウイルスインサートは、その5’末端でAd35のpIXプロモーターを含有する。このことは、pAdApt535に基づいたアダプタープラスミドで生成されたウイルスへのAd35pIXプロモータの再挿入を招く可能性があった。従って、pIXプロモーター配列を欠く、Ad35骨格コスミドの新しい種類を作製する。この点に関して、PIXcosF−2及びAdapt35−3のプライマーでPCR断片を生成した。各プライマー(100μMのストック)3μl、2mMのdNTP混合物5μl、10x完全緩衝液(Mg2+を含む)5μl、DMSO1.5μl、Pwo酵素0.5μl及び10ngのpAdApt35IP1鋳型を含有する50μl容積において、PwoDNAポリメラーゼ(2.5単位/μl、ゲナキシス)による増幅を行った。PCRプログラムは、94℃にて2分間、その後、(94℃で30秒間、58℃で30秒間及び72℃で1.5分間)を5サイクル、次いで(94℃で30秒間、60℃で30秒間及び72℃で1.5分間)を25サイクルに設定し、68℃で8分間の最終工程で終了した。得られた1.2kbのPCR産物は、5’末端に結合したAatII部位及びNotI部位を持つヌクレオチド3481〜ヌクレオチド4663(WO 00/70071に公開されているAd35の配列に基づいた番号付け)由来のAd35配列を含有する。製造元の指示書に従って、PCR精製キット(キアゲン)を用いてPCR産物を精製し、製造元の指示書に従って、pPCR−ScriptAmpベクター(ストラタジーン)にクローニングした。次いで配列決定によってクローニングした断片の配列を確認し、AatII及びAgeIによる消化によってコンストラクトから取り出した。製造元の指示書に従って、ジーンクリーンスピンキット(バイオ101社)を用いて、ゲルから、得られた780bpの断片を精製する。
コンストラクトpIG35.55Kは、ヒトのホスホグリセリン酸キナーゼプロモーター(hPGK)に操作可能に接続されたAd35のE1B−55K遺伝子のコーディング配列及びHBVのポリアデニル化配列を含有する。加えて、それは、RSVプロモーター及びHBVpAに操作可能に接続されたネオマイシン耐性遺伝子を含有する。pIG35.55Kの構築は以下のとおりである。
pIX遺伝子の発現を促進する異種プロモーターの例として、RSVプロモーターを、LacZ又はルシフェラーゼのリポーター遺伝子を含有するAd35のアダプタープラスミドに挿入した。RSVプロモーターは、pRc−RSV(インビトロゲン)から入手可能なNruI/ApaLI断片に相当する。製造元の指示書に従って、クレノウ酵素(ニューイングランドバイオラボズ)を用いて突出末端を埋めた。QIAクイック・ゲルエクストラクションキット(キアゲン)を用いて、RSVプロモーターを含有する388bpの断片をアガロースゲルから単離した。アダプタープラスミドpAdApt35.Luc及びpAdApt35.LacZをBglIIで直鎖状にし、その後、クレノウ処理により末端を平滑化した。BglIIは、導入遺伝子発現カセットのSV40ポリアデニル化配列のすぐ後ろで消化する。pAdApt35を基にしたアダプタープラスミドの記載についてはWO 00/70071を参照のこと。次いで、製造元(ロシュ)の指示書に従って、シュリンクアルカリホスファターゼ(SAP)を用いて、処理されたアダプタープラスミドを脱リン酸化した。次いで、単離したRSVプロモーターを処理したベクターのそれぞれに連結し、DH5αコンピテント細菌(インビトロゲン)を形質転換した。pIX遺伝子に対して正方向に挿入されたRSVプロモーターについてコロニーを解析し、pAdApt35.Luc.rsv及びpAdApt35.LacZ.rsvを生じた。
前の実施例は、E1A及びE1Bをコードする領域が完全に取り除かれているA35組換えアデノウイルスは、ゲノムサイズが増すと、漸次より不安定になることを示している。我々はこの応用において、pIXの発現を駆動する異種のプロモータを添加することによって不安定性を克服できることを示す。WO 02/40665及び上記において、我々は、完全なE1B−55Kのコーディング配列を保持するAd35ウイルスをPER.C6細胞で産生されることができ、安定であることを開示した。完全なE1Bコーディング配列を保持するウイルスについても同じことが真実である(WO 00/70071;Abrahamsen et al., 1997)。共にこれらの結果は、サブグループCにおけるpIX遺伝子に比べて、サブグループBのウイルスではpIX遺伝子の発現が異なって調節されている可能性を生じる。E1Bプロモーターにより駆動されるE1B−55K遺伝子を保持するウイルスは安定なので(上記を参照のこと)、pIX制御配列はおそらくこの領域に位置する。これを調べるために、我々は、55K配列の様々な長さの3’末端を保持する一連のコンストラクトを生成する。この点に関して、pBr.Ad35.ΔAM.AdAptLacZが以下のように先ず生成される。SnaBI及びMfeIによりコンストラクトpBr.Ad35.lITR−pIX(WO 00/70071に記載)を消化し、クレノウで平滑化し、SAF酵素(ロシュ)により脱リン酸化する。次いで、4.4kbのベクター断片をアガロースゲルから単離する。コンストラクトpAdApt.LacZ(LacZ導入遺伝子インサートを持つAd5を基にしたアダプタープラスミドpAdApt;WO99/55132)を、AvrII及びBglII(及び断片サイズの差異を高めるために、必要に応じてSalI)により消化し、クレノウ酵素で平滑化する。次いで、4.2kbのCMV.LacZ.pAインサートをゲルから単離する。次いで、単離した双方の断片を連結してpBr.Ad35ΔSM.AdAptLacZを得る(図7)。正しい方向で連結するとAvrII部位及びMfeI部位の双方が復元するので、制限消化によって配向性をチェックすることができる。コンストラクトpBr.Ad35ΔSM.AdAptLacZは、pIX遺伝子に対して野生型の位置に、0.6kbの3’E1B−55K配列(wt Ad35はヌクレオチド2804〜3400を含む)を保持する。以前我々は、PER.C6細胞における増殖が可能であるとは思われなかったので、これらの55K配列は、機能を有するE1B−55Kタンパク質の発現へ至らないことを示した(pBr.Ad35ΔSM;WO 02/40665)。pBr.Ad35ΔSM.AdAptLacZから出発して、680bp(0.7kb)のE1B−55K領域の様々な欠失を作製することができ、それはそれぞれ、MfeI(MunIのアイソシゾマー)及びStuI、NsiI又はBglIIのいずれかによって消化し、続いて、クレノウを用いて突出末端を平滑化することにより、または5’又は3’の突出の場合はT4DNAポリメラーゼを用いて平滑化することにより行うことができる。消化したDNAの再連結により機能を有するアダプタプラスミドが与えられ、その後それは上述のように、pWE.Ad35.pIX−rITRと共にコトランスフェクションすることにより、PER55K細胞で組換えウイルスを生成するのに使用される。酵素DraIII、Bsu36I、BssHII又はBamHIを用いて、本分野で公知の方法を用いて部分的にベクターを消化し(LacZ遺伝子もこれら酵素の認識部位を含有する)、次いで正しいクローンを選抜することにより、追加のコンストラクトを作製する。上述のように、Ad35.AdApt.LacZ.rsvコンストラクトについて安定性を調べる。安定である(すなわち、導入遺伝子領域において欠失を獲得しない)コンストラクトは、pIXの発現に対する適切な制御領域を含有する。さらに、リポーター遺伝子の上流に配列を挿入することにより所定の配列におけるプロモーター活性を直接調べることができる。pGL3ベーシック(プロメガ)はそのようなリポーター遺伝子のコンストラクトである。MunIとpIX遺伝子の開始部位の間に領域を、プライマーセットAd3555KmfeFとAd35pIXNcoRを用いて増幅した。このPCR[94℃で2分間、次いで(94℃で30秒間、59℃で30秒間、72℃で60秒間)を30サイクル、次いで68℃で8分間;酵素:追加の3%DMSOと共に、製造元の指示書に従ってPwo(ジェネキシス)]は、2804〜3491(wt Ad35におけるような番号付け)のAd35配列を増幅し、それによってpIXの開始コドン付近の配列をNcoI部位に変え、5’末端にHindIII部位を挿入した。この増幅した断片をHindIII及びNocIで消化し、同じ酵素で消化したpGL3ベーシックにクローニングし、pGL3−MNを生成した。次いで、pGL3−MNを用いてルシフェラーゼのコーディング領域の上流の配列を欠失させ、それは、HindIIIの消化と、たとえば、PacI、NsiI、StuI、Bsu36I、BssHII又はBglIIの消化を組み合わせて、続いて突出末端を平滑化し、再連結することによって行う。製造元の指示書に従って、リポフェクトアミン試薬を用いて、PER.C6細胞に得られたコンストラクトの一時的なトランスフェクションすることにより、プロモーター活性を調べる。製造元の指示書に従って、ステディ−Gloルシフェラーゼアッセイシステム(プロメガ)を用いて、トランスフェクションの2日後にルシフェラーゼ活性を解析する。別の方法では、pGL3ベーシックベクターに異なった領域が挿入され、その領域はAd35のE1B−55領域中の特定の配列に向けられ、Nco−pIXrevプライマーと組み合わされた5’末端に結合したHindIII部位を有する5’(正方向)プライマーを用いたPCR増幅によって生成される。この方法では、クローニングのための特異な制限部位が存在することによって限定されない。
上記実施例は、例としてAd35ウイルスを使用する。サブグル−プBにおけるその他のメンバーは互いにかなりの相同性を有し、匹敵するpIXの調節を有することがある。
アデノウイルス抗原型35ゲノムの配列決定のみならず、プラスミドを基にしたベクター系の構築及びAd35を基にした組換えウイルスの生成は、WO 00/70071に詳細に記載されている。
プライマーDF35−1及び35FRを用いて第1のPCR断片を増幅した。追加のDMSO(シグマ、最終濃度3%)と共にPwoDNAポリメラーゼ(ロシュ)を用いて鋳型DNAとしてのpWE.Ad35.pIX−rITR(WO 00/70071を参照のこと)により増幅を行った。プログラムは以下のとおりであった:94℃で2分間、続いて(94℃で30秒間、52℃で30秒間及び72℃で3分間)を30サイクル、及び確実に完全な断片にするための72℃で8分間の最終工程である。増幅により、Ad35の配列のヌクレオチド30224からヌクレオチド31805に相当する1.6kbの断片を生じた。BamHI部位を3’末端に導入した。ジーンクリーンキットを用いて増幅したDNAをゲルから精製し、pCRScript/Ampクローニングベクターキット(ストラタジーン)に連結した。エレクトロコンピーテントなDH10B細胞を形質転換した後、さらなる解析のために白色のコロニーを選択した。これにより、コンストラクトpCR−fiber35を生じた。平滑クローニングのために、PCR断片を2方向で挿入できた。5’末端にてpCRScript/AmpベクターのポリリンカーでBamHI部位を持つインサートを有するクローンを選択した。従って、BamHIによる消化は1.6kbの断片を生じる。配列決定によって、PCR断片の正しい増幅が裏付けられた。プライマー5E4F及び5E4Rを用いて第2のPCR断片を増幅した。pWE.Ad5.Af1II−rITR(ECACC寄託番号P97082116、WO 02/40665に記載)中の余分のPacI部位を含有するコスミドベクターである、pWE.Ad5.Af1II−rITRspにより増幅を行った。上記のようなPwoDNAポリメラーゼを用いて、pWE.Ad5.Af1II−rITRspは鋳型として作用したが、pWE.Ad5.Af1II−rITRは同じ目的で使用することもできた。ゲルからの精製の後、SstI及びBamHI(PCR中に導入された双方の部位)でDNAを消化し、ジーンクリーンキットを用いてアガロースゲルから3kbの断片を精製した。増幅されたAd5のE4領域は、Ad5の配列の塩基対32794から塩基対35828に相当する。プライマー:355ITR及び353ITRを用いて、pWE.Ad35.pIX−rITR上で第3のPCR断片を生成した。上述のようにPCR増幅を行った。得られる160bpの断片には、SstI部位(5’末端)及びEcoRI部位(3’末端)が隣接する。上記のようにゲルから精製した後、SstIとEcoRIでDNAを消化した。次いでAd35の右ITRに相当する160bpの断片を低融点アガロースゲル上で消化した末端から分離し、ゲル中に回収した。次に、BamHIとEcoRIでpUC119を消化し、ジーンクリーンキットを用いてゲルから3.1kbの断片を精製した。次いで、上記で処理した第2のPCR断片及び第3のPCR断片をBamHI/EcoRIで消化したpUC119と連結して、pUC.Ad5E4−35ITRを生じた。クローニングしたPCR由来のインサートを配列決定して正しい増幅を確認した。次にpCR−fiber35における1.6kbのインサートをBamHIで切り取り、上記のようにゲルから断片を精製した。pUC.Ad5E4−35ITRもBamHIで消化し、直鎖状の断片をゲルから精製した。双方の断片を連結し、互いに対して正しい方向を有するクローンを選択することによって、pUC.35−5E4を生じた(図14)。pUC.35−5E4の構築を導く工程を図13に模式的に表す。pUC.35−5E4中のアデノウイルスインサートを、Ad35の3’配列を有するpBr.Ad35.PRn(図15;WO 00/70071を参照のこと)中にサブクローニングした。この点に関して、コンストラクトpUC.35−5E4をMluIとNotIで消化し、ジーンクリーンキットを用いて4.7kbの断片をゲルから精製する。次いで、コンストラクトpBr.Ad35.PRnのMluI及びNotIによる消化により生じる断片に、この断片を連結する。アガラーゼ酵素(ロシュ)を用いて、この16.3kbの断片をゲルから精製した。次いで、連結物でコンピテントDH10B細胞を形質転換した。得られたコンストラクトを、pBr.Ad35.PR5E4(図16、ECACC寄託番号P02041229)と命名した。最後の工程は、ウイルスコスミドクローンpWE.Ad35.pIX−rITRへの、Ad35配列の修飾された3’末端のクローニングを伴う。この点に関して、製造元の指示書に従って、λファージパッケージング反応(ストラタジーン)において2つの断片を組み合わせる。第1は、PacIとSwaIによる消化によって得られるpBr.Ad35.PR5E4からの16.8kbの修飾されたA35のインサートであり、第2は、PacIとSwaIによるpWE.Ad35.pIX−rITRの消化によって得られる22.8kbの断片である。2つの断片の正しい組み合わせによって、pWE.Ad35.pIX−rITR5E4(図17)が得られる。従って、このコンストラクトにおいて、Ad35の骨格におけるE4領域は、Ad5に由来する相当する領域で置き換えられる。
pIX遺伝子(Ad35の配列のヌクレオチド3401)から右ITRの末端を含有するが、Ad5の相当する配列について変換されているE4−orf6及びE4−orf6/7を持つアデノウイルスの骨格コンストラクトを得るために、以下に記載するように、Ad35及びAd5をPCRで増幅し、組み合わせた。3%までのDMSOを加えて、PwoDNAポリメラーゼによってPCR断片を生成した。第1のPCRは、pBr.Ad35.PRn(図15;WO 00/70071を参照のこと)を鋳型として、プライマーE4−F1及びE4−R2を用いて行った。プログラムは、94℃にて2分間、(94℃で30秒間、50℃で30秒間及び72℃で1分間)を5サイクル、次いで(94℃で30秒間、60℃で30秒間及び72℃で1分間)を30サイクルに設定し、68℃で8分間の最終工程で終了した。ジーンクリーンキットを用いて、得られた1.8kbの断片を精製した。第2のPCRは、pWE.Ad5.Af1II−rITR(ECACC供託番号P97082116、WO 02/40665に記載)におけるPacI部位を含有するコスミドベクターである、鋳型としてのpWE.Ad5.Af1II−rITRspならびにプライマーのE4−F3及びE4−R4により行った。プログラムは以下のように:94℃にて2分間、次いで(94℃で30秒間、62℃で30秒間及び72℃で1分間)を30サイクルに設定し、68℃で8分間の最終工程で終了した。1.1kbの断片を上記のように精製した。第3のPCRは、鋳型としてのpBr.Ad35.PRnならびにプライマーのE4−F5及びE4−R6により行った。プログラムは以下のように:94℃にて2分間、(94℃で30秒間、48℃で30秒間及び72℃で45秒間)を5サイクル、次いで(94℃で30秒間、56℃で30秒間及び72℃で45秒間)を30サイクルに設定し、68℃で8分間の最終工程で終了した。上記のように366bpの断片を精製した。精製した断片の試料をゲルにかけて濃度を推定し、次いで、断片を一緒に混合し、合計30μl中に700ngのPCR−1、650ngのPCR−2及び430ngのPCR−3を含むようにした。この混合物に、EcoPol緩衝液(ニューイングランドバイオラボズ)3μl、2mMのdNTP溶液3μl及びミリQ水3μlを加えた。得られた混合物を94℃で3分間インキュベートし、PCR機にて0.5℃/秒の割合で65℃に冷却した。65℃で10分間のインキュベートに続いて、0.05℃/秒の割合で20℃まで、混合物をさらに冷却し、20℃にて10分間インキュベートした。次いで、1μl(5単位)のクレノウ酵素(ニューイングランドバイオラボズ)を加え、続いて37℃にて60分間インキュベートした。このクレノウ混合物5μlを鋳型として用いて、以下のように2つの断片を別途増幅した。最終濃度3%にDMSOを加えたPwoDNAポリメラーゼ(ロシュ)を用いた反応において、プライマーセット1:NF−1及びNcoI−Rを使用し、PCR機の以下の設定:94℃にて2分間、次いで(94℃で30秒間、66℃で30秒間及び72℃で3分間)を30サイクル、続いて68℃で8分間の最終工程を使用した。最終濃度3%にDMSOを加えたPwoDNAポリメラーゼ(ロシュ)を用いた反応において、プライマーセット2:NcoI−F及びNR−2を使用し、PCR機の以下の設定:94℃にて2分間、次いで(94℃で30秒間、62℃で30秒間及び72℃で90秒間)を30サイクル、続いて68℃で8分間の最終工程を使用した。ジーンクリーンキットを用いて、2.7kb(プライマーセット1)及び1.1kb(プライマーセット2)の得られた断片をゲルから精製し、それぞれをpCRscriptAmpベクター(ストラタジーン)に連結し、エレクトロコンピーテントなDH10B細胞を形質転換した。これにより、コンストラクトpCRscriptAmp.NFI−NcoIR(図18)及びコンストラクトpCRscriptAmp.NcoIF−NR2(図19)が得られた。インサートは平滑末端を含有していたので、各クローニングについて2つの配向性が得られた。KpnI消化を用いて、さらなるクローニングに必要な配向性を持つコンストラクトを選択した(図18及び19を参照のこと)。次いで、インサートを配列決定して正しい増幅を確認した。次に、BamHIとNcoIを用いて、pCRscriptAmp.NcoIF−NR2のインサートの一部を切り出し、上記のようにゲルから精製した。同じ酵素でpCRscriptAmp.NFI−NcoIRを消化し、断片を含有するベクターをゲルから精製した。これらの断片を連結してpCR.NF1−NR2(図20)を得た。pCR.NF1−NR2は、ジェンバンクにおいて公開されているAd5の配列(受入番号M73260)の32968から34077の間に位置する配列に由来するAd5について置き換えられたヌクレオチド31879〜ヌクレオチド32974の間のE4−orf6及びE4−orf6/7の配列を持つAd35の配列のヌクレオチド30162からヌクレオチド33234の間のAd35の配列を含有する。従って、図21及び22に提示されるアミノ酸アラインメントに見ることができるように、クローニングしたE4−orf6タンパク質のアミノ酸配列は、Ad5に見い出されるE4−orf6の配列(配列番号61;Ad35のE4−orf6のアミノ酸配列は配列番号62)と同一であり、E4−orf6/7のアミノ酸配列の大部分は、Ad5中に存在するE4−orf6/7と同一である(E4−orf6/7の配列は、Ad5については配列番号63、Ad35については配列番号64、クローニングした融合タンパク質については配列番号65)。明らかに、本発明から逸脱することなく、上記で概説した一般的な方法を用いて、様々なハイブリッドAd35−Ad5のE4コンストラクトを設計することができる。次いでpCR.NF−NR−2に由来するこのキメラインサートをpWE.Ad35.pIX−rITRにクローニングし:pCR.NF−NR−2をMluIとNdeIで消化し、ジーンクリーンキットを用いて、得られた2.8kbの断片をゲルから精製した。コンストラクトpBr.Ad35.PRnもMluIとNdeIで消化し、アガラーゼ酵素(ロシュ)を用いて18kbのベクター断片をゲルから単離した。双方の断片の連結によりコンストラクトpBr.Ad35.PR.5Orf6(図23、ECACC寄託番号P02041227)を生じた。次いで、上記のようなλファージパッケージングを用いて、このコンストラクト中におけるキメラE4領域を含有するPacIおよびSwaIの間のA35配列を、コンストラクトpWE.Ad35.pIX−rITRにクローニングする。次いで、得られるpWE.Ad35pIX−rITR.5Orf6(図24)を用いて、A35アダプタープラスミドと共に、PER.C6パッケージング細胞上にコトランスフェクションすることにより、Ad35を基にした組換えウイルスを生成する。
E3配列の欠失によりad35の骨格をさらに改変した。E3タンパク質は、アデノウイルス感染に対する宿主免疫応答を調節することが知られているので、組換えウイルスの生体外増殖には必要ではない。さらに、E3配列の欠失により、パッケージング効率を落とすことなく、さらに大きな異種の配列を挿入することができる。また、ワクチン用ベヒクルとしてのアデノウイルスベクターの応用については、E3にコードされる免疫調節性遺伝子の発現は好ましくない。
pBr.Ad35.PRnを基にしたコンストラクトを用いた相補細胞株であるPER.C6において組換えAd35ウイルスの生成を可能にするために、我々は、先ず、Ad35配列(WO 00/70071)を、Ad35の配列の塩基対3401から塩基対24650まで、従って、アダプタープラスミド及びpBr.Ad35.PRnを基にしたコンストラクトの双方との重なり合いを含有する新しいコンストラクトを作製した。これら3つのプラスミドのPER.C6細胞へのトランスフェクション及び二重相同組換え事象によって、図18に概略するように、完全なウイルスゲノム及び組換えウイルスの複製がもたらされる。必要なプラスミドは、pWE.Ad35.pIX−rITRにおけるAd35配列の大部分の欠失によって作製した。この点に関して、pWE.Ad35.pIX−rITRをEcoRVで消化し、ジーンクリーンスピンキットを用いて、29kbのベクターを含有する断片を低融点ゲルから精製した。精製したDNAを自己連結し、電気的にエレクトロコンピーテントなDH10B細菌(インビトロゲン/LTI)を形質転換するのに用い、pWE.Ad35.pIX−EcoRV(図29)を生じた。
完全にE1を欠失したウイルスにおけるpIX遺伝子を活性化する異種プロモーターの配列の効果を調べるために、一連のアダプタープラスミドを用いて組換えAd35ウイルスを生成した。この点に関して、pAdApt35LacZ、pAdApt35.LacZ.rsv(実施例9)、pAdApt535.LacZ(実施例5)及びpAdApt35BLacZ(pIX遺伝子の前でAd35のE1Bプロモーター配列を含有する;以下に記載)をpIPsp−Iで消化し、実施例2(及びWO 00/70071)に記載するように、NotIで消化したコスミドpWE/Ad35−3481及びpWE/Ad35−3481ΔE3(実施例7)を持つウイルスを生成するのに使用した。さらにアダプタープラスミドpBr.Ad35ΔSM.AdAptLacZ(図7;実施例10)によりウイルスを生成した。このアダプタープラスミドは、E1A配列及びE1B配列の大部分を欠失する。それは3’E1B−55K配列の0.6kbを保持し、55Kの停止コドンとpIXの開始コドンとの間の野生型配列も有する。
上に記載された結果に基づいて、我々は、サブグループBのウイルスにおけるpIXプロモーターは、E1B−55Kのコーディング領域に位置するであろうと予測した。これを直接調べるために、我々は、実施例10に記載されるように、pIXのmRNAのキャップ部位を同定しようと計画した。この点に関して、野生型Ad35、野生型Ad11及びA35.E1B+.AdApt.Lucのウイルスを用いて、50VP/細胞のMOIにてPER55Kクローン16細胞を感染させた。このウイルスのプロモータ−及びmRNA開始部位が分かっているので、対照として、野生型Ad5を共に用いた。製造元により記載されているように、TRIzol試薬(インビトロゲン)を用いて、感染後16〜18時間での感染培養物からRNAを単離した。手順の終わりに、100%ホルムアミド中に単離したRNAを保存した。転写開始の位置を決定するために、ジーンレーサーキット(インビトロゲン)を用いて、pIX転写物の5’末端を増幅した。ジーンレーサーのプロトコールを開始する前に、ジーンレーサーのプロトコールに記載されるように酢酸ナトリウム沈殿によって5μgのRNAをホルムアミドから精製した。pIXmRNAの5’末端を増幅するための製造元のプロトコールに従って、精製されたRNAを処理した。ホスファターゼ処理、それに続くタバコ酸性ピロホスファターゼによるキャップ構造の除去及びジーンレーサーRNAオリゴの連結の後、cDNAの合成にキットのSuperScriptTMII逆転写酵素を用いた。pIXのための遺伝子特異的な(逆)プライマーを用いた逆転写によってcDNAを合成した。Ad35(野生型及びE1B+.Lucウイルス)及び野生型Ad11についてはプライマーpIXrevを用い、Ad5については、プライマーpIXrev−Ad5を用いた。得られたcDNA(未知の濃度で1μl)をPCRの鋳型として用い、dsDNAを生成した。このPCRは、製造元の指示書に従ってPwoDNAポリメラーゼ(ロシュ)を用い、DMSO(シグマ;3%v/v)を添加して行った。mRNAの5’末端に連結されるオリゴヌクレオチドに特異的なキット由来のジーンレーサー5’プライマー、及び上述のような遺伝子特異的な逆プライマーによって増幅を行った。反応条件は以下のとおりであった:94℃で2分間の変性、続いて、(94℃で30秒間、60℃で30秒間及び72℃で2分間)を30サイクル、及び68℃で8分間の伸長で終了した。1.0%のアガロースゲル上での電気泳動によって得られたDNA断片のサイズ分離を行った。Ad5については480bpの断片が得られ、Ad35(両方のウイルス)及びAd11については、200bpの断片が得られた。Ad11は2kbの断片も示した。断片をすべて切り出し、アガロースゲルから精製した。精製したDNA断片をpCR4Blunt−TOPO(登録商標)ベクター(インビトロゲン)にクローニングした。市販のM13正方向プライマー及び逆方向プライマーを用いてベクターの配列決定を行った。野性型配列に対して得られた配列を並べ、pIXの転写開始の位置を決定した。Ad35及びAd11のcDNA調製物から単離された200bpのバンドは、本物のpIXmRNAを構成しており、Ad11から単離された2kbの断片はE1Bプロモーターをもたらすことが判明した。図30は、野生型のAd35の配列(配列番号60)と共にAd35(配列番号59)及びAd11(配列番号58)のcDNAの配列を示す。配置は、pIXmRNAにおいてスプライシングされたイントロンの配列の位置を示す。図31において、同定されたキャップ部位及びスプライシング部位の位置がAd35のE1B−pIX領域に模式的に示されている。Ad5については、予想されるpIXmRNA(Babiss and Vales, 1991)は、3580位(示さず;ジェンバンク受入番号M73260の番号付け)に位置するキャップ部位と共に同定された。Ad35については、キャップ部位は、A残基における3339位(Ad35の配列、WO 00/70071)におけるE1B−55K遺伝子の3’末端に位置していた。Ad11では、キャップ部位は、同様にT残基で見い出された(ジェンバンク受入番号AY163756の3339位)。興味深いことに、55K遺伝子の停止コドンとpIXの開始コドンの間の配列には、Ad5ウイルスにおいてpIXのプロモーターが位置しているが、その配列は、Ad35ウイルス及びAd11ウイルスでスプライシングを受けている。これらの結果は、Ad35及びAd11では、pIX遺伝子の発現は、55K遺伝子の3’末端に位置するプロモーターに調節されているという強い証拠を提供している。
pIXmRNAのキャップ部位の同定と共に、正しいpIX発現のために天然のAd35プロモーターを包含することも、ウイルスベクターにおいてE1B−55Kの配列をできるだけ多く限定することも可能になる。ここで、我々は、非限定例として、55Kコーディング配列の3’末端の166bpを保持するAd35のアダプタープラスミド(pAdApt35Bsu.Luc)の構築、及び安定性及び/又はパッケージング容量を高めたE1欠失のAd35Lucウイルスの生成を示す。この166bpの配列は、機能的な55K遺伝子産物をコードしているのではないが、pIXのコーディング配列に対するその天然の位置において、前の実施例で同定したpIXmRNAのキャップ部位を含有する。
表V:実施例において述べたようにPER.C6細胞上でE1-欠失したAd35に基づいたウイルスを作製するのに使用したコンストラクトのリスト。アダプタープラスミドをPacIで消化し、pWE.Ad35.pIX-EcoRVはNotIとEcoRVで消化し、E4-改変したpBrに基づいたコンストラクトはPacIとNotIで消化した。
Claims (11)
- E1領域において欠失を持つ組換え型サブグループBアデノウイルスであり、およびそのアデノウイルスは機能的なE1B55K遺伝子生成物をコード化しないものであって、次の
(i)E1B55Kコード配列の停止コドンおよびpIXコード配列の開始コドンの間に位置付けられるアデノウイルス配列、および
(ii)プロモーター活性を持つ配列の調節下の機能的なpIXコード配列であり、プロモーター活性を持つ前記配列は、pIX開始コドンの直接的に上流にあるアデノウイルス配列の0.7kbまでから構成され、前記配列は、E1B55Kコード配列の終わりの166bp内に存在する、プロモーター活性のために最小限に要求される配列を備えるもの
を具える、組換え型アデノウイルス。 - プロモーター活性を持つ前記配列は、pIXコード配列の直接的に上流にあるアデノウイルス配列の600ヌクレオチドまでを含む、請求項1記載の組換え型アデノウイルス。
- プロモーター活性を持つ前記配列は、pIXコード配列の直接的に上流にあるアデノウイルス配列の300ヌクレオチドまでを含む、請求項1記載の組換え型アデノウイルス。
- プロモーター活性を持つ前記配列は、pIXコード配列の直接的に上流にあるアデノウイルス配列の250ヌクレオチドまでを含む、請求項1記載の組換え型アデノウイルス。
- 前記アデノウイルスは、アデノウイルス血清型35またはアデノウイルス血清型11から導かれる、請求項1〜4のいずれか1項記載の組換え型アデノウイルス。
- さらに、サブグループCのアデノウイルスから導かれる機能的なE4−orf6タンパク質をコード化する配列を具えることを特徴とする、請求項1〜5のいずれか1項記載の組換え型アデノウイルス。
- E1領域において欠失を持つ組換え型サブグループBアデノウイルスの安定性および/またはパッケージング容量を増加させるための方法であって、パッケージング細胞において前記組換え型アデノウイルスのウイルス粒子への生成および組立てのために必要な要素を発現させる工程を具え、次の要素
(i)E1B55Kコード配列の停止コドンおよびpIXコード配列の開始コドンの間に位置付けられるアデノウイルス配列、および
(ii)プロモーター活性を持つ配列の調節下の機能的なpIXコード配列であり、プロモーター活性を持つ前記配列は、pIX開始コドンの直接的に上流にあるアデノウイルス配列の0.7kbまでから構成され、前記配列は、E1B55Kコード配列の終わりの166bp内に存在する、プロモーター活性のために最小限に要求される配列を備えるもの
が前記アデノウイルスにおいて保有され、または再導入される
ことを特徴とする、方法。 - プロモーター活性を持つ前記配列は、pIXコード配列の直接的に上流にあるアデノウイルス配列の600ヌクレオチドまでを含む、請求項7記載の方法。
