Description

【発明の詳細な説明】 〔産業上の利用分野〕 本発明はイブプロフェンを含有する薬剤学的製剤および
その製造法に関する。
〔従来技術〕
口腔及び咽頭腔内で強く粘膜を刺激する酸から成る作用
物質、又はこの部分を強く刺激する酸基を含む作用物質
が存在することは公知であり、この種の更に難溶性又は
不溶性の作用物質にイブプロフェンがある。
イブプロフェンはりウマチ及び関節炎治療用の作用物質
として益々重要視されるようになった。
この物質は水に不溶であるが極めて不快な味を有し、又
おそらく化合物２−（４−イソブチルフェニル）プロピ
オン酸の存在により特に食道の粘膜を刺激する。同様に
文献で公知のように、イブプロフェンは、アセチルサリ
チル酸に僚で、この物質が錠剤の形であれカプセルの形
であれ高濃度で胃壁に達した場合は特に、胃腸の出血を
引き起すおそれがある。従って胃腸内で局部的な高濃度
を生ずることのないようにこの物質を水に懸濁させてか
ら服用させるのが治療上有利である。しかしこの水に懸
濁させた液は、その作用物質の一部が口や食道に残留し
、そこでひりひり刺激するような感覚を与えるので、何
か適当な対策をとらない限り味覚の点から服用不能であ
る。
〔発明が解決しようとする問題点〕 本発明の目的は、前述した味覚上のマイナスの効果を除
去し、イブプロフェンを含有する薬剤の経口服用を問題
な（実施できるようにする、冒頭に述べた種類の薬剤学
的製剤右よびその製造法を提供することにある。
〔問題点を解決するための手段〕
本発明によれば、前述の種類の薬５剤学的製剤に於ける
上記の目的は、イブプロフェン粒子を少くとも一種のプ
ソイドコロイド及びフマル酸から成る外被で被覆するこ
とにより達成される。
このプソイドコロイドはキサンテン及び／又はマルトデ
キストリンを含有するものとすることができる。
更に本発明による薬剤学的製剤は場合によっては発泡性
混合物を含有することを特徴とする。
この発泡性混合物としては、クエン酸及び炭酸カルシウ
ムを含有するものとすることができ−その際炭酸カルシ
ウムは、炭酸カルシウムとクエン酸の結晶の表面に近い
層との反応により生成した接着層の接着作用により、ク
エン酸を被覆する。
本発明による種類の薬剤学的製剤の製造法は、本発明に
よればイブプロフェン粒子を真空混合機の中で真空下で
少くとも一種のプソイドコロイドとフマル酸とから成る
外被で被覆し、次いで真空乾燥することを特徴とする。
この場合特に、先ずイブプロフェンとプソイドコロイド
又はプソイドコロイドと水とを真空混合機の中で約０．
１バールの真空下で混合し、次に約０．２バールで乾燥
した後でフマル酸を添加し、次いで完全に乾燥するよう
にすることができる。
本発明は、従来のイブプロフェン製剤の味覚上のマイナ
ス効果をイブプロフェンをプソイドコロイドとフマル酸
で被覆することにより除去できるという知見に基づくも
ので、イブプロフェン結晶をプソイドコロイドで、それ
より低いｐＨ−値を有するフマル酸の存在下で被覆すれ
ば、この味覚上のマイナス効果を防止できる。ここでプ
ソイドコロイドは水に不溶性のイブプロフェンと難溶性
のフマル酸とを固定する機能を果すだけである。
本発明による薬剤学的製剤の！！Ｉｉ造乃至本発明によ
る方法の実施を、真空混合機を用いへ西独公＠特許公報
Ｐ３４３４７７ｔ７号の主題をなす方法によって行なう
のが、本目的のみならずその鉋の点でも有利である。こ
の点に関する本発明思想の補足的説明には前記西独公ａ
特許公報が全面的に引用される。
次に実施例により本発明の詳細な説明する。
実施例　ｌ イフフロフェン２００部を、キサンテンｌＯ謬とマルト
デキスト９７１０部とを５０ｓの水に溶かした溶液で処
理し、真空中で完全に乾燥する前に、約２００ミリバー
ルで７マル酸２０部を加える。完全に乾燥すると粒状物
が得られ、これを粒径約（１３關に粉砕する。
この粒子を瞬間技法（ｉｎｓｔａｎｔａｎｅｏｕｓ　Ｌ
ｅｃｋｉｏｉｏｌｃｙ）により砂糖の結晶に付着させる
ことがてき、この場合好ましくは１０−１５倍の量の砂
糖を使用し、又２倍量のクエン酸を加えてもよい。
−回置２０■のイブプロフェンのこの種の混合物を作れ
ば、適当に香料を添加することにより飲み易い飲料が得
られ、これは粘膜に何等の刺激を与えることがない。
去範炭−１ 前述と同じシステムは、これにクエン酸と炭酸カルシウ
ムとの発泡性混合物を追加することにより更に改良でき
る。
クエン酸５０部と炭酸カルシウム５０部とに５０％エタ
ノール５部を加えて真空下で反応させる。
この発泡性混合物を乾燥し、実施例１の方法で処理した
イブプロフェンに５倍量加える。
こうして作った調合物は緩やかに泡を発生する混合物に
よる自己懸濁効果を示し、この際存在するカルシウムイ
オンは明らかにイブプロフェンの味覚を更に改善させる
効果がある。

Claims (7)

    【特許請求の範囲】
  1. （１）イブプロフェン粒子を少くとも一種のプソイドコ
    ロイドおよびフマル酸から成る外被で被覆することを特
    徴とする、イブプロフェンを含有する薬剤学的製剤。
  2. （２）プソイドコロイドがキサンテンを含有することを
    特徴とする特許請求の範囲第１項記載の製剤。
  3. （３）プソイドコロイドがマルトデキストリンを含有す
    ることを特徴とする特許請求の範囲第１項又は第２項記
    載の製剤。
  4. （４）一種の発泡性混合物を含有することを特徴とする
    特許請求の範囲第１乃至３項の何れかに記載の製剤。
  5. （５）発泡性混合物がクエン酸と炭酸カルシウムを含有
    し、その際炭酸カルシウムは、炭酸カルシウムとクエン
    酸の結晶の表面に近い層との反応により生成した接着層
    の接着作用によりクエン酸を被覆することを特徴とする
    特許請求の範囲第４項記載の製剤。
  6. （６）イブプロフェン粒子を真空混合機の中で真空下で
    、少くとも一種のプソイドコロイドとフマル酸とから成
    る外被で被覆し、次いで真空乾燥することを特徴とする
    薬剤学的製剤の製造法。
  7. （７）先ずイブプロフェンとプソイドコロイド類と水と
    を真空混合機の中で約０．１バールの真空下で混合し、
    次に約０．２バールで乾燥した後でフマル酸を添加し、
    次いで完全に乾燥することを特徴とする特許請求の範囲
    第６項記載の方法。
