Claims (1)
- 【特許請求の範囲】 1.哺乳動物細胞で通常発現する蛋白質をコードする合成遺伝子であって、該 哺乳動物蛋白質をコードする天然の遺伝子における好ましくない、または比較的 好ましくない少なくとも1個のコドンを、同じアミノ酸をコードする好ましいコ ドンに置き換えた合成遺伝子。 2.同一条件下のインビトロ哺乳動物細胞培養系において天然遺伝子によって 発現されるレベルの少なくとも110%のレベルで哺乳動物蛋白質を発現させるこ とができる、請求項1の合成遺伝子。 3.同一条件下のインビトロ哺乳動物細胞培養系において天然遺伝子によって 発現されるレベルの少なくとも150%のレベルで哺乳動物蛋白質を発現させるこ とができる、請求項1の合成遺伝子。 4.同一条件下のインビトロ哺乳動物細胞培養系において天然遺伝子によって 発現されるレベルの少なくとも200%のレベルで哺乳動物蛋白質を発現させるこ とができる、請求項1の合成遺伝子。 5.同一条件下のインビトロ哺乳動物細胞培養系において天然遺伝子によって 発現されるレベルの少なくとも500%のレベルで哺乳動物蛋白質を発現させるこ とができる、請求項1の合成遺伝子。 6.同一条件下のインビトロ哺乳動物細胞培養系において天然遺伝子によって 発現されるレベルの少なくとも10倍のレベルで哺乳動物蛋白質を発現させること ができる、請求項1の合成遺伝子。 7.天然遺伝子におけるコドンの少なくとも10%が好ましくないコドンである 、請求項1の合成遺伝子。 8.天然遺伝子におけるコドンの少なくとも50%が好ましくないコドンである 、請求項1の合成遺伝子。 9.天然遺伝子に存在する好ましくないコドン及び比較的好ましくないコドン の少なくとも50%が好ましいコドンに置き換えられている、請求項1の合成遺伝 子。 10.天然遺伝子に存在する好ましくないコドン及び比較的好ましくないコドン の少なくとも90%が好ましいコドンに置き換えられている、請求項1の合成遺伝 子。 11.蛋白質がレトロウイルスまたはレンチウイルスの蛋白質である、請求項1 の合成遺伝子。 12.蛋白質がHIVの蛋白質である、請求項11の合成遺伝子。 13.蛋白質がgag、pol及びenvからなる群より選択されたものである、請求項1 2の合成遺伝子。 14.蛋白質がgp120又はgp160である、請求項13の合成遺伝子。 15.蛋白質がヒト蛋白質である、請求項1の合成遺伝子。 16.通常哺乳動物細胞によって発現される蛋白質をコードする合成遺伝子を調 製する方法であって、該蛋白質をコードする天然の遺伝子における好ましくない コドン及び比較的好ましくないコドンを同定し、1つ又は複数の該好ましくない コドン及び比較的好ましくないコドンを置換コドンとして同じアミノ酸をコード する好ましいコドンに置き換えることを含む方法。
