Description
Claims (13)
- 1.水性担体液体中に懸濁されたリポソーム小胞に封入すべき物質を充填する方 法であって、前記リポソーム小胞は層状リン脂質と場合により非リン脂質両親媒 性化合物を含む膜形成成分の1または複数の膜により取り囲まれた水性液体相に 満ちたコアから成り、前記方法は前記封入すべき物質を前記担体液体中に導入し そして均一に分散させ、膜脂質転移温度Tcよりも高い温度において一定時間の 間インキュベートすることを含んで成り、それによって前記封入すべき物質か経 膜透過により前記小胞のコア中に浸透するであろう方法であって、充填前の小胞 内部コア相中に含まれる液体相の重量オスモル濃度か200mOsm/kgを越 えないことを特徴とする方法。
- 2.少なくとも1つの成分がイオン的に荷電しているリポソーム形成脂質の混合 物が、前記脂質を水和させそして積層化させるのに十分な時間の間水性液体相と 接触して置かれ、前記接触は、撹拌しながらまたは撹拌せずに1重量部の微粉砕 された形の前記脂質混合物が少なくとも20重量部の前記水性液体相中に導入さ れそして均一に分散される段階において行われ、そしてこの段階中は前記液相の 温度が前記水和形の脂質の転移温度(Tc)より数℃上の温度範囲に維持される ことを特徴とし、それによって高い比率の液体相が中に封入され且つ経膜充填容 量が増加されたリポソーム小胞が形成するであろう方、法。
- 3.前記リポソーム小胞のコアを満たす水性液体が、リポソームが懸濁される水 性担体液体も構成する、請求項1の方法。
- 4.前記封入すべき物質がイオン性または非イオン性である、請求項1の方法。
- 5.前記リポソーム分散液を、リポソームの外側とそのコアの内側の担体相中の 溶解物質の濃度が実質的に平衡になるまでの時間、Tcと約150℃の間の温度 に加熱することによりインキュベーションが行われる、請求項1の方法。
- 6.前記加熱温度が100℃より高く、そして加熱時間がリポソームの滅菌を保 証するのに十分である、請求項5の方法。
- 7.リポソーム内部コア中に取り込まれる容量対リポソーム小胞壁を構成する脂 質の重量の比が5μl/mg以上である、請求項2の方法。
- 8.前記担体相中に脂質を分散する段階が撹拌下で行われ、こうして得られるリ ポソーム小胞の平均サイズが前記撹拌の程度に反比例する、請求項2の方法。
- 9.前記リポソームが形成される水性液体が純水であり、それによって形成する リポソームが水のみを含む空のリポソームである、請求項1の方法。
- 10.繰り返し凍結−解凍段階および/または脱水−再水和段階によりリポソー ム内部コア中に取り込まれる容量が増加される、請求項2の方法。
- 11.前記小胞のサイズ分布を均一化するためにリポソーム分散液が孔径調整さ れた多孔質膜を通して押し出される、請求項2の方法。
- 12.前記封入すべき物質が一般の薬剤および注入可能な診断薬から選択される 、請求項1の方法。
- 13.前記診断薬が有機ヨウ素化X線造影剤であって、該薬剤は、脂質1mgあ たりのヨウ素のmg(I/L)により表わされる充填効率が3より大きい値に達 することができる程度にインキュベーションの間にリポソーム内に充填される、 請求項11の方法。
