Description
本発明は、試料を少なくとも2つの成分に分離するための遠心分離機と、この遠心分離機が使用される、試料を少なくとも2つの成分に分離するための方法とに関する。
本発明は、例えば、以下を提供する。
(項目1)
試料を少なくとも2つの成分に分離する遠心分離機であって、
遠心分離される試料を受容するチャンバを備えており、
該試料の分離の進展を制御する手段が、該チャンバに位置することを特徴とする、遠心分離機。
(項目2)
前記チャンバは、
円形のベースプレートであって、該ベースプレートの中心が回転軸に対して実質的に垂直に配向される、ベースプレートと、
該ベースプレートに対して実質的に垂直に配向される、外装材と、
円形のカバープレートであって、該ベースプレートから遠く離れている該外装材の縁の上に配置可能である、カバープレートと
を備えることを特徴とする、項目1に記載の遠心分離機。
(項目3)
前記試料の前記分離の進展を制御する前記手段は、前記ベースプレートまたは前記カバープレートに配置されることを特徴とする、項目1または2に記載の遠心分離機。
(項目4)
前記試料の前記分離の進展を検出する前記手段は、
光源からの光が、分離されつつある該試料の少なくとも一部を、少なくとも部分的に透過することと、
該試料の少なくとも一部を通過する光を、光検出器によって検出することと
が可能であるように配置されることを特徴とする、項目1〜3のうちのいずれか一項に記載の遠心分離機。
(項目5)
前記試料の前記分離の進展を制御する前記手段は、該試料が検出可能となるように遠心分離中に進入することができる、前記ベースプレートまたは前記カバープレート内に位置するチャネルまたは間隙に配置されることを特徴とする、項目1〜4のうちのいずれか一項に記載の遠心分離機。
(項目6)
前記分離の進展を検出する前記手段は、回転容器の回転軸に対して本質的に垂直に、該回転容器に位置することを特徴とする、項目1〜5のうちのいずれか一項に記載の遠心分離機。
(項目5)
前記チャネルまたは間隙は、前記ベースプレートまたは前記カバープレートの前記回転軸に位置する領域から該ベースプレートまたは該カバープレートの周囲に位置する領域まで、放射状に延在するように配向されることを特徴とする、項目4に記載の遠心分離機。
(項目6)
前記試料の前記分離の進展を制御する前記手段は、光源からの光が、該試料の少なくとも一部を透過させられることができるように位置する、窓、鏡、またはプリズムであり、該窓、鏡、またはプリズムから出る該光は、各々、光検出器によって検出可能であることを特徴とする、項目1〜5のうちのいずれか一項に記載の遠心分離機。
(項目7)
前記窓、プリズム、または鏡は、前記チャネルまたは間隙を覆うように配置されることを特徴とする、項目1〜6のうちのいずれか一項に記載のシステム。
(項目8)
前記チャンバは、細胞の培養のための容器としての機能を果たすことができるように構成されることを特徴とする、項目1〜7のうちのいずれか一項に記載の遠心分離機。
(項目9)
前記チャンバは、その上で細胞を増殖させるための少なくとも1つの層を備えることを特徴とする、項目1〜8のうちのいずれか一項に記載のシステム。
(項目10)
前記チャンバは、使い捨てであるか、および/または、滅菌することができることを特徴とする、項目1〜9のうちのいずれか一項に記載のシステム。
(項目11)
前記チャンバは、プラスチック、ポリスチロール(PS)、ポリスチレン、ポリ塩化ビニル、ポリカーボネート、ガラス、ポリアクリレート、ポリアクリルアミド、ポリメチルメタクリルレート(PMMA)、ポリエチレンテレフタレート(PET)、ポリテトラフルオロエチレン(PTFE)、熱可塑性ポリウレタン(TPU)、シリコーンから成る群より選択される、材料を備えるか、または材料で作ることができ、該チャンバはまた、ポリエチレン(PE)、コラーゲン、キチン、アルギン酸塩、ヒアルロン酸誘導体、ポリラクチド(PLA)、ポリグリコリド(PGA)、およびそれらの共重合体、ポリスチロール(PS)、ポリスチレン、ポリカーボネート、ポリアクリレート、セラミック、ハイドロキシアパタイト(HA)およびリン酸カルシウムのようなガラス材料、および上記の材料のうちの1つ以上を備える組成物で作ることもできることを特徴とする、項目1〜10のうちのいずれか一項に記載のシステム。
(項目12)
少なくとも2つの成分に分離されることができる試料を提供することと、
項目1〜11のうちのいずれか一項に記載の遠心分離機の中で該試料を遠心分離することと
を含む、試料を少なくとも2つの成分に分離するための方法。
(項目13)
特に、前記試料の層の形成、pH値の変化、および/または温度の変化を検出することによって、前記分離の進展を検出することをさらに含む、項目12に記載の方法。
(項目14)
前記試料は、血液、骨髄、細胞、細胞および/または細胞成分を含む組成物、または類似物等の生体試料である、項目12または13に記載の方法。
本発明の遠心分離チャンバは、細胞培養フラスコまたはバッグと同様に、細胞の培養に使用することができる。
Claims (16)
- 試料を少なくとも2つの成分に分離する遠心分離機であって、
遠心分離される試料を受容するチャンバ(170;190;201;220;250;500)と、
該チャンバに位置する該試料の分離の進展を検出する手段(188;237;810)と
を備えており、
該チャンバ(170;190;201;220;250;500)は、
円形のベースプレートであって、該ベースプレートの中心が回転軸に対して実質的に垂直に配向される、ベースプレートと、
該ベースプレートに対して実質的に垂直に配向される、外装材と、
円形のカバープレート(192;800)であって、該ベースプレートから遠く離れている該外装材の縁の上に配置可能である、カバープレートと
を含み、
該試料の該分離の進展を検出する該手段(188;237;810)は、該ベースプレートまたは該カバープレート(192;800)に配置されており、
該試料の該分離の進展を検出する該手段(188;237;810)は、該試料が検出可能となるように遠心分離中に進入することができる、該ベースプレートまたは該カバープレート(192;800)内に位置するチャネル(186;205;801)または間隙(801)に配置されることを特徴とし、
該チャネル(186;205;801)または間隙(801)は、該ベースプレートまたは該カバープレート(192;800)の該回転軸に位置する領域から該ベースプレートまたは該カバープレート(192;800)の周囲に位置する領域まで、放射状に延在するように配向されることを特徴とする、遠心分離機。 - 前記分離の進展を検出する前記手段(188;237;810)は、
光源からの光が、分離されつつある試料の少なくとも一部を、少なくとも部分的に透過することと、
該試料の少なくとも一部を通過する光を、光検出器によって検出することと
が可能であるように配置されることを特徴とする、請求項1に記載の遠心分離機。 - 前記分離の進展を検出する前記手段(188;237;810)は、前記チャンバ(170;190;201;220;250;500)の回転軸(176)に対して本質的に垂直に、該チャンバ(170;190;201;220;250;500)に位置することを特徴とする、請求項1または2に記載の遠心分離機。
- 前記試料の前記分離の進展を検出する前記手段(188;237;810)は、光源からの光が、該試料の少なくとも一部を透過させられることができるように位置する、窓、鏡、またはプリズムであり、該窓、鏡、またはプリズムから出る該光は、各々、光検出器によって検出可能であることを特徴とする、請求項1〜3のうちのいずれか一項に記載の遠心分離機。
- 前記窓、プリズム(188;237;810)、または鏡は、前記チャネル(186;205;801)または間隙(801)を覆うように配置されることを特徴とする、請求項4に記載の遠心分離機。
- 前記チャンバ(170;190;201;220;250;500)は、細胞の培養のための容器であることを特徴とする、請求項1〜5のうちのいずれか一項に記載の遠心分離機。
- 前記チャンバ(170;190;201;220;250;500)は、その上で細胞を増殖させるための少なくとも1つの層状構造を備えることを特徴とする、請求項1〜6のうちのいずれか一項に記載の遠心分離機。
- 前記チャンバ(170;190;201;220;250;500)は、使い捨てであることを特徴とする、請求項1〜7のうちのいずれか一項に記載の遠心分離機。
- 前記チャンバ(170;190;201;220;250;500)は、滅菌することができることを特徴とする、請求項1〜8のうちのいずれか一項に記載の遠心分離機。

- 前記ベースプレートは、少なくとも1つのセンサパッドを備えている、請求項1〜11のいずれか一項に記載の遠心分離機。
- 少なくとも2つの成分に分離されることができる試料を提供することと、
請求項1〜12のうちのいずれか一項に記載の遠心分離機の中で該試料を遠心分離することと
を含む、試料を少なくとも2つの成分に分離するための方法。 - 特に、前記試料の層の形成、pH値の変化、および/または温度の変化を検出することによって、前記分離の進展を検出することをさらに含む、請求項13に記載の方法。
- 前記試料は、生体試料である、請求項13または14に記載の方法。
- 前記生体試料は、血液、骨髄、細胞、または、細胞および/もしくは細胞成分を含む組成物である、請求項15に記載の方法。
