Description
ホップ葉からの冷水抽出:
ホップの葉(国産フラノ18号)を刻み10倍量(w/v)の水に浸して5℃にて一晩静置した後、7000rpmで15分遠心分離し、上清を回収して冷水抽出物を得た。
上清を分液ロートに移し、ヘキサンを加えヘキサン移行成分を廃棄した。さらに、水層に酢酸エチルを加え、酢酸エチル移行成分を廃棄した。最後に水層にn−ブタノールを加えブタノール抽出操作を3回繰り返して得たブタノール層を合併し減圧濃縮してフラボノール画分(ホップの組織の冷水抽出物から分離されたフラボノイド配糖体)を得た。
ホップペレットからの冷水抽出:
ホップペレット(チェコ産ザーツ種:タイプ90)1kgを蒸留水10Lに入れ、5℃にて時々攪拌しペレット状態を消失させながら一晩静置した。7000rpmで15分遠心分離した後、上清を回収し、それをさらに濃縮して150gの冷水抽出物を得た。
上清を実施例1と同様の方法で、ブタノール抽出成分を取得し、HPLCで分析し、成分の同定を行った。HPLC分析の結果を図2に示す。図2に示すように、ホップペレット抽出物のフラボノール画分には主要ピークが3つ存在し、これらはケンフェロール配糖体(アストラガリン及びケンフェロールマロニルグルコシド)とケルセチンマロニルグルコシドであることが同定された。詳しくは、図2の1で示すピークはケンフェロールマロニルグルコシド、2で示すピークはアストラガリン、3で示すピークはケルセチンマロニルグルコシドであった。なお、図2の4で示すピークはルチン、5で示すピークはイソケルシトリン、6で示すピークはケンフェロールルチノシドであった。
ホップペレット(チェコ産ザーツ:タイプ90)を1%(w/w)となるようにクロロホルム−メタノール溶液(クロロホルム:メタノール=3:1)で抽出した。抽出はクロロホルム−メタノール溶液の沸点にて2時間行った。収率は約15%であった。
ホップペレット(チェコ産ザーツ:タイプ90)を1%(w/w)となるようにして煮沸水で2時間抽出を行った。収率は約23%であった。
実施例2及び比較例1〜2で得られたホップ抽出物を用いて、以下のようにしてヒスタミン遊離抑制作用の確認を行った。
ホップペレット(チェコ産ザーツ種:タイプ90)100gを蒸留水1Lに入れ、15分間、30分間、60分間煮沸し、それを7000rpmで15分遠心分離した後、上清を回収し抽出物(熱水抽出物)とした。抽出を15分行ったものが比較例3、30分行ったものが比較例4、60分行ったものが比較例5である。
用いるホップの種類又は組織を変化させて実施例1又は2と同様にして冷水抽出物を得、上記と同様にしてヒスタミン遊離抑制率(%)を算出した。ドイツ産ハラタウ・トラディション種のペレットを用いたのが実施例3、国産フラノβのペレットを用いたのが実施例4、国産ホップの若葉の葉を用いたのが実施例5である。ヒスタミン遊離抑制率(%)の結果を図6に示す。なお、実施例2と同様のホップ(チェコ産ザーツ種のペレット)を用いたものについては、図6において実施例2と記載してある。図6に示すように、ホップ種としては、チェコ産ザーツ種、ドイツ産ハラタウ・トラディション種、国産フラノβが優れており、組織として葉も優れていることがわかった。
マウスDNP−HSAによる耳介浮腫におけるホップ水抽出物の抑制作用
6週齢のICR系マウス(雄又は雌;日本チャールス・リバー)を、1ケージあたり4〜5匹とし、室温24±2℃、湿度55±15%、12時間ごとの明暗サイクル(明期8:00−20:00)の条件下、飼育した。一週間以上の予備飼育後、試験に供した。試験群にはそれぞれホップペレット水抽出液、ホップ葉水抽出液、又はフマル酸ケトチフェン水溶液を、対照群には蒸留水をそれぞれ10mL/kgで強制経口投与した。ここで、ホップペレット水抽出液は、ホップペレット(チェコ産ザーツ種)を10%(w/w)となるように蒸留水中に加え4℃で一晩抽出した後、3000rpmで10分間遠心分離しろ過することによって調製した。ホップ葉水抽出液は、乾燥ホップ葉(国産ホップ若葉)を10%(w/w)となるように蒸留水中に加え4℃で一晩抽出した後、3000rpmで10分間遠心分離し、ろ過することによって調製した。また、フマル酸ケトチフェン水溶液は、0.5mg/mLの濃度となるように蒸留水に溶解することによって調製した。
Claims (15)
- ホップの組織の冷水抽出物からなる抗アレルギー組成物であって、前記冷水の温度は0℃超10℃以下であり、前記抽出物は有効成分としてケンフェロール配糖体を含む、抗アレルギー組成物。
- 前記ホップは、ビール醸造用ホップである、請求項1記載の抗アレルギー組成物。
- 前記組織は、茎、毬花又は葉である、請求項1又は2記載の抗アレルギー組成物。
- 前記組織は、乾燥された苞の粉砕物である、請求項1又は2記載の抗アレルギー組成物。
- 前記組織は、乾燥された毬花の粉砕物から、ルプリンの大きさ以下の粉砕物の少なくとも一部が除かれたものである、請求項1又は2記載の抗アレルギー組成物。
- 前記乾燥された毬花の粉砕物は、乾燥された毬花の凍結物の粉砕物である、請求項5記載の抗アレルギー組成物。
- 前記組織は、乾燥された毬花から、有機溶媒抽出又は超臨界流体抽出される物質の少なくとも一部が除かれたホップ残渣である、請求項1又は2記載の抗アレルギー組成物。
- 前記ケンフェロール配糖体は、ケンフェロールルチノシド、アストラガリン及びケンフェロールマロニルグルコシドからなる群より選ばれる少なくとも1つである、請求項1〜7のいずれか一項記載の抗アレルギー組成物。
- 前記フラボノール配糖体として、ケルセチンマロニルグルコシドを更に含有する、請求項8記載の抗アレルギー組成物。
- 前記冷水の温度が2℃〜8℃である、請求項1〜9のいずれか一項記載の抗アレルギー組成物。
- 前記冷水抽出物は、前記冷水によって前記ホップの組織を抽出した抽出液である、請求項1〜10のいずれか一項記載の抗アレルギー組成物。
- 前記冷水抽出物は、前記抽出液に対してさらにケンフェロール配糖体を濃縮したものである、請求項11記載の抗アレルギー組成物。
- 前記冷水抽出物は、前記抽出液に対してさらにヘキサン抽出及び酢酸エチル抽出を行い、ヘキサン抽出成分及び酢酸エチル抽出成分が除かれた抽出物である、請求項11記載の抗アレルギー組成物。
- 前記冷水抽出物は、前記ヘキサン抽出成分及び酢酸エチル抽出成分が除かれた抽出物に対してさらにブタノール抽出を行ったブタノール抽出物である、請求項13記載の抗アレルギー組成物。
- 請求項1〜14のいずれか一項記載の抗アレルギー組成物を有効成分とする抗アレルギー剤。
