Description
(ホップ組織の冷水抽出物の調製)
ホップ組織の冷水抽出物の口内清涼作用を調べるために、乾燥されたホップ毬花から製造され、ルプリンを豊富に含有するホップペレット(チェコ産ザーツ種:SSAタイプ90)の製造過程で副産物として生じたスペントホップからホップ冷水抽出物を調製した。スペントホップとは、乾燥されたホップ毬花の粉砕物からルプリンの大きさ以下の粉砕物の少なくとも一部が取り除かれた画分のことである。
健常人19名を、A班(10名)とB班(9名)の2グループにランダムに分け、A班には餃子を、B班にはコンソメスープを一口ずつ飲食させた後に盲目的に水又は0.2%ホップ冷水抽出物を順不同で飲ませ、口内のさっぱり度を5段階で評価させた。この評価では、数値が大きい程さっぱり度が高いことを意味している。その後、引き続いてA班には餃子を、B班にはコンソメスープを飲食させて、それぞれを美味しく感じたか否かを3段階で評価させた。餃子又はコンソメスープが美味しくなったと感じたとき+1、まずくなったと感じたときは−1、変わらないと感じたときは±0とした。尚、温度の影響を取り除くために、水及び0.2%ホップ冷水抽出物は、いずれも予め4℃に調整しておいて、飲む直前に提供した。
ホップ組織の冷水抽出物が水溶液中の脂肪酸に及ぼす影響について、コク・キレセンサー(相互薬工(株)製)を用いて調べた。かかるコク・キレセンサーは、1Hzの振動数の変化が成分約1ngの着脱に相当するよう設計されており、ビール成分のみでなく、脂肪酸のセンサー部への吸着性についても判断できる特性を有している。したがって、コク・キレセンサーでは、50ppmのオレイン酸又はリノール酸を添加すれば、これらの脂肪酸のセンサー部への吸着量の増加を振動数の増加として捉えることができる。本実験では、センサー部に吸着した脂肪酸を舌に存在する味覚受容体細胞の化学受容体を覆っている脂肪酸であると想定して、50ppmのオレイン酸又はリノール酸に0.02%となるようにホップ冷水抽出物を混合してコク・キレセンサーに添加した場合の脂肪酸のセンサー部への吸着量を測定した。
(ホップ冷水抽出物中に含まれるフラボノイドのLC−MSによる分析)
ホップ冷水抽出物中に含まれるフラボノイド成分を調べるため、以下の条件下で、ホップ冷水抽出物のLC−MS分析を行った。
LC−MSの分析条件:
・溶離液:A液 0.05%TFA水溶液、B液 アセトニトリル
・グラジエント条件:0〜16min、B液10%〜50%
・流量:0.2mL/min
・カラム温度:40℃
・カラム:Waters Symmetry C18 2.1×150mm 3.5μm
・マススペクトル:(SIR;m/z197、m/z211、m/z287、m/z303)
(ホップペレット又はスペントホップの冷水抽出物の調製)
チェコ産ザーツ種ホップのスペントホップから調製されたホップ冷水抽出物には、脂肪吸着阻害作用及び脂肪吸着除去作用のあることが明らかとなったので、その他の品種のホップペレット又はスペントホップから調製したホップ冷水抽出物についても同様の作用があるか否かについて試験した。その際、チェコ産ザーツ種のホップについては、スペントホップ熱水抽出物も同時に調製して調べた。
まず、50ppmのリノール酸に0.02%となるようにホップペレット冷水抽出物、スペントホップ冷水抽出物又はスペントホップ熱水抽出物を加えて十分に混合し、その後、この混合液を上記のコク・キレセンサー(相互薬工(株)製)に添加して、リノール酸のセンサー部への吸着量を測定した。
Claims (5)
- ホップ組織の冷水抽出物を有効成分とし、
アストラガリン、アストラガリンマロニルグルコシド、イソケルシトリン、イソケルシトリンマロニルグルコシド、ケルセチンマロニルグルコシド、ケンフェロールルチノシド、ケンフェロールマロニルグルコシド、ルチン及びフロロアシルフェノン配糖体からなる群より選ばれるフラボノイド配糖体の少なくとも一つを含有し、
脂肪吸着除去剤である、口内清涼剤。 - 前記ホップ組織は、乾燥されたホップ苞の粉砕物である、請求項1記載の口内清涼剤。
- 前記ホップ組織は、乾燥されたホップ毬花の粉砕物から、ルプリンの大きさ以下の粉砕物の少なくとも一部が除かれたものである、請求項1記載の口内清涼剤。
- 前記乾燥されたホップ毬花の粉砕物は、乾燥されたホップ毬花の凍結物の粉砕物である、請求項3記載の口内清涼剤。
- 前記ホップ組織は、乾燥されたホップ毬花から、有機溶媒抽出又は超臨界流体抽出される物質の少なくとも一部が除かれたホップ残渣である、請求項1記載の口内清涼剤。
