Description

【発明の詳細な説明】
本発明の要旨はイチョウ（ginkgo）エキスと少なくと
も１種のポリフェノール誘導体との相乗作用を有する組
合せ体を抗酸化剤として含有する化粧料又は製薬組成物
に関する。 化粧料又は製薬組成物の大部分は脂肪相よりなり、そ
の油状生成物は室温でさえ酸化する或る傾向がある。こ
の酸化は特性特に嗅覚特性を大いに変性する結果であ
り、これによって該組成物を可変期間後には使用不能と
させる。 これらの酸化現象に関して組成物を保護するために
は、抗酸化剤として使用する保護剤を配合することが普
通実施されている。 抗酸化剤は化粧料又は製薬組成物における脂肪の良好
な保護に特に有用であるけれども、これらの若干はまた
大気汚染源及び紫外線によって特に発生するフリーラジ
カルの作用下に生成される酸化性物質の有害作用を制御
し得ることは今や知られている。 これらの有害作用は外部環境と接触している皮膚及び
粘膜の細胞に特に生ずる。 即ち取り返しのつかない細胞損傷を生起してしまう酸
化現象を調節することができしかもフリーラジカルの形
成を抑制できる入手可能な抗酸化剤を有し得ることが重
要である。 イチョウエキスと少なくとも１種のポリフェノール化
合物との組合せ体を用いることにより易酸化性の脂肪を
含有する化粧料又は製薬組成物の良好な保存を達成でき
しかも皮膚又は粘膜を有効に保護できることが今般見出
された。この組合せ体は驚くべきことには相乗特性を有
することも見出された。 “ポリフェノール化合物”なる表現は少なくとも１個
のジフェノール芳香族環を含有する化合物を意味すると
理解され、該フェノール基は場合によってはエーテル化
又はエステル化できる。以下においてはかゝる化合物は
単に“ポリフェノール”と呼び得る。 本発明の要旨はイチョウエキスと少なくとも１種のポ
リフェノール化合物との組合せよりなる相乗作用のある
抗酸化系を含有する化粧料又は製薬組成物に関する。 イチョウエキス（ginkgo extract）の１種以上の活性
物質は知られていないがこの活性物質は非極性溶剤を用
いてイチョウ植物等に葉を抽出することにより得られ
る。“非極性抽出物”なる表現はかゝる抽出物を表わす
か又はかゝる抽出物に含有される１種以上の活性物質で
より完全な精製によって抽出物から単離し得る活性物質
の何れかを以下では表わす。活性物質はこゝでは抗酸化
活性を有する物質（例えば以下の実験部分に記載される
如くビタミンＦの自己酸化試験により表わし得る）を表
わし、これとポリフェノールとの組み合せによって相乗
的な抗酸化作用を有し得る。 Ginkgo bilobaの葉からの抽出物（エキス）が特に使
用される。 非極性のイチョウエキスは非極性溶剤でイチョウの葉
の抽出から得られるフラクションを蒸発乾固することに
より得られる。直鎖、分枝鎖又は環式C6-14アルカン類
を非極性溶剤として挙げ得る。ｎ−ヘキサンが特に好ま
しい。かゝる抽出物は日本特許公開91−014,007に特に
記載される。 本発明の組成物で用いたポリフェノール化合物は実験
部分で以下に記載される如くビタミンＦの自己酸化試験
で抗酸化活性を有する化合物から選択できる。 ポリフェノールは例えば次の化合物から選択できる： ａ）フラボノイド、 ｂ）カルノシン酸又はカルノゾール、 ｃ）場合によっては置換された2,5−ジヒドロキシ安息
香酸及び（2,5−ジヒドロキシフェニル）アルキレンカ
ルボン酸及びそれらの誘導体、特にそれらの塩、エステ
ル又はアミド、 ｄ）カフェイン酸のエステル又はアミド、 ｅ）タンニン酸 使用し得るポリフェノール類のうちでは、特に一般式
（Ｉ）： 又は（II）： 〔式中Ａ″,B″,C″及びＤ″は互いに個々にＨ又はOHを
表わし;E″はH,OH又はOX′を表わし、Ｘ′は を表わし、 Ｆ″,G″及びＪ″は互いに個々にＨ又はOHを表わし;X1
は−CH2−，−CO−又は−CHOH−を表わし、Ａ′,C′及
びＤ′は互いに個々にH,OH又はOCH3を表わし； Ｅ′はH,OH又はOR′を表わし、Ｒ′は式Ｒ′OHの糖の残
基を表わし； Ｂ′,F′,G′及びＪ′は互いに個々にH,OH,OCH3又は−O
CH2−CH2−OHを表わす〕に相当するフラボノイドが挙げ
られる。糖Ｒ′OHのうちではルチノースが挙げられる。 前記式（Ｉ）及び（II）の化合物は既知である。これ
らは特に“The Flavonoides"Harborne J.B.,Mabry T.J.
Helga Marbry（1975）１〜45頁に記載された方法により
得られる。 本発明により使用し得るフラボノイドのうちでは、タ
キシホリン、カテキン、エピカテキン、エリオジクチオ
ール、ナリンゲニン、ルチン、トロキセルチン、クリシ
ン、タンゲレチン、ルテオリン、エピガロカテキン及び
エピガロカテキンガレート、クエルセチン、フィセチ
ン、カエンプフェロール、ガランギン、ガロカテキン及
びエピカテキンガレートが特に挙げられる。 使用し得る或るポリフェノール類は、既知の仕方で抽
出し得る植物中に存在する。茶の葉（Camellia sinensi
s又はCamellia japonica）からの抽出物を使用できる。
特にニッコー社によりサンフェノン（Sunphenon）の名
称で市販されフラボノイドを特に含有する緑茶エキスを
挙げ得る。 使用し得るポリフェノール類としては、抽出続いて蒸
留により（ChangらのJOSC,Vol.61,No.6（1984））ある
いは欧州特許出願EP−307,626に記載される如く芳香性
物質を除去するのにヘキサンの如き非極性溶剤を用いて
抽出し続いてエタノールの如き極性溶剤での抽出によ
り、例えばマンネンロウから抽出し得るカルノシン酸及
びカルノゾールの如きポリフェノールが挙げられる。 使用し得るポリフェノールはまた式（III）： 〔式中Ｒ″１は−Ｏ−AlK、OH又は−Ｎ（ｒ′）
（ｒ″）を表わし、Alkは、場合によっては１個以上の
ヒドロキシル又はアルコキシ基により置換された直鎖又
は分枝鎖C1-20アルキル基であるか又はC2-20アルケニル
基であり、 ｒ′及びｒ″は個々にＨ、C1-20アルキル、C2-6ヒドロ
キシアルキル又はC3-6ポリヒドロキシアルキルを表わす
か又は別法としてｒ′及びｒ″はそれらが結合している
窒素原子と一緒になって複素環式基を表わし、 ｒは−（CH2）ｒ−COR″１連鎖が精々21個の炭素原子を
含有するような個数（零も含む）であり、 Ｒ″２及びＲ″３は個々にＨ又はC1-4アルキル基を表わ
し、追加的にＲ″２はC1-4アルコキシ基を表わし得る〕
の（2,5−ジヒドロキシフェニル）アルキレンカルボン
酸及びそれらの誘導体（特にエステル及びアミド）から
選択できる。 前記式（III）の化合物は既知であるか又は例えばフ
ランス特許FR−2,400,358及びFR−2,400,359に記載され
た方法と同様な既知の方法により製造できる。 本発明により使用できるポリフェノール類のうち、カ
フェイン酸のエステル又はアミドが挙げられる。カフェ
イン酸のエステルのうち、次式（IV）： （式中ＺはC1-8アルキル例えばメチル又はフィトール
（phytol）の残基を表わす）の化合物を特に挙げ得る。 カフェイン酸のアミドのうち、次式（Ｖ）： （式中Ｚ′はC1-8アルキル特にC6-8アルキル基を表わ
す）の化合物が特に挙げられる。 前記式（IV）又は（Ｖ）の化合物は既知であるか又は
既知の方法により製造できる。 タンニン酸は、Sochibo社からスペクトラット（Supex
trat）の名称で市販される五倍子エキスに特に存在す
る。 本発明の化粧料又は製薬組成物において、非極性のイ
チョウエキスは組成物の全重量に関して１〜10重量％の
濃度で一般に存在する。 前述の如きポリフェノールは組成物の全重量に関して
0.1〜１重量％の割合で存在する。 非極性のイチョウエキスとポリフェノールとの最適な
相対的割合は簡単な定常実験によって各々の型式の組成
物について測定できる。非極性イチョウエキスとポリフ
ェノールとの重量比は一般に１〜100であり、特に10の
付近にある。 本発明の化粧料又は製薬組成物は前記した活性成分の
組合せに加えて、適当な担体及びかゝる組成物を製造す
るのに普通用いた成分又は助剤を含有できる。該組成物
は特に溶剤例えば水、有機溶剤（例えばアルコール又は
油類）又はシリコーン、増粘剤、表面活性剤、重合体、
固体の脂肪（例えばワックス又はラノリン）、加湿剤、
防腐剤、pH改質剤、金属イオン封鎖剤、着色剤、芳香
剤、固体の充填剤（粉末及び顔料）、紫外線吸収物質、
即時のなめし剤（例えばジヒドロキシアセトン）等を含
有できる。 小のう分散物の形の組成物は、例えば脂質の二重層に
配合した少なくとも１種の活性成分即ちミセルを含有
し、これは水性相を包蔵化できしかも水性溶剤に分散さ
れ得る。 脂質特にイオン系又は非イオン系両親媒性脂質の小の
う分散物は既知の方法により製造され、例えばBanghan,
Standish及びWatkinsのJ.Mol.Biol.,13,238（1965）に
よる文献又はフランス特許FR 2,315,991及び2,416,008
に記載される如く、水性媒質に分散した小球を形成する
ために水溶液中に脂質を膨潤させることにより製造され
る種々の製造方法の記載はまたINSERM/John Libbery Eu
rotext出版の“Les Liposomes en biologie cellulaire
et pharmacologie"〔Liposomes in cell biology and p
harmacology〕（1987）６〜18頁に見出される。 組成物は微小粒子（nanoparticles）の分散物の形で
も提供される。用語“微小粒子”は一方では微小球（na
nospheres）を包含し、他方では微小カプセル（nanocap
sules）を包含し；用語“微小球”は活性成分が吸収及
び／又は吸着された多孔質重合体母材よりなる微小粒子
を表わし、用語“微小カプセル”は活性成分によって形
成された芯部を包囲する重合体膜よりなる微小粒子を表
わす。かゝる組成物形は例えば欧州特許出願EP−274,91
6及びフランス特許出願FR−2,659,554に記載されてい
る。 本発明の組成物は特にヒトの皮膚、毛髪及び粘膜を保
護する化粧料又は製薬組成物、皮膚をメーキャップする
組成物及び歯みがきの如き口腔衛生用途の外骨格組成物
又は洗眼剤の如き眼用組成物である。 本発明の化粧料組成物を毛髪の保護に用いる時はシャ
ンプー、ローション、ゲル組成物の形あるいはシャンプ
ー前後に、染色又は漂白前後に又はパーマネントウェー
ブ又は毛髪のストレート処理の前に、間に又は後に施用
されるリンス組成物の形で提供し得る。更にはスタイリ
ング又は処置用ゲル又はローションの形、ブロー乾燥又
は毛髪のセット用ローション又はゲルの形、ヘアーラッ
カーの形、パーマネントウエーブ又は毛髪のストレート
処理組成物又は毛髪の染色又は漂白用組成物の形で提供
し得る。 本発明の組成物をまつげ、まゆ毛又は皮膚をメーキャ
ップする生成物として用いる時には、例えば皮膚の処置
用クリーム又はファンデーションクリーム、口紅、アイ
シャドー、ブラッシャー、アイライナー又はマスカラの
形で提供される。 本発明の組成物が製薬組成物である時は、特にエマル
ジョン（ミルク又はクリーム）、ゲル、ローション、軟
膏、小のう分散物又は微粒子分散物形で提供できしかも
前記した組合せに加えて別の製薬活性成分を含有でき
る。 本発明の組成物が含有する相乗作用のある組合せ体に
よって、該組成物は皮膚、外骨格及び粘膜に特に施用さ
れるのに意図した化粧料又は製薬組成物を構成し、これ
によって例えば大気汚染源及び紫外線によって特に誘発
されるフリーラジカルによって生起する損傷を防止又は
処置し得る。特に本発明の化粧料組成物は皮膚の老化促
進現象を防止又は処置し得る。 本発明の別の要旨は、大気汚染源及び／又は紫外線に
より特に誘発されるフリーラジカルによって皮膚、頭皮
又は粘膜上に生起される細胞の損傷を防止又は処置する

のに意図した化粧料又は製薬組成物を製造するのに、相
乗活性のある組合せ体として少なくとも１種のイチョウ
エキスと少なくとも１種のポリフェノールとの組合せ体
の使用である。 本発明を次の実施例により説明する。 これらの実施例において、用いたイチョウエキスは以
下に示した方法で調製された。緑茶エキス“Sunphenon"
はニッコーケミカル社によって市販されている。水性／
アルコール性五倍子エキスはSupextratの名称でSochibo
社により市販されている。 イチョウエキスを取得する方法 植物粉末の形に換えたイチョウ（Ginkgo biloba）の
葉を多孔質セルロースカードボード製のカートリッジに
装入した。 このカートリッジを“ソックスレー”型の抽出器に導
入し；この抽出器はその基部に沸点（69℃）に加熱した
ヘキサンを含有する丸底フラスコを備えている。溶剤蒸
気は分岐を通過し、還流コンデンサーで凝縮され、カー
トリッジに液体状態で落下返送され、カートリッジを徐
々に浸漬する。全体の浸漬後に、植物エキスを含んだ溶
剤はサイフォンにより最初の丸底フラスコに流出する。
該方法は連続的に行ない、丸底フラスコ中の液体には植
物エキスが増々含有される。 抽出は12時間行った。次いでヘキサンフラクションを
減圧下に蒸発乾固した。 実施例１ O/Wボディローション このローションは次の組成（重量％）を有する： グリセリルステアレート 2％ トゥイーン60（ICI社によって市販の、20モルのエチ
レンオキシドを含有するソルビタンモノステアレート）
1％ ステアリン酸 1.4％ トリエタノールアミン 0.7％ カルボポール940（トリエタノールアミンによって中
和した） 0.2％ スウィートアーモンドの油 3％ 流動パラフィン 8％ 水性／アルコール性五倍子エキス 0.1％ イチョウエキス 1％ 無菌脱塩水＋防腐剤 全体を100％にする十分量 O/Wは水中油型エマルジョンを意味する。 カルボポール940はグリッドリッチ社により市販の架
橋したポリアクリル酸である。 グリセリルステアレート、トゥイーン60、ステアリン
酸及び油を75〜80℃に加熱し、トリエタノールアミンを
添加し、この混合物を60gの水の存在下に中和済みカル
ボポールに注加した。温度を40℃に低下させ、抽出液、
残りの水及び防腐剤を添加した。 実施例２ O/Wボディケアクリーム 次のクリームを同様に製造した： グリセリルステアレート 2％ トゥイーン60（ICI社により市販の、20モルのエチレ
ンオキシドを含有するソルビタンモノステアレート） 1％ セチルアルコール 0.5％ ステアリン酸 1.4％ トリエタノールアミン 0.7％ カルボポール940（トリエタノールアミンによって中
和した） 0.4％ カリテ（karite）脂肪の液体フラクション 12％ 合成パーヒドロスクアレン 12％ 水性／アルコール性五倍子エキス 0.1％ イチョウエキス 1％ 無菌脱塩水＋防腐剤 全体を100％にする十分量 実施例３ ハンドローション イチョウエキス 1％ 緑茶エキス 0.1％ レシノール S10 0.375％ ゲネロール122 ES 0.625％ Dow社により市販のD5（シクロメチコーン） 2％ グリセロール 10％ メチルｐ−ヒドロキシベンゾエート 0.3％ 水 全体を100％にする十分量 レシノールS10はニッコー社により市販の水添レシチ
ンの商品名である。 ゲネロール122 ESはHenkel社により市販の５モルのエ
チレンオキシドでオキシエチレン化したフィトステロー
ルの商品名である。 実施例４ 小のう分散物 非イオン系アンフィフィル（amphiphile）＊ 1.5％ コレステロール 1.5％ ナトリウムアシルグルタメートHS21（アジノモト）0.
5％ グリセロール 3％ 緑茶エキス（Sunphenon） 0.1％ イチョウエキス 1％ パーヒドロスクアレン 10％ メチルｐ−ヒドロキシベンゾエート 0.2％ カルボポール940（グッドリッチ社） 0.4％ トリエタノールアミン pH＝７にする十分量 水 全体を100％にする十分量 （＊）非イオン系アンフィフィル（amphiphile）は次
式： C12H25−〔Ｏ−C2H3（Ｒ）−Ｏ−C3H5（OH）−Ｏ〕ｎ−Ｈ 〔式中ｎは単位の統計数を表わし、2.7に等しく、 −OC2H3（Ｒ）−基は、 及び を表わし、−C3H5（OH）−Ｏ−基は 及び を表わし、 Ｒ基はC14H29とC16H33との等モル混合物を表わす〕に相
当する生成物の混合物である。 “アシルグルタメートHS21"の名称で市販の生成物は
ジナトリウムステアリルグルタメートである。 この小のう状分散物を次の仕方で製造した。 非イオン系のアンフィフィル化合物を100℃の温度で
コレステロール及びアシルグルタメートに添加した。 温度を90℃に低下させ、グリセロール、緑茶エキス、
イチョウエキス及び水（10g）をこの温度で添加した。 該混合物を50℃に冷却し次いでビルチス（Virtis）60
ホモジナイザー（40,000rpm）を用いて４分間２回均質
化した。 カルボポールゲル（カルボポール940及び100にする十
分量の水）を30秒間10,000rpmで分散させ、合した混合
物を次いでトリエタノールアミンで中和した。 平滑で光沢のあるクリームが得られた。これはフリー
ラジカルによる有害な作用から毛髪を保護するのに頭皮
上で用いた。 抗酸化活性の研究 本発明による抗酸化系の効果は、酸化に対して特に鋭
敏な物質であるビタミンＦの促進酸化法によって証明さ
れた。 この研究についてはメトロール社の自動装置“ランシ
マット（Rancimat）”（A.SeherらのFette,Seifen,Anst
richmittel 88（１）１〜６頁（1986）参照）を使用し
た。 ビタミンＦと緑茶エキス“Sunphenon"のみとの混合
物、ビタミンＦとイチョウのヘキサン抽出物のみとの混
合物、及びビタミンＦと緑茶エキスとイチョウのヘキサ
ン抽出物との混合物を調製した。 空気（20／時）を散布しながら各々の試料を100℃
に生起させた。次いでビタミンＦのハイドロパーオキシ
ド及びアルデヒドの分解から得られる揮発性酸の濃度
を、白金電極を浸漬した水を充填したセル中で連続的に
監視した。この電極は時間の関数として揮発性酸の濃度
の上昇によって生起される導電度の増大を測定する。誘
発時間は得られた指数酸化曲線の２本の漸近線の交点に
よって測定される。 この時間はビタミンＦの自己酸化に先立つ潜伏期間に
相当する。この潜伏期間が長ければ長い程、ビタミンＦ
の自己酸化に対する耐性がより良いこととなる。 結果は次の通りである： 試験した生成物 誘発時間 緑茶0.1％ 33分 イチョウ１％ 48分 緑茶0.1％＋イチョウ１％ 258分

Claims (10)

    (57)【特許請求の範囲】
  1. 【請求項１】イチョウエキスと少なくとも１種のポリフ
    ェノール化合物との組合せ体よりなりしかも相乗作用を
    有する抗酸化系を含有し、イチョウエキスは非極性溶剤
    を用いてイチョウ植物の葉を抽出することにより得られ
    ることを特徴とする化粧料又は製薬組成物。
  2. 【請求項２】前記の非極性溶剤はＣ６〜14アルカンであ
    ることを特徴とする請求の範囲１記載の組成物。
  3. 【請求項３】前記の非極性溶剤はｎ−ヘキサンであるこ
    とを特徴とする請求の範囲２記載の組成物。
  4. 【請求項４】ポリフェノール化合物は少なくとも１種の
    ジフェノール芳香族環を含有し、フェノール基は場合に
    よってはエーテル化又はエステル化され得ることを特徴
    とする請求の範囲１〜３の何れかに記載の組成物。
  5. 【請求項５】ポリフェノール化合物はフラボノイド、カ
    ルノシン酸、カルノゾール、場合によっては置換された
    2,5−ジヒドロキシ安息香酸及び（2,5−ジヒドロキシフ
    ェニル）アルキレンカルボン酸及びそれらの誘導体、カ
    フェイン酸のエステル又はアミド及びタンニン酸から選
    択されることを特徴とする、請求の範囲１〜４の何れか
    に記載の組成物。
  6. 【請求項６】イチョウエキスは組成物の全重量に関して
    １〜10重量％の濃度で存在することを特徴とする請求の
    範囲１〜５の何れかに記載の組成物。
  7. 【請求項７】ポリフェノール化合物は組成物の全重量に
    関して0.1〜１重量％の割合で存在することを特徴とす
    る請求の範囲１〜６の何れかに記載の組成物。
  8. 【請求項８】イチョウエキスとポリフェノール化合物と
    の重量比は１〜100であり、特に10の付近にあることを
    特徴とする、請求の範囲１〜７の何れかに記載の組成
    物。
  9. 【請求項９】大気の汚染源によって及び／又は紫外線に
    よって特に誘発されるフリーラジカルにより皮膚、頭皮
    又は粘膜において生起される細胞損傷を防止又は処置す
    るのに意図された及び／又は皮膚の老化促進現象を調節
    するのに意図された化粧料又は製薬組成物を製造するに
    際して、相乗作用のある組合せ体として、非極性溶剤で
    イチョウの葉を抽出することにより得られる、少なくと
    も１種のイチョウエキスと、少なくとも１種のポリフェ
    ノール化合物との組合せ体の使用。
  10. 【請求項１０】請求の範囲９記載の組成物は請求の範囲
    ２〜８の何れか１つに定義される如き特性を有すること
    を特徴とする、請求の範囲第９記載の使用。
