多くの疾患の基礎をなす(ウイルス感染及び悪性細胞を含む)異常細胞の出現及び持続は、前記異常細胞に対する不十分な免疫応答によって可能になる。免疫療法の目標は、患者の免疫系の応答を開始又は増強する例えば免疫細胞、例えばナチュラルキラー(NK)細胞が損傷又は異常細胞を損傷させるか、殺滅するか、又は他に阻害する能力を後押しすることである。1つの免疫療法アプローチは、異常細胞の認識及び破壊を標的にするための免疫細胞におけるキメラ受容体の組換え発現である。一般に、キメラ受容体は、標的細胞上のリガンドを認識する細胞外受容体ドメイン、アンカー膜貫通ドメイン及びリガンド結合時のシグナルの活性化を伝達するエフェクタードメインを含む。本明細書で開示されるいくつかの実施形態では、その一般的構造を有するか又はその一般的構造におけるバリエーションを有するキメラ受容体が用いられる。加えて、いくつかの実施形態では、膜貫通ドメイン及びエフェクタードメインは、一緒に融合された別々のペプチドである。いくつかの他の実施形態では、膜貫通及びエフェクタードメインは、同じペプチドに由来する。いくつかのかかる実施形態では、膜貫通及びエフェクタードメインは、単一のペプチド(例えば、膜を貫通することに加え、平衡が保たれ、シグナルカスケードを開始させる1つのペプチド)を含む。以下により詳細に考察される通り、免疫細胞(例えばNK細胞)において所望の程度の発現を示し、非標的細胞に対する有害作用を回避する程度の標的結合活性と平衡した、NK細胞から細胞傷害活性を誘導するキメラ受容体構築物を作製するため、切断、突然変異、追加的なリンカー/スペーサーエレメント、二量体などが用いられる。免疫細胞の表面上での本明細書で開示されるようなキメラ受容体の組換え発現により、目的の異常細胞に対する免疫細胞の標的化が再誘導され、且つ会合時の免疫活性化が増強される可能性がある。
1つの免疫療法アプローチは、キメラ受容体を発現するように操作されたT細胞を患者に投与し、陽性免疫応答を誘発することを含む。しかし、この手法の欠点は、患者における移植片対宿主病の誘導を予防するために自家細胞の使用が必要である点である。本明細書で開示されるいくつかの実施形態において提供される通り、操作されたNK細胞を含む組成物は、いくつかの利点を享受する。例えば、自家又はドナー由来同種細胞のいずれかは、NK細胞アプローチで用いることができる。加えて、いくつかの実施形態によると、本明細書で提供されるような操作されたNK細胞は、正常細胞に対して細胞傷害性を増強することが有意でない。さらに、NK細胞は、活性化されると、有意な細胞傷害性効果を有する。これを考慮すると、本明細書で提供されるような操作されたNK細胞が、その細胞傷害性効果をさらに高めることにより、罹患した標的細胞を選択的に殺滅するといったさらにより有効な手段を提供し得ることは、想定外である。したがって、いくつかの実施形態では、癌又は感染性疾患を治療又は予防する方法であって、治療有効量の、本明細書に記載のキメラ受容体を発現するNK細胞を投与することを含む方法が提供される。一実施形態では、投与されるNK細胞は、自家細胞である。さらなる実施形態では、投与されるNK細胞は、ドナー由来(同種)細胞である。
上記の通り、いくつかの実施形態では、NK細胞は、腫瘍細胞及びウイルス感染細胞を含む異常細胞を認識及び破壊する。これらの天然免疫細胞の細胞傷害活性は、細胞表面上に存在する阻害性及び活性化受容体の各々からのシグナル伝達の平衡によって調節される。前者は、健常細胞の表面上に発現される自己分子に結合する一方、後者は、異常細胞上に発現されるリガンドに結合する。阻害性受容体に対する活性化受容体の会合の増加により、NK細胞の活性化及び標的細胞の溶解がもたらされる。ナチュラルキラーグループ2メンバーD(NKG2D)は、ストレス及び異常細胞上に発現されるいくつかのリガンドを認識する重要なNK細胞の活性化受容体である。様々なNKG2Dリガンドの表面発現は、健常細胞において一般に低いが、悪性形質転換又はウイルス感染時に上方制御される。NKG2Dによって認識されるリガンドの非限定例として、限定されないが、MICA、MICB、ULBP1、ULBP2、ULBP3、ULBP4、ULBP5及びULBP6並びにNK細胞の細胞溶解又は細胞傷害性機能を制御する、標的細胞上に発現される他の分子が挙げられる。
上記の通り、一般的なキメラ抗原受容体構造は、リガンド結合ドメインをシグナル伝達ドメインに連結する少なくとも1つの膜貫通ドメインを含む。いくつかの実施形態では、しかし、膜貫通ドメインは、シグナル伝達機能を提供することにも役立ち得る。
本明細書に記載された本開示を考慮して、特定の標的細胞(例えば、疾患細胞又は癌細胞)に標的を定めて破壊するために、NK細胞中で生成及び発現され得る様々なキメラ受容体が存在する。そのようなキメラ受容体の非限定的な例を下記でより詳細に論じる。
以下の実験方法及び材料は、以下に開示される非限定的な実験例において用いられる。
ヒト急性リンパ芽球性白血病細胞株REH、ヒト骨肉腫細胞株U−2 OS及びヒト胚性腎線維芽細胞293T(HEK293T)細胞は、アメリカ合衆国培養細胞系統保存機関(ATCC;Manassas,Virginia)から入手した。REH細胞は、10%ウシ胎仔血清(FBS;Hyclone,Logan,Utah)及び1%ペニシリン−ストレプトマイシンを添加したRoswell Park Memorial Instituteシリーズ1640(RPMI−1640;Gibco,Carlsbad、California)中で維持し、増殖させた。HEK293T及びU−2 OS細胞の両方は、10%FBS及び1%ペニシリン−ストレプトマイシンを添加したダルベッコ修飾イーグル培地(DMEM;Hyclone)中で維持し、増殖させた。すべての哺乳動物細胞を5%CO2下において37℃でインキュベートした。
キメラ受容体NKG2D−DAP10−CD3ζを含有するDNAプラスミドを前述のように作製した(Chang et al.Cancer research,Vol.73(6):2013を参照されたい)。伸長ポリメラーゼ連鎖反応(SOE−PCR)を重複させることによるスプライシングを用いて、NKG2D−41BB−CD3ζ構築物を形成する個々のドメインを融合させた。次に、その構築物をマウス幹細胞ウイルス(MSCV)レトロウイルスベクターに挿入した(図3A)。NKG2D−CD16及びNKG2D−CD16−41BBにおける構築物は、GenScript(Nanjing,China)により、コドン最適化し、MSCVベクターに挿入した(図3B)。構築物の配列は、DNA配列決定により検証した。
ヒト末梢血単核球(PBMC)は、健常成人ドナーからの血液サンプルのフィコール密度遠心分離により入手した。NK細胞を拡大するため、PBMCを膜結合性IL−15及び4−1BBリガンド(K562−mb15−41BBL)で遺伝子組換えしたK562とともに培養した。細胞は、40IUのIL−2/mlを2日ごとに添加した幹細胞増殖培地(SCGM;Cell Genix,Freiburg,Germany)中で培養した。
HEK293T細胞にレトロウイルスパッケージングプラスミドを一過性にトランスフェクトすることにより、レトロウイルスの産生を実施した。HEK293T細胞をトランスフェクションの18時間前、まず12ml中2.5×106個の細胞の濃度のDMEMに播種した。次に、この細胞に、各NKG2Dキメラ受容体を含有する3.5μgのMSCVベクター(非限定的な構築物を図1B〜1C及び2A〜2Bに概略的に示す)、3.5μgのpEQ−PAM3及び3.0μgのpRDFをトランスフェクトした。対照として、GFPを含有する空のMSCVベクターを用いた。X−tremeGENE 9 DNAトランスフェクション試薬(Roche,Basel,Switzerland)をトランスフェクションのために用いた。トランスフェクションから24時間後にDMEMを馴化RPMI−1640と交換した。
形質導入したNK細胞を、アルブミンを含有するリン酸緩衝食塩水で1回洗浄し、ウサギ血清2μlを添加した。次に、暗所で細胞をペリジニンクロロフィル(PerCP)コンジュゲート抗ヒトNKG2D抗体(クローン149810;R&D Systems,Minneapolis,USA)で10分間染色した。対照として、形質導入したNK細胞を各々のPerCPコンジュゲートIgGアイソタイプ抗体で染色した。Accuri C6フローサイトメーター(BD,Franklin Lakes,NewJersey)を用いる分析前、すべてのNK細胞を再洗浄し、0.5%ホルムアルデヒド300μlで固定した。対応t検定を用いて、データを分析した。
REH細胞をカルセインAMの赤−オレンジ(Thermo Fisher Scientific,Waltham,Massachusetts)で染色した。REH細胞を96ウェル丸底プレート(CoStar,Corning,New York)に播種した。次に、形質導入したNK細胞を様々なエフェクター:標的(E:T)比で添加した。細胞培養物を37℃及び5%CO2で4時間インキュベートした。Accuri C6フローサイトメーターを用いて、染色した生存可能標的細胞をカウントした。U−2 OS細胞を96ウェル平底白色プレート(Costar)に播種し、4時間インキュベートした。次に、形質導入したNK細胞を異なるE:T比に応じて添加した。次に、細胞培養物をさらに4時間インキュベートした。分析前、Bright−Glo基質(Promega,Madison,Wisconsin)をこの細胞に添加した。生存可能標的細胞からの発光の強度を、FL×800 Fluorescence Reader(Bio Tek,Winooski,Vermont)を用いて測定した。発光の強度と対照との差を細胞傷害性の百分率に変換した。
NK細胞によって産生されるIFNγの量を判定するため、エフェクター及び標的細胞をまず96ウェル丸底プレート内、(1:1のE:T)又はREH不在下で培養した。GolgiPlug(ブレフェルジンA;BD Biosciences)の添加前、細胞を1時間インキュベートした。さらに5時間の培養後、細胞をフィコエリトリン(PE)コンジュゲート抗ヒトCD56抗体(クローンMY31,BD Biosciences)で標識した。専売透過処理試薬を用いて細胞を透過処理し、暗所で40分間インキュベートした。次に、この細胞を専売洗浄緩衝液で洗浄した。細胞内IFNγを、アロフィコシアニン(APC)コンジュゲートIFNγ抗体(クローン25723.11;BD Biosciences)を用いて45分間検出した。次に、この細胞を固定し、Accuri C6フローサイトメーターを用いて分析した。
本明細書で開示される通り、様々な膜貫通及び/又はシグナル伝達ドメインと共役されたNKG2D及び/又はNKG2D変異体を含む様々な構築物を準備する。CD3ζシグナル伝達ドメインを含む構築物の発現及び細胞傷害活性を評価するため、本実験を実施した。上記の方法及び材料に従い、2つのCD3ζ構築物を調製し、試験した。構築物に応じて、用いる方法を容易に調節することで、構築物の生成、発現及び試験にとって必要とされるバリエーションを構成することができる。2つの構築物は、NKG2D−DAP10−CD3ζ及びNKG2D−41BB−CD3ζであった。参照として、図1Aは、内因性NKG2Dを概略的に示す。NK細胞では、NKG2Dの膜貫通領域間のイオン相互作用により、そのアダプタータンパク質DAP10との会合が可能になる(Wu et al.,1999)。リガンド結合時、NKG2Dシグナルは、DAP10上に見出されるシグナル伝達モチーフYxNMを通じて伝達される。CD3ζは、その免疫受容体チロシンに基づく活性化モチーフ(ITAM;Lanier,2008)を通じてシグナルを伝達する。2つの実験構築物をそれぞれ図1B及び1Cに概略的に示す。図1Bは、YxNM及びITAMモチーフの両方を通じてシグナル伝達が生じるNKG2D−DAP10−CD3ζを示す。図1Cは、膜貫通ドメインとしてCD8aヒンジ領域、またシグナル伝達ドメインとして4−1BB及びCD3ζを用いるNKG2D−41BB−CD3ζ構築物を示す。
発現、細胞傷害性及びサイトカイン産生を評価するため、追加的な構築物を作製した。本明細書で提供される通り、いくつかの実施形態は、CD16膜貫通及び/又はシグナル伝達ドメインを用いる、(いくつかの実施形態ではコドン最適化された)切断されたNKG2Dを含む構築物に関する。この実験における評価のために作製した構築物を図2A〜2Bに概略的に示し、それは、A)NKG2D−CD16、及びB)NKG2D−CD16−41BBキメラ受容体の構造を示す。両方のキメラ受容体は、CD16の膜貫通領域に依存し、CD3ζ又はFcRγのいずれかと会合する。これらの構築物を作製するために用いるプラスミドを図3Bに示す。上で考察の通り、いくつかの実施形態では、用いる構築物は、CD3ζ又はFcRγの内因性発現に依存するが、いくつかの実施形態では、キメラ受容体をコードするプラスミド(又は分離プラスミド)は、NK細胞によるCD3ζ及び/又はFcRγの発現を高め、それによりこの細胞の効力を増強するように設計する。
異なる細胞外ドメイン、膜貫通ドメイン及び細胞内エフェクタードメインを有する追加的な構築物を作製し、それらの発現及び細胞傷害性を評価した。この実験において評価のために作製した12の構築物を図14に概略的に示す。これらキメラ受容体変異体の一部は、CD16膜貫通領域に依存し、CD3ζ又はFcRγのいずれかと会合する。上で考察の通り、いくつかの実施形態では、用いる構築物は、CD3ζ又はFcRγの内因性発現に依存するが、いくつかの実施形態では、このキメラ受容体をコードするプラスミド(又は分離プラスミド)は、NK細胞によるCD3ζ及び/又はFcRγの発現を高め、それによりこの細胞の効力を増強するように設計する。上記のように、この構築物の発現レベルを評価した。モックトランスフェクトNK細胞は、MFIによる評価として、低レベルのNKG2D発現を示す(図16A)。それに対し、上記のNKG2D変異体構築物を形質導入したNK細胞は、異なるレベルのNKG2D発現を示し、操作した変異体構築物4及び9は、NK細胞における有意に増強された発現を示した。図16Bは、NKG2D変異体構築物1、4、8、9の、2つのドナーのNK細胞への形質導入後についての代表的なフローサイトメトリーデータを示す。モック形質導入NK細胞と比べて、変異体8及び9の形質導入NK細胞は、キメラ受容体の特に強力な発現を示した。変異体構築物の発現は、形質導入後7日間、2つのドナーのNK細胞において持続し、変異体8及び9は、MFIによる評価として特に上昇したレベルを示した(図16C)。これらのデータは、構築物がNK細胞に有効に導入可能であり、発現されることを実証するために重要である。構築物の発現を樹立しており、それらが細胞傷害性効果を形質導入NK細胞に送達する能力についても評価した。NKG2D変異体構築物4、8及び9の細胞傷害性を、1:1のE:T比でNK細胞に形質導入してから14日後に評価した(図17)。
本明細書で開示される実施形態に記載の異なる細胞外ドメイン、ヒンジ、膜貫通ドメイン及び細胞内エフェクタードメインを有する追加的な構築物を図22に概略的に示す。本実施例において、これらの7つの構築物によって媒介される発現、細胞傷害性、持続性及びサイトカイン産生を、実施例3に記載のNK39構築物の3つ(NK39−5、NK39−6、NK39−10)並びに膜結合性インターロイキン15をバイシストロン性に発現するNK16の一バージョン(NK26−8)に対して評価した。本明細書で開示されるいくつかの実施形態によると、複数のシグナル伝達領域を用い得る。これらのキメラ受容体変異体の一部は、CD3ζ及び/又は別のシグナル伝達ドメイン(例えば、OX40、CD28及び/又は4−1BB同時刺激ドメイン)を含むエフェクタードメインに依存し、リガンド結合時にシグナルを伝達する一方、他のキメラ受容体変異体は、二量体化を通じて完全長CD3ζ分子をシナプスに動員するCD3ζ膜貫通ドメインを含む。本明細書で開示される通り、これらの構築物は、膜結合性IL15を同時発現するようにさらに設計する。

- NK細胞の細胞傷害性の増強を、それを必要とする哺乳動物において行うための方法であって、NK細胞を前記哺乳動物に投与することを含み、前記NK細胞は、請求項35〜112のいずれか一項に記載のポリヌクレオチドによってコードされるキメラ受容体を発現する、方法。
- 前記NK細胞は、患者から単離された自家細胞である、請求項118に記載の方法。
- 前記NK細胞は、ドナーから単離された同種細胞である、請求項118に記載の方法。
