Description

多くの疾患の基礎をなす(ウイルス感染及び悪性細胞を含む)異常細胞の出現及び持続は、前記異常細胞に対する不十分な免疫応答によって可能になる。免疫療法の目標は、患者の免疫系の応答を開始又は増強する例えば免疫細胞、例えばナチュラルキラー(NK)細胞が損傷又は異常細胞を損傷させるか、殺滅するか、又は他に阻害する能力を後押しすることである。1つの免疫療法アプローチは、異常細胞の認識及び破壊を標的にするための免疫細胞における活性化キメラ受容体の組換え発現である。一般に、キメラ受容体は、標的細胞上のリガンドを認識する細胞外受容体ドメイン、アンカー膜貫通ドメイン及びリガンド結合時のシグナルの活性化を伝達するエフェクタードメインを含む。本明細書で開示されるいくつかの実施形態では、その一般的構造を有するか又はその一般的構造におけるバリエーションを有する活性化キメラ受容体が用いられる。加えて、いくつかの実施形態では、膜貫通ドメイン及びエフェクタードメインは、一緒に融合された別々のペプチドである。いくつかの他の実施形態では、膜貫通及びエフェクタードメインは、同じペプチドに由来する。いくつかのかかる実施形態では、膜貫通及びエフェクタードメインは、単一のペプチド(例えば、膜を貫通することに加え、平衡が保たれ、シグナルカスケードを開始させる1つのペプチド)を含む。以下により詳細に考察される通り、免疫細胞(例えばNK細胞)において所望の程度の発現を示し、非標的細胞に対する有害作用を回避する程度の標的結合活性と平衡した、NK細胞から細胞傷害活性を誘導する活性化キメラ受容体構築物を作製するため、切断、突然変異、追加的なリンカー/スペーサーエレメント、二量体などが用いられる。免疫細胞の表面上での本明細書で開示されるような活性化キメラ受容体の組換え発現により、目的の異常細胞に対する免疫細胞の標的化が再誘導され、且つ会合時の免疫活性化が増強される可能性がある。
1つの免疫療法アプローチは、活性化キメラ受容体を発現するように操作されたT細胞を患者に投与し、陽性免疫応答を誘発することを含む。しかし、この手法の欠点は、患者における移植片対宿主病の誘導を予防するために自家細胞の使用が必要である点である。本明細書で開示されるいくつかの実施形態において提供される通り、操作されたNK細胞を含む組成物は、いくつかの利点を享受する。例えば、自家又はドナー由来同種細胞のいずれかは、NK細胞アプローチで用いることができる。加えて、いくつかの実施形態によると、本明細書で提供されるような操作されたNK細胞は、正常細胞に対して細胞傷害性を増強することが有意でない。さらに、NK細胞は、活性化されると、有意な細胞傷害性効果を有する。これを考慮すると、本明細書で提供されるような操作されたNK細胞が、その細胞傷害性効果をさらに高めることにより、罹患した標的細胞を選択的に殺滅するといったさらにより有効な手段を提供し得ることは、想定外である。したがって、いくつかの実施形態では、癌又は感染性疾患を治療又は予防する方法であって、治療有効量の、本明細書に記載の活性化キメラ受容体を発現するNK細胞を投与することを含む方法が提供される。一実施形態では、投与されるNK細胞は、自家細胞である。さらなる実施形態では、投与されるNK細胞は、ドナー由来(同種)細胞である。
上記の通り、いくつかの実施形態では、NK細胞は、腫瘍細胞及びウイルス感染細胞を含む異常細胞を認識及び破壊する。これらの天然免疫細胞の細胞傷害活性は、細胞表面上に存在する阻害性及び活性化受容体の各々からのシグナル伝達の平衡によって調節される。前者は、健常細胞の表面上に発現される「自己」分子に結合する一方、後者は、異常細胞上に発現されるリガンドに結合する。根底にある論理は、「自己」分子と相互作用するNK細胞上の阻害性受容体が活性化されることがなく、それによりNK細胞による「自己」分子を有する細胞の破壊が免れることである。それに対し、活性化受容体を発現するNK細胞が、例えば異常細胞上の「非自己」リガンドと相互作用するとき、NK細胞は活性化され、それに続いて細胞傷害性効果が現れる。したがって、阻害性受容体と比べての活性化受容体の会合の増加により、NK細胞の活性化及び標的細胞の溶解がもたらされる。
上記の通り、一般的な活性化キメラ受容体構造は、リガンド結合ドメインをシグナル伝達ドメインに連結する少なくとも1つの膜貫通ドメインを含む。いくつかの実施形態では、しかし、膜貫通ドメインは、シグナル伝達機能を提供することにも役立ち得る。
本明細書に記載された本開示を考慮して、特定の標的細胞(例えば、疾患細胞又は癌細胞)に標的を定めて破壊するために、NK細胞中で生成及び発現され得る様々な活性化キメラ受容体が存在する。そのようなキメラ受容体の非限定的な例を下記でより詳細に論じる。
以下の実験方法及び材料は、下に開示する非限定的な実験例において用いた。
ヒト腫瘍細胞株SKBR3、HT-29、U2-OSは、アメリカ合衆国培養細胞系統保存機関(American Type Culture Collection)(ATCC;Rockville,MD)から購入した。ユーイング肉腫細胞株ES8及びEW8は、St.Jude Research Hospital(Memphis,TN)の組織貯蔵所から入手した。細胞株をRPMI-1640(ThermoFisher,Waltham,MA)中で維持し;培地に10%ウシ胎仔血清(FBS;GE Healthcare,Chicago,IL)及び抗生物質を添加した。細胞株に、緑色蛍光タンパク質(GFP)又はルシフェラーゼのいずれかを有するマウス幹細胞ウイルス(MSCV)レトロウイルスベクター(St.Jude Children’s Research HospitalのVector Development and Production Shared Resource,Memphis,TNから入手)を形質導入した。
末梢血サンプルは、National University Hospital Blood Bank,Singaporeでの健常成人ドナーからの血小板輸血の匿名扱いの廃棄副生物から得た。
すべての構築物をコードするプラスミドは、Genescript(Nanjing,China)により合成した。構築物を有するARD114シュードタイプMSCVレトロウイルスを用いて、NK細胞に形質導入した。レトロウイルスベクター培養上清を、レトロネクチン(Takara、大津、日本)でコーティングしたポリプロピレン管に添加し;遠心分離及び上清の除去後、増殖したNK細胞(3×105個)を該管に添加し、37℃で12時間放置し;新しいウイルス上清を12時間ごとに全部で6回添加した。次に、実験の時期まで、FBS、抗生物質及び400IU/mlのIL-2を有するRPMI中で細胞を維持した。
抗NKG2Cの表面発現は、抗NKG2C-フィコエリスリン(PE)抗体(Miltenyi Biotech)を用いて検出した。NKG2Aの発現は、PE又はAPC(Miltenyi Biotech)にコンジュゲートされた抗NKG2A抗体を用いて検出した。CD94の発現は、抗CD94-APC(BD Biosciences)を用いて検出した。
GFP/ルシフェラーゼを形質導入した標的細胞を、10%FBSを有するRPMI-1640に懸濁し、96ウェル平底プレート(Costar,Corning,NY)に蒔いた。プレートをインキュベーター内に少なくとも4時間置き、NK細胞を添加する前に細胞接着を可能にした。次に、10%FBSを有するRPMI-1640に懸濁した、様々な受容体又はGFP単独を発現する増殖したNK細胞を様々なエフェクター対標的(E:T)比で添加し、標的細胞とともに4時間共培養した。培養の終了時、Flx 800プレートリーダー(BioTek,Winooski,VT)を用いて、BrightGlo(Promega,Fitchburg,WI)をウェルに添加後、生細胞の数を測定した。
本明細書に開示の通り、様々な膜貫通ドメインと共役された細胞外受容体ドメインを含む様々な活性化キメラ受容体構築物を準備する。本実験は、HLA-E/ペプチド複合体に対する高親和性について改変した活性化ナチュラルキラーグループ2メンバーC(NKG2C)変異体の発現及び細胞傷害性活性を評価するために実施した。活性化NKG2C受容体構築物は、上記の方法及び材料に従い、調製し、試験した。構築物に応じて、使用方法は、構築物の作製、発現、及び試験に要求されるバリエーションに対処するため、容易に調節することができる。
本明細書に開示の通り、様々な膜貫通ドメインと共役された細胞外受容体ドメインを含む様々な活性化キメラ受容体構築物を準備する。本実験は、活性化ナチュラルキラーグループ2メンバーA(NKG2A)/CD94受容体構築物の発現及び細胞傷害性活性を評価するため、実施した。活性化NKG2A/CD94受容体構築物は、上記の方法及び材料に従い、調製し、試験した。構築物に応じて、使用方法は、構築物の作製、発現、及び試験に要求されるバリエーションに対処するため、容易に調節することができる。
本明細書に開示される活性化NK受容体構築物が標的細胞(例えば腫瘍細胞)に対する細胞傷害性効果を発揮する能力を実証するため、結腸直腸腺癌の異種移植片モデルを用いた。HT-29細胞は、不死化した結腸直腸腺癌細胞株である。HT-29細胞に、本実施例中で用いる改変活性化受容体によって認識される標的の一部であるHLA-Eを発現するように、HT-29細胞にGpHLA-Eとルシフェラーゼの双方を形質導入した(D12(SIIS)2C/94構築物をHLA-E/ペプチド複合体に対する高親和性について改変する(165-168SIIS))。形質導入HT-29細胞を、1×105個の細胞/マウスの用量で、NOD-SCIDIL2RGヌルマウス12匹の各々に腹腔内注射した。その後、NK細胞(1×107個の細胞)を、3、6、10及び13日目に注射した。NK細胞に、GFPのみ(「対照」)又はD12(SIIS)2C/94構築物のいずれかを形質導入した。マウスにはまた、IL-2(20,000IU)を、腹腔内注射により週3回投与した。Xenogen Spectrum装置を用いて、腹側及び背側の生物発光を測定し、腫瘍量を評価した(各記号は、腹側及び背側の読み出し値の平均である)。

から選択される追加的なポリヌクレオチドを含む遺伝子改変ナチュラルキラー細胞。

NK細胞の細胞傷害性の増強を、それを必要とする哺乳動物において行うための方法であって、NK細胞を前記哺乳動物に投与することを含み、前記NK細胞は、項1~14のいずれか一項に記載のポリヌクレオチドによってコードされる活性化キメラ受容体を発現する、方法。

Claims (25)
- HLAクラスI組織適合性抗原、α鎖E(HLA-E)/ペプチド複合体に結合する活性化キメラ受容体を発現する遺伝子改変ナチュラルキラー(NK)細胞であって、前記活性化キメラ受容体が、前記HLA-E/ペプチド複合体への結合後に活性化及び/又は同時刺激シグナルを伝達し、ここで前記活性化キメラ受容体が、
a)ナチュラルキラーグループ2メンバーA(NKG2A)の断片を含む細胞外受容体ドメイン、
ここでNKG2Aの前記断片が、膜貫通領域及び細胞内シグナル伝達ドメインを含むエフェクタードメインに共役され、
天然NKG2Aが、HLA-E/ペプチド複合体への結合後、阻害性シグナルを伝達し、且つ
NKG2Aの前記断片が、HLA-E/ペプチド複合体への結合後、活性化及び/又は同時刺激シグナルを伝達し、
前記活性化キメラ受容体が、配列番号61の核酸配列によってコードされ、且つ
前記細胞外受容体ドメインが、CD8a膜貫通ドメイン並びに4-1BB及びCD3ζを含むエフェクタードメインに共役されている;又は
b)ナチュラルキラーグループ2メンバーC(NKG2C)の改変変異体を含む細胞外受容体ドメイン、
ここで非改変NKG2Cが、HLA-E/ペプチド複合体に対して低い結合親和性を有し、
前記改変NKG2C変異体が、HLA-E/ペプチド複合体に対して、天然NKG2Cと比べて増強された結合親和性を有し、且つ
前記改変NKG2C変異体が、配列番号63の核酸配列によってコードされ、且つ
前記細胞外受容体ドメインが、膜貫通領域及び細胞内シグナル伝達ドメインを含むエフェクタードメインに共役されている;
を含む、遺伝子改変ナチュラルキラー(NK)細胞。 - DNAX活性化タンパク質12(DAP12)をコードするポリヌクレオチドをさらに含む、請求項1に記載の遺伝子改変NK細胞。
- 配列番号59の核酸配列によってコードされるキメラCD94をさらに含む、請求項1又は2に記載の遺伝子改変NK細胞。
- 前記遺伝子改変NK細胞が、(a)ナチュラルキラーグループ2メンバーD(NKG2D)の天然リガンドに結合するペプチドを含む細胞外受容体ドメイン;並びに(b)膜貫通領域及び細胞内シグナル伝達ドメインを含むエフェクタードメインを含むキメラ受容体をさらに含む、請求項1~3のいずれか一項に記載の遺伝子改変NK細胞。
- 前記遺伝子改変NK細胞が、天然NKG2Aの転写又は翻訳を特異的に阻害する短いヘアピンRNA(shRNA)をコードするポリヌクレオチドをさらに含み、
ここで前記shRNAが、前記天然NKG2A遺伝子にストリンジェントな条件下でハイブリダイズするヌクレオチド配列を含み、前記shRNAが、前記NKG2A断片に不在である前記NKG2A遺伝子における標的配列に対して実質的に同一なヌクレオチド配列を含むセンス断片、及びアンチセンス断片を含み、ここで前記センス及びアンチセンス断片が、ループ断片によって分離される、請求項1~4のいずれか一項に記載の遺伝子改変NK細胞。 - 前記NK細胞が、膜結合型インターロイキン15(mbIL15)を追加的に発現する、請求項1~5のいずれか一項に記載の遺伝子改変NK細胞。
- ドナーから単離された同種細胞である、請求項1~6のいずれか一項に記載の遺伝子改変NK細胞。
- 前記NK細胞が、天然NKG2Aの表面発現を欠く、請求項1~7のいずれか一項に記載の遺伝子改変NK細胞。
- 請求項1~8のいずれか一項に記載の遺伝子改変NK細胞を含む、NK細胞の細胞傷害性の増強を必要とする哺乳動物においてNK細胞の細胞傷害性を増強するための医薬組成物。
- HLAクラスI組織適合性抗原、α鎖E(HLA-E)/ペプチド複合体に結合する活性化キメラ受容体であって、前記活性化キメラ受容体が、前記HLA-E/ペプチド複合体への結合後に活性化及び/又は同時刺激シグナルを伝達し、ここで前記活性化キメラ受容体が、
a)ナチュラルキラーグループ2メンバーA(NKG2A)の断片を含む細胞外受容体ドメイン、
ここでNKG2Aの前記断片が、膜貫通領域及び細胞内シグナル伝達ドメインを含むエフェクタードメインに共役され、
天然NKG2Aが、HLA-E/ペプチド複合体への結合後、阻害性シグナルを伝達し、且つ
NKG2Aの前記断片が、HLA-E/ペプチド複合体への結合後、活性化及び/又は同時刺激シグナルを伝達し、
前記活性化キメラ受容体が、配列番号61の核酸配列によってコードされ、且つ
前記細胞外受容体ドメインが、CD8a膜貫通ドメイン並びに4-1BB及びCD3ζを含むエフェクタードメインに共役されている;又は
b)ナチュラルキラーグループ2メンバーC(NKG2C)の改変変異体を含む細胞外受容体ドメイン、
ここで非改変NKG2Cが、HLA-E/ペプチド複合体に対して低い結合親和性を有し、
前記改変NKG2C変異体が、HLA-E/ペプチド複合体に対して、天然NKG2Cと比べて増強された結合親和性を有し、且つ
前記改変NKG2C変異体が、配列番号63の核酸配列によってコードされ、且つ
前記細胞外受容体ドメインが、膜貫通領域及び細胞内シグナル伝達ドメインを含むエフェクタードメインに共役されている;
を含む、活性化キメラ受容体。 - 前記細胞外受容体ドメインが、天然NKG2C膜貫通領域及び天然NKG2C細胞内シグナル伝達ドメインに共役された改変NKG2C変異体を含む、請求項10に記載の活性化キメラ受容体。
- 前記活性化受容体が、DNAX活性化タンパク質12(DAP12)をさらに含む、請求項11に記載の活性化キメラ受容体。
- 配列番号59の核酸配列によってコードされるキメラCD94をさらに含む、請求項10~12のいずれか一項に記載の活性化キメラ受容体。

天然NKG2Aが、HLA-E/ペプチド複合体への結合後、阻害性シグナルを伝達し、且つ
NKG2Aの前記断片が、HLA-E/ペプチド複合体への結合後、活性化及び/又は同時刺激シグナルを伝達し、
前記活性化キメラ受容体が、配列番号61の核酸配列によってコードされ、且つ
前記細胞外受容体ドメインが、CD8a膜貫通ドメイン並びに4-1BB及びCD3ζを含むエフェクタードメインに共役されている;又は
b)ナチュラルキラーグループ2メンバーC(NKG2C)の改変変異体を含む細胞外受容体ドメイン、

前記細胞外受容体ドメインが、膜貫通領域及び細胞内シグナル伝達ドメインを含むエフェクタードメインに共役されている;
を含む、ポリヌクレオチド。 - 前記ポリヌクレオチドが、DNAX活性化タンパク質12(DAP12)をさらにコードする、請求項14に記載のポリヌクレオチド。

- 前記ポリヌクレオチドが、(a)ナチュラルキラーグループ2メンバーD(NKG2D)の天然リガンドに結合するペプチドを含む細胞外受容体ドメイン;並びに(b)膜貫通領域及び細胞内シグナル伝達ドメインを含むエフェクタードメインを含むキメラ受容体をさらにコードする、請求項14~16のいずれか一項に記載のポリヌクレオチド。
- 前記ポリヌクレオチドが、天然NKG2Aの転写又は翻訳を特異的に阻害する短いヘアピンRNA(shRNA)をさらにコードし、
ここで前記shRNAが、前記天然NKG2A遺伝子にストリンジェントな条件下でハイブリダイズするヌクレオチド配列を含み、前記shRNAが、前記NKG2A断片に不在である前記NKG2A遺伝子における標的配列に対して実質的に同一なヌクレオチド配列を含むセンス断片、及びアンチセンス断片を含み、ここで前記センス及びアンチセンス断片が、ループ断片によって分離される、請求項14~17のいずれか一項に記載のポリヌクレオチド。 - 前記ポリヌクレオチドが、膜結合型インターロイキン15(mbIL15)をコードする追加的な構築物と同時発現される、請求項14~18のいずれか一項に記載のポリヌクレオチド。
- 請求項14~19のいずれか一項に記載のポリヌクレオチドを含むベクターであって、前記ポリヌクレオチドは、前記活性化キメラ受容体の発現のための少なくとも1つの調節エレメントに作動可能に連結されている、ベクター。
- 前記ベクターは、レトロウイルスベクターである、請求項20に記載のベクター。
- 対象における癌の治療のための医薬組成物であって、前記組成物は、活性化キメラ受容体を含む遺伝子改変ナチュラルキラー(NK)細胞を含み、前記活性化キメラ受容体が、
a)ナチュラルキラーグループ2メンバーA(NKG2A)の断片を含む細胞外受容体ドメイン、
ここでNKG2Aの前記断片が、膜貫通領域及び細胞内シグナル伝達ドメインを含むエフェクタードメインに共役され、
天然NKG2Aが、HLA-E/ペプチド複合体への結合後、阻害性シグナルを伝達し、
前記活性化キメラ受容体が、配列番号61の核酸配列によってコードされ、且つ
NKG2Aの前記断片が、HLA-E/ペプチド複合体への結合後、活性化及び/又は同時刺激シグナルを伝達し、且つ

(i)(a)ナチュラルキラーグループ2メンバーD(NKG2D)の
天然リガンドに結合するペプチドを含む細胞外受容体ドメ
イン;及び
(b)膜貫通領域及び細胞内シグナル伝達ドメインを含むエフェ
クタードメイン
を含む、キメラ受容体;
(ii)膜結合型インターロイキン15(mbIL15);
(iii)天然NKG2Aの転写又は翻訳を特異的に阻害する短いヘアピンRNA(shRNA);並びに
(iv)(i)、(ii)、及び(iii)のいずれかの組み合わせ
