技術分野
本発明は、一般に、ヒト人工多能性幹細胞(iPSC)を生成し、培養するためのフィーダーフリー条件を含めた、幹細胞を培養するための細胞培養条件、培地、および培養プラットフォームに関する。

PARP阻害剤は、ポリ(ADP−リボース)ポリメラーゼ(「PARP」)を阻害する。PARPタンパク質は、細胞においてDNA修復経路を調節するように機能するDNA修復酵素である。PARPは、塩基除去修復(BER)経路に関与し、PARPを阻害することにより、再プログラミングの間の細胞のゲノム安定性または多能性細胞の維持が促進され得る。本発明の細胞培養培地において使用するための例示的なPARP阻害剤としては、限定することなく、イニパリブ(iniparib)、ベリパリブ(veliparib)、およびオラパリブ(olaparib)(AZD−2281)が挙げられる。
下記の表には、単細胞継代および細胞選別および富化の手順を受けている細胞の生存能力および多能性を増強するため、再プログラミングプロセスを増強するため、ならびに多能性幹細胞を未分化の状態に維持するために使用することができる分子、およびそれらが影響を及ぼすシグナル伝達経路の例が列挙されている。
本発明のいくつかの実施形態では、本発明の細胞培養培地は、サイトカインおよび/または成長因子を実質的に含まず、必要に応じて、フィーダーフリー環境である。他の実施形態では、細胞培養培地は、補充物、例えば、血清、抽出物、成長因子、ホルモン、サイトカインなどを含有する。
任意の適切な容器または細胞培養容器を、基本培地および/または細胞培養補充物中の細胞培養物の支持体として使用することができる。支持体上の基質コーティングは必要ない。しかし、培養容器の表面を、接着促進性基層(例えば、コラーゲン、フィブロネクチン、RGDを含有するポリペプチド、ゼラチンなど)でコーティングすることにより、細胞の付着が促進され、それにより、本明細書に開示されている細胞培養培地および補充物の効果が増強され得る。細胞を培養し、継代するための適切な基質は当技術分野で公知であり、それらとしては、限定することなく、ゼラチン、ラミニン、フィブロネクチン、コラーゲン、エラスチン、オステオポンチン、天然に存在する細胞系により産生されたマトリックス、例えば、Matrigel(商標)の混合物、ならびに、合成または人工の表面、例えば、ポリアミン単層およびカルボキシ末端単層が挙げられる。
フィーダーフリー環境
細胞は、一般には、フィーダー細胞上でまたはフィーダー細胞で予備馴化され、ウシ胎仔血清を含有する培養環境で培養されているが、そのような環境は、臨床的な使用および治療的な使用のための細胞を作製するためには不適当な場合がある。例えば、そのような異物が混入した環境において培養された細胞は、一般に、動物の構成成分に曝露することにより、免疫拒絶反応および処置される患者に未確認の病原体が伝染する重大な危険性がもたらされる可能性があり、また、動物レトロウイルスを潜在的に再活性化する恐れがあるので、ヒト細胞移植には不適当であるとみなされる。動物を含まない培地を用いた培養系、例えば本発明のフィーダーフリー環境により、臨床グレードの細胞系、特にhESC細胞系およびiPSC細胞系を作製することが容易になる。
細胞を単細胞、例えば、単細胞懸濁物に解離させることは、酵素的または機械的な手段によって実現することができる。細胞を単細胞に解離させることができる当技術分野で公知の任意の酵素剤を、本発明の方法において使用することができる。本発明の一実施形態では、解離作用剤は、トリプシン/EDTA、TrypLE−Select、コラゲナーゼIVおよびディスパーゼから選択される。

フィーダーフリーに適合させる前に、ヒト人工多能性幹細胞(hiPSC)を、フィーダー細胞(マイトマイシンCで処理したマウス胚線維芽細胞(MEF)細胞(Millipore))上で維持し、従来の培地で培養した。本出願において使用される場合、「従来の培地」とは、DMEM/F12(Mediatech)、10ng/mLのbFGF(Invitrogen)、20%v/vのノックアウト血清代替物(Invitrogen)、1%v/vの非必須アミノ酸(Mediatech)、2mMのL−グルタミン(Mediatech)および100μMのβ−メルカプトエタノール(Invitrogen)を含有する基本ヒト胚性幹細胞(hESC)培地を指す。従来の培地は、表1の最初の項にも記載されている。hiPSCを5〜7日ごとに、先の細いガラスピペットを使用してコロニーを小さな片に機械的に切り、こすり取ることによって継代し(凝集塊継代)、採取し、新しく播種した、マイトマイシンC処理したMEF細胞上に1:3〜1:6に希釈継代し、hESC培地を毎日添加した。細胞培養物を、加湿インキュベーターセットにおいて37℃、5%CO2で維持した。フィーダー細胞を伴う従来の培地中で、凝集塊継代を用いて細胞を培養するステップは、本明細書では、「従来の培養」と称される。
B.再プログラミングの誘導
再プログラミングプロセスを開始するために、再プログラミング因子の異所性発現(ヒトOct4、Sox2、Klf4、c−Myc、Lin28、およびNanogの多様な組合せで)をレンチウイルスの形質導入または他の方法、例えば、タンパク質のみの処理を用いて実現した。ほとんどの場合、出発細胞は、10%集密度(すなわち、6ウェルプレートのウェル当たり細胞1×105個)で、ゼラチン(Mediatech)でコーティングした表面上にプレーティングした。ウイルス感染の方法については、新しく採取したレンチウイルスを出発細胞に1:2希釈で加え、4μg/mLのポリブレン(Millipore)を補充し、32℃、650gで1.5時間スピン感染させた。培養物を37℃、5%CO2に移してさらに7時間置いた。インキュベーションが達成されたら、細胞をPBSで3回洗浄し、新鮮な培地を供給した。細胞、例えば、IMR90線維芽細胞をフィーダーフリー培養系において感染させることは難しく、このプロセスを、最初の感染の48時間後にもう1回繰り返した。再プログラミングの誘導の非遺伝学的方法、例えば、細胞に直接タンパク質を適用することの使用について、8μg/mLの再プログラミング因子からなるタンパク質混合物、またはカクテルを細胞溶液に添加し、24時間維持してから培地を換えた。このステップをさらに2〜4回繰り返した。出発細胞は全て、それら自体のそれぞれの体細胞用培地で、最初にタンパク質を添加した後4日目まで培養し、その時点で培地を、一部は体細胞用培地に、そして一部は従来のhESC培地に切り換えた。集密になったら(通常4〜6日)、細胞をトリプシン処理し、等量の培地と混合し、300gで5分間遠心沈澱させ、1:1の体細胞/従来のhESC培地に再懸濁させ、より大きな培養プレートへと1:4〜1:6で拡大した。例えば、6ウェルプレートの2つのウェル中の細胞を通常10cmディッシュ上に拡大する。拡大した次の日に、培地を従来のhESC培地に完全に切り換える。拡大した細胞が集密に達したら(通常8〜12日)、細胞を、富化(独特の集団富化を参照されたい)するために処理する。全ての場合において、培地を常套的に1日おきに交換した。
出発細胞を、Oct4および/またはKlf4および/またはSox2および/またはMycを含有する個々のレンチウイルス構築物またはポリシストロニックベクターを含めた種々の戦略を用いて再プログラミングされるように誘導し、およそ8〜12日間(上記を参照されたい)培養した後、細胞を単細胞に解離させ(多能性幹細胞の細胞培養を参照されたい)、種々の多能性の表面マーカー、体細胞のマーカーおよび/または不完全な再プログラミングのマーカーで染色した。簡単に述べると、解離細胞を、ハンクス平衡塩類溶液(Invitrogen)、4%ウシ胎仔血清(Invitrogen)および10mMのHepes(Invitrogen)を含有する染色溶液に再懸濁させ、氷上で保持した。製造者推奨の希釈に従って、結合体化させた一次抗体を細胞溶液に添加し、溶液を氷上で15分間インキュベートした。細胞溶液を洗浄し、染色緩衝液に再懸濁させ、氷上で維持した。この時点で、蛍光活性化細胞選別(Fluorescent Activated Cell Sorting)(BD Biosciences、以下を参照されたい)および磁気細胞選別(Magnetic Cell Sorting)(Miltenyi Biotec、以下を参照されたい)を含めた種々の富化/枯渇戦略をとった。
細胞を4%v/vのパラホルムアルデヒド(Alfa Aesar)中に30秒間固定し、PBSで3回洗浄し、アルカリホスファターゼ染色キット(Alkaline Phosphatase Staining Kit)(Sigma−Aldrich)で染色した。簡単に述べると、亜硝酸ナトリウム溶液(Sodium Nitrite Solution)1mLをFRV−アルカリ性溶液(FRV−Alkaline Solution)1mLに加え、混合し、25℃で2分間インキュベートした。次いで、この溶液を45mLのH2Oと混合し、その後、ナフトールAS−BIアルカリ性溶液(Naphthol AS−BI Alkaline Solution)1mLを加えた。アルカリ性色素混合物を固定した細胞に加え、25℃で15分間インキュベートした後、PBS洗浄した。次いで、細胞をアルカリホスファターゼの存在についてスコア化した。
細胞を、4%v/vのパラホルムアルデヒド(Alfa Aesar)を使用して15分間固定し、0.2%v/vのTween(PBST)(Fisher Scientific)を含有するPBSで3回洗浄し、PBS中0.15%v/vのトリトンX−100(TritonX−100)(Sigma−Aldrich)を用いて25℃で1時間透過性を上げた。透過性を上げた後、細胞を、PBST(Fisher Scientific)中1%v/vのBSA(Invitrogen)(PBSTB)を用いて25℃で30分間ブロッキングした。PBSTBを穏やかに除去した後、細胞を、PBSTB中の一次抗体と一緒に4℃で一晩インキュベートした。この試験において使用した一次抗体は、Nanog(Abcam)、Tra−1−60(BD Biosciences)、Tra−181(BD Biosciences)、SSEA4(BD Biosciences)、β−III チューブリン(β−III Tubulin)(R&D Systems)、α−平滑筋アクチン(α−Smooth Muscle Actin)(Sigma)およびSox17(R&D Systems)を含んだ。一晩インキュベートした後、細胞をPBSTで3回洗浄し、PBSTB中に1:200希釈した二次抗体(Alexa 488または555;Invitrogen)を用いて25℃で1時間染色した。細胞をPBST中で3回洗浄し、Hoechst色素(Invitrogen)で染色した。染色された細胞の画像を蛍光顕微鏡検査およびCCDカメラを使用して捕捉した。
フィーダーフリーiPSCを、単層として、および胚様体としての両方で分化させた。単層分化については、細胞は通常、分化の際にそれらの増殖を減少させるので、iPSCを集密近くまで到達させた後に分化培地に切り換えた。簡単に述べると、集密になったら、SMC4培地を、DMEM/F12(Mediatech)、20% ウシ胎仔血清(Invitrogen)、1% 非必須アミノ酸(Mediatech)、2mMのL−グルタミン(Mediatech)および100μMのβ−メルカプトエタノールを含有する分化培地に切り換えた。培地を切り換えたら、iPSCを14日間分化させた。培地を2〜3日ごとに替えた。胚葉体(「EB」)の形成および分化については、hiPSCを、アキュターゼ(Millipore)を用いて単細胞に解離し、分化培地に再懸濁させて、最終濃度を1mL当たり細胞75,000個にし、5uMのチアゾビビン(Thiazovivan)を加えた。細胞を、V底96ウェルの非組織培養プレート(Nunc)にウェル当たり100μLで播種し、950gで5分間遠心分離した。翌日、密集した「球様凝集塊」を超低結合6ウェルプレート(Corning)にP1000を用いておよそウェル当たりEB30〜40個で移した。7日後に、移したEBをMatrigelでコーティングした6ウェルプレートに1:1で移した。3週間培養した後、細胞を固定し、染色した。
RNAを、PicoPure RNA Isolationキット(MDS Analytical Technologies)を使用して単離し、0.5μgのRNAを用いて、iScript cDNA Synthesis Kit(Bio−Rad)を使用して第一鎖cDNAを生成した。相対的な遺伝子発現レベルを、TaqMan Fast Universal PCR Master Mix(Applied Biosystems)および下で表3に列挙されているFAMで標識したTaqManプローブを使用して決定した。
全RNAを、Pico Pure RNA Isolation Kit(Molecular Devices,Sunnyvale,CA)を使用して細胞から単離した。簡単に述べると、ビオチン化したaRNAを、MessageAmp II aRNA Amplification Kit(Applied Biosystems/Ambion、Austin、TX)用の標準のプロトコールを使用して、任意選択のSecond Round Amplificationを利用して調製し、次いで、MessageAmp II Biotin Enhanced Kit(Applied Biosystems/Ambion、Austin、TX)を使用し、標準のプロトコールを用いてビオチン標識したaRNAへと転写した。ビオチン標識したaRNAをAffymetrixの推奨に従って精製し、断片化した。20μgの断片化したaRNAを使用して、Human Genome U133 Plus2.0チップ(Affymetrix Inc. Santa Clara、CA)と、45℃で16時間にわたってハイブリダイズさせた。アレイを洗浄し、Affymetrix Fluidics Station 450で染色し、Affymetrix GeneChip Scanner 3000 7Gを使用してスキャンした。画像データを、Affymetrix Expression Consoleソフトウェアを使用し、初期状態の解析設定を用いて解析した。アレイを、対数尺度ロバストマルチアレイ解析(log scale robust multi−array analysis)(RMA)によって正規化し、Spotfire for Genomics 3.1(Tibco Spotfire、Palo Alto、CA)で可視化した。
20のG−バンド染色した中期細胞に対する細胞遺伝学的分析を、Madison、WIに所在するCell Line Geneticsが実施した。
フィーダーフリー培養環境および酵素による単細胞への解離および多能性幹細胞の継代を可能にするための細胞培養条件および方法
本実施例は、多能性細胞集団の培養および解離に関する。そのような細胞集団としては、これらに限定されないが、胚性幹細胞(ESC)および人工多能性細胞、例えば、体細胞核移植(SCNT)によって、または多能性因子を導入することによって生成されるもの−人工多能性幹細胞(iPSC)が挙げられる。多能性幹細胞の培養条件には、伝統的に、放射線照射またはマイトマイシンC処理によって有糸分裂が不活性にされているが、幹細胞の培養を支持するために必要な成長因子および栄養分を提供するフィーダー細胞の使用が含まれていた。フィーダー細胞を使用せずに幹細胞集団を培養するステップは、幹細胞の均一な集団が必要である研究および工業的な適用にとって、または拡大した工業的活動が幹細胞生成物のための異種を含まない定義済みの培養条件を必要とする、研究および工業的な適用にとって有利になる。本実施例では、特定の細胞シグナル伝達経路のいくつかの低分子修飾因子を試験して、個々の低分子または低分子の組合せ(「カクテル」)をフィーダーフリー系における多能性細胞の培養を増強するために使用することができるかどうかを確立した。
多能性および細胞の生存能力を維持しながら多能性細胞からの単細胞選別を可能にするための方法および培養条件
実施例2に記載の通り、ESCまたはiPSCの幹細胞培養物は、常套的にフィーダー細胞上で培養し、細胞コロニーを手動で選択することによって継代し、その後それを機械的に解離させた後に再プレーティングする。熟練した研究者は、多能性、非分化特性を有する幹細胞コロニーを、コロニー形態に基づいて認識し、これを、多能性細胞を選択する方法として用いることができる。したがって、多能性集団または所望の特性を有する集団を、一部の細胞があまり望ましくない特性を有する、例えば、培養物または死細胞の凝集塊中の分化の徴候を示す細胞の細胞集団から選び取り、手動で富化することができる。このプロセスは、労力を要し、所望の細胞集団を選び取ることは熟練した研究者に依存する。したがって、細胞を所望の特性に基づいて個別に選択する場合に細胞富化または選別技術を用いることは、この分野に大きな利益を与えることになる。現在利用可能な技法、例えば、磁気活性化細胞選別(MACS)または蛍光活性化細胞選別(FACS)を用いたそのような富化ステップには、多能性細胞集団を単細胞様式へと酵素により継代した後に、富化し、また培養物に播種することが必要になる。さらに、フィーダーにより支持した培養物を使用することは、これらの技法にはあまり望ましくないことになり、フィーダーフリー培養系を用いることが必要である。
フィーダーフリー培養系において細胞を多能性の状態に効率的に再プログラミングさせる方法および条件
工業的適用および/または臨床的適用のためのiPSCの使用には、完全に定義済みの培養条件、詳細には、異種を含まない条件において細胞を生成し、選択し、維持することが必要である。したがって、フィーダーフリー培養条件において細胞を再プログラミングさせることが非常に望ましい。しかし、線維芽細胞およびケラチノサイトは皮膚生検材料または毛包を介して入手するので、これらが再プログラミングに最も一般的に使用される細胞型であるが、これらの細胞型の再プログラミングの効率は非常に低く、これらの細胞をフィーダーフリー培養において再プログラミングさせるための効率的な方法はまだ実証されていない。
ナイーブな状態の多能性細胞の生成および維持において使用する細胞培養組成物
最近の研究により、後成的な再プログラミングを通じて、最終分化細胞は、前駆体様の状態(Xie, H.、Ye, M.、Feng, R.、およびGraf, T. 2004年)、異なる分化した細胞の状態(Szabo、Bhatia 2010年)または、元の胚様の状態(Takahashiら、2006年)例えば、iPSCにまで戻って再度コース設定する(recourse)能力を有することが実証された。iPSCの生成はより常套的になってきたが、所与の実験において、非常に低い百分率の体細胞のみがiPSCに再プログラミングされる。この低効率には、体細胞の増殖性の状態、遺伝子の活性化または抑制を導くさらなる変異誘発、遺伝子送達の様式および環境要因を含めたいくつかのパラメータが起因する。iPSCとして同定された全ての細胞がESCに類似した挙動をするとは限らないことも報告されている。例えば、遺伝子発現プロファイリングにより、多くのiPSCが、それらのESC対応物に対して発現プロファイルの有意差を示すことが実証された。さらに、Xist活性の試験およびX染色体の再活性化の分析により、一部のESCはナイーブな状態(すなわち、多能性の基底状態)にあるが、得られたiPSCの全てではないが大部分はプライムされた状態(すなわち、分化するようにプライムされた)にあることが示されている。まとめると、これらの差異が、多能性の低下およびiPSCの特定の細胞型への分化の効率が低いことの一因である可能性があり、これにより再生医療におけるiPSCの価値が下がっている。
多能性幹細胞表面マーカーを調査した。SSEA4およびTra181の発現に加えて、CD30およびCD50の発現も同定し、さらなる多能性の表面マーカーを示すとみなした(図10)。Oct4、Klf4およびSox2を発現しているポリシストロニックなレンチウイルスを用いて再プログラミングされた細胞は、種々の能力の状態を示し、真の多能性のマーカー、例えば、Nanogの発現を担持するわずかな細胞を用いて同定される。現在まで、表面マーカーの発現に基づいて真に多能性のhiPSCを同定する信頼できる方法は存在していない。例えば、図11において認められるように、SSEA4、Tra181およびCD9について陽性と同定された一部のクローン集団は、Nanogを発現せず、hiPSCであると不正確に同定されたことになる。
分化細胞からの人工多能性幹細胞の生成における単細胞選別の使用
図8Aにおいて認めることができるように、Oct4、Klf4、Sox2およびmycを発現している個々のレンチウイルスを用いて誘導した再プログラミングプロセスの初期にある線維芽細胞培養物は、形態学的に異なる細胞のコロニーを含有した。これらの細胞の一部は多能性のマーカーに対して陽性に染色されたが、他はただ単に形質転換された成長が速い細胞であった。再プログラミングプロセスのこの段階では、細胞形態単独では、どの細胞コロニーがiPSCを形成するようになるかは明らかにならない。成長がより速く、形質転換されたが多能性ではない細胞がすぐに培養物を引き継ぐ。したがって、再プログラミングプロセスの初期にあるiPSCを選択する富化ステップを有することが有利であることになる。さらに、図8に示されている通り、一部のコロニーは再プログラミングプロセスの間にいくつかの多能性マーカーを発現し、それは、真のiPSCであったが、一部のコロニーは完全には再プログラミングされておらず、一部の多能性のマーカーのみを発現した。図8Bのコロニー1は、SSEA4とTra181の両方に対して陽性であったが、コロニー4および5は、どちらかのマーカーに対してのみ陽性であった。したがって、コロニー形態によるのではなく、多能性の細胞表面マーカーまたはいくつかのマーカーを同時に用いて多能性細胞を選択することがより効率的であり、技術的な困難が少ないことになる。
複数のiPSCクローンを迅速に生成するための方法および培養組成物
ヒトiPSCを、多能性遺伝子、例えば、Oct4、Sox2、Klf4、c−myc、Lin28およびNanogを異所性発現させることによって生成することは、非効率的かつ技術的に困難なプロセスである。レンチウイルスまたはレトロウイルスによる多能性因子導入遺伝子の宿主細胞のゲノムへの組込みを、フィーダー細胞支持体を含む培養系と組み合わせてもたらす戦略が、iPSCを生成するための伝統的に最も効率的な方法であった。ヒトiPSCを生成するためのウイルスおよびフィーダー細胞方法体系を用いた歴史的研究についての文献レビューにより、0.001%〜0.01%の効率で感染細胞がiPS細胞になることが示されており、潜在的な多能性細胞は感染後21〜30日の期間で認められ、これらは、感染後30日間〜45日間の間に手動の「凝集塊」継代によって、クローンで得られる(表4)。

単細胞選別および富化を用いて多能性細胞集団を分化細胞集団から枯渇させる方法
薬物スクリーニングおよび幹細胞生物学の一部の臨床的適用には、ESCまたはiPSCなどの多能性細胞から特定の系統に分化した均一の細胞集団を生成することが必要である。分化細胞集団に多能性細胞が混入することにより、誤ったスクリーニング結果がもたらされる可能性がある、または、in vivoにおいて腫瘍/奇形腫の形成がもたらされる可能性さえある。細胞集団を分化細胞について富化するか、または多能性細胞を細胞集団から枯渇させるかのいずれかの方法は、本明細書の実施例3および6に記載の選別技術を含んでよい。細胞培養培地において低分子添加物を使用することにより、特に、本発明によって提供されるように、単細胞選別プロセスの間に多能性細胞が分化または部分的に分化することが妨げられ、これにより、完全に分化した細胞の集団から多能性細胞を陰性選択することが可能になる。逆に、細胞選別によって分化細胞を細胞集団から陽性選択することは、多能性細胞が多能性の表面マーカーに対して完全に陽性のままである培養条件下ではより有効であり得る。図20において認めることができるように、完全に分化した線維芽細胞と多能性細胞の混成集団は、表1に記載の培養環境において細胞を前培養した場合には、FACSを用いて有効に分離された。選別手順の間に低分子培養添加物も使用することができ、それにより単細胞懸濁物が安定化される。図20から、多能性マーカーに対して陰性であるとして選択された細胞は、その後の培養およびアルカリホスファターゼについての染色において完全に多能性細胞を含まないことが確かめられたことが理解され得る。
多能性幹細胞のフィーダーフリー培養におけるサイトカインおよび成長因子を含まない培養
実施例2において考察されている通り、従来のヒト多能性培養系は、フィーダー細胞、および、ヒト多能性幹細胞を未分化の状態に維持するための外因性の刺激として機能するbFGFなどのサイトカインを含む。しかし、フィーダー細胞および組換えサイトカインを産生するためのプロセスは、異種汚染物質の供給源として機能する。さらに、フィーダー細胞から分泌される重要な因子(複数可)およびサイトカインによって刺激される正確な細胞経路はまだ同定されていない。したがって、ヒト多能性幹細胞の従来の培養は、不明確な系を表し、臨床グレードの製造への移行を妨げる可能性がある。
多能性幹細胞の生成および維持におけるゲノム安定性
複数の試験により、多能性幹細胞の再プログラミングプロセスおよびその後の培養により、ゲノムの異常の傾向がより高くなり得ることが示唆されている。さらに、フィーダーフリー培養により、核型異常細胞のクローン成長を生じることが示されている。図17A、17Bおよび18Cにおいて実証されている通り、本発明は、再プログラミングさせてゲノム安定性を有する細胞を得る方法、ならびに再プログラミングされたゲノム安定性を有する細胞を維持する方法を提供する。本発明の方法では、細胞を本発明者らのOct4、Klf4およびSox2を含有する3因子ポリシストロニック構築物を使用して再プログラミングさせ、SMC4培地中で培養した場合に、再プログラミングプロセスの間、ならびに長期フィーダーフリー培養の間の両方でゲノム安定性が維持された。図17Aにおいて認められるように、高分解能比較ゲノムハイブリダイゼーションにより、長期フィーダーフリー培養において、SMC4培地を使用してhiPSCを生成し、維持した場合に、最小のコピー数の変動が検出されたことが実証された。さらに、図17Bおよび18Cにおいて実証されている通り、常套的な長期フィーダーフリー培養および単細胞培養の間、ゲノム安定性が維持された。
細胞表面マーカーを使用した、培養している間の多能性細胞の細胞集団の維持
多くの場合、幹細胞培養物の多能性を維持するために、分化細胞を多能性細胞培養物から除去することが有用である。現在まで、このプロセスには、分化細胞を細胞培養物から手動で選び取ること、または未分化細胞を実質的に分化した集団から採取することが必要である。どちらのプロセスも重労働であり、技術訓練を必要とし、常に培養物中の細胞の真の多能性の状態を示すとは限らない形態に基づいた細胞の選択に依拠する(図8)。

- 能力が増大している前記細胞が、従来法で培養したiPSCと比較して少なくとも2倍低いXist活性を有する、請求項32に記載の方法。
- 能力が増大している前記細胞が、密集したドーム形のコロニー形態を有する、請求項32に記載の方法。
- 能力が増大している前記細胞が、TFGβ、アクチビン、およびMEKシグナル伝達経路の外因性の刺激の不在下、および必要に応じてbFGF経路の外因性の刺激の不在下で複製し、かつ多能性を維持する、請求項32に記載の方法。
- 能力が増大している前記細胞を、フィーダーフリー環境において、i)TFGβ阻害剤、ii)GSK3阻害剤、iii)MEK阻害剤、またはiv)Rock阻害剤のうちの少なくとも1種の存在下で培養して、該細胞の該能力を維持しながら少なくとも1回の細胞分裂を可能にするステップをさらに含む、請求項32に記載の方法。

iv)Rock阻害剤からなる群から選択される少なくとも1種の低分子作用剤と、再プログラミングを誘導するために十分な条件下で接触させるステップであって、それにより、再プログラミングの効率が、該細胞集団を該低分子作用剤と接触させていない場合の再プログラミングの効率と比較して少なくとも10%、少なくとも50%、少なくとも100%、少なくとも300%、または少なくとも500%改善するステップを含む方法。 - 再プログラミング前の前記細胞集団が非多能性細胞を含む、請求項50に記載の方法。
- 前記再プログラミングの効率を、再プログラミングのために必要とされる時間または再プログラミングされた細胞の数によって測定する、請求項50に記載の方法。
- 前記条件が、前記細胞集団を、Oct4、Sox、Klf、Myc、Lin28、もしくはNanogポリペプチド、またはOct4、Sox、Klf、Myc、Lin28、もしくはNanogをコードするポリヌクレオチドからなる群から選択される少なくとも1種の外因性転写因子と接触させることを含む、請求項50に記載の方法。
