Abstract
【解決手段】本発明は、単離した哺乳動物マイナス鎖RNAウイルス、すなわちパラミクソウイルス科のニューモウイルスサブファミリーに含まれるメタニューモウイルス(MPV)を提供する。特に本発明は、哺乳動物MPVとそのサブグループおよび変種を提供する。本発明は、哺乳動物メタニューモウイルス、特にヒトメタニューモウイルスの、種々の単離株のゲノムヌクレオチド配列に関する。本発明は、診断および治療法を目的とした、哺乳動物メタニューモウイルスの種々の単離株の配列情報の使用に関する。本発明は、哺乳動物メタニューモウイルスとトリメタニューモウイルスの両方を含めたメタニューモウイルスのゲノムまたはその一部をコードする、ヌクレオチド配列に関する。本発明はさらに、前記ヌクレオチド配列によってコードされたキメラウイルスまたは組換えウイルスを包含する。

Description
トリニューモウイルス(APV)によって生じる呼吸器疾患は、まず南アフリカで1970年代後半に記述され(Buys他、1980、Turkey 28:36〜46)、シチメンチョウ産業に壊滅的な影響を与えた。このシチメンチョウの疾患は副鼻腔炎および鼻炎を特徴とし、シチメンチョウ鼻気管炎(TRT)と呼ばれた。APVのヨーロッパ単離株も、ニワトリの頭部膨張症候群(SHS)の要因として大きく関係している(O'Brien、1985、Vet.Rec. 117:619〜620)。当初この疾患は、ニューカッスル病ウイルス(NDV)に感染したブロイラー集団に見られ、ニューカッスル病(ND)に伴う2次的な問題と考えられていた。SHS発症後の罹患したニワトリでヨーロッパAPVに対する抗体が検出され(Cook他、1988、Avian Pathol. 17:403〜410)、したがってAPVに原因があることが示された。
パラインフルエンザウイルスに感染すると、乳児および児童は重篤な呼吸器疾患に罹る(Tao他、1999、Vaccine 17:1100〜08)。感染性パラインフルエンザウイルス感染は、世界中の呼吸器感染に罹患した小児入院患者全ての約20%を占める(同上)。
呼吸器合胞体ウイルス(RSV)は、乳児および児童の重篤な下気道疾患の主な原因である(Feigen他編、1987、Textbook of Pediatric Infectious Diseases、WB Saunders、Philadelphia、第1653〜1675頁; New Vaccine Development,Establishing Priorities、Vol.1、1985、National Academy Press、Washington DC、第397〜409頁; およびRuuskanen他、1993、Curr.Probl.Pediatr. 23:50〜79)。RSV感染が毎年流行するという性質は世界中で明らかであるが、所与の季節でのRSV疾患の発生率および重症度は地域ごとに様々である(Hall、1993、Contemp.Pediatr. 10:92〜110)。北半球の温暖な地域では、通常晩秋から始まって晩春で終わる。1次RSV感染は、6週齢から2才の小児に最もよく生じ、まれに院内感染で4週齢の乳児にも生じる(Hall他、1979、New Engl.J.Med. 300:393〜396)。RSV感染の危険性が高い小児には、早期産児(Hall他、1979、New Engl.J.Med. 300:393〜396)と、気管支肺異形成(Groothuis他、1988、Pediatrics 82:199〜203)、先天性心疾患(MacDonald他、New Engl.J.Med. 307:397〜400)、先天性または後天性免疫欠損(Ogra他、1988、Pediatr.Infect.Dis.J. 7:246〜249; およびPohl他、1992、J.Infect.Dis. 165:166〜169)、嚢胞性線維症(Abman他、1988、J.Pediatr. 113:826〜830)の小児が含まれるが、これらに限定されない。RSV感染で入院している心疾患または肺疾患の小児における致死率は、3%〜4%である(Navas他、1992、J.Pediatr. 121:348〜354)。

哺乳動物メタニューモウイルスの構造的特徴
本発明は、哺乳動物MPVを提供する。哺乳動物MPVは、パラミクソウイルス科ニューモウイルス亜科に属するネガティブセンス1本鎖RNAウイルスである。さらに哺乳動物MPVは、系統学的にメタニューモウイルス属に相当するものとして同定可能であり、この哺乳動物MPVは、トリ鼻気管炎の病因物質であるシチメンチョウ鼻気管炎ウイルスよりも、I-2614としてCNCM(パリ)(配列番号19)に寄託されたウイルス単離株のほうに系統学上より密接に関係している。ウイルスは、例えばそのウイルスの核酸配列情報を得て系統学的分析により試験することによって、系統学上メタニューモウイルス属に相当するものとして同定可能である。ウイルスの株同士の系統学的関係を決定するには、当業者に知られている任意の技法を使用することができる。例示的な方法に関しては、セクション5.9を参照されたい。その他の技法は、参照によりその全体を本明細書に援用するWO 02/057302として公開された国際特許出願PCT/NL02/00040に開示されている。特にPCT/NL/02/00040は、第12頁第27行から第19頁第29行に系統学的分析に適した核酸配列を開示しており、これを本明細書に参照により援用する。ウイルスはさらに、以下により詳細に述べるように、配列の類似性に基づいて哺乳動物MPVとして同定可能である。
上式で(Tm)は融解温度であり、[Na+]はナトリウム濃度であり、G+Cはグアニンおよびシトシンの含量であり、lは塩基対中のハイブリッドの長さである。配列同士のミスマッチの作用は、Bonner他による式を使用して計算することができ(Bonner他、1973、J.Mol.Biol. 81:123〜135)、ハイブリッド中の塩基対のミスマッチ1%ごとに、融解温度が1〜1.5℃ずつ低下する。
哺乳動物MPVの構造的定義の他、哺乳動物MPVは、その機能的特徴によっても定義することができる。ある実施形態で、本発明の哺乳動物MPVは、哺乳動物宿主に感染することが可能である。哺乳動物宿主は、哺乳動物の細胞、組織、器官、または哺乳類でよい。特定の実施形態で、哺乳動物宿主はヒトであり、あるいはヒトの細胞、組織、または器官である。哺乳動物宿主が哺乳動物MPVに感染しているどうか試験をするには、当業者に知られている任意の方法を使用することができる。ある実施形態では、ウイルスが哺乳動物細胞に結合できるかどうか試験をする。その他のある実施形態では、ウイルスがそのゲノムを哺乳動物細胞内に移すことができるかどうかを試験する。例示的な実施形態では、ウイルスのゲノムを検出可能に標識し、例えば放射性標識する。次いでウイルスを、哺乳動物細胞と共に少なくとも1分間、少なくとも5分間、少なくとも15分間、少なくとも30分間、少なくとも1時間、少なくとも2時間、少なくとも5時間、少なくとも12時間、または少なくとも1日インキュベートする。その後、細胞を洗浄して細胞からウイルス粒子を全て除去し、次いでその細胞について、検出可能な標識を用いてウイルスゲノムが存在するかどうか試験をする。別の実施形態で、細胞内のウイルスゲノムの存在は、哺乳動物MPV特異的プライマーを使用したRT-PCRを使用して検出する(本明細書に参照により援用するPCT WO 02/057302、第37〜44頁参照)。
本発明は、哺乳動物変種およびトリ変種を含む、メタニューモウイルスのゲノムから得られたウイルスベクターによってコードされた組換えまたはキメラウイルスを包含する。本発明によれば、組換えウイルスは、内在性のまたは天然のゲノム配列あるいは非天然のゲノム配列によってコードされた哺乳動物MPVおよびAPVから得られたものである。本発明によれば、非天然配列は、天然のまたは内在性のゲノム配列とは異なるものであるが、それは表現型の変化をもたらしてももたらさなくてもよいゲノム配列に対する点変異、再配列、挿入、欠失などを含むがこれらに限定されない1つまたは複数の変異があることに起因する。本発明の組換えウイルスは、哺乳動物変種およびトリ変種を含む、メタニューモウイルスのゲノムから得られたウイルスベクターによってコードされたウイルスを包含し、ウイルスゲノムに対して非天然の核酸を含んでも含まなくてもよい。本発明によれば、メタニューモウイルスのゲノムから得られたウイルスベクターは、哺乳動物メタニューモウイルスの1つのORFの少なくとも一部をコードする核酸配列を含有するものであり、ORFによってコードされたポリペプチドは、前掲のセクション5.1および表1で述べたアミノ酸配列同一性を有している。
ある実施形態では、ウイルスベクターが、本発明のヒトまたは哺乳動物メタニューモウイルスのゲノムから得られる。その他の実施形態では、ウイルスベクターはトリニューモウイルスのゲノムから得られる。ある実施形態では、ウイルスベクターは哺乳動物MPVおよびAPVから得られた配列を含有し、したがってキメラヒトMPV/APVウイルスはこのウイルスベクターによってコードされるようになる。例示的な実施形態では、キメラhMPV/APVウイルスが構成されるように、ヒトメタニューモウイルスのF遺伝子および/またはG遺伝子をトリニューモウイルスのF遺伝子および/またはG遺伝子に置き換えている。その他の実施形態では、ウイルスベクターが、APVおよび哺乳動物MPVから得られた配列を含有し、その結果、キメラAPV/hMPVウイルスがウイルスベクターによってコードされるようになる。さらに例示的な実施形態では、キメラAPV/hMPVウイルスが構成されるように、トリニューモウイルスのF遺伝子および/またはG遺伝子がヒトメタニューモウイルスのF遺伝子および/またはG遺伝子に置き換えられている。
本発明によれば、本発明のウイルスベクターはさらに、異種配列を発現するよう設計製作することができる。本発明の実施形態で、異種配列は、ウイルスベクター以外の供給源から得られる。限定するのではなく一例として、異種配列は、ウイルスベクターが得られる種、サブグループ、または変種よりも、メタニューモウイルスの様々な種、サブグループ、または変種に属するウイルスの抗原タンパク質、ポリペプチド、またはペプチドをコードする。限定するのではなく一例として、異種配列は、メタニューモウイルス以外のウイルスの抗原タンパク質、ポリペプチド、またはペプチドをコードする。限定するのではなく一例として、異種配列は、その出所源がウイルスではない。この実施形態によれば、異種配列は、所望の生物学的性質または活性を有する部分、ペプチド、ポリペプチド、またはタンパク質をコードすることができる。そのような異種配列は、タグまたはマーカーをコードすることができる。そのような異種配列は、生体応答調節物質、すなわちその例にはリンホカイン、インターロイキン、顆粒球マクロファージコロニー刺激因子、および顆粒球コロニー刺激因子が含まれる生体応答調節物質をコードすることができる。
本発明のウイルスベクターへの外来遺伝子配列の挿入は、ウイルスコード領域を異種配列で完全置換することによって、または部分置換することによって、またはウイルスゲノムに異種ヌクレオチド配列を付加することによって行うことができる。完全置換は、PCR誘導型の変異誘発性を使用することにより、おそらく最良に行われると考えられる。簡単に言うと、PCRプライマーAは、5'末端から3'末端まで、クラスIIS制限酵素部位などの固有の制限酵素部位(すなわち「シフター」酵素; 特定の配列を認識するがその配列の上流または下流でDNAを切断する); 置換される遺伝子の領域に相補的なヌクレオチドの伸長部; および異種配列のカルボキシ末端コード部分に相補的なヌクレオチドの伸長部を含有すると考えられる。PCRプライマーBは、5'末端から3'末端まで、固有の制限酵素部位; 置換される遺伝子に相補的なヌクレオチドの伸長部; および異種または非天然遺伝子の5'コード部分に対応するヌクレオチドの伸長部を含有すると考えられる。これらのプライマーを使用して、クローニングした異種または非天然遺伝子のコピーについてPCR反応させた後、固有の制限部位を使用して生成物を切り出しクローニングすることができる。クラスIIS酵素による消化、および精製したファージポリメラーゼによる転写によって、結果的に異種または非天然遺伝子の挿入を持つようになったウイルス遺伝子の正確な非翻訳末端を含有するRNA分子が生成されると考えられる。代替の実施形態では、PCRによる刺激を受けた反応を使用して、バクテリオファージプロモーター配列およびハイブリッド遺伝子配列を含有する2本鎖DNAを調製することができ、その結果、RNA鋳型をクローニングすることなく直接転写できるようになる。
Gタンパク質は、メタニューモウイルスの膜貫通タンパク質である。特定の実施形態では、ウイルスエンベロープの表面に発現するように、外部ドメインをコードするG-ORFの領域に異種配列を挿入する。1つの手法では、いかなるウイルス配列も欠失させることなく、異種配列を抗原部位に挿入することができる。別の手法では、異種配列がG-ORFの配列に取って代わる。そのような構成の発現産物は、外来抗原に対するワクチンに役立てることができ、ワクチン接種した宿主での組換えウイルスの増殖に伴う問題を確かに回避することができる。抗原部位のみが置換されている無傷のG分子によって、G機能が可能になり、したがって生存可能なウイルスの構成が可能になる。したがってこのウイルスは、さらなるヘルパー機能を必要とせずに、増殖することができる。ウイルスは、あらゆる偶発的な逸脱の危険性が回避されるように、その他の方法で弱毒化してもよい。
2シストロン性mRNAは、ウイルス配列の翻訳の内部開始が可能になるように、また規則的な末端開始部位から外来タンパク質コード配列の発現が可能になるように、構成することができる。あるいは2シストロン性mRNA配列は、ウイルス配列が規則的な末端オープンリーディングフレームから翻訳されるがその一方で外来配列が内部部位から開始するものを構成することができる。ある特定の内部リボソーム進入部位(IRES)配列を利用することができる。選択されるIRES配列は、MPVパッケージングの限界に影響を与えないように、十分短いものであるべきである。したがって、そのような2シストロン的手法を目的として選択されるIRESは、その長さが500ヌクレオチド以下であることが好ましい。特定の実施形態で、IRESは、1つのピコルナウイルスから得られ、いかなる追加のピコルナウイルス配列も含まない。特定のIRES要素には、哺乳動物BiP IRESとC型肝炎ウイルスIRESが含まれるが、これらに限定されない。
上述のように調製されたウイルスベクターおよび組換え鋳型は、適切な宿主細胞に異種遺伝子産物が発現するように、または異種遺伝子産物を発現するキメラウイルスが生成されるように、様々な方法で使用することができる。一実施形態では、組換えcDNAを使用して適切な宿主細胞にトランスフェクトすることができ、得られたRNAを用いて異種遺伝子産物の発現を高いレベルで実施することができる。高レベルの発現をもたらす宿主細胞系は、それぞれAPVまたはMPVに重感染した細胞系、APVまたはMPV機能を補完するよう設計製作された細胞系など、ウイルス機能を提供する連続した細胞系を含む。
本発明のキメラおよび組換えウイルスを調製するために、プラスまたはマイナスセンスで本発明の組換えまたはキメラウイルスのゲノムをコードするcDNAを使用して、複製およびレスキューに必要なウイルスタンパク質および機能をもたらす細胞にトランスフェクトすることができる。あるいは、本発明の組換えウイルスをコードするDNAまたはRNA分子によるトランスフェクションの前、間、または後に、細胞にヘルパーウイルスをトランスフェクトすることができる。本発明の合成組換えプラスミドDNAおよびRNAは、例えば1992年11月24日発行の米国特許第5,166,057号; 1998年12月29日発行の米国特許第5,854,037号; 1996年2月20日公開の欧州特許公開EP0702085A1; 米国特許出願第09/152,845号; 1997年4月3日公開の国際特許公開PCT WO97/12032; 1996年11月7日公開のWO96/34625; 欧州特許公開EP-A780475; 1999年1月21日公開のWO 99/02657; 1998年11月26日公開のWO 98/53078; 1998年1月22日公開WO 98/02530; 1999年4月1日公開のWO 99/15672; 1998年4月2日公開のWO 98/13501; 1997年2月20日公開のWO 97/06270; および1997年6月25日公開のEPO 780 47SA1であって、その全体が参照により本明細書に援用されているものに記載されているような、当技術分野で知られている数多くの技法によって、感染性ウイルス粒子に複製しまたはレスキューすることができる。
本発明の組換えウイルスはさらに、弱毒化表現型を示すよう遺伝子組換えすることができる。特に本発明の組換えウイルスは、ウイルスがワクチンとして投与される対象において、弱毒化表現型を示す。弱毒化は、当業者に知られている任意の方法によって実現することができる。理論に拘泥するものではないが、組換えウイルスの弱毒化表現型は、例えば意図される宿主において自然な状態では十分に複製されないウイルスを使用する(例えばヒトにおけるAPVを使用する)ことによって、また、野生型のウイルス株に比べ、ウイルスゲノムの複製を減少させることによって、ウイルスが宿主細胞に感染する能力を低下させることによって、またはウイルスタンパク質が感染性ウイルス粒子にアセンブルする能力を低下させることによって、引き起こすことができる。リーダー配列やトレラー配列など、ウイルスのある配列の生存能力は、ミニゲノムアッセイを使用して試験することができる(セクション5.8参照)。

感染率は、例えば臨床サンプル(例えば鼻腔ぬぐい分泌液)について本発明のウイルスが存在するかどうか、抗APV抗原抗体、抗hMPV抗原抗体、抗APV抗原抗体、および/または抗体であって異種ヌクレオチド配列の遺伝子産物に特異的なものを使用する免疫蛍光アッセイ(IFA)によってそれぞれ試験をすることであるがこれに限定することのない、当技術分野で周知の任意の方法によって決定することができる。
感染した細胞の細胞質内にある小封入体で強調された、明るい蛍光を特徴とする。
抗体血清力価は、当技術分野で周知の任意の方法によって決定することができ、例えば、血清サンプル中の抗体または抗体断片の量をサンドイッチELISAで定量することができるが、これに限定するものではない。簡単に言うと、ELISAは、血清中の抗体または抗体断片を認識する抗体で、4℃で一晩マイクロタイタープレートを被覆することからなる。次いでこのプレートを、室温で約30分間、PBS-Tween-0.5%BSAで遮断する。PBS-TWEEN-BSA中に希釈した精製済みの抗体または抗体断片を使用して標準曲線を構成し、サンプルをPBS-BSA中に希釈する。サンプルおよび標準液を、アッセイプレートの2つ組のウェルに加え、室温で約1時間インキュベートする。次に非結合抗体をPBS-TWEENで洗い落とし、結合抗体については、室温で約1時間、標識二次抗体(例えばホースラディッシュペルオキシダーゼ結合ヤギ抗ヒトIgG)で処理する。標識に特異的な色素産生基質を添加し、例えば分光光度計によって基質の代謝回転速度を測定することにより、標識抗体の結合を検出する。血清中の抗体または抗体断片レベルの濃度は、サンプルに関する基質の代謝回転速度と標準曲線での基質の代謝回転速度とを、ある希釈率で比較することによって決定する。
本発明のある実施形態では、哺乳動物MPVの構成要素に結合する抗体が存在するかどうか検出する。特に、哺乳動物MPVのタンパク質を対象とする抗体の存在は対象内で検出することができ、それによって、対象内の哺乳動物MPVの存在を診断する。哺乳動物MPVの構成要素を対象とする抗体の存在を検出するには、当業者に知られている任意の方法を使用することができる。
抗体結合の動態パラメータの決定は、例えば、0.05%Tween-20を含有するHBS緩衝液にモノクローナル抗体(「mAb」)を様々な濃度で溶かしたもの250μLを、抗原が固定されているセンサチップ表面に注入することによって、決定することができる。抗原は、哺乳動物MPVの任意の構成要素でよい。特定の実施形態で、抗原は哺乳動物MPVのFタンパク質またはGタンパク質でよいが、これらに限定されない。流量は、75uL/分で一定に維持する。解離データを15分間、または必要に応じてそれ以上収集する。各注入/解離サイクルの後、希釈酸、典型的な場合には10〜100mM HClの短い1分間のパルスを使用して、抗原表面から結合mAbを除去するが、状況が許すならその他の再生剤を使用する。
抗体またはその抗原結合断片がウイルス感染力を中和する能力を、微量中和アッセイにより決定する。この微量中和アッセイは、Anderson他(1985、J.Clin.Microbiol. 22:1050〜1052、その開示全体を参照により本明細書に援用する)により記述された手順に変更を加えたものである。この手順は、その開示全体を参照により本明細書に援用するJohnson他、1999、J.Infectious Diseases 180:35〜40にも記載されている。
このアッセイは、抗体を添加する前に細胞をそれぞれのウイルスに4時間感染させ、読取りが細胞の融合の存否に関するものであること以外、基本的に微量中和アッセイと同一である(Taylor他、1992、J.Gen.Virol. 73:2217〜2223)。
熱力学的結合親和性およびエンタルピーを、抗体とそれに対するそれぞれの抗原との相互作用に関して等温滴定熱量測定(ITC)から決定する。
抗体の結合熱力学を、Microcal,Inc. VP滴定熱量計を使用したITC測定により決定する。VP滴定熱量計は、断熱性エンクロージャ内に封入されたサンプル用と参照用容器(1.409ml)の一対の組合せと、このサンプル容器に対してリガンド溶液を滴定するための回転スターラ-シリンジとからなる。ITC測定は、25℃および35℃で行う。サンプル容器には、抗体を加えたリン酸緩衝液が入っていたが、参照容器には緩衝溶液のみ入っている。リン酸緩衝溶液は、HyClone,Inc.製の生理食塩水67mM PO4(pH7.4)である。5〜10μlという一定量の0.05〜0.1mM RSV-抗原、PIV-抗原、および/またはhMPV-抗原溶液を、3〜4分間あけて、熱交換シグナルが失われることからわかるように結合が飽和するまで抗体サンプル溶液中に滴定する。

本発明のある実施形態では、本発明の治療および診断法を目的とした標的集団を、年齢によって定義する。ある実施形態では、本発明の治療および/または診断法を目的とした標的集団は、呼吸器感染の他に疾患または障害によって特徴付けられる。
in vitroアッセイおよび動物モデルで試験をする本発明のワクチンまたはその断片は、さらに、健常な成人志願者群において、安全性、耐性、および薬物動態について調べることができる。志願者に、筋肉内、静脈内、または肺送達系によって、単回用量の本発明の組換えウイルスおよび/または本発明のワクチンを投与する。単回用量の本発明の組換えウイルスおよび/または本発明のワクチンを受ける前に少なくとも24時間、各志願者をモニターし、その投与を受けた後少なくとも48時間は、各志願者の臨床部位をモニターする。次いで志願者を、外来患者として投与後3日、7日、14日、21日、28日、35日、42日、49日、および56日目にモニターする。

6.1 実施例1: 検体の採取、ウイルスの単離、ウイルスの特性解析
ウイルスの存在についてアッセイするため、また同定されたウイルスを特性解析するために宿主から鼻咽頭吸引液の試料を得た。鼻咽頭吸引液は、気道感染症(RTI)を罹患している子供から採取した。病気の原因が何であるかを決定するために、A型およびB型インフルエンザウイルス、hRSV、ならびに1、2および3型ヒトパラインフルエンザウイルス(hPIV)に対する蛍光標識抗体を用いた直接免疫蛍光アッセイ(DIF)ですべての鼻咽頭吸引液を試験した(実施例9の方法を参照)。ウイルスはまた、VERO細胞、第3カニクイザル腎臓(tMK)細胞、ヒト内皮肺(HEL)細胞およびマービンドック(marbin dock)腎臓(MDCK)細胞を含めた様々な細胞系で、高速シェルバイアル技法(Rothbarth他、1999、J of Virol. Methods、78:163-169)を用いて鼻咽頭吸引液から単離した。DIFアッセイで陰性であった2または3回の継代後に細胞変性効果(CPE)を示す試料を、A、BおよびC型インフルエンザウイルス、AおよびB型hRSV、麻疹ウイルス、流行性耳下腺炎ウイルス、1〜4型ヒトパラインフルエンザウイルス(hPIV)、センダイウイルス、5型サルウイルス、ならびにニューカッスル病ウイルスに対するウイルス特異的抗体を用いた間接免疫蛍光アッセイ(IFA)(実施例11の方法を参照)によって試験した。多くの場合に病原体を同定することができたが、一部の検体は試験したすべてのウイルスに対して陰性であった。
ヒト集団におけるこのウイルスの血清有病率を調査するために、様々な年齢区分のヒト由来の血清を、未同定のウイルス単離株のうち1種に感染させたtMK細胞を用いた間接IFAによって分析した。調査により、このウイルスに対する抗体が6カ月齢〜12カ月齢の子供の25%で検出されることが明らかとなった。さらに、5歳までには、子供のほぼ100%が血清陽性であった。全体で56個の血清試料を間接IFAおよびVNアッセイで観察した。試料の51個または91%で、VNアッセイの結果、すなわち8を超える力価が、間接IFAの結果、すなわち32を超える力価と一致した。IFAで陽性であることが判明した4個の試料試料がVNアッセイで陰性であった、すなわち力価が8未満であった。1個の血清試料がIFAで陰性であった、すなわち、力価が32未満であり、VN試験で陽性であった、すなわち、力価が16であった(図2)。
未知のウイルス単離株の配列情報を得るために、RAP-PCR(Welsh他、1992、NAR、20:4965-4970)として知られるランダムPCR増幅戦略(実施例19参照)。手短に述べると、tMK細胞をウイルス単離株のうち1種(単離株00-1)ならびに陽性対照として役割を果たすhPIV-1に感染させた。両方の培養物が同等のレベルのCPEを示した後、培養物上清中のウイルスを連続的な20〜60%スクロース勾配で精製した。EMによって勾配画分をウイルス様粒子について検査し、約50%のスクロースを含む分画からRNAを単離し、これ中にヌクレオカプシドが観察された。両方のウイルス分画から単離したRNA単離株を等量ずつRAP-PCRで使用し、その後、試料を隣り合わせで3% NuSieveアガロースゲルに流した。次いで、未同定のウイルスに特異的な20個の示差的に現れたバンドをゲルから精製し、プラスミドpCR2.1(Invitrogen)にクローニングし、ベクター特異的プライマーを用いて配列決定した(実施例20参照)。National Library of Medicineから入手可能なBLASTプログラムを用いて、Genbankデータベースで配列に対する相同性を検索することによって、20個の断片のうち10個がAPV/TRTV配列との類似性を示していることが判明した。
MPVの核タンパク質(N)、リン酸化タンパク質(P)、基質タンパク質(M)および融合タンパク質(F)遺伝子の配列分析により、米国において主としてトリで見つかるAPV血清型Cとの最も高い度合の配列相同性が明らかとなった。また、これらの分析により、ウイルスゲノムの3'末端に非構造タンパク質NS1およびNS2が存在しないこと、ならびに融合タンパク質が基質タンパク質のすぐ隣に位置することも明らかになった。22K(M2)遺伝子、小さな疎水性(SH)遺伝子、結合(G)遺伝子、ポリメラーゼ(L)遺伝子、遺伝子間領域、およびトレーラー配列の配列を決定した。既に記載した配列と組み合わせると、ここで提示する配列により、ゲノム末端の最末端の12〜15個のヌクレオチドを除いてMPVのゲノム配列が完成し、MPVのゲノム機構が確立した。MPVゲノムの配列をAPVサブタイプA、BおよびC、RSVサブタイプAおよびB、PVMならびに他のパラミクソウイルスの配列と並べて比較することにより、メタニューモウイルス属におけるMPVの分類の強力な証拠が提供される。
hMPVの別の単離株(1-99)も同定かつ配列決定した。これを行うために、hMPV単離株1-99を第3サル腎臓細胞で、既に記載された方法と全く同じように増殖させた(van den Hoogen他、2001、Nature Medicine、7(6):719-724)。MagnaPure LC単離システム(Roche Applied Science)および全核酸キットのプロトコルを用いてウイルスRNAを単離した。標準のプロトコルを用い、ランダム6量体(Progema Inc. Leiden)をプライマーとして使用して、RNAをcDNAへと変換した。このcDNAは、配列分析で使用するまで-20℃またはそれより低い温度に維持した。このプロジェクトの全体で使用したプライマーは、プロトタイプhMPV 1-00株から入手可能な配列に基づいているか、またはhMPV株1-99を用いて配列分析の後に得られた。
ウイルス群の間、すなわちMPVと他のウイルス間の関係を同定するために系統的な手法を用いることができる。さらに、同型のウイルスの様々な単離株について系統的関係を決定することができる。MPVと他のウイルスとの間の関係性を決定するために、kまたhMPVの様々な単離株間の関係性を決定するために、系統樹を決定した。たとえば、MPVと様々なウイルスとの間の関係性を例示するために、ヌクレオチドまたはタンパク質の配列データを使用して系統樹を作製することができる。あるいは、hMPVの様々な単離株間の関係性を例示するために、ヌクレオチドまたはタンパク質の配列データを使用して系統樹を作製することができる。
ゲノム組成の大部分が決定したが、最末端にある真の(authentic)の末端配列が欠けていた。ウイルスRNAのライゲーション、次いでライゲートした接合点のPCR増幅ならびにポリアデニル化および3'RACE法を組み合わせて用いて、真のヌクレオチド配列を決定した(図54)。ウイルスRNA末端のライゲーションによって作製したPCR断片の配列分析により、図26に示すリーダー配列およびトレーラー配列が明らかとなった(配列番号18〜21参照)。このようにして得たトレーラー配列は、APVを含めた他のパラミクソウイルスのトレーラー配列から予測された配列と矛盾がなかった。しかし、配列分析した71個のクローンのうち2個のリーダー配列しか、現在までにすべてのパラミクソウイルスで見つかっている末端ヌクレオチド残基としてのACを含んでいなかった。したがって、次いでポリアデニル化および3'RACE法を組み合わせて用いてhMPV/NL/1/00リーダーの末端ヌクレオチド配列を確認した。さらに、hMPV NL/1/00の3'リーダー末端に2つの追加のヌクレオチドが同定された。
hMPVのウイルス単離株を血清型または遺伝型によって区別できるかどうかを決定するために、ウイルス中和アッセイを使用した(たとえば実施例16参照)。ウイルス特異的抗血清を産生させるために、ウイルス単離株00-1および99-1をフェレットに接種した。00-1単離株では、個別の加圧したグローブボックス内で飼育した2匹の特定病原体除去フェレットおよび2匹のモルモットを実験的に鼻腔内感染させることによって、ウイルスに対するフェレットおよびモルモット特異的抗血清を産生させた。2〜3週間後、すべての動物を心室穿刺によって出血させ、その血清を対照血清として用いた。血清は、以下に記載のように間接IFAで、既に記載したすべてのウイルスについて試験した。これらの抗血清と99-1単離株を用いて調製した抗血清とを、両方のウイルスでのウイルス中和アッセイで使用した(表10)。
7.1 実施例9: 直接免疫蛍光アッセイ(DIF)法
RTIを罹患している患者由来の鼻咽頭吸引液試料を、記載のようにDIFによって分析した(Rothbarth他、1999、J. of Virol. Methods、78:163-169)。試料は-70℃で保存した。手短に述べると、鼻咽頭吸引液を5mlのダルベッコMEM(BioWhittaker、Walkersville, MD)で希釈し、ボルテックスミキサーで1分間、よく攪拌した。懸濁液を10分間、840×gで遠心した。沈降物をマルチスポットスライド(Nutacon、Leimuiden, The Netherlands)上に広げ、上清をウイルス単離に使用した。乾燥させた後、細胞をアセトン中で1分間、室温で固定した。スライドを洗浄した後、これをインフルエンザAおよびB、hRSVおよびhPIV 1〜3などのウイルスに特異的な市販のFITC標識抗血清(Dako、Glostrup, Denmark)と共に15分間37℃でインキュベートした。PBSで3回、水道水で1回洗浄した後、スライドをグリセロール/PBS溶液(Citifluor、UKO、Canterbury, UK)に浸して覆った。その後、Axioscop蛍光顕微鏡を用いてスライドを分析した。
宿主由来の培養試料中におけるウイルスの検出は、宿主が現在かつ/または過去にウイルスに曝されたまたは感染していたことの直接の指標である。
感染細胞または組織中のウイルスタンパク質を可視化するために抗体を使用することができる。間接免疫蛍光アッセイ(IFA)とは、蛍光指標にカップリングさせた第2の抗体がウイルスに特異的な抗体上の共通のエピトープを認識する感受性のある手法である。IFAは、その感度のレベルが高いのでDIFよりも有利である。
ウイルスの検出には、ウイルスの様々な特徴を利用することができる。たとえば、多くのウイルスが赤血球に結合することができるタンパク質を含み、これにより格子が生じる。この特性は赤血球凝集と呼ばれ、ウイルスを検出するための赤血球凝集アッセイで使用することができる。電子顕微鏡(EM)の下でウイルスを可視化することもでき、またPCR技法によって検出することもできる。
感染/病気中には、ウイルスに対する特異的抗体が産生される。したがって、宿主におけるウイルスに特異的な抗体の検出は、宿主が現在かつ/または過去にそのウイルスに感染していたことの指標である。
ウイルスに特異的な抗体の検出には、MPVを感染させたtMK細胞を(上述のように)アセトンでカバーガラス上に固定し、PBSで洗浄し、1〜16の希釈率の血清試料と共に30分間37℃でインキュベートした。PBSで2回、水道水で1回洗浄した後、スライドを37℃で30分間、FITC標識の使用した種に対する二次抗体(Dako)と共にインキュベートした。上述のようにスライドを処理した。
パラミクソウイルス科ではNタンパク質が最も豊富なタンパク質であり、このタンパク質に対する免疫応答が感染の初期に起こる。そのため、MPVに対する抗体を検出するためのELISAアッセイを開発するためにNタンパク質の組換え源を使用することが好ましい。抗体の検出に適した抗原には、MPVウイルスに曝されたまたはそれに感染した患者の任意のMPV特異的抗体と結合する、任意のMPVタンパク質が含まれる。本発明の好ましい抗原には、MPVに曝された患者において免疫応答を優先的に引き起こし、したがって典型的には患者の抗体によって最も容易に認識されるものが含まれる。特に好ましい抗原には、MPVのN、F、MおよびGタンパク質が含まれる。免疫学的技法に用いられる抗原は、未変性抗原であっても、それを改変した形であってもよい。分子生物学における周知の技術を用いてMPV抗原のアミノ酸配列を変化させ、免疫学的技法で使用し得る改変した形の抗原を生成することができる。
対象がウイルスに感染すると、そのウイルスに対する抗体のアレイが産生させる。これらの抗体の一部はウイルス粒子に結合してその感染力を中和することができる。ウイルス中和アッセイ(VN)は通常、血清またはモノクローナル抗体の希釈液をウイルスと混合し、これをインキュベートし、培養細胞、孵化卵、または動物を用いて残存感染力をアッセイすることによって実施する。中和抗体はウイルス粒子上の型特異的な抗原ウイルス粒子を定義するために使用することができる。たとえば、ウイルスの血清型を定義するために中和抗体を使用することができる。さらに、幅広く中和する抗体も存在し得る。
宿主におけるウイルスの存在は、宿主から採取した試料中でウイルス核酸を検出することによっても診断することができる(たとえば実施例18のRT-PCRおよび実施例19のRAP-PCR参照)。
生体試料中のウイルスの検出は、ウイルスのゲノム物質を複製または増幅する方法を用いて行うことができる。既知のパラミクソウイルスにおけるRT-PCRアッセイのためのウイルスに特異的なオリゴヌクレオチド配列を本実施例中で以下に記載する。50mMのTris.HCl pH8.5、50mMのNaCl、4mMのMgCl2、2mMのジチオトレイトール、200μMずつの各dNTP、10単位の組換えRNAsin(Promega、Leiden, the Netherlands)、10単位のAMV RT(Promega、Leiden, the Netherlands)、5単位のAmplitaq Gold DNAポリメラーゼ(PE Biosystems、Nieuwerkerk aan de Ijssel, The Netherlands)、および5μlのRNAを含む50μlの反応液中で、1ステップのRT-PCRを行った。サイクル条件は、42℃で45分間および95℃で7分間を1回、95℃で1分間、42℃で2分間、および72℃で3分間を40回繰り返し、72℃で10分間を1回である。プライマーの配列は配列表で提供する。より具体的には、核タンパク質の遺伝子に使用したプライマーはそれぞれ配列番号28および29に対応するヌクレオチド配列を有するN3およびN4であり、これらを用いて151個のヌクレオチドの断片を増幅した。基質タンパク質の遺伝子に使用したプライマーはそれぞれ配列番号30および31に対応するヌクレオチド配列を有するM3およびM4であり、これらを用いて252個のヌクレオチドの断片を増幅した。ポリメラーゼタンパク質の遺伝子に使用したプライマーはそれぞれ配列番号34および35に対応するヌクレオチド配列を有するL6およびL7であり、これらを用いて173個のヌクレオチドの断片を増幅した。Fタンパク質の遺伝子に使用したプライマーはそれぞれ配列番号32および33に対応するヌクレオチド配列を有するF7およびF8であり、これらを用いて221個のヌクレオチドの断片を増幅した。
MPVまたは他のウイルスの遺伝物質は、ゲノム内の領域にハイブリダイズし、遺伝物質が増幅されるように特定の方向に伸張するプライマーを用いて検出または増幅することができる。この種の技法は、特定の配列情報が入手不可であるか、また遺伝物質の最初の増幅を試料中で行う場合に有用である。このような技法の1つはRAP-PCRと呼ばれている。
RAP-PCRを用いてセグメントを増幅した後、増幅したセグメントについて配列情報を得ることができる。これを行うためには、配列決定の前に作製した断片をベクター内にクローニングすることが有利である。
RAP-PCR方法は配列中に、増幅または複製されていないギャップを残す場合がある。完全な配列を得るために、RT-PCRを用いてギャップの配列情報を得ることができる。
hMPVウイルスを検出するために、様々な宿主内で抗体の存在を検出する免疫学的アッセイ。最も豊富に産生されるタンパク質であるので、一例では、Nタンパク質に対する抗体を使用する。この特徴は、ウイルスのゲノムに渡って起こる転写の勾配によるものである。
8.1 実施例22: 様々な細胞系におけるHMPVの増殖
各ウイルスの特徴を検査するためにウイルス単離株を様々な細胞系で培養することができる。たとえば、培養物中で測定される力価レベルに基づいて様々なウイルス単離株の感染力を特徴とし、かつ区別することができる。あるいは、さらに分析するために、細胞を使用して培養物中のウイルス株を増殖または増幅させることができる。
ミニレプリコン構築体を、アンチセンスレポーター遺伝子を含むように作製することができる。ミニレプリコンの例であるCAT-hMPVを図24に示す。ミニレプリコン構築体の作製に使用したリーダー配列およびトレーラー配列を図26に示す。比較のために、APV、RSVおよびPIV3のリーダー配列およびトレーラー配列のアラインメントも図26に示す。
hMPVウイルスの遺伝子を発現する完全長cDNAを構築して、感染性のウイルスを生成することができる。たとえば、hMPVのすべての遺伝子またはすべての必須の遺伝子をコードしているcDNAを構築することができる。その後、ゲノムを発現させて感染性ウイルスを生成することができる。
MPV株の病原性を特性解析すために動物宿主を感染させることができる。たとえば、それぞれの株が生物内でどれほど感染性があるかを決定するために、様々な宿主を用いることができる。
APVは、ヒトMPVによる感染を予防するため、またはヒト宿主におけるヒトMPVの感染力を低減させるためのワクチンとして使用することができる。ワクチンはAPV全体あるいはAPVの配列および加えて別のメタニューモウイルスの異種配列からなるそのキメラもしくは組換え型または誘導体であることができる。組換えウイルスワクチンを作製するバックボーンとしてAPVのゲノムを使用することができる。たとえば、APVのF遺伝子および/またはG遺伝子をヒトMPVのF遺伝子またはG遺伝子で置換したワクチンを作製することができる。あるいは、APVバックボーンの配列をPIV由来の配列をAPVバックボーンに置換したまたは付加した配列を含むワクチンを作製することができる。組換え/キメラワクチンのさらなる情報については、たとえばConstruction of the Recombinant cDNA and RNAを参照されたい。
ヒトMPVは、APVによる感染を予防するため、またはトリ宿主におけるAPVの感染力を低減させるためのトリ用ワクチンとして使用することができる。ワクチンはMPV全体あるいはMPVの配列および加えて別のメタニューモウイルスの異種配列からなるそのキメラもしくは組換え型または誘導体であることができる。組換えウイルスワクチンを作製するバックボーンとしてMPVのゲノムを使用することができる。たとえば、ヒトMPVのF遺伝子および/またはG遺伝子をAPVのF遺伝子またはG遺伝子で置換したワクチンを作製することができる。組換え/キメラワクチンのさらなる情報については、たとえばConstruction of the Recombinant cDNA and RNAを参照されたい。
Claims (15)
- 組換えヒトメタニューモウイルス変種A1、A2又はB1、および医薬品として許容される担体を含む免疫原性組成物。
- 組換えヒトメタニューモウイルスが配列番号18、19又は20の核酸配列と少なくとも99%同一であるヌクレオチド配列を含む、請求項1記載の免疫原性組成物。
- 組換えヒトメタニューモウイルスが弱毒化されている、請求項2記載の免疫原性組成物。
- 組換えヒトメタニューモウイルスが配列番号18、19又は20の核酸配列を有するゲノムを含む、請求項1記載の免疫原性組成物。
- 組換えヒトメタニューモウイルスのゲノムが遺伝的変異を含む、請求項1〜4のいずれか1項に記載の免疫原性組成物。
- 遺伝的変異が、置換、挿入、欠失またはその組合せである、請求項5記載の免疫原性組成物。
- 挿入が、異種核酸の挿入である、請求項6記載の免疫原性組成物。
- 呼吸器感染を治療または予防するための医薬品を製造するための、組換えヒトメタニューモウイルス変種A1、A2又はB1の使用。
