Abstract
【解決手段】腫瘍細胞において特異的に発現するゲノムインプリンティング遺伝子に着目し、この遺伝子の調節領域を利用して細胞傷害性の産物をコードする異種遺伝子を特異的に発現させ、腫瘍細胞の増殖を抑制する方法を開発した。具体的遺伝子としては、H19遺伝子、及びインスリン様増殖因子遺伝子のプロモーター、エンハンサー領域の利用である。また、細胞傷害性の産物としては、ジフテリア毒素、コレラ毒素、オンコジーン形態からなる群より選択される遺伝子配列に特異的にハイブリダイズするアンチセンス配列、該配列をコードするRNAを特異的に切断するリボザイム等である。

Description
1.発明の技術分野
本発明は、腫瘍細胞生理学および癌の治療分野に関する。さらに詳しくは、本発明は、腫瘍細胞における異種遺伝子、特に細胞傷害性生成物をコードする遺伝子の特異的発現に関する。
2.1 H19遺伝子
H19遺伝子は、ヒトにインプリンティングされている(imprinted)ことが知られている数少ない遺伝子の一つである(Hurst et al., 1996, Nature Genetics 12:234-237)。H19は、胚形成のごく初期に両染色体の対立遺伝子から発現される(DeGroot et al., 1994, Trophoblast 8:285-302)。その後間もなく、父性の対立遺伝子は沈黙(silencing)する一方で、母性遺伝対立遺伝子だけは転写される。
IGF-2は、発現がその親の由来に依存するもう一つのインプリンティングされた遺伝子である。しかしながら、H19とは異なり、マウスおよびヒトのIGF-2は母系インプリンティングであり、従って父系遺伝対立遺伝子から発現される(Rainier et al., 1993, Nature 363:747-749)。ヒトIGF-2遺伝子は複雑な転写パターンを示す。組織および発生に特異的な方法で活性化されるIGF-2プロモーターは4つある。そのプロモーターのうちの3つ、すなわち、P2、P3およびP4だけが胎児発生中および癌組織でインプリンティングされ活性となる。4つ目のプロモーターP1はインプリンティングされることはなく、成人肝および脈絡集網だけで活性化される(Holthuizen et al., 1993, Mol. Reprod. Dev. 35:391-393参照)。IGF-2遺伝子のP3プロモーターは肝硬変および肝細胞癌の進行に関与している(Kim & Park, 1998, J. Korean Med. Sci. 13:171-178)。
腫瘍関連遺伝子からの調節配列は、腫瘍由来細胞において自殺遺伝子の発現を選択的にねらい打ちするのに用いられてきた。例えば、アルファーフェトプロテイン発現は肝細胞癌で誘導される。Huber ら(1991, Proc. Natl. Acad. Sci. USA 88:8039-8043)は、アルブミン遺伝子またはアルファーフェトプロテイン遺伝子のどちらかからの調節配列を用いて、肝癌細胞配列をコードする水痘帯状疱疹チミジンキナーゼ(VZV TK)遺伝子の発現を標的にした。in vitroでこれらの発現構築物の1つを含むレトロウイルスベクターに感染させた肝癌細胞はVZV TKを発現し、通常は非毒性のプロドラッグ、6-メトキシプリンアラビノヌクレオシド(araM)感受性となった。Kanekoら(1995, Cancer Res. 55:5283-5287)は、アルファーフェトプロテイン調節配列の制御下でHSV TKを発現するアデノウイルスベクターを構築した。このベクターを含有する組換えアデノウイルス粒子を、無胸腺マウスに発生させた肝細胞癌由来の腫瘍に直接注射した。次いでガンシクロビルを腹腔内投与すると、肝細胞癌由来の腫瘍退行が起こった。
本発明は、腫瘍細胞において異種遺伝子の選択的発現を誘導する方法および組成物に関する。本発明は特に、異種遺伝子の腫瘍特異的発現をもたらす異種遺伝子と機能的に連結された調節転写配列を含むポリヌクレオチドに関する。さらに詳しくは、この調節転写配列は、H19のような癌細胞で特異的に発現されるゲノムインプリンティングされた遺伝子およびIGF-2 P3、P4に由来するものであり、異種遺伝子は細胞傷害性タンパク質または細胞増殖抑制遺伝子の産物をコードする。本発明の他の態様によれば、IGF-Iプロモーターは異種遺伝子と機能的に連結され、当該遺伝子の腫瘍特異的発現をもたらす。調節配列は、多数の異なる癌細胞タイプにおける遺伝子発現を指示するものである。そのような方法および組成物は、広範にわたる多様な癌および過剰増殖症状の治療に有用である。
本発明は、一部は、癌細胞で発現されるゲノムインプリンティング遺伝子の調節領域を癌細胞における目的のコード配列発現を標的とするのに使用できるという発見に基づくものである。特に、以下の例示に限定されるものではないが膀胱癌、肝細胞癌、肝芽細胞腫、横紋筋肉腫、卵巣癌、子宮頸癌、肺癌、乳癌、頭部および頸部の扁平上皮癌、食道癌、甲状腺癌、神経膠星状細胞腫、神経節芽細胞腫および神経芽細胞腫を含む、広範で多様な癌においてH19の発現が活性化されることを見出した。さらに、異種遺伝子と機能的に連結されたH19プロモーター領域、すなわち、異種遺伝子と機能的に連結されたIGF-2 P3若しくはP4プロモーターを含む構築物、または、下流のH19エンハンサーと組み合わせてそのようなプロモーターを含む構築物が腫瘍細胞において特異的に活性化されることを見出した。本発明の他の態様によれば、IGF-1プロモーターは異種遺伝子の発現を命令するのに使用される。
本明細書中に記載されているのは、異種コード配列の腫瘍細胞特異的な発現を指令するために使用できるH19調節配列である。これらのH19調節配列には、上流のH19プロモーター領域および/または下流のH19エンハンサー領域が含まれる。ある1つのH19プロモーター領域のヌクレオチド配列を図1A〜1C(配列番号1)に示す。この830ヌクレオチド配列は(Brannanら、上掲に記載されたように)キャップ部位から−837〜−7位のヌクレオチドにわたる。共通TATA配列はヌクレオチド−27〜−35位に見出される。2つの共通AP2結合部位(8/9一致)は、転写開始部位から約−500〜−40ヌクレオチド上流に見出される。コード領域の上流を異種遺伝子と置き換えた場合は、以下により詳細に論じるように、癌細胞(該細胞もまた内在性H19を発現する)中に機能的に連結された異種遺伝子の発現を指令するには、約830塩基対の調節領域で十分である。さらに、ヌクレオチド−819〜+14間の別のH19プロモーター領域(図5、配列番号2)もまた、癌細胞中に機能的に連結された異種遺伝子の発現を指令するには十分である。
インプリンティングされた遺伝子の発現を再活性化する細胞は、異種遺伝子に機能的に連結されたかかるインプリンティングされた遺伝子の調節領域を含有する発現構築物を特異的に活性化することもできる。そうした細胞、特に腫瘍細胞は、本発明の遺伝子治療法の適切な標的である。腫瘍および細胞系の両方におけるH19、およびIGF-2 P3およびP4の特異的な発現は、RNA解析技術(in situハイブリダイゼーションおよびレポーター遺伝子構築物)を用いて測定することができる。さらに、活性化されたIGF-1遺伝子発現を伴う腫瘍細胞も同様に測定し、IGF-1プロモーターを使用して異種遺伝子の発現を指令する遺伝子治療において標的とすることができる。

従って、上記癌は本発明の方法により治療することができるものである。実際に、H19発現を活性化するいずれの腫瘍も本発明の方法によって治療することができる。
本発明はまた、異種遺伝子に機能的に連結された調節領域を含むポリヌクレオチドでトランスフェクトした宿主細胞に関する。かかる宿主細胞は培養物中で維持されてもよく、または動物(好ましくは哺乳動物)の部分であってもよい。典型的には、対象のポリヌクレオチドを多様なベクターに挿入し、このベクターを次に本明細書中に開示されている方法および材料を用いて送達する。このようなベクターは確立された分子生物学的手法(概要はSambrookら、(1989) Molecular Cloning Vols. I-III, Cold Spring Harbor Laboratory Press, Cold Spring Harbor, New YorkならびにCurrent Protocols in Molecular Biology(1989) John WileyおよびSons, all Vols.ならびにその定期的な最新情報(引用により本明細書中に組み込む)を用いて製造することができる。典型的には、翻訳が所望される場合は、必要であれば対象の異種遺伝子も遺伝子操作して適当な3'ポリアデニル化配列を含ませる。
非相同遺伝子に作動的に連鎖されたインプリントされた遺伝子調節領域を含有するポリヌクレオチドによるトランスフェクションを受けた宿主細胞は、あらゆる原核又は真核細胞であってよい。ポリヌクレオチドをベクターのような遺伝子構成体内にライゲートさせ真核細胞(酵母、鳥類、昆虫又は哺乳動物)又は原核細胞(細菌細胞)を宿主細胞へと形質転換又はトランスフェクションすることは、微生物又は組織培養技術において広く使用される標準的手順である。
本発明は同様に、ガン及び高増殖性疾患を治療する目的で遺伝子療法において使用するための非相同性遺伝子に作動的に連鎖された遺伝子調節領域を含有するポリヌクレオチドの使用をも包含するものである。遺伝子療法を目的として、本発明の発現構成体は、in vivoで細胞に対し組換え型遺伝子を有効に送達する能力をもつあらゆる製剤又は組成物といった生物学的に有効な任意の担体内で投与することができる。アプローチとしては、組換え型レトロウイルス、アデノウイルス、アデノ関連性ウイルス及び単純ヘルペスウイルス−1又は組換え型細菌又は真核性プラスミドを含むウイルスベクター内への対象遺伝子の挿入が含まれる。ウイルスベクターは、細胞を直接トランスフェクションする:すなわち、プラスミドDNAは、例えばカチオン重合体、カチオン性リポゾーム(例えばリポフェクチン、コレステロール誘導体、例えば、D.D.A.B.及びカチオンリン脂質)、又は誘導体化された(例えば抗体接合された)ポリリシン接合体、グラミシジンS、人工ウイルスエンベロープ又はその他のこのような細胞内担体ならびに裸の遺伝子構成体の直接注入、in vivo で行なわれるCaPO4沈降法又は電気穿孔法を援用して、送達されうる。ガン遺伝子療法のための遺伝子の移入及び発現系の最近の論評としては、Cooper, 1996,Seminars in Oncology(腫瘍学セミナー)23;172−187がある。
当業者であればわかるように、医療従事者又は患者の観点から見ると、ガンによる身体条件(例えば、痛み、感受性、体重減少など)に関連する望ましくない症状の軽減又は予防は事実上全て望ましいものである。さらに、腫瘍質量又は成長速度の低下も、腫瘍の組織病理学像の改善と共に望まれる。従って、本出願においては、本書で用いられている「治療」、「治療的使用」又は「医療的使用」という語は、疾病状態又は症候を改善するか又はその他の方法で疾病又はその他の望ましくない症候の進行を何らかの形で防止、妨害、遅延又は逆行させる請求対象の組成物のあらゆる使用を意味するものとする。
この節では、H19調節配列の制御下に置かれたCATリポータ遺伝子を含むさまざまな発現構成体の構築及び複数の異なる膀胱ガン細胞株内へのそれらの移入について記述する。
6.1.1 細胞株及びトランスフェクション
膀胱ガン細胞株HT−1376、EJ28、T24P、1197及びUM−UC−3は米国標準培養収集機関(ATCC)から得たものであり、ATCC推奨事項に従って維持された。
(CATリポータ遺伝子とそれに先行する多重クローニング部位を含有する)pCAT−basic、(SV40プロモータの制御下でCATリポータ遺伝子を含む)pCAT−promoter、(CATリポータ遺伝子の下流のSV40エンハンサ及びCATリポータ遺伝子の下流のプロモータの挿入のための多重クローニング部位を含む)pCAT enhancer 及び(SV40プロモータとエンハンサの両方の制御下でCATリポータ遺伝子を含む)pCAT−control といったプラスミドは全て Promega(Madison,WI)から市販されていたものである。

5つの異なる膀胱ガン細胞株HT−1376、EJ28、T24P、1197及びUM−UC−3を各々、pCAT−basic(図2でP−Eとして呼称されているもの)、pCAT−control(図2でpSV40ESV40と呼称されているもの)、pH19E、pH19EH19D、及びpH19EH19Rでトランスフェクションした。各構成体の発現結果を、図3A〜3Eに示した。各々の細胞株において、CAT活性の最高のレベルは、SV40エンハンサとSV40プロモータの両方を含有するpCAT−control プラスミドで観察された。この構成体は、SV40調節配列が遺伝子発現の誘発因子として立証されていることから、正の対照として役立った。しかしながら、SV40調節配列はその遺伝子発現を誘発する能力において腫瘍細胞特異的ではない。H19プロモータの制御下でCATリポータ遺伝子を含有する、pH19Eでのトランスフェクションを受けた細胞株も同様にバックグラウンドに比べ著しく増大したCAT発現を示した。H19プロモータによるCAT活性の誘発レベルは、HT−1376細胞株内の5倍からUM−UC−3細胞株内の10倍までの範囲にわたっていた。H19プロモータ/CATリポータ遺伝子構成体に対するH19エンハンサの添加はさらに、ある種の細胞株において発現レベルを増大させた。例えば、細胞株FJ28、T24P及び1197においては、H19エンハンサは、発現をH19プロモータ/CATリポータ遺伝子から著しく増大させた。しかしながら、エンハンサの向きは、異なる細胞株内で異なる結果を与えた。細胞株HT−1376及びUM−UC−3においては、エンハンサは、発現に対しほとんど又は全く効果を示さなかった。
7.1 材料と方法
以上の第6節で記述した発現構成体を、CATではなく毒性産物又はプロドラッグをコードする配列を発現するよう修飾する。例えば、CAT遺伝子産物をコードする配列を除去し、当該技術分野において周知の標準的クローニング方法を用いて単純ヘルペスウイルスチミジンキナーゼ(HSV−TK)で置換させる。
8.1. 材料及び方法
生後5週目の雌のC3H/Heマウス(Charles River)70匹を、カゴ1個あたり6匹の割合で収容し、空調された室内において12時間明/12時間暗のサイクルで環境順化させた。生後8週間目で実験を開始し、マウスを対照群(10匹)と実験群(60匹)に無作為に分けた。実験群のマウスには、飲料水中で随意に溶解させた0.05%のN−ブチル−N−(4−ヒドロキシブチル)ニトロサミン(BBM)(東京化成工業(株)日本国東京)を与えた。対照マウスには、水道水を与えた。両方の群からの動物を、実験開始後4週目、8週目、12週目、16週目、20週目及び26週目に屠殺した。標準的な手順を用いて膀胱を切除し、定着させ、パラフィンブロック内に埋め込んだ。
マウスH19コーディング領域を含む2.1kbのフラグメントを、T7及びT3RNAポリメラーゼ結合部位の後ろでpBluescript II KSプラスミド(Stratagene, La Jolla, CA)内にサブクローニングした。〔35S〕で標識づけされたアンチセンスH19RNAを、T7ポリメラーゼ(Boehringer Mannheim)及び AmershamRPN2006キットを用いて Hind III で線状化されたプラスミドDNAから in vitro で産生させた。 in vitro で生成された写しは、107cpm/μgの比活性を有していた。T3ポリメラーゼ(Boehringer Mannheim)及びEcoRI−線状化された鋳型を用いて調製したセンスH19mRNAを対照として使用した。
ホルマリン定着された組織のパラフィンワックス切片(5μM)を3アミノプロピルトリエトキシラン(Tespa, Sigma) でコーティングされた顕微鏡スライド上に乗せ、37℃で一晩乾燥させた。切片をキシレンで脱ろうし、4%のパラホルムアルデヒドで定着させ、プロテイナーゼK(Sigma)で処理した。プローブの非特異的結合を減少させるべくスライドをアセチル化し、エタノール系列を通して脱水した。
26週までに、生存した実験群のマウスの全てが触知可能な膀胱腫瘍を発生させていた。H19の広範な発現が、化学的に誘発された膀胱腫瘍の中で観察された。これとは対照的に、正常な成熟した膀胱内ではH19の発現は全く見られなかった。従って、この化学的に誘発された膀胱ガンのマウスモデルは、毒素遺伝子に作動的に連鎖されたH19の調節領域を含む構成体の in vivoでの腫瘍特異的細胞障害性を実証するための動物モデルとして使用することができる。
H19/毒素又はプロドラッグ発現プラスミドを、in vivo でのマウス膀胱への送達のためリポソーム内に(本書に参考として内含されている Takashita et al.,1993,J. Clin. Invest.93:652−651によって記述されているとおりに)取込ませる。この実験のために使用されるマウスは、第8節で上述したとおり化学的に誘発された膀胱腫瘍を有している。
10.1 材料と方法
この実験においては、4つの異なるIGF−2プロモータのうちの1つの制御下に置かれたルシフェラーゼリポータ遺伝子を含むさまざまな発現構成体を構築し、複数の異なる膀胱ガン細胞株内に移入した。以下のヒトIGF−2プロモータ/ルシフェラーゼ構成体を作成した:
IGF−2プロモータ配列は、本書に参考として内含されているSussenbach et al.,1992 Growth Reg, 2:1−9の中で記述されている。ルシフェラーゼリポータベクターは、Promega Madison, WI(カタログ#E1641)から市販されている。
得られた発現プラスミドを、以上の第6節で記述されているようにヒトの膀胱ガン細胞株HT−1376、EJ28、T24P、1197及びUM−UC−3の中にトランスフェクションした。ルシフェラーゼ活性を、市販のキット(Promega, Madison, WI,カタログ#E1500)を用いて検定した。図4A〜4Eに示された結果は、IGF−2P4プロモータが、テストされた各々の膀胱ガン細胞株内のルシフェラーゼリポータ遺伝子の発現を導いたことを立証している。細胞株1197において、IGF−2 P3プロモータは同様に、ルシフェラーゼリポータ遺伝子の発現をも導いた。その後の実験において、IGF−2 P3及びP4プロモータは、繊毛上皮腫細胞及び横紋筋肉腫細胞を含むその他の腫瘍細胞内のルシフェラーゼ遺伝子発現の発現を導くことが示された。
11.1. 材料と方法
4つのルシフェラーゼリポータベクター、pGL3−Basic、pGL3−Promoter、 pGL3−Enhancer 及びpGL3−Control を Promegaから得た。これらのベクターを、lipofetamine(Gibco/BRL)、fugene (Boehringer)、 8つの脂質試薬から成る Perfect Transfection Kit (Invitrogen)、 TFX−10、TFX−20、transfast (Promega) を含む一定数の異なるトランスフェクション試薬及びリン酸カルシウム法(Gorman et al.,1982,Mol. 培養された細胞株内に4つのルシフェラーゼ遺伝子含有ベクターを導入するのに複数のトランスフェクション試薬が使用された場合、リン酸カルシウム沈降は、テストされた細胞株の大部分について最高のトランスフェクション効率を生み出した。従って、その後、さまざまな発現ベクターをトランスフェクションするためにカルシウム沈降が使用された。さらに、プラスミドDNAの濃度の増加は、安定水準より高い濃度で使用した場合でさえ、トランスフェクション効率を阻害しなかった。

Claims (51)
- 細胞傷害性の遺伝子産物をコードする異種配列に機能的に連結された調節配列を含むポリヌクレオチドを含有する、腫瘍細胞において配列を発現させるためのベクターであって、前記調節配列がIGF-2 P4プロモーターおよびIGF-1プロモーターから選択されるものである、上記ベクター。
- 異種配列がβ−ガラクトシダーゼ、ジフテリア毒素、シュードモナス毒素、リシン、コレラ毒素、網膜芽細胞腫遺伝子、p53、単純ヘルペスチミジンキナーゼ、水痘−帯状疱疹チミジンキナーゼ、シトシンデアミナーゼ、ニトロレダクターゼ、シトクロムp-450 2B1、チミジンホスホリラーゼ、プリンヌクレオシドホスホリラーゼ、アルカリホスファターゼ、カルボキシペプチダーゼAおよびG2、リナマラーゼ(linamarase)、β−ラクタマーゼおよびキサンチンオキシダーゼからなる群より選択されるタンパク質をコードする、請求項1記載のベクター。
- ヒトにおける増殖性疾患の予防または治療への使用に意図されたものである、請求項1または2記載のベクター。
- ヒトに対するリポソーム投与に意図されたものである、請求項1〜3のいずれか1項記載のベクター。
- 請求項1〜4のいずれか1項記載のベクターを含む宿主細胞。
- IGF-2 P4プロモーターおよびIGF-1プロモーターから選択される配列に由来する調節配列に機能的に連結された細胞傷害性または細胞増殖抑制性の遺伝子産物をコードするポリヌクレオチドを含む、被験体の癌治療のための医薬組成物。
- 細胞傷害性の遺伝子産物がジフテリア毒素、シュードモナス毒素、リシン、コレラ毒素、網膜芽細胞腫遺伝子およびp53からなる群より選択される、請求項6記載の医薬組成物。
- 癌が膀胱癌、肝細胞性癌、肝芽細胞腫、横紋筋肉腫、卵巣癌、子宮頸癌、肺癌、乳癌、頭部および頸部の扁平上皮癌、食道癌、甲状腺癌、神経膠星状細胞腫、神経節芽細胞腫および神経芽細胞腫から選択される、請求項6または7記載の医薬組成物。
- 癌が膀胱癌または肝細胞性癌である、請求項8記載の医薬組成物。
- 増殖性疾患が膀胱癌、肝細胞性癌、肝芽細胞腫、横紋筋肉腫、卵巣癌、子宮頸癌、肺癌、乳癌、頭部および頸部の扁平上皮癌、食道癌、甲状腺癌、神経膠星状細胞腫、神経節芽細胞腫および神経芽細胞腫から選択される、請求項3記載のベクター。
- 増殖性疾患が膀胱癌または肝細胞性癌である、請求項10記載のベクター。
- 増殖性疾患または障害を治療または予防するための医薬として使用するための核酸であって、
a) 少なくとも1つのH19調節配列、IGF-2調節配列および少なくとも1つのH19調節配列断片より選択される少なくとも1つの調節配列と、
b) 細胞傷害性または細胞増殖抑制性の遺伝子産物をコードする異種配列と、
を含み、前記少なくとも1つの調節配列が異種配列に機能的に連結されており、且つ前記少なくとも1つの調節配列が腫瘍細胞で特異的に発現するゲノムインプリンティングされた遺伝子に由来するものである、前記核酸。 - H19調節配列がH19プロモーター配列である、請求項12記載の核酸。
- H19調節配列がH19エンハンサー配列である、請求項12記載の核酸。
- IGF-2調節配列がIGF-2 P4プロモーターである、請求項12記載の核酸。
- IGF-2調節配列がIGF-2 P3プロモーターである、請求項12記載の核酸。
- 腫瘍細胞が膀胱癌細胞である、請求項12記載の核酸。
- 膀胱癌細胞が、Ht-1376、EJ28、T24P、1197およびUm-UC-3からなる群より選択される、請求項17記載の核酸。
- 少なくとも1つのH19調節配列が、H19プロモーター、H19エンハンサー、およびH19プロモーターとH19エンハンサーとの組合わせから選択される、請求項12記載の核酸。
- 異種配列がβ−ガラクトシダーゼ、ジフテリア毒素、シュードモナス毒素、リシン、コレラ毒素、網膜芽細胞腫遺伝子、p53、単純ヘルペスチミジンキナーゼ、水痘−帯状疱疹チミジンキナーゼ、シトシンデアミナーゼ、ニトロレダクターゼ、シトクロムp-450 2B1、チミジンホスホリラーゼ、プリンヌクレオシドホスホリラーゼ、アルカリホスファターゼ、カルボキシペプチダーゼAおよびG2、リナマラーゼ(linamarase)、β−ラクタマーゼおよびキサンチンオキシダーゼのコード配列からなる群より選択される、請求項12記載の核酸。
- 異種配列が、cdk2、cdk8、cdk2l、cdc25A、サイクリンDl、サイクリンE、サイクリンA、cdk4並びにp53、c-fos、c-jun、Kr-rasおよびHer2/neuのオンコジーン形態からなる群より選択される遺伝子をコードする配列に特異的にハイブリダイズするアンチセンス配列である、請求項12記載の核酸。
- 異種配列が、cdk2、cdk8、cdk2l、cdc25A、サイクリンDl、サイクリンE、サイクリンA、cdk4並びにp53、c-fos、c-jun、Kr-rasおよびHer2/neuのオンコジーン形態からなる群より選択される遺伝子をコードするRNAを特異的に切断するリボザイムをコードする、請求項12記載の核酸。
- H19プロモーターが配列番号1のヌクレオチド1〜830からなる、請求項19記載の核酸。
- H19プロモーターが配列番号2の配列からなる、請求項19記載の核酸。
- H19エンハンサーがプラスミドpH19EH19(ATCC受託番号209322)中にクローン化されたH19エンハンサーの配列からなる、請求項19記載の核酸。
- H19エンハンサーが配列番号3の配列からなる、請求項19記載の核酸。
- H19エンハンサーが配列番号4の配列からなる、請求項19記載の核酸。
- H19エンハンサーが配列番号5の配列からなる、請求項19記載の核酸。
- H19エンハンサーが異種配列の3'側に配置される、請求項19記載の核酸。
- 請求項12記載の核酸を含有する、腫瘍細胞において配列を発現させるためのベクターであって、少なくとも1つの調節配列がIGF-2調節配列を含むものである、前記ベクター。
- H19プロモーター配列をさらに含む、請求項30記載のベクター。
- IGF-2調節配列がIGF-2 P4プロモーターである、請求項30記載のベクター。
- IGF-2調節配列がIGF-2 P3プロモーターである、請求項30記載のベクター。
- H19プロモーターとH19エンハンサーとをさらに含み、且つ異種配列がβ−ガラクトシダーゼ、ジフテリア毒素、シュードモナス毒素、リシン、コレラ毒素、網膜芽細胞腫遺伝子、p53、単純ヘルペスチミジンキナーゼ、水痘−帯状疱疹チミジンキナーゼ、シトシンデアミナーゼ、ニトロレダクターゼ、シトクロムp-450 2B1、チミジンホスホリラーゼ、プリンヌクレオシドホスホリラーゼ、アルカリホスファターゼ、カルボキシペプチダーゼAおよびG2、リナマラーゼ(linamarase)、β−ラクタマーゼおよびキサンチンオキシダーゼからなる群より選択されるタンパク質をコードする、請求項31記載のベクター。
- 請求項30記載のベクターを含む宿主細胞。
- 請求項12記載の核酸と製薬上許容される担体とを含む、被験体の癌治療のための医薬組成物であって、少なくとも1つの調節配列がIGF-2調節配列を含むものである、前記医薬組成物。
- IGF-2調節配列がIGF-2 P4プロモーターである、請求項36記載の医薬組成物。
- IGF-2調節配列がIGF-2 P3プロモーターである、請求項36記載の医薬組成物。
- 細胞傷害性の遺伝子産物がジフテリア毒素、シュードモナス毒素、リシン、コレラ毒素、網膜芽細胞腫遺伝子およびp53からなる群より選択される、請求項36記載の医薬組成物。
- 癌が膀胱癌、肝細胞性癌、肝芽細胞腫、横紋筋肉腫、卵巣癌、子宮頸癌、肺癌、乳癌、頭部および頸部の扁平上皮癌、食道癌、甲状腺癌、神経膠星状細胞腫、神経節芽細胞腫および神経芽細胞腫から選択される、請求項36記載の医薬組成物。

- 患者における増殖性疾患または障害を治療または予防するための医薬の調製方法であって、

患者への投与に適した製剤中に前記核酸を製剤化する工程と、

- 増殖性疾患が癌である、請求項44記載の使用。
- 癌が膀胱癌、肝細胞性癌、肝芽細胞腫、横紋筋肉腫、卵巣癌、子宮頸癌、肺癌、乳癌、頭部および頸部の扁平上皮癌、食道癌、甲状腺癌、神経膠星状細胞腫、神経節芽細胞腫および神経芽細胞腫から選択される、請求項45記載の使用。
- 医薬がリポソーム投与に適合したものである、請求項44記載の使用。
- 無菌充填バイアルまたはアンプルに請求項12記載の核酸を含有するベクターを含む、増殖性疾患または障害を治療するためのキット。
- 増殖性疾患が癌である、請求項48記載のキット。
- 癌が膀胱癌、肝細胞性癌、肝芽細胞腫、横紋筋肉腫、卵巣癌、子宮頸癌、肺癌、乳癌、頭部および頸部の扁平上皮癌、食道癌、甲状腺癌、神経膠星状細胞腫、神経節芽細胞腫および神経芽細胞腫から選択される、請求項49記載のキット。
- 請求項12記載の核酸を含むウイルスベクターを含んでなる増殖性疾患または障害を治療するための剤形であって、1×107プラーク形成単位〜1×1010プラーク形成単位を含む、前記剤形。
