Abstract
Description
(少なくとも一部が)架橋可能なポリマーから形成される高分子ミクロスフィア、即ちミクロスフィアは、医学的及び工業的分野において多様な用途が見出されている。それらは、例えばドラッグデリバリー剤、組織充填剤、組織工学剤及び塞栓剤として使用可能である。従って、高分子ミクロスフィアの調合に向けて方向付けられた多くの方法がある。これらの方法には、モノマーの分散重合、乳化液に溶解された架橋可能なポリマーの電位差分散後の溶媒蒸発、静電気的にコントロールされた押出し、及び乳化液に溶解された架橋可能なポリマーの多孔質膜を通しての注入後における溶媒蒸発が含まれる。
一態様において、本発明の方法は、適切な鋳型用ポリマーから所望の又は規定のサイズの球状ビーズ又は粒子を生成する工程と、前記ビーズ又は粒子をポリビニルアルコールのような構造用ポリマーに接触させる工程と、前記構造用ポリマーを前記ビーズ又は粒子内に架橋する工程とを含む。前記鋳型用ポリマー材料はその後に取り除かれ、高分子ミクロスフィアをもたらすことが可能となる。
本発明の好ましい実施形態において、得られる高分子ミクロスフィアの好ましいサイズは、直径1−50ミクロンの範囲内である。高分子ミクロスフィアの他の好ましい粒径範囲は、直径50−100ミクロンの粒径範囲内のミクロスフィア、直径100−600ミクロンの粒径範囲内のミクロスフィア、及び直径600−1000ミクロンの粒径範囲内のミクロスフィアを含む。

Claims (36)
- 球状の高分子粒子を製造する方法であって、
鋳型用ポリマーからなる出発材料から所望のサイズの球状ビーズを生成する生成工程と、
前記ビーズを構造用ポリマーに接触させる接触工程と、
前記構造用ポリマーを架橋させる架橋工程とからなり、それによって高分子球状粒子を製造する方法。 - 請求項1に記載の方法において、前記生成工程は液滴生成器の使用からなる方法。
- 請求項1に記載の方法において、前記生成工程はスフィロナイゼーションからなる方法。
- 請求項1に記載の方法において、前記接触工程は前記構造用ポリマーを前記ビーズ内に拡散させる拡散工程からなる方法。
- 請求項4に記載の方法において、前記ビーズは多孔質である方法。
- 請求項5に記載の方法において、前記ビーズは、アルギン酸塩、多糖類、カラギナン、キトサン、ヒアルロン酸、及びカルボキシル基、硫酸基又はアミン基を備えた機能的ポリマーからなる群より選ばれる鋳型用ポリマーからなる方法。
- 請求項1に記載の方法において、前記接触工程は前記ビーズを前記構造用ポリマーで被覆する被覆工程からなる方法。
- 請求項7に記載の方法において、前記ビーズは実質的に非多孔質である方法。
- 請求項8に記載の方法において、前記ビーズはメタクリル樹脂の鋳型用ポリマーからなる方法。
- 請求項9に記載の方法において、前記ビーズはさらに充填剤を含む方法。
- 請求項10に記載の方法において、前記充填剤はデンプンである方法。
- 請求項2に記載の方法において、前記生成工程は、(i)前記液滴生成器を通して前記鋳型用ポリマーと溶媒とからなる混合物を集中させることによって液滴を形成させる工程と、(ii)前記液滴をビーズへと凝固させるために前記液滴をゲル化イオンからなるゲル化溶液内に沈降させる工程とからなり、前記ビーズは多孔性勾配を有している方法。
- 請求項12に記載の方法において、前記溶媒は前記構造用ポリマーの架橋に影響を与えない方法。
- 請求項12に記載の方法において、前記ゲル化溶液はさらに前記多孔性勾配を制限するための非ゲル化イオンを含む方法。
- 請求項12に記載の方法において、前記ゲル化溶液は前記多孔性勾配を制限しないために非ゲル化イオンを含まない方法。
- 請求項4に記載の方法において、拡散工程は前記構造用ポリマーからなる溶液に前記ビーズを浸す工程からなる方法。
- 請求項7に記載の方法において、被覆工程は前記構造用ポリマーからなる溶液に前記ビーズを浸す工程からなる方法。
- 請求項7に記載の方法において、被覆工程は前記構造用ポリマーからなる溶液を前記ビーズにスプレーする工程からなる方法。
- 請求項1に記載の方法において、さらに前記架橋工程の後に前記球状の高分子粒子を前記鋳型用ポリマーのみに選択的な溶媒に曝すことにより前記構造用ポリマーを除去する除去工程を含む方法。
- 請求項19に記載の方法において、前記溶媒はヘキサメタリン酸ナトリウムの溶液からなる方法。
- 請求項19に記載の方法において、前記溶媒はエチレンジアミンテトラ酢酸の溶液からなる方法。
- 請求項19に記載の方法において、前記溶媒はアセトンからなる方法。
- 請求項1に記載の方法において、得られる高分子球状粒子は直径1−50ミクロンの範囲内にある方法。
- 請求項1に記載の方法において、得られる高分子球状粒子は直径50−100ミクロンの範囲内にある方法。
- 請求項1に記載の方法において、得られる高分子球状粒子は直径100−600ミクロンの範囲内にある方法。
- 請求項1に記載の方法において、得られる高分子球状粒子は直径600−1000ミクロンの範囲内にある方法。
- 請求項1に記載の方法において、前記鋳型用ポリマーは形態形成剤である方法。
- 請求項1に記載の方法において、前記構造用ポリマーはポリビニルアルコール、ポリアクリルアミド、ポリエチレングリコール、ポリアミド、ポリウレア、ポリウレタン及びその誘導体からなる群より選ばれる方法。
- 請求項1に記載の方法において、前記架橋工程は架橋剤の適用からなる方法。
- 請求項29に記載の方法において、前記架橋剤は前記構造用ポリマーと共有結合を形成する方法。
- 請求項1に記載の方法において、前記架橋工程は放射線の適用からなる方法。
- 請求項29に記載の方法において、前記出発材料は前記架橋剤からなる方法。
- 球状の高分子粒子を製造する方法であって、氷のビーズを生成する生成工程と、前記ビーズを構造用ポリマーに接触させる接触工程と、前記構造用ポリマーを架橋させる架橋工程とからなり、それによって高分子球状粒子を製造する方法。
- 請求項33に記載の方法において、さらに前記架橋工程の後に前記粒子を高温に曝すことにより前記氷を除去する除去工程を含む方法。
- 請求項33に記載の方法において、前記接触工程は前記ビーズを前記構造用ポリマーで被覆する被覆工程からなる方法。
- 請求項33に記載の方法において、前記構造用ポリマーはポリビニルアルコール、ポリアクリルアミド、ポリエチレングリコール、ポリアミド、ポリウレア、ポリウレタン及びその誘導体からなる群より選ばれる方法。
