Fra tysk patentskrift nr. 1.493.574 kendes forbindelser med den almene formel: r1 5 5 fV C - CH - CH, - N/R r-s/ 0 R4 ' CH - CH— R2 r3 CH3 R6 12 3 I denne formel er E , R og R , der er ens eller forskel-10 lige, hydrogen eller en hydroxygruppe, en methoxygruppe 4 eller chlor. R er hydrogen eller en methyl- eller ethyl- C β gruppe. R er hydrogen eller en methylgruppe. R er hy- 7 8 drogen eller en hydroxygruppe. R og R , der er ens eller forskellige, er hydrogen, en methylgruppe, en metho-15 xygruppe eller chlor.
Disse forbindelser er farmakodynamisk virksomme og forbedrer navnlig hjertefunktionen.
Opfindelsen angår en analogifremganasmåde til fremstilling af hidtil ukendte cycloaliphatiske keto- eller 20 hydraxyaminer med den almene formel I: r. Λ 3

3 \=/ Λ hvor X er en gruppe >CO eller >CH(OH), & er hydrogen 3 eller en C^-C^-alkyIgruppe, og R er hydrogen eller en 25 hydroxygruppe, og R1 er en adamantylgruppe eller en mættet eller enkelt umættet C^-C^g-cycloalkylgruppe, idet denne C^-C^g-cycloalkylgruppe også kan være substitueret med en C^-C^-alkylgruppe eller med et halogenatom, eller syreadditionssalte deraf, hvilken fremgangsmåde er ejen-30 dommelig ved det i kravets kendetegnende del anførte.
De enkelte fremstillingsmetoder er omtalt mere detaljeret nedenfor.
2 R er fortrinsvis en C^-C^-alkylgruppe, navnlig methyl eller ethyl. Den mættede eller umættede C3 C16 35 cycloalkylgruppe består fortrinsvis af 3 til 12 C-ato- 2 146687 mer, navnlig 3 til 8 C-atomer. Hvis denne cycloalkyl-gruppe er substitueret, drejer det sig fortrinsvis om methyl, ethyl, chlor, brom eller fluor.
De omhandlede forbindelser er farmakodynamisk virk-5 somme og bevirker for eksempel en sænkning af blodtrykket samt en forbedring af den cerebrale eller perifere blodgennemstrømning. Endvidere har de en antiphlogistisk virkning og udmærker sig ved en god resorption. For eksempel opnås ved intraduodenal anvendelse (narkotiseret lo hund) allerede efter 15 minutter den maksimale biologiske effekt (f.eks. ved forbedring af femoralisblodgennem-strømning). Den udmærkede resorption viser sig også ved, at de optrædende biologiske effekter (f.eks. forøgelse af blodgennemstrømningen i Arteria Femoralis) ved samme 15 dosis er så at sige identiske ved intraduodenal anvendelse og ved intravenøs anvendelse.
Ved de angivne fremstillingsmetoder er det ofte hensigtsmæssigt i udgangsforbindelserne at beskytte den phenoliske hydroxygruppe med i og for sig kendte beskyt-2o telsesgrupper. Ofte er sådanne beskyttelsesgrupper allerede nødvendige for fremstillingen af udgangsforbindelserne selv. Disse beskyttelsesgrupper kan let fraspaltes fra slutprodukterne. Det drejer sig enten om let solvo-lytisk fraspaltelige acylgrupper eller ved hydrogenering 25 fraspaltelige grupper, såsom benzylgruppen. De solvoly-tisk fraspaltelige beskyttelsesgrupper fraspaltes for eksempel ved forsæbning med fortyndede mineralsyrer i et opløsnings- eller suspensionsmiddel (lavere alkoholer) ved temperaturer mellem 10° og 150°C. Alt efter arten af 3o beskyttelsesgruppen foregår der imidlertid også fraspalte ning allerede under fremstillingsreaktionen. Sidstnævnte er for eksempel tilfældet, når den phenoliske hydroxygruppe er beskyttet med en benzylgruppe eller carboben-zoxygruppe, og fremgangsmåden omfatter et.hydrogenerings-35 trin. Hvis beskyttelsesgruppen ikke fraspaltes under reaktionen, kræves en simpel efterbehandling af reaktionsproduktet, hvorved da fraspaltningen af beskyttelses- 3 1A6687 gruppen foregår for eksempel under betingelser som de ovenfor anførte.
Som beskyttelsesgrupper kommer for eksempel følgende i betragtning: benzylgruppen, a-phenylethylgruppen, i 5 benzenkernen substituerede benzylgrupper, såsom p-brom-eller p-nitro-benzylgruppen, carbobenzoxygruppen, carbo-benzthiogruppen, trifluoracetylgruppen, phthalylgruppen, tritylgruppen eller p-toluensulfonylgruppen, men endvidere er her også simple acylgrupper, såsom acetylgruppen, 10 formylgruppen eller tert.-butylcarboxygruppen egnede.
Reduktionen af ketogruppen i forbindelser, hvor X er gruppen >C0, til forbindelser, hvor X er gruppen^CHOH, samt reduktionen af en dobbeltbinding i gruppen gennemføres i almindelighed ved katalytisk hydrogenering.
15 Som katalysatorer kommer for eksempel de sædvanlige findelte metalkatalysatorer, såsom ædelmetalkatalysatorer, f.eks. Raney-nikkel, platin eller navnlig palladium i betragtning. Fremgangsmåden kan gennemføres ved normale temperaturer eller ved forhøjede temperaturer. Der arbej-20 des hensigtsmæssigt inden for temperaturområdet fra ca.
40 til ca. 200°C, eventuelt ved forhøjet tryk (1-100, navnlig 1-50 bar). Hvis den phenoliske hydroxygruppe indeholder en benzylbeskyttelsesgruppe, bliver denne fraspaltet samtidigt med den katalytiske hydrogenering, når 25 der for eksempel anvendes en palladiumkatalysator.
Reduktionen af ketogruppen kan imidlertid foregå på andre måder, for eksempel ved hjælp af komplekse metalhy-drider (f.eks. lithiumaluminiumhydrid, natriumborhydrid, cyanoborhydrid , lithium-tri-tert.-butoxy-aluminiumhy-30 drid) eller ved hjælp af aluminiumalkoholater ifølge Meerwein og Ponndorf (f.eks. ved hjælp af aluminiumiso-propylat) ved temperaturer mellem 0° og 150°C, navnlig 20-100°C. Som opløsningsmiddel eller suspensionsmiddel for denne reaktion kommer for eksempel følgende i be-35 tragtning: Lavere aliphatiske alkoholer, dioxan, tetrahy-drofuran, vand eller aromatiske carbonhydrider, såsom benzen, toluen samt blandinger af disse midler.

En selektiv reduktion af en dobbeltbinding i gruppen R1 er for eksempel mulig under skånende betingelser ved hydrogenering i nærværelse af ædelmetalkatalysatorer (Pd, Pt) eller Raney-nikkel.
5 Vedrørende de af kravet omfattede fremgangsmåder a), b) og c) til fremstilling af de omhandlede forbindelser skal bemærkes følgende:
Fremgangsmåde a):
Denne fremgangsmåde gennemføres i almindelighed i 10 et indifferent opløsningsmiddel eller suspensionsmiddel ved temperaturer mellem 5° og 250°C. Som opløsningsmiddel kommer for eksempel følgende i betragtning: Lavere ali-phatiske alkoholer (ethanol, methanol, isopropanol, propanol) , mættede alicycliske og cycliske ethere (dioxan, 15 tetrahydrofuran, diethylether), lavere aliphatiske ketoner (acetone), lavere aliphatiske carbonhydrider eller halogencarbonhydrider (chloroform, 1,2-dichlorethan), aromatiske carbonhydrider (benzen, xylen, toluen), iseddikesyre, vand eller blandinger af disse midler. Hvis 20 der som udgangsforbindelse anvendes en forbindelse med formlen III, hvor E er en gruppe arbejdes for det meste i den nedre del af det angivne temperaturområde (5-80°C), idet der her som opløsningsmiddel navnlig anvendes lavere alkoholer, ethere, acetone, dioxan eller chloro-25 form. Ved anvendelse af en forbindelse III, hvor E er hydrogen og gruppen -C^-NRgR^, arbejdes for det meste i et højere temperaturområde (80-250°C, navnlig 80-150°C, hvis X = CO), idet der som opløsningsmiddel navnlig anvendes vand, alkohol/vand eller et tofasesystem, såsom 20 vand/benzen eller vand/toluen.
Hvis der anvendes en udgangsforbindelse III, hvor E repræsenterer to hydrogenatomer, arbejdes navnlig mellem 20° og 150°C.
Som formaldehyd-afgivende stoffer kommer for eksem-25 pel formaldehydacetaler i betragtning, ud fra hvilke der i nærværelse af syreanioner frigøres formaldehyd, såsom polyformaldehyd, paraformaldehyd, hexamethylentetramin.
I stedet for formaldehyd og aminen med formlen II kan 146687 5 også en forud ud fra disse stoffer fremstillet oxazoli-din med formlen II': ^CH2- 0 R3' I ri/ 111 5 ^"CH -CH —<' y ^ A=/ 3' hvor R har ovennævnte betydning, eller et mineralsyre- additionssalt (for eksempel hydrochlorid eller hydrobrom- 10 id) deraf omsættes med forbindelsen med formlen R -CO-2 C(R )H2. Hvis oxazolidinderivatet ikke benyttes som salt, er det hensigtsmæssigt at arbejde i nærværelse af fortyndede mineralsyrer (for eksempel 20%'s alkoholisk saltsyre eller hydrogenbromidsyre).

Udgangsforbindelserne med formlen R -CO-C(R )H9, 1 ^ hvor R er en mættet cycloalkylgruppe, kan for eksempel 20 vindes ud fra de tilsvarende umættede forbindelser (der kan vindes ifølge J.org.Chem. 15, 1147 (1950)) ved katalytisk hydrogenering i nærværelse af metalliske hydrogeneringskatalysatorer (Pd) i lavere alkoholer mellem 20° og 100°C.
1 2 25 Udgangsforbindelserne med formlen R -X-C(R )H-CH2” NR R, kan for eksempel vindes ved Mannichreaktion af ke-a b 12 toner med formlen R -CO-C(R )H2 med dimethylaminhydro- chlorid i nærværelse af formaldehyd. Udgangsforbindelser 1 2 med formlen R - X-C(R )=CH2 kan for eksempel vindes ud 30 fra ketoner med formlen R^-CO-C (R^)H-CH2~NRaRj;) ved fra-spaltning af amindelen (for eksempel ved vanddampdestillation) eller ved termisk HCl-fraspaltning fra de tilsvarende cycloaliphatiske 3-chlor—ethylketoner med formlen 1 2 R —CO—CH(R )-CH„C1. En tilstedeværende dobbeltbinding i 1 ^ 35 rx og/eller CH-gruppen X kan derefter eventuelt på sædvanlig måde reduceres (gruppen^ CO til gruppen } CH (OH)). Fremgangsmåde b):
Denne fremgangsmåde gennemføres hensigtsmæssigt i 146687 6 et opløsnings- eller suspensionsmiddel, såsom aromatiske carbonhydrider (benzen, toluen, xylen), aliphatiske alkoholer (ethanol, propanol, butanol), lavere aliphatiske syreamider (dimethylformamid), tetramethylurinstof, di-5 methylsulfoxid, alicycliske og cycliske mættede ethere (diethylether, dioxan) ved temperaturer mellem 20° og 200°C, navnlig 50° til 140°C. Det tilrådes at arbejde i nærværelse af en syreacceptor (kaliumcarbonat, natrium-carbonat, alkalimetalalkoholater, alkalimetalamider, al-10 kalimetalcarbonater, tertiære aminer), hvis for eksempel Z indeholder et halogenatom og W en aminogruppe (eller omvendt). Som halogenatomer kommer for eksempel chlor, brom og iod i betragtning. Som syreacceptor kan også tjene et overskud af amin.
15 Fremgangsmåden gennemføres i nærværelse af hydrogen, hvis Z er en aminomethylgruppe, og W er en 1-oxo-ethyl-gruppe, eller hvis Z er en formylgruppe, og W er en 1-aminoethylgruppe.
Som katalysatorer kommer her de sædvanlige hydro-20 generingskatalysatorer i betragtning, fortrinsvis metalliske hydrogeneringskatalysatorer, såsom Raney-nikkel, platin, palladium. Der kan imidlertid også anvendes al-kalimetalborhydrider (NaBH^).
Hvis der samtidigt med den hydrogenerende konden-25 sation skal fjernes en benzylbeskyttelsesgruppe eller en anden ved hydrogenering fraspaltelig beskyttelsesgruppe, anvendes fortrinsvis palladium-katalysatorer.

1 2 15 Udgangsforbindelser med formlen R -X-C(R )H-CH2- NH2 kan for eksempel vindes ud fra de tilsvarende halogenforbindelser og phthalimidkalium i et opløsningsmiddel (toluen, xylen) ved kogetemperatur og påfølgende fraspaltning af phthalimidoresten med mineralsyre (25% 20 HC1) i et opløsnings- eller suspensionsmiddel (iseddike-syre/^O, alkohol/E^O) mellem 80° og 200°C.
Fremgangsmåde c);
Denne fremgangsmåde gennemføres i et opløsningseller suspensionsmiddel ved temperaturer mellem 0° og 25 150°C. Som opløsningsmiddel kommer for eksempel de til
Grignard-reaktioner sædvanlige midler i betragtning, såsom lavere aliphatiske ethere (diisobutylether), tetra-hydrofuran, aromatiske carbonhydrider (benzen, toluen, xylen).

5 Som opløsningsmiddel kommer for eksempel følgende i betragtning: Lavere alkoholer, ethere, dioxan, benzen. Ved forsæbning eller sædvanlig omsætning med halogenerings-midler vindes derudfra· sådanne udgangs forbindelser, hvori A er en carboxygruppe eller halogencarbonylgruppe.
10 Alt efter fremgangsmådebetingelserne og udgangs forbindelserne vindes slutprodukterne med formlen I i fri form eller i form af deres salte. Saltene af slutprodukterne kan på i og for sig kendt måde, for eksempel med alkalimetalbaser eller ionbyttere, atter overføres i baserne. Ud fra sidst-15 nævnte kan der ved omsætning med organiske eller uorganiske syrer, navnlig sådanne der er egnede til dannelse af terapeutisk anvendelige salte, vindes salte. Som sådanne syrer kan for eksempel nævnes: Hydrogenhalogenid-syrer, svovlsyre, phosphorsyrer, salpetersyre, perchlor-20 syre, organiske mono-, di- eller tricarboxylsyrer fra den aliphatiske, alicycliske, aromatiske eller hetero-cycliske række samt sulfonsyrer. Eksempler herpå er: Myresyre, eddikesyre, propionsyre, ravsyre, glycolsyre, mælkesyre, æblesyre, vinsyre, citronsyre, ascorbinsyre, 25 maleinsyre, fumarsyre, hydroxymaleinsyre eller pyrodrue-syre, phenyleddikesyre, benzoesyre, p-amino-benzoesyre, anthranilsyre, p-hydroxy-benzoesyre, salicylsyre eller p-amino-salicylsyre, embonsyre, methansulfonsyre, ethan-sulfonsyre, hydroxyethansulfonsyre, ethylensulfonsyre, 30 halogenbenzensulfonsyre , toluensulfonsyre, naphthalen-sulfonsyre eller sulfanilsyre eller også 8-chlor —theo-phyllin.
De forbindelser, der indeholder asymmetriske car-bonatomer og i reglen fremkommer som racemater, kan på i 35 og for sig kendt måde, for eksempel ved hjælp af en optisk aktiv syre, opdeles i de optisk aktive isomere. 10 Farmakologiske eller farmaceutiske data.
De omhandlede forbindelser er egnede til fremstilling af farmaceutiske præparater. De farmaceutiske præparater eller lægemidler indeholder som virksomt stof én eller flere af de omhandlede forbindelser, eventuelt i 15 blanding med andre farmakologisk eller farmaceutisk virksomme stoffer. Fremstillingen af lægemidlet foregår på i og for sig kendt måde, idet de kendte og sædvanlige farmaceutiske hjælpestoffer samt andre sædvanlige bære- og fortyndingsmidler kan anvendes.
20 Anvendelsen af det virksomme stof eller lægemidlet kan ske på hud eller slimhud eller i legemets indre, f. eks. oralt, enteralt, pulmonalt, rectalt, nasalt, vaginalt, lingualt, intravenøst, intraarterielt, intracardi-alt, intramuskulært, intraperitonealt, intracutant eller 25 subcutant.
De omhandlede forbindelser viser på narkotiseret hund en stærk og vedvarende udvidelse af de perifere kar (målt med elektromagnetisk flowmeter, se f.eks. A.Kolin og R.T.Kado, Proc. Natl.Akad. Sc. £5, 1312 (1959), eller 30 Best-Taylor, The Physiological Basis of Medical Practice, Baltimore 1966, 8.udgave, side 702 ff.) under samtidig forøgelse af hjerteydelsen målt ved hjertetidsvolumen. Hjertetidsvolumenet fandtes ved kuldefortyndingsmetoden ifølge H.Slama og J.Piiper, Kreislaufforschung 53, 322 35 (1964).
For eksempel fandtes ved ovennævnte forsøgsmetode ved en dosis på 1,5 mg/kg legemsvægt på hund intraduode-nalt en forøgelse af blodgennemstrømningen i Arteria femoralis på 40% (middelværdi over 180 minutter).

Denne karudvidende virkning kan sammenlignes med virkningen af det kendte lægemiddel cinnarizin.
Den laveste, endnu hæmodynamisk virksomme dosis ved det ovennævnte dyreforsøg var for eksempel: 5 0,3 mg/kg oralt og 0,03 mg/kg intravenøst.
Som almindeligt dosisområde for den karudvidende virkning (dyreforsøg som ovenfor) kommer for eksempel følgende i betragtning: 1-20 mg/kg oralt, navnlig fra 3 til 10 mg/kg, eller ' 10 0,5-5 mg/kg intravenøst, navnlig fra 0,3 til 3 mg/kg.
Indikationer for hvilke de omhandlede forbindelser kommer i betragtning:
Perifere blodgennemstrømningsforstyrrelser, cerebrale blodgennemstrømningsforstyrrelser, akut, 15 glycosidrefraktær insufficiens, diabetiske angio- pathier, Ulcus cruris, funktionelle blodgennemstrømningsforstyrrelser, hæmning af veer ved truende abort.
De farmaceutiske præparater indeholder i alminde-2Q lighed mellem 1 og 50 mg af den eller de omhandlede aktive komponenter.
Anvendelsen kan for eksempel ske i form af tabletter, kapsler, piller, dragée, stave, salver, geler, cremer, pudderprodukter, støvpulver, aerosoler eller i fly-25 dende form.
Den akutte toksicitet af de omhandlede forbindelser på mus (udtrykt ved LD 50 mg/kg, metode ifølge Miller og Tainter, Proc. Soc. Exper. Biol. a. Med. 57 (1944) 261) ligger for eksempel ved oral anvendelse mellem 250 og 30 600 mg/kg.
Fremgangsmåden ifølge opfindelsen beskrives nærmere gennem følgende eksempler, hvori de angivne udbytter er baseret på det anvendte norephedrin eller hydroxynorephe-drin.

Til isolering af slutproduktet er det ofte hensigtsmæssigt, at det for det meste som hydrochlorid fremkommende umiddelbare reaktionsprodukt ved behandling med for eksempel fortyndet ammoniak overføres i den frie ba-5 se, der eventuelt på sædvanlig måde atter overføres i et salt.
Eksempel 1 -i- [3-Hydroxy-3-phenyl-propyl- (2) ]- [3- (1-cyclohexen-l-yl)-10 3-oxo-propyl]-amin.

Yderligere 10 g parafomaldehyd blev inden for reaktionstiden tilsat i portioner på 2 gram. Det ved afkøling udkrystalliserende produkt blev frasuget på nutsch og omkrystalliseret af ethanol/vand.
Udbytte 48%. Smp. af hydrochlorid: 203-204°C.

AlClj ved -5°C efterfulgt af 4 timers gennemledning af éthylen under -5°C, påfølgende hydrolyse med 500 ml vand ved stuetemperatur og inddampning af den med Na2S0^ tørrede organiske fase i vakuum (analogt med USA-patentskrift 20 nr. 2.792.406)vundne olieagtige cyclohexyl-3-chlorethyl-keton blev sat til en opløsning af 164 g (1,09 mol) Z-norephedrin i 1000 ml dioxan. Det ønskede produkt udkrystalliserede i løbet af natten, blev frasuget på nutsch og omkrystalliseret af ethano1/vand 1:1.
25 Udbytte: 67% (beregnet på cyclohexancarboxylsyrechlorid).

Eksempel 18 £-erythro-[3-Hydroxy-3-(3-hydroxyphenyl)-propyl-(2)]—(3— cyclohexy1-3-oxo-propy1)-amin.
30 _ O CH~ OH _ C-CH2CH2-NH-CH-CH — 17 146687 10 g (0,057 mol) Cyclohexyl-3-chlorethyl-keton, opløst i 30 ml acetonitril, blev ved 75°C dryppet til en blanding af 18 g (0,057 mol) /-erythro-m-hydroxy-nore-phedrin-hydrogentartrat og 23,8 ml triethylamin i 150 ml 5 acetonitril. Der blev opvarmet 1 time under tilbagesvaling, opløsningsmidlet blev afdestilleret, og til ind-dampningsresten blev der sat 50 ml varmt vand. Efter tre ganges udrystning med methylisobutylketon blev der efter tørring med natriumsulfat tilsat den for saltdannelsen 10 teoretisk nødvendige mængde oxalsyre, opløst i 20 ml methylisobutylketon. Det ved afkøling udkrystalliserende oxalat blev renset ved omkrystallisation af isopropanol.
Udbytte: 22%.
Smp. af oxalat: 174-175°C.
15 Eksempel 19 threo-[3-Hydroxy-3-phenyl-propyl-(2)]-(3-cyclohexyl-3-oxo-propyl)-amin.
O ch3 OH __ / h \—- c - ch2ch2-nh-ch-ch “V^J/ 20 8,6 g (0,057 mol) £-ψ-Norephedrin og 15,8 g kalium- carbonat blev i 150 ml xylen opvarmet under omrøring til 100°C, og der blev dråbevis tilsat en opløsning af 10 g cyclohexy1-3-(chloro-ethyl)-keton i 30 ml xylen. Efter endt tildrypning blev der opvarmet yderligere 1 time un- 25 der tilbagesvaling, og opløsningen blev filtreret varmt.
Den ved afkøling udkrystalliserende base blev opløst i methylethylketon, og der blev tilsat 10 ml 5N isopropa-nolisk saltsyre. Det vundne hydrochlorid blev omkrystalliseret af isopropanol.

Eksempel 20 l-[3-Hydroxy-3-phenyl-propyl-(2)]-(3-cyclohexyl-3-oxo-propyl)-amin.
Til en cyclohexylmagnesiumbromid-etheropløsning, 5 fremstillet ud fra 6,1 g (0,25 mol) magnesium og 48 g (0,295 mol) cyclohexylbromid, blev en opløsning af 25,2 g (0,1 mol) ^-2-norephredrino-propionsyreethylester i 150 ml tetrahydrofuran dryppet ved 5-10°C. Efter fire timers opvarmning under tilbagesvaling blev reaktionsblandingen 10 hældt på is-kogsalt (300 g is, 100 g H20, 100 g NH^Cl) , og den organiske fase blev fraskilt. Den vandige fase blev ekstraheret flere gange med ether, tørret over magnesiumsulfat, opløsningsmidlet blev afdestilleret i vakuum, og hydrochloridet blev fremstillet med 8N isopropano-15 lisk HC1. Dette blev omkrystalliseret to gange af etha-nol/vand (1:1).
Udbytte: 6%.
Smp. af hydrochlorid: 219-221°C.
Eksempel 21 20 (j- [3-Hydroxy-3-phenyl-propyl- (2) ] - (3-cyclohexyl-3-oxo- propy1)-amin.
^ CO-CH2-CH2-NH-CH (CH3) -CH (OH) —^ ^ 25 g £-(3-Hydroxy-3-phenyl-propyl-(2)]-[3-(l-cyclo-hexen-l-yl)-3-oxo-propyl]-amin-hydrochlorid blev opløst 25 i 250 ml methanol/vand (2:1), og der blev tilsat 2,5 g Pd-C (10%*s) og hydrogeneret ved 50°C og 5 bar indtil endt hydrogenoptagning. Derefter blev katalysatoren frafiltreret, opløsningsmidlet blev afdestilleret i vakuum, og produktet blev omkrystalliseret af ethanol.
30 Udbytte: 85%.

Eksempel 35 λ-[3-Hydroxy-3-(4-hydroxyphenyl)-propyl-(2)]-(3-cyclo-hexyl-3-oxo-propyl)-amin.

En blanding af 6 g (0,037 mol) β-chlorethylcyclo-10 hexylketon, 9,5 g (0,037 mol) i-(4-benzyloxy)norephedrin og 10 g blev i 500 ml xylen opvarmet 4 timer til kogning. Efter frafiltrering af dannet KC1 blev filtratet inddampet i vakuum. Fremstillingen af hydrochloridet ud fra det udfældede råprodukt foregik på sædvanlig måde 15 med isopropanolisk saltsyre. Den herved vundne forbindelse svarede til ovennævnte formel, bortset fra at hydroxy-gruppen på phenylringen var beskyttet af en benzylgruppe. Hydrochloridet af denne mellemproduktforbindelse havde smp. 2Q6-2Q7°C, udbytte 57%.
2Q Til hydrolytisk fraspaltning af O-benzylgruppen blev 8 g (Q,Q2 mol) af den beskyttelsesgruppeholdige forbindelse opløst i 150 ml isopropanol og 20 ml vand, og der blev tilsat 30 ml 6N isopropanolisk saltsyre. Reaktionsblandingen blev kogt 24 timer under tilbagesvaling.
25 Opløsningsmidlet blev i vakuum inddampet til 4/5. Det ved afkøling udkrystalliserede produkt blev fraskilt på nutsch og omkrystalliseret af isopropanol. Hydrochloridet havde smp. 212^214°C, udbytte 40%.
Ved anvendelse af en udgangsforbindelse, der i ste-3Q det for benzylgruppen indeholdt en carbobenzoxy- eller carbobenzthiogruppe, en p-toluensulfonylgruppe eller en lavere aliphatisk acylgruppe, blev der opnået samme resultat .

146687 Analogifremgangsmåde til fremstilling af cycloali-phatiske keto- eller hydroxyaminer med den almene formel Is R1 2 3 4 5 6 7 8 r1-x-ch-ch2-nh-ch(ch3)-ch(oh)—^ I R2 2 hvor X er en gruppe>CO eller ^CH (OH), R er hydrogen el-10 ler en C-j-Cg-alkylgruppe, R1 er hydrogen eller en hy-droxygruppe, og R1 er en adamantylgruppe eller en mættet eller enkelt umættet C^C^-cycloalkylgruppe, idet denne Cs-Cig-eycloalkylgruppe også kan være substitueret med en C,-CA-alkylgruppe eller med et halogenatom, eller sy-15 1 4 readditipnssalte deraf,· kendetegnet ved, at a) en amin med den almene formel II: R1'
20 H2N-CH (CH3) -CH (OH) 9 35 1 3 2 hvor R har samme betydning som R ovenfor eller er en 3 med en sædvanlig beskyttelsesgruppe beskyttet hydroxy- 4 25 gruppe, omsættes med en forbindelse med den almene formel 5 III: R1-X-C=E III R2 6 30 12 7 hvor X, R og R har ovennævnte betydninger, og E er en methylengruppe eller et hydrogenatom og gruppen -CH2-NR og Ra og er lavmolekylære alkylgrupper, der også kan være sammensluttet til en ring, eller idet E 8 også kan betyde to hydrogenatomer, når X er gruppen CO, 9 og der arbejdes i nærværelse af formaldehyd eller et formaldehydafgivende stof, eller der anvendes i stedet for forbindelsen II og formaldehyd en oxazolidin med 146687 formlen II': /CH2-°| 3' HN\ I
