Description
- Die vorliegende Erfindung betrifft ein Verfahren zur Stabilisierung einer pharmazeutischen Zusammensetzung durch Kontaktierung dieser Zusammensetzung mit einem ein Antioxidans enthaltenden Polymermaterial, die entsprechende Verwendung dieses Antioxidans, eine nach diesem Verfahren erhaltene pharmazeutische Zusammensetzung und einen Gegenstand, der aus einem ein Antioxidans enthaltenden Polymermaterial hergestellt worden ist.
- In
JP 08 165 240 A - Die
EP 0 827 741 A bezieht sich auf eine Zusammensetzung, die ein nicht steroidales Arzneimittel enthält, das in Klebstoffen auf Basis eines nicht polaren Polymers dispergiert ist, wobei eine oxidative Zusammensetzung des Polymers durch Zusatz von Antioxidantien verhindert ist. - Die
US 3 966 905 A betrifft eine Kombination von Catecholamin mit einem Polyvinylpyrrolidonpolymer und einem Antioxidans. - Die
US 4 959 208 A betrifft eine Vorrichtung zur Abgabe eines arzneilichen Wirkstoffs, die auf einem mikroporösen Polyolefin (Polyethylen, Polypropylen) unter Einschluss eines Antioxidans (Irganox®) basiert. - Allerdings besteht noch immer Bedarf an einer Stabilisierung pharmazeutischer Zusammenset zungen, insbesondere wässriger pharmazeutischer Zusammensetzungen und vor allem wässriger Augentropfen, die gegenüber einer Zersetzung stabil sind, welche durch Einwirkung von Hitze, Licht und/oder Sauerstoff verursacht wird.
- Dieses Problem wird nun erfindungsgemäß durch den Hauptanspruch gelöst, nämlich insbesondere durch die Verwendung einer Flasche aus Kunststoff, in welchem ein Antioxidans enthalten ist. Der Vorteil einer solchen Stabilisierung besteht in der Möglichkeit einer Weglassung eines zusätzlichen Antioxidans und/oder eines Stabilisators, der sonst in solchen pharmazeutischen Zusammensetzungen benötigt werden würde. Solche pharmazeutische Zusammensetzungen verfügen daher gewöhnlich über eine verbesserte Verträglichkeit im Vergleich zu in üblicher Weise stabilisierten Zusammensetzungen, da das Antioxidans hierdurch nicht an den Organismus verabreicht wird, der eine solche pharmazeutische Zusammensetzung braucht.
- Ein erstes Ziel der vorliegenden Erfindung ist daher ein Verfahren zur Stabilisierung einer phar mazeutischen Zusammensetzung, was den Zusatz eines Antioxidans erfordern würde, damit diese Zusammensetzung im wesentlichen genauso stabil wie nach diesem Verfahren wäre, gekennzeichnet durch Kontaktierung der Zusammensetzung, die frei an Antioxidans ist, mit einem Polymermaterial, das in der pharmazeutischen Zusammensetzung unlöslich ist und ein Antioxidans enthält, welches in das Polymermaterial während des Herstellungsverfahrens dieses Materials eingearbeitet worden ist, wobei das Polymermaterial in Form eines Gegenstands vorliegt, der aus einem Schlauch, einem Gefäß und einer Flasche ausgewählt ist.
- Ein weiteres Ziel der vorliegenden Erfindung ist die Verwendung eines Antioxidans, das in ein Polymermaterial während des Herstellungsverfahrens dieses Materials zwecks Stabilisierung einer pharmazeutischen Zusammensetzung eingearbeitet worden ist, die ausgewählt ist aus einer Lösung, einer Suspension, einem Gel, einer Salbe, einer Emulsion oder einem Gemisch hiervon, was den Zusatz eines Antioxidans erfordern würde, damit diese Zusammensetzung im wesentlichen genauso stabil wäre, wie dies durch diese Verwendung verursacht worden wäre, die aber frei davon ist.
- Ein wiederum weiteres Ziel der Erfindung ist eine pharmazeutische Zusammensetzung, die nach dem oben erwähnten Verfahren erhalten worden ist. Ein wiederum anderer Aspekt der Erfindung betrifft einen Gegenstand, ausgewählt aus einem Schlauch, einem Gefäß und einer Flasche und hergestellt aus einem Polymermaterial, das ein Antioxidans enthält, welches in das Polymermaterial während des Herstellungsverfahrens dieses Materials eingearbeitet worden ist, wobei dieser Gegenstand eine pharmazeutische Zusammensetzung enthält, damit diese Zusammensetzung im wesentlichen genauso stabil wäre, wie dies durch diese Verwendung verursacht worden wäre, die aber frei davon ist.
- Innerhalb der vorliegenden Erfindung bezieht sich die Angabe Stabilisierung auf die Stabilität einer pharmazeutischen Zusammensetzung insgesamt und vor allem auf die Stabilität des Wirkstoffs selbst, wenn dieser einer Lagerung, Sauerstoff und/oder Luft (Sauerstoffradikale) Licht (UV) und/oder Hitze (beispielsweise einer Sterilisationsstufe bei 121°C) ausgesetzt ist. Unter einer Hitzesterilisation wird auch eine Behandlung in einem Autoklav verstanden.
- Unter einem Polymermaterial wird ein Polymer verstanden, das vorzugsweise unlöslich ist in der flüssigen erfindungsgemäßen pharmazeutischen Zusammensetzung und das ferner irgendeine willkürliche Form haben kann. Beispiele für solche Formen sind insbesondere Pellets, Perlen, Stäbe, Riegel, Platten, Schläuche oder Behältnisse und besonders bevorzugt eine Flasche. Ein bevorzugtes Polymermaterial umfasst Polyethylen (PE), Polypropylen (PP) und/oder Gemische hiervon oder besteht daraus.
- Die Angabe Flasche aus Kunststoff bezieht sich daher insbesondere auf eine Flasche aus Polyethylen (PE) und/oder Polypropylen (PP). Optional können diese Materialien weitere Hilfsstoffe enthalten, wie ein lichtabsorbierendes Material, beispielsweise Titandioxid, ein Farbpigment, einen UV-Absorber und/oder dergleichen.
- Unter einem Antioxidans wird erfindungsgemäß eine Verbindung verstanden, die aus der Gruppe ausgewählt ist, welche besteht aus 2,2',2'',6,6',6''-Hexa(1,1-dimethylethyl)-4,4',4''-[(2,4,6-trimethyl-1,3,5-benzoltriyl)trismethylen]triphenol (Irganox®1330), 1,3,5-Tris[3,5-di(1,1-dimethylethyl)-4-hydroxybenzyl]-1H,3H,5H-1,3,5-triazin-2,4,6-trion, Pentaerythrityltetrakis[3-[3,5-di(1,1-dimethylethyl)-4-hydroxyphenyl]propionat], Octadecyl-3-[3,5-di(1,1-dimethylethyl)-4-hydroxyphenyl]propionat, Tris[2,4-di(1,1-dimethylethyl)phenyl]phosphit, 2,2'-Di(octadecyloxy)-5,5'-spirobi(1,3,2-dioxaphosphorinan), Dioctadecyldisulfid, Didodecyl-3,3'-thiodipropionat, Dioctadecyl-3,3'-thiodipropionat, Butylhydroxytoluol, Ethylenbis[3,3-di[3-(1,1-dimethylethyl)-4-hydroxyphenyl]butyrat] und Gemischen hiervon. Ein bevorzugtes Antioxidans ist Irganox®1330.
- Die Menge an im Polymermaterial enthaltenem Antioxidans liegt typischerweise im Bereich der Empfehlungen der Europäischen Pharmakopöe und reicht typischerweise von 0,05 bis 1,0 Gew.-%, bevorzugter von 0,1 bis 0,7 Gew.-%, und noch stärker bevorzugt von 0,12 bis 0,55 Gew.-%.
- Es können auch andere Antioxidantien vorhanden sein, wie Ascorbinsäure, Acetylcystein, Cystein, Natriumhydrogensulfit, Butylhydroxyanisol und alpha-Tocopherolacetat.
- Ein Vorteil eines erfindungsgemäßen Polymermaterials, das ein Antioxidans enthält, besteht darin, dass es damit typischerweise zu einer nur minimalen oder praktisch gar keinen Freisetzung des Antioxidans in ein Pharmazeutikum und insbesondere in eine wässrige ophthalmische Zusammensetzung kommt, das oder die sich mit dem Polymermaterial in Kontakt befindet. Hierdurch ergibt sich typischerweise ein praktischer Ausschluss des Antioxidans von der pharmazeutischen Zusammensetzung.
- Ein Antioxidans wird typischerweise während des Herstellungsverfahrens eines Polymermaterials verwendet, um dessen Zersetzung und/oder Oxidation zu verhindern. Ein solches Polymermaterial entspricht daher insbesondere den erfindungsgemäßen Definitionen eines ein Antioxidans enthaltenden Polymermaterials.
- Ein pharmazeutisch wirksames Mittel ist ein Mittel, das beispielsweise ausgewählt ist aus der Gruppe, die besteht aus Acetylcholinchlorid, Acyclovir, Adrenalin, Amethocain, Aminocapronsäure, Antazolinphosphat, Arachidonsäure, Atropin, Betaxolol, Bupivacain, Carbachol, Carteolol, Chloramphenicol, Chlortetracyclin, Chymatrypsin, Clonidin, Cocain, Corynanthin, Cromolyn, Cyclopentolat, Demecarium, Dexamethason, Dibutolin, Dichlorphenamid, Diclofenac, Dipivefrin, Echodtiophat, Ephedrin, Erythromycin, Ethambutol, Etidocain, Eucatropin, Fluormethalon, Fluormetholon, Gentamycin, Gramicidin, H-Thymidin, Homatropin, Hyaluronsäure, Hydrocortison, Idoxuridin, Indomethacin, Isofluorphat, Isosorbid, Ketorolac, Ketotifen, Lachesin, Levobunolol, Levocabastin, Lidocain, Lignocain, Medryson, Mepivacain, Methacholin, Methazolamid, Naphtazolin, Natamycin, Neomycin, Neostigmin, Noradrenalin, Ofloxacin, Oxybuprocain, Oxymetazolin, Oxyphenonium, Pheniramin, Phenylephrin, Physostigmin, Pilocarpin, Polymyxin B, Prednisolon, Proparacain, Proxymethacain, Pyrilamin, Retinolsäure, Retinol, Retinolacetat, Retinolpalmitat, Scopolamin, Sorbinil, Sulfacetamid, Tamoxifen, Tetracain, Tetracyclin, Tetrahydrozolin, Timolol, Trifluridin, Tropicamid, Vidarabin und pharmazeutisch annehmbaren Salzen und Gemischen hiervon.
- Bevorzugte pharmazeutisch wirksame Verbindungen sind aus der Gruppe ausgewählt, die besteht aus Betaxolol, Chloramphenicol, Diclofenac, Dipivefrin, Ephedrin, Erythromycin, Gentamycin, Indomethacin, Ketotifen, Levobunolol, Levocabastin, Ofloxacin, Pilocarpin, Polymyxin B, Prednisolon, Retinolsäure, Retinol, Retinolacetat, Retinolpalmitat, Tetracyclin und pharmazeutisch annehmbaren Salzen hiervon.
- Stärker bevorzugte pharmazeutisch wirksame Verbindungen sind aus der Gruppe ausgewählt, die besteht aus Betaxolol, Chloramphenicol, Diclofenac, Ketotifen, Levobunolol, Levocabastin, Pilocarpin, Retinolsäure, Retinol, Retinolacetat, Retinolpalmitat und pharmazeutisch annehmbaren Salzen hiervon.
- Hoch bevorzugt sind Ketotifen, Retinolsäure, Retinol, Retinolacetat, Retinolpalmitat und pharmazeutisch annehmbare Salze hiervon.
- Ganz besonders bevorzugt sind Ketotifen und pharmazeutisch annehmbare Salze hiervon.
- Innerhalb der vorliegenden Erfindung ist eine pharmazeutische Zusammensetzung charakterisiert durch den Träger, in welchem die pharmazeutisch wirksame Verbindung vermischt, suspendiert, gelöst und/oder teilweise gelöst ist, und der ausgewählt ist aus der Gruppe, die besteht aus Wasser, Gemischen aus Wasser und mit Wasser nicht mischbaren Lösemitteln, C1-C7-Alkanole, Pflanzenöle oder Mineralöle, die 0,5 bis 5 Gew.-% Hydroxyethylcellulose, Ethyloleat, Carboxymethylcellulose, Polyvinylpyrrolidon und sonstige nicht toxische wasserlösliche Polymere enthalten, insbesondere für ophthalmische Anwendungen, beispielsweise Cellulosederivate, wie Methylcellulose, Alkalimetallsalze von Carboxymethylcellulose, Hydroxymethylcellulose, Hydroxyethylcellulose, Methylhydroxypropylcellulose und Hydroxypropylcellulose, Acrylate oder Methacrylate, wie Salze von Polyacrylsäure oder Ethylacrylat, Polyacrylamide, Naturprodukte, wie Gelatine, Alginate, Pectine, Tragacanth, Karayagummi, Xanthangummi, Carrageenin, Agar und Akazie, Stärkederivate, wie Stärkeacetat und Hydroxypropylstärke, und auch andere Syntheseprodukte, wie Polyvinylalkohol, Polyvinylpyrrolidon, Polyvinylmethylether, Polyethylenoxid, vorzugsweise vernetzte Polyacrylsäure, wie neutrales Carbopol, oder Gemische dieser Polymere. Bevorzugte Träger sind Wasser, Cellulosederivate, wie Methylcellulose, Alkalimetallsalze von Carboxmethylcellulose, Hydroxymethylcellulose, Hydroxyethylcellulose, Methylhydroxypropylcellulose und Hydroxypropylcellulose, neutrales Carbopol oder Gemische hiervon. Ein hoch bevorzugter Träger ist Wasser. Die Konzentration des Trägers macht typischerweise das 1- bis 100.000-fache der Konzentration des Wirkstoffs aus. Unter der Angabe wässrig wird typischerweise eine wässrige Zusammensetzung verstanden, in welcher der Träger in einer Menge von > 50 Gew.-%, bevorzugter von > 75 Gew.-% und insbesondere von > 90 Gew.-% Wasser vorhanden ist.
- Innerhalb der vorliegenden Erfindung bezieht sich die Angabe Zusammensetzung insbesondere auf eine Lösung, eine Suspension, ein Gel, eine Salbe, eine Emulsion und/oder ein Gemisch hiervon.
- Weiter sind pharmazeutische Zusammensetzungen bevorzugt, die für eine Verabreichung an das Auge geeignet sind. Eine solche pharmazeutische Zusammensetzung enthält daher vorzugsweise auch weitere Bestandteile, welche für eine Verträglichkeit durch die Augen sorgen. Bei einem besonderen Aspekt bezieht sich die vorliegende Erfindung daher auf die Stabilisierung einer ophthalmischen Zusammensetzung und insbesondere einer wässrigen ophthalmischen Zusammensetzung.
- Diese weiteren Zusätze können Tonizitätsverbesserungsmittel, Konservierungsmittel und Mittel zur Einstellung des pH Werts enthalten.
- Zur Einstellung des pH Werts, vorzugsweise auf einen physiologischen pH, eignen sich vor allem Puffer. Beispiele für Puffersubstanzen sind Acetat, Ascorbat, Borat, Hydrogencarbonat/Carbonat, Citrat, Gluconat, Lactat, Phosphat, Propionat und TRIS (Trimethamin). Trimethamin und Boratpuffer sind bevorzugte Puffer. Die Menge an zuzusetzender Puffersubstanz entspricht typischerweise derjenigen Menge, die zur Sicherstellung und Aufrechterhaltung eines physiologisch verträglichen pH Bereichs erforderlich ist. Der pH Bereich liegt im allgemeinen in einem Bereich von 4 bis 9, vorzugsweise von 4,5 bis 8,5, und bevorzugter von 5,0 bis 8,2.
- Die Tonizität wird erforderlichenfalls typischerweise durch Mittel zur Erhöhung der Tonizität eingestellt. Solche Mittel können beispielsweise ionisch und/oder nicht ionisch sein. Beispiele für ionische Tonizitätsverbesserungsmittel sind Alkalimetall- oder Erdalkalimetallhalogenide, wie CaCl2, KBr, KCl, LiCl, NaI, NaBr oder NaCl, Na2SO4 oder Borsäure. Nicht ionische Tonizitätsverbesserungsmittel sind beispielsweise Harnstoff, Glycerin, Sorbit, Mannit, Propylenglycol oder Dextrose. Typischerweise kann eine derartige Menge eines Tonizitätsverbesserungsmittels zugegeben werden, das sich für die obige ophthalmische Zusammensetzung eine Osmolalität von etwa 50 bis 1.000 mOsmol, vorzugsweise von 100 bis 400 mOsmol, bevorzugter von 200 bis 400 mOsmol, und noch stärker bevorzugt von 250 bis 350 mOsmol, ergibt.
- Das Konservierungsmittel kann typischerweise ausgewählt werden aus quaternären Ammoniumverbindungen, wie Benzalkoniumchlorid, Benzoxoniumchlorid und dergleichen. Benzalkoniumchlorid wird besser beschrieben als N-Benzyl-N-(C8-C18-alkyl)-N,N-dimethylammoniumchlorid. Beispiele für von quaternären Ammoniumsalzen unterschiedliche Konservierungsmittel sind Alkylquecksilbersalze von Thiosalicylsäure, wie Thiomersal, Phenylmercurinitrat, Phenylmercuriacetat oder Phenylmercuriborat, Parabene, wie Methylparaben oder Propylparaben, Alkohole, wie Chlorbutanol, Benzylalkohol oder Phenylethanol, Guanidinderivate, wie Chlorhexidin oder Polyhexamethylenbiguanid, Natriumperborat, Germal®II oder Sorbinsäure. Bevorzugte Konservierungsmittel sind quaternäre Ammoniumverbindungen, insbesondere Benzalkoniumchlorid, Alkylquecksilbersalze und Parabene. Geeignetenfalls wird die ophthalmische Zusammensetzung mit einer solchen Menge eines Konservierungsmittels versetzt, dass ein Schutz gegen Sekundärkontaminationen während der Benutzung sichergestellt ist, die durch Bakterien und Fungi verursacht werden.
- Eine pharmazeutische Zusammensetzung kann zusätzlich auch die Gegenwart eines Lösungsvermittlers erfordern, insbesondere dann, wenn die Wirkstoffkomponenten oder die inaktiven Komponenten zur Bildung einer Suspension oder einer Emulsion neigen. Ein für eine obige Zusammensetzung geeigneter Lösungsvermittler wird beispielsweise aus der Gruppe ausgewählt, die besteht aus Tyloxapol, Fettsäureglycerinpolyethylenglycolestern, Fettsäurepolyethylenglycolestern, Polyethylenglycolen, Glycerinethern, einem Cyclodextrin (beispielsweise α-, β- oder γ-Cyclodextrin, beispielsweise alkylierten, hydroxyalkylierten, carboxyalkylierten oder alkyloxycarbonylalkylierten Derivaten oder Mono- oder Diglycosyl-α-, -β- oder -γ-cyclodextrin, Mono- oder Dimaltosyl-α-, -β- oder -γ-cyclodextrin oder Panosylcyclodextrin), Polysorbat 20, Polysorbat 80 oder Gemischen dieser Verbindungen. Ein spezielles Beispiel für einen besonders bevorzugten Lösungsvermittler ist ein Reaktionsprodukt aus Ricinusöl und Ethylenoxid, wie beispielsweise die Handelsprodukte Cremophor EL® oder Cremophor RH 40®. Reaktionsprodukte aus Ricinusöl und Ethylenoxid haben sich als besonders gute Lösungsvermittler erwiesen, da sie vom Auge äußerst gut vertragen werden. Ein weiterer bevorzugter Lösungsvermittler ist ausgewählt aus Tyloxapol und einem Cyclodextrin. Die zu verwendende Konzentration des Lösungsvermittlers ist insbesondere von der Konzentration des Wirkstoffs abhängig. Die zuzusetzende Menge reicht typischerweise zur Solubilisierung des Wirkstoffs aus. Beispielsweise beträgt die Konzentration des Lösungsvermittlers das 0,1- bis 5.000-fache der Konzentration des Wirkstoffs.
- Eine obige pharmazeutische Zusammensetzung kann auch nicht toxische Hilfsstoffe enthalten, beispielsweise Emulgiermittel, Netzmittel oder Füllstoffe, wie die Polyethylenglycole mit den Bezeichnungen 200, 300, 400 und 600, oder die Carbowaxe mit den Bezeichnungen 1000, 1500, 4000, 6000 und 10000. Sonstige Hilfsstoffe, die gewünschtenfalls verwendet werden können, werden im folgenden aufgelistet, ohne dass hierdurch der Umfang der möglichen Hilfsstoffe irgendwie beschränkt werden soll. Menge und Art des zuzusetzenden Hilfsstoffes entsprechen den besonderen Bedürfnissen, wobei der Hilfsstoff im allgemeinen in einem Mengenbereich von etwa 0,0001 bis etwa 90 Gew.-% verwendet wird.
- Zu bevorzugten Ausführungsformen für das erfindungsgemäße Verfahren zur Stabilisierung einer pharmazeutischen Zusammensetzung, wie sie oben beschrieben worden ist, gehören:
- – Ein Verfahren, worin die pharmazeutische Zusammensetzung eine wässrige Zusammensetzung ist,
- – ein Verfahren, worin die pharmazeutische Zusammensetzung eine ophthalmische Zusammensetzung ist,
- – ein Verfahren, worin die Zusammensetzung eine Lösung, eine Suspension oder ein Gel und vorzugsweise eine Lösung ist,
- – ein Verfahren, worin das Polymermaterial ausgewählt ist aus Polyethylen, Polypropylen und einem Gemisch hiervon und vorzugsweise Polypropylen,
- – ein Verfahren, worin das Polymermaterial die Form einer Flasche hat,
- – ein Verfahren, worin das Antioxidans ausgewählt ist aus der Gruppe, die besteht aus 2,2',2'',6,6',6''-Hexa(1,1-dimethylethyl)-4,4',4''-[(2,4,6-trimethyl-1,3,5-benzoltriyl)trismethy len]triphenol (Irganox®1330), 1,3,5-Tris[3,5-di(1,1-dimethylethyl)-4-hydroxybenzyl]-1H,3H,5H-1,3,5-triazin-2,4,6-trion, Pentaerythrityltetrakis[3-[3,5-di(1,1-dimethylethyl)-4-hydroxyphenyl]propionat], Octadecyl-3-[3,5-di(1,1-dimethylethyl)-4-hydroxyphenyl]propionat, Tris[2,4-di(1,1-dimethylethyl)-phenyl]phosphit, 2,2'-Di(octadecyloxy)-5,5'-spirobi(1,3,2-dioxaphosphorinan), Dioctadecyldisulfid, Didodecyl-3,3'-thiodipropionat, Dioctadecyl-3,3'-thiodipropionat, Butylhydroxytoluol, Ethylenbis[3,3-di[3-(1,1-dimethylethyl)-4-hydroxyphenyl]butyrat] und Gemischen hiervon, insbesondere, falls das Polymermaterial ausgewählt ist aus Polypropylen und Polyethylen und das Antioxidans ausgewählt ist aus der Gruppe, die besteht aus 2,2',2'',6,6',6''-Hexa(1,1-dimethylethyl)-4,4',4''-[(2,4,6-trimethyl-1,3,5-benzoltriyl)trismethylen]triphenol (Irganox®1330), 1,3,5-Tris[3,5-di(1,1-dimethylethyl)-4-hydroxybenzyl]-1H,3H,5H-1,3,5-triazin-2,4,6-trion, Pentaerythrityltetrakis[3-[3,5-di(1,1-dimethylethyl)-4-hydroxyphenyl]propionat], Octadecyl-3-[3,5-di(1,1-dimethylethyl)-4-hydroxyphenyl]propionat und Tris[2,4-di(1,1-dimethylethyl)phenyl]-phosphit, 2,2'-Di(octadecyloxy)-5,5'-spirobi(1,3,2-dioxaphosphorinan), insbesondere, falls das Polymermaterial Polypropylen ist und das Antioxidans ausgewählt ist aus Butylhydroxytoluol und vorzugsweise aus 2,2',2'',6,6',6''-Hexa(1,1-dimethylethyl)-4,4',4''-[(2,4,6-trimethyl-1,3,5-benzoltriyl)trismethylen]triphenol (Irganox®1330), besonders bevorzugt, falls das Polymermaterial Polypropylen ist und das Antioxidans ausgewählt ist aus 2,2',2'',6,6',6''-Hexa(1,1-dimethylethyl)-4,4',4''-[(2,4,6-trimethyl-1,3,5-benzoltriyl)trismethylen]triphenol (Irganox®1330) und vorzugsweise auch Butylhydroxytoluol und die Zusammensetzung eine ophthalmische Zusammensetzung ist,
- – ein Verfahren, worin die ophthalmische Zusammensetzung ferner einen pharmazeutisch wirksamen Bestandteil enthält, der ausgewählt ist aus der Gruppe, die besteht aus Ketotifen, Retinolsäure, Retinol, Retinolacetat, Retinolpalmitat und pharmazeutisch annehmbaren Salzen hiervon und worin diese ophthalmische Zusammensetzung vorzugsweise auch einen Puffer, ein Mittel zur Verbesserung der Tonizität und insbesondere ein Konservierungsmittel enthält. Besonders bevorzugt ist die Verwendung des Antioxidationsmittels zur Stabilisierung einer erfindungsgemäßen pharmazeutischen Zusammensetzung, falls diese Zusammensetzung einen pharmazeutischen Wirkstoff enthält, der aus der Gruppe ausgewählt ist, welche besteht aus Ketotifen, Retinolsäure, Retinol, Retinolacetat, Retinolpalmitat und pharmazeutisch annehmbaren Salzen hiervon.
- Eine weitere bevorzugte Ausführungsform der vorliegenden Erfindung ist eine wässrige ophthalmische Zusammensetzung, die Ketotifen oder ein pharmazeutisch annehmbares Salz hiervon enthält, worin das Antioxidans in ein Polymermaterial eingearbeitet ist, das aus der Gruppe ausgewählt ist, die besteht aus Polyethylen, Polypropylen und einem Gemisch hiervon, insbesondere, falls das Antioxidans aus der Gruppe ausgewählt ist, die besteht aus 2,2',2'',6,6',6''-Hexa(1,1-dimethylethyl)-4,4',4''-[(2,4,6-trimethyl-1,3,5-benzoltriyl)trismethylen]triphenol (Irganox®1330), 1,3,5-Tris[3,5-di(1,1-dimethylethyl)-4-hydroxybenzyl]-1H,3H,5H-1,3,5-triazin-2,4,6-trion, Pentaerythrityltetrakis[3-[3,5-di(1,1-dimethylethyl)-4-hydroxyphenyl]propionat], Octadecyl-3-[3,5-di(1,1-dimethylethyl)-4-hydroxyphenyl]propionat, Tris[2,4-di(1,1-dimethylethyl)phenyl]phosphit, 2,2'-Di(octadecyloxy)-5,5'-spirobi(1,3,2-dioxaphosphorinan), Dioctadecyldisulfid, Didodecyl-3,3'-thiodipropionat, Dioctadecyl-3,3'-thiodipropionat, Butylhydroxytoluol, Ethylen bis[3,3-di[3-(1,1-dimethylethyl)-4-hydroxyphenyl]butyrat] und Gemischen hiervon, und vorzugsweise, worin das Antioxidans Irganox®1330 ist.
- Zu bevorzugten Ausführungsformen der vorliegenden Erfindung, die aus einem ein Antioxidans enthaltenden Polymermaterial hergestellt worden sind, das in das Polymermaterial während des Herstellungsverfahrens dieses Materials eingearbeitet worden ist, gehören diejenigen, bei welchen die pharmazeutische Zusammensetzung eine ophthalmische Zusammensetzung ist, und diejenigen, bei denen das Polymermaterial ausgewählt ist aus Polyethylen, Polypropylen und einem Gemisch hiervon, vorzugsweise aus Polypropylen.
- Zu einem weiteren Ziel der vorliegenden Erfindung gehört die Verwendung einer Flasche aus Kunststoff entsprechend der Beispiele zur Stabilisierung einer darin beschriebenen Zusammensetzung. Beispiel 1: Ketotifen 0,025% Augentropfen Beispiel 2: Ketotifen 0,05% Augentropfen Beispiel 3: Bestandteile in einer 0,025% Ketotifen enthaltenden ophthalmischen Zusammensetzung Beispiel 4: Bestandteile in einer 0,05% Ketotifen enthaltenden ophthalmischen Zusammensetzung Beispiel 5: Vitamin A enthaltende Augentropfen Beispiel 6: Bestandteile in Vitamin A enthaltenden Augentropfen (Gel)

Claims (20)
- Verfahren zur Stabilisierung einer pharmazeutischen Zusammensetzung, was den Zusatz eines Antioxidans erfordern würde, damit diese Zusammensetzung im wesentlichen genauso stabil wie nach diesem Verfahren wäre, gekennzeichnet durch Kontaktierung der Zusammensetzung, die frei an Antioxidans ist, mit einem Polymermaterial, das in der pharmazeutischen Zusammensetzung unlöslich ist und ein Antioxidans enthält, welches in das Polymermaterial während des Herstellungsverfahrens dieses Materials eingearbeitet worden ist, wobei das Polymermaterial in Form eines Gegenstands vorliegt, der aus einem Schlauch, einem Gefäß und einer Flasche ausgewählt ist.
- Verfahren nach Anspruch 1, worin die pharmazeutische Zusammensetzung eine wässrige Zusammensetzung ist.
- Verfahren nach Anspruch 1 oder 2, worin die pharmazeutische Zusammensetzung eine ophthalmische Zusammensetzung ist.
- Verfahren nach einem der Ansprüche 1 bis 3, worin die Zusammensetzung eine Lösung, eine Suspension oder ein Gel und vorzugsweise eine Lösung ist.
- Verfahren nach einem der Ansprüche 1 bis 4, worin das Polymermaterial ausgewählt ist aus Polyethylen, Polypropylen und einem Gemisch hiervon und vorzugsweise Polypropylen.
- Verfahren nach Anspruch 5, worin der Gegenstand eine Flasche ist.
- Verfahren nach Anspruch 1, worin das Antioxidans ausgewählt ist aus der Gruppe, die besteht aus 2,2',2'',6,6',6''-Hexa-(1,1-dimethylethyl)-4,4',4''-[(2,4,6-trimethyl-1,3,5-benzoltriyl)-trismethylen]-triphenol (Irganox®1330), 1,3,5-Tris[3,5-di(1,1-dimethylethyl)-4-hydroxybenzyl]-1H,3H,5H-1,3,5-triazin-2,4,6-trion, Pentaerythrityl-tetrakis[3-[3,5-di(1,1-dimethylethyl)-4-hydroxyphenyl]-propionat], Octadecyl-3-[3,5-di(1,1-dimethylethyl)-4-hydroxyphenyl]-propionat, Tris[2,4-di(1,1-dimethylethyl)-phenyl]-phosphit, 2,2'-Di(octadecyloxy)-5,5'-spirobi(1,3,2-dioxaphosphorinan), Dioctadecyldisulfid, Didodecyl-3,3'-thiodipropionat, Dioctadecyl-3,3'-thiodipropionat, Butylhydroxytoluol, Ethylen-bis[3,3-di[3-(1,1-dimethylethyl)-4-hydroxyphenyl]butyrat] und Gemischen hiervon.
- Verfahren nach Anspruch 1 oder Anspruch 6, worin das Polymermaterial ausgewählt ist aus Polypropylen und Polyethylen und das Antioxidans ausgewählt ist aus der Gruppe, die besteht aus 2,2',2'',6,6',6''-Hexa-(1,1-dimethylethyl)-4,4',4''-[(2,4,6-trimethyl-1,3,5-benzoltriyl)-trismethylen]-triphenol (Irganox®1330), 1,3,5-Tris[3,5-di(1,1-dimethylethyl)-4-hydroxybenzyl]-1H,3H,5H-1,3,5-tnazin-2,4,6-trion, Pentaerythrityl-tetrakis[3-[3,5-di(1,1-dimethylethyl)-4-hydroxyphenyl]-propionat], Octadecyl-3-[3,5-di(1,1-dimethylethyl)-4-hydroxyphenyl]-propionat und Tris[2,4-di(1,1-dimethylethyl)-phenyl]-phosphit, 2,2'-Di(octadecyloxy)-5,5'-spirobi(1,3,2-dioxaphosphorinan).
- Verfahren nach Anspruch 1 oder 6, worin das Polymermaterial Polypropylen ist und das Antioxidans ausgewählt ist aus Butylhydroxytoluol und vorzugsweise aus 2,2',2'',6,6',6''-Hexa-(1,1-dimethylethyl)-4,4',4''-[(2,4,6-trimethyl-1,3,5-benzoltriyl)-trismethylen]-triphenol (Irganox®1330).
- Verfahren nach Anspruch 1, worin die Zusammensetzung eine ophthalmische Zusammensetzung ist, das Polymermaterial Polypropylen ist und das Antioxidans ausgewählt ist aus 2,2',2'',6,6',6''-Hexa-(1,1-dimethylethyl)-4,4',4''-[(2,4,6-trimethyl-1,3,5-benzoltriyl)-trismethylen]-triphenol (Irganox®1330) und vorzugsweise aus Butylhydroxytoluol.
- Verfahren nach Anspruch 10, worin die ophthalmische Zusammensetzung ferner einen pharmazeutischen Wirkstoff enthält, der ausgewählt ist aus der Gruppe, die besteht aus Ketotifen, Retinoesäure, Retinol, Retinolacetat, Retinolpalmitat und pharmazeutisch annehmbaren Salzen hiervon.
- Verfahren nach Anspruch 10, worin die Zusammensetzung ferner einen Puffer, einen Tonizitätsverbesserer und insbesondere ein Konservierungsmittel enthält.
- Verwendung eines Antioxidans, das in ein Polymermaterial während des Herstellungsverfahrens dieses Materials zwecks Stabilisierung einer pharmazeutischen Zusammensetzung eingearbeitet worden ist, die ausgewählt ist aus einer Lösung, einer Suspension, einem Gel, einer Salbe, einer Emulsion oder einem Gemisch hiervon, was den Zusatz eines Antioxidans erfordern würde, damit diese Zusammensetzung im wesentlichen genauso stabil wäre, wie dies durch diese Verwendung verursacht worden wäre, die aber frei davon ist.
- Verwendung nach Anspruch 13, worin diese Zusammensetzung einen pharmazeutischen Wirkstoff enthält, der aus der Gruppe ausgewählt ist, die besteht aus Ketotifen, Retinoesäure, Retinol, Retinolacetat, Retinolpalmitat und pharmazeutisch annehmbaren Salzen hiervon.
- Pharmazeutische Zusammnensetzung, erhalten nach dem Verfahren eines der Ansprüche 11 bis 12.
- Wässrige ophthalmische Zusammensetzung nach Anspruch 15, die Ketotifen oder ein pharmazeutisch annehmbares Salz hiervon enthält, worin das Antioxidans in ein Polymermaterial eingearbeitet worden ist, das ausgewählt ist aus der Gruppe, die besteht aus Polyethylen, Polypropylen und einem Gemisch hiervon.
- Zusammensetzung nach Anspruch 16, worin das Antioxidans ausgewählt ist aus der Gruppe, die besteht aus 2,2',2'',6,6',6''-Hexa-(1,1-dimethylethyl)-4,4',4''-[(2,4,6-trimethyl-1,3,5-benzoltriyl)-trismethylen]-triphenol (Irganox®1330), 1,3,5-Tris[3,5-di(1,1-dimethylethyl)-4-hydroxybenzyl]-1H,3H,5H-1,3,5-triazin-2,4,6-trion, Pentaerythrityl-tetrakis[3-[3,5-di(1,1-dimethylethyl)-4-hydroxyphenyl]-propionat], Octadecyl-3-[3,5-di(1,1-dimethylethyl)-4-hydroxyphenyl]-propionat, Tris[2,4-di(1,1-dimethylethyl)-phenyl]-phosphit, 2,2'-Di(octadecyloxy)-5,5'-spirobi(1,3,2-dioxaphosphorinan), Dioctadecyldisulfid, Didodecyl-3,3'-thiodipropionat, Dioctadecyl-3,3'-thiodipropionat, Butylhydroxytoluol, Ethylen-bis[3,3-di[3-(1,1-dimethylethyl)-4-hydroxyphenyl]butyrat] und Gemischen hiervon, und wobei das Antioxidans vorzugsweise Irganox®1330 ist.
- Gegenstand, ausgewählt aus einem Schlauch, einem Gefäß und einer Flasche und hergestellt aus einem Polymermaterial, das ein Antioxidans enthält, welches in das Polymermaterial während des Herstellungsverfahrens dieses Materials eingearbeitet worden ist, wobei dieser Gegenstand eine pharmazeutische Zusammensetzung enthält, damit diese Zusammensetzung im wesentlichen genauso stabil wäre, wie dies durch diese Verwendung verursacht worden wäre, die aber frei davon ist.
- Gegenstand nach Anspruch 18, worin die pharmazeutische Zusammensetzung eine ophthalmische Zusammensetzung ist.
- Gegenstand nach Anspruch 18 oder 19, worin das Polymermaterial ausgewählt ist aus Polyethylen, Polypropylen und einem Gemisch hiervon und vorzugsweise Polypropylen.
