Description
- Die Erfindung betrifft eine Tablette, die sich im Mundraum schnell zersetzen kann.
- Es gibt verschiedene Arten von oral verabreichbaren Medikamenten, z.B. Tabletten, Kapseln, Granulate, Pulver, Sirups usw. Solche oral verabreichbare Medikamente bringen jedoch einige nachfolgend aufgeführte ernsthafte Probleme mit sich. Das Herunterschlucken von Tabletten und Kapseln kann z.B. für ältere Personen oder Kinder, deren Kraft beim Schlucken gering ist, schwer sein. Granulate und Pulver können nach der Dosierung im Mund zu einem unangenehmen Gefühl führen, oder sie können fälschlicherweise in die Atemwege oder die Lunge gelangen.
- Außerdem können sie nicht ohne Wasser eingenommen werden, da Wasser gewöhnlich für die Dosierung erforderlich ist. Bei Sirups ist das Abmessen beim Dosieren problematisch, und es ist nicht zu erwarten, daß ältere Personen oder Kinder diese exakt abmessen. Andererseits können feste Medikamente, die sich im Mundraum schnell lösen oder zersetzen können, ohne Abmessen oder ohne Wasser eingenommen werden, so daß sie von derartigen älteren Personen oder Kindern leicht eingenommen werden können.
- Es wurden z.B. einige Arten von Medikamenten entwickelt, die sich nach dem Dosieren leicht im Mundraum lösen oder zersetzen können. In der JP-B-62-50445 sind z.B. feste Medikamente offenbart, die unter Anwendung eines Gefriertrocknungsverfahrens aus einer wäßrigen Lösung erzeugt werden können, die hauptsächlich Gelatine enthält, die einen Wirkstoff enthält. In der WO 93/12769 werden auch feste Medikamente offenbart, die durch Trocknen einer Suspension hergestellt werden können, die Agar enthält.
- Die mit dem vorstehend genannten herkömmlichen Verfahren hergestellten Medikamente weisen jedoch keine ausreichende Härte auf, damit sie aus PTP-Packungen (Packungen zum Herausdrücken), die diese Medikamente enthalten, durch ein Herausdrücken entnommen werden können. Sie erfordern eine spezielle pharmazeutische Herstellungstechnologie und auch einen enormen Aufwand bei Anlagen und Ausrüstungen.
- Die JP-A-5-271054 und WO 93/15724 offenbaren Herstellungsverfahren für Tabletten, bei denen aus Saccharid bestehende Tabletten so hergestellt werden, daß ein geeignet mit Wasser ergänztes Saccharidgemisch bei einem geringen Druck gepreßt und dann getrocknet wird, so daß feste Tabletten hergestellt werden.
- Solche Verfahren erfordern jedoch eine spezielle pharmazeutische Herstellungstechnologie und zeigen den Mangel, daß das Pulver, aus dem die Tabletten bestehen, beim Preßverfahren bei feuchten Bedingungen an der Oberfläche der Metallform haften kann. Es kann deshalb problematisch sein, diese Verfahren für die Herstellung in industriellem Umfang zu verwenden.
- Die EP-A-0 553 777 offenbart die Herstellung von Tabletten, die einen Wirkstoff und einen Zucker oder Zuckeralkohol mit einer bevorzugten Partikelgröße von 30 μm bis 50 μm enthalten. Es wird auch die wahlfreie Aufnahme eines Sprengmittels offenbart. Zu dem Sprengmittel gehören Maisstärke, Kartoffelstärke und andere Stärken, Carmellose-Calcium, Carmellose-Natrium und Polyvinylalkohol.
- Die Erfinder der vorliegenden Erfindung haben eine sich schnell im Mund zersetzende Tablette überprüft, die keine spezielle pharmazeutische Herstellungstechnologie erfordert und mit einer normalen Anlage einfach und leicht hergestellt werden kann. Im Ergebnis haben sie festgestellt, daß gepreßte Tabletten, die im wesentlichen aus Zuckeralkohol oder Saccharid, wie D-Manitol oder Lactose, mit einem mittleren Partikeldurchmesser von nicht mehr als 30 μm als wesentlichem Bestandteil, einem Wirkstoff und einem Sprengmittel bestehen, sich innerhalb einer Minute im Mund zersetzen können und eine für praktische Zwecke angemessene Härte aufweisen, obwohl eine solche Tablette lange Zeit als nicht herstellbar angesehen worden ist.
- Folglich betrifft die vorliegende Erfindung eine Tablette, die Zuckeralkohol oder Saccharid mit jeweils einem mittleren Partikeldurchmesser von nicht mehr als 30 μm, einen Wirkstoff und ein Sprengmittel, ausgewählt aus der Gruppe von Crospovidon, Croscarmellose-Natrium und niedrig substituierter Hydroxypropylcellulose, aufweist, wobei der Zuckeralkohol oder das Saccharid in einer Menge von 60 bis 95 Gew.-% der Tablette und das Sprengmittel in einer Menge von 1 bis 10 Gew.-% der Tablette vorliegen.
- Ferner betrifft die vorliegende Erfindung das Verfahren zur Herstellung einer Tablette, das dadurch gekennzeichnet ist, daß ein Gemisch gepreßt wird, das Zuckeralkohol oder Saccharid mit jeweils einem mittleren Partikeldurchmesser von nicht mehr als 30 μm, einen Wirkstoff und ein Sprengmittel, ausgewählt aus der Gruppe von Crospovidon, Croscarmellose-Natrium und niedrig substituierter Hydroxypropylcellulose, aufweist, wobei der Zuckeralkohol oder das Saccharid in einer Menge von 60 bis 95 Gew.-% der Tablette und das Sprengmittel in einer Menge von 1 bis 10 Gew.-% der Tablette vorliegen.
- In der vorliegenden Erfindung kann z.B. D-Manitol, Sorbitol oder dgl., das weitverbreitet für Medikamente und Lebensmittel eingesetzt wird, als Zuckeralkohol verwendet werden, und Lactose und Glucose oder dgl., die ebenfalls weitverbreitet für Medikamente und Lebensmittel eingesetzt werden, können als Saccharid verwendet werden. Es wird mindestens eine Art von Zuckeralkohol oder Saccharid verwendet.
- Als Wirkstoffe werden die folgenden verwendet, es können jedoch auch andere Bestandteile für die orale Verabreichung verwendet werden.

- Bei der vorliegenden Erfindung können als Sprengmittel Crosspovidon, Crosscarmellose-Natrium und niedrig substituierte Hydroxypropylcellulose verwendet werden, die weitverbreitet für Medikamente und Lebensmittel eingesetzt werden. Es wird zumindest eine Art von Sprengmitteln verwendet.
- Nachstehend wird das erfindungsgemäße Verfahren zur Herstellung von Tabletten beschrieben.
- Die erfindungsgemäße Tablette kann erhalten werden, indem nach dem Granulieren eine gemischte pulverförmige Komponente, die Zuckeralkohol oder Saccharid mit einem mittleren Partikeldurchmesser von nicht mehr als 30 μm aufweist, die mit einer Hammermühle oder einer Strahlenmühle oder dgl. gemahlen worden ist, ein Wirkstoff und ein Sprengmittel gepreßt und zu Tabletten verarbeitet werden.
- Andererseits kann die erfindungsgemäße Tablette auch erhalten werden, indem nach dem Granulieren eine gemischte pulverförmige Komponente, die Zuckeralkohol oder Saccharid mit einem mittleren Partikeldurchmesser von nicht mehr als 30 μm aufweist, die mit einer Hammermühle, einer Strahlenmühle oder dgl. gemahlen worden ist, ein Wirkstoff und ein Sprengmittel in Gegenwart eines leicht flüchtigen, sich zersetzenden Adjuvans gepreßt und zu Tabletten verarbeitet wird und das sich zersetzende Adjuvans dann ausgetrieben wird.
- Die Menge des Zuckeralkohols oder Saccharids beträgt 60 bis 95%, vorzugsweise etwa 80 bis 95% pro einer Tablette.
- Die Menge des Wirkstoffs ist in Abhängigkeit von der Art und Dosierungsmenge der Wirkstoffe unterschiedlich, bevorzugt sind jedoch 0,01 bis 30% und stärker bevorzugt 0,01 bis 10% pro einer Tablette.
- Die vorhandene Menge des Sprengmittels beträgt 1 bis 10% pro einer Tablette.
- Das leicht flüchtige, sich zersetzende Adjuvans ist z.B. sublimierender Campher, Urethan, Harnstoff, Ammoniumbicarbonat, Benzoesäure oder dgl., Campher ist jedoch besonders bevorzugt. Die Menge des leicht flüchtigen, sich zersetzenden Adjuvans beträgt vorzugsweise 1 bis 20% und stärker bevorzugt 1 bis 10% pro einer Tablette.
- Es kann vorzugsweise ein Naßgranulierverfahren unter Verwendung von gereinigtem Wasser, Ethanol oder dgl. angewendet werden. Bei diesem Verfahren kann das Granulieren z.B. mit einem allgemeinen Granulator, wie z.B. einem Wirbelbettgranulator, einem Drehrührgranulator oder einem extrudierenden Granulator, durchgeführt werden. Das granulierte Material wird getrocknet und mit einem Gleitmittel gemischt und danach zu der vorher festgelegten Form gepreßt. Ein Bindemittel, ein Säuerungsmittel, ein Schaumbildner, ein süßendes Mittel, ein Geschmacksstoff oder ein Färbemittel können als Zusatz zugegeben werden.
- Als Gleitmittel können z.B. Magnesiumstearat, Stearinsäure, Stearylalkohol, Saccharoseester von Fettsäure, Talcum, schwache wasserfreie Kieselsäure oder dgl. verwendet werden. Das Bindemittel ist z.B. Hydroxypropylcellulose, Polyvinylpyrrolidon, Hydroxypropylmethylcellulose, teilweise verseifte Polyvinylalkohol-Methylcellulose, Pullulan oder dgl. Das Säuerungsmittel ist Citronensäure, Äpfelsäure, Adipinsäure, Ascorbinsäure oder dgl.
- Der Schaumbildner ist Natriumbicarbonat, Natriumcarbonat, Calciumcarbonat oder dgl. Das süßende Mittel ist Aspartam®, Saccharin, Glycyrrhetinsäure oder dgl. Der Geschmacksstoff ist Zitrone, Orange, Kiefer, Minze, Menthol oder dgl. Das Färbemittel ist gelbes Eisensesquioxid, rotes Eisensesquioxid, Teerfarbe oder dgl. Die Menge des Gleitmittels beträgt vorzugsweise 0,01 bis 1% und stärker bevorzugt 0,01 bis 0,5% pro einer Tablette.
- Obwohl das Preßverfahren in der vorliegenden Erfindung nicht eingeschränkt ist, kann stärker bevorzugt eine rotierende Tablettiermaschine, eine hydraulische Preßmaschine oder eine Tablettiermaschine mit einem einzigen Preßstempel verwendet werden, die eine hohe Produktivität haben. Wenn ein leicht flüchtiges, sich zersetzendes Adjuvans verwendet wird, wird die Tablette nach dem Preßverfahren durch Erwärmen getrocknet.
- Die Gleitmittel können beim Granulierverfahren aus dem pulverförmigen Gemisch ausgenommen sein, in diesem Fall können sie vor dem Preßverfahren auf die Oberflächen der Preßstempel und Formen der Tablettiermaschine verteilt werden. Dadurch wird die vorliegende Erfindung effektiver. Der Preßdruck einer rotierenden Tablettiermaschine kann vorzugsweise mehr als 300 kg betragen.
- Die Form der in der vorliegenden Erfindung erhaltenen Tablette kann eine Pillenform oder andere Formen, wie eine Tablette mit einer normalen R-Oberfläche, eine Tablette mit einer mit Zucker überzogenen R-Oberfläche, eine Tablette mit rechteckigen Rändern, eine Tablette mit abgerundeten Rändern oder eine Tablette mit zwei R-Oberflächen oder dgl. sein.
- Außerdem kann die Tablette eine Bruchlinie haben.
- Die vorliegende Erfindung wird anhand der nachstehenden Beispiele und Bezugsbeispiele genau beschrieben.
- Bezugsbeispiel 1
- 1890 g D-Manitol (Towa Kasei Co., Ltd.; mittlerer Partikeldurchmesser etwa 60 μm) und 100 g Crosspovidon (POLYPLASDONE® XL-10, GAF Co., Ltd.) wurden in einen Wirbelbett-Granulationstrockner (Glatt Co., Ltd., WSG-Typ 5) gegeben, gereinigtes Wasser wurde eingesprüht, nach Granulier- und Trocknungsverfahren wurde dann das granulierte Material erhalten. 10 g Magnesiumstearat wurden zugesetzt und mit dem granulierten Material gemischt, und dieses wurde mit einer rotierenden Tablettiermaschine (Kikusui Seiko Co., Ltd., CLEAN PRESS COLLECT TYPE 12) gepreßt und zu Tabletten verarbeitet. Die Bedingungen bei der Tablettenherstellung waren wie folgt: das Tablettengewicht betrug 200 mg, die Metallform war vom flachen Typ mit einem Durchmesser von 8 mm, und der Preßdruck wurde geändert, wie z.B. mit den Werten 150 kg, 300 kg, 450 kg, 600 kg und 800 kg.
- Beispiel 1
- D-Manitol (Towa Kasei Co., Ltd., mittlerer Partikeldurchmesser etwa 60 μm) wurde vorher mit einer Strahlmühle (Japan Pneumatic Co., Ltd., Typ PJM-1-1.5) gemahlen, und es wurde das pulverisierte D-Manitol mit einem mittleren Partikeldurchmesser von 20 μm erhalten. 1890 g des pulverisierten D-Manitols und 100 g Crosspovidon (POLYPLASDONE® XL-10, GAF Co., Ltd.) wurden in einen Wirbelbett-Granulationstrockner (Glatt Co., Ltd., WSG-Typ 5) gegeben, gereinigtes Wasser wurde eingesprüht, und nach Granulier- und Trocknungsverfahren wurde das granulierte Material erhalten. Es wurden 10 g Magnesiumstearat zugesetzt und mit dem granulierten Material gemischt, und dieses wurde mit einer rotierenden Tablettiermaschine (Kikusui Seiko Co., Ltd., CLEAN PRESS COLLECT TYPE 12) gepreßt und zu Tabletten verarbeitet. Die Bedingungen bei der Tablettenherstellung waren die gleichen wie im Bezugsbeispiel 1.
- Bezugsbeispiel 2
- 100 g Domperidon, ein Mittel gegen Erbrechen, 1790 g Lactose (DMV Co., Ltd., mittlerer Partikeldurchmesser etwa 80 μm) und 100 g Crosspovidon (POLYPLASDONE® XL-10, GAF Co., Ltd.) wurden in einen Wirbelbett-Granulationstrockner (Glatt Co., Ltd., WSG-Typ 5) gegeben, es wurde gereinigtes Wasser eingesprüht, und nach Granulier- und Trocknungsverfahren wurde das granulierte Material erhalten. Es wurden 10 g Magnesiumstearat zugesetzt und mit dem granulierten Material gemischt, und dieses wurde mit einer rotierenden Tablettiermaschine (Kikusui Seiko Co., Ltd., CLEAN PRESS COLLECT Typ 12) gepreßt und zu Tabletten verarbeitet. Die Bedingungen bei der Tablettenherstellung waren die gleichen wie im Bezugsbeispiel 1.
- Beispiel 2
- Lactose (DMV Co., Ltd., mittlerer Partikeldurchmesser etwa 80 μm) wurde vorher mit einer Strahlmühle (Japan Pneumatic Co., Ltd., Typ PJM-1-1,5) gemahlen, und es wurde pulverisierte Lactose mit einem mittleren Partikeldurchmesser von 15 μm erhalten. 1790 g der pulverisierten Lactose, 100 g Domperidon und 100 g Crosspovidon (POLYPLASDONE® XL-10, GAF Co., Ltd.) wurden in einen Wirbelbett-Granulationstrockner (Glatt Co., Ltd., WSG-Typ 5) eingeführt, gereinigtes Wasser wurde eingesprüht, und nach Granulier- und Trocknungsverfahren wurde das granulierte Material erhalten. Es wurden 10 g Magnesiumstearat zugesetzt und mit dem granulierten Material gemischt, und dieses wurde mit einer rotierenden Tablettiermaschine (Kikusui Seiko Co., Ltd., CLEAN PRESS COLLECT TYPE 12) gepreßt und zu Tabletten verarbeitet. Die Bedingungen bei der Tablettenherstellung waren die gleichen wie im Bezugsbeispiel 1.
- Beispiel 3
- Das in Beispiel 1 erhaltene granulierte Material wurde bei folgenden Bedingungen zu Tabletten verarbeitet: das Tablettengewicht betrug 200 mg, der Preßdruck lag bei 50 kg/cm2, und etwas Magnesiumstearat wurde auf die Metallform (8 mm Durchmesser, flacher Typ) und die Formen der hydraulischen Preßmaschine (Riken Seiki Co., Ltd., Typ P-1B) aufgebracht.
- Beispiel 4
- 140 g des in Beispiel 1 verwendeten pulverisierten D-Manitols, 10 g Domperidon, 10 g Crosspovidon (POLYPLASDONE XL-10, GAF Co., Ltd.) und 40 g Campher wurden in einem Vinylbeutel gemischt und unter folgenden Bedingungen zu Tabletten verarbeitet: das Tablettengewicht lag bei 200 mg, der Durchmesser der Metallform betrug 9 mm, es wurde eine Tablettiermaschine mit einem einzigen Preßstempel (Okada Seiko Co., Ltd., Preßpulver-Tablettiermaschine Typ N-20E) verwendet, und der Preßdruck betrug 1500 kg/cm2. Die gepreßte Tablette wurde 10 Minuten bei 80°C in einem Vakuumtrockner vakuumgetrocknet.
- Nachfolgend werden die Härte und die Zersetzungszeit der erfindungsgemäßen Tablette anhand eines Versuchsbeispiels beschrieben.
- Versuchsbeispiel
- Es wurden die Härte und die Zersetzungszeit der in den Beispielen 1 und 2 und den Bezugsbeispielen 1 und 2 erhaltenen Tabletten gemessen. Die Härte der Tablette wurde mit einem zerstörenden Festigkeitsmeßgerät für Tabletten (Toyama Sangyo Co., Ltd., Typ TH-203CP) gemessen. Die Zersetzungszeit der Tablette wurde gemessen, indem die Tablette auf ein Drahtgewebe Nr. 10 gelegt wurde, Wasser mit einer Rate von 4 ml/min auf die Tablette tropfte und die Zeit gemessen wurde, bis die Tablette durch das Drahtgewebe fiel. Die Zeit wurde als Zersetzungszeit bestimmt.
- Die Ergebnisse sind in Tabelle 1 zusammengefaßt.
- In den Bezugsbeispielen 1 und 2 war die Tablettenherstellung bis zu einem Preßdruck von 450 kg schwer, und das Formen der Tabletten wurde bei einem Preßdruck von etwa 600 kg möglich. Die Härte der Tablette war jedoch nicht ausreichend. In den Beispielen 1 und 2 konnte bei einem Preßdruck von mehr als 300 kg eine ausreichende Härte der Tablette erreicht werden, und die Zersetzungszeit war sehr kurz. Beim Dosieren der bei einem Preßdruck von 450 kg gemäß Beispiel 1 hergestellten Tablette zersetzte sich die Tablette im Mundraum innerhalb von 10 Sekunden.
- Bei den in den Beispielen 3 und 4 erhaltenen Tabletten wurden ebenfalls die Härte und die Zersetzungszeit in der gleichen Weise gemessen. Die in Beispiel 3 erhaltenen Tabletten hatten eine ausreichende Härte von etwa 6,5 kgf und deren Zersetzungszeit betrug etwa 10 Sekunden. Die Härte der Tablette von Beispiel 4 betrug etwa 4 kgf und deren Zersetzungszeit lag bei etwa 2 Sekunden, was sehr kurz ist.
- Industrielle Anwendbarkeit
- Gemäß der vorliegenden Erfindung kann eine sich im Mundraum schnell zersetzende Tablette bereitgestellt werden.
Claims (10)
- Tablette, die folgendes aufweist: – Zuckeralkohol oder Saccharid mit jeweils einem mittleren Partikeldurchmesser von nicht mehr als 30 μm, – einen Wirkstoff und – ein Sprengmittel, das aus der Gruppe ausgewählt ist, die aus Crospovidon, Croscarmellose-Natrium und niedrig substituierter Hydroxypropylcellulose besteht, wobei der Zuckeralkohol oder das Saccharid in einer Menge von 60 bis 95 Gew.-% der Tablette vorliegt und wobei das Sprengmittel in einer Menge von 1 bis 10 Gew.-% der Tablette vorliegt.
- Tablette nach Anspruch 1, wobei der Zuckeralkohol oder das Saccharid in einer Menge von 80 bis 95 Gew.-% der Tablette vorliegt.
- Tablette nach Anspruch 1 oder 2, wobei der Zuckeralkohol D-Manitol ist.
- Tablette nach Anspruch 1 oder 2, wobei das Saccharid Lactose ist.
- Tablette nach einem der Ansprüche 1 bis 4, wobei der Wirkstoff in einer Menge von 0,01 bis 30 Gew.-% der Tablette vorliegt.
- Verfahren zum Herstellen einer Tablette, gekennzeichnet durch das Pressen eines Gemischs, das folgendes aufweist: – Zuckeralkohol oder Saccharid mit jeweils einem mittleren Partikeldurchmesser von nicht mehr als 30 μm, – einen Wirkstoff und – ein Sprengmittel, das aus der Gruppe ausgewählt ist, die aus Crospovidon, Croscarmellose-Natrium und niedrig substituierter Hydroxypropylcellulose besteht, wobei der Zuckeralkohol oder das Saccharid in einer Menge von 60 bis 95 Gew.-% der Tablette vorliegt und wobei das Sprengmittel in einer Menge von 1 bis 10 Gew.-% der Tablette vorliegt.
- Herstellungsverfahren nach Anspruch 6, wobei der Zuckeralkohol oder das Saccharid in einer Menge von 80 bis 95 Gew.-% der Tablette vorliegt.
- Herstellungsverfahren nach Anspruch 6 oder 7, wobei der Zuckeralkohol D-Manitol ist.
- Herstellungsverfahren nach Anspruch 6 oder 7, wobei das Saccharid Lactose ist.
- Herstellungsverfahren nach einem der Ansprüche 6 bis 9, wobei der Wirkstoff in einer Menge von 0,01 bis 30 Gew.-% der Tablette vorliegt.
