Description
- Hintergrund der Erfindung
- Diese Erfindung betrifft Rapamycin-42-ester mit 2,2-Bis-hydroxymethyl)propionsäure mit Antitumorwirksamkeit.
- Rapamycin ist ein makrocyclisches Trien-Antibiotikum, hergestellt durch Streptomyces Hygroscopicus, von welchem befunden wurde, dass es Antipilzwirksamkeit aufweist, insbesondere gegen Candida albicans, und zwar in vitro, als auch in vivo [C. Vezina et al., J. Antibiot. 28, 721 (1975); S. N. Sehgal et al., J. Antibiot. 28, 727 (1975); H. A. Baker et al., J. Antibiot. 31, 539 (1978);
US-A-3929992 undUS-A-3993749 ]. - Von Rapamycin allein (
US-A-4885171 ) oder in Kombination mit Picibanil (US-A-Patent 4401653 ) ist gezeigt worden, dass es Antitumorwirksamkeit hat. R. Martel et al. [Can. J. Physiol. Pharmacol. 55, 48 (1977)] offenbarten, dass Rapamycin wirksam im experimentellen allergischen Enzephalomyelitismodell ist, einem Modell für Multiple Sklerose; im Adjuvanz-Arthritis-Modell, einem Modell für Rheumatoidarthritis; und wirksam die Bildung von IgE-ähnlichen Antikörpern hemmte. - Die immunsuppressiven Wirkungen von Rapamycin sind in FASEB 3, 3411 (1989) offenbart worden. Von Cyclosporin A und FK-506, anderen makrocyclischen Molekülen, ist ebenfalls gezeigt worden, dass sie als Immunsuppressivmittel wirksam und daher brauchbar beim Verhindern von Transplantatabstoßung sind [FASEB 3, 3411 (1989); FASEB 3, 5256 (1989); R. Y. Calne et al., und Lancet 1183 (1978); und
US-A-Patent 5100899 ]. - Von Rapamycin ist ebenfalls gezeigt worden, dass es brauchbar beim Verhindern oder Behandeln von systemischem Lupus erythematosus [
US-A-5078999 ], pulmonarer Entzündung [US-A-5080899 ], Insulin-abhängiger Diabetes mellitus [Fifth. Int. Conf. Inflamm. Res. Assoc. 121 (Abstract), (1990)], Glattmuskelzellen-Proliferation und intimaler Verdickung nach vaskulärer Verletzung [Morris, R. J. Heart Lung Transplant 11 (pt. 2): 197 (1992)], Erwachsenen T-Zellen-Leukämie/Lymphom [EP-A-525960 EP-A-532862 - Von Mono- und diacylierten Derivaten von Rapamycin (verestert an den 28- und 43-Positionen) ist gezeigt worden, dass sie brauchbare Antipilzmittel sind (
US-A-Patent 4316885 ) und sie wurden verwendet, um wasserlösliche Aminoacyl-Proarzneien von Rapamycin herzustellen (US-A-4650803 ). Kürzlich ist die Nummerierungskonvention für Rapamycin geändert worden; daher wären gemäß der Chemical Abstracts Nomenklatur die oben beschriebenen Ester an den 31- und 42-Positionen. Rapamycin-42-carbonsäureester, welche bei Transplantatabstoßung brauchbar sind, werden inWO-A-9205179 US-A-5233 03 6 offenbart. Die Herstellung von Rapamycin-42-hemisuccinat durch Lipase-vermittelte Hydrolyse von Rapamycin-42-hemisuccinat-benzyl und -methylester wird in Tetrahedron Letters, Vol. 35, No. 7, pp. 1019–1022, 1994 beschrieben. - Beschreibung der Erfindung

- Der Begriff Alkyl mit 1-6 Kohlenstoffatomen schließt sowohl geradkettige, als auch verzweigte Kohlenstoffketten ein.
- Die Verbindung dieser Erfindung kann durch Acylierung von Rapamycin unter Verwendung der geschützten, entsprechenden Dihydroxysäure oder Dialkoxy-carbonsäure, die aktiviert worden ist, gefolgt von Entfernung der Alkoholschutzgruppen hergestellt werden. Mehrere Verfahren zur Carboxylat-Aktivierung sind nach Stand der Technik bekannt, aber die bevorzugten Verfahren nutzen Carbodiimide, gemischte Anhydride oder Säurechloride. Zum Beispiel kann die entsprechende Carbonsäure als gemischtes Anhydrid mit einer Acylierungsgruppe wie 2,4,6-Trichlorbenzoylchlorid aktiviert werden. Behandlung von Rapamycin mit dem gemischten Anhydrid unter leicht basischen Bedingungen sieht die gewünschte Verbindung vor. Alternativ kann die Acylierungsumsetzung mit 1-(3-Dimethylaminopropyl)-3-ethylcarbodiimid-hydrochlorid und Dimethylaminopyridin erreicht werden. Gemische aus 42- und 31, 42-Estern können durch Chromatographie getrennt werden.
- Entsprechend sieht die Erfindung ein Verfahren zum Herstellen von Rapamycin-42-ester mit 2,2-Bis(hydroxymethyl)propionsäure der Formel I vor, welches umfasst:
Acylieren von Rapamycin mit einem Acylierungsmittel, welches ein reaktives Derivat einer Säure der Formel (II) ist:HO-COCCCH3R1R2 (II),
R für Tri-(C1-6-alky)silyl, Tri-(C1-6-alkyl)silylethyl, Triphenylmethyl, Benzyl, Alkoxymethyl mit 2-7 Kohlenstoffatomen, Tri-(C1-6-alkyl)silylethoxymethyl, Chlorethyl oder Tetrahydropyranyl steht;
und nach der Umsetzung, Entfernen aller vorhandenen Schutzgruppen. - Die Umsetzung kann in Gegenwart eines Kopplungsmittels durchgeführt werden, wie einem geeignet substituierten Carbodiimid-Kopplungsmittel. Die oben erwähnte Verbindung dieser Erfindung kann auch durch Acylierung unter Verwendung von reaktiven Derivaten der Säure der Formel II hergestellt werden, wie einem Anhydrid, einem gemischten Anhydrid oder einem Säurehalogenid wie dem Chlorid.
- Die immunsuppressive Wirkung der Verbindung dieser Erfindung wurden in einem pharmakologischen Standardtestverfahren in vitro, um die Hemmung von Lymphozytenproliferation (LAF) zu messen, und zwei pharmakologischen verfahren in vivo gemessen. Das Pinch-Hauttransplantat-Testverfahren misst die immunsuppressive Wirksamkeit der getesteten Verbindung, als auch die Fähigkeit der getesteten Verbindung, Transplantatabstoßung zu hemmen oder zu behandeln. Das pharmakologische Adjuvansarthritis-Standardtestverfahren misst die Fähigkeit der getesteten Verbindung, immunvermittelte Entzündung zu hemmen. Das Adjuvans-arthritis-Testverfahren ist ein pharmakologisches Standardtestverfahren für Rheumatoidarthritis. Die Verfahren für diese pharmakologischen Testverfahren werden unten vorgesehen.
- Das Komitogen-induzierte Thymozytenproliferationsverfahren (LAF) wurde als ein in vitro Maß der immunsuppressiven Wirkungen der Verbindung verwendet. Kurz: Zellen vom Thymus von normalen BALB/c-Mäusen wurden für 72 Stunden mit PHA und IL-1 kultiviert und mit tritiummarkiertem Thymidin während der letzten sechs Stunden gepulst. Die Zellen werden mit und ohne verschiedene Konzentrationen von Rapamycin, Cyclosporin A oder Testverbindung kultiviert. Die Zellen werden geerntet und die eingeschlossene Radioaktivität wird bestimmt. Die Hemmung von Lymphproliferation wird als prozentuale Veränderung in Zählungen pro Minute gegenüber nicht mit Arzneimittel behandelten Kontrollen bewertet. Für jede bewertete Verbindung wurde Rapamycin ebenfalls zu Vergleichszwecken bewertet. Ein IC50 wurde für Testverbindungen erhalten, als auch für Rapamycin. Wenn es als Vergleich für die repräsentativen Verbindungen dieser Erfindung bewertet wurde, hatte Rapamycin einen IC50 im Bereich von 0,6–1,5 nM. Die erhaltenen Ergebnisse werden als IC50 und als prozentuale Hemmung von T-Zellen-Proliferation bei 0,1 μM vorgesehen. Die für die Verbindung dieser Erfindung erhaltenen Ergebnisse wurden ebenfalls als Quotient im Vergleich mit Rapamycin ausgedrückt. Ein positiver Quotient zeigt immunsuppressive Wirksamkeit an. Ein Quotient größer als 1 zeigt an, dass die Testverbindung Thymozytenproliferation in größerem Ausmaß als Rapamycin hemmte. Die Berechnung für den Quotienten wird unten gezeigt:
- Die Verbindung dieser Erfindung wurde ebenfalls in einem in vivo Testverfahren bewertet, welches konstruiert war, die Überlebenszeit von Pinch-Hauttransplantat von männlichen BALB/c Spendern, transplantiert auf männliche C3H(H-2K) Empfänger zu bestimmen. Das Verfahren wird von Billingham R. E. und Medawar P. B., J. Exp. Biol. 28: 385–402 (1951) angepasst. Kurz: ein Pinch-Hauttransplantat vom Spender wurde auf den Rücken des Empfängers als Allograft transplantiert und ein Isograft wurde als Kontrolle in der gleichen Region verwendet. Die Empfänger wurden mit entweder variierenden Konzentrationen von Testverbindungen intraperitoneal oder oral behandelt. Rapamycin wurde als Testkontrolle verwendet. Unbehandelte Empfänger dienen als Abstoßungskontrolle. Das Transplantat wurde täglich überwacht und die Beobachtungen wurden aufgezeichnet, bis das Transplantat trocken wurde und einen geschwärzten Schorf bildete. Dies wurde als Abstoßungstag angesehen. Die mittlere Transplantat-Überlebenszeit (Anzahl von Tagen ± S. D.) der mit Arzneimittel behandelten Gruppe wurde mit der Kontrollgruppe verglichen. Die folgende Tabelle zeigt die Ergebnisse, die erhalten wurden. Die Ergebnisse werden als mittlere Überlebenszeit in Tagen ausgedrückt. Unbehandelte(Kontroll-)Pinch-Hauttransplantate werden üblicherweise innerhalb von 6–7 Tagen abgestoßen. Die Verbindungen wurden unter Verwendung einer Dosis von 4 mg/kg getestet.
- Das pharmakologische Adjuvansarthritis-Standardtestverfahren misst die Fähigkeit von Testverbindungen, immunvermittelte Entzündung zu verhindern und Rheumatoidarthritis zu hemmen oder zu behandeln. Das Folgende beschreibt kurz das verwendete Testverfahren. Eine Gruppe Ratten (männliche Inzucht Wistar Lewis Ratten) wurde mit der zu testenden Verbindung (1 Std. vor Antigen) vorbehandelt und dann mit Freud's Complete Adjuvant (FCA) in die rechte Hinterpfote injiziert, um Arthritis auszulösen. Die Ratten werden dann nach einem Montag, Mittwoch, Freitag Plan von Tag 0–14 für insgesamt 7 Dosen oral dosiert. Beide Hinterpfoten werden an Tagen 16, 23 und 30 gemessen. Die Differenz im Pfotenvolumen (ml) von Tag 16 gegenüber Tag 0 wird bestimmt und eine prozentuale Veränderung gegenüber Kontrolle wird erhalten. Die Entzündung der linken Hinterpfote (nicht injizierte Pfote) wird durch T-Zellen vermittelte Entzündung verursacht und wird in der Tabelle oben aufgezeichnet (% Veränderung gegenüber Kontrolle). Die Entzündung der rechten Hinterpfote wird andererseits durch nicht spezifische Entzündung verursacht. Verbindungen wurden bei einer Dosis von 5 mg/kg getestet. Die Ergebnisse werden als prozentuale Veränderung in der nicht injizierten Pfote am Tag 16 gegenüber Kontrolle ausgedrückt; je negativer die prozentuale Veränderung, desto wirksamer ist die Verbindung. Rapamycin sah zwischen –70% und –90% Veränderung gegenüber Kontrolle vor, was darauf hinwies, dass mit Rapamycin behandelte Ratten zwischen 70–90% weniger immunvermittelte Entzündung hatten als Kontrollratten.
- Die in diesen pharmakologischen Standardtestverfahren erhaltenen Ergebnisse werden nach dem Verfahren zum Herstellen der spezifischen Verbindung vorgesehen, die getestet wurde.
- Die Ergebnisse dieser pharmakologischen Standardtestverfahren zeigen immunsuppressive Wirksamkeit für die Verbindung dieser Erfindung sowohl in vitro, als auch in vivo. Die im LAF-Testverfahren erhaltenen Ergebnisse weisen auf Suppression von T-Zellen-Proliferation hin und zeigen dadurch die immunsuppressive Wirksamkeit der Verbindungen dieser Erfindung. Weitere Demonstration der Brauchbarkeit der Verbindung dieser Erfindung als Immunsuppressivmittel wurde durch die in den pharmakologischen Hauttransplantat- und Adjuvansarthritis-Standardtestverfahren erhaltenen Ergebnisse gezeigt. Zusätzlich zeigen die in dem Hauttransplantat-Testverfahren erhaltenen Ergebnisse ferner die Fähigkeit der Verbindung dieser Erfindung Transplantationsabstoßung zu behandeln oder zu hemmen. Die in dem pharmakologischen Adjuvansarthritis-Standardtestverfahren erhaltenen Ergebnisse zeigen ferner die Fähigkeit der Verbindung dieser Erfindung, Rheumatoidarthritis zu behandeln oder zu hemmen.
- Basierend auf den Ergebnissen dieser pharmakologischen Standardtestverfahren ist die Verbindung dieser Erfindung brauchbar bei der Behandlung oder Hemmung von Transplantationsabstoßung wie Nieren-, Herz-, Leber-, Lungen-, Knochenmark-, Bauchspeicheldrüsen-(Inselzellen), Cornea-, Dünndarm- und Hautallografts und Herzklappen-Xenografts; bei der Behandlung oder Hemmung von Autoimmunerkrankungen wie Lupus, Rheumatoidarthritis, Diabetes mellitus, Myasthenia gravis und Multipler Sklerose; und Entzündungserkrankungen wie Psoriasis, Dermatitis, Ekzem, Seborrhöe, entzündlicher Darmerkrankung, pulmonarer Entzündung (einschließlich Asthma, chronischer obstruktiver pulmonarer Erkrankung, Emphysem, akutem Atemnotsyndrom, Bronchitis und ähnlichem) und Augen-Uveitis.
- Aufgrund des erhaltenen Wirksamkeitsprofils wird von der Verbindung dieser Erfindung ebenfalls angenommen, dass sie Antitumor-, Antipilzwirksamkeiten und antiproliferative Wirksamkeiten haben. Die Verbindung dieser Erfindung ist daher ebenfalls brauchbar beim Behandeln von festen Tumoren, Erwachsenen T-Zellen Leukämie/Lymphom, Pilzinfektionen und hyperproliferativen vaskulären Erkrankungen wie Restenose und Atherosklerose. Wenn sie für Restenose verwendet wird, wird es bevorzugt, dass die Verbindung dieser Erfindung verwendet wird, um Restenose zu behandeln, die nach angioplastischen Verfahren auftritt. Wenn sie für diesen Zweck verwendet wird, kann die Verbindung dieser Erfindung vor dem Verfahren, während dem Verfahren, nach dem Verfahren oder jeder Kombination des Obigen verabreicht werden.
- Wenn sie für die Behandlung oder Hemmung der obigen Krankheitszustände verabreicht wird, kann die Verbindung dieser Erfindung einem Säuger oral, parenteral, intranasal, intrabronchial, transdermal, topisch, intravaginal oder rektal verabreicht werden.
- Es wird in Erwägung gezogen, dass, wenn die Verbindung dieser Erfindung als Immunsuppressiv- oder Antientzündungsmittel verwendet wird, sie zusammen mit einem oder mehreren anderen immunregulativen Wirkstoffen verabreicht werden kann. Solche anderen immunregulativen Wirkstoffe schließen ein, aber sind nicht begrenzt auf Azathioprin, Corticosteroide wie Prednison und Methylprednisolon, Cyclophosphamid, Rapamycin, Cyclosporin A, FK-506, OKT-3 und ATG. Durch Kombinieren der Verbindung dieser Erfindung mit solchen anderen Arzneimitteln oder Wirkstoffen zum Herbeiführen von Immunsuppression oder Behandeln entzündlicher Zustände werden geringere Mengen von jedem dieser Wirkstoffe benötigt, um die gewünschte Wirkung zu erreichen. Die Basis für solch eine Kombinationstherapie wurde durch Stepkowski begründet, dessen Ergebnisse zeigten, dass die Verwendung einer Kombination von Rapamycin und Cyclosporin A bei subtherapeutischen Dosen die Herz-Allograft-Überlebenszeit deutlich verlängerte. [Transplantation Proc. 23: 507 (1991)].
- Die Verbindung dieser Erfindung kann allein oder mit einem pharmazeutischen Träger an einen Säuger formuliert werden, welcher sie benötigt. Der pharmazeutische Träger kann fest oder flüssig sein. Bei oraler Formulierung ist herausgefunden worden, dass 0,01% Tween 80 in PHOSAL PG-50 (Phospholipid-Konzentrat mit 1,2-Propylenglycol, A. Nattermann & Cie. GmbH) eine annehmbare orale Formulierung vorsieht.
- Ein fester Träger kann eine oder mehrere Substanzen einschließen, welche auch als Geschmackmittel, Schmiermittel, Aufschlussmittel, Suspensionsmittel, Füllstoffe, Gleitmittel, Kompressionshilfen, Bindemittel oder Tablettenaufschlussmittel fungieren können; es kann auch ein Einkapselungsmaterial sein. In Pulvern ist der Träger ein fein geteilter Feststoff, welcher sich in Vermischung mit dem fein geteilten Wirkstoff befindet. In Tabletten wird der Wirkstoff mit einem Träger mit den notwendigen Kompressionseigenschaften in geeigneten Anteilen gemischt und in die gewünschte Form und Größe gepresst. Die Pulver und Tabletten enthalten vorzugsweise bis zu 99% des Wirkstoffs. Geeignete feste Träger schließen zum Beispiel Calciumphosphat, Magnesiumstearat, Talk, Zucker, Lactose, Dextrin, Stärke, Gelatine, Cellulose, Methylcellulose, Natriumcarboxymethylcellulose, Polyvinylpyrrolidin, niedrig schmelzende Wachse und Ionenaustauschharze ein.
- Flüssige Träger werden beim Herstellen von Lösungen, Suspensionen, Emulsionen, Sirupen, Elixieren und unter Druck stehenden Zusammensetzungen verwendet. Der Wirkstoff kann in einem pharmazeutisch annehmbaren flüssigen Träger wie Wasser, einem organischen Lösungsmittel, einem Gemisch aus beidem oder pharmazeutisch annehmbaren Ölen oder Fetten gelöst oder suspendiert werden. Der flüssige Träger kann andere geeignete pharmazeutische Zusatzstoffe, wie Aufschlussmittel, Emulgatoren, Puffer, Konservierungsstoffe, Süßstoffe, Geschmacksstoffe, Suspensionsmittel, Verdickungsmittel, Farbstoffe, Viskositätsregulatoren, Stabilisatoren oder Osmo-Regulatoren enthalten. Geeignete Beispiele für flüssige Träger zur oralen und parenteralen Verabreichung schließen ein: Wasser (welches teilweise Zusatzstoffe wie oben enthält, z. B. Cellulosederivate, vorzugsweise Natriumcarboxymethylcelluloselösung), Alkohole (einschließlich einwertige Alkohole und mehrwertige Alkohole, z. B. Glycole) und ihre Derivate, Lecithine und Öle (z. B. fraktioniertes Kokosnussöl und Arachisöl). Zur parenteralen Verabreichung kann der Träger auch ein öliger Ester sein, wie Ethyloleat und Isopropylmyristat. Sterile flüssige Träger sind in Zusammensetzungen mit steriler flüssiger Form zur parenteralen Verabreichung brauchbar. Der flüssige Träger für unter Druck stehende Zusammensetzungen kann halogenierter Kohlenwasserstoff oder ein anderes pharmazeutisch annehmbares Treibmittel sein.
- Flüssige pharmazeutische Zusammensetzungen, welche sterile Lösungen oder Suspensionen sind, können zum Beispiel für intramuskuläre, intraperitoneale oder subkutane Injektion verwendet werden. Sterile Lösungen können auch intravenös verabreicht werden. Die Verbindung kann auch oral entweder in flüssiger oder fester Zusammensetzungsform verabreicht werden.
- Die Verbindungen dieser Erfindung können rektal in Form eines herkömmlichen Zäpfchens verabreicht werden. Zur Verabreichung durch intranasale oder intrabronchiale Inhalation oder Insufflation kann die Verbindung dieser Erfindung in eine wässerige oder teilweise wässerige Lösung formuliert werden, welche dann in Form eines Aerosols genutzt werden kann. Die Verbindung dieser Erfindung kann auch transdermal durch die Verwendung eines transdermalen Pflasters verabreicht werden, welches den Wirkstoff und einen Träger enthält, der gegenüber dem Wirkstoff inert ist, nicht toxisch gegenüber der Haut ist und die Abgabe des Wirkstoffs zur systemischen Absorption in den Blutstrom durch die Haut erlaubt. Der Träger kann eine Anzahl an Formen annehmen, wie Cremes und Salben, Pasten, Gels und Okklusionsmittel. Die Cremes und Salben können viskos flüssige oder halbfeste Emulsionen des entweder Öl-in-Wasser oder Wasser-in-Öl Typs sein. Pasten, welche sich aus absorptiven Pulvern, dispergiert in Petroleum oder hydrophilem Petroleum zusammensetzen, welche den Wirkstoff enthalten, können auch geeignet sein. Eine Vielfalt an Okklusionsmitteln kann verwendet werden, um den Wirkstoff in den Blutstrom abzugeben, wie eine halbdurchlässige, ein Reservoir bedeckende Membran, welches den Wirkstoff mit oder ohne Träger enthält, oder eine Matrix, welche den Wirkstoff enthält. Andere Okklusionsmittel sind in der Literatur bekannt.
- Zusätzlich kann die Verbindung dieser Erfindung als Lösung, Creme oder Lotion durch Formulierung mit pharmazeutisch annehmbaren Trägern eingesetzt werden, welche 0,1–5 Prozent, vorzugsweise 2% an Wirkstoff enthalten, was an einen von Pilz befallenen Bereich verabreicht werden kann.
- Die Dosierungsanforderungen variieren mit den speziellen eingesetzten Verbindungen, dem Verabreichungsweg, der Schwere der vorhandenen Symptome und dem speziellen zu behandelnden Subjekt. Basierend auf Ergebnissen, welche in den pharmakologischen Standardtestverfahren erhalten werden, wären geplante tägliche Dosierungen an Wirkstoff 0,1 μg/kg–100 mg/kg, vorzugsweise zwischen 0,001–25 mg/kg und bevorzugterweise zwischen 0,01 und 5 mg/kg. Die Behandlung wird im Allgemeinen mit kleinen Dosierungen begonnen werden, welche geringer sind, als die optimale Dosierung der Verbindung. Danach wird die Dosierung erhöht, bis die optimale Wirkung unter den Umständen erreicht ist; genaue Dosierungen zur oralen, parenteralen, nasalen oder intrabronchialen Verabreichung werden durch den verabreichenden Arzt basierend auf der Erfahrung mit dem einzelnen behandelten Subjekt bestimmt werden. Vorzugsweise befindet sich die pharmazeutische Zusammensetzung in Einheitsdosierungsform, z. B. als Tabletten oder Kapseln. In solch einer Form wird die Zusammensetzung unterteilt in Einheitsdosen, welche passende Mengen des Wirkstoffs enthalten; die Einheitsdosierungsformen können verpackte Zusammensetzungen sein, zum Beispiel verpackte Pulver, Vialen, Ampullen, vorgefüllte Spritzen oder Sachets, welche Flüssigkeiten enthalten. Die Einheitsdosierungsform kann zum Beispiel eine Kapsel oder Tablette selbst sein, oder sie kann die passende Anzahl einer jeden solchen Zusammensetzung in verpackter Form sein.
- Die folgenden Beispiele veranschaulichen die Herstellung und biologischen Wirksamkeiten der Verbindung dieser Erfindung.
- Beispiel 1 ((Zwischenverbindung)
- Rapamycin-42-ester mit 2,2,5-Trimethyl[1,3]dioxan-5-carbonsäure
- Zu einer Lösung aus dem 2,2-Bis(hydroxymethyl)propionsäureisopropylidenketal (1,041 g, 5,98 mmol) (hergestellt gemäß dem Verfahren von Bruice, J. Am. Chem. Soc. 89: 3568 (1967)) und Triethylamin (0,83 ml, 5,98 mmol) in 20 ml wasserfreiem THF bei 0°C unter Stickstoff wurde 2,4,6-Trichlorbenzoylchlorid (0,93 ml, 5,98 mmol) zugegeben und die sich ergebende weiße Suspension wurde 5 Std. bei Raumtemperatur gerührt. Der Niederschlag wurde durch Vakuumfiltration, Spülen des Kolbens und Filterkuchens mit zusätzlichen 10 ml trockenem THF entfernt. Das Filtrat wurde durch Rotationsverdampfung zu einem weißen Feststoff konzentriert. Der Rückstand wurde in 20 ml trockenem Benzol gelöst, dann wurden Rapamycin (5,47 g, 5,98 mmol) und DMAP (0,731 g, 5,98 mmol) zugegeben. Nach Rühren über Nacht bei Raumtemperatur wurde das Gemisch mit EtOAc verdünnt, mit H2O und gesättigtem NaCl (aq) gewaschen, über trockenem MgSO4 getrocknet, filtriert und zu einem gelben Öl eingedampft. Blitzchromatographie (5 x mit 60% EtOAc-Hexan) ergab die Titelverbindung (2,2 g, 34%) als einen weißen Feststoff.

- Rapamycin-42-ester mit 2,2-Bis-(hydroxymethyl)propionsäure
- Eine Lösung aus dem Produkt von Beispiel 1 (2,8 g, 2,65 mmol) in 50 ml THF und 25 ml 1N HC1 wurde bei Raumtemperatur für 4 Stunden gerührt. Das Gemisch wurde mit Wasser verdünnt und drei mal mit EtOAc extrahiert. Die vereinigten organischen Phasen wurden mit gesättigter NaHCO3-Lösung, gesättigter NaCl-Lösung gewaschen, über MgSO4 getrocknet, filtriert und zu einem gelben öligen Feststoff eingedampft. Reinigung durch Blitzchromatographie (3 x mit EtOAc) ergab die Titelverbindung (1,6 g, 59%).

Hauttransplantat-Überlebenszeit: 11,4 ± 1,5 und 12,0 ± 1,1 Tage
Prozentuale Veränderung bei Adjuvansarthritis gegenüber Kontrolle: –88%.

- Verwendung einer Verbindung nach Anspruch 1 zur Herstellung eines Medikaments zur Behandlung eines Tumors bei einem Säugetier.
- Pharmazeutische Zusammensetzung, die eine Verbindung nach Anspruch 1 und einen pharmazeutischen Träger umfasst.
- Verfahren zur Herstellung von Rapamycin-42-ester mit 2,2-Bis-(hydroxymethyl)propionsäure der Struktur nach Anspruch 1, welches umfasst: Acylieren von Rapamycin mit einem Acylierungsmittel, das ein reaktives Derivat einer Säure der Formel (II) ist:
