Description
- Die vorliegende Erfindung betrifft Zwischenprodukte, die in Podophyllotoxin und verwandte Verbindungen, welche als antineoplastische Mittel bekannt sind, überführt werden können. Insbesondere betrifft die Erfindung eine effiziente Totalsynthese von Podophyllotoxin. Außerdem betrifft die Erfindung Verfahren zur Herstellung derartiger Zwischenprodukte und Verfahren zur Umwandlung der Zwischenprodukte in bekannte Zwischenprodukte, welche leicht in Podophyllotoxin und verwandte Verbindungen überführt werden können.
- Podophyllotoxin (I), ein bekanntes aus einigen Spezien von Podophyllum isoliertes Lignanlacton, ist ein wirksames cytotoxisches Mittel. Zahlreiche andere verwandte Verbindungen mit der charakteristischen Aryltetralin-Ringstruktur, welche entweder natürlich vorkommen oder von natürlich vorkommenden Verbindungen abgeleitet sind, sind bekannt. Einige dieser Verbindungen besitzen antineoplastische Aktivität, während andere zur Umwandlung in Verbindungen brauchbar sind, welche eine derartige Aktivität besitzen. Podophyllotoxin ist ein wichtiges Zwischenprodukt zur Herstellung des Antitumormittels Etoposid und dessen Analoga. Podophyllotoxin hat die folgende Struktur:
- Die charakteristischen Merkmale im Molekül von Podophyllotoxin sind (1) die Anwesenheit des C2-C3-trans-Laktons und (2) die cis-Stellung der C1- und C2-Substituenten. Für die Synthese von Etoposid kann die C4-Hydroxygruppe entweder in α- (Podophyllotoxin) oder β- (Epipodophyllotoxin) Orientierung vorliegen, weil in der Glycosylierungsstufe nur das C4-β-Glycosid erhalten wird.
- In J.Org.Chem., 31, 4004-4008 (1966) beschreiben W.J.Gensler und C.D.Gatsonis die Vervollständigung der Totalsynthese von Podophyllotoxin durch Epimerisierung mittels Enolat - Quenchen des O-Tetrahydropyranylderivats von Picropodophyllin. Diese Epimerisierung verläuft jedoch nicht vollständig, so daß die Trennung einer etwa 45:55-Mischung von Podophyllotoxin und Picropodophyllin erforderlich ist.
- Picropodophyllin ist das cis-Lacton-Isomer von Podophyllotoxin und hat folgende Struktur:
- In J.Org.Chem., 46, 2826-2828 (1981) beschreiben A.S. Kende et al. eine verbesserte Totalsynthese von Podophyllotoxin, ausgehend von Piperonal in 12 Stufen mit einer Gesamtausbeute von 4,5%. Die Kende-Synthese erfordert jedoch die Herstellung und anschließende Epimerisierung von Picropodophyllin in ähnlicher Weise wie die oben beschriebene Gensler-Synthese.
- In J.Am.Chem.Soc., 103, 6208-6209 (1981) beschreiben D.Rajapaksa und R.Rodrigo eine neue Synthese von Podophyllo-
- toxin, welche die in der Umwandlung von Picropodophyllin zu Podophyllotoxin liegende thermodynamische Hürde, wie früher in den oben erwähnten Publikationen von Gensler et al. und Kende et al. beschrieben, vermeidet. Diese Synthese erfordert jedoch die Herstellung einer bicyclischen Prekursorverbindung und eine zufriedenstellende Ausbeute kann nur durch Recycling- Verfahren erzielt werden.
- Die erfindungsgemäße Herstellung von Podophyllotoxin vermeidet ebenfalls das Picropodophyllin-Zwischenprodukt und stellt darüber hinaus eine neue und effiziente stereospezifische Synthese, ausgehend vom Gensler-Keton der Formel:
- zur Verfügung.
- Die Erfindung basiert darauf, daß das cis-Gensler- Keton der Formel:
- eine begrenzte Stabilität besitzt. Diese Stabilität erlaubt die Einführung einer Hydroxymethylgruppe am C3 ohne Epimerisierung am C2.
- Kende et al., [J. Amer. Chem. Soc., 99, 7082-7083 (1977) und J. Org. Chem., 46, 2826-2828 (1981)] und Murphy et al [J.C.S. Perkins I, 271-276 (1982)] haben früher die Hydroxymethylierung am C3 unter Verwendung einer Base und Formaldehyd versucht. Es wurde hauptsächlich das Bisalkylierungsprodukt erhalten und die Ausbeute war nicht hoch. Die vorliegende Erfindung umgeht die Bisalkylierung und liefert ein Produkt mit der gewünschten Stereochemie am C2.
- Die erfindungsgemäße Synthese von Podophyllotoxin und verwandten Verbindungen ist in Schema I zusammengestellt: SCHEMA I SCHEMA I (Fortsetzung) :
- worin R¹ und R² jeweils unabhängig voneinander ein Wasserstoffatom oder Niedrigalkoxy bedeuten oder R¹ und r² zusammen für Methylendioxy stehen; R ein Wasserstoffatom oder eine Carboxylschutzgruppe bedeutet; R&sup4; und R&sup6; jeweils unabhängig voneinander ein Wasserstoffatom oder Niedrigalkoxy bedeuten; R&sup5; für ein Wasserstoffatom oder eine Phenolschutzgruppe steht; R&sup7;, R&sup8; und R&sup9; jeweils unabhängig voneinander für Niedrigalkyl oder Phenyl stehen; und R¹&sup0; für ein Wasserstoffatom, Niedrigalkyl, Niedrigalkoxy oder Nitro steht.
- Die Verbindungen V, VI, VII und VIII in Schema I sind neue Zwischenverbindungen.
- Die in der Beschreibung und in den Ansprüchen verwendeten Ausdrücke "Niedrigalkyl" und "Niedrigalkoxy" bedeuten geradkettige oder verzweigte Alkyl- oder Alkoxygruppen mit 1 bis 6 Kohlenstoffatomen, wie Methyl, Ethyl, Propyl, Isopropyl, Butyl, Isobutyl, t-Butyl, Amyl, Hexyl, etc.. Vorzugsweise enthalten diese Gruppen 1 bis 4 Kohlenstoffatome und insbesondere bevorzugt 1 oder 2 Kohlenstoffatome. Wenn nicht anders angegeben, bedeutet der Ausdruck "Halogen" in der Beschreibung und den Ansprüchen Chlor, Fluor, Brom und Jod.
- Carboxylschutzgruppen, die erfindungsgemäß zur Blockierung oder zum Schutz von Carboxylgruppen brauchbar sind, sind dem Fachmann bekannt. Dazu zählen Reste, wie Niedrigalkyl, Phenylniedrigalkyl, ringsubstituiertes Phenylniedrigalkyl, Methoxymethyl, Benzyloxymethyl, Allyl, Diphenylmethyl und dergleichen.
- Phenolschutzgruppen, die erfindungsgemäß zur Blockierung und zum Schutz einer Phenolfunktion brauchbar sind, sind dem Fachmann ebenfalls bekannt. Dazu zählen Reste wie Niedrigalkyl, Phenylniedrigalkyl, ringsubstituiertes Phenylniedrigalkyl, Benzyloxycarbonyl, 2,2,2-Trichlorethoxycarbonyl, Methoxymethyl, Allyl und dergleichen. Weitere geeignete Schutzgruppen sind in "Protective Groups in Organic Synthesis", Theodora W. Greene (John Wiley & Sons, 1981) in Kapitel 3 für Phenolgruppen und Kapitel 5 für Carboxylgruppen beschrieben. Auf diese Publikation wird hiermit Bezug genommen.
- Das Aryltetralon-Ausgangsmaterial, eine Verbindung der Formel III, worin R¹ und R² zusammen für Methylendioxy stehen, R³ ein Wasserstoffatom, CH&sub3; oder C&sub2;H&sub5; bedeutet, R&sup4; und R&sup6; für Methoxy stehen und R&sup5; eine Methylgruppe bedeutet, kann man nach der in J. Am. Chem. Soc., 82, 1714-1727 (1960), W.J. Gensler et al. beschriebenen allgemeinen Methode herstellen. Die Verbindungen der Formel III kann man auch nach einer in J. C. S. Perkin I, 271-276 (1982), W.S.Murphy und S.Wattanasin beschriebenen verbesserten Methode herstellen, wobei R¹ für Methoxy und R² für ein Wasserstoffatom stehen, oder R¹ und R² zusammen für Methylendioxy stehen, R³ ein Wasserstoffatom oder Ethyl bedeutet, R&sup4; und R&sup6; ein Wasserstoffatom bedeuten oder R&sup4; und R&sup6; für Methoxy stehen und R&sup5; für Methyl steht.
- Eine Verbindung der Formel III mit der Estergruppe in trans-Konfiguration wird zu einem cis-Aryltetralon der Formel IV bei niedrigen Temperaturen, d.h. bei etwa -80ºC bis -20ºC und vorzugsweise bei etwa -78ºC, durch Enolquenchen unter Verwendung einer nicht-nukleophilen starken Base, wie Lithiumhexamethyldisilazid, Natriummethylsulfinylmethid, Lithium-1,1,6,6-tetramethylpiperid und Lithiumdiisopropylamid, in einem inerten organischen Lösungsmittel, wie Tetrahydrofuran (THF), Dioxan, Dimethoxymethan, Hexamethylphosphoramid und Tetramethylharnstoff und anschließender Zugabe einer Mineralsäure, beispielsweise Salzsäure, epimerisiert. Dies ist die Reaktion A im Schema I.
- Die Reaktion B im Schema I erläutert die Herstellung des Enolsilylethers, einer Verbindung der Formel V, aus einer Verbindung der Formel IV. Dies erfolgt durch Umsetzung einer Verbindung der Formel IV mit einem Reagens der Formel:
- worin R&sup7;, R&sup8; und R&sup9; die oben angegebenen Bedeutungen besitzen und X ein Halogenatom oder eine Trifluormethansulfonatgruppe bedeutet. Die Reaktion erfolgt bei einer Temperatur von -78ºC bis 25ºC in Anwesenheit einer nicht-nukleophilen Base, wie Triethylamin oder Hexamethyldisilazan. Zur Vermeidung einer Epimerisierung am C2 sollten möglichst milde Bedingungen zur Anwendung kommen. Das Silylreagens XI kann beispielsweise Trimethylsilyltrifluormethansulfonat (TMSOTf), Trimethylsilyljodid, t-Butyl-dimethylsilyljodid oder Triethylsilyljodid sein.
- Die Verbindungen der Formel V können ohne Reinigung oder Isolierung in die Verbindungen der Formel VI (Reaktion C im Schema I) überführt werden. Dies erfolgt durch Umsetzung einer Verbindung der Formel V mit einem Bisbenzyloxymethan der Formel:
- oder einem Benzyloxymethylhalogenid der Formel:
- worin R¹&sup0; die oben angegebenen Bedeutungen besitzt und X ein Halogenatom bedeutet. Diese Reaktion erfolgt in Anwesenheit einer Lewis-Säure, wie TMSOTf, ZnCl&sub2;, ZnBr&sub2; und TiCl&sub4;. Die Reaktion sollte bei einer Temperatur von etwa -50ºC bis 20ºC und vorzugsweise bei etwa 0ºC durchgeführt werden.
- Die Verbindungen der Formel VI werden durch Reduktion der C4-Carbonylgruppe bei einer Temperatur von -20ºC bis 20ºC unter Verwendung eines Reduktionsmittels, wie LiBH&sub4;, NaBH&sub4;, NaBH&sub3;CN, LiBHC&sub2;H&sub5; oder Zn(BH&sub4;)&sub2;, in die Verbindungen der Formel VIII überführt. Die Verbindungen der Formel VII fallen als Nebenprodukt dieser Reaktion an.
- Die Reaktion F im Schema I erläutert die Debenzylierung der Verbindungen der Formel VIII zu den Verbindungen der Formel IX. Die Debenzylierung kann unter üblichen Hydrierungsbedingungen durchgeführt werden, d.h. unter Verwendung von Katalysatoren, wie Pt, Raney-Ni, Rh und Lösungsmitteln, wie THF, Ethanol und Ethylacetat. Diese Reaktion erfolgt bei einer Temperatur von 0ºC bis 50ºC, vorzugsweise etwa 25ºC.
- Die Verbindungen der Formel IX werden durch Behandlung mit einer wäßrigen Base und anschließender Behandlung mit einem Kondensationsmittel, wie Dicyclohexylcarbodiimid, 1-Cyclohexyl-3-(2-morpholinoethyl)-carbodiimid-metho-p- toluolsulfonat, Isopropylchloroformat, etc. in die Verbindungen der Formel X (Reaktion G im Schema I) überführt. Diese Reaktion wird bei einer Temperatur von etwa 0ºC bis etwa 50ºC, insbesondere etwa 25ºC durchgeführt.
- Bei dem erfindungsgemäßen Verfahren erfolgt keine Epimerisierung des C2-Anions, weil es als Lacton gebunden ist und, sobald der Lactonring geöffnet wird, das Carboxylatsalz die Deprotonierung am C2 unterdrückt.
- Bei den folgenden Beispielen 1 bis 6 wurden die Schmelzpunkte mit einem Thomas-Hoover-Kapillarschmelzpunktapparat aufgenommen, sie sind unkorrigiert. Protonen-NMR-Spektren wurden mit einem Bruker WM 360-Spektrometer unter Verwendung von Tetramethylsilan als internem Standard aufgenommen. Die Infrarotspektren wurden mit einem Nicolet 5DX FT IR-Spektrophotometer aufgenommen. Chromatographische Trennungen erfolgten unter Verwendung von Woelm-Silikagel (0,040 - 0,063 mm) zur Flashchromatographie oder 0,5 mm E.Merck-Silikagelplatten (60F-254).
- Eine nBuLi-Lösung in Hexan (1,1M, 16,5 ml, 28 mmol) wurde bei -78ºC zu einer Lösung von Diisopropylamin (3,94 ml, 28 mmol) in 10 ml THF getropft. Nach 20-minütigem Rühren bei -78ºC wurde eine THF-Lösung von IIIa (Verbindung III, worin R¹ - R&sup6; wie oben für die Titelverbindung dieses Beispiels angegeben definiert sind) (3,0 g, 7,0 mmol) zugegeben. Die Lösung wurde 1 h auf -40ºC erwärmt und anschließend mit konzentrierter Salzsäure (5 ml konzentrierte Salzsäure, verdünnt mit 10 ml THF) behandelt. Die Reaktionsmischung wurde mit Eiswasser verdünnt und mit Ethylacetat extrahiert. Trocknen über Na&sub2;SO&sub4; und Entfernen des Lösungsmittels lieferte einen weißen Feststoff. Dieser wurde aus heißem Ethanol umkristallisiert, wobei man 2,11 g (70%) der Titelverbindung erhielt; Schmelzpunkt 146-148ºC: NMR (CDCl&sub3;) δ 1.22 (t, 3H, J= 7Hz), 2.74 (dd, 1H, J = 18.1, 4.2 Hz), 2.93 (dd, 1H, J = 18.0, 13.3 Hz), 3.72 (s, 6H), 3.80 (s, 3H), 4.10 (m, 2H), 4.66 (d, 1H, J = 4.7 Hz), 6.03 (s, 1H), 6.04 (s, 1H), 6.17 (s, 2H), 6.62 (s, 1H), 7.54 (s, 1H); IR (KBr) 1731, 1674, 1590, 1504, 1481, 1255, 1130, 1038, cm&supmin;¹.

- Zu einer Lösung von IVa (856 mg, 2 mmol) in 10 ml trockenem CH&sub2;Cl&sub2; wurden bei 3ºC Triethylamin (304 mg, 3 mmol) und Trimethylsilyltrifluormethansulfonat (TMSOTf, 667 mg, 3 mmol) gegeben. Die Mischung wurde 1 h bei Raumtemperatur gerührt. Die Mischung wurde erneut gekühlt und die gleichen Mengen an Triethylamin und TMSOTf wurden zugegeben. Die Mischung wurde 1 h bei Raumtemperatur gerührt und anschließend mit kalter gesättigter NaHCO&sub3;-Lösung gewaschen. Die organische Schicht wurde über Na&sub2;SO&sub4; getrocknet und verdampft, wobei man die rohe Titelverbindung erhielt. NMR (CDCl&sub3;) δ 0.15 (m, 9H), 1.32 (t, 3H, J = 7.321 Hz), 3,76 (s, 6H), 3.78 (s, 3H), 4.01 (m, 2H), 4.23 (1H, dd, J = 7.439 Hz), 5.17 (d, 1H, J = 2.776 Hz), 5.90 (d, 1H, J = 9.623 Hz), 6.44 (s, 2H), 6.6 (s, 1H), 7.0 (s, 1H).
- A. Das rohe gemäß Beispiel 2 erhaltene Va wurde in 5 ml CH&sub2;Cl&sub2; gelöst und bei -20ºC zu einer Lösung von Bisbenzyloxymethan (912 mg, 5 mmol) in 10 ml CH&sub2;Cl&sub2;, enthaltend 0,1 ml der TMSOTf-Lösung (die TMSOTf-Lösung wurde durch Lösen von 10 g TMSOTf in 10 ml CH&sub2;Cl&sub2; hergestellt) gegeben. Nach 18- stündigem Rühren bei Raumtemperatur wurde die Reaktionsmischung mit gesättigter NaHCO&sub3;-Lösung gewaschen und über Na&sub2;SO&sub4; getrocknet. Entfernen des Lösungsmittels und Chromatographie des Rückstandes an SiO&sub2; (4% Ethylacetat - CH&sub2;Cl&sub2;) lieferte 613 mg (70%, bezogen auf wiedergewonnenes Ausgangsmaterial, 170 mg) der Titelverbindung, Schmelzpunkt: 107-109ºC: NMR (CDCl3) δ 1.19 (t, 3H, J = 7 Hz), 2.99 (dt, 1H, J = 12.2, 1 Hz), 3.63 (dd, 1H, J = 9.2, 3.3 Hz), 3.71 (s, 6H), 3.78 (s, 3H), 3.83 (dd, 1H, J = 12.5, 5.0 Hz), 4.01 (m, 2H), 4.30 (dd, 1H, J = 9.0, 2.3 Hz), 4.36 (d, 1H, J = 12.2 Hz), 4.50 (d, 1H, J = 12.1 Hz), 4.56 (d, 1H, J = 5.1 Hz), 6.01 (s, 1H), 6,02 (s, 1H), 6.10 (s, 2H), 6.59 (s, 1H), 7.29 (m, 5H), 7.55 (s, 1H); IR (KBr) 1733, 1672, 1590, 1560, 1532, 1250, 1130, 1035 cm&supmin;¹.

- B. Zu einer Lösung von IVa (140 mg, 0,33 mmol) in 5 ml CH&sub2;Cl&sub2; wurden bei 0ºC Triethylamin (55 µl, 0,40 mmol) und TMSOTf (80 µl, 0,40 mmol) gegeben. Nach 0,5-stündigem Rühren bei 0ºC wurden die gleichen Mengen an Triethylamin und TMSOTf zugegeben. Die Reaktionsmischung wurde nach weiterem 0,5- stündigen Rühren mit gesättigter NaHCO&sub3;-Lösung gewaschen. Die CH&sub2;Cl&sub2;-Schicht wurde über Na&sub2;SO&sub4; getrocknet und verdampft, wobei man rohes Va erhielt. Dieses Material wurde erneut in 5 ml CH&sub2;Cl&sub2; gelöst und auf -78ºC gekühlt. Nach Zugabe von Benzyloxymethylchlorid (100 mg, 0,8 mmol) wurde TiCl&sub4; (44 µl, 0,4 mmol) zugetropft. Man rührte eine weitere Stunde bei -78ºC und über Nacht bei Raumtemperatur. Die Reaktionsmischung wurde mit CH&sub2;Cl&sub2; verdünnt und mit Kochsalzlösung gewaschen. Der nach dem Trocknen über Na&sub2;SO&sub4; und Verdampfen des Lösungsmittels erhaltene Rückstand wurde an SiO&sub2; (5% EtOAc-CH&sub2;Cl&sub2;) chromatographiert, wobei man 85 mg VIa (74% Ausbeute, bezogen auf wiedergewonnenes Ausgangsmaterial, 50 mg) erhielt. Dieses Material war hinsichtlich NMR- und Massenspektrum identisch mit dem gemäß Teil A dieses Beispiels erhaltener Produkt.
- Zu einer Lösung von Verbindung VIa (400 mg, 0,55 mmol) in 5 ml trockenem THF wurden bei 0ºC 0,6 ml LiBH&sub4;-Lösung (2M in THF) gegeben. Nach Rühren bei Raumtemperatur wurde die Reaktion durch Zugabe von gesättigter NH&sub4;Cl-Lösung gequencht. Das Produkt wurde mit Ethylacetat extrahiert und die Ethylacetatschicht wurde über Na&sub2;SO&sub4; getrocknet. Der nach Verdampfen des Lösungsmittels erhaltene Rückstand wurde an SiO&sub2;- Platten (10% Ethylacetat-CH&sub2;Cl&sub2;) chromatographiert, wobei man 120 mg (43%) der zweiten Titelverbindung, nämlich Neopodophyllotoxinbenzylether erhielt; Schmelzpunkt 194-196ºC. NMR (CDCl&sub3;) δ 2.96 (t, 1H, J = 4.5 Hz), 3.25 (m, 1H), 3.39 (dd, 1H, J = 7.9, 7.5 Hz), 3.55 (dd, 1H, J = 7.4, 7.2 Hz), 3.71 (s, 6H), 3.83 (s, 3H), 4.10 (d, 1H, J = 4.6 Hz), 4.45 (s, 1H), 5.16 (d, 1H, J = 4.8 Hz), 5.96 (s, 2H), 6.21 (s, 2H), 6.42 (s, 1H), 6.71 (s, 1H), 7.29 (m, 5H); IR (KBr) 1775, 1590, 1508, 1485, 1330, 1255, 1125 cm&supmin;¹.

- Eine stärker polare Bande ergab 70 mg (25%) der ersten Titelverbindung (VIIa), NMR (CDCl&sub3;) δ 1,08 (t, 3H, J = 7.1 Hz), 2.75 (m, 1H), 2.99 (dd, 1H, J = 11.9, 5.4 Hz), 3.50 (t, 1H, J = 8.5 Hz), 3.68 - 3.89 (m, 4H), 3.75 (s, 6H), 3.80 (s, 3H), 4.27 (d, 1H, J = 5.5 Hz), 4.49 (d, 1H, J = 12.0 Hz), 4.57 (d, 1H, J = 12.0 Hz), 4.77 (d, 1H, J = 7.7 Hz), 5.90 (s, 1H), 5.91 (s, 1H), 6.23 (s, 2H), 6.38 (s, 1H), 7.07 (s, 1H), 7.33 (m, 5H). Verbindung VIIa kann in Gegenwart einer milden Base in die Verbindung VIIIa überführt werden.
- Neopodophyllotoxin, (IXa), (Verbindung IX, worin R¹ und R² zusammen = Methylendioxy, R&sup4; und R&sup6; = OCH&sub3; und R&sup5; CH&sub3;)
- Eine Lösung der Verbindung VIIIa (80 mg, 0,16 mmol) in 10 ml Ethylacetat, die wenige Tropfen Methanol und 40 mg 10%iges Pd/C enthält, wurde unter 30 psi H&sub2; 3 h bei Raumtemperatur hydriert. Nach Filtrieren durch Celit wurde das Lösungsmittel entfernt, wobei man einen weißen Feststoff erhielt. Dieses Material wurde an SiO&sub2;-Platten (CH&sub2;Cl&sub2;) chromatographiert, wobei 36 mg (55%) der Titelverbindung erhalten wurden, Schmelzpunkt 235 - 237ºC: NMR (CDCl&sub3;) δ 3.02 (t, 1H, J=4.5 Hz), 3.17 (m, 1H), 3.67 (t, 1H, J=4.8 Hz), 3.76 (m, 1H), 3.78 (s, 6H), 3.85 (s, 3H), 4.25 (d, 1H, J=4.7 Hz), 5.18 (d, 1H, J=4.8 Hz), 5.95 (d, 1H, J=1.0 Hz), 5.97 (d, 1H, J=1.0 Hz, 6.27 (s, 2H), 6.49 (s, 1H), 6.74 (s, 1H); IR (KBr) 3425, 1764, 1592, 1508, 1489, 1333, 1255, 1240, 1125, 1037 cm&supmin;¹.

- Neopodophyllotoxin (IXa) (33 mg, 0,08 mmol) wurde in 1 ml THF und 2 ml Wasser gelöst und mit 0,2 ml 1N NaOH-Lösung 2,5 h bei Raumtemperatur behandelt. Die erhaltene Lösung wurde gekühlt und mit 1N HCl-Lösung angesäuert. Sie wurde rasch mit Ethylacetat extrahiert und die Ethylacetatschicht wurde mit Kochsalzlösung gewaschen und über Na&sub2;SO&sub4; getrocknet. Der nach Verdampfen des Lösungsmittels erhaltene Rückstand wurde in 2 ml trockenem THF gelöst und mit Dicyclohexylcarbodiimid (25 mg, 0,12 mmol) 2 h bei Eisbadtemperatur behandelt. Die erhaltene Mischung wurde verdampft und an SiO&sub2;-Platten (20% Ethylacetat-CH&sub2;Cl&sub2;) chromatographiert, wobei 15 mg (45% der Titelverbindung; Schmelzpunkt 2345-236ºC erhalten wurde. NMR (CDCl&sub3;) δ 1.99 (d, 1H, J = 8.1 Hz), 2.77-2.87 (m, 2H), 3.76 (s, 6H), 3.81 (s, 3H), 4.09 (t, 1H, J=9z), 4.59 (d, 1H, J=5.1 Hz), 4.61 (t, 1H, J=6.9 Hz), 4.77 (t, 1H, J=8 Hz), 5.97 (d, 1H, J=1.2 Hz), 5.98 (d, 1H, J=1.2 Hz), 6.37 (s, 2H), 6.51 (s, 1H), 7.11 (s, 1H); IR (KBr) 3420, 1765, 1592, 1508, 1485, 1240, 1130 cm&supmin;¹.

- Verbindungen der Formelworin R¹ und R² jeweils unabhängig voneinander für ein Wasserstoffatom oder (Niedrig)alkoxy stehen, oder R¹ und R² zusammen für Methylendioxy stehen; R³ ein Wasserstoffatom oder eine Carboxyl-Schutzgruppe ist; R&sup4; und R&sup6; jeweils unabhängig voneinander für ein Wasserstoffatom oder (Niedrig)alkoxy stehen; R&sup5; ein Wasserstoffatom oder eine Phenol-Schutzgruppe ist; und R&sup7;, R&sup8; und R&sup9; jeweils unabhängig voneinander Niedrig(alkyl) oder Phenyl darstellen.
- 2. Verbindungen nach Anspruch 1, worin R¹ und R² zusammen für Methylendioxy stehen, R³ für Ethyl steht, R&sup4; und R&sup6; jeweils für Methoxy stehen, R&sup5; für Methyl steht und R&sup7;, R&sup8; und R&sup9; jeweils für Methyl stehen. 3. Verbindungen der Formel

- 6. Verbindungen nach Anspruch 5, worin R¹ und R² zusammen für Methylendioxy stehen, R³ für Ethyl steht, R&sup4; und R&sup6; jeweils Methoxy darstellen, R&sup5; für Methyl und R¹&sup0; für ein Wasserstoffatom steht.
- 7. Verbindungen der Formelworin R¹ und R² jeweils unabhängig voneinander für ein Wasserstoffatom oder (Niedrig)alkoxy stehen, oder R¹ und R² zusammen für Methylendioxy stehen; R&sup4; und R&sup6; jeweils unabhängig voneinander für ein Wasserstoffatom oder (Niedrig)alkoxy stehen; R&sup5; ein Wasserstoffatom oder eine Phenol-Schutzgruppe darstellt; und R¹&sup0; für ein Wasserstoffatom, Niedrig(alkyl), Niedrig(alkoxy) oder Nitro steht.
- 8. Verbindungen nach Anspruch 7, worin R¹ und R² zusammen für Methylendioxy stehen, R&sup4; und R&sup6; jeweils Methoxy darstellen, R&sup5; für Methyl und R¹&sup0; für ein Wasserstoffatom steht.
- 9. Verfahren zur Herstellung von Verbindungen nach Anspruch 3, wobei man eine Verbindung der Formelworin R¹ und R² jeweils unabhängig voneinander für ein Wasserstoffatom oder (Niedrig)alkoxy stehen, oder R¹ und R² zusammen für Methylendioxy stehen; R³ ein Wasserstoffatom oder eine Carboxyl-Schutzgruppe darstellt; R&sup4; und R&sup6; jeweils unabhängig voneinander für ein Wasserstoffatom oder (Niedrig)alkoxy stehen; R&sup5; ein Wasserstoffatom oder eine Phenol-Schutzgruppe darstellt; und R&sup7;, R&sup8; und R&sup9; jeweils unabhängig voneinander für Niedrig(alkyl) oder Phenyl stehen, mit einem Bisbenzyloxymethan der Formeloder einem Benzyloxymethylhalogenid der Formelworin R¹&sup0; für ein Wasserstoffatom, Niedrig(alkyl), Niedrig(alkoxy) oder Nitro und X für Halogen steht, umsetzt.
- 10. Verfahren nach Anspruch 9, worin die Reaktion bei einer Temperatur von etwa -50ºC bis 20ºC durchgeführt wird.
- 11. Verfahren nach Anspruch 9, worin die Reaktion in Gegenwart einer katalytischen Menge einer Lewis Säure durchgeführt wird.
- 12. Verfahren nach Anspruch 9, worin R¹ und R² zusammen für Methylendioxy stehen, R³ für Ethyl steht, R&sup4; und R&sup6; jeweils Methoxy darstellen, R&sup5; für Methyl steht, R&sup7;, R&sup8; und R&sup9; jeweils Methyl darstellen, R¹&sup0; für ein Wasserstoffatom und X für Chlor steht. 13. Verfahren zur Herstellung von Verbindungen nach Anspruch 5, wobei man eine Verbindung der Formel
- worin R¹ und R² jeweils unabhängig voneinander für ein Wasserstoffatom oder (Niedrig)alkoxy stehen, oder R¹ und R² zusammen für Methylendioxy stehen; R³ ein Wasserstoffatom oder eine Carboxyl-Schutzgruppe darstellt; R&sup4; und R&sup6; jeweils unabhängig voneinander für ein Wasserstoffatom oder (Niedrig)alkoxy stehen; R&sup5; ein Wasserstoffatom oder eine Phenol-Schutzgruppe darstellt; und R¹&sup0; für ein Wasserstoffatom, Niedrig(alkyl), Niedrig(alkoxy) oder Nitro steht, reduziert und die Verbindungen nach Anspruch 5 von der Reaktionsmischung abtrennt.
- 14. Verfahren nach Anspruch 13, worin die Reduktion bei einer Temperatur von -20ºC bis 20ºC in Gegenwart eines Reduktionsmittels durchgeführt wird.
- 15. Verfahren nach Anspruch 14, worin das Reduktionsmittel ausgewählt ist unter LiBH&sub4;, NaBH&sub4;, NaBH&sub3;CN, LiBHC&sub2;H&sub5; und Zn(BH&sub4;)2.
- 16. Verfahren nach Anspruch 13, worin R¹ und R² zusammen für Methylendioxy stehen, R³ für Ethyl steht, R&sup4; und R&sup6; jeweils Methoxy darstellen, R&sup5; für Methyl steht und R¹&sup0; für ein Wasserstoffatom steht.
- 17. Verfahren zur Herstellung einer Verbindung nach Anspruch 7, wobei man eine Verbindung der Formelworin R¹ und R² jeweils unabhängig voneinander für ein Wasserstoffatom oder (Niedrig)alkoxy stehen, oder R¹ und R² zusammen für Methylendioxy stehen; R³ ein Wasserstoffatom oder eine Carboxyl-Schutzgruppe darstellt; R&sup4; und R&sup6; jeweils unabhängig voneinander für ein Wasserstoffatom oder (Niedrig)alkoxy stehen; R&sup5; ein Wasserstoffatom oder eine Phenol-Schutzgruppe darstellt; und R¹&sup0; für ein Wasserstoffatom, Niedrig(alkyl), Niedrig(alkoxy) oder Nitro steht, reduziert und die Verbindung nach Anspruch 7 von der Reaktionsmischung abtrennt.
- 18. Verfahren nach Anspruch 17, worin die Reduktion bei einer Temperatur von -20ºC bis 20ºC in Gegenwart eines Reduktionsmittels durchgeführt wird.
- 19. Verfahren nach Anspruch 18, worin das Reduktionsmittel ausgewählt ist unter LiBH&sub4;, NaBH&sub4;, NaBH&sub3;CN, LiBHC&sub2;H&sub5; und Zn(BH&sub4;)&sub2;.
- 20. Verfahren nach Anspruch 17, worin R¹ und R² zusammen für Methylendioxy stehen, R³ für Ethyl steht, R&sup5; und R&sup6; jeweils Methoxy darstellen, R&sup5; für Methyl steht und R¹&sup0; für ein Wasserstoffatom steht.
- 21. Verfahren zur Herstellung von Verbindungen der Formelworin R¹ und R² jeweils unabhängig voneinander für ein Wasserstoffatom oder (Niedrig)alkoxy stehen, oder R¹ und R² zusammen für Methylendioxy stehen; R&sup4; und R&sup6; jeweils unabhängig voneinander für ein Wasserstoffatom oder (Niedrig)alkoxy stehen und R&sup5; ein Wasserstoffatom oder eine Phenol-Schutzgruppe darstellt, wobei man die Verbindungen gemäß Anspruch 7 hydriert.
- 22. Verfahren nach Anspruch 21, worin die Hydrierung in Gegenwart eines Katalysators, ausgewählt unter Pt, Raney-Ni und Rh, durchgeführt wird.
- 23. Verfahren nach Anspruch 22, worin R¹ und R² zusammen für Methylendioxy stehen, R³ für Ethyl steht, R&sup4; und R&sup6; jeweils Methoxy darstellen und R&sup5; für Methyl steht.
- 24. Verfahren zur Herstellung von Podophyllotoxin, wobei man(a) cis-Ethyl-1,2,3,4-tetrahydro-6,7-methylendioxy-4oxo- 1-(3,4,5-trimethoxyphenyl)naphthalin-2-carboxylat mit Hexamethyldisilazan und Trimethylsilyljodid behandelt, wobei man den entsprechenden Enolsilylether erhält; (b) den Enolsilylether mit Bisbenzyloxymethan in Gegenwart einer katalytischen Menge Triethylsilyl-triflat umsetzt, wobei man d,1-[1α, 2α,3β]-Ethyl-1,2,3,4-tetrahydro-3- benzyloxymethyl-4-oxo-1-(3,4,5-trimethoxyphenyl)napthalin- 2-carboxylat erhält;(c) die C4-Carbonylgruppe des d,1-[1α, 2α,3β]-Ethyl- 1,2,3,4-tetrahydro-3-benzyloxymethyl-4-oxo-1-(3,4,5-trimethoxyphenyl)naphthalin-2-carboxylat reduziert, wobei man den Neopodophyllotoxinbenzylether erhält;(d) den Neopodophyllotoxinbenzylether hydriert, wobei man Neopodophyllotoxin erhält; und(e) das Neopodophyllotoxin hydrolysiert und mit einem Kondensationsmittel behandelt, wobei man Podophyllotoxin erhält.
- 25. Verfahren zur Herstellung der Verbindungen der Formel V nach Anspruch 1, wobei man eine Verbindung der Formel IVmit einer Verbindung der Formel XIworin R&sup7;, R&sup8; und R&sup9; jeweils unabhängig voneinander für Niedrig(alkyl) oder Phenyl stehen und X ein Halogenatom oder eine Trifluoromethansulfonatgruppe ist, umsetzt.
- 26. Verfahren nach Anspruch 25, worin die Reaktion bei einer Temperatur von -78ºC bis 25ºC in Gegenwart einer nicht-nucleophilen Base durchgeführt wird.
- 27. Verfahren nach den Ansprüchen 25 oder 26, worin die Verbindung der Formel XI Trimethylsilyljodid, t-Butyl- dimethylsilyljodid oder Triethylsilyljodid ist. my
