R H oder eine für ein Stickstoffatom geeignete Schutzgruppe und
X O, S oder N-R8, worin R8 eine Alkyl-,
Cycloalkyl-, Aryl-, Alkoxycarbonylgruppe oder eine Dialkylaminogruppe ist, bedeuten.
Der Erfindung lag die Aufgabe zugrunde, ein neues Verfahren für die Herstellung optisch aktiven D-(+)-Biotins bereitzustellen, welches die Durchführung einer RacematSpaltung und damit das Verwerfen oder Rückführen des unerwünschten Enantiomeren vermeidet.
Verfahren zur stereospezifischen Synthese von D-(+)-Biotin aus Zuckern geeigneter Konfiguration sind bekannt. So wird in Tetrahedron Letters Nr. 32, S. 2765-2766 (1975), als Ausgangsmaterial D-Hannose, in Agric. Biol. Chem. Nr. 42, S. 465 (1978), D-Glucose und in den
DE-OS 3122 562 und DE-OS 33 20 140 D-Arabinose als chirales Ausgangsmaterial verwendet.
Alle diese Verfahren sind jedoch durch eine hohe Zahl von Syntheseschritten mit folglich geringer Gesamtausbeute gekennzeichnet. Die aufgrund ihrer Zuckernatur meist nicht kristallisierbaren Zwischenstufen werden oft nur in unbefriedigender Reinheit erhalten und erfordern, bedingt durch ihre Polyfunktionalität und der damit verbundenen chemischen Labilität die Einhaltung vergleichsweise enger Reaktionsparameter. Eine Reihe von Zuckern ist auch aus natürlichen Quellen nicht zugänglich, was einen hohen Preis zur Folge hat.
Die Verwendung von L-Cystein, wie aus US-4009172,
US-4130713, US-4337345 und Journal of the American
Chemical Society Nr. 99, S. 7020 (1977) bekannt, vermeidet zwar die Handhabung labiler Zwischenstufen, führt hingegen über insgesamt 18 Reaktionsstufen unter Abtrennung unerwünschter Isomere nur in unbefriedigender Ausbeute zu optisch aktivem D-(+)-Biotin.
Eine sehr elegante Synthese von D-(+)-Biotin aus Cystein bzw. Cystin über ein optisch aktives bicyclisches
Hydantoin-Derivat als Zwischenprodukt wurde von
E. Poetsch und M. Casutt (Chimia, 41, 148 (1987) und EP-A2-0 242 686/EP-A2-0243 734) beschrieben.
Der einzige Nachteil dieser Tolansynthese ist, daß man eine Cyano-Gruppe in ein Zwischenprodukt einführen muß und somit mit hochgiftigem Alkalicyanid oder Trimethylsilylcyanid arbeiten muß.
Die von Corey et al. (Tetrahedron Letters 29, 57
(1988)) beschriebene Totalsynthese von D-(+)-Biotin aus dem Hydantoin von L-Cystin ergibt über viele Syntheseschritte lediglich 12 % Ausbeute an D-(+)-Biotin und ist somit für die technische Durchführung nicht geeignet.
In einem weiteren Verfahren werden in Journal of the American Chemical Society Nr. 105, S. 5946 (1983) und in der EP-OS 0 094 776 substituierte 3H,5H-Imidazo
[1,5-c]-tetrahydrothiazole beschrieben, aus denen nach Racematspaltung optisch aktives Biotin erhalten wird.
Da die vergleichsweise hohe Stufenzahl verbunden mit teilweise mäßigen Ausbeuten und der Notwendigkeit einer optischen Trennung auch diese Ausgangsstoffe als zur Herstellung von D-(+)-Biotin wenig geeignet erscheinen lassen, bestand weiterhin Bedarf an einem geeigneten Verfahren zur einfachen, ökonomischen und stereospezifischen Herstellung von D-(+)-Biotin.
Es wurde nun überraschend gefunden, daß D-(+)-Biotin ohne zusätzliche Racematspaltung aus dem chiralen (4R/S, 5R)-5-Mercaptomethyl-imidazolidin-4-ol der Formel II erhältlich ist. Dieses kann aus den natürlich vorkommenden Aminosäuren L-Cystein oder L-Cystin über ein Cysteinhydantoin der Formel I, hergestellt werden.
Gegenstand der Erfindung ist daher ein Verfahren
zur Herstellung von D-(+)-Biotin, dadurch gekennzeichnet, daß man ein Cysteinhydantoin der Formel I,
wobei R1 H, eine Gruppe R3R4CH- oder SiR5R6R7, worin
R3 und R4 jeweils unabhängig voneinander
H, unsubstituiertes oder substituiertes Alkyl, Cycloalkyl, Aryl,

und R7 jeweils unabhängig voneinander unsubstituiertes oder substituiertes Alkyl, Cycloalkyl, Aryl, Aralkyl oder Heteroaryl bedeuten, R2 H oder eine für ein Stickstoffatom geeignete Schutzgruppe und
X O, S oder N-R8, worin R8 eine Alkyl-,
Cycloalkyl-, Aryl-, Alkoxycarbonylgruppe oder eine Dialkylaminogruppe ist, bedeuten, unter Einwirkung eines komplexen Reduktionsmittels, zu einem Alkohol der Formel II reduziert.

Weiterhin ist ein solches Verfahren, wobei man zur Herstellung des Cysteinhydantoins der Formel I das optisch aktive bicyclische Hydantoin der
Formel VII
worin R2, R3, R4 und X die angegebene Bedeutung besitzen, mit einer Säure und/oder einem Reduktionsmittel spaltet, Gegenstand der Erfindung.
Die bei dem erfindungsgemäßen Verfahren durchlaufenen Zwischenprodukte der Formeln II, IV, IVa und V sind neu. Die neuen Zwischenprodukte sind ebenfalls Gegenstand der Erfindung, nämlich die Verbindungen der Formel II,

worin R1, R2, R9, R10, Z und X die angegebene Bedeutung besitzen, insbesondere worin R1 und R2 Benzyl, Z O und R9 -(CH2)3-COOR, worin R niederes Alkyl ist, bedeuten, sowie die Verbindungen Formel V,
worin R1, R2, R9, Z und X die in angegebene Bedeutung besitzen, insbesondere worin R1 und R2 Benzyl, Z O, und R9 -(CH2)3-COOR, worin R niederes Alkyl ist, bedeuten. Die Herstellung der Ausgangsverbindungen der Formel I erfolgt nach an sich bekannten Methoden, wie sie in der Literatur (z.B. in Standardwerken wie Houben-Weyl, Methoden der Organischen Chemie, Georg-Thieme-Verlag, Stuttgart) beschrieben sind, und zwar unter Reaktionsbedingungen, wie sie für die genannten Umsetzungen bekannt und geeignet sind. Dabei kann man auch von an sich bekannten, hier nicht näher erwähnten Varianten Gebrauch machen.
Sie können z.B. nach E.J. Corey et al. (Tetrahedron Letters 29, 57 (1988)) aus L-Cystin oder aus dem bicyclischen Hydantoin der Formel VII durch Spaltung mit einer Säure und/oder einem Reduktionsmittel erhalten werden. Die bicyclischen Hydantoine der
Formel VII sind bekannt oder können nach bekannten Methoden aus L-Cystein oder L-Serin (EP-A2-0243 734) hergestellt werden.
Zur Spaltung der Bicyclen der Formel VII eignen sich beispielsweise Säuren wie Salzsäure oder Schwefelsäure, die, zweckmäßigerweise in einem Lösungsmittel, z.B. Wasser oder Alkoholen, zu den Hydantoinen der Formel Ia, worin R1 = H, führen.
Als Spaltungsreagenz weiterhin anwendbar sind Schwermetallsalze wie Silbernitrat, Quecksilber(II)-chlorid oder Quercksilber(II)-acetat. Die anschließende Zersetzung der intermediär gebildeten Metallmercaptide erfolgt zweckmäßig mit Schwefelwasserstoff. Für diese Spaltungsreaktion geeignete Lösungsmittel finden sich beispielsweise unter den Alkoholen wie Methanol oder Ethanol, Amiden wir Dimethylformamid oder Ethern wir Tetrahydrofuran.
Mit Reduktionsmitteln können bicyclische Hydantoine der Formel VII zu Zwischenprodukten der Formel I, worin R1 CHR3R4 bedeutet, umgesetzt werden. Geeignete Reduktionsmittel sind beispielsweise Metalle wie Zink. Die Spaltung der Säurederivate durch Reduktion mit
Metallen erfolgt vorzugsweise in saurem Milieu. Hierfür geeignete Säuren sind beispielsweise Mineralsäuren wie Salzsäure oder Schwefelsäure oder organische
Säuren wir Ameisensäure oder Essigsäure. Zweckmäßig werden diese Säuren als Lösungsmittel oder im Gemisch mit anderen Lösungsmitteln wie z.B. Alkoholen verwendet. Die Reaktionstemperatur liegt üblicherweise
zwischen 0 °C und 200 °C, vorzugsweise zwischen 20 °C und 150 °C. Bei diesen Temperaturen sind die Reaktionen in der Regel nach 15 Minuten bis 24 Stunden beendet.
Aus den Cysteinhydantoinen der Formel I lassen sich durch Einwirkung eines komplexen Reduktionsmittels die Alkohole der Formel II erhalten. Bevorzugte kom
plexe Reduktionsmittel sind beispielsweise Borane wie Diboran, Natriumboranat, Lithium-cyano-trihydroborat, Lithium-, Natrium- oder Kaliumtriethylboranat, Metallhydride wie Natriumhydrid oder Aluminiumhydrid, Siliciumhydride wie Triethylsilan, Tributylzinnhydrid und gemischte Hydride wie Lithiumalanat, Natriumalanat, Natrium-bis(2-methoxy-ethoxy)-dihydridoaluminat,
Kaliumboranat oder Lithiumboranat, insbesondere Diisobutylaluminiumhydrid (DIBAH). Die Umsetzung der Oxoverbindungen der Formel I mit den Reduktionsmitteln erfolgt zweckmäßig in einem geeigneten Lösungsmittel bei Temperaturen zwischen etwa -100 °C und +150°, insbesondere zwischen etwa -10° und 0°. Als Lösungsmittel kommen je nach der chemischen Natur des Reduktionsmittel z.B. Wasser, Alkohole wie Methanol, Ethanol, Isopropanol oder Butanol, Ether wie Tetrahydrofuran, Dioxan, Diethylether oder Ethylenglykoldimethylether und Kohlenwasserstoffe wie Pentan, Cyclohexan, Benzol oder Toluol in Betracht. Vor- und nachstehend besitzen R1, R2, R3, R4, R9,
R10, X, Y, Z und n die angegebene Bedeutung, sofern nicht ausdrücklich etwas anderes vermerkt ist.
Der Rest R1 bedeutet vorzugsweise einen Rest der
Formel R3R4CH oder SiR5R6R7. Darin bedeuten die Reste R3 und R4 vorzugsweise H, C1-C4-Alkyl, unsubstituiertes oder durch C1-C3-Alkyl und/oder Alkoxy ein oder mehrfach substituiertes Phenyl oder Benzyl, insbesondere bevorzugt sind gleichzeitig R3 = H und R4 = Phenyl.
Die Reste R5, R4 und R7 bedeuten vorzugsweise C1-C4- Alkyl unsubstituiertes oder durch C1-C3 substituiertes Phenyl, insbesondere bevorzugt sind R5 und R6 gleichzeitig Methyl oder Phenyl und R7 Methyl oder tert.-Butyl. R2 bedeutet vorzugsweise eine Schutzgruppe für das mit ihr verknüpfte Stickstoffatom, die es ermöglicht, die Verbindung der Formel I in ein D-(+)-Biotin-Derivat, dessen Stickstoffatom mit R2 geschützt ist, mit dem erfindungsgemäßen Verfahren überzuführen, und anschließend diese Schutzgruppe unter milden Bedingungen selektiv abzuspalten.
Solche Schutzgruppen sind dem Fachmann geläufig
(z.B. Protective Groups in Organic Chemistry, Plenum Press, New York, 1973) und können nach bekannten
Methoden eingeführt und abgespalten werden. Bevorzugte Reste R2 sind beispielsweise Benzyl, Methoxybenzyl, 3,4-Dimethoxybenzyl, 4-Methylbenzyl, Allyl,
Methallyl, Crotyl, Trimethylsilyl oder tert.-Butyldimethylsilyl, Diphenylmethyl, Trityl, 9-H-Fluorenyl,
9-Phenyl-9-Fluorenyl, Methoxymethyl.
X bedeutet O oder S, vorzugsweise O.
Der Alkohol der Formel II wird in Gegenwart einer Base in einem inerten Lösungsmittel mit einem Elektrophil der Formel III umgesetzt, wobei man unter Alkylierung der Sulfhydryl-Gruppe das Alkylierungsprodukt der Formel IV erhält. Im Alkylierungsmittel der Formel III bedeutet Y eine Fluchtgruppe, vorzugsweise ein Halogen wie Cl, Br, oder J oder einen Sulfonsäurerest -O-SO2-R", worin R" vorzugsweise p-Toluyl, Methyl oder Trifluormethyl bedeutet. Insbesondere bevorzugt bedeutet Y Cl oder Brom. In den Ver
bindungen der Formeln III, IV, IVa und V bedeutet Z bevorzugt O oder N-R', worin R' eine Alkylgruppe oder eine Dialkylaminogruppe bedeutet. Falls Z N-R' bedeutet, erhält man durch Hydrolyse der entsprechenden Imine oder Hydrazone in an sich bekannter Weise die entsprechenden Ketone (Z = O).
In den Verbindungen der Formeln III, IV, IVa, V und VI bedeutet R9 vorzugsweise Alkyl oder Alkenyl mit bis zu 8 C-Atomen, -(CH2)n-CN, -(CH2)n-COOR oder
-(CH2)n-OR worin R niederes Alkyl bedeutet und n 2, 3, 4, 5, 6, 7 oder 8 ist. Vorzugsweise stellt R9
einen Rest dar, der sich in an sich bekannter Weise in einen Rest der Formel -(CH2)3-CO2R umwandeln läßt. Insbesondere ist R9 n-Butyl, 3-Cyanopropyl, 3-(Alkoxycarbonyl)-propyl, 2-Alkoxyethyl, 3-Alkoxypropyl oder 4-Alkoxybutyl, wobei Alkoxy vorzugsweise Methoxy,
Ethoxy, n-Propoxy oder i-Propoxy bedeutet. Die Verbindungen der Formel II werden in Gegenwart einer
Base mit den Elektrophilen der Formel III alkyliert. Hierfür geeignete Basen sind beispielsweise Alkalimetallhydroxide wie Natrium- oder Kaliumhydroxid,
Alkalimetallcarbonate wie Natriumhydrogencarbonat, Kaliumcarbonat oder Kaliumhydrogencarbonat, Alkalimetallacetate wie Natrium- oder Kaliumacetat, Erdalkalimetallhydroxide und - oxide wie Calciumhydroxid, Calciumoxid oder Aluminiumoxid oder organische Basen wie Triethylamin, Diisopropylethylamin, Pyridin,
Lutidin, Piperidin, Morpholin, Piperazin, Collidin, Chinolin oder N,N-Dimethylaminopyridin, insbesondere Triethylamin oder Pyridin. Zweckmäßig führt
man die Reaktion in einem inerten Lösungsmittel
durch. Als inerte Lösungsmittel eignen sich vorzugsweise Kohlenwasserstoffe wie Cyclohexan, Benzol oder
Toluol, Ether wie Tetrahydrofuran oder Dioxan, Amide wie Dimethylformamid oder Hexamethylphosphorsäuretriamid, Sulfoxide wie Dimethylsulfoxid, Alkohole wie Methanol oder Ethanol oder Ester wie Ethylacetat.
Auch ein Überschuß an organischer Base eignet sich als Lösungsmittel.
Die Reaktionstemperaturen liegen zweckmäßig zwischen etwa 0° und 200 °C, vorzugsweise zwischen 20 und
150 °C, die Reaktionszeiten zwischen etwa 1 und
48 Stunden.
Die Alkylierungsprodukte der Formel IV werden vorzugsweise zu den Ethern der Formel IVa verethert. In den Ethern der Formel IVa bedeutet R10 vorzugsweise Methyl, Ethyl, n-Propyl, n-Butyl, n-Pentyl, n-Hexyl, Trimethylsilyl, Triisopropylsilyl oder tert. Butyldimethylsilyl sowie weiterhin i-Propyl und tert. Butyl.
Die Herstellung der Alkylether der Formel IVa (R10 = C1-C6-Alkyl) kann nach den für Veretherungen bekannten und gebräuchlichen Methoden erfolgen, wie sie beispielsweise in Houben-Weyl, Methoden der Organischen Chemie Bd. 6/3, S. 1 ff beschrieben sind.
Vorzugsweise werden die entsprechenden Alkohole der Formel R10-OH mit dem Alkohol der Formel IV unter
Zusatz einer katalytischen Säuremenge oder eines wasserentziehenden Mittels wie z.B. einer Säure wie Schwefelsäure, Phosphorsäure, Chlorwasserstoff oder p-Toluolsulfonsäure, einem Säurederivat wie Phosphorpentoxid, Phosphortrichlorid, oder Phosphoroxychlorid, einem sauren Ionenaustauscher oder Molekularsieben umgesetzt.
Die Veretherung wird in einem geeigneten Lösungsmittel, dem reinen Alkohol R10-OH oder einem Gemisch
aus einem geeigneten Lösungsmittel und dem Alkohol
R10-OH durchgeführt. Geeignete Lösungsmittel sind z.B. Benzol, Toluol,
Chloroform, Dichlorethan, Diethylether und Tetrahydrofuran. Die Veretherung wird vorzugsweise bei Temperaturen zwischen -20 °C und +150 °C durchgeführt. Insbesondere bevorzugt wird die Veretherung im schwach
sauren Milieu, d.h. bei einem pH zwischen 2 und 4,
vorzugsweise zwischen 2,5 und 3,5, durchgeführt, um
eine Dehydratisierung des Alkohols der Formel IV zu
unterdrücken.
Die Herstellung der Trialkylsilylenolether erfolgt
durch Umsetzung des Alkohols der Formel IV bzw. des
Ethers der Formal IVa, ggf. nach Überführung der
von O verschiedenen Gruppe Z (Z = S, N-R') in O, mit
einem Silylierungsreagenz in Gegenwart einer Base
in einem inerten Lösungsmittel. Geeignete Silylierungsreagenzien sind die Trialkylsilylhalogenide wie z.B. Trimethylchlorsilan (TMCS), Triisopropylchlorsilan (TIPCS), tert. Butyldimethylchlorsilan (TBDMCS), Trimethylbromsilan (TMBS) und Trimethyljodsilan (TMJS), Trialkylsilyltriflate wie z. B. Trimethylsilyltriflat (TMSOTf), Thexyldimetethylsilyltriflat (ThDMSOTf) und Triisopropylsilyltriflat (TIPSOTf) und Trimethylsilylessigsäure ethylester (ETSA), N,O-Bis(trimethylsilyl)acetamid (BSA) N-Methyl-N-trimethylsilyltrifluoracetamid (MSTFA) und Hexamethyldisilazan (HMDS), Insbesondere bevorzugt sind die Trialkylsilyltriflate, da diese eine Reaktionsführung unter kinetischer Kontrolle bei Temperaturen zwischen -60 °
und 10 °C, vorzugsweise zwischen -30 und 0 °C, erlauben. Bei diesen Temperaturen sind die Reaktionen üblicherweise nach 10 Minuten bis 48 Stunden beendet. Als Basen sind organische Amine geeignet. Geeignete organische Amine sind z.B. Pyridin, Triethylamin, Diisopropylamin, Piperidin, Morpholin, tert. Butylamin, Collidin, Chinolin, 4-N,N-Dimethylaminopyridin, Diisopropylethylamin, 1,8-Diazabicyclo[5.4.0]undec-7-en (DBU) und 1,6-Diazabicyclo[4.3.0]non-5-en (DBN).
Der so erhaltene Silylenolether der Formel IVb,
worin R1, R2, R9, R10 und X die angegebene Bedeutung besitzen, und R11 jeweils unabhängig voneinander eine
Alkylgruppe mit 1-6 C-Atomen bedeutet, wird in situ mittels einer Lewis-Säure zu einem Biotinderivat der
Formel V cyclisiert.
Geeignete Lewis-Säuren sind Aluminium-, Zinn-, Zinkund Titanhalogenide bzw. -triflate, wie z.B. Aluminiumchlorid, Zinntetrachlorid, Zinkchlorid und Titantetrachlorid, Borhalogenide und Borsäureester, wie z.B. Bortrifluorid, Bortrichlorid und Trimethylborat oder Trialkylsilyltriflate wie z.B. Trimethylsilyltriflat und Triisopropylsilyltriflat.
Die Reaktionstemperatur liegt üblicherweise zwischen -90° und 0 °C vorzugsweise zwischen -90° und -50 °C. Bei diesen Temperaturen sind die Reaktionen üblicherweise nach 15 Minuten bis 4 Stunden beendet. In einer bevorzugten Ausführungsform des erfindungsgemäßen Verfahrens wird der Alkohol der Formel IV, bzw. der Ether der Formel IVa in einem inerten Lösungsmittel in Gegenwart einer Base bei einer Temperatu zwischen -30 °C und 0 °C mit etwa einem Äguivalent an Trialkylsilyltriflat in den Silylenolether der Formel IVb überführt und nach Abkühlen auf eine Temperatur zwischen 90 °C und -50 °C durch Zugabe einer weiteren unterstöchiometrischen Menge desselben Trialkylsilyltriflates in situ zum Biotinderivat der Formel V cyclisiert. Das Biotinderivat der Formel V liegt als Gemisch der Diastereomeren der Formeln Va und Vb vor.
Durch geeignete Reaktionsführung läßt sich das Verhältnis dieser Diastereomere beeinflussen.
Insbesondere erhält man nach Überführung des Alkohols der Formel IV bzw. des Ethers der Formel IVa in den Silylenolether der Formel IVb bei tiefen Temperaturen mit Trialkylsilyltriflaten und der Cyclisierung höhere Anteile des Diastereomers der Formel Vb.
Die Biotinderivate der Formel Vb können mit geeigneten Reduktionsmitteln zu Biotinderivaten der
Formel VIII,
worin R1, R2, R9, und X die angegebene Bedeutung besitzen, reduziert werden. Diese Biotinderivate der
Formel VIII sind bekannt und können nach an sich bekannten Methoden wie z.B. in DE-2058234, EP-0 036 030, EP-0 084377 und U.S. Pat. 3,859,167 offenbart, zu
D-(+)-Biotin umgesetzt werden.
Geeignete Reduktionsmittel zur Reduktion der Biotinderivate der Formel Vb zu solchen der Fomel VIII sind z.B. Zink-Amalgam in wäßriger Salzsäure (ClemmensenReduktion), Hydrazin-Hydrat in Gegenwart von Basen
(Wolff-Kishner, bzw. Huang Minlon-Reduktion), Wasserstoff in Gegenwart von Hydrierkatalysatoren bei 180 bis 250 °C oder nach Überführung der Ketogruppe (Z = O) in das entsprechende Tosylhydrazon (Z = N-NH-SO2-C7H7) auch komplexe Hydride wie z.B. Lithiumalanat oder Natrium-boranat.
Weiterhin kann das Diasteromerengemisch der Formeln Va und Vb durch Reduktion zum Alkohol der Formel Vc,
worin R1, R2, R9 und X die angegebene Bedeutung besitzen, mit einem komplexen Reduktionsmittel und anschließender Dehydratisierung zu einem DehydrobiotinDerivat der Formel VI überführt werden. Als komplexe Reduktionsmittel sind insbesondere jene geeignet, die auch zur Reduktion der Cysteinhydantoine in der Formel I Verwendung finden.
Man führt die Umsetzung der komplexen Hydride mit den Verbindungen der Formel V vorteilhaft in einem inerten Lösungsmittel, einem protischen Lösungsmittel oder einem Gemisch aus einem inerten und einem protischen
Lösungsmittel durch. Als inerte Lösungsmittel eignen sich vorzugsweise Kohlenwasserstoffe wie Pentan,
Hexan, Cyclohexan, Benzol oder Toluol, Ether wie
Diethylether, Tetrahydrofuran, Dioxan oder Ethylenglykoldimethylether sowie Gemische dieser Lösungsmittel untereinander. Als protische Lösungsmittel eignen sich vorzugsweise Alkohole wie Methanol, Ethanol, Isopropanol und Butanol. Die Reaktionstemperaturen liegen zweckmäßig zwischen etwa -120 °C und +150 °C, vorzugsweise zwischen -80 °C und +100 °C, die Reaktionszeiten zwischen etwa 30 Minuten und 24 Stunden.
Zur Dehydratisierung der Alkohole der Formel Va eignen sich beispielsweise Säuren wie Schwefelsäure, ChlorWasserstoffsäure, Phosphorsäure, Toluolsulfonsäure oder Basen wie Natrium- oder Kaliumhydroxid. Man kann dabei in Gegenwart oder Abwesenheit eines inerten Lösungsmittels bei Temperaturen zwischen etwa 0 °C und 150 °C, vorzugsweise zwischen 20 °C und 100 °C, arbeiten. Als Lösungsmittel kommen z.B. Alkohole wie Methanol, Ethanol, Isopropanol oder Butanol in Betracht. Vorzugsweise werden die Alkohole der Formel Vc mit Mesylchlorid in das entsprechende Mesylat überführt und anschließend in Gegenwart einer Base erhitzt.
Die Dehydrobiotin-Derivate der Formel VI sind bekannt und können nach an sich bekannten Verfahren (z.B.
EP 0 084 377) zu D-(+)-Biotin umgesetzt werden.
Das erfindungsgemäße Verfahren erlaubt somit die HerStellung von optisch aktivem D-(+)-Biotin auf einfache und stereospezifische Weise in hohen Ausbeuten aus leicht zugänglichen, wohlfeilen Ausgangsstoffen in wenigen, zum Teil in Eintopfverfahren durchführbaren Syntheseschritten und stellt damit einen wesentlichen Fortschritt auf dem Gebiet der Biotinsynthese dar.
Die folgenden Beispiele sollen das erfindungsgemäße Verfahren näher erläutern ohne es zu begrenzen. Folgende Abkürzungen werden verwendet:

Die spezifischen Drehwerte sind auf einem Perkin-Elmer Polarimeter im jeweils angegebenen Lösungsmittel gemessen.
Die Säulenchromatographischen Trennungen werden auf Silica 60 230-400 mesh durchgeführt.
Beispiel 1
Eine Lösung von 324,4 g (1,0 Mol) (7aR)-3-Phenyl-6-benzyl-1H,3H-imidazo[1,5-c]thiazol-5,7-dion in 3200 ml wasserfreier Essigsäure wird bei 85 °C portionsweise mit 261,6 g (4,0 Mol) Zinkpulver versetzt, weitere
6 Stunden bei 85 °C gerührt, filtriert und unter
vermindertem Druck eingeengt.
Den Rückstand löst man in 1000 ml Toluol, wäscht nacheinander mit 300 ml 1 N Salzsäure und 400 ml Wasser, trocknet über Natriumsulfat und engt unter vermindertem Druck ein.
Man erhält 308,5 g N,N,-Dibenzyl-L-cysteinhydantoin als farbloses Öl.
[α]3 20 65 = -190°, c = 1 (Methanol) Beispiel 2
Ein Gemisch aus 9,8 g (30 mmol) N,N'-Dibenzyl-L-cysteinhydantoin (hergestellt nach Beispiel 1) und 200 ml THF wird bei -70 °C mit 66 ml einer Lösung von DIBAH in
THF (1 mol/l) versetzt. Nach einstündigem Rühren bei -70 °C werden nacheinander 50 ml Wasser und 200 ml einer 2,5 % wäßrigen Natronlauge zugetropft.
Nach einstündigem Rühren bei Raumtemperatur werden die Phasen getrennt und nach üblichem Aufarbeiten und
chromatographischer Reinigung (EtOAc/Hexan 1/1) erhält von 9,82 g (99 % d. Th. ) (4 RS, 5 R)-1,3-Dibenzyl-2-oxo-5-mercaptomethyl-imidazolidin-4-ol, Rf = 0,40 (EtOAc/Hexan 1/1),
[α]D = -14,8 °, c = 0,870 (CHCl3)
Beispiel 3
Ein Gemisch aus 1,85 g (5,63 mmol) (4 RS, 5 R) 1,3-Dibenzyl-2-oxo-5-mercaptomethyl-imidazolidin-4-ol (hergestellt nach Beispiel 2) und 7,0 ml THF wird bei 0-5 °C mit 0,855 g (1,18 ml, 8,45 mmol) TEA und 0,547 g (0,47 ml, 5,91 mmol) Chloraceton versetzt. Das Gemisch wird
2 Stunden bei 0 °C und anschließend 2 Stunden bei
Raumtemperatur gerührt. Der Niederschlag wird ab filtriert und das Lösungsmittel abdestilliert. Nach Flashchromatographie erhält man 1,956 g (90 % d. Th.)
(4 RS, 5 R)-1,3-Dibenzyl-2-oxo-5-(4-oxo-2-thiapentyl)-imidazolidin-4-ol, Rf = 0,24 (EtOAc/Hexan 3/2)
Analog werden hergestellt:
(4 RS, 5 R)-1,3-Dibenzyl-2-oxo-5-(4-oxo-4-phenyl-2-thiabutyl)-imidazolidin-4-ol, Ausbeute: 88 % d. Th, Rf = 0,44 (EtOAc/ Hexan 1/1)
(4 RS/5 R)-1,3-Dibenzyl-2-oxo-5-(4-oxo-7-(methoxycarbonyl)-2-thiaheptyl)-imidazolidin-4-ol, Ausbeute 85 % d. Th, Rf = 0,41 (EtOAc/Hexan 3/2) [α]D = +34,8 ° , c = 1,150 (CHCl3)
Beispiel 4
Ein Gemisch aus 2,12 g (4 RS, 5 R)-1,3-Dibenzyl-2-oxo-5-(4-oxo-4-phenyl-2-thiabutyl)-imidazolidin-4-ol (hergerstellt nach Beispiel 3) und 100 ml Ethanol wird bei 0-5 °C mit 2 normaler Schwefelsäure in Ethanol auf pH 3,1 (gegen Methylorange) gebracht. Nach zweistündigem Rühren bei 0-5 °C wird mit 2 %igem Kaliumhydroxid in Ethanol auf pH 5 bis 6 gebracht. Nach üblicher Aufarbeitung und Flashchromatographie erhält
man 2,043 g (91 % d. Th. ) (4 RS, 5 R)-1,3-Dibenzyl-2-oxo-5-(4-oxo-4-phenyl-2-thiabutyl)-4-ethoxy-imidazolidin, Rf = 0,69 (EtOAc/Hexan 3/7). Analog werden hergestellt: (4 RS, 5 R)-1,3-Dibenzyl-2-oxo-5-(4-oxo-7-(methoxycarbonyl)-2-thiaheptyl)-4-ethoxy-imidazolidin, Ausbeute 85 % d. Th, Rf = 0,62 (EtOAc/Hexan 1/1)
(4 RS, 5 R)-1,3-Dibenzyl-2-oxo-4-ethoxy-5-mercaptomethyl-imidazolidin, Ausbeute 80 % d.Th. [a]D 20=
+ 20,1°, c = 1 (Methanol) (4 RS, 5 R)-1,3-Dibenzyl-2-oxo-4-ethoxy-5-(4-oxo-2-thiapentyl)-imidazolidin
Beispiel 5
Unter Stickstoffatmosphäre werden bei -20 °C 328 mg
TEA zu einem Gemisch aus 761 mg (4 RS, 5 R)-1,3-Dibenzyl-2-oxo-5-(4-oxo-7-(methoxycarbonyl)-2-thiaheptyl)-imidazolidin-4-ol (hergestellt nach Beispiel 3) und
10 ml trockenem Dichlormethan getropft. Zu diesem
Reaktionsgemisch tropft man ein Gemisch aus 722 mg
TMSOTf und 2 ml trockenem Dichlormethan. Nach Aufwärmen auf Raumtemperatur wird noch 1,5 Stunden weiter gerührt. Nach Abkühlen auf -60 °C wird ein Gemisch auf 180 mg TMSOTf und 1 ml trockenem Dichlormethan hinzugetropft.
Nach einer halben Stunde gießt man daß Reaktionsgemisch in eine wäßrige Natriumhydrogencarbonat-Lösung und extrahiert mit Dichlormethan. Die organischen Phasen werden über Kaliumcarbonat getrocknet und eingedampft. Nach Säulenchromatographie und Umkristallisation erhält man 305 mg (42 % d. Th) (3aS, 4S, 6aR)-1,3-Dibenzyl-4-
(1-oxo-4-methoxycarbonylbutyl)-tetrahydro-thieno[3,4-d] imidazolidin-2(3H)-on (Cyclisierungsprodukt) mit einem Schmelzpunkt von 80-81 °C [α]D 20 = -15,6°, C = 0,58 (CHCl3), Rf = 0,42 (EtOAc/
Hexan 1:1 + 1 % TEA).
Daneben erhält man als Nebenprodukt 293 mg (40 % d. Th) 1,3-Dibenzyl-2-oxo-4-(4-oxo-7-methoxycarbonyl-2-thiaheptyl)imidazolidin-4-en (Eliminierungsprodukt)
Rf = 0,25 (EtOAc/Hexan 1:1 + 1 % TEA) Beispiel 6
384 mg (4 RS, 5 R)-1,3-Dibenzyl-2-oxo-5-(4-oxo-2-thiapentyl) imidazolidin-4-ol in 2 ml Dichlormethan wird bis -20 °C gekühlt und mit 212 mg Triethylamin und 456 mg TMSOTf versetzt. Nach 1 1/2 h Rühren bei Raumtemperatur wird die Lösung auf -70 °C gekühlt.
Man gibt 222 mg TMSOTf zu und läßt noch 1 h bei -70 ° rühren. Das Gemisch wird in ag. NaHCO3 gegossen und die organische Phase über K2CO3 getrocknet. Nach Entfernung des Lösungsmittels wird der Rückstand mit
Flashchromatographie getrennt. Man erhält 108 mg
(30 % d. Th) (3 aS, 4S, 6 aR)-1,3-Dibenzyl-4-acetyltetrahydrothieno[3,4-d]imidazolidin-2(3H)-on, Schmelzpunkt: 1195-1215 °C, Rf = 0,61 (EtOAc/Hexan 1:1).
[α]D = 59,2°, c = 1.195, (CHCl3) Beispiel 7
In Analogie zur zuvor beschriebenen Umsetzung werden 223 mg (4 RS, 5 R)-1,3-Dibenzyl-2-oxo-5-(4-oxo-4-phenyl- 2-thiabutyl)-imidazolidin-4-ol in Dichlormethan (2 ml) mit 160 mg Diazabicyclo[5.4.0]undec-7-en (DBU) 160 mg TMSOTf bei -20 °C und weiteren 56 mg TMSOTf umgesetzt.
Nach Aufarbeiten erhält man 98 mg (46 %) (3aS, 4S, 6aR)-1,3-Dibenzyl-4-benzoyltetrahydrothieno[3,4-d] imidazolidin-2(3H)-on, Rf = 0,60 (EtOAc/Hexan 1:1). Schmelzpunkt: 127-1285 °C [α]D = +91,2°, c = 1.04, (CHCl3).
Beispiel 8
Eine Mischung aus 237 mg (4 RS, 5 R)-1,3-Dibenzyl-2-oxo-5-(4-oxo-4-phenyl-2-thiabutyl)-4-ethoxyimidazolidin (hergestellt nach Beispiel 4) und 2 ml Dichloromethan wird analog Beispiel 7 mit 53 mg TEA und 111 mg TMSOTf bei -20 °C versetzt und 1,5 Stunden bei Raumtemperatur gerührt, anschließend werden nochmals 111 mg TMSOTf bei -60 °C zugesetzt. Man erhält 165 mg (77 % d. Th) (3aS, 6aR, 4R)-1,3-Dibenzyl-4-benzoyltetrahydrothieno [3,4 d]imidazolidin-2(3H)-on, Rf = 0,51 (EtOAc/Hexan 3/2) mit einem Schmelzpunkt von 143-144 °C.
[α]D = -77,6°, c = 1,135 (CHCl3).
Analog erhält man aus (4 RS, 5 R)-1,3-Dibenzyl-2-oxo-4-ethoxy-5-(4-oxo-2-thiapentyl)-imidazolidin (hergestellt nach Beispiel 4)
(3aS, 6aR, 4R)-1,3-Dibenzyl-4-acetyltetrahydrothieno [3,4 d]imidazolidin-2(3 H)-on
Beispiel 9
Eine Mischung aus 500 mg (4 RS, 5 R)-1,3-Dibenzyl-2-oxo-5-(4-oxo-7-(methoxycarbonyl)-2-thiaheptyl)-4-ethoxyimidazolidin (hergestellt nach Beispiel 4) und 4 ml Dichlormethan wird analog Beispiel 5 mit 111 mg
TEA und insgesamt 467 mg TMSOTf umgesetzt. Man erhält in 310 mg (69 % d. Th) eines 1:1-Diastereomerengemisches aus (3aS, 6aR, 4S)-und (3aS, 6aR, 4R)-1,3Dibenzyl-4-(1-oxo-4-methoxycarbonylbutyl)-tetrahydrothieno[3,4d]imidazolidin-2(3 H)-on Rf = 0,65 (EtOAC/Hexan = 3/2).
Beispiel 10
Eine Mischung aus 500 mg (4 RS, 5 R)-1,3-Dibenzyl-2-oxo-5-(4-oxo-7-(methoxycarbonyl)-2-thiaheptyl)-4-ethoxyimidazolidin (hergestellt nach Beispiel 4) und 4 ml
Dichlormethan wird bei -20 °C und 111 mg TEA und 222 mg TMSOTf 10 Stunden gerührt. Nach Abkühlen auf -60 °C, Zugabe von weiteren 245 mg TMSOTf und weiterem halbstündigen Rühren erhält man 310 mg (69 % d. Th.) eines 2:1 Diastereomerengemisches aus (3aS, 6aR, 4R)-und (3aS, 6aR, 4S)-1,3-Dibenzyl-4-(1-oxo-4-methoxycarbonylbutyl)-tetrahydrothieno[3,4 d]imidazolidin-2(3H)-on. Durch Umkristallisation erhält man das reine (3aS,6aR,4R)-Diastereomer mit einem Schmelzpunkt von 101-102 °C.
[α]D = -10,7°, C = 0,205 (CHCl3)
Dieses wird mit Tosylhydrazin in das entsprechende
Tosylhydrazon überführt, welches mit Natriumboranat reduziert wird. Man erhält (3aS, 6aR, 4R)-1,3-Dibenzyl-4-(4-methoxycarbonylbutyl)-tetrahydrothieno[3,4-d] imidazolidin-2(3H)-on (Dibenzylbiotinmethylester).
Beispiel 11
Ein Gemisch aus 452,6 mg (3aS, 6aR)-1,3-Dibenzyl-4- (1-oxo-4-methoxycarbonylbutyl)-tetrahydrothieno [3,4-d] imidazolidin-2(3H)-on (1:1 (4 RS)-Diasteriomerengemisch, hergestellt nach Beispiel 9) und 40 ml Methanol wird
bei Raumtemperatur mit 38 mg Natriumboranat versetzt und 60 Minuten gerührt. Anschließend wird unter vermindertem Druck eingeengt in 40 ml Wasser aufgenommen und zweimal mit je 30 ml Diethylether extrahiert.
Die vereinigten organischen Phasen werden über Natriumsulfat getrocknet und unter vermindertem Druck eingeengt.
Als Rückstand wird (3aS, 6aR)-1,3-Dibenzyl-4-(1-hydroxy-4-methoxy-carbonylbutyl)-tetrahydrothieno[3,4-d]imidazolidin-2 (3H)-on erhalten.
Das Rohprodukt wird anschließend in einem Gemisch aus 15 ml Dichlormethan und 15 ml Triethylamin gelöst und bei Raumtemperatur mit 573 mg Methansulfonylchlorid ( MesylChlorid) versetzt. Die Lösung wird 90 Minuten bei 35 °C gerührt, filtriert und unter vermindertem Druck eingeengt.
Das rohe Mesylat wird anschließend in 15 ml DBU gelöst und 2 Stunden bei 60 °C gerührt. Danach kühlt man auf Raumtemperatur ab, verdünnt das Reaktionsgemisch mit 75 ml Toluol und wäscht nacheinander mit je 30 ml 2N Salzsäure sowie Wasser.
Man trocknet über Natriumsulfat, engt unter vermindertem Druck ein und chromatographiert den Rückstand an Kieselgel (Diethylether). Es werden 388 mg (3aS, 6aR)-1,3-Dibenzyl-4-(4-methoxycarbonylbutyliden)-tetrahydrothieno[3,4-d]imidazolidin-2(3H)-on erhalten.
[α]D 25 = +227°, c = 1 (Benzol)
Dieses wird durch Verseifen in alkoholischer Kalilauge in die entsprechende Carbonsäure überführt, Schmelzpunkt 81-83 °C
[α]D 22 : + 217°, c = 1 (Methanol) welche nach EP-00 84 377 in D-(+)-Biotin überführt wird.
Beispiel 12:
Unter Stickstoffatmosphäre werden bei -70 °C 168 mg ETSA zu einem Gemisch aus 249 mg (4RS, 5R)-1,3-Dibenzyl-2-oxo-5-(4-oxo-7-(methoxycarbonyl)-2-thiaheptyl-4-ethoxy-imidazolin (hergestellt nach Beispiel 4) und 1 ml trockenem
Tetrahydrofuran getropft. Zu diesem Reaktionsgemisch tropft man eine katalytische Menge TBAF in trockenem Tetrahydrofuran. Nach Aufwärmen auf Raumtemperatur wird noch 24 Stunden weiter gerührt und unter vermindertem Druck eingeengt. Den Rückstand löst man in 2 ml Dichlormethan. Nach Abkühlen auf -70 °C wird ein Gemisch aus 165 mg TMSOTf und 1 ml Dichlormethan hinzugetropft.
Nach einstündigem Rühren bei -70 °C und weiterem Aufarbeiten erhält man 176 mg (78 % d. Th. ) eines 3:2 Diastreomerengemisches aus (3αS, 6αR, 4R)- und (3αS, 6αR, 4S)-1,3-Dibenzyl-4-(1-oxo-4-methoxycarbonylbutyl)-tetrahydrothieno [3,4,d]imidazolidin-2(3H)-on. (Cyclisierungsprodukt).
Daneben erhält man als Nebenprodukt 33 mg (7 % d. Th. ) 1,3-Dibenzyl-2-oxo-4-(4-oxo-7-(methoxycarbonyl)-2-thiaheptyl)- imidazolidin-4-en. (Eliminierungsprodukt).
Claims
1. Verfahren zur Herstellung von D-(+)-Biotin, dadurch gekennzeichnet, daß man ein Cysteinhydantoin der Formel I,
wobei
R1 H, eine Gruppe R3R4CH- oder SiR5R6R7, worin R3 und R4 jeweils unabhängig voneinander
H, unsubstituiertes oder substituiertes Alkyl, Cycloalkyl, Aryl, Aralkyl oder Heteroaryl oder zusammengenommen unsubstituiertes oder substituiertes Alkylen oder
Heteroalkylen,
R5, R6
und R7 jeweils unabhängig voneinander
unsubstituiertes oder substituiertes Alkyl, Cycloalkyl, Aryl, Aralkyl oder Heteroaryl bedeuten, R2 H oder eine für ein Stickstoffatom geeignete Schutzgruppe und
X O, S oder N-R8, worin R8 eine Alkyl-,
Cycloalkyl-, Aryl-, Alkoxycarbonylgruppe oder eine Dialkylaminogruppe ist, bedeuten, unter Einwirkung eines komplexen Reduktionsmittels, zu einem Alkohol der Formel II reduziert.

worin R1, R2, R9, Z und X die angegebene Bedeutung besitzen und R10 Alkyl mit 1-6 C-Atomen oder eine Trialkylsilylgruppe bedeutet, nach Überführung in einen Silylenolether mit einem Silylierungsreagenz in Gegenwart einer Base mittels einer Lewis-Säure zu einem Biotinderivat der Formel V cyclisiert,
worin R1, R2, R9 und X die angegebene Bedeutung besitzen, und dieses Derivat durch Reduktion mit einem komplexen Reduktionsmittel und anschließende Eliminierung zu einem Dehydrobiotin-Derivat der Formel VI umsetzt,
worin R1, R2, R9 und X die angegebene Bedeutung besitzen, das nach an sich bekannten Verfahren in D-(+)-Biotin überführt wird.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß man zur Herstellung des Cysteinhydantoins der Formel I das optisch aktive bicyclische Hydantoin der Formel VII
worin R2, R3, R4 und X die angegebene Bedeutung besitzen, mit einer Säure und/oder einem Reduktionsmittel spaltet.
3. Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet, daß man als komplexes Reduktionsmittel zur Reduktion der Verbindung der Formel I Lithiumborhydrid oder Diisobutylaluminiumhydrid verwendet.
4. Verfahren nach mindestens einem der Ansprüche 1-3, dadurch gekennzeichnet, daß man als Base bei der Umsetzung der Verbindung der Formel II mit dem
Elektrophil der Formel III Triethylamin oder Pyridin verwendet.
5. Verfahren nach mindestens einem der Ansprüche 1-4, dadurch gekennzeichnet, daß man als Silylierungsmittel ein Trialkylsilyltriflat einsetzt.
6. Verfahren nach mindestens einem der Ansprüche 1-5, dadurch gekennzeichnet, daß man als Lewis-Säure zur Herstellung der Verbindung der Formel V ein Trialkylsilyltriflat einsetzt.
7. Verfahren nach Anspruch 5 oder 6, dadurch gekennzeichnet, daß das Trialkylsilyltriflat Trimethylsilyltriflat ist.
8. Eine Verbindung der Formel II,
worin R1 , R2 und X die in Anspruch 1 angegebene Bedeutung besitzen.
9. Verbindung nach Anspruch 8, dadurch gekennzeichnet, daß R1 und R2 Benzyl bedeuten.
10. Eine Verbindung der Formel IV,
worin R1, R2, R9, Z und X eine in Anspruch 1 angegebene Bedeutung besitzen.
11. Verbindung nach Anspruch 10, dadurch gekennzeichnet, daß R1 und R2 Benzyl, Z O und R9 -(CH2)3-COOR, worin R niederes Alkyl ist, bedeuten.
12. Eine Verbindung der Formel IVa,
worin R1, R2, R9, R10, Z und X die in Anspruch 1 angegebene Bedeutung besitzen.
13. Verbindung nach Anspruch 12, dadurch gekennzeichnet, daß R1 und R2 Benzyl, Z O und R9 -(CH2)3-COOR, worin
R niederes Alkyl ist, bedeuten.
14. Eine Verbindung der Formel V,
worin R1, R2, R9, Z und X die in Anspruch 1 angegebene Bedeutung besitzen,
15. Verbindung nach Anspruch 11, dadurch gekennzeichnet, daß R1 und R2 Benzyl, Z O, und R9 -(CH2)3-COOR, worin R niederes Alkyl ist, bedeuten.
