Verfahren zur Herstellung von Penem-Verbindungen
Die Erfindung betrifft die Herstellung von Penem-Verbindungen der allgemeinen Formel (I)

R1 eine organische Gruppe, R2 ein Wasserstoffatom oder eine Carboxylschutzgruppe
und X
(i) eine gewünschtenfalls geschützte Hydroxygruppe, (ii) ein Halogenatom,
(iii) eine Azidogruppe, (iv) eine gewünschtenfalls geschützte Pyridyl-
gruppe, (v) eine Formyloxygruppe, eine unsubstituierte Acyl·
gruppe mit 2 bis 6 Kohlenstoffatomen oder eine

Acyloxygruppe mit 2 bis 6 Kohlenstoffatomen, die durch ein oder mehrere Ilalogenatomc, Acylgruppen
mit 2 bis 6 Kohlenstoffatomen, Amino-, Hydroxy- oder Mercaptogruppen, wobei die Amino-,
Hydroxy- oder Mercaptogruppen frei oder geschützt sind, substituiert ist, (vi) eine Carbamoyloxy- oder N-Alkyl-carbamoyl-
oxygruppe,
(vii) eine Alkoxy- oder Alkylthiogruppe, jeweils mit
1 bis 12 Kohlenstoffatomen, die unsubstituiert
ist oder die substituiert ist durch ein oder mehrere Halogenatome, Formylgruppen, Acylgruppen
mit 2 bis 6 Kohlenstoffatomen, Amino-, Hydroxy- oder Mercaptogruppen, wobei die Amino-,
Mercapto- oder Hydroxygruppen frei sind
oder geschützt sind,
(viii) eine Triarylmethylthiogruppe oder (ix) eine heterocyclische Thiogruppe
oder eines pharmazeutisch annehmbaren Salzes davon. Die erfindungsgemäßen Verbindungen der Formel (I) sind als
Antibiotika geeignet oder als Zwischenprodukte zu deren Herstellung, indem man die O- und N-Schutzgruppen entfernt
oder indem man die Gruppe X gemäß der Definition unter (i) bis (iii) in davon verschiedene Gruppen X,
gemäß den Definitionen unter (iv) bis (ix) umwandelt.
Die organische Gruppe, für welche R. steht, schließt gewünschtenfalls
substituierte aliphatische, cycloaliphatische und aromatische Kohlenwasserstoffgruppen ein. Die aliphatischen
Kohlenwasserstoffgruppen sind vorzugsweise Alkylgruppen mit 1 bis 12 Kohlenstoffatomen. Methyl- und
Ethylgruppen, insbesondere Ethylgruppen, werden besonders bevorzugt. Bevorzugte Substituenten, von denen einer oder
mehrere vorliegen können, sind Hydroxy-, Amino-, Cyano-
und Mercaptogruppen. Die Hydroxy-, Amino- und Mercaptosubstituenten können frei sein oder geschützt sein. Die
cycloaliphatischen Kohlenwasserstoffgruppen sind vorzugsweise Monocycloalkylgruppen mit 4 bis 7 Ringkohlenstoffatomen,
wobei Cyclohexyl besonders bevorzugt wird. Bevorzugte Substituenten, die wiederum einfach oder mehrfach
vorliegen können, sind außer den bereits vorher erwähnten Substituenten Alkylgruppen mit 1 bis 6 Kohlenstoffatomen,
insbesondere Methyl- und Ethylgruppen. Die bevorzugte aromatische Kohlenwasserstoffgruppe ist eine Phenylgruppe,
die durch ein oder mehrere Substituenten, wie sie im Zusammenhang mit den cycloaliphatischen Kohlenwasserstoffgruppen
erwähnt wurden, substituiert sein kann. 1-Hydroxyethyl, frei oder geschützt, ist eine bevorzugte Gruppe
für R- .
Die Carboxyschutzgruppen, die für R„ stehen, schließen ein:
(a) Alkylgruppen mit 1 bis 6 Kohlenstoffatomen,
(b) Haloalkylgruppen mit 1 bis 6 Kohlenstoffatomen,
(c) Alkenylgruppen mit 2 bis 4 Kohlenstoffatomen,
(d) gewünschtenfalls substituierte Arylgruppen,
(e) gewünschtenfalls substituierte Aralkylgruppen, wobei
der Alkylteil davon 1 bis 6 Kohlenstoffatomen enthält und
(f) Aryloxyalkylgruppen.
Beispiele hierfür sind:
(a) Methyl-, Ethyl- und t-Butylgruppen, (b) die 2,2,2-Trichloroethylgruppe,
(c) die Allylgruppe,
(d) Phenyl- und p-Nitrophenylgruppen,
(e) Benzyl-, p-Nitrobenzyl-, p-Methoxybenzyl-, Diphenylmethyl-
und Di-(o-nitrophenyl)-methylgruppen und
(f) die Phenoxymethylgruppe.
Weitere Gruppen, die als typische Carboxyschutzgruppen erwähnt werden können, sind Acetonyl-, Trimethylsilyl-,
Diphenyl-t-butylsilyl- und Dimethyl-t-butyl-silylgruppen.
Eingeschlossen sind auch Reste, die in vivo hydrolysieren und die günstige pharmakologische Eigenschaften aufweisen
wie Acetoxymethyl-, Pivaloyloxymethyl- und Phthalidylgruppen.
Bedeutet X eine geschützte Hydroxygruppe, dann sind bevorzugte
Schutzgruppen p-Nitrobenzyloxycarbonyl; Diphenylt-butyl-silyl;
Trimethylsilyl; 2,2,2-Trichloroethoxy-carbonyl;
Triphenylmethyl und Pyranyl.
Bedeutet X ein Halogenatom, dann ist es vorzugsweise ein Chlor-, Brom- oder Jodatom. Bedeutet X eine unsubstituierte
Pyridylgruppe, dann ist es vorzugsweise eine 1-Pyridylgruppe. Bedeutet X eine substituierte Pyridylgruppe, dann
ist es vorzugsweise eine 1-Pyridylgruppe, die durch eine Carbamoylgruppe und insbesondere ein 4-Carbamoylgruppe
substituiert ist. Die Acyloxygruppe, für die X stehen kann, ist vorzugsweise eine aliphatische Acyloxygruppe mit 2 bis
6 Kohlenstoffatomen, insbesondere Acetyl. Im Falle der
Substitution mit einer Acylgruppe ist die Acylgruppe vorzugsweise eine aliphatische Acylgruppe mit 2 bis 6 Kohlenstoffatomen,
insbesondere eine Acetylgruppe. Ist X eine N-Alkyl-carbamoyloxygruppe, dann enthält der Alkylsubstituent
vorzugsweise 1 bis 6 Kohlenstoffatome, wobei besonders
eine Methyl- oder Ethylgruppe bevorzugt ist. Die Alkoxy-
oder Alkylthiogruppen, für die X stehen kann, haben vorzugsweise
1 bis 6 Kohlenstoffatome. Bevorzugt werden Methoxy-,
Ethoxy-, Methylthio- und Ethylthiogruppen. Triphenylmethylthio
ist eine bevorzugte Triarylmethylthiogruppe, für welche X stehen kann.
Bedeutet X eine Heterocyclylthiogruppe, dann ist der Heteroring vorzugsweise:
A) ein fünfatomiger oder sechsattomiger heteromonocyclischer
Ring enthaltend wenigstens eine Doppelbindung und wenigstens ein Heteroatom, ausgewählt aus N, S
und O, unsubstituiert oder substituiert durch eine oder mehrere der nachfolgenden Substituenten, ausgewählt
aus
a) Hydroxy, C -Cg-Alkoxy, Halogen, C^-Cg-aliphatisches
Acyl;
b) Cj-Cg-Alkyl, unsubstituiert oder substituiert durch
einen oder mehrere Substituenten, ausgewählt aus Hydroxy und Halogen;
c) Cρ-Cg-Alkenyl, unsubstituiert oder substituiert durch
einen oder mehrere Substituenten, ausgewählt aus Hydroxy und Halogen;
d) -S-R3, worin R3 Wasserstoff oder Cj-Cg-Alkyl bedeu-
tet oder -S-CH0-COOR,, worin R. Wasserstoff, C1-C,-

Alkyl oder eine Carboxyschutzgruppe bedeutet;
e) -(CH2J1n-COOR4 oder -CH=CH-COOR4, worin m Null, 1, 2
oder 3 bedeutet und R. die oben angegebene Bedeutung hat;
-(CH9) -CN oder -(CH9) -CONH9, worin m die vorher

-(CH„) -SO3H, worin m die vorher angegebene Bedeutung
hat, oder
f) -(CHj)-N , worin m die vorher angegebene Be-
deutung hat, wobei jedes R5 und Rg, die gleich oder
verschieden sein können, Wasserstoff, C.-C,--Alkyl
oder eine aliphatische Acylgruppe bedeuten oder wenn einer der Reste Rc und R, Wasserstoff ist, der an-

dere auch eine Aminoschutzgruppe bedeuten kann oder
B) ein heterobicyclischer Ring, enthaltend wenigstens zwei Doppelbindungen, wobei jeder der kondensierten heteromonocyclischen
Ringe, die gleich oder verschieden sein können, ein fünfatomiger oder sechsatomiger heteromonocyclischer
Ring ist, enthaltend wenigstens ein Heteroatom, ausgewählt aus N, S und O, und wobei der heterobicyclische
Ring unsubstituiert ist oder substituiert ist durch einen oder mehrere Substituenten ausgewählt
aus a) , b) , c) , d) , e) und f) .
Bei den obigen Definitionen A) und B) sind bevorzugte HaIogene
Chlor, Brom und Jod; bevorzugte C1-C-.-Alkylgruppen Methyl
und Ethyl; eine bevorzugte C~-Cfi-Alkenylgruppe Allyl;
eine bevorzugte aliphatische Acylgruppe Acetyl. Eine Carboxyschutzgruppe
kann eine der zuvor für den R„-Substituenten angegebene sein und die freien Sulfo- und Carboxygruppen
können gegebenenfalls neutralisiert sein, z. B. als Natriumoder Kaliumsalze.
Ein heteromonocyclischer Ring der obigen Klasse A) kann beispielsweise ein gegebenenfalls substituierter Thiazolyl-,
Triazolyl-, Thiadiazolyl-, Tetrazolyl- oder Triazinylring sein. Bevorzugte Substituenten an diesen Ringen
sind ein oder mehrere Substituenten, ausgewählt aus Amino-,
Hydroxy-, Oxo- und C1-C-.-Alkylgruppen, vorzugsweise Methyl
oder Ethyl, worin die C1 -C,-Alkylgruppe gewiinschtenfalls
substituiert sein kann durch einen Substituenten, ausgewählt
aus Carobxy, SuIfο, Cyano, Carbamoyl, Amino, Methylamino
oder Dimethylamino.
Ein heterobicyclischer Ring der obigen Klasse B) kann beispielsweise
ein Tetrazolopyridazinylrest, der gewünschtenfalls
durch Amino- oder Carboxy substituiert ist, sein.
In der obigen Formel (I) können die Amino-, Hydroxy- oder Mercaptoschutzgruppen, die gegebenenfalls vorhanden sind,
solche sein, wie sie üblicherweise in der Chemie der Penicilline und Cephalosporine verwendet werden. Sie können
beispielsweise gegebenenfalls substituierte, insbesondere
halogensubstituierte Acylgruppen, z. B. Acetyl-, Monochloroacetyl-, Dichloroacetyl-, Trifluoroacetyl-, Benzoyl-
oder p-Bromophenacylgruppen sein, oder Triarylmethylgruppen, insbesondere Triphenylmethylgruppen; Silylgruppen, insbesondere
Trimethylsilyl, Dimethyl-t-butyl-silyl, Diphenylt-butyl-silyl;
oder auch Gruppen wie t-Butoxycarbonyl, p-Nitro-benzyloxycarbonyl, 2,2,2-Trichloroethoxycarbony1,
Benzyl und Pyranyl.
Ist der R1-Substituent in der Formel (I) eine Alkylgruppe,
die durch Hydroxy substituiert ist, dann ist eine bevorzugte Schutzgruppe für die Hydroxyfunktion p-Nitrobenzyloxycarbonyl;
Dimethyl-t-butyl-silyl; Diphenyl-t-butylsilyl;
Trimethylsilyl; 2,2,2-Trichloroethoxycarbonyl;
Benzyl-p-bromo-phenacyl; Triphenylmethyl und Pyranyl.
Alle Alkyl- und Alkenylgruppen, einschließlich des aliphatischen
Kohlenwasserstoffrestes der Alkoxy-, Alkylthio-
und Acyloxygruppen können verzweigt- und geradkettig sein.
Eine bevorzugte Ausführungsform der Erfindung ist auf ein
Verfahren zur Herstellung von Verbindungen der Formel (I) gerichtet, worin R CH3-CH- bedeutet, worin R3 ein Was-

serstoffatom oder eine Hydroxyschutzgruppe ist und X eine gewünschtenfalls geschützte Hydroxy- oder Carbamoyloxygruppe
ist und insbesonsere auch die Herstellung der vorstehenden Verbindungen, worin R- eine a-Hydroxyethylgruppe
ist;
X eine Carbamoyloxygruppe und R2 eine Acetoxymethylgruppe
bedeutet.
Eine besonders bevorzugte Ausführungsform der Erfindung
besteht in der Herstellung der Verbindungen der nachfolgenden Formel (I1) mit der Stereokonfiguration 5R,6S[I(R)]
der folgenden sterischen Konfiguration:

worin R2 und R3 die vorher angegebenen Bedeutung haben
und X1 eine gewünschtenfalls geschützte Hydroxy- oder Carbamoyloxygruppe bedeutet.
Wie bereits erwähnt, sind die Verbindungen der Formel (I) antibakterielle Mittel und werden beispielsweise in der
GB-PS 2 043 639 und in den britischen Patentanmeldungen 2097786-A und 2118181-A beschrieben, oder wenn X die unter
(i) bis (iii) angegebene Bedeutung haben, können sie
in brauchbare Antibiotika der Formel (I), worin X die unter (iv) bis (ix) angegebene Bedeutung haben, umgewandelt
werden, wie dies ausführlich in den GB-Patentanmeldungen 2111496-A und 2118181-A beschrieben wird.

Die Erfindung betrifft ein Verfahren zur Herstellung von Penem-Verbindungen der allgemeinen Formel (I) wie vorher
angegeben, und das Verfahren betrifft die Umsetzung eines Azetidinonderivats der allgemeinen Formel (II)

worin R1 und R„ uns X die vorher angegebene Bedeutung haben,
mit bis zu 2 Moläquivalenten einer dreiwertigen Organophosphorverbindung in einem inerten Lösungsmittel bei einer
Temperatur von 110 bis 150°C während einer Zeitspanne von
2 h bis zu einigen Tagen.
Die Organophosphorverbindungen können ein cyclisches oder acyclisches Trialkylphosphit, Triarylphosphit, gemischtes
Arylphosphit oder Phosphoramid sein. Trialkylphosphite,
insbesondere Triethylphosphit, werden bevorzugt.
Geeignete inerte Lösungsmittel sind beispielsweise Toluol, Xylol und Dimethylformamid, wobei Toluol und Xylol bevorzugt
werden. Die Umsetzung wird bei etwa 1400C während 5 bis
15h durchgeführt.
Die allgemeine Formel (II) umfaßt alle optischen Formen (razemische Verbindungen oder optisch aktive Verbindungen).
Die bevorzugte Konfiguration is 3S, 4R; insbesondere,
wenn R. Hydroxyethyl bedeutet, ist die (R) Konfiguration
für das Kohlenstoffatom, welches die Sauerstoffunktion
trägt, bevorzugt, um die bevorzugte endgültige (5R, 6S, 8R) Stereochemie des Penem-Kerns zu erhalten.
Die Umwandlung einer Verbindung der Formel (I), worin X eine geschützte Hydroxygruppe ist, in eine andere Verbindung
der Formel (I), worin X davon verschieden ist, kann nach bekannten Verfahren erfolgen, z. B. indem man
die Hydroxyschutzgruppe in der 2-Hydroxymethylgruppe entfernt und die erhaltene Verbindung der Formel (I), worin
X OH bedeutet, mit Trichloroacetylisocyanat umsetzt.
Die vorhandenen Schutzgruppen können nach literaturbekannten Verfahren abgespalten werden, wodurch man dann die
freie Penem-Verbindung I als Salz (R2=Na) erhält. Das erfindungsgemäße
Verfahren stellt einen erheblichen Fortschritt gegenüber dem Stand der Technik zur Herstellung
von Penem-Verbindungen I dar.

Aus dem obigen Sehern geht hervor, daß man die Penemverbindungen
I durch eine kürzere Route herstellen kann, und daß man umständige Stufen vermeiden kann, und wobei
man eine sehr erwünschte Ausbeutesteigerung erzielt. Es wurden keine Veränderungen in der stereochemischen
Konfiguration festgestellt, wobei auch in dem vorliegenden
Falle keine diasteromeren Mischungen erhalten werden.
Das Ausgangsmaterial der allgemeinen Formel (II) kann auf verschiedene Weise hergestellt werden:
1) Durch Ozonolyse der Verbindung (III), wie dies in Stufe
(h) im obigen Schema gezeigt wird,und dies in den vorerwähnten Patentschriften beschrieben wird (Y bedeutet
ein Wasserstoffatom oder eine Alkyl-, substituierte Alkyl-, Alkoxycarbonyl- oder Cyanogruppe);
2) durch Ozonolyse der Verbindung (V), hergestellt gemäß Tetrahedron Letters 24, Seite 1623 bis 1629 (1983).

Die Verbindung (II) erhält man, indem man eine N-H-freie
Verbindung (IV) mit einem geeigneten Oxalyloylderivat der allgemeinen Formel (VI) umsetzt, worin Z ein Halogenatom,

Die folgenden Herstellungsbeispiele und Beispiele beschreiben

Herstellung A: 4ß-Acetylglykollylthio-3a-[1(R)-t-butyldimethylsilyloxyethyl]-1-(p-nitrobenzyloxyoxalyl)-azetidin-2-on
1,5 g 4ß-Acetoxymethylvinylthio-3a-[1(R)-t-butyldimethylsilyloxy-ethyl]-1
-(1-p-nitrobenzyloxycarbonyl-2-methyl-1-propenyl)-azetidin-2-on
wurden in 80 ml Dichloromethan gelöst und auf -78°C gekühlt. Ozon und
Sauerstoff wurde durch die Lösung geperlt, bis eine blaue Farbe eintrat. Die organische Phase wurde mit Ethylacetat
verdünnt, mit einer gesättigten Natriummetabisulfitlosung
und dann mit Wasser gewaschen und über wasserfreiem Natriumsulfat getrocknet. IR 1820, 1780 - 1720 breit
Herstellung B:
4ß-t-Butyldiphenylsilylglykolloylthio-3a-[1(R)-t-butyl-0
dimethylsilyloxy-ethyl]-azetidin-2-on
Methode A
30 g 3a-[1(R)-t-Butyldimethyloxy-ethyl]-4-acetoxyazetidin-2-on
wurden in 80 ml Aceton und 45 ml Wasser gelöst. Zu dieser Lösung wurden 32 g t-Butyldiphenylsilyloxythioessigsäure,
gelöst in 80 ml Aceton und ml 1N Natriumhydroxid bei 00C zugegeben. Nach 2 h wurde
der Niederschlag gesammelt und mit kaltem Aceton:Wasser-Gemisch gewaschen. Man erhielt 18 g der Titelverbindung
als weißen Feststoff. F. 118 bis 122°C.

Methode B
1,2 g 4ß-Benzothiazolyldithio-3a~[1(R)-t-butyldimethylsilyloxy-ethyl]-azetidin-2-on
wurden in 30 ml Dichlormethan gelöst und 0,87 g t-Butyldiphenylsilyloxythioessigäsure
wurden bei Raumtemperatur zugegeben. Das rohe Reaktionsgemisch wurde dann mit 0,74 g Triphenylphosphin
behandelt und die Titelverbindung wurde kieselgelsäulenchromatographisch gereinigt, wobei man 1,15 g eines Produktes
erhielt, das identisch war mit der gemäß Methode A erhaltenen Verbindung.
Herstellung C:
4ß-t-Butyldiphenylsilylglykollöylthio-3a-[1(R)-t-butyldimethylsilyloxyethyl]-i-allyloxyoxalyl-azetidin-2-on
1,37 g 4ß-t-Butyldiphenylsilylglykolloylthio-3a-[1(R)-tbutyldimethylsilyloxy-ethyl]-azetidin-2-on
wurden in 10
ml Dichloromethan gelöst und auf 100C gekühlt. 1 g Calciumcarbonat,
0,445 g Allyloxyoxalylchlorid und 0,54 ml Diisopropylethylamin,
gelöst in 2 ml Dichlormethan, wurden zugegeben. Das Reaktionsgemisch wurde filtriert, mit ethanolfreiem
Chloroform verdünnt und zweimal mit Eiswasser gewaschen. Die organische Phase wurde über wasserfreiem
Natriumsulfat getrocknet und eingedampft. Das Rohprodukt wurde für die nächste Stufe ohen weitere Reinigung verwendet.

Methode ergab die Titelverbindung.
Herstellung E:
4ß-Glykolloylthio-3a-[1(R)-t-butyldimethylsilyloxyethyl]-1-(1-p-nitrobenzyloxycarbonyl^-methyl-i-propenyl)
-azetidin-2-on
Ausgehend von 4ß-Benzothiazolyldithio-3a-[1(R)-tbutyldimethylsilyloxy-ethyl]-1
-(1-p-nitrobenzyloxycarbonyldimethylsilyloxy-ethyl]-1-(1-p-nitrobenzyloxycarbonyl-2-methyl-1-propenyl)-azetidin-2-on
und weitere Bearbeitung gemäß Herstellung B, Methode B, wobei man jedoch Hydroxythioessigsäure anstelle von t-Butyldiphenylsilyloxythioessigsäure
verwendete, ergab die Titelverbindung.
Herstellung F:
4ß-Glykollylthio -3a-[1(R)-t-butyldimethylsilyloxyethyl]-1~(p-nitrobenzyloxyoxalyl)-azetidin-2-on
Methode A
Ausgehend von 4ß-Hydroxymethylvinylthio-3a-[1(R)-t~ butyldimethylsilyloxy-ethyl]-1 -(1-p-nitrobenzyloxycarbonyl-2-methyl-1-propenyl)-azetidin-2-on,
wobei man gemäß Herstellung A arbeitete, ergab die Titelverbindung.
Methode B
Ausgehend von 4ß-Glykolloylthio-3a-[ 1 (R)-t-butyldimethylsilyloxy-ethyl]-1-[1-p-nitrobenzyloxycarbonyl-2-methyl-1-propenyl]-azetidin-2-on,
wobei man gemäß Herstellung A arbeitete, ergab die Titelverbindung. Diese war in jeder
Hinsicht identisch mit der gemäß Methode A erhaltenen Verbindung.
Herstellung G:
4ß-[(1-Methyl-1H-tetrazol-5-ylthio)-3a-[1(R)-t-butyldimethylsilyloxy-ethyl]-1-(p-nitrobenzyloxyoxalyl)-azetidin-2-on
Ausgehend von 4ß-[Ci-Methyl-IH-tetrazol-5-ylthio)-methylvinylthio]-3a-[1(R)-t-butyldimethylsilyloxy-ethyl]-1-(1-p-nitrobenzyloxycarbonyl-2-methyl-i-propenyl)-azetidin-2-on,
wobei man gemäß Herstellung A arbeitete, ergab die Titelverbindung.
Herstellung H:
4ß-Carbamoylglykolloylthio-3a-[1(R)-t-butyldimethylsilyloxy-ethyl]
-1 -(p-nitrobenzyloxyoxalyl)-azetidin-2-on
Ausgehend von 4ß-Carbamoyloxymethylvinylthio-3a-[1(R)-tbutyldimethylsilyloxy-ethyl]-1-(1-p-nitrobenzyloxycarbonyl-2-methyl-1-propenyl)-azetidin-2-on
und weitere Verarbeitung gemäß Herstellung A ergab die Titelverbindung,
Herstellung I:
4ß-Carbamoylglykolloylthio-3a-[1(R)-trichlorethoxycarbonyloxyethyl]-1-(acetoxymethyloxyoxalyl)-azetidin-2-on
1,1 g 4ß-(1-Carbamoyloxymethylvinyl)thio-3a-[1(R)-trichlorethoxycarbonyloxyethyl]-1-(1-acetoxymethyloxycarbonyl-2-methyl-1-propenyl)-azetidin-2-on
wurden in 30 ml Dichlormethan gelöst, auf -78°C gekühlt und ein Strom aus Ozon und Sauerstoff wurde durch die Lösung gegeben,
bis die Beendigung der Umsetzung durch TLC-Analyse angezeigt wurde.
Der Ozonüberschuß wurde durch einen Stickstoffstrom entfernt
und die Lösung wurde bei Raumtemperatur mit weiterem Dichlormethan verdünnt und mit einer wäßrigen Lösung
aus Natriummetabisulfit und dann mit Kochsalzlösung gewaschen.
Die über Na3SO4 getrocknete Lösung wurde eingedampft,
wobei man 1 g der rohren Titelverbindung als weißen Schaum erhielt. IR (cm"1) 1825, 1770, 1730.
p-Nitrobenzyl-(5R,6S)-2-acetoxymethyl-6-[1(R)-t-butyldimethylsiloxy-ethyl]-penem-3-carboxylat
1,2 g rohes 4ß-Acetylglykolylthio-3a[1(R)-t-butyldimethylsilyloxy-ethyl]-1-(p-nitrobenzyloxyoxalyl)-azetidin-2-on,
hergestellt gemäß Herstellung A, wurde in 60 ml Xylol gelöst und dazu wurden 1,5 ml Triethylphosphit gegeben,
und die Reaktionsmischung wurde 12h rückflußbehandelt.
Die Reinigung des Reaktionsproduktes durch Kieselgelsäu-0 lenchromatographie ergab 0,85 g der Titelverbindung.

Beispiel 2:
p-Nitrobenzyl- (5R, 6S) ^-acetoxymethyl-ö- [ 1 (R) -hydroxyethyl]-penem-3-carboxylat
Ausgehend von p-Nitrobenzyl- (5R, 6S) ^-acetoxymethyl-S-[1(R)-t-butyldimethylsilyloxy-ethyl]-penem-3-carboxylat,
hergestellt gemäß Beispiel 1, und weiterer Verarbeitung gemäß dem nachfolgenden Beispiel 8 ergab die Titelverbindung

Natrium-(5R, 6S) ^-acetoxymethyl-ö- [1 (R) -hydroxyethyl] penem-3-carboxylat
0,7 g p-Nitrobenzyl-(5R,6S)-2-acetoxymethyl-5-[1 (R) hydroxyethyl]-penem-3-carboxylat,
hergestellt gemäß Beispiel 2, wurden in 50 ml Ethylacetat gelöst. 30 ml einer gesättigten wäßrigen Natriumbicarbonatlösung wurden zugegeben,
und die Reaktionsmischung wurde unter Wasserstoff bei Raumtemperatur geschüttelt. Die rohe wäßrige
Phase wurde durch Umkehrphasensäulenchromatographie gereinigt, wobei man 0,250 g der reinen Titelverbindung
erhielt.

Beim Abdampfen des Lösungsmittels erhielt man 0,3 g eines Festproduktes. Beim Umkristallisieren aus Chloroform-Hexan
erhielt man 0,27 g der reinen Titelverbindung mit einem F. von 127-128°C.

hergestellt gemäß Herstellung C, wurden in 30 ml Xylol gelöst und rückflußbehandelt. 1,5 mg Triethylphosphit,
gelöst in 2 ml Xylol, wurden zugegeben, und die Mischung wurde unter Rückfluß 5 h behandelt. Das Titelprodukt
wurde durch Kieselgelentspannungschromatographie mit CyclohexanrEthylacetat gereinigt, wobei man 55 0 mg
des reinen Produktes erhielt.

Beispiel 7:
Allyl-(5R,6S)-2-(N-trichloracetylcarbamoyloxy-methyl)-6-[1(R)-t-butyldimethylsilyloxy-ethyl]-penem-3-carboxylat
3,1 g Allyl-(5R,6S)-2-hydroxymethyl-6-[1(R)-t-butyldimethylsilyloxy-ethyl]
-penem-3-carboxylat, hergestellt gemäß Beispiel 6, wurden in 100 ml Dichlormethan gelöst.
Die Lösung wurde auf -4 0 C gekühlt und dazu wurden tropfenweise 1,38 ml Trichloracetylisocyanat gegeben. Das Reaktionsgemisch
wurde auf Raumtemperatur erwärmt und mit einer 2 %igen wäßrigen Lösung von Natriumbicarbonat und
dann mit Wasser gewaschen. Dann wurde es über wasserfreiem Natriumsulfat getrocknet und eingedampft, wobei
man 4,8 g eines weißen Wachses erhielt, das ohne weitere Reinigung für die nächste Stufe verwendet wurde.
Allyl- (5R, 6S) ^-carbamoyloxymethyl-e- [ 1 (R) -hydroxyethyl]-penem-3-carboxylat
4,8 g rohes Allyl-5R,6S)-2-(N-trichloracetylcarbamoyloxymethyl-6-[1(R)-t-butyldimethylsilyloxy-ethyl]-penem-3-carboxylat,
hergestellt gemäß Beispiel 7, wurden in 40 ml Tetrahydrofuran gelöst. 4,43 ml Essigsäure und
7,35 g Tetrabutylammoniumfluorid wurden bei Raumtemperatur zugegeben. Das Reaktionsgemisch wurde 24 h unter
Stickstoff stehen gelassen. Das Lösungsmittel wurde abgedampft und der Rückstand wurde in 100 ml Ethylacetat
0 gelöst. Die Lösung wurde zuerst mit einer gesättigten Lösung von Natriumbicarbonat gewaschen und dann über
wasserfreiem Natriumsulfat getrocknet und auf ein klei-
nes Volumen eingedampft. Das Produkt wurde durch Zugabe
von Tolul ausgefällt, wobei man 1,56 g eines weißen Feststoffes
erhielt.

1,56 g Allyl-(5R,6S)-2-carbamoyloxymethyl-6-[1(R)-hydroxyethyl]-penem-3-carboxylat,
hergegestellt gemäß Beispiel 8, wurden in 20 ml Tetrahydrofuran gelöst und
mit 0,15 g Triphenylphosphin, 1,29 g Natriumethylhexanoat
und 0,15 g Tetrakistriphenylphosphin-Pd bei Raumtemperatur behandelt. Es fiel ein Niederschlag aus, der

nach 30 min durch Zentrifugieren gesammelt wurde. Dann wurde über Lichroprep RP-18 (Merck) die Reinigung durchgeführt
und schließlich das erhaltene Produkte lyophilisiert. "Merck" ist ein Handelsname. 1,34 g einer weißen
lyophilisierten Verbindung wurden erhalten.

hergestellt gemäß Herstellung F und weitere. Verfahrensweise gemäß Beispiel 1 ergab die Titelverbindung.

6-[1(R)-hydroxyethyl]-penem-3-carboxylat, hergestellt gemäß Beispiel 12 und weitere Verarbeitung wie in Beispiel 3,
ergab die Titelverbindung.
25 Beispiel 14:
p-Nitrobenzyl-(5R,6S)-2-[(1-methyl-IH-tetrazol-5-ylthio)
methyl]-6-[1(R)-t-butyldimethylsilyloxy-ethyl]-penem-3-carboxylat
Ausgehend von 4ß-[(1-Methyl-1H-tetrazol-5-ylthio)-methylthio-3a-[1
(R)-t-butyldimethylsilyloxy-ethyl]-1 -(p-nitrobenzyloxyoxalyl)-azetidin-2-on,
hergestellt gemäß Herstellung G und weitere Verarbeitung gemäß Beispiel 1,
ergab die Titelverbindung.
p-Nitrobenzyl-(5R,6S)-2-[(1-methyl-IH-tetrazol-5-ylthio)
methyl]-6-[1(R)-hydroxyethyl]-penem-3-carboxylat
Ausgehend von p-Nitrobenzyl-(5R,6S)-2-[(1-methyl-1H-tetrazol-5-ylthio)-methyl[-6-[1(R)-t-butyldimethylsilyloxy-ethyl]
-penem-3-carboxylat, hergestellt gemäß Beispiel 14 und weitere Verarbeitung gemäß Beispiel 8,
ergab die Titelverbindung.
Natrium-(5R,6S)-2-[(1-methyl-IH-tetrazol-5-ylthio) methyl]-6-[1(R)-hydroxyethyl]-penem-3-carboxylat
0,7 g p-Nitrobenzyl-(5R,6S)-2-[(1-methyl-IH-tetrazol-S-
-methyl]-6-[1(R)-hydroxyethyl]-penem-3-carboxylat, hergestellt gemäß Beispiel 15, wurden in 40 ml Tetrahydrofuran
gelöst und auf 0 C gekühlt. Eine wäßrige Lösung von Ammoniumchlorid wurde zusammen mit Eisenstaub zugegeben.
Nach Beendigung der Umsetzung wurde das unlösliche Material abfiltriert und das Produkt durch Umkehrphasensäulen-Chromatographie
gereinigt. Man erhielt 0,28 g der Titelverbindung .

Ausgehend von 4ß-Carbamoylglykolloylthio-3a-[1(R)-tbutyldiraethylsilyloxy-ethyl]-1-(p-nitrobenzyloxyoxalyl)-
azetidin-2-on, hergestellt gemäß Herstellung H und weitere Verarbeitung gemäß Beispiel 1, ergab die Titelverbindung.
p-Nitrobenzyl-(5R,6S)-2~carbamoyloxymethyl-6-[1(R)-hydroxyethyl]-penem-3-carboxylat
Ausgehend von p-Nitrobenzyl-(5R,6S)-2-carbamoyloxymethyl-6-[1(R)-t-butyldimethylsilyloxy-ethyl]-penem-3-carboxylat,
hergestellt gemäß Beispiel 17 und weitere Verarbeitung gemäß Beispiel 8, ergab die Titelverbindung. Die spektroskopischen
Daten werden in Beispiel 12 gezeigt.
Claims (8)
1. Verfahren zur Herstellung einer Verbindung der allgemeinen
Formel (I)
(I)
worin bedeuten
R. eine organische Gruppe,
R2 ein Wasserstoffatom oder eine Carboxylschutzgruppe
R. eine organische Gruppe,
R2 ein Wasserstoffatom oder eine Carboxylschutzgruppe
und
X
X
(i) eine gewünschtenfalls geschützte Hydroxygruppe,
(ii) ein Halogenatom,
(iii) eine Azidogruppe,
(iii) eine Azidogruppe,
(iv) eine gewünschtenfalls geschützte Pyridyl-
gruppe,
(v) eine Formyloxygruppe, eine unsubstituierte Acylgruppe mit 2 bis 6 Kohlenstoffatomen oder eine Acyloxygruppe mit 2 bis 6 Kohlenstoffatomen, die durch ein oder mehrere Halogenatome, Acylgruppen mit 2 bis 6 Kohlenstoffatomen, Amino-, Hydroxy- oder Mercaptogruppen, wobei die Amino-, Hydroxy- oder Mercaptogruppen frei oder geschützt sind, substituiert ist,
(v) eine Formyloxygruppe, eine unsubstituierte Acylgruppe mit 2 bis 6 Kohlenstoffatomen oder eine Acyloxygruppe mit 2 bis 6 Kohlenstoffatomen, die durch ein oder mehrere Halogenatome, Acylgruppen mit 2 bis 6 Kohlenstoffatomen, Amino-, Hydroxy- oder Mercaptogruppen, wobei die Amino-, Hydroxy- oder Mercaptogruppen frei oder geschützt sind, substituiert ist,

(vi) eine Carbamoyloxy- oder N-Alkyl-carbamoyloxygruppe,
(vii) eine Alkoxy- oder Alkylthiogruppe, jeweils mit 1 bis 12 Kohlenstoffatomen, die unsubstituiert
ist oder die substituiert ist durch ein oder mehrere Halogenatome, Formylgruppen, Acylgruppen
mit 2 bis 6 Kohlenstoffatomen, Amino-, Hydroxy- oder Mercaptogruppen, wobei die Amino-,
Mercapto- oder Hydroxygruppen frei sind oder geschützt sind,
(viii) eine Triarylmethylthiogruppe oder (ix) eine heterocyclische Thiogruppe
dadurch gekennzeichnet , daß man Azetidinonderivat
der allgemeinen Formel (II)

mit bis zu 2 Moläquivalenten einer dreiwertigen Organophosphorverbindung in einem inerten Lösungsmittel bei
einer Temperatur von 110 bis 1500C während 2 h bis zu

ist, wodurch man Allyl-(5R,6S)-2-t-butyldiphenylsilyloxymethyl-
-6-[1(R)-t-butyldimethylsilyloxyethyl]-penem-3-carboxylat
erhält.
8. Verfahren gemäß Anspruch 6, dadurch gekennzeichnet,
daß das Azetidinonderivat der Formel (II1)
4ß-Carbamoylglykolloylthio-3-a[1(R)-trichlorethoxycarbonyloxyethyl]-1-(acetoxymethyloxyoxalyl)-azetidin-2-on
ist, wodurch man Acetoxymethyl-(5R,6S)-2-carbamoyloxymethyl-6-[1(R)-trichlorethoxycarbonyloxyethyl]-penem-3-carboxylat
erhält.
